157. Update on the role of alternatively activated macrophages in asthma

Alveolar macrophages (AMs) and interstitial macrophages are two different phenotypes that differentially exert immunological function under ­physiological and pathological conditions. Exposure to pathogen induces polarization of AM cells into classically activated macrophages (M1 cells) and alternatively activated macrophages (M2 cells). M1 cells dominantly express proinflammatory cytokines such as TNF-α and IL-1 β and induce lung inflammation and tissue damage. M2 cells are further divided into M2a and M2c subsets. M2a cells dominantly produce allergic cytokines IL-4 and IL-13, but M2c cells dominantly produce anti-inflammatory cytokine IL-10. M2a and M2c cells are differently involved in initiation, inflammation resolution, and tissue remodeling in the different stages of asthma. Microenvironment dynamically influences polarization of AM cells. Cytokines, chemokines, and immune-regulatory cells interplay and affect the balance between the polarization of M1 and M2 cells, subsequently influencing disease progression. Thus, modulation of AM phenotypes through molecular intervention has therapeutic potential in the treatment of asthma and other allergic inflammatory diseases. This review updated recent advances in polarization and functional specialization of these macrophage subtypes with emphasis on modulation of polarization of M2 cells in asthma of human subjects and animal models. Abstract

Surface collection, exposed sections and the use of irrigation wells and channels enabled the authors to map the settlement pattern of the elusive Dian kingdom before it became a subsidiary of the Han empire. The pattern showed that the Dian were already hierarchical, with settlements of different sizes and a political centre in which ritual bronzes featured. The empire redrew the landscape, with settlement migrating away from the wetlands into the hills where it could oversee the routes of communication into Southeast Asia.

165. Single-shot optical multiple-image encryption by jointly using wavelength multiplexing and position multiplexing

Abstract

A single shot large-capacity optical multiple-image encryption method based on wavelength multiplexing and position multiplexing is proposed. In the encryption process of the proposed method, multiple plane waves of different wavelengths are adopted to illuminate secret images that are placed at different positions along the optical axis. All the secret images are encoded into a single grayscale intensity-only image that is recorded by a monochromic camera by applying a diffractive-imaging based double random phase encoding (DRPE) system. In the decryption process, high accuracy images are decrypted without crosstalk from the intensity image through a multimode phase retrieval algorithm and a two-step iterative shrinkage/thresholding (TwIST) algorithm. The feasibility of the proposed method is demonstrated by numerical simulations.

169. Highly Stable Spherical Metallo-Capsule from a Branched Hexapodal Terpyridine and Its Self-Assembled Berry-type Nanostructure

Abstract

Discrete spherical metallo-organic capsules at the nanometer scale, especially those constructed from unique building blocks, have received significant attention recently because of their fascinating molecular aesthetics and potential applications due to their compact cavities. Here, the synthesis and characterization of a hexapodal, branched terpyridine ligand are presented along with the nearly quantitative self-assembly of the resulting tetrameric metallo-nanosphere. This metallo-nanosphere exhibited four quasi-triangular and six rhombus-like facets, all of which were made by the same hook-like bis-terpyridine. Collision-induced dissociation experiments were done to investigate overall stability. The metallo-architecture and host–guest chemistry were investigated with coronene and fully characterized by 1D and 2D NMR, ESI-MS, and transmission electron microscopy. Furthermore, this metallo-nanosphere was observed to hierarchically self-assemble into berry-type structures in an acetonitrile/methanol mixture, by virtue of counterion-mediated attractions. The functional molecular metallo-nanosphere presented here expands the reach of terpyridine coordination systems into molecular containers and other model systems.

Abstract

Safety design of high voltage part is the foundation of the whole automobile design; it is based on safety design requirements of the electric car, and the security function of hardware. The design the different functions of hardware modules are presented, through circuit principle diagram layout of PCB, and circuit board is made, the systems hardware test board is design according to the functional test of the system. Test results show that relative errors between the measurement of insulation resistance value of the safety system and the theoretical value is only 0.3%, thus the rationality of the design calculation model of the insulation resistance is verified. The response times of DC (direct current) contactor are higher than the national standard requirements, so the system security design meets the design requirements. Abstract

In this study we use difference characteristics between the transmission modes of PP and PS waves, to generate the converted 3D seismic wave, which is then applied to exploration in the of karst reservoir development zone of the northern Tarim Basin. A 3D recording geometry is designed based on the CMP and CCP bins and the stacked mutual correlation of the common geophone stations, which are controlled by PP and PS wave structure times, and are used to calculated the converted wave statics. This process improves the converted wave static correction method. The research results show that both the converted wave seismic data and the P-waves data can resolve the shoestring reflection characteristics of the dissolved cavern reservoir.

