Bronchiolitis obliterans syndrome ( BOS) is associated with lack of immunosuppression of T cell proinflammatory cytokines and increased T cell granzyme B. Repeated antigen-driven proliferation down-regulates T cell CD28. We hypothesized that down-regulation of CD28 and up-regulation of alternate co-stimulatory molecules ( CD134, CD137, CD152 and CD154) on T cells may be associated with BOS. Co-stimulatory molecules, granzyme B, perforin and intracellular cytokines were measured by flow cytometry on T cells from stable lung transplant patients ( n = 38), patients with BOS ( n = 20) and healthy controls ( n = 10). There was a significant increase in the percentage of CD4/28null and CD8/28null T cells producing granzyme B, interferon ( IFN)-γ and tumour necrosis factor ( TNF)-α in BOS compared with stable patients. Down-regulation of CD28 was associated with steroid resistance and up-regulation of CD134, CD137, CD152 and CD154 on CD4+ T cells and CD137 and CD152 on CD8+ T cells. There was a significant correlation between increased CD28null/ CD137 T cells producing IFN-γ, TNF-α with BOS grade ( r = 0·861, P < 0·001 for CD28null/ CD137 IFN-γ/ CD8) and time post-transplant ( r = 0·698, P < 0·001 for CD28null/ CD137 IFN-γ/ CD8). BOS is associated with down-regulation of CD28 and up-regulation of alternate co-stimulatory molecules on steroid-resistant peripheral blood proinflammatory CD4+ and CD8+ T cells. Therapeutic targeting of alternate co-stimulatory molecules on peripheral blood CD28null T cells and monitoring response using these assays may help in the management of patients with BOS. Summary Chronic obstructive pulmonary disease (COPD) is a chronic inflammatory lung disease; it is a leading cause of death and existing treatments have no proven disease-modifying effect. The mechanisms underlying this resistance are largely unknown, but suggest the presence of some self-maintaining pathogenic process, possibly initiated by cigarette smoking, that prevents the normal resolution of inflammation. We have previously reported increased production of proinflammatory cytokines and granzyme b by CD8+ T cells in COPD; costimulatory receptor/ligand interactions required include CD80:86/CD28, B7-1/CTLA4, 4-1BB/1BBL and OX40/OX40L. We hypothesized that a dysregulated expression/function of these molecules may play a role in inflammatory/autoimmune components of COPD. We analysed T cell co-stimulatory molecules in blood from 34 controls, 15 smokers and 48 COPD subjects. We assessed the potential functional relevance of CD8/CD28null cells in COPD by measuring their production of proinflammatory cytokines, co-stimulatory molecules, granzyme and perforin. A smoke-exposed murine model was applied to investigate the relative expression of CD8/CD28null T cells in blood, lung tissue and airway. CD8/CD28null cells were increased in both current- and ex-smoker COPD groups; these cells expressed significantly more interferon (IFN)-γ, OX40, 4-1BB, CTLA4, granzyme and perforin when stimulated than CD8/CD28+ T cells. There were no changes in CD4/CD28null T cells. In mice exposed to cigarette smoke for 12 weeks, CD8/CD28null T cells were significantly increased in the airway with a trend for an increase in lung tissue and blood. Increased production of proinflammatory cytokines and expression of alternative co-stimulatory molecules by CD8/CD28null T cells may play a role in inflammatory or autoimmune responses in COPD and identify therapeutic targets. Eucalyptus urophylla is one of the most commercially important forest species in the world, primarily as a hybrid parent. However, the conservation status of the majority of the populations where it naturally occurs on seven islands in eastern Indonesia range from critically endangered to vulnerable. We examine the evolutionary forces that might have caused genetic variation within and between E. urophylla populations and link these findings to international provenance trial results and molecular marker studies. We demonstrate that one climatic type does not describe all E. urophylla populations. We suggest that volcanism played an important role in its distribution and levels of genetic diversity on the islands. We report significant provenance variation for survival and growth in trials established in Brazil, Colombia, Mexico, South Africa and Venezuela. Provenance differences for growth within a country are sometimes >50%. Based on these assessments, we develop an ex situ conservation strategy to protect populations that are most threatened but also show the greatest productivity across countries. We conclude that populations of E. urophylla will only be conserved ex situ if traditional and molecular tree breeders convince private industry of the economic importance to do so. Diagnosis of congenital or neonatal infection is often based on clinical signs. However, clinical symptoms of infections may not be specific, and for this reason early diagnosis is often determined on results of laboratory tests, which may not currently be adequate. A more reliable method of detection of infection may be the demonstration of activated lymphocytes, which can be conducted rapidly and before the isolation of the infected organism. We have shown that detection of up-regulation of CD45RO, an activated/memory