151. French Version of the User Mobile Application Rating Scale: Adaptation and Validation Study

Abstract: Abstract BackgroundManaging noncommunicable diseases effectively requires continuous coordination and monitoring, often facilitated by eHealth technologies like mobile health (mHealth) apps. The end-user version of the Mobile Application Rating Scale is a valuable tool for assessing the quality of mHealth apps from the user perspective. However, the absence of a French version restricts its use in French-speaking countries, where the evaluation and regulation of mHealth apps are still lacking, despite the increasing number of apps and their strong relevance in health care. ObjectiveThis study aims to translate and culturally adapt a French version of the user Mobile Application Rating Scale (uMARS-F) and to test its overall and internal reliability. MethodsCross-cultural adaptation and translation followed the universalist approach. The uMARS-F was evaluated as part through a cohort study using the French mHealth app “MonSherpa” (Qare). Participants were French-speaking adults with Apple or Android phones, excluding those with difficulty understanding French, prior app use, or physical limitations. They assessed the app using the uMARS-F twice (T1 and T2) 1 week apart. Scores for each section and overall were assessed for normal distribution using the Shapiro-Wilk test and presented as mean (SD), and potential floor or ceiling effects were calculated accordingly. Overall reliability was evaluated using intraclass correlation coefficients and internal reliability using Cronbach α. Concordance between the 3 subscales (objective quality, subjective quality, and perceived impact), 4 sections, and 26 items at T1 and T2 was evaluated using the paired t ResultsIn total, 167 participants assessed the app at both T1 and T2 (100% compliance). Among them, 49.7% (n=83) were female, and 50.3% (n=84) were male, with a mean age of 43 (SD 16) years. The uMARS-F intraclass correlation coefficients were excellent for objective quality (0.959), excellent for subjective quality (0.993), and moderate for perceived impact (0.624). Cronbach α was good for objective quality (0.881), acceptable for subjective quality (0.701), and excellent for perceived impact (0.936). The paired tPrP ConclusionsThe uMARS-F is a valid tool for end users to assess the quality of mHealth apps in French-speaking countries. The uMARS-F used in combination with the French version of the Mobile Application Rating Scale could enable health care professionals and public health authorities to identify reliable, high-quality, and valid apps for patients and should be part of French health care education programs.

152. Breast and endometrial safety of micronised progesterone versus norethisterone acetate in menopausal hormone therapy (PROBES): study protocol of a double-blind randomised controlled trial

Abstract: Introduction Data suggest that micronised progesterone (mP) in menopausal hormone therapy is safer for the breast than synthetic progestins, while protection of the endometrium appears to be less effective. However, comparative randomised trial data are lacking. The objective of the Progesterone Breast Endometrial Safety Study is to investigate breast and endometrial safety of mP versus norethisterone acetate (NETA) in continuous combination with oral oestrogen.Methods and analysis This multicentre trial, conducted at three University Hospitals in Stockholm and Uppsala, Sweden, consists of two phases: part 1 focuses on breast safety and is designed as a double-blind, randomised controlled trial. 260 postmenopausal women will be randomised to 100 mg mP or 0.5 mg NETA per day in continuous combination with 1 mg oestradiol. The primary objective is to compare the treatments with respect to percentage change in mammographic breast density after 12-month treatment. Secondary outcomes are breast proliferation, endometrial histology and proliferation, bleeding pattern, gut and vaginal microbiome, hormone levels and coagulation and metabolic factors, mood, and health-related quality of life. Part 2 features an open, single-arm design to study endometrial safety of 1-year treatment with mP in continuous combination with oestradiol on endometrial pathology (hyperplasia and cancer). We will treat 260 additional women with 100 mg mP/1 mg oestradiol resulting in an endometrial safety population of 390 women. The total number of participants in part 1 and part 2 will be 520.Ethics and dissemination The study protocol was approved by the Swedish Ethical Review Authority (2021-03033) on 29 June 2021 with amendment (2023-01480-02, protocol version 3.1) on 14 March 2023. Results of the study will be published in peer-reviewed journals and presented at scientific meetings.Trial registration number NCT05586724.

Abstract: A healthy vaginal microbiome (VMB) is dominated by Lactobacillus spp. and provides the first line of defense against invading pathogens. Vaginal dysbiosis, characterized by the loss of Lactobacillus dominance and increase of microbial diversity, has been linked to an increased risk of adverse genital tract diseases, including bacterial vaginosis, aerobic vaginitis, vulvovaginal candidiasis, sexually transmitted infections, and pregnancy complications such as preterm birth. Currently, antibiotics and antifungals are recommended first-line treatments with high cure rates, but they also can lead to high recurrence and resistance development. As an alternative, lactobacilli have been utilized to restore the vaginal microbiota. In this review article, we discuss vaginal dysbiosis in various gynecological infections and potential interventions based on Live Biotherapeutic Products (LBPs) with a focus on those that use intravaginal treatment modalities to modulate the VMB. Based on these, we provide insights on key factors to consider in designing phenotypic and genotypic screens for selecting bacterial strains for use as vaginally administered microbiome-directed therapeutics. Lastly, to highlight current progress within this field, we provide an overview of LBPs currently being developed with published clinical trial completion for recurrent BV, VVC, and UTI. We also discuss regulatory challenges in the drug development process to harmonize future research efforts in VMB therapy.

154. Influence of rearing method on the behaviour and welfare of Assaf lambs during the neonatal period

Abstract: The present study aimed to determine the influence of rearing method on behaviour and welfare in Assaf lambs during the first two weeks of the neonatal period, in an intensive rearing system. The experiment was conducted with 24 Assaf lambs, in which two methods of management were applied – natural rearing (NR) and artificial rearing (AR). Forced separation of lambs from their mothers caused 74% fewer suckling attempts during the first three days of the neonatal period, which reduced suckling time by 57%. The lambs reared under NR, spent more time moving and less time standing and lying. Overall, it was found that the type of rearing affects the suckling time. Artificial rearing decreased lying and playing time, which is indicative of the improved welfare of NR lambs. The two rearing methods (NR and AR) did not affect blood total protein or albumin, indicating no significant effect of different rearing methods on these haematological variables. Artificial rearing does not disturb the welfare and behaviour of the animals when it is done correctly and stress appears transient and animals quickly adapt with similar growth rates.