186. Pulmonary Aspergillus Overlap Syndromes.

Abstract

Background: Broadly, there are three main categories in pulmonary aspergillosis: chronic forms of aspergillosis; allergic bronchopulmonary aspergillosis; and invasive aspergillosis (IPA). IPA has been further subdivided into angioinvasive and airway-invasive aspergillosis. Aspergillus overlap syndromes is defined as the occurrence of more than one form aspergillus disease in a single individual.Objectives: To help clinicians correctly deal with AOS.Methods: Retrospectively study the clinical findings of nine patients presenting with AOS.Results: Four cases were diagnosed as angioinvasive aspergillosis complicated with ABPA, three cases as IPA overlap aspergilloma, and two cases as ABPA with AWIA. All the patients presented with cough and expectoration. In three patients with IPA overlap aspergilloma, two had hemoptysis, two had wheezing and fever. All of patients with IPA overlap ABPA had wheezing, dyspnea, and fever, three had sputum plugs, two had hemoptysis, and five patients had mucopurulent discharge and rhonchi in auscultation. Their total IgE ranged from 129 to 2124 IU/ml (676.5 ± 557.33 IU/ml). Fungal culture in sputum showed A. Fumigatus in three patients. All the six patients with IPA overlap ABPA applied steroid therapy and antifungal therapy. Three of them received two or more antifungal drugs successively, and three received combinational therapy. All the patients improved except one diagnosed ABPA overlap IPA.Conclusions: Clinical manifestation of AOS is not typical. Poor first-line therapeutic effects and complicated diagnosis criteria require clinicians to be aware of AOS when facing patients with aspergillosis. [ABSTRACT FROM AUTHOR]

Abstract

Background: SOCS3 (suppressor of cytokine signaling 3) is a negative regulator of JAK/STAT3 signaling pathway and participates in the regulation of lung inflammation in a mouse model with acute lung injury (ALI). However, it is not well understood how SOCS3 regulates lung inflammation in the ALI mouse model.Method: In the present study, we investigated the effects of SOCS3 on modulation of Ly6C(+) monocyte phenotypes in a mouse model with lipopolysaccharide (LPS)-induced ALI. Conditional SOCS3(Lyz2cre) mice with myeloid cell-restricted depletion of SOCS3 gene were created by breeding transgenic Lyz2Cre mice with SOCS3(fl/fl) mice. Wilde-type (WT) and SOCS3(Lyz2cre) mice were intratracheal instilled with 5 mg/kg LPS for 2 days. Lung, bronchoalveolar lavage (BAL) and blood were collected for analysis by flow cytometry, ELISA, qRT-PCR and Western blot analysis.Results: The studies in the ALI mouse model revealed that myeloid cell-restricted SOCS3 deficiency exacerbated the severity of ALI as compared to the WT mice. The increased severity of ALI in SOCS3-deficient mice was associated with higher populations of neutrophils, T lymphocytes and Ly6C(+) monocytes in the inflamed lung tissues. In addition, CCR2 and CXCL15 were elevated, and accompanied by greater expression and activation of STAT3 in the lung of SOCS3-deficient mice. SOCS3-deficient bone marrow-derived macrophages (BMDMs) expressed a higher amount of TNF-alpha, and adoptive transfer of the SOCS3-deficient Ly6C(+) BMDMs into WT mice enhanced the severity of ALI than adoptive transfer of WT control BMDMs. However, depletion of Ly6C(+) circulating monocytes by anti-Ly6C(+) neutralizing antibody moderately attenuated neutrophil infiltration and resulted in lower prevalence of Ly6C(+) cells in the lung of treated mice.Conclusion: Myeloid cell-restricted lack of SOCS3 induced more severe ALI through modulation of Ly6C(+) subtype macrophages. The results provide insight into a new role of SOCS3 in modulation of Ly6C(+) monocyte phenotypes and provide a novel therapeutic strategy for ALI by molecular intervention of macrophages subtypes. Abstract

