A series of tetraamino-bisthiourea chiral macrocycles containing two diarylthiourea and two chiral diamine units were synthesized by a fragment-coupling approach in high yields. Different chiral diamine units, including cyclohexanediamines and diphenylethanediamines were readily incorporated by both homo and hetero [1 + 1] macrocyclic condensation of bisamine and bisisothiocyanate fragments. With the easy synthesis, gram-scale of macrocycle products can be readily obtained. These chiral macrocycles were applied in catalyzing bioinspired decarboxylative Mannich reactions. Only 5 mol % of the optimal macrocycle catalyst efficiently catalyzed the decarboxylative addition of a broad scope of malonic acid half thioesters to isatin-derived ketimines with excellent yields and good enantioselectivity. The rigid macrocyclic framework and the cooperation between the thiourea and tertiary amine sites were found to be crucial for achieving efficient activation and stereocontrol. As shown in control experiments, catalysis with the acyclic analogues having the same structural motifs were non-selective.

A thorough evaluation of the recently released Global Satellite Mapping of Precipitation (GSMaP) is critical for both end-users and algorithm developers. In this study, six products from three versions of GSMaP version 8, including real time (NOW-R and NOW-C), near real time (NRT-R and NRT-C), and post-real time (MVK-R and MVK-C), are systematically and quantitatively evaluated based on time-by-time observations from 2167 stations in mainland China. Among each version, both products with and without gauge correction are adopted to detect the gauge correction effect. Error quantification is carried out on an hourly timescale. Three common statistical indices (i.e., correlation coefficient (CC), relative bias (RB), and root mean square error (RMSE)) and three event detection capability indices (i.e., probability of detection (POD), false alarm ratio (FAR), and critical success index (CSI)) were adopted to analyze the inversion errors in precipitation amount and precipitation event frequency across the various products. Additionally, in this study, we examine the dependence of GSMaP errors on rainfall intensity and elevation. The following main results can be concluded: (1) MVK-C exhibits the best ability to retrieve rainfall on the hourly timescale, with higher CC values (0.31 in XJ to 0.47 in SC), smaller RMSE values (0.14 mm/h in XJ to 0.99 mm/h in SC), and lower RB values (−4.78% in XJ to 16.03% in NC). (2) Among these three versions, the gauge correction procedure plays a crucial role in reducing errors, especially in the post-real-time version. After being corrected, MVK-C demonstrates an obvious CC value improvement (>0.3 on the hourly timescale) in various sub-regions, increasing the percentage of sites with CC values above 0.5 from 0.03% (MVK-R) to 28.47% (MVK-C). (3) GSMaP products generally exhibit error dependencies on precipitation intensity and elevation, particularly in areas with drastic elevation changes (such as 1200–1500 m and 3000–3300 m), where the accuracy of satellite precipitation estimates is significantly affected. (4) CC values decreased with an increasing rainfall intensity; RB and RMSE values increased with an increasing rainfall intensity. The results of this study may be helpful for algorithm developers and end-users and provide a scientific reference for different hydrological applications and disaster risk reduction.

The spatial distribution of gas emitted from an odor source provides valuable information regarding the composition, size, and localization of the odor source. Surface-enhanced Raman scattering (SERS) gas sensors exhibit ultra-high sensitivity, molecular specificity, rapid response, and large-area detection. In this paper, a SERS gas sensor array was developed for visualizing the spatial distribution of gas evaporated from benzaldehyde and 4-ethylbenzaldehyde odor sources. The SERS spectra of the gas were collected by scanning the sensor array using an automatic detection system. The non-negative matrix factorization algorithm was employed to extract feature and concentration information at each spot on the sensor array. A heatmap image was generated for visualizing the gas spatial distribution using concentration information. Gaussian fitting was applied to process the image for localizing the odor source. The size of the odor source was estimated using the processed image. Moreover, the spectra of benzaldehyde, 4-ethylbenzaldehyde, and their gas mixture were simultaneously detected using one SERS sensor array. The feature information was recognized using a convolutional neural network with an accuracy of 98.21%. As a result, the benzaldehyde and 4-ethylbenzaldehyde odor sources were identified and visualized. Our research findings have various potential applications, including odor source localization, environmental monitoring, and healthcare.