isoform of CD45 present on T cells, provides a reasonably sensitive screening test for neonatal infection. We also showed that dual expression of CD45RA/CD45RO was up-regulated early during the infective process in neonates with documented infection. However, other leucocytes are also activated during the infective process. To improve the sensitivity of the neonatal infection screening test and to identify the types of leucocytes involved in the immune response to the infective organism, we studied further the up-regulation of a comprehensive range of surface activation markers on T cells, monocytes and natural killer (NK) cells from a group of 17 newborn patients with positive culture, a group of 40 possibly infected patients based on clinical signs and a control group. ‘Normal’ ranges were established for each activation marker for each leucocyte subset from 1 to 7 and 7-14-day-old newborns <35 weeks’ gestation and 35-40 weeks’ gestation. There was a significant increase in the percentage of T cells expressing CD25 in the peripheral blood from infants at 2 weeks of age. Expression of HLA-DR on T cells, CD25 and CD69 on monocytes and HLA-DR on NK cells was also increased significantly in the peripheral blood from infants at 2 weeks of age and may reflect a maturation of these functional surface molecules. Up-regulation of CD69 on NK cells was the most sensitive marker for neonatal sepsis (positive in 13/16 patients). CD69 and CD25 expression was increased significantly on T cells in 11/17 and 10/17 patients, respectively. A combination of CD45RA/CD45RO and CD45RO identified 11/16 infected patients. Measurement of CD69 expression on NK cells with CD45RA, CD45RO, CD25 and CD69 expression on T cells resulted in a significant increase in at least two leucocyte activation markers from infected patients. In conclusion, this is the first report of the up-regulation of CD69 on NK cells as a sensitive marker of neonatal infection. (1) Salt sensitive hypertension, which occurs as a result of treatment with nitric oxide synthase inhibitors, is associated with a loss of the usual down-regulatory effect of dietary sodium on angiotensin II (Ang II) synthesis. In the spontaneous hypertensive rat (SHR), which suffers a relative NO deficiency, the hypertension is in part salt sensitive. We sought to determine therefore whether the salt sensitive component to the hypertension was associated with a loss of the regulatory effect of dietary sodium on Ang II synthesis. (2) Male SHR were placed on low, intermediate or high salt diets for 4 weeks and their blood pressure recorded. After 4 weeks, blood was collected for determination of renin, angiotensinogen, Ang I, Ang II and aldosterone concentrations, as well as ACE activity. (3) The increase in systolic blood pressure in rats on the high salt diet was significantly greater than in those on the low (P < 0.005) and intermediate salt diets (P < 0.0005). Plasma renin and aldosterone concentrations and ACE activity decreased with increasing dietary sodium. However, the concentrations of Ang II and angiotensinogen both increased in the rats on the high salt diet (Ang II: P < 0.05; angiotensinogen: P < 0.05). (4) We conclude that the hypertension in the SHR is in part salt sensitive and that this salt sensitive component is associated with a loss of the normal down-regulatory effect of dietary sodium on Ang II and angiotensinogen synthesis. Inhibitor antibody formation in patients with haemophilia receiving factor VIII (FVIII) concentrate is a serious problem. T helper type 2 (Th2) cytokines are necessary for antibody production by B cells and have been shown to be produced predominantly by CD30+/CD45RO+/CD3+ cells. We have previously shown that the Th2 cytokine, interleukin (IL)-6, is inhibited but IL-10 is upregulated, in the presence of plasma-derived FVIII (pdFVIII). To clarify further the overall effect of FVIII on Th2 cytokine production, the percentage of T cells expressing the CD30+/CD45RO+/CD3+ Th2 phenotype was studied over 72 h and the production of the Th2 cytokines, IL-4 and IL-5, determined at 24 h in the presence of FVIII following whole-blood stimulation using multiparameter flow cytometry. The production of IL-4 and IL-5 by T cells was significantly inhibited in the presence of pdFVIII. The percentage of CD30+/CD45RO+/CD3+ increased with stimulation of whole blood cultures over 72 h but was significantly inhibited by the presence of pdFVIII or TGF-β at 72 h. The combined inhibitory effect of prednisolone (a commonly used immunosuppressive agent used to treat patients with inhibitors) with pdFVIII on T-cell CD30+/CD45RO+ upregulation, was additive. There was no significant alteration in Th2 cytokine production or phenotype noted in the presence of recombinant FVIII (rFVIII) concentrate. Neutralizing antibody to TGF-β significantly abrogated the inhibitory effects of pdFVIII on Th2 upregulation, indicating TGF-β to be a major inhibitory component of pdFVIII on Th2 cytokine production. The cellular immune system of the newborn infant is immature and hypo-responsive when compared with adults. The extent to which immaturity of the leucocyte function underlies hyporesponsiveness in the newborn is incompletely understood. In this study flow cytometric techniques were applied to investigate