Abstract

Variation in nutrition is a key determinant of growth, body composition, and the ability of animals to perform to their genetic potential. Depending on the quality of feed available, animals may be able to overcome negative effects of prior nutritional restriction, increasing intake and rates of tissue gain, but full compensation may not occur. A 2 × 3 × 4 factorial serial slaughter study was conducted to examine the effects of prior nutritional restriction, dietary energy density, and supplemental rumen undegradable protein (RUP) on intake, growth, and body composition of lambs. After an initial slaughter (n = 8), 124 4-mo-old Merino cross wethers (28.4 ± 1.8 kg) were assigned to either restricted (LO, 500 g/d) or unrestricted (HI, 1500 g/d) intake of lucerne and oat pellets. After 8 wk, eight lambs/group were slaughtered and tissue weights and chemical composition were measured. Remaining lambs were randomly assigned to a factorial combination of dietary energy density (7.8, 9.2, and 10.7 MJ/kg DM) and supplemental RUP (0, 30, 60, and 90 g/d) and fed ad libitum for a 12- to 13-wk experimental period before slaughter and analysis. By week 3 of the experimental period, lambs fed the same level of energy had similar DMI (g/d) and MEI (MJ/d) (P > 0.05), regardless of prior level of nutrition. Restricted-refed (LO) lambs had higher rates of fat and protein gain than HI lambs (P < 0.05) but had similar visceral masses (P > 0.05). However, LO lambs were lighter and leaner at slaughter, with proportionally larger rumens and livers (P < 0.05). Tissue masses increased with increasing dietary energy density, as did DMI, energy and nitrogen (N) retention (% intake), and rates of protein and fat gain (P < 0.05). The liver increased proportionally with increasing dietary energy density and RUP (P < 0.05), but rumen size decreased relative to the empty body as dietary energy density increased (P < 0.05) and did not respond to RUP (P > 0.05). Fat deposition was greatest in lambs fed 60 g/d supplemental RUP (P < 0.05). However, lambs fed 90 g/d were as lean as lambs that did not receive supplement (P0, P > 0.05), with poorer nitrogen retention and proportionally heavier livers than P0 lambs (P < 0.05). In general, visceral protein was the first tissue to respond to increased intake during refeeding, followed by non-visceral protein and fat, highlighting the influence of differences in tissue response over time on animal performance and body composition.

152. Asynchronous and Rhetorical: Appointment Forms and Their Effect on Writer-Consultant Exchanges

Abstract

Especially in the wake of the recent pandemic, asynchronous consulting has become increasingly central to writing center work. Yet writing center scholarship has little attended to the significant impact writer input can have on asynchronous writer-consultant exchanges. Drawing on asynchronous consultation data collected before and after our 2019 redesign of our writing center's asynchronous system, this comparative study examines the specific effect of the writer appointment form on the nature of both writers' requests for feedback (RFFs) and consultants' resulting comments. Our findings suggest that differently designed appointments forms can scaffold significantly different kinds of asynchronous writer-consultant exchanges, especially visible in the different emphases writers and consultants put on issues of correctness, clarity, organization, and the writer's rhetorical situation. We argue that, particularly in the case of asynchronous consulting--which can easily devolve to a "fix-it" model of consulting--it is important for writing center administrators to design asynchronous platforms that encourage both writers and consultants to more explicitly consider how the specific rhetorical features of a writing task can shape revising goals.

153. Decision-Making by and for Academic Libraries during COVID-19

Abstract

Academic libraries are fundamental in promoting equitable access to education but are often overlooked and underfunded. The COVID-19 pandemic amplified these inequities. This study investigates how 39 library deans and directors perceived decision-making by university administration during COVID-19's onset. Open-ended survey questions were sent to deans and directors asking them to describe their experiences working with university administration to adjust library services during the crisis. Some library leaders reported that working closely with other departments strengthened their campus connections. Others commented that disagreement between library personnel and university administration caused discord. Some deans and directors were forced to cut staff funding or felt pressured to reopen, while others were trusted to choose their budget and service priorities. The authors recommend that library leaders be more consistently relied upon for their expertise during university decision-making.