Abstract: Trichoptera larvae and adults were collected as part of an overall assessment of aquatic macroinvertebrate diversity in the major headwater tributaries of the Okavango and Zambezi River basins in Angola. This report documents one of the National-Geographic-funded, Okavango-Wilderness-Project expeditions undertaken between October and November 2016. A total of 21 sites on the Cuanavale, Cuito, Cuando, and Cuembo Rivers and some of their tributaries were sampled. Biotopes sampled in source lakes, headwater streams, pools, and large rivers included mostly sandy substrates, aquatic emergent and submerged vegetation, marginal vegetation and (rarely) bedrock, stones, or gravel. Light trap collecting was possible at only seven sites. With both adults and larvae collected, at least 55 species in seven families (Philopotamidae, Dipseudopsidae, Ecnomidae, Hydropsychidae, Hydroptilidae, Leptoceridae, and Sericostomatidae) were recorded. The Leptoceridae were the most abundant family, represented by ten genera and 22 species. There are a number of unnamed species which will be described in a separate paper.

Abstract: Objective: To investigate the prevalence and factors associated with no intention to exclusively breastfeed for the first 6 months of life in a sample of women in the first 24 h postpartum during the hospital stay. Methods: Cross-sectional study with data from screening phase of a birth cohort. The proportion of mothers who did not intend to breastfeed exclusively for 6 months (primary outcome) derived from a negative response to the question “Would you be willing to try to breastfeed exclusively for the first 6 months?”, in an interview conducted by previously trained interviewers. Crude and adjusted prevalence ratios (PR) with 95% confidence intervals were obtained by Poisson regression with robust variance. Results: A total of 2964 postpartum women were interviewed. The overall prevalence of mothers who did not intend to breastfeed exclusively for 6 months was 17.8% (16.4–19.1%). After adjusting for maternal age and type of pregnancy (singleton or multiple), no intention to exclusively breastfeed was higher in mothers with a monthly household income < 3 minimum wages (PR, 1.64; 1.35–1.98) and in those who intended to smoke 4–7 days/week after delivery (PR, 1.42; 1.11–1.83). The presence of significant newborn morbidity (PR, 0.32; 0.19–0.54) and intention to breastfeed up to 12 months (PR, 0.46; 0.38–0.55) had a protective effect against not intending to breastfeed exclusively for 6 months. Conclusions: Approximately 1 in every 5 mothers did not intend to breastfeed exclusively for 6 months. Strategies aimed at promoting exclusive breastfeeding should focus attention on mothers from lower economic strata and smokers.

159. Comparison of the forms of carbohydrate metabolism in children with cystic fibrosis: A review

Abstract: Background. Forms of glucose metabolism disorders are one of the key processes in the course of cystic fibrosis (CF) and can be variable, starting with impaired glucose tolerance (IGT) and ending with cystic fibrosis-related diabetes (CFRD), which is the most dangerous complication, worsening the prognosis and outcome of disease. Decreased lung function and poor nutritional status are the leading causes of morbidity and mortality in patients with carbohydrate metabolism disorders (CMD). Materials and methods. The study included 85 children with CF. Patients underwent measurement of anthropometric parameters (height, weight) to assess nutritional status (NS), and spirometry test was used to assess pulmonary function (PF). Of these, 78 patients were assessed with an oral glucose tolerance test (OGTT) at 5 points (0 minutes, 30 minutes, 60 minutes, 90 minutes, 120 minutes) assessing the concentration of venous plasma glucose in mmol/l. The results of the OGTT were assessed in accordance with the diagnostic criteria for CFRD. Results. According to the Kruskal–Wallis test, the maximum volumetric flow rate during exhalation of 75% FVC (FEF 75) differed between different forms of carbohydrate metabolism (p=0.031). Other indicators of respiratory function didn’t have any significant differences. Using the Mann–Whitney test, pairwise comparisons were performed, so the FEF 75 indicator was statistically significantly lower in the group of children with CFRD than in other groups. There were no statistically significant differences when assessing Z-scores weight/age (WAZ) and height/age (HAZ) depending on the forms of carbohydrate metabolism. However, BMI/age Z-scores (BAZ) were statistically significantly lower in groups of children with CFRD than in other forms of carbohydrate metabolism. Conclusion. Children with CF in the period of prediabetes don’t have significant impairments in lung function and NS. These disorders are typical for children with CFRD.

Abstract: Abstract We propose a novel two-scale (meso-macro-scale) approach to computationally capture cancellous bone remodelling allowing for efficient and effective numerical implementation. Therein, the macro-scale is governed by the well-established kinematics and kinetics of one-scale continuum bone remodelling. However, the constitutive behaviour is not postulated phenomenologically at the macro-scale, but rather follows from the meso-scale. There, for the sake of computational efficiency, the trabecular architecture is idealised as a truss network with the cross-sectional area of the trabeculae adapting to mechanical loading. Then, the meso- and the macro-scale are coupled through up- and down-scaling. Computational results on benchmark problems from bio-mechanics demonstrate that the proposed two-scale approach is effective from a modelling perspective and efficient from a computational perspective. In particular, it automatically captures anisotropy resulting from the irregular trabecular architecture at the meso-scale, and, most importantly, enables the direct investigation of different trabecular structures at the meso-scale, thereby serving as a virtual “magnifiying glass”. As an outlook, the proposed two-scale approach to cancellous bone remodelling provides an excellent launch pad for further extension, e.g., by considering more complex trabecular architectures and/or through inclusion of micro-scale bone cellular activities.

