171. Correlations of miR-203a-3p and PHOX2B expression in glioma tissues with clinical features and prognosis.

Abstract

Objective To investigate the correlations microRNA-203a-3p (miR-203a-3p) and paired allographic box protein 2B (PH0X2B) expression levels in glioma tissues with clinical features and prognosis. Methods Ninety-six glioma patients admitted to our hospital from June 2017 to April 2020 were included in the study. The resected glioma tissues and corresponding paracancerous tissues from the patients were collected intraoperatively. qRT-PCR and immunohistochemistry were conducted to detect the expression of miR-203a-3p and PHOX2B mRNA in glioma tissues and paracancerous groups. Pearson's method was used to analyze the correlation between miR-203a-3p and PHOX2B mRNA. Kaplan-Meier survival curves were used to analyze the correlations of miR-203a-3p and PHOX2B mRNA with the prognosis of glioma patients. COX regression was used to analyze the risk factors affecting the prognosis of glioma patients. Results The miR-203a-3p level and positive expression rate of glioma tissues were significantly lower than that of paracancerous tissues (P < 0.05), and the PHOX2B mRNA level and positive expression rate were significantly higher than that of paracancerous tissues (P < 0.05). By Pearson correlation analysis, the miR-203a-3p and PHOX2B mRNA levels of glioma patients were negatively correlated ( P < 0.05 ). miR-203a-3p and PHOX2B were associated with KPS score, tumor tissue necrosis, and WHO grading ( P < 0.05 ) . Kaplan-Meier curve analysis yielded the results that the survival rate of the miR-203a-3p highly expressed group was significantly higher than that of the lowly expressed group (P < 0.05), and the survival rate of the PHOX2B mRNA highly expressed group was significantly lower than that of the lowly expressed group (P < 0.05 ). COX regression analysis showed that miR-203a-3p, PHOX2B mRNA, tumor tissue necrosis, and WHO grading were the risk factors affecting the prognosis of glioma patients (P < 0.05). Conclusion The expression level of miR-203a-3p is obviously reduced and the expression level of PHOX2B mRNA is obviously increased in glioma, both showing close correlation with clinical characteristics and prognosis. 173. A dynamic virtual try-on simulation framework for speed skating suits.

Abstract

Speed skating suits have important effects on performance of athletes, and they also protect human body to avoid injury in competitions. Appropriate patterns including structure and shape influence the wearing comfort, which is a main factor of athletes' action performance. Sport wears provide different pressure on human body because of their surface tension, and suitable pressure is helpful to athletes to perform actions freely. However, design and adjustment of tight-fitting suits to achieve satisfying press distribution are hard works, because the pressure distribution cannot be measured directly in dynamic process of sport actions. This paper proposes a dynamic virtual try-on simulation framework for speed skating suit. First, a skeleton supported virtual human model with fixed structure skin mesh is created, and the model can be deformed according to speed skating actions. Second, a typical sport suit including 5 parts is created on the human body skin model as a virtual try-on framework, thus all the suit's parts are deformed driven by human model. Finally, deformations of suit parts are calculated frame-by-frame in dynamic process, and the deformation rate is considered as the tense distribution of fabric which can be transformed to pressure by curvature radius. The deformation rate distribution of different area on three-dimensional suit part provides a direct visual reference to the pattern adjustment. 174. Modulations of the South China Sea Ocean Circulation by the Summer Monsoon Intraseasonal Oscillation Inferred from Satellite Observations.

Abstract

The South China Sea (SCS) displays remarkable responses and feedback to the summer monsoon intraseasonal oscillation (ISO). This study investigates how the SCS summer ocean circulation responds to the monsoon ISO based on weekly satellite data. In summer, the largest amplitudes for intraseasonal (30–90 days) sea surface height variations in the SCS occur around the northeastward offshore current off southeast Vietnam between a north–south eddy dipole. Our results show that such strong intraseasonal sea surface height variations are mainly caused by the alternate enhancement of the two eddies of the eddy dipole. Specifically, in response to the intraseasonal intensification of southwesterly winds, the northern cyclonic eddy of the eddy dipole strengthens within 1–2 weeks, and its southern boundary tends to be more southerly. Afterwards, as the wind-driven southern anticyclonic gyre spins up, the southern anticyclonic eddy gradually intensifies and expands its northern boundary northward, while the northern cyclonic eddy weakens and retreats northward. Besides the local wind forcing, westward propagations of the eastern boundary-originated sea surface height anomalies, which exhibit latitude-dependent features that are consistent with the linear Rossby wave theory, play an important role in ocean dynamical adjustments to the monsoon ISO, especially in the southern SCS. Case studies further confirm our findings and indicate that understanding this wind-driven process makes the ocean more predictable on short-term timescales. [ABSTRACT FROM AUTHOR]

