The in-depth study of protein-protein interactions (PPIs) is of key importance for understanding how cells operate. Therefore, in the past few years, many experimental as well as computational approaches have been developed for the identification and discovery of such interactions. Here, we present UniReD, a user-friendly, computational prediction tool which analyses biomedical literature in order to extract known protein associations and suggest undocumented ones. As a proof of concept, we demonstrate its usefulness by experimentally validating six predicted interactions and by benchmarking it against public databases of experimentally validated PPIs succeeding a high coverage. We believe that UniReD can become an important and intuitive resource for experimental biologists in their quest for finding novel associations within a protein network and a useful tool to complement experimental approaches (e.g. mass spectrometry) by producing sorted lists of candidate proteins for further experimental validation. ACTH is a pituitary hormone important for proper function of adrenal glands, cortisol production as well as human physiology in general. It is involved in the pathogenesis of several endocrine disorders like Cushing syndrome and can be a useful diagnostic tool for diseases like primary adrenal insufficiency. Although popular as a hormone in endocrine system physiology and testing, ACTH has been used as a drug since the 1950s. Except for steroid-releasing properties, its mechanism of action involves a steroid-independent anti-inflammatory and possible immune-modulatory effect. Pharmaceutic ACTH has a wide range of indications approved by FDA and usually comes in the form of subcutaneous injections. In this narrative review, we accumulated what we considered as important data from reviews, cases and trials involving the most basic FDA-approved ACTH indications. A special emphasis was given on rheumatologic indications of ACTH. More large data studies need to be performed to assess ACTH usefulness, efficacy, safety and cost-effectiveness as a drug.

Photodissolution is a process that is well known for its ability to cause inclusion of silver into the matrix of a chalcogenide layer, changing its optical properties. In this paper, using e-beam deposition, we developed Ag (74 nm)/As2S3 (355 nm) bilayers and characterized the photodissolution kinetics when exposed to actinic radiation. We showed that local complete silver photodissolution at the micron scale can be achieved. Based on this result, we then developed amplitude-based computer-generated holograms using direct laser writing. We review the establishment of computational biology in Greece and Cyprus from its inception to date and issue recommendations for future development. We compare output to other countries of similar geography, economy, and size—based on publication counts recorded in the literature—and predict future growth based on those counts as well as national priority areas. Our analysis may be pertinent to wider national or regional communities with challenges and opportunities emerging from the rapid expansion of the field and related industries. Our recommendations suggest a 2-fold growth margin for the 2 countries, as a realistic expectation for further expansion of the field and the development of a credible roadmap of national priorities, both in terms of research and infrastructure funding.

The little owl (Athene noctua) is a common raptor in Mediterranean habitats. To acquire more information on its diet, this study identified cranial and post-cranial skeletal material from 70 owl pellets collected during the 2016 and 2017 breeding seasons. The material was used to quantify the little owl’s relative prey abundance using MNI (minimum number of individuals), a taphonomical unit. This study is the first to examine the diet of the little owl in the Peloponnese (southern Greece). After examining 3,691 isolated skeletal and exoskeletal remains from the processed pellets, a total of 78 and 108 prey items were recorded for the two consecutive years. This study, in congruence with previous research, showed that in both years the little owl favoured primarily small mammals and arthropods, with a clear predominance of Thomas’s pine vole (Microtus thomasi) and arthropods from the class Diplopoda. Finally, a redundancy discriminant analysis (RDA) was applied to our two-year results, along with those from similar studies in the Mediterranean region, to examine the relationship between habitat types and prey taxa, which supported the little owl’s opportunistic feeding behaviour, depending on variation of ecological factors.