the concurrent expression of a range of surface and intracellular leucocyte functional molecules and cytokines in resting and stimulated cord and adult blood. Production of interleukin (IL)-2 and expression of the components of its receptor, IL-2Rα/β/γ, were investigated. No differences in the proportion of leucocytes producing IL-2Rα and IL-2Rγ were observed for newborns and adults. A lower proportion of T cells and natural killer (NK) cells from newborns expressed IL-2Rβ and upregulation of expression was slower. We hypothesize that reduced IL-2Rβ may curtail early autocrine IL-2 activation of immune responses in the newborn. This hypothesis was supported by the observation that an increased proportion of stimulated T cells from newborns produced IL-2 at 4 h poststimulation, but at 24 h the proportion was lower than for adult T cells. The very low levels of interferon (IFN)-γ produced by neonatal T cells and NK cells may also be partly explained by a curtailment of early autocrine activation of T cells. Expression and kinetics of upregulation for other functional molecules were studied. CD71, HLA-DR, tissue factor and CD152 levels were not significantly different for adults and newborns, suggesting that cord blood leucocytes, in some respects, may demonstrate functional maturity. IL-6 secretion by stimulated monocytes was also comparable in cord and adult blood. However, IL-1α and IL-1β were produced by a lower proportion of monocytes from newborns than adults. Similarly, tumour necrosis factor (TNF)-α production for monocytes and T cells was lower in cord blood. The mean fluorescence intensity for IL-1α, IL-1β and TNF-α was also lower for leucocytes from cord blood. These findings are significant in relation to the inability of newborn infants to mount a febrile response to infection. The findings of lower expression of IL-2Rβ and lower production of inflammatory cytokines IL-1α, IL-1β and TNF-α is a basis for improved understanding of the immunological immaturity of leucocytes in the newborn. Inhibitor formation in patients with haemophilia receiving factor VIII (FVIII) concentrate is a common problem requiring tolerance induction therapy. Immune tolerance is dependent on defective T cell/antigen-presenting cell (APC) interactions and inhibitor antibody formation is associated with effective T-cell/B-cell interaction. We studied the expression of the cell-surface molecules involved with these interactions using multiparameter flow cytometry and a whole blood stimulation assay–phytohaemaglutinin (PHA) to activate T cells and Escherichia coli lipopolysaccharide (LPS) to activate monocytes and B cells. Up-regulation of T-cell co-stimulatory receptors CD11a, CD40 ligand (CD40L) and CTLA4 were inhibited in a dose-dependent manner by plasma-derived (pd)FVIII, but CD28 was unchanged. Up-regulation of monocyte and B-cell co-stimulatory ligands CD4O, B7-1 (CD80) and B7-2 (CD86) were also inhibited in a dose-dependent manner by pdFVIII, but LFA-3 (CD58) was unchanged. The combined inhibitory effect of prednisolone, an immunosuppressive agent used in several tolerance induction protocols, with pdFVIII on co-stimulatory molecules, was additive. There was no significant alteration in T-cell/APC adhesion or co-stimulatory molecules noted in the presence of recombinant (rh)FVIII concentrate. The inhibitory effect of pdFVIII on molecules involved in interaction between T cells and APCs may result in immune tolerance in recipients of pdFVIII concentrate. The inhibitory effect of pdFVIII on CD40/CD40L up-regulation may result in defective antibody formation. Diagnosis of perinatal infection in the newborn is difficult; there may be few clinical signs and current tests are slow or non-specific. Detection of organisms, antigen or specific antibody to common pathogens often requires repeat samples and does not give immediate results. Haematological parameters, although relied upon frequently to diagnose infection in the neonate prior to a positive bacterial isolation, are unreliable and insensitive. Indicators such as an increase in neutrophil band cell counts are highly variable between morphologists. Infection induces the expression of a number of T lymphocyte surface markers, including CD45RA/CD45RO and CD45RO. The use of changed expression of surface markers as a laboratory test for detection of infection in neonates was evaluated. We used multiparameter flow cytometry to detect expression of early (CD45RA/CD45RO) and late (CD45RO) activation markers. In the respective groups of 50 full term (including 25 normal vaginal deliveries and 25 caesarean deliveries) and 30 premature, i.e. < 36 weeks gestation (born by either normal vaginal delivery or caesarean delivery) the CD45RA isoform was brightly expressed on newborn ‘naive’ CD4+ T cells, whereas the CD45RO isoform (including both ‘bright’ and ‘dim’ populations) was present on < 19% of CD4+ T cells from these newborn infants. In a group of 37 infants, tested to evaluate possible effects of non-infective parameters such as respiratory distress and iso-immunization, no significant changes in surface marker expression were found and specificity of the test was confirmed. In 14 neonates with documented sepsis, up-regulation of dual staining CD45RA/CD45RO isoforms on CD4+ T cells