Abstract

A series of replacement experiments of [C-14]-triazines, [C-14]-atrazine and [7-C-14]-2-benzylamino-4-methyl-6-trifluoromethyl-1,3,5-triazine, bound to thylakoids isolated from wild-type and atrazine-resistant Chenopodium album (lambsquarters) were conducted. Replacement experiments of [C-14]-triazines bound to wild-type Chenopodium thylakoids with non-labeled atrazine and 2-benzylamino-4-methyl-6-trifluoromethyl-1,3,5-triazine were carried out, to elucidate whether benzylamino-1,3,5-triazines use the same binding niche as atrazine. [C-14]-Atrazine and [7-C-14]-2-benzylamino-4-methyl-6-trifluoromethyl-1,3,5-triazine bound to wild-type thylakoids were replaced by non-labeled 2-benzylamino-4-methyl-6-trifluoromethyl-1,3,5-triazine and non-labeled atrazine, respectively. The above two replacements showed mutual competition. To clarify further whether benzylamino-1,3,5-triazines bind at the D1-protein to amino acid residue( s) different from atrazine or not, experiments to replace [7-C-14]-2-benzylamino-4-methyl-6-trifluoromethyl-1,3,5-triazines bound to atrazine-resistant Chenopodium thylakoids by non-labeled atrazine, 2-(4-bromobenzylamino)-4-methyl-6-trifluoromethyl-1,3,5-triazine, DCMU and DNOC were carried out. Although the bound [7-C-14]-2-benzylamino-4-methyl-6-trifluoromethyl-1,3,5-triazine was difficult to be replaced even with high concentrations of atrazine, [C-14]-labeled 1,3,5-triazine was competitively replaced by non-labeled 2-(4-bromobenzylamino)-4-methyl-6-trifluoromethyl-1,3,5-triazine, DCMU or DNOC. Thus, 2-benzylamino-4-methyl-6-trifluoromethyl-1,3,5-triazine herbicides are considered to bind to the same niche at the D1 protein as atrazine, but use amino acid residue(s) different from those involved with atrazine binding.

Abstract

Calorectal adenomas are macroscopically visible morphological changes of the mucosa that can develop focal carcinoma in the absence of surgical intervention. The successive molecular changes proposed to occur at different: stages in the adenoma-carcinoma sequence were primarily based on DNA studies of exophytic; polypoid-type adenomas. Nor all colorectal lesions, however, display an exophytic phenotype and a presumed distinct colorectal neoplasm, the nonpolypoid adenoma, was subsequently described as a precursor of colorectal cancer. The low incidence of KRAS mutations in nonpolypoid colorectal adenomas reported previously suggested a different genetic basis for the transformation process in these lesions. We have pursued the identification of genetic changes in benign sporadic nanpolypoid colorectal adenomas in a selected Swedish patient group with no family history of colarectal cancer. Mutation screening of the adenomarous polyposis coli (APC), KRAS, and TP53 genes was conducted using the protein truncation test, hereroduplex-single-strand conformation polymorphism analysis, and denaturing gradient: gel electrophoresis on PCR-amplified fragments. Fourteen mutations in the APC gene were characterized in 10/20 samples. Mutations in the KRAS and TP53 genes were identified in 3/57 and 4/51 adenomas, respectively. The mutation frequencies and distribution of mutations in APC correlate with published data on exophytic adenomas. The low mutation frequency of the TP53 gene is consistent with the benign nature of the research material. KRAS activation (an early event in polypoid colorectal adenomas) apparently does not play a significant role in nonpolypoid adenoma development bur may result in the development of a polypoid configuration. Abstract

The effects of nine novel 2-benzylamino-1,3,5-triazines on photosynthetic reactions were measured in thylakoids isolated from wild-type and atrazine-resistant plants of Chenopodium album. The resistant plants have a mutation of serine for glycine at position 264 of the D1 protein. The measurement of oxygen evolution and chlorophyll a fluorescence induction indicated a 2–4-fold stronger inhibition by the 6-trifluoromethyl analogues of Photosystem II-dependent electron flow than atrazine. Analogues having a 6-methyl-, 6-monofluoromethyl or 6-difluoromethyl substitution were weak inhibitors, indicating that the 6-trifluoro group is very important for strong inhibition. All the nine novel 2-benzylamino-1,3,5-triazines were almost as active in wild-type as in atrazine-resistant thylakoids, indicating that the benzylamino substitution may be important for the lack of resistance in the atrazine-resistant plants

Abstract

The international programme "THORPEX: a World Weather Research Programme" provides a framework in which to tackle the challenge of improving the forecast skill of high-impact weather through international collaboration between academic institutions, operational forecast centres, and users of forecast products. The objectives of the THORPEX Data Assimilation and Observation Strategy Working Group (DAOS-WG) are two-fold. The primary goal is to assess the impact of observations and various targeting methods to provide guidance for observation campaigns and for the configuration of the Global Observing System. The secondary goal is to setup an optimal framework for data assimilation, including aspects such as targeted observations, satellite data, background error covariances and quality control. The Atlantic THORPEX Regional campaign, ATReC, in 2003, has been very successful technically and has provided valuable datasets to test targeting issues. Various data impact experiments have been performed, showing a small but very slightly positive impact of targeted observations. Projects of the DAOS-WG include working on the AMMA field experiment, in the context of IPY and to prepare the future THORPEX-PARC field campaign in the Pacific by comparing sensitivity of the forecasts to observations between several groups.
