157. Na+,K+-ATPase concentration in rodent and human heart and skeletal muscle:apparent relation to muscle performance

Abstract

To investigate whether the sodium-potassium pump or Na+,K+-ATPase concentration in muscles is related to the capacity for muscle performance, the 3H-ouabain binding site concentration or 3-O-methylflourescein phosphatase activity was determined in samples of heart ventricular muscles from trained rats, cardiomyopathic hamsters, and human subjects. These methods have earlier been shown to quantify the Na+,K+-ATPase concentration in muscle tissue with high accuracy. When rats were swim trained for six weeks the heart ventricular muscle Na+,K+-ATPase concentration was increased by 20% (p less than 0.02) and the heart to body weight ratio by 14% (p less than 0.005). The increase in Na+,K+-ATPase concentration was only slowly reversible. After three weeks of deconditioning an increase of 12% (p less than 0.05) was still observed. In comparison skeletal muscle Na+,K+-ATPase concentration was increased by up to 46% (p less than 0.001) and decreased by up to 30% (p less than 0.005) after training and immobilisation respectively. Cardiomyopathic hamsters showed a reduction of 33% (p less than 0.005) in the heart ventricular Na+,K+-ATPase concentration compared with normal hamsters. This decrease was associated with a heart to body weight increase of 19% (p less than 0.01) and congestive heart failure. In six patients with a mean ventricular ejection fraction of 68% the mean Na+,K+-ATPase concentration in endomyocardial biopsy specimens obtained by left heart catheterisation was 505(41) compared with 322(32) pmol.g-1 wet wt (p less than 0.001) in nine patients with a mean ejection fraction of 29%. Taken together the present data indicate a relation between the Na+,K+-ATPase concentration and the capacity for muscle performance.

158. Training increases the concentration of [3H]ouabain-binding sites in rat skeletal muscle

Abstract

Exercise is associated with a net loss of K+ from the working muscles and an increased plasma K+ concentration, indicating that the capacity for intracellular reaccumulation of K+ is exceeded. Training reduces the exercise-induced rise in plasma K+, and an increased plasma [K+] may interfere with physical performance. Since the clearing of K+ from the extracellular space depends on the capacity for active K+ uptake in skeletal muscle, the effects of training and inactivity on the total concentration of (Na+ + K+)-ATPase was determined. Following 6 weeks of swim training, the concentration of [3H]ouabain-binding sites in rat hindlimb muscles was up to 46% (P less than 0.001) higher than in those obtained from age-matched controls. Whereas muscle Na+, K+ contents remained unchanged, the concentration of citrate synthase increased by up to 76% (P less than 0.001). Training induced no change in the [3H]ouabain-binding-site concentration in the diaphragm, but in the heart ventricles, the K+-dependent 3-O-methylfluorescein phosphatase activity increased by 20% (P less than 0.001). Muscle inactivity induced by denervation, plaster immobilisation or tenotomy reduced the [3H]ouabain-binding-site concentration by 20-30% (P less than 0.02-0.001) within 1 week. In conclusion, training leads to a significant and reversible rise in the concentration of (Na+ + K+)-ATPase in muscle cells. This may be of importance for the beneficial effects on physical performance by improving the maximum capacity for K+ clearance.

Abstract

The Greenland Ice Sheet loses mass through surface meltwater runoff and discharge from marine terminating outlet glaciers. The spatial variability and magnitude of these processes have been studied and described in detail for the past decades. Here, we combine the mass loss between the LIA to 2010 with a SMB model extending back to ~1900 in order to investigate the spatial distribution of mass loss processes.We use high quality aerial stereo photogrammetric imagery recorded between 1978 and 1987 to map morphological features such as trim lines and end moraines marking the maximum ice extent of the LIA, which enables us to obtain vertical point-based differences associated with former ice extent. These point measurements are combined with contemporary ice surface differences derived using NASA's Airborne Topographic Mapper (ATM) from 2003-2010, NASA's Ice, Cloud, and land Elevation Satellite (ICESat) from 2003-2009, NASA's Land, Vegetation, and Ice Sensor (LVIS) from 2010, and ASTER (Silcast AST14DMO) co-registered to ICESat, to estimate mass loss throughout the 20th and early 21st Century. The mass balance estimates of the GrIS since retreat from maximum LIA is combined with a SMB model for the period for three intervals, LIAmax (~1900) - 1978/87, 1978/87 - 2003, and 2003 - 2010. Across the GrIS the total mass loss if found to be spatially- and temporally variable. However, when assessing the mass loss due to SMB and mass loss due to dynamic ice loss, we find that that the ratios between these components are variable between the different sectors of the GrIS, e.g. in the southeast sector of the GrIS we find substantial mass loss, possibly driven by high precipitation rates but also the presence of a large number of marine terminating glaciers. Furthermore many areas currently undergoing changes correspond to those that experienced considerable thinning throughout the 20th century. Consequently, comparing the 20th century thinning pattern to that of the last decade, and assuming a similar warming pattern, we argue that the present sensitivity distribution will hold also for future ice sheet mass loss until marine outlet glaciers become grounded.

Abstract

The Greenland Ice Sheet (GrIS) has been one of the largest contributors to global sea level rise over the last 20 years, accounting for c. 0.5 of a total of c. 3.2 mm yr-1. A significant portion of this contribution is associated with the speed up of glaciers in southeast and northwest Greenland. Here, we reveal that the Northeast Greenland Ice Stream (NEGIS), which extends more than 600 km into the interior of the ice sheet, is now undergoing dynamic thinning after more than a quarter of a century of stability. This sector of the GrIS is of particular interest in sea level projections, because the glacier flows into a large submarine basin with a negative bed slope near the grounding line.Our findings unfold the next step in mass loss of the GrIS as we show a heightened risk of rapid sustained loss from Northeast Greenland on top of the thinning in Southeast and Northwestern Greenland.

Abstract

Glaciers and ice caps peripheral to the main Greenland Ice Sheet contribute markedly to sea-level rise1,2,3. Their changes and variability, however, have been difficult to quantify on multi-decadal timescales due to an absence of long-term data4. Here, using historical aerial surveys, expedition photographs, spy satellite imagery and new remote-sensing products, we map glacier length fluctuations of approximately 350 peripheral glaciers and ice caps in East and West Greenland since 1890. Peripheral glaciers are found to have recently undergone a widespread and significant retreat at rates of 12.2 m per year and 16.6 m per year in East and West Greenland, respectively; these changes are exceeded in severity only by the early twentieth century post-Little-Ice-Age retreat. Regional changes in ice volume, as reflected by glacier length, are further shown to be related to changes in precipitation associated with the North Atlantic Oscillation (NAO), with a distinct east–west asymmetry; positive phases of the NAO increase accumulation, and thereby glacier growth, in the eastern periphery, whereas opposite effects are observed in the western periphery. Thus, with projected trends towards positive NAO in the future5,6, eastern peripheral glaciers may remain relatively stable, while western peripheral glaciers will continue to diminish.