161. Compositional and functional differences of the vaginal microbiota of women with and without cervical dysplasia

Abstract: Abstract Alterations in the vaginal microbiota, including both species composition and functional pathways, have been associated with HPV infection and progression of dysplasia to cervical cancer. To further explore this, shotgun metagenomic sequencing was used to taxonomically and functionally characterize the vaginal microbiota of women with and without cervical dysplasia. Women with histologically verified dysplasia (n = 177; low grade dysplasia (LSIL) n = 81, high-grade dysplasia (HSIL) n = 94, cancer n = 2) were compared with healthy controls recruited from the cervical screening programme (n = 177). Women with dysplasia had a higher vaginal microbial diversity, and higher abundances of Gardnerella vaginalis, Aerococcus christensenii, Peptoniphilus lacrimalis and Fannyhessea vaginae, while healthy controls had higher relative abundance of Lactobacillus crispatus. Genes involved in e.g. nucleotide biosynthesis and peptidoglycan biosynthesis were more abundant in women with dysplasia. Healthy controls showed higher abundance of genes important for e.g. amino acid biosynthesis, (especially L-lysine) and sugar degradation. These findings suggest that the microbiota may have a role in creating a pro-oncogenic environment in women with dysplasia. Its role and potential interactions with other components in the microenvironment deserve further exploration.

162. Establishment of Reference Value of 20 Amino Acids for Toddlers by High Performance Liquid Chromatography Tandem Mass Spectrometry

Abstract: BACKGROUND: Amino acids are one of the essential metabolites, especially the 20 amino acids that are preserved as the building blocks of protein. Alterations in amino acid concentrations are related to disease such as inborn error of metabolism, cancer, as well as nutritional status. Hence, it is necessary to define reference values of 20 plasma-free amino acids for Indonesian toddlers and to establish a robust measurement technique using chromatography with tandem mass spectrometry (MS). METHODS: This study was a cross-sectional preliminary study to establish reference values. The sample was prepared by mixing plasma with 20% sulfosalicylic acid. Plasma-free amino acids were measured with high-performance liquid chromatography (HPLC) non-derivatization technique using column XTerra for chromatographic separation coupled with tandem MS. Amino acids reference values were taken from 101 healthy Indonesian toddlers aged 1-3 years old. Since amino acids data were not Gaussian distributed, the lower and upper of the reference value was established from the 5th percentile and the 95th percentile, respectively. RESULTS: Analysis for 20 amino acids was validated. The accuracy ranged from 90.53-105.39% and the precision ranged from 0.06-3.80%. The limit of detection range was 1-2 nmol/mL, and the limit of quantification range was 2-4 nmol/mL. The result was linear, with R2 higher than 0.998. There was no significant difference between boys and girls for all amino acids except for glycine. CONCLUSION: HPLC with tandem MS method can be used to evaluate amino acids in clinical practice. The reference values obtained are specific for aged 1-3 years old from urban areas in Indonesia. The study suggests that for each population, the reference values for amino acids should be established. 163. Enhancing astaxanthin biosynthesis and pathway expansion towards glycosylated C40 carotenoids by Corynebacterium glutamicum

Abstract: Abstract Astaxanthin, a versatile C40 carotenoid prized for its applications in food, cosmetics, and health, is a bright red pigment with powerful antioxidant properties. To enhance astaxanthin production in Corynebacterium glutamicum, we employed rational pathway engineering strategies, focused on improving precursor availability and optimizing terminal oxy-functionalized C40 carotenoid biosynthesis. Our efforts resulted in an increased astaxanthin precursor supply with 1.5-fold higher β-carotene production with strain BETA6 (18 mg g−1 CDW). Further advancements in astaxanthin production were made by fine-tuning the expression of the β-carotene hydroxylase gene crtZ and β-carotene ketolase gene crtW, yielding a nearly fivefold increase in astaxanthin (strain ASTA**), with astaxanthin constituting 72% of total carotenoids. ASTA** was successfully transferred to a 2 L fed-batch fermentation with an enhanced titer of 103 mg L−1 astaxanthin with a volumetric productivity of 1.5 mg L−1 h−1. Based on this strain a pathway expansion was achieved towards glycosylated C40 carotenoids under heterologous expression of the glycosyltransferase gene crtX. To the best of our knowledge, this is the first time astaxanthin-β-d-diglucoside was produced with C. glutamicum achieving high titers of microbial C40 glucosides of 39 mg L−1. This study showcases the potential of pathway engineering to unlock novel C40 carotenoid variants for diverse industrial applications.

164. Development of Food Multi-Mix Using a Linear Programming Approach to Fill the Nutrient Gap of Amino Acids and Micronutrients for Stunted Non-Wasted Children.

Abstract: Food-based approaches using locally available food escalates the feasibility and the sustainability of nutrition intervention. A complementary feeding recommendation (CFR) integrated with the food multi-mix (FMM) formulation was targeted to fulfill micronutrient and amino acid requirements for stunted non-wasted (SNW) children aged 12–23 months living in agricultural areas. A seven-day estimated food record (EFR) of 87 children was used to design the CFR and 4 identified underutilized foods were integrated as the FMM. A linear programming approach using Optifood was applied to optimize the CFR and FMM. CFR alone successfully fulfills the vitamin C, riboflavin, iron, and zinc, but it cannot fulfill calcium, thiamin, niacin, vitamin B6, folate, vitamin B12, and histidine. With the incorporation of the selected underutilized cowpea, buncis batik, wader fish, and cows' milk in the FMM development, the nutrients that are challenging in CFR development, can be fulfilled. Therefore, these findings present evidence that food multi-mix developed based on locally available nutrient-dense food sources can help to meet the nutrient gaps, which often remained even after a complementary feeding diet is optimized. Efficacy study using the developed CFR and FMM is recommended to assess effect in improving intake of micronutrients and amino acids and improving the linear growth of stunted, non-wasted children. Abstract: Understanding the biological changes that occur prior to onset of late-life depression (LLD) is key to its prevention. To investigate potential predictors of LLD, we assessed cognitive scores and neurodegenerative and vascular biomarkers in healthy older adults who later developed depression.Longitudinal data from the Alzheimer's Disease Neuroimaging Initiative of 241 cognitively unimpaired and non-depressed older adults aged 56-90 at baseline with at least 4 years of follow-up were included. Participants were classified based on whether they developed an incident depression (n = 96) or not (n = 145). Cognitive measures of memory, executive functioning, and language, and biomarkers proposed to be related to LLD: hippocampal volume, white matter hyperintensity volume (WMH), and cortical and cerebrospinal fluid (CSF) amyloid beta levels, were compared between the incident depression and the never-depressed groups at four time points: at baseline, the visit prior to onset, at onset, and after the onset of depression.In the incident depression group, there was a mild decline in cognitive scores from baseline to the visit before depression onset compared with the never-depressed group. The cognitive differences between the groups became more marked after depression onset. Baseline cortical amyloid burden, CSF amyloid beta levels, and WMH were significant predictors of incident depression. Compared to the non-depressed group, hippocampal volume was not reduced before onset, but was reduced following depression.Amyloid pathology and WMH can predict future development of LLD in cognitively unimpaired individuals and may be involved in precipitating vulnerability for depression in older adults.