Classical differential interference contrast (DIC) microscopy is an excellent tool for rendering high-contrast images to observe transparent specimen. Though the monochromatic shadow-cast image qualitatively reflects the specimen configuration, its phase can hardly be extracted, inevitably limiting its application in quantitative measurements. In order to extend its application, we propose a quantitative DIC (qDIC) microscopy technique, which can extract the specimen phase from phase-shifting beam-shearing interference images combining principal component analysis (PCA) and phase integration methods. Proved by both simulations and experiments, the PCA-based qDIC microscopy can rapidly and accurately retrieve the quantitative specimen phase. The new proposed technique shares with conventional DIC the advantage of high-contrast imaging, especially for transparent label-free specimens, but transforms DIC microscopy into a quantitative phase microscopy technique. We envision PCA-based qDIC microscopy as a future quantitative biological imaging modality.

Abstract

Intravenous (i.v.) administration of encephalitogenic peptide can effectively prevent experimental autoimmune encephalomyelitis (EAE), an animal model of multiple sclerosis; however, the underlying cellular and molecular mechanisms are not fully understood. In this study, we induced i.v. tolerance to EAE by administration of MOG35-55 peptide and determined the effect of this approach on intracellular signaling pathways of the IL-23/IL-17 system, which is essential for the pathogenesis of MS/EAE. In tolerized mice, phosphorylation of Janus kinase (Jak)/signal transducer and activator of transcription (STAT)-1, -4, ERK1/2 and NF-κBp65 were significantly reduced in splenocytes and the central nervous system. MOG i.v. treatment led to significantly lower production of IL-17, and administration of exogenous IL-17 slightly broke immune tolerance, which was associated with reduced activation of STAT4 and NF-κB. Suppressed phosphorylation of these pathway molecules was primarily evident in CD11b+ and small numbers of CD4+, CD8+ and CD11c+ cells. More importantly, adoptive transfer of CD11b+ splenocytes of tolerized mice effectively delayed onset and reduced clinical severity of actively induced EAE. This study correlates MOG i.v. tolerance with modulation of Jak/STATs signaling pathways and investigates novel therapeutic avenues for the treatment of EAE/MS.

A novel metallo-organic ligand [RuL2] with four uncomplexed coordination sites was created, and upon treatment with Zn2+ generated a dimetallic 3D metallosupramolecule in nearly quantitative yield. This self-assembly process gave stable intermediates, opening the pathway to the first 3D metallo-assembly possessing two Ru2+, two Fe2+, and two Zn2+ ions precisely located in the different edges. Abstract

ABSTRACT Background and objective The American Thoracic Society ( ATS) and European Respiratory Society ( ERS) emphasize a satisfactory start in maximal expiratory flow-volume ( MEFV) curves and highlight subjective parameters: performance without hesitation and expiration with maximum force. We described a new parameter, angle β for characterization of the start to the MEFV curve. Methods Subjects completed the MEFV curve at least three times and at least two curves met ATS/ ERS quality. Subjects were divided into normal, restrictive and obstructive groups according to pulmonary function test results. The tangent line was drawn at the start of the MEFV curve's ascending limb to the x-axis and the angle β between the tangent line and x-axis was obtained. The relationships between tangent of β, pulmonary function parameters ( PFPs) and anthropometric data were assessed. The MEFV curves with insufficient explosion at the start were considered as poor-quality MEFV curves. Results In 998 subjects with high-quality spirometry, although PFP varied in relation to the three aspects: the angle β and its tangent were similar ( P > 0.05), the tangent of β did not correlate with PFP or anthropometric measurements ( P > 0.05) and the lower limit of normal ( LLN) of the angle β was 80° in the group with high-quality spirometry ( P < 0.05). Angle β derived from poor-quality MEFV curves was smaller than that from good quality one ( P < 0.05). Conclusion Angle β may function as a parameter to assess the expiratory efforts, which can be used to assess the quality of the MEFV curve start. [ABSTRACT FROM AUTHOR]