154. Teacher Professional Development in K-12 Schools: A School Leader's Perspective

Abstract

This study examines teacher professional development as part of the required K-12 continuous school improvement process. Through semi-structured, open-ended interviews and data (school improvement plans, PD agendas), this qualitative case drew from organizational theory to identify and describe information to better understand the role teacher professional development takes in the continuous school improvement process, the barriers to the implementation of effective teacher professional development, and how principals assess the effectiveness of teacher professional development. Findings indicate that principals desire to support their schools for student success and teacher support in an educational environment that is conducive to learning. Continuous school improvement included a balance of dedicated commitment from principals, reform compliance, utilizing data in decision-making, and human emotion to navigate and continuously assess effective teacher professional development. Abstract

Antibiotic resistance is a serious ongoing threat to both the medical and agricultural sectors, with far-reaching and diverse consequences. Historically, antibiotic resistance has been thought to be driven by the anthropogenic use of antibiotics in clinical and agricultural settings. However, there is a growing body of evidence that clearly demonstrates that antimicrobial resistance developed naturally, millions of years ago. Understanding these natural drivers of antibiotic resistance is key to managing the ongoing antimicrobial resistance problem. One potential natural driver of antibiotic resistance are phytochemicals which are chemicals produced by plants for environmental interaction. Phytochemicals are being explored for their potential role as antibiotic replacements in the clinic, and in agriculture. We propose that the use of phytochemicals as antimicrobial replacements in multiple industries will have an equal, or worse impact on the problem of antibiotic resistance, due to their potential to select for cross-resistance to other antimicrobials such as antibiotics. Phytochemicals could drive antibiotic resistance in one of three ways: (1) The selection of genes which confer cross resistance to antibiotics. (2) The selection of plasmids carrying both phytochemical and antibiotic resistance genes and (3) the selection of bacteria which are known antimicrobial resistance reservoirs. In this study, two antimicrobial phytochemicals, quercetin and berberine were explored for their role as natural drivers of antibiotic resistance. This study was split into three different experimental methodologies. The first experimental methodology developed a novel phytochemical screening assay, which was then successfully used to screen an oral metagenomic library, and the Swab and Send bacterial isolate library. Screening the metagenomic library uncovered three isolates with tolerance to either quercetin or berberine. These isolates contained inserts with multiple antibiotic resistance genes, including a glycosyltransferase and transpeptidase. Screening the Swab and Send library uncovered 20 isolates from 11 bacterial species containing antibiotic resistance genes with a phytochemical tolerant phenotype. The second experimental methodology evolved P. aeruginosa NCTC 7244 and NCTC 9433 in sub-inhibitory concentrations of the phytochemicals for 30 days. This experiment was conducted see how environmental bacteria responded to phytochemical selective pressure. Eleven of the 12 isolates developed a 2-fold phytochemical tolerance after 30 days. The isolates contained multiple genetic mutations including mutations in a glycosyltransferase and transpeptidase gene. The final experimental methodology explored the impact of phytochemical addition to chicken feed on the microbiome of chickens through two in vitro model experiments, and an in vivo chicken study. The studies also compared the viability of the two in vitro models as replacement for in vivo chicken experiments. Addition of both phytochemicals impacted microbiome development in all models compared to the control. Specifically increasing the levels of Escherichia-shigella and Turicibacter genera and decreasing levels of Lactobacillus pontis which may indicate an increase in disease incidence due to phytochemical supplementation. This study shows that berberine and quercetin can act as a selective pressure for the maintenance and mutation of genes which confer antimicrobial resistance, and the selection of antimicrobial resistant isolates in isolate libraries and the chicken gastrointestinal system. Therefore, serious consideration should be given to the risks of using phytochemicals as antimicrobial replacements in both agricultural and clinical sectors.

