158. An in vivoscreening system to identify tumorigenic genes

Abstract

Screening for oncogenes has mostly been performed by in vitrotransformation assays. However, some oncogenes might not exhibit their transforming activities in vitrounless putative essential factors from in vivomicroenvironments are adequately supplied. Here, we have developed an in vivoscreening system that evaluates the tumorigenicity of target genes. This system uses a retroviral high-efficiency gene transfer technique, a large collection of human cDNA clones corresponding to ~70% of human genes and a luciferase-expressing immortalized mouse mammary epithelial cell line (NMuMG-luc). From 845 genes that were highly expressed in human breast cancer cell lines, we focused on 205 genes encoding membrane proteins and/or kinases as that had the greater possibility of being oncogenes or drug targets. The 205 genes were divided into five subgroups, each containing 34–43 genes, and then introduced them into NMuMG-luc cells. These cells were subcutaneously injected into nude mice and monitored for tumor development by in vivoimaging. Tumors were observed in three subgroups. Using DNA microarray analyses and individual tumorigenic assays, we found that three genes, ADORA2B, PRKACBand LPAR3, were tumorigenic. ADORA2Band LPAR3encode G-protein-coupled receptors and PRKACBencodes a protein kinase A catalytic subunit. Cells overexpressing ADORA2B, LPAR3or PRKACBdid not show transforming phenotypes in vitro, suggesting that transformation by these genes requires in vivomicroenvironments. In addition, several clinical data sets, including one for breast cancer, showed that the expression of these genes correlated with lower overall survival rate.

Abstract

Ion heating experiments are performed in a fast flowing plasma with an ion Mach number of nearly unity. RF waves with an ion cyclotron range of frequency is excited by a pair of loop antennas located at a divergent magnetic nozzle. Increase of plasma thermal energy W(perpendicular to) measured by a diamagnetic coil is observed when the waves are excited with various azimuthal mode numbers in several magnetic nozzle configurations. It is most effective to heat ions of plasma flow to excite the waves with an azimuthal mode number of m= 1. Abstract

Malignant fibrous histiocytoma (MFH) was induced in rats by injection of 9,10-dimethyl-1,2-benzanthracene. Using cell suspensions prepared from the heterotransplanted nude mouse tumor as immunogen, a monoclonal antibody, (MAb), MEP-1, against fibroblastlike MFH tumor cells was generated. In the primary rat tumors and transplanted rat or nude mouse tumors, MEP-1 reacted specifically with the fibroblastlike cells but not with the histiocytelike cells or xanthoma cells. Anti-rat macrophage MAbs RM-1 and TRPM-3 did not stain the fibroblastlike cells, but both were reactive with the histiocytelike cells. Double stainings with both MEP-1 and RM-1 or TRPM-3 did not detect any double positive cells. Immunoelectron microscopy using these MAbs showed that the fibroblastlike cells were the major cell component of the primary and transplanted rat tumors and that their cell membrane was stained positively with MEP-1, but not for RM-1 or TRPM-3. By the double staining method using a MAb against prolyl 4-hydroxylase beta and MEP-1 or TRPM-3, this enzyme was demonstrated in MEP-1-positive cells but not in TRPM-3-positive cells. Results obtained by DNA cytofluorometry with 4,6-diamidino-2-phenylindole dihydrochloride staining or by the combined method of DNA cytofluorometry and indirect immunofluorescence, using MEP-1, RM-1, and TRPM-3, indicate that MEP-1-positive cells are neoplastic cells of rat MFH having proliferation activity. In the transplanted nude mouse tumors, no differentiation of MEP-1-positive rat tumor cells into histiocytelike cells was detected, and all histiocytelike cells were immunostained by F4/80 and most of them were positive for M5/114. These results suggest that fibroblastlike cells and intermediate cells are tumor cells of 9,10-dimethyl-1,2-benzanthracene-induced rat MFH showing differentiation toward fibroblasts and that histiocytelike cells are infiltrated macrophages.

Abstract

For waterfront development in inner bays, it is essential to improve water quality so as to meet social needs for comfortable coast environments. A concept of creating a new water-affinity space along a shallow coast is discussed with the consideration of sea surface as an attractive environmental resource. Economical water purification should harness natural energies (tide, waves, wind, etc.) as well as biological activities (microbial film, macrophytes on tidal marsh, shells, etc.).