Abstract: OBJECTIVES: The relationship between gut microbiota and growth during infancy has been substantiated, but whether this stands true in later childhood is under investigation. We used a new clustering methodology that improves interpretability of later childhood growth patterns. Three distinct patterns were identified (Increasing, Stable, Declining) for BMI-for-age (BMIZ) and height-for-age Z-scores (HAZ) in a birth cohort of 4231 Brazilian children. Here, we investigate the association between these growth patterns and gut microbiota diversity in late childhood. METHODS: We sampled 337 children from the 2004 Pelotas birth cohort (Brazil) grouped into Increasing (n = 78; 68), Stable (n = 161; 170), or Declining (n = 98; 99) patterns for BMIZ and HAZ, respectively. Feces and anthropometrics were collected at age 13 years. The fecal microbiota was analyzed via Illumina MiSeq (16S rRNA V3-V4 region) and the UNOISE pipeline. 169. Influence of Gestational Age Correction and Shape-Based Trajectories in the Identification of Children's Growth Patterns From Birth to 11 Years

Abstract: OBJECTIVES: Childhood growth reflects nutritional status such as under- or overnutrition and is a determinant of life health. Thus, early detection of sub-optimal growth patterns can help identify modifiable risk factors. Current approaches rely on the trajectory intercept and struggle to capture differences in trajectory shape, which better reflects change over time. This study implemented a novel approach to identify shape-based growth patterns in children from birth to 11 years of age. METHODS: We used anthropometric measures from 3,945 children at postnatal age 3 months, 1, 2, 4, 6 and 11 years from the 2004 Pelotas Birth Cohort (Brazil). Weight-, height- and BMI-for-age Z-scores were calculated using INTERGROWTH21st and WHO growth standards. The impact of gestational age-correction (CA) in preterm-born children (n = 461) was tested, and trajectories were either baseline anchored at zero or not. This produced 4 analytical pipelines for pattern detection (k-means, Frechet's distance): 1) no CA–no baseline anchoring; 2) no CA–with baseline anchoring; 3) with CA–no baseline anchoring; 4) with CA–with baseline anchoring. RESULTS: We found 3 patterns for each Z-score, with “High” (increasing), “Middle” (stable) and “Low” (declining) trajectories. The use of both CA and baseline anchoring highlighted sub-groups of children with specific patterns related to early growth (first 3 months of life). In the non-anchored approach, CA influenced the mean trajectories detected by increasing the intercept and reducing the slope between birth and 3 months. Baseline anchoring identified mean trajectories with steeper slopes during this period, thus, capturing differential growth patterns in early life. CONCLUSIONS: Depending on the question, incorporating CA with or without baseline anchoring when analyzing growth patterns can better highlight aspects of early growth versus overall patterns across childhood and can improve growth analyses standardization in population-based studies. Abstract: Previous studies have proven that red hybrid tilapia (Oreochromis sp.) vaccinated with a feed-based bivalent vaccine incorporating the formalin-killed whole organisms Streptococcus agalactiae and Aeromonas hydrophila mixed with 10 % palm oil showed good protection against streptococcosis and aeromoniasis diseases. However, the molecular mechanisms related to the induction of fish's immunological responses after vaccination are poorly investigated. Therefore, a transcriptomic study using the hindgut of red hybrid tilapia after vaccination was conducted, as the gut plays a role in antigen uptake and nutrient absorption. The transcriptome dataset has the potential to provide an understanding of the early induction of immunological responses in red hybrid tilapia after vaccination. 172. Tick-borne piroplasms and trypanosomes incidentally detected in eastern grey kangaroos (Macropus giganteus) during a mortality and morbidity event in southern New South Wales, Australia

Abstract: Tick-borne haemoparasites, including piroplasms and trypanosomes, are almost ubiquitous in Australian wildlife, with some associated with health impacts to individual animals and declining wildlife populations. An array of ecologically distinct piroplasm and trypanosome species occur throughout Australia although many of these species and their sylvatic ecologies are poorly characterised. Between May 2022 and October 2023, an anecdotally reported localised eastern grey kangaroo (Macropus giganteus) morbidity/mortality event occurred in coastal southern New South Wales, Australia, characterised by animals presenting with blindness, emaciation, lethargy, ataxia, and astasia. Here we used molecular techniques to identify tick-borne piroplasms (Babesia and Theileria) and trypanosomes in affected animals. Blood (n = 89) and liver (n = 19) samples were collected after the humane euthanasia of wild animals due to welfare concerns, and brief notes on the animal's health were recorded. In total, 20 (22.5%) animals were infected with tick-borne haemoparasites, including a novel Theileria sp. nov. (14, 15.7%), Babesia macropus (2, 2.2%), Trypanosoma gilletti (5, 5.6%), and Trypanosoma vegrandis (1, 1.1%). Liver samples were also screened for Wallal and Warego viruses due to animals' blindness, but were negative. This is the first report of T. gilletti and T. vegrandis in eastern grey kangaroos, although they have been previously reported in high numbers in ticks which commonly parasites this host. The novel Theileria sp. was previously reported in questing Ixodes holocyclus and in ticks from an opportunistically collected eastern grey kangaroo and red-necked wallaby (Notamacropus rufogriseus). However, we show for the first time this Theileria sp. can occur widely in eastern grey kangaroos. Ultimately, this small study did not intend, and is not able to draw inference regarding the pathogenicity of these haemoparasites to eastern grey kangaroos and it is likely that other factors, such as chronic Phalaris grass toxicity, had a role in this localised mortality/morbidity event.

