Abstract

A 73-year-old Japanese man was histologically diagnosed with lung adenocarcinoma harboring an exon 19 deletion in the epidermal growth factor receptor. The patient was treated with gefitinib for 6 weeks until he developed substantially elevated hepatic enzyme levels that resulted in the discontinuation of gefitinib. Gefitinib was reintroduced with an intermittent treatment schedule after the transaminase levels normalized, but the patient's enzyme levels rose again, and the cancer progressed. Gefitinib was eventually replaced with erlotinib. There was stable disease for 7 weeks without any signs of liver toxicity. Thus, erlotinib may be a beneficial and well-tolerated treatment option for patients with gefitinib-related hepatotoxicity.

Abstract

Dentin sialophosphoprotein (DSPP) is critical for proper mineralization of tooth dentin, and mutations in DSPP cause inherited dentin defects. Dentin phosphoprotein (DPP) is the C-terminal cleavage product of DSPP that binds collagen and induces intrafibrillar mineralization. We isolated DPP from individual pigs and determined that its N-terminal and C-terminal domains are glycosylated and that DPP averages 155 phosphates per molecule. Porcine DPP is unstable at low pH and high temperatures, and complexing with collagen improves its stability. Surprisingly, we observed DPP size variations on SDS-PAGE for DPP isolated from individual pigs. These variations are not caused by differences in proteolytic processing or degrees of phosphorylation or glycosylation, but rather to allelic variations in Dspp. Characterization of the DPP coding region identified 4 allelic variants. Among the 4 alleles, 27 sequence variations were identified, including 16 length polymorphisms ranging from 3 to 63 nucleotides. None of the length variations shifted the reading frame, and all localized to the highly redundant region of the DPP code. The 4 alleles encode DPP domains having 551, 575, 589, or 594 amino acids and completely explain the DPP size variations. DPP length variations are polymorphic and are not associated with dentin defects.

Abstract

The population dynamics of species interactions provides valuable information for life sciences. Lotka–Volterra equations (LVEs) are known to be the most popular model, and they are mainly applied to the systems of predation and competition. However, LVEs often fail to catch the population dynamics of mutualism; the population sizes of species increase infinitely under certain condition (divergence problem). Furthermore, LVEs never predicts the Allee effect in the systems of obligate mutualism. Instead of LVEs, several models have been presented for mutualism; unfortunately, they are rather complicated. It is, therefore, necessary to introduce a simpler theory for mutualism. In the present paper, we apply the lattice gas model which corresponds to the mean-field theory of the usual lattice model. The derived equations are cubic and contain only essential features for mutualism. In the case of obligate mutualism, the dynamics exhibits the Allee effect, and it is almost the same as in the male–female systems. In our model, the population sizes never increase infinitely, because our model contains not only intra- but also interspecific competitions. If the density of one species increases disproportionately in respect of its mutual partners, then this might imply downward pressure on the population abundance of the mutual partner species and such feedback would eventually act as a controlling influence on the population abundance of either species. We discuss several assumptions in our model; in particular, if both species can occupy in each cell simultaneously, then the interspecific competition disappears.