In the latest years, there is an increasing emphasis on DICOM medical images, especially in digital medical images transmission to e-health services. Besides, the confidentiality and protection of patients’ secure information need more attention and research. In this paper, we propose an enhanced medical image steganographic algorithm to hide patients’ secure information in their medical images. The proposed algorithm has a higher level of confidentiality while maintaining a higher quality of the medical image and a higher embedding capacity and robustness. The proposed algorithm has two main steps. The medical cover image is decomposed using an integer wavelet filter in the first step. Moreover, the patients’ confidential information is processed using the Arithmetic Coding (AC) and Data Encryption Standard (DES) algorithms to be compressed and encrypted before of the embedding process. In the second step, the insignificant coefficients are selected from the integer wavelet high-frequency subbands of the transformed medical image, and finally, both the target cover bits and the secret bits are grouped during the injection process. The performance of our algorithm was evaluated using different evaluation metrics. The obtained results show that our algorithm outperforms existing ones, with a higher embedding capacity, and a lower imperceptibility. The proposed algorithm can be applied in hospitals and health care canters worldwide.

Antisense therapy is widely used as an alternative therapeutic option for various diseases. RNA interference might be effective in infections, through the degradation of messenger RNA and, therefore, translation process. Hence, proteins essential for microorganisms and viruses’ proliferation and metabolism are inhibited, leading to their elimination. The present study aimed to evaluate the use of oligonucleotide in patients infected by Epstein–Barr (EBV) or Herpes Simplex Viruses 1/2 or with Lyme Disease caused by Borrelia burgdorferi. Blood samples were collected from 115 patients and the different species were characterized using molecular biology techniques. Then, SOT molecules (Supportive Oligonucleotide Therapy), which are specific small interfering RNA (siRNA), were designed, produced, and evaluated, for each specific strain. Oligonucleotides were administered intravenously to patients and then a quantitative Polymerase Chain Reaction was used to evaluate the effectiveness of SOT. This study revealed that for Lyme Disease, one or two SOT administrations can lead to a statistically significant decrease in DNA copies, while for viruses, two or three administrations are required to achieve a statistically significant reduction in the genetic material. These preliminary results indicate that antisense SOT therapy can be considered a potential treatment for viral as well as Lyme diseases.

Wolf (Canis lupus) populations have recovered and expanded across many parts of the world thanks to conservation efforts, including improved legal status and restoration of their prey. Concurrently, public concerns regarding the risk of wolf attacks on humans and livestock are increasing as wolves occupy human-dominated landscapes. We examined a unique case in Europe allegedly involving wolves in the death of a female British tourist, aged 64, in northern Greece in September 2017. This incident received extensive international media attention and yet many fundamental details of the case area are lacking, including whether local livestock guarding dogs played a role. To assist in resolving the case, we conducted an extensive literature review which documented 13 criteria linked to the risk of either a wolf and/or a dog attacking a human. We also conducted a camera trap survey (October to December 2017) soon after the fatal attack to calculate the activity overlap among humans, dogs and wolves. Sufficient data were available for assessing 11 of the 13 criteria. For the remaining two, the required data were either not analysed (i.e. canid DNA collected from the attack site), not appropriately collected (i.e. DNA from the mouths of suspected dogs) or were collected, but misinterpreted (i.e. the post-consumption patterns of the victim’s corpse). Via this combination of evidence, we conclude that this case involved a fatal dog attack. This assertion is supported by evidence such as the: a) high dog-human activity overlap at the attack site which peaked during the attack time as opposed to near zero wolf-human activity overlap at the same time, b) presence of a large pack of unsupervised dogs, c) high ratio of male dogs in the dog pack, d) close vicinity of the attack site to dog owner’s property and e) previous documented aggression of these dogs towards humans. The consumption patterns, time scale and location of the victim’s remains indicate a posthumous consumption of the corpse possibly by the same dogs and/or by wild scavengers including wolves. A multidisciplinary approach, such as this one, in the assessment of putative wildlife attacks on humans can reduce misidentifications of the responsible species by forensic authorities and, therefore, prevent unfounded decrease in public tolerance for large carnivores.

The management of septic non-unions with associated bone necrosis is challenging, especially when the resulting bone defect after the debridement is extensile. Different techniques have been described in the literature for the treatment of these demanding cases, with the most prominent being free vascularized Fibular graft and bone transport with distraction osteogenesis principles. Recently, 3D printing technology has been increasingly utilized in many complex orthopaedic pathologies. However, the application of those advancements regarding septic non-unions with residual bone defect has not been previously studied. This study presents a novel 3D printing technique for the management of an infected critical bone deficit of the tibia. Queries, challenges and future perspectives concerning the recruiting of 3D printing technology in limb reconstruction are also being discussed.Clinical Evidence Level: IV.

Cutaneous drug concentration quantification after topical application remains an active, yet challenging research area for topical drug development. Macroscale approaches quantify cutaneous pharmacokinetics 30 min to hours after application and miss rapid temporal and spatial dynamics that are vital to comprehend drug disposition. We have developed a 3D-printed applicator coupled with an inverted microscope and a rapidly-tunable fiber optic laser to quantify active pharmaceutical ingredients via sparse spectral sampling stimulated Raman scattering. The 3D-printed applicator is cost-effective (< $0.70/applicator) and utilizes a small formulation volume (20 µL). Ruxolitinib was formulated in two known permeation enhancers (propylene glycol and diethylene glycol monoethyl ether) that are known to display different permeation profiles to validate device capabilities. Results indicated that the applicator enabled relative-concentration monitoring immediately following drug product application. This approach has significant potential for investigating novel excipients, active pharmaceutical ingredients, and formulations to understand the permeation and biodistribution of these compounds.

BackgroundOutcomes after lung transplant (LTx) in children have slowly improved. Although atrial arrhythmia (AA) is a common and adverse complication following LTx among adults, there is limited data on pediatric recipients. We detail our pediatric single-center experience while providing further insights on occurrence and management of AA following LTx.MethodsA retrospective analysis of LTx recipients at a pediatric LTx program from 2014 to 2022 was performed. We investigated timing of occurrence and management of AA following LTx, and its effect on post-LTx outcome.ResultsThree out of nineteen (15%) pediatric LTx recipients developed AA. The timing of occurrence was 9–10 days following LTx. Those patients in the older age group (age >12 years old) were the only ones who developed AA. Developing AA did not have a negative effect on hospital stay duration or short-term mortality. All LTx recipients with AA were discharged home on therapy that was discontinued at 6 months for those who was on mono-therapy without recurrence of AA.ConclusionsAA is an early post-operative complication in older children and younger adults undergoing LTx at a pediatric center. Early recognition and aggressive management can mitigate any morbidity or mortality. Future investigations should explore factors that place this population at risk for AA in order to prevent this complication post-operatively.

Background: Achilles tendon rupture can be treated nonoperatively with functional rehabilitation. However, prolonged immobilization has associated risk of venous thromboembolism (VTE). Early weightbearing may reduce VTE risk, and this was introduced to our rehabilitation protocol. We investigated the prevalence of symptomatic VTE events before and after the introduction of the early weightbearing protocol. Methods: Adults with ultrasonography-confirmed complete tendo-Achilles ruptures between January 2017 and June 2020 were included. Preprotocol, patients were instructed to not weightbear for 4 weeks. In 2018, immediate weightbearing was introduced to the treatment protocol. All patients in both cohorts were given low-molecular-weight heparin for 4 weeks. Patients with symptomatic VTE events were investigated with duplex ultrasonographic scan or chest computed tomography. Two independent anonymized examiners collected data from electronic records. Rates of symptomatic VTEs were compared. Results: A total of 296 patients were included. Sixty-nine patients were managed with the nonweightbearing protocol, and 227 patients were managed with the early-weightbearing protocol. Two patients in each group developed deep vein thrombosis and 1 developed pulmonary embolism in the early-weightbearing group. Rates of VTEs were lower in the early-weightbearing group (1.3% vs 2.9%) but did not reach statistical significance ( P = .33). Conclusion: In this cohort we found that symptomatic VTE after nonoperatively treated Achilles tendon rupture was uncommon. We did not demonstrate a reduction in symptomatic VTE between our early weightbearing and nonweightbearing rehabilitation protocols. We believe a larger study may help clarify whether early weightbearing is beneficial in VTE reduction. Level of Evidence: Level III, retrospective cohort study.