177. Operationalizing a Definition: Rhode Island's Learning Disabilities Identification Process.

Abstract: To reduce Rhode Island's high percentage of students being classified as learning disabled, an advisory committee recommended development of a statewide Learning Disabilities Identification Process (L.D.I.D.), to prepare evaluation teams to consistently consider regulatory criteria and to justify their decisions when making learning disability determinations. L.D.I.D. helps teams share and analyze information, focus on the severe discrepancy criterion, keep distractors out of the way, and maintain team professional decision-making responsibility. The approach features analysis and recording of information to concentrate on ability and achievement, and appropriate use of a statistical comparison of scores. Evaluation team members are informed of appropriate evaluation practices through district workshops, a network of contact persons, and a newsletter. The process is being implemented in over 90% of Rhode Island's school districts. A Guideline Chart, which indicates how the student's performance compares to a benchmark level of 1.5 standard deviations of difference on ability and achievement test scores, is used as a legitimate tool by teams but does not dominate decision making. Rhode Island's learning disability incidence figures are declining. Seven references and three related documents are listed. (JDD)

No. of bitstreams: 0 Previous issue date: 2021-03-09 Wellcome Trust From 1985 to 2016, the prevalence of underweight decreased, and that of obesity and severe obesity increased, in most regions, with significant variation in the magnitude of these changes across regions. We investigated how much change in mean body mass index (BMI) explains changes in the prevalence of underweight, obesity, and severe obesity in different regions using data from 2896 population-based studies with 187 million participants. Changes in the prevalence of underweight and total obesity, and to a lesser extent severe obesity, are largely driven by shifts in the distribution of BMI, with smaller contributions from changes in the shape of the distribution. In East and Southeast Asia and sub-Saharan Africa, the underweight tail of the BMI distribution was left behind as the distribution shifted. There is a need for policies that address all forms of malnutrition by making healthy foods accessible and affordable, while restricting unhealthy foods through fiscal and regulatory restrictions. 179. Associated factors for accelerated growth in childhood: a systematic review

Abstract: Several studies have shown that accelerated growth in the postnatal period is critical for the development of chronic diseases. The term catch-up has been used for the accelerated growth of children who have suffered some sort of restriction of nutrition or oxygen supply. However, accelerated growth has been observed among children who have an appropriate birth weight for their gestational age (AGA) and with no apparent morbidity. Therefore, this systematic review was carried out on the associated factors of accelerated growth, or catch-up, using the Medline/ Pubmed database. Only cohort studies written in Portuguese, English or Spanish, with children between zero and 12 years old who presented accelerated growth or catch-up as the outcome were included. Out of the 2,155 articles found, 9 were selected. There is no uniformity in the operational definition of accelerated growth, or in the concept of catchup. According to this review, accelerated growth is associated with primiparity, maternal smoking during pregnancy, lower birth weight, and early weaning. The main limitations in the available literature are the high number of follow-up losses and the lack of control for confounding factors. The determinants of accelerated growth still need to be studied further, especially among AGA children. 180. Functional traits in soil-living oribatid mites unveil trophic reorganization in belowground communities by introduced tree species

Abstract: Biodiversity loss and its potential threat on ecosystem functions call for a critical evaluation of human impacts on forest ecosystems. Management practices based on stand diversification offer a possible solution to biodiversity loss due to monoculture plantations, and these practices often involve planting introduced tree species. Although introduced non-native tree species may provide high economic returns, they may also form novel ecosystems and threaten local biodiversity, but this has been little studied. Here, we combined a taxonomic and trait-based approach and investigated communities of oribatid mites (Oribatida, Acari) across forest types of both native and introduced tree species in Northern Germany. Both trophic and life-history traits of oribatid mites were evaluated using native European beech (Fagus sylvatica) as reference, compared to native Norway spruce (Picea abies), introduced Douglas fir (Pseudotsuga menziesii) and beech-conifer mixtures. The abundance and diversity of oribatid mites were generally similar among monocultures of European beech, Norway spruce and Douglas fir. By contrast, species and trait compositions of oribatid mite communities were shifted to include more primary decomposers and more surface-living oribatid mites in Douglas fir, resulting in a trophic reorganization with less predators than in European beech forests. These results suggest that oribatid mites maintain a similar level of trait diversity regardless of forest type, but the changes in trophic guild composition and vertical distribution indicate greater availability of litter-based resources in Douglas fir than in European beech forests. The similar abundance and diversity of oribatid mite communities in Douglas fir mixed stands as in native European beech forests points to mixed forests as a promising management option for future forestry. Overall, our trait-based analyses provided insights into the changes of soil biota composition, revealing the impact of introduced tree species on the structure and functions of soil animal communities.

181. The first finding of Dictyocaulus cervi and Dictyocaulus skrjabini (Nematoda) in feral fallow deer (Dama dama) in Australia

Abstract: Feral deer are widespread throughout Australia with the capacity to impact livestock production via transmission of parasites. Samples of Dama dama (fallow deer), Rusa unicolor (sambar deer), Cervus elaphus (red deer) and an unidentified deer were sourced from various locations in south-eastern Australia for examination for parasites. Adult nematodes were collected from the lungs of all deer species across four separate geographical locations. The nematodes were identified as species of Dictyocaulus through both morphological and molecular means. Species identification based on morphological features was difficult, with many measurements from described species overlapping. Molecular analyses targeting three markers, namely 18S rRNA, ITS2, and cox1 revealed the presence of two distinct species: Dictyocaulus cervi and Dictyocaulus skrjabini. These are the first genetically confirmed reports of species of Dictyocaulus in feral deer in Australia, and although cross-transmission of species of Dictyocaulus with livestock has not yet been reported, it cannot be completely discounted without further research.