Thermal crosstalk between array structures is a key factor in limiting the sensitivity of micro-nano array sensors. We propose a two-stage thermally isolated structure with thermal holes and heat dissipation layer and pulsed voltage heating to reduce thermal crosstalk. Through finite element thermal simulation analysis as well as thermal interference test, the results show that the thermal crosstalk of the two-stage structure is reduced by 12.89% and 39.67%, respectively, in the steady state compared to the structure with no thermal isolation, and pulsed voltage heating leads to the thermal crosstalk of the two-stage structure to be

This paper reviews the researches on boiler combustion optimization, which is an important direction in the field of energy saving and emission reduction. Many methods have been used to deal with boiler combustion optimization, among which evolutionary computing (EC) techniques have recently gained much attention. However, the existing researches are not sufficiently focused and have not been summarized systematically. This has led to slow progress of research on boiler combustion optimization and has obstacles in the application. This paper introduces a comprehensive survey of the works of intelligent optimization algorithms in boiler combustion optimization and summarizes the contributions of different optimization algorithms. Finally, this paper discusses new research challenges and outlines future research directions, which can guide boiler combustion optimization to improve energy efficiency and reduce pollutant emission concentrations.

When a steel box girder is constructed using the jacking method, the contact area between the jack and the bottom of the girder is subjected to complex forces, and it is very critical to ensure the local stability of the girder. When the phenomenon of unsynchronized jacking occurs, it will lead to changes in the contact area and affect the structural safety. In order to solve the above problems, this paper takes the background of the incremental launching construction of the main bridge across the Yellow River on Jiao Ping Expressway, adopts the Midas FEA NX 2021 finite element software to establish a finite element hybrid unit model under the maximum cantilever condition for the first time, and analyzes the local stresses in this state. The results show that the local maximum equivalent stress of the steel box girder is 198.301 MPa, which meets the requirements. The effect of jacking asynchrony on the structural forces is analyzed by simulating jacking asynchrony in the local model. The results show that both vertical jacking asynchrony and lateral deflection will lead to an increase in local stresses in the steel box girder and even steel yielding. On the basis of the above single-parameter study, a two-parameter correlation analysis is carried out to obtain the two-parameter control equation of jacking, the control threshold of the vertical jacking height difference is formulated to be 15 mm, and the dynamic control of lateral deflection is realized according to the control equation. Through comparison, it is found that the two-parameter control threshold of jacking synchronization is reduced, which can supplement the unfavorable state missed during single-parameter control and is a safer and more effective means of control.

To reduce the economic losses caused by debt evasion amongst lost-link borrowers (LBs) and improve the efficiency of finding information on LBs, this paper focuses on the cross-platform information collaborative search optimization problem for LBs. Given the limitations of platform/system heterogeneity, data type diversity, and the complexity of collaborative control in cross-platform information search for LBs, a collaborative search model for LBs’ information based on multi-agent technology is proposed. Additionally, a multi-agent Q-learning algorithm for the collaborative scheduling of multi-search subtasks is designed. We use the Q-learning algorithm based on function approximation to update the description model of the LBs. The multi-agent collaborative search problem is transformed into a reinforcement learning problem by defining search states, search actions, and reward functions. The results indicate that: (i) this model greatly improves the comprehensiveness and accuracy of the search for key information of LBs compared with traditional search engines; (ii) during searching for the information of LBs, the agent is more inclined to search on platforms and data types with larger environmental rewards, and the multi-agent Q-learning algorithm has a stronger ability to acquire information value than the transition probability matrix algorithm and the probability statistical algorithm for the same number of searches; (iii) the optimal search times of the multi-agent Q-learning algorithm are between 14 and 100. Users can flexibly set the number of searches within this range. It is significant for improving the efficiency of finding key information related to LBs.

Femtosecond laser drilling is extensively used to create film-cooling holes in aero-engine turbine blade processing. Investigating and exploring the impact of laser processing parameters on achieving high-quality holes is crucial. The traditional trial-and-error approach, which relies on experiments, is time-consuming and has limited optimization capabilities for drilling holes. To address this issue, this paper proposes a process design method using machine learning and a genetic algorithm. A dataset of percussion drilling using a femtosecond laser was primarily established to train the models. An optimal method for building a prediction model was determined by comparing and analyzing different machine learning algorithms. Subsequently, the Gaussian support vector regression model and genetic algorithm were combined to optimize the taper and material removal rate within and outside the original data ranges. Ultimately, comprehensive optimization of drilling quality and efficiency was achieved relative to the original data. The proposed framework in this study offers a highly efficient and cost-effective solution for optimizing the femtosecond laser percussion drilling process.