Out-of-autoclave processes of carbon fiber thermoplastic composites are gaining interest as they can drastically reduce the economic cost. To optimize consolidation, the flow behavior of these highly filled composites has to be characterized. Here, we propose to measure viscosity of carbon fiber/polyetherketoneketone through squeeze flow experiments in a rheometer. A modified Stefan's law assuming a power law fluid behavior with full anisotropy is developed for square and circular geometries to model the data. Values of the power law parameters K and n are obtained, on the order of 15,000 Pa.sn and 0.02 at 1 bar. Though as expected independent of the samples' geometry and tapes' stacking, K and n depend on the applied pressure and plate size. This is due to localized shear which results in a shear-banding-like phenomenon. Finally, squeeze flow is compared to dynamic measurements and invalidate the Cox-Merz rule for such materials.

This study delves into the sedimentation mechanisms governing mud deposits in shallow marine and tidal environments, with a particular focus on elucidating the versatile therapeutic applications of these muds. This research provides valuable insights for optimizing the selection of mud as a cosmetic resource that can positively influence human health and well-being by utilizing a comprehensive analysis involving CaCO3, TOC, grain size, and statistical parameters across six outcrops situated on the Kefalonia and Corfu islands. The research reveals that the CaCO3 content of mud deposits on both islands is comparable. Despite the average value (26.71%) significantly exceeding the recommended value (10%) for optimal plasticity, no discernible impact on the mechanical behavior and plasticity of the clay was observed, rendering it a neutral quality criterion. In the last decade, the publication of research studies in the field of groundwater ecosystem services and their classification has increased dramatically. Several academic and other institutions have developed ecosystem service classification systems for use in environmental policy research. However, the existing classification systems need to be optimized in the following areas: (a) the weak correlation between concepts and the definitions assigned, which creates double counting in economic valuation, (b) the absence of correlation of the concept of use value with the ecosystem service, (c) the non-categorization of final and intermediate services, and (d) the overlapping of wetland, soil, and groundwater regulating ecosystem services. Our research aims, through the results of a literature analysis and the synthesis of the results of an expert consultation process, to develop a unified categorization system for groundwater ecosystem services. In the context of the conceptualization of groundwater ecosystem services, this research implemented an expert judging elicitation process where subject discussions and targeted interviews were performed, combined with a literature review analysis. Through the completion of a specific questionnaire and expert interviews, a new groundwater ecosystem services classification system, namely GROUNDWES, was established.

The problem of remaining useful life estimation (RULE) of hollow worn railway vehicle wheels in terms of remaining mileage via wheel tread depth estimation using on-board vibration signals from a single accelerometer on the bogie frame is presently investigated. This is achieved based on the introduction of a statistical time series method that employs: (i) advanced data-driven stochastic Functionally Pooled models for the modeling of the vehicle dynamics under different wheel tread depths in a range of interest until a critical limit, as well as tread depth estimation through a proper optimization procedure, and (ii) a wheel tread depth evolution function with respect to the vehicle running mileage that interconnects the estimated hollow wear with the remaining useful mileage. The method’s RULE performance is investigated via hundreds of Simpack-based Monte Carlo simulations with an Attiko Metro S.A. vehicle and many hollow worn wheels scenarios which are not used for the method’s training. The obtained results indicate the accurate estimation of the wheels tread depth with a mean absolute error of ∼0.07 mm that leads to a corresponding small error of ∼3% with respect to the wheels remaining useful mileage. In addition, the comparison with a recently introduced Multiple Model (MM)-based multi-health state classification method for RULE, demonstrates the better performance of the postulated method that achieves 81.17% True Positive Rate (TPR) which is significantly higher than the 45.44% of the MM method.