182. Applicability and perspectives for DNA barcoding of soil invertebrates

Abstract: Belowground invertebrate communities are dominated by species-rich and very small microarthropods that require long handling times and high taxonomic expertise for species determination. Molecular based methods like metabarcoding circumvent the morphological determination process by assigning taxa bioinformatically based on sequence information. The potential to analyse diverse and cryptic communities in short time at high taxonomic resolution is promising. However, metabarcoding studies revealed that taxonomic assignment below family-level in Collembola (Hexapoda) and Oribatida (Acariformes) is difficult and often fails. These are the most abundant and species-rich soil-living microarthropods, and the application of molecular-based, automated species determination would be most beneficial in these taxa. In this study, we analysed the presence of a barcoding gap in the standard barcoding gene cytochrome oxidase I (COI) in Collembola and Oribatida. The barcoding gap describes a significant difference between intra- and interspecific genetic distances among taxa and is essential for bioinformatic taxa assignment. We collected COI sequences of Collembola and Oribatida from BOLD and NCBI and focused on species with a wide geographic sampling to capture the range of their intraspecific variance. Our results show that intra- and interspecific genetic distances in COI overlapped in most species, impeding accurate assignment. When a barcoding gap was present, it exceeded the standard threshold of 3% intraspecific distances and also differed between species. Automatic specimen assignments also showed that most species comprised of multiple genetic lineages that caused ambiguous taxon assignments in distance-based methods. Character-based taxonomic assignment using phylogenetic trees and monophyletic clades as criteria worked for some species of Oribatida but failed completely for Collembola. Notably, parthenogenetic species showed lower genetic variance in COI and more accurate species assignment than sexual species. The different patterns in genetic diversity among species suggest that the different degrees of genetic variance result from deep evolutionary distances. This indicates that a single genetic threshold, or a single standard gene, will probably not be sufficient for the molecular species identification of many Collembola and Oribatida taxa. Our results also show that haplotype diversity in some of the investigated taxa was not even nearly covered, but coverage was better for Collembola than for Oribatida. Additional use of secondary barcoding genes and long-read sequencing of marker genes can improve metabarcoding studies. We also recommend the construction of pan-genomes and pan-barcodes of species lacking a barcoding gap. This will allow both to identify species boundaries, and to cover the full range of variability in the marker genes, making molecular identification also possible for species with highly diverse barcode sequences.

183. Changes of adolescent sleep patterns during the COVID-19 pandemic

Abstract: ABSTRACT OBJECTIVE The COVID-19 pandemic has raised numerous concerns regarding its effects on individuals’ health and lifestyle. We aim to analyze potential changes in adolescent sleep patterns from before and during the pandemic and identify specific predictors of changes. METHODS A subgroup of adolescents from a population-based birth cohort from Pelotas, Brazil, was assessed pre-pandemic (T1, November-2019 to March-2020) and peri-pandemic (T2, August-2021 to December-2021) in in-person interviews (n = 1,949). Sleep parameters, including sleep duration and latency time on workdays and free days, as well as social jetlag (SJL), were assessed using the Munich ChronoType Questionnaire (MCTQ). Socio-demographic, pre-pandemic, and pandemic-related predictors were analyzed. Changes in sleep parameters from T1 to T2 were estimated by multivariate latent change score modeling. RESULTS The latent change factor shows a significant mean increase in workday sleep duration (M = 0.334, p < 0.001), workday sleep latency (M = 0.029, p = 0.002), and free day sleep latency (M = 0.021, p = 0.034), and a decreased in SJL (M = −0.758, p < 0.001) during the pandemic. Female adolescents presented higher increases in workday sleep duration. Adolescents who adopted a stricter social distancing level during the pandemic presented greater increases in workday sleep duration and smaller reductions in SJL. Self-evaluated insomnia during the pandemic predicted lower increases in workday and free day sleep duration and higher increases in workday and free day sleep latency. CONCLUSION The COVID-19 outbreak brought certain advantages regarding increased sleep duration and reduced SJL. However, the observed increase in sleep latency and the influence of self-reported insomnia could be related to psychological distress inherent to the pandemic.

Abstract: BackgroundDespite mounting evidence of gut-brain involvement in psychiatric conditions, functional data remain limited, and analyses of other microbial niches, such as the vaginal microbiota, are lacking in relation to mental health. This aim of this study was to investigate if the connections between the gut microbiome and mental health observed in populations with a clinical diagnosis of mental illness extend to healthy women experiencing stress and depressive symptoms. Additionally, this study examined the functional pathways of the gut microbiota according to the levels of psychological symptoms. Furthermore, the study aimed to explore potential correlations between the vaginal microbiome and mental health parameters in young women without psychiatric diagnoses.MethodsIn this cross-sectional study, 160 healthy Danish women (aged 18-40 years) filled out questionnaires with validated scales measuring symptoms of stress and depression and frequency of dietary intake. Fecal and vaginal microbiota samples were collected at the beginning of the menstrual cycle and vaginal samples were also collected at cycle day 8-12 and 18-22. Shotgun metagenomic profiling of the gut and vaginal microbiome was performed. The Kyoto Encyclopedia of Genes and Genomes (KEGG) was used for functional profiling and 56 Gut Brain Modules were analyzed in the fecal samples.ResultsThe relative abundance in the gut of the genera Escherichia, Parabacteroides, and Shigella was higher in women with elevated depressive symptoms. Women with high perceived stress showed a tendency of increased abundance of Escherichia, Shigella, and Blautia. Amongst others, the potentially pathogenic genera, Escherichia and Shigella correlate with alterations in the neuroactive pathways such as the glutamatergic, GABAeric, dopaminergic, and Kynurenine pathways. Vaginosis symptoms were more prevalent in women reporting high levels of stress and depressive symptoms.ConclusionsThe findings of this study support the concept of a microbiota-associated effect on the neuroactive pathways even in healthy young women. This suggest, that targeting the gut microbiome could be a promising approach for future psychiatric interventions.

Abstract: The microalgae-bacteria interaction in enriched culture media could affect the growth and nutritional properties of Artemia. The addition of carbon sources to the media improves the heterotrophic bacterial population, which in turn interacts with microalgae to improve the health and production of Artemia. The aim of this study was to assess the impacts of the microbial consortium developed with different fermented carbon sources and microalgae on Artemia. Three different microalgae, Chlorella sp./A1, Nannochloropsis oceanica/A2 and Chaetoceros calcitrans/A3 were mixed with three fermented carbon sources (rice bran/C1, tapioca flour/C2 and molasses/C3) to produce nine treatments (A1C1, A1C2, A1C3, A2C1, A2C2, A2C3, A3C1, A3C2, A3C3) and three controls with microalgae alone (A1, A2, A3). Enrichment was carried out for 24 h with Artemia instar I at an initial density of 25 individuals mL-1. Two-way ANOVA revealed that the survival, length, protein and lipid contents of Artemia nauplii were significantly affected (P < 0.05) by carbon source and species of microalgae. The carbon source–microalgae interaction also significantly affects Artemia survival, length, biomass and protein content. This study illustrated that bacteria associated with different carbon sources and microalgae consortia improved Artemia growth, survival, protein and lipid content.