174. Understanding the lived experience of older men in prison

Abstract

Older prisoners are the fastest growing prison population with the number of older offenders rapidly increasing in recent years. Whilst this trend can be seen across both male and female offenders, the overwhelming majority of older people in prison are men. Despite this, older men in prison have received limited attention within policy, practice, and academic research. Insight into how they make sense of their everyday realities in prison is particularly scant. This thesis presents a constructivist grounded theory study exploring the lived experiences of older men in prison. A total of 24 in-depth interviews were conducted with 17 older men who had experience of prison in later life, either in prison or recently released, and 7 prison officers. Central to the men's experiences of prison were the nature of the environment, their relationships, and identities, and preparing for and managing release. An overarching theoretical account was constructed from analysis of the empirical data, with two concepts central to this: prison climate and capital. The older men described various factors that came together to shape the overall feel of the prison environment, or climate, and the extent to which this was suitable for their needs as older people. Whilst there was a sense of commonality in that the prison climate was perceived as challenging and hard to navigate, the way in which it was experienced was also shaped by the presence of various individual resources, or capital. Based on this, some were afforded more ways in which to negotiate the environment and make sense of their situation constructively. This study therefore contributes an original way of understanding the experiences of older men in prison. The implications of these findings are discussed in relation to both policy and practice. The importance of a unifying policy strategy for older offenders is particularly stressed.

175. Transdermal delivery of heavy chain only antibody binding domains using hydrogel-forming microarray patches

Abstract

Since the licensing of the first monoclonal antibody (mAb) over 30 years ago, the mAb industry has expanded exponentially, with six of the top 10 blockbuster drugs in 2018 being antibody-based. However, rather than resting on their laurels, drug developers have continued to innovate. This has led to the development of 'next generation' biologics which have been designed to overcome the size and stability limitations associated with mAbs. This new era of biotechnology incorporates heavy chain only antibodies (HCAbs) such as Nanobody® molecules and VNARs, derived from camelids and sharks, respectively. Owing to their small size (~13 kDa) and reduced complexity, they are less expensive to produce than mAbs. Furthermore, their potency has been shown to be an order of magnitude higher than the best-selling mAb-based drug currently on the market, Humira®. This has enabled other methods of administration to be investigated, such as the oral, pulmonary and ocular routes. However, transdermal delivery of HCAbs has yet to be examined. Therefore, this thesis explores the transdermal delivery of both VNARs and Nanobody® molecules using hydrogel-forming microarray patches (MAPs). Using this MAP technology, both HCAbs could permeate through dermatomed (350 µm) neonatal porcine skin in vitro. Furthermore, two Nanobody® molecules were successfully delivered in a Sprague Dawley rat model for the first time. Therefore, the MAP system developed in this thesis has supported the transdermal delivery of 'next generation' biotherapeutics. This thesis also aimed to address the requests of patients from a previous study by designing a novel feedback mechanism to confirm complete MAP skin insertion. With further modification, this could help facilitate successful MAP commercialisation.

The possibility of two interaction regions (IRs) is a design requirement for the Electron Ion Collider (the EIC). There is also a significant interest from the nuclear physics community in a 2nd IR with measurements capabilities complementary to those of the first IR. Abstract

Many scientific high-throughput applications can benefit from the elastic nature of Cloud resources, especially when there is a need to reduce time to completion. Cost considerations are usually a major issue in such endeavors, with networking often a major component; for data-intensive applications, egress networking costs can exceed the compute costs. Dedicated network links provide a way to lower the networking costs, but they do add complexity. In this paper we provide a description of a 100 fp32 PFLOPS Cloud burst in support of IceCube production compute, that used Internet2 Cloud Connect service to provision several logically-dedicated network links from the three major Cloud providers, namely Amazon Web Services, Microsoft Azure and Google Cloud Platform, that in aggregate enabled approximately 100 Gbps egress capability to on-prem storage. Abstract

Acute environmental stressors such as short-term exposure to pollutants can have lasting effects on organisms, potentially impacting future generations. Parental exposure to toxicants can result in changes to the epigenome (e.g., DNA methylation) that are passed down to subsequent, unexposed generations. However, it is difficult to gauge the cumulative population-scale impacts of epigenetic effects from laboratory experiments alone. Here, we developed a size- and age-structured delay-coordinate population model to evaluate the long-term consequences of epigenetic modifications on population sustainability. The model emulated changes in growth, mortality, and fecundity in the F0, F1, and F2 generations observed in experiments in which larval Menidia beryllina were exposed to environmentally relevant concentrations of bifenthrin (Bif), ethinylestradiol (EE2), levonorgestrel (LV), or trenbolone (TB) in the parent generation (F0) and reared in clean water up to the F2 generation. Our analysis suggests potentially dramatic population-level effects of repeated, chronic exposures of early-life stage fish that are not captured by models not accounting for those effects. Simulated exposures led to substantial declines in population abundance (LV and Bif) or near-extinction (EE2 and TB) with the exact trajectory and timeline of population decline dependent on the combination of F0, F1, and F2 effects produced by each compound. Even acute one-time exposures of each compound led to declines and recovery over multiple years due to lagged epigenetic effects. These results demonstrate the potential for environmentally relevant concentrations of commonly used compounds to impact the population dynamics and sustainability of an ecologically relevant species and model organism.