was detected early in the infection. The INTELSAT Operations Center TDMA facility (IOCTF) was built by COMSAT Laboratories to monitor and control the operation of the INTELSAT time-division multiple-access (TDMA) system. This paper describes the overall monitoring and control requirements, and the role played by the IOCTF in satisfying those requirements. Genetic-parameter estimates and parental breeding-value predictions were compared from open-pollinated and control-pollinated progeny populations of Eucalyptus globulus and two populations of E. nitens. For E. globulus there were two types of open-pollinated populations (native stand open-pollinated and seed orchard open-pollinated) and two types of control-pollinated populations (intra-provenance and interprovenance full-sib families). For E. nitens there were two populations, a seed orchard open-pollinated population and intra-provenance full-sib families. Progeny tests were established across multiple sites and 2-year height and diameter were measured and volume calculated. Genetic parameters from native stand open-pollinated E. globulus were unlike the parameters from the other three E. globulus populations; heritability estimates were severely inflated, presumably due to high levels, and possibly differential levels, of inbreeding depression relative to the other populations. Estimates of dominance variance in the E. globulus full-sib populations were high, but were zero in the E. nitens population. Correlations among parental breeding values, predicted using data from the different populations, were generally low and non-significant, with two exceptions: predictions from the two E. globulus full-sib populations were significantly correlated (r=0.54, P = 0.001), as were predictions from the E. nitens seed orchard OP and full-sib population (r = 0.61, P = 0.08). There was some indication that superior parents of E. globulus native stand open-pollinated families also tended to have above-average breeding values based on the performance of intra-provenance full-sib offspring. The application and underlying assumptions of formulae used to estimate the variance of variance components and ratios of variance components are fully described for (1) variance components estimated using Henderson's Method 3 (HM3) and Restricted Maximum Likelihood (REML) and (2) ratios of variance components commonly used in genetic tests - biased and unbiased heritabilities. A first-order Taylor series approximation is often used to estimate the variance of a ratio of two random variables (e.g., heritability), however the formula is complicated, thus making calculations prone to errors. Dickerson's approximation is considerably simpler, though relatively rarely used. In case studies using data from 148 slash pine full-sib progeny tests, Dickerson's method was found to be slightly more conservative than the Taylor series approximation when estimating the variance of heritability estimates, regardless of test size, age, or the trait (volume, which is a continuous trait, and rust resistance, which is a bernoulli trait). Both the Taylor series and Dickerson approximations compared favorably with an empirical estimate of the variance of heritability estimates, however there is some evidence of small-sample bias associated with the use of the asymptotic variance-covariances from REML variance component estimation. Computer simulation was used to compare minimum variance quadratic estimation (MIVQUE), minimum norm quadratic unbiased estimation (MINQUE), restricted maximum likelihood (REML), maximum likelihood (ML), and Henderson's Method 3 (HM3) on the basis of variance among estimates, mean square error (MSE), bias and probability of nearness for estimation of both individual variance components and three ratios of variance components. The investigation also compared three procedures for dealing with negative estimates and included the use of both individual observations and plot means as the experimental unit of the analysis. The structure of data simulated (field design, mating designs, genetic architecture and imbalance) represented typical analysis problems in quantitative forest genetics. Results of comparing the estimation techniques demonstrated that: estimates of probability of nearness did not discriminate among techniques; bias was discriminatory among procedures for dealing with negative estimates but not among estimation techniques (except ML); sampling variance among estimates was discriminatory among procedures for dealing with negative estimates, estimation techniques and unit of observation; and MSE provided no additional information to variance of the estimates. HM3 and REML were the closest competitors under these criteria; however, REML demonstrated greater robustness to imbalance. Of the three negative estimate procedures, two are of practical significance and guidelines for their application are presented. - Many patients with Peyronie's disease suffer severe penile curvature, impotence, and respond poorly to conservative management. Sixteen patients with intractable Peyronie's disease had surgical incision of Peyronie's plaque and insertion of semi-rigid or inflatable penile implants. Patients had satisfactory results with elimination of curvature and resumption of coitus.