Abstract: Abstract Objectives This study aimed to assess the effectiveness of a continuous quality improvement initiative at the University of Florida Health Physicians practice in reducing the time to administer factor replacement therapy (FRT) for hemophilia patients presenting with bleeding in the emergency department (ED). Methods The study, a quasi‐experimental, interventional design, was conducted between January 2020 and January 2023. The intervention, implemented in September 2021, involved training ED physicians, creating a specialized medication order set within the electronic health record (EHR), and a rapid triage system. The effectiveness was measured by comparing the time from ED arrival to factor administration before and after the intervention and benchmarking it against the National Bleeding Disorders Foundation's Medical and Scientific Advisory Council (MASAC)‐recommended 1‐hour timeline for factor administration. An interrupted time series (ITS) analysis with a generalized least squares model assessed the intervention's impact. Results A total of 43 ED visits (22 pre‐intervention and 21 post‐intervention) were recorded. Post‐intervention, the average time from ED arrival to factor administration decreased from 5.63 to 3.15 hours. There was no significant increase (27% vs. 29%) in the patients receiving factor within 1‐hour of ED arrival. The ITS analysis predicted a 20‐hour reduction in the average quarterly time to administer factor by the end of the study, an 84% decrease. Conclusions The quality improvement program decreased the time to administer FRT for patients with hemophilia in the ED. However, the majority of patients did not achieve the 1‐hour MASAC‐recommended timeline for factor administration after ED arrival.

Abstract: Background Hypertension can be detected at the primary health-care level and low-cost treatments can effectively control hypertension. We aimed to measure the prevalence of hypertension and progress in its detection, treatment, and control from 1990 to 2019 for 200 countries and territories. Methods We used data from 1990 to 2019 on people aged 30-79 years from population-representative studies with measurement of blood pressure and data on blood pressure treatment. We defined hypertension as having systolic blood pressure 140 mm Hg or greater, diastolic blood pressure 90 mm Hg or greater, or taking medication for hypertension. We applied a Bayesian hierarchical model to estimate the prevalence of hypertension and the proportion of people with hypertension who had a previous diagnosis (detection), who were taking medication for hypertension (treatment), and whose hypertension was controlled to below 140/90 mm Hg (control). The model allowed for trends over time to be non-linear and to vary by age. Findings The number of people aged 30-79 years with hypertension doubled from 1990 to 2019, from 331 (95% credible interval 306-359) million women and 317 (292-344) million men in 1990 to 626 (584-668) million women and 652 (604-698) million men in 2019, despite stable global age-standardised prevalence. In 2019, age-standardised hypertension prevalence was lowest in Canada and Peru for both men and women; in Taiwan, South Korea, Japan, and some countries in western Europe including Switzerland, Spain, and the UK for women; and in several low-income and middle-income countries such as Eritrea, Bangladesh, Ethiopia, and Solomon Islands for men. Hypertension prevalence surpassed 50% for women in two countries and men in nine countries, in central and eastern Europe, central Asia, Oceania, and Latin America. Globally, 59% (55-62) of women and 49% (46-52) of men with hypertension reported a previous diagnosis of hypertension in 2019, and 47% (43-51) of women and 38% (35-41) of men were treated. Control rates among people with hypertension in 2019 were 23% (20-27) for women and 18% (16-21) for men. In 2019, treatment and control rates were highest in South Korea, Canada, and Iceland (treatment >70%; control >50%), followed by the USA, Costa Rica, Germany, Portugal, and Taiwan. Treatment rates were less than 25% for women and less than 20% for men in Nepal, Indonesia, and some countries in sub-Saharan Africa and Oceania. Control rates were below 10% for women and men in these countries and for men in some countries in north Africa, central and south Asia, and eastern Europe. Treatment and control rates have improved in most countries since 1990, but we found little change in most countries in sub-Saharan Africa and Oceania. Improvements were largest in high-income countries, central Europe, and some upper-middle-income and recently high-income countries including Costa Rica, Taiwan, Kazakhstan, South Africa, Brazil, Chile, Turkey, and Iran. Interpretation Improvements in the detection, treatment, and control of hypertension have varied substantially across countries, with some middle-income countries now outperforming most high-income nations. The dual approach of reducing hypertension prevalence through primary prevention and enhancing its treatment and control is achievable not only in high-income countries but also in low-income and middle-income settings. 191. Risk factors for executive function impairment in adolescence: an analysis of data from the 2004 Pelotas Birth Cohort study

Abstract: Objective: To investigate risk factors associated with impaired attention-related executive functions (EFs) at age 11 and working memory at age 15. Methods: Data from participants of the population-based 2004 Pelotas Birth Cohort at ages 11 (n=3,582) and 15 (n=1,950) were analyzed. The study measured attentional control, cognitive flexibility, and selective attention using the Test of Everyday Attention for Children (TEA-Ch). Spatial working memory was assessed by the Cambridge Neuropsychological Test Automated Battery (CANTAB). Logistic regression was employed to explore the relationship between perinatal and childhood exposures and EF impairment. Results: Low maternal education had a significant negative impact on EFs. At age 11, it was associated with decreased attentional control (OR = 3.04; 95%CI 2.09-4.43), and at age 15, it was linked to impaired spatial working memory (OR = 2.21; 95%CI 1.58-3.09). Additional risk factors included low household income, black or brown maternal skin color, high parity, prematurity, low birth weight, and multiple siblings. Breastfeeding, regardless of duration, was found to be a protective factor against impaired cognitive flexibility (OR = 0.38; 95%CI 0.22-0.65). Conclusion: This study underscores the lasting impact of perinatal exposures on EF development. Policies that mitigate the negative effects of risk factors and promote EF development, especially among vulnerable populations, are needed.

