The Pacific oyster, Crassostrea gigas, is the main oyster species cultivated in Korea. Despite a long history of hatchery practices, little is known about the population genetics of native populations of C. gigas in Korea. This study examined the population structure and genetic diversity of 297 individuals of C. gigas in six populations, representing two major geographically separated habitats along the coast of Korea, using multiplex assays with nine highly polymorphic loci. All populations showed very high genetic diversity, with a deficit of heterozygotes and a recent bottleneck. No significant differences were found in the genetic diversity of the two geographically separated oyster groups, although significant, but minimal, genetic differentiation was observed (FST = 0.003, p = 0.002). This was mainly due to genetic differences between the oyster populations from Taean and Gaduk, suggesting that the oyster is not panmictic. This result was corroborated by an analysis of molecular variance (AMOVA) and multidimensional scaling analysis. It was inconsistent with a previous finding of no population differentiation using mitochondrial sequence analysis, implying rather recent separation of the two geographic populations. This genetic differentiation might have resulted from the combined of various factors, such as historical dispersal, the local environment, and ocean currents. Nonetheless, these results mean that the two geographical groups should be treated as independent units for proper resource management. Ruditapes philippinarum and R. variegate are commercially important shellfish in Korea. In order to understand the processes and organization of genetic diversity and genetic resources for the sustainable management of fisheries resources we developed 13 primer pairs for microsatellite loci in R. philippinarum and tested their cross-amplification in R. variegate. Twelve primers amplified in both species. Nine loci were polymorphic in R. philippinarum and eight in R. variegate, each with nine to 26 alleles per locus. Unbiased expected heterozygosity levels varied from 0.73 to 0.94 in R. philippinarum. All polymorphic loci possessed species-specific alleles. No linkage disequilibrium was found. Discovering new drug candidates with high efficacy and few side effects is a major challenge in new drug development. The two evolutionarily related peptides oxytocin (OXT) and arginine vasopressin (AVP) are known to be associated with a variety of physiological and psychological processes via the association of OXT with three types of AVP receptors. Over decades, many synthetic analogs of these peptides have been designed and tested for therapeutic applications; however, only a few studies of their natural analogs have been performed. In this study, we investigated the bioactivity and usefulness of two natural OXT/AVP analogs that originate from the marine invertebrate Octopus vulgaris, named octopressin (OTP) and cephalotocin (CPT). By measuring the intracellular Ca2+ or cyclic AMP increase in each OXT/AVP receptor subtype–overexpressing cell, we found that CPT, but not OTP, acts as a selective agonist of human AVP type 1b and 2 receptors. This behavior is reminiscent of desmopressin, the most widely prescribed antidiuretic drug in the world. Similar to the case for desmopressin, a single intravenous tail injection of CPT into Sprague-Dawley rats reduced urine output and increased urinary osmolality. In conclusion, we suggest that CPT has a significant antidiuretic effect and that CPT might be beneficial for treating urological conditions such as nocturia, enuresis, and diabetes insipidus. We investigated the study on the endemic and endangered species of Dawkinsia tambraparniei were confined only the areas of the river Tamiraparani. These species are under threats due to the menace of anthropogenic stress. To recognize the crisis behind the particular species, it was analyzed histologically and molecularly from the five pollutant levels of river Tamiraparani. Histologically, the microscopic examinations were also carried out from the crucial organs such as the brain, gill, heart, kidney, and liver, which confirm the spiky survivability of the endemic fish. Assessment of fish organ damages was observed highly in Kokkirakulam and Vannarapettai. Probably with conserved molecular sequences, the species can be identified out from the encountered surveillance of the particular taxa leading to the evolutionary circumstances. The phylogenetic analysis of Dawkinsia tambraparniei populations showed that Cheranmadevi and Vallandau sites populations were closely distributed. Even though the species have similarity sequences of each population were shown that the closely related with same genus but other sub-species. The observed results emphasize the conventional measures to conserve the endemic species and more effectual planning to the proximity of endurances in inhabited zone. [ABSTRACT FROM AUTHOR]

The small abalone, Haliotis diversicolor supertexta, of the family Haliotidae, is one of the most important species of marine shellfish in eastern Asia. Over the past few decades, this species has drastically declined in Korea. Thus, hatchery-bred seeds have been released into natural coastal areas to compensate for the reduced fishery resources. However, information on the genetic background of the small abalone is scarce. In this study, 20 polymorphic microsatellite DNA markers were identified using next-generation sequencing techniques and used to compare allelic variation between wild and released abalone populations in Korea. Using high-throughput genomic sequencing, a total of 1516 (2.26%; average length of 385 bp) reads containing simple sequence repeats were obtained from 86,011 raw reads. Among the 99 loci screened, 28 amplified successfully, and 20 were polymorphic. When comparing allelic variation between wild and released abalone populations, a total of 243 different alleles were observed, with 18.7 alleles per locus. High genetic diversity (mean heterozygosity = 0.81; mean allelic number = 15.5) was observed in both populations. A statistical analysis of the fixation index (FST) and analysis of molecular variance (AMOVA) indicated limited genetic differences between the two populations (FST = 0.002, p > 0.05). Although no significant reductions in the genetic diversity were found in the released population compared with the wild population (p > 0.05), the genetic diversity parameters revealed that the seeds released for stock abundance had a different genetic composition. These differences are likely a result of hatchery selection and inbreeding. Additionally, all the primer pair sets were effectively amplified in another congeneric species, H. diversicolor diversicolor, indicating that these primers are useful for both abalone species. These microsatellite loci may be valuable for future aquaculture and population genetic studies aimed at developing conservation and management plans for these two abalone species. Kelp grouper (Epinephelus bruneus Bloch 1793) is a commercially important fish in Korea. In recent years, the catch of kelp grouper in the coastal waters of Korea has significantly declined. Despite its importance, little is known about its genetic diversity and conservation efforts are hampered. In this study, we isolated and characterized 12 microsatellite loci using an enrichment method based on magnetic/biotin capture of microsatellite sequences from a size-selected genomic library. All loci were readily amplified and contained TG/CA denucleotide repeats. To characterize each locus, 30 individuals from a natural E. bruneus population in the coastal waters of Jeju Island, Korea, were genotyped. All loci except three, KEm118, KEm154, and KEm219, were polymorphic, with an average of 8.1 alleles per locus (range 2–18). The mean observed and expected heterozygosities were 0.47 (range 0.19–1.00) and 0.61 (range 0.29–0.92), respectively. A significant deviation from Hardy-Weinberg equilibrium was observed at three loci (KEm134, KEm184, and KEm283). These findings will be useful for effective monitoring and management of genetic variation of kelp grouper as well as for the implementation of a fisheries conservation program. [ABSTRACT FROM AUTHOR]