This thesis focuses on designing and synthesis of novel molecular imaging probes based on non-covalent funtionalisation of pure single-walled carbon nanotubes (SWNTs). Several synthetic strategies for the supramolecular chemistry functionalisation of SWNTs, cytotoxicity measurements and cellular imaging of supramolecularly functionalised carbon nanotube probes are discussed. Chapter one is a literature review as the thesis Introduction. This describes aspects of the physical and chemical properties, structural importance and synthesis methods of single-walled carbon nanotubes (SWNTs), also opens the discussion of the different functionalisation methods to enhance the solubility and biocompatibility of SWNTs for biomedical applications. Several approaches for the design of functionalisation SWNTs for molecular imaging reported in the current literature are highlighted. Techniques and facilities for accessing cell imaging ability and behavior of these synthesized molecular imaging probes, including confocal laser scanning microscopy (CLSM) and fluorescence-lifetime imaging microscopy (FLIM) are described briefly. Chapter two explores the synthesis of specifically designed naphthalene diimide (NDI). These molecules are known to form 3 dimensional (3D) helical organic nanotubes through hydrogen bonding. In this work an iodine-tagged NDI was allowed to self-assemble onto the surface of SWNTs. The cavities of the NDI organic nanotubes can accommodate SWNTs strands in their hydrophobic interior as observed high-resolution transmission electron microscopy (HR TEM). A new hybrid material, NDI@SWNT, was prepared and characterised as dispersed in organic solvents and aqueous media and in the solid state by HR TEM, tapping mode atomic force microscopy (TM AFM), scanning electron microscopy (SEM), circular dichroism, Raman and fluorescence spectroscopies (steady-state single and two-photon techniques). These measurements indicate that amino acid-functionalised NDI interacts strongly with SWNTs in dispersions and forms a donor-acceptor complex denoted NDI@SWNT. The interaction of this nanohybrid with cancer cells was explored using fluorescence microscopies. Chapter three describes the synthesis of series of molecular imaging agents based on two cancer targeting peptides (bombesin and RGDfK). Two types of NDIpeptide conjugates (Iodine-tagged NDI-Bombesin and Tryptophan-NDI-RGDfK) were designed and synthesized through EDC-coupling method. New compounds synthesized were characterised by mass spectroscopy and also HPLC. Fluorescence lifetime imaging microcopy and confocal laser scanning microscopy were utilised for investigating cellular behaviors (stability, fluorescence intensity and localization) of these molecular imaging probes. Chapter four describes the synthesis of amphiphilic conjugated thiophenes (dodecathiophene, denoted as T12). In this system, the thiophene backbone structure was chosen as a biocompatible coating for carbon nanotubes as simple molecular mechanics modeling suggested that it would be perfectly fitted to the curvature of SWNTs. T12 showed very good capability for debundling of SWNTs and forming corresponding solution dispersions of T12@SWNTs describes the potential of T12@SWNTs as a stable fluorescent bioimaging nano-probe for tracking cancer cells. Chapter five describes the successful filling of SWNTs with Cu2+ by radiochemistry methods (using 'hot' 64Cu ions anchored onto NaOAc) and also by a 'cold' optimised procedure for excess Cu(OAc)2. Filling with other metal ions was also tested, for example KReO4 and Zr(OAc)4. The filling experiments with Zr(OAc)4 in solution did not prove successful at normal pH but results were promising when pH was adjusted to ca. 2 by adding H2SO4. Any significant leakage of metal ions from open SWNTs was avoided by a simultaneous encapsulation of C70 molecules at the ends of SWNTs. Functionalisation of SWNTs by the supramolecular wrapping the surface of SWNTs in aqueous media with a naturally occurring glucan (β-1,3-1,4-Dglucan, denoted here as β-D-glucan) was also explored. Several boronic acid fluorophores were successfully synthesized and tested for the labeling of β-D-glucan @SWNTs by molecular recognition between boronic acids and this polysaccharide. Their cellular translocation behaviour and fluorescence properties were investigated by confocal fluorescence imaging and fluorescence lifetime imaging. Both methods show that localisation in sub-cellular (MCF-7 cells) regions and that the glucan coating significantly enhances the cell membrane translocation of SWNTs. Chapter six reports an efficient and economical strategy of supramolecular complex formulation of thermally reduced graphene. Naphthalene diimide (NDI) was used to form a stable and energy transfer complex which showed efficient quenching and significant red-shift of fluorescence of NDI when adsorbed onto graphene surfaces. The effect of thermally reducing annealing procedure to convert graphene oxides in graphene-nanoflake like materials was investigated. A new hybrid material (denoted here as NDI@TRG) synthesized was characterised by transmission electron microscopy (TEM), Raman spectroscopy, thermogravimetric analysis (TGA) and fluorescence microscopy in the dispersed phase Chapter seven contains full experimental details for the work described in this thesis. The Appendix contains details of the crystallographic data and supplementary information on cell imaging photos and fluorescence lifetime point decay data for SWNT nanocomposites.

Abstract

The effects of ash and sulfur content on the morphology of particulate matter (PM) in diesel particle filter (DPF) were investigated with five different components of lubricants. The aggregate morphology of primary particles in diesel were analyzed using transmission electron microscopy (TEM). The fractal dimensions of carbon particles were calculated by box-counting method (BCM), differential box-counting method (DBC), relative differential box-counting method (RDBC) and MAD-based box counting method (MAD-DBC), and the results were compared. The results showed that the microstructure of PM developed from chain-like structure to agglomerate structure with the increase of sulfur and ash content in lubricating oil. The fractal dimension of carbon particles increased with the increase of sulfur and ash content. The SSE of RDBC fitting results was smaller, and the R-square is larger. MAD-DBC fitting results had stronger anti-noise interference performance.

Abstract

The filtration efficiencies of the particulate matter and the particulate number in DPF, the no-load pressure drop of DPF, and the load pressure drop of DPF were selected as key performance parameters of DPF. The influence of the cell density and the wall thickness of the DPF on these key performance parameters of DPF was studied in detail. Firstly, a calculation model of DPF was established, and the compensation mathematical model was added. Secondly, the key performance parameters of DPF were defined. Finally, based on the model, the key performance parameters are numerically investigated under different cell density and wall thickness of the DPF. Consequently, with the increase of cell density of DPF, the filtration efficiency of DPF increases, the load pressure drop of DPF decreases, but the no-load pressure drop of DPF increases. With the decrease of the wall thickness of the DPF, the no-load pressure drop of the DPF decreases, the load pressure drop of the DPF decreases, but the filtration efficiency of the DPF decreases. The most suitable combination scheme is a cell density of 200-300 CPSI and a wall thickness of 11-16 mil.