Background: Corneal fluorescein staining is a key biomarker in evaluating dry eye disease. However, subjective scales of corneal fluorescein staining are lacking in consistency and increase the difficulties of an accurate diagnosis for clinicians. This study aimed to propose an automatic machine learning-based method for corneal fluorescein staining evaluation by utilizing prior information about the spatial connection and distribution of the staining region. Methods: We proposed an end-to-end automatic machine learning-based classification model that consists of staining region identification, feature signature construction, and machine learning-based classification, which fully scrutinizes the multiscale topological features together with conventional texture and morphological features. The proposed model was evaluated using retrospective data from Beijing Tongren Hospital. Two masked ophthalmologists scored images independently using the Sjögren’s International Collaborative Clinical Alliance Ocular Staining Score scale. Results: A total of 382 images were enrolled in the study. A signature with six topological features, two textural features, and two morphological features was constructed after feature extraction and selection. Support vector machines showed the best classification performance (accuracy: 82.67%, area under the curve: 96.59%) with the designed signature. Meanwhile, topological features contributed more to the classification, compared with other features. According to the distribution and correlation with features and scores, topological features performed better than others. Conclusions: An automatic machine learning-based method was advanced for corneal fluorescein staining evaluation. The topological features in presenting the spatial connectivity and distribution of staining regions are essential for an efficient corneal fluorescein staining evaluation. This result implies the clinical application of topological features in dry-eye diagnosis and therapeutic effect evaluation.

Abstract The gut microbes play important roles in human longevity and the gut microbiota profile of centenarians shows some unique features from young adults. Nowadays, most microbial studies on longevity are commonly based on metagenomic sequencing which may lose information about the functional microbes with extremely low abundance. Here, we combined in-depth metagenomic sequencing and large-scale culturomics to reveal the unique gut microbial structure of a Chinese longevity population, and to explore the possible relationship between intestinal microbes and longevity. Twenty-five healthy Hainan natives were enrolled in the study, including 12 centenarians and 13 senior neighbors. An average of 51.1 Gb raw sequencing data were obtained from individual fecal sample. We assembled 1778 non-redundant metagenomic assembled genomes (MAGs), 33.46% of which cannot be classified into known species. Comparison with the ordinary people in Hainan province, the longevous cohort displayed significantly decreased abundance of butyrate-producing bacteria and largely increased proportion of Escherichia coli, Desulfovibrio piger and Methanobrevibacter smithii. These species showed a constant change with aging. We also isolated 8,030 strains from these samples by large-scale culturomics, most of which belonged to 203 known species as identified by MALDI-TOF. Surprisingly, only 42.17% of the isolated species were also detected by metagenomics, indicating obvious complementarity between these two approaches. Combination of two complement methods, in-depth metagenomic sequencing and culturomics, provides deeper insights into the longevity-related gut microbiota. The uniquely enriched gut microbes in Hainan extreme decades population may help to promote health and longevity.

Abstract Background Epithelial-to-mesenchymal transition (EMT) is a process linked to metastasis and drug resistance with non-coding RNAs (ncRNAs) playing pivotal roles. We previously showed that miR-100 and miR-125b, embedded within the third intron of the ncRNA host gene MIR100HG, confer resistance to cetuximab, an anti-epidermal growth factor receptor (EGFR) monoclonal antibody, in colorectal cancer (CRC). However, whether the MIR100HG transcript itself has a role in cetuximab resistance or EMT is unknown. Methods The correlation between MIR100HG and EMT was analyzed by curating public CRC data repositories. The biological roles of MIR100HG in EMT, metastasis and cetuximab resistance in CRC were determined both in vitro and in vivo. The expression patterns of MIR100HG, hnRNPA2B1 and TCF7L2 in CRC specimens from patients who progressed on cetuximab and patients with metastatic disease were analyzed by RNAscope and immunohistochemical staining. Results The expression of MIR100HG was strongly correlated with EMT markers and acted as a positive regulator of EMT. MIR100HG sustained cetuximab resistance and facilitated invasion and metastasis in CRC cells both in vitro and in vivo. hnRNPA2B1 was identified as a binding partner of MIR100HG. Mechanistically, MIR100HG maintained mRNA stability of TCