152. EXPRESSION OF XANTHINE OXIDASE IN TESTICULAR CELLS

Abstract

Objective : Previous studies showed that xanthine oxidase-related active oxygen generation was involved in heat stress-induced apoptosis in testicular cells. Hence, in the present study, the expressionof xanthine oxidase in experimental cryptorchidism and heat-stressed testicular cells was assessed to determine the involvement of xanthine oxidase-related active oxygen generation in heat stress-induced apoptosis in testicular cells. Methods : (1) Immunohistological examinationof xanthine oxidase was performed in a rat model of cryptorchidism. (2) mRNA expression of xanthine oxidase in cultured rat testicular cells was assessed by RT-PCR. (3) Expression of xanthine oxidase protein in cultured rat testicular cells was assessed by western blot analysis.Results : The presence of xanthine oxidase in testicular cells was confirmed by immunohistochemistry, especially after the spermatid stage. mRNA expression of xanthine oxidase in cultured testicular cells was similar with and without heat stress as determined by semiquantitative RTPCR. Expression of xanthine oxidase protein in cultured testicular cells was similar under both conditions, i.e., at 32.5˚C and at 37.0˚C, as determined by western blot analysis. Conclusions : mRNA and protein expression of xanthine oxidase was confirmed in testicular cells. Quantitative changes in xanthine oxidase expression cause by heat stress have not been clarified. These findings suggest that activate oxygen generated by xanthine oxidase is involved in apoptosis of testicular cells induced by heat stress.

153. Collecting or observation of the nutria (Myocaster coypus) on Honjima, Teshima, Oteshima, Shyodoshima and Teshima (Marugame city, or Shyozu-gun, Kagawa prefecture), Seto inland sea, Japan

Abstract

application/pdf, A total number of 35 specimens of Myocaster coypus were collected from Honjima, Teshima, and Oteshima (Marugame City), and Shyodoshima and Teshima (Shyozu-gun), Kagawa Prefecture. These islands are distributed in the Seto Inland Sea. Among 10 females, four ones were pregnant with four to ten embryos in their uterus collected in June, September, October, and November. Among 15 males, eight ones had sexually active testes collected in September, December, January and February. There has been no information of the nutria on the breeding or the living on these islands and/or Shikoku. The nutria has been killed more than 1500 individuals as harmful animals since 1998 in Okayama Prefecture. Some of these nutrias would migrate or disperse through the sea from Oakayama Prefecure, where the nutria is abundant around the mouth of Rivers Takahashi, Asahi, and Yoshii., 1) 2007年8月31日に香川県丸亀市本島でヌートリアの死亡個体(骨格のみ)が発掘された。 2) 2008年6月にヌートリアの有害鳥獣申請後, 箱ワナを用いた駆除捕獲の結果, 2009年2月までに岡山に面する香川県側のほとんどの島嶼部, すなわち小豆島, 本島, 手島, 小手島で捕獲され, 計35頭であった。また, 豊島では1頭死亡個体が目撃された。 3) 捕獲した採集月でみると2008年6月4頭, 8月3頭, 9月7頭, 10月2頭, 11月5頭, 12月3頭, 2009年1月5頭, 計35頭であった。捕獲総個体中, 雄22頭, 雌13頭(うち妊娠が確認されたのは6頭)であった。 4) 肉眼で胎児を持った妊娠雌の出現月, および副精巣尾部が管状を示し性的成熟に達している雄の出現月から推測すると, ヌートリアの繁殖活動は春と, 秋から冬にかけておこなわれていると推測される。 5) 手島では鞍部の200m x 200m程度のわずかな畑地で小さな小川しかなく, 計21頭が捕獲されたということは, 外部から絶えず侵入していることが考えられる。 6) 1994～2005年度までの「岡山県環境白書」ではほぼ1500頭以上が捕獲され, 上述の捕獲・目撃された島嶼部以外に直島での捕獲記録もあることから, これらのヌートリアは自力遊泳かあるいは何かの浮遊物を使って今回捕獲・目撃された島嶼部へたどり着いたという推測が可能である。

Abstract

A new species of the fossil murine rodent of the genus Rattus is described by comparing with the living endemic species of Diplothrix legata on the islands of Okinawa and Amamioshima. The murine fossil specimens aged as the later part of Pleistocene were excavated from the Pinza-Ave cave, the Miyako Island of the Ryukyu Archipelago, Japan in 1983., 齧歯目ネズミ科ネズミ亜科のRattus属の化石新種を記載した. 記載に当たっては沖縄島と奄美大島産の現生固有種であるケナガネズミDiplothrix legataと比較した. このネズミ亜科化石は1983年に琉球列島宮古島のピンザアブ洞窟から発掘され, 更新世後期とされるものである.

Abstract

We reported a case of recurrent hemangiopericytoma that arose primarily in the lumbar spinal canal. Treatments including preoperative embolization, tumor resection via anterior and posterior approaches, and postoperative radiation failed to completely eradicate the tumor, resulting in further recur-rences and neurological deterioration. By reviewing the 21 cases in the litera-ture along with this case, we discussed the clinical features and treatment outcome of intraspinal hemangiopericytoma.