Abstract

Accurate lesion segmentation is critical in stroke rehabilitation research for the quantification of lesion burden and accurate image processing. Current automated lesion segmentation methods for T1-weighted (T1w) MRIs, commonly used in stroke research, lack accuracy and reliability. Manual segmentation remains the gold standard, but it is time-consuming, subjective, and requires neuroanatomical expertise. We previously released an open-source dataset of stroke T1w MRIs and manually-segmented lesion masks (ATLAS v1.2, N = 304) to encourage the development of better algorithms. However, many methods developed with ATLAS v1.2 report low accuracy, are not publicly accessible or are improperly validated, limiting their utility to the field. Here we present ATLAS v2.0 (N = 1271), a larger dataset of T1w MRIs and manually segmented lesion masks that includes training (n = 655), test (hidden masks, n = 300), and generalizability (hidden MRIs and masks, n = 316) datasets. Algorithm development using this larger sample should lead to more robust solutions; the hidden datasets allow for unbiased performance evaluation via segmentation challenges. We anticipate that ATLAS v2.0 will lead to improved algorithms, facilitating large-scale stroke research.

185. Teacher-Student Relationships: The Lived Experiences of Four K-12 Music Educators

Abstract

The purpose of this phenomenological study was to explore four K-12 public school music educators' lived experiences with teacher-student relationships. Research questions were the following: (a) How do music educators describe teacher-student relationships in their classrooms? (b) What strategies do music educators use to encourage positive, caring relationships with their students? and (c) What benefits and challenges exist for educators in relationship-focused classrooms? Participant selection was purposive; all participants were known to prioritize teacher-student relationships in their teaching. Interview data, coded in multiple phases, revealed six themes that influenced teacher-student relationships: structure; time; family and community connections; choice, voice, and ownership; authenticity; and enthusiasm. The essence of participants' experiences involved moments of caring connection facilitated by awareness, empathy, and attention to individuals. Implications for music teacher educators include the intentional design of curriculum and instruction, approaches to classroom interactions, and focus on students' experiences.

187. School Constituents' Ratings of the Leadership Dimensions of High-Performing Principals Based on School Regions

Abstract

In Jamaica, schools are placed in six regions which involve one to three parishes, but in one instance, two regions are partially located in one parish. While schools in any jurisdiction should perform in accordance with stated standards and established criteria, there are many factors which seem to mitigate against this expectation. This failure usually results in weaker schools located in some regions whose performance differ based on factors such as location-urban versus rural, the levels and types of schools, the overall population of these schools, and quality of human resources, including teachers and principals, among others. In Jamaica, Region 1 comprises three adjoining parishes, Kingston, St. Andrew and St. Thomas. Five of the regions are essentially in rural Jamaica, except region 1 and even then substantial parts of St. Andrew are rural and St. Thomas is fully rural. The fact that schools are located in mainly rural settings means that the leadership displayed by the principal will be influenced by the given context and his/her ability to use personal abilities and characteristics to influence change. This study seeks to determine if school constituents' ratings of the leadership dimensions of high-performing principals based on the regional location of schools. A knowledge of the ratings of high-performing principals is important because the leadership provided by the principal is central to school performance generally. This paper locates the study of high-performing principal in the Jamaican school context and provides a review of the relevant literature with emphasis on the phases of leadership development and the four dimensions related to the performance of high-performing principals. The overview of the methodology presents the sample, procedures and statistical tests which were used and the findings presented and discussed in regards to the four dimensions related to the performance of high-performing principals. The conclusion makes specific suggestions and recommendations regarding strategies and approaches to address some of the problems related to the differences in the ratings of the performance dimensions of high-performing principals based on school regions.