However, offspring that have experienced contrasting levels of long-term food intake may beg differently for a given level of short-term need, perhaps as a result of developmental differences in behaviour and/or physiology. We tested for the effects of both short- and long-term need on begging and digestion by manipulating food intake rates of the junior and senior nestlings from broods of pied flycatchers, Ficedula hypoleuca. Artificial food intake at natural levels created stable and normal levels of begging in both senior and junior nestlings, over 10h of hand-feeding individuals in isolation. In both classes of nestling, higher than natural levels of food intake led to similar progressive linear declines in begging effort, while lower than natural levels of food intake led to similar progressive increases in begging. However, there were no corresponding changes in digestive efficiency (assimilable mass coefficient). Consistent with previous studies, junior nestlings begged at consistently higher rates throughout compared with senior nestlings. There was also evidence that digestion by junior nestlings was slightly more efficient, but individual variation in begging effort (and change in begging effort) did not correspond to differences in digestive efficiency. These results show that begging does not reflect digestive efficiency across a range of natural food intake rates, and suggest that the honesty of offspring solicitation signals, such as the differences in begging between junior and senior nestlings, are not maintained by contrasting digestive development or function. Abstract: Providing all infants with the best start to life is a universal but challenging goal for the global community. Historically, the size and shape of infants, quantified by anthropometry and commencing with birthweight, has been the common yardstick for physical growth and development. Anthropometry has long been considered a proxy for nutritional status during infancy when, under ideal circumstances, changes in size and shape are most rapid. Developed from data collected in the Multicentre Growth Reference Study (MGRS), WHO Child Growth Standards for healthy infants and children have been widely accepted and progressively adopted. In contrast, and somewhat surprisingly, much less is understood about the ‘quality’ of growth as reflected by body composition during infancy. Recent advances in body composition assessment, including the more widespread use of air displacement plethysmography (ADP) across the first months of life, have contributed to a progressive increase in our knowledge and understanding of growth and development. Along with stable isotope approaches, most commonly the deuterium dilution (DD) technique, the criterion measure of total body water (TBW), our ability to quantify lean and fat tissue using a two-compartment model, has been greatly enhanced. However, until now, global reference charts for the body composition of healthy infants have been lacking. This paper details some of the historical challenges associated with the assessment of body composition across the first two years of life, and references the logical next steps in growth assessments, including reference charts.

Abstract: Background/Objective: Available evidence on infant body composition is limited. This study aimed to investigate factors associated with body composition at 6 and 24 months. Subjects/Methods: Multicenter study with data from a 0 to 6-mo cohort (Australia, India and South Africa) and a 3 to 24-mo cohort (Brazil, Pakistan, South Africa, and Sri Lanka). For the 0–6-mo cohort, body composition was assessed by air-displacement plethysmography (ADP) and for the 3–24-month cohort by the deuterium dilution (DD) technique. Fat mass (FM), fat-free mass (FFM), FM index (FMI), and FFM index (FFMI) were calculated. Independent variables comprised the Gini index of the country, maternal and infant characteristics, and breastfeeding pattern at 3 months. For the 3–24-mo cohort, breastfeeding, and minimum dietary diversity (MDD) at 12 months were also included. Crude and adjusted analyses stratified by sex were conducted by multilevel modelling using mixed models. Results: At 6 months, every 1 kg increase in birth weight was associated with an increase of 0.716 kg in FFM and 0.582 kg/m2in FFMI in girls, whereas in boys, the increase was of 0.277 kg in FFM. At 24 months, compared to those weaned before 12 months, girls still breastfed at 12 months presented a decrease of 0.225 kg in FM, 0.645 kg in FFM and 0.459 kg/m2in FFMI, and in boys the decreases were of 0.467 kg in FM, 0.603 kg in FFM and 0.628 kg/m2in FFMI. Conclusion: Birth weight and breastfeeding are independent predictors of body composition in early life, irrespective of sex.

Abstract: The study examines contemporary theories of the business environment and family entrepreneurship and shows what external effects the business environment has and what impact it can have on family businesses. Theories outlining the main external factors of the business environment and the different aspects of family businesses are highlighted. The business environment theories of Professors Campbell D. J. & Craig T. provide, in our opinion, the most comprehensive insight into the analysis of the external factors of the business environment, while Professors T. Zellwegers, K. E. Gersick and St. R. Covey highlight the various dimensions of the family business and its operational organisation. Our task in this article is to examine these two areas and how they interact and influence each other. The viability of family businesses requires stability and security and is determined by the environment in which the business operates. The external factors of the business environment are mostly those that the company cannot influence and that lead to uncertainty and instability. Early identification of undesirable conditions can help family businesses to respond more quickly. The European Commission's definition of a family business is at odds with that of academics. With this study, we invite you to update the discussion on the modern legal definition of a family business, which is the basis for policy planning documents, ensuring legal protection, assumption or inheritance of liabilities, etc. The results of the study could be significant for researchers dealing with business and economic regularity, as well as for entrepreneurs and professionals whose field of activity is related to business or public administration.

Abstract: Alzheimer’s disease (AD) is highly heritable and recent studies have identified over 20 disease-associated genomic loci. Yet these only explain a small proportion of the genetic variance, indicating that undiscovered loci remain. Here, we performed a large genome-wide association study of clinically diagnosed AD and AD-by-proxy (71,880 cases, 383,378 controls). AD-by-proxy, based on parental diagnoses, showed strong genetic correlation with AD (rg = 0.81). Meta-analysis identified 29 risk loci, implicating 215 potential causative genes. Associated genes are strongly expressed in immune-related tissues and cell types (spleen, liver, and microglia). Gene-set analyses indicate biological mechanisms involved in lipid-related processes and degradation of amyloid precursor proteins. We show strong genetic correlations with multiple health-related outcomes, and Mendelian randomization results suggest a protective effect of cognitive ability on AD risk. These results are a step forward in identifying the genetic factors that contribute to AD risk and add novel insights into the neurobiology of AD. © 2019.