Abstract

Three kinds of bis(aromatic fluoride) compounds, 3,5-bis(4-fluorobenzoyl)biphenyl (1), 3,5-bis(4-fluorobenzoyl)-1,1′:4′,1″-terphenyl (2), and 3,5-bis(4-fluorobenzoyl)-1,1′:4′,1″:4″,1′′′-quaterphenyl (3), were synthesized by cross-coupling of the corresponding triflates with phenylboronic acid. Linear poly(ether ketone)s (1x, 2x, and 3x) having phenyl, biphenyl, and terphenyl side groups, respectively, were prepared by the polycondensation of the bis(aromatic fluoride) compounds with bisphenol A (a) and 4,4′-dihydroxybiphenyl (b) in N-methyl-2-pyrrolidone. The obtained poly(ether ketone)s were characterized by X-ray diffraction, differential scanning calorimetry, and thermogravimetry. The structure–property relationships of these poly(ether ketone)s were examined and compared with those of poly(ether ketone)s (4x) having no side group, which were prepared from 3,5-bis(4-fluorobenzoyl)benzene (4). The properties of the poly(ether ketone)s, solubilities, and thermal properties were compared and discussed based on the length of the side groups. [ABSTRACT FROM AUTHOR]

Abstract

Aim: The aim of this study was to underscore problems associated with the dipstick test and determination of protein concentration alone in spot-urine (P-test) compared with spot-urine protein-to-creatinine ratio (P/Crtest) and to determine whether urine collection for 24-h test was complete. Material and Methods: Dipstick and P/Cr tests were performed simultaneously in 357 random spot-urine specimens from 145 pregnant women, including 35 with pre-eclampsia. Positive results were defined as 1+ on dipstick test, protein concentration 30 mg/dL on P-test, and P/Cr ratio 0.27 (mg/mg) on P/Cr test. Sixty-four 24-h urine tests (quantification of protein in urine collected during 24 h) were performed in 27 of the 145 women. We assumed that P/Cr ratio 0.27 predicted significant proteinuria (urinary protein 0.3 g/day). The 24-h urine collection was considered incomplete when urinary creatinine excretion was <11.0 mg/kg/day or >25.0 mg/kg/day. Results: Forty-four percent (69/156) of specimens with a positive test result on dipstick test contained protein < 30 mg/dL. Dipstick test was positive for 25.7% (69/269) of specimens with protein < 30 mg/dL and for 28.8% (79/274) of specimens with P/Cr ratio < 0.27. P-test results were positive for 7.3% (20/274) and negative for 18.1% (15/83) of specimens with P/Cr ratio < 0.27 and 0.27, respectively. Incomplete 24-h urine collection occurred in 15.6% (10/64) of 24-h urine tests. Daily urinary creatinine excretion was 702–1397 mg, while creatinine concentration varied from 16 mg/dL to 475 mg/dL in spot-urine specimens. Conclusion: Dipstick test and P-test were likely to over- and underestimate risks of significant proteinuria, respectively. The 24-h urine collection was often incomplete.

Abstract

Background: Complete automation of high-performance liquid chromatography (HPLC) for determination of hemoglobin F (%Hb F) and hemoglobin A1c (%Hb A1c) levels has made this procedure available in many clinical laboratories. However, the physiological changes in %Hb F during pregnancy and the effects of physiological and supraphysiological levels of %Hb A1c on measurement of %Hb F have not been studied extensively. Methods: Simultaneous determination of %Hb F and %Hb A1c was conducted in 490 blood samples obtained before (n=21), during the 1st (n=150), 2nd (n=116), and 3rd (n=192) trimesters of pregnancy, and postpartum (n=11) from 357 women, including 60 women with hyperglycemia but unaffected by clinical fetomaternal hemorrhage, by HPLC. Results: Mean (SD) Hb F levels were 0.71% (0.25%) before pregnancy. The value of 0.82% (0.47%) during the 1st trimester decreased significantly to 0.66% (0.35%) during the 2nd trimester and to 0.58% (0.38%) during the 3rd trimester. The level was 0.62% (0.31%) approximately one year after delivery. Thus, %Hb F was highest during the 1st trimester of pregnancy. The effects of varied %Hb A1c levels on %Hb F measurements were clinically negligible. Conclusions: The data presented in this work may be used as reference intervals of %Hb F determined with HPLC during pregnancy.

Abstract

Introduction: The differences in the D-dimer rise between women with singleton and multifetal pregnancies have not been studied extensively. Materials and Methods: D-Dimer levels were determined in 1089 blood specimens from 1089 women in various stages of pregnancy, including 977 and 112 women with singleton and multifetal pregnancies, respectively. None of the 1089 women developed hypertension or clinical venous thromboembolism during pregnancy or in the postpartum period. Results: The D-dimer levels were significantly and positively correlated with gestational week at examination in women with singleton or multifetal pregnancies. The D-dimer levels (mu g/ml, mean +/- SD [number of specimens]) determined at the 1st trimester did not differ significantly (0.81 +/- 0.82 [102] for singleton vs. 1.20 +/- 0.77 [7] for multifetal), but those at the 2nd (1.61 +/- 1.45 [216] vs. 2.62 +/- 2.26 [59]) and 3rd (2.37 +/- 2.22 [659] vs. 4.02 +/- 2.14 [46]) trimesters were significantly higher in women with multifetal than singleton pregnancies. The 90th percentile value was 4.31 mu g/ml for 1089 specimens. A significantly greater number of women exceeded 4.31 mu g/ml during the 2nd (16.9% vs. 5.6%, P = 0.0043) and 3rd (34.8% vs. 10.6%, P < 0.0001) trimesters among those with multifetal than with singleton pregnancies. Conclusions: The degree of D-dimer rise in pregnancy was greater in women with multifetal than with singleton pregnancies. (C) 2013 Elsevier Ltd. All rights reserved.

Neuritic abnormalities within the epithelium give more information regarding Alzheimer's disease. Random sampling using biopsy cannot indicate abnormalities as the location of the pathological changes in the superficial epithelium is unpredictable. Staining of nerve bundles in the deeper basal lamina with tau antibodies does not provide information on Alzheimer's disease.
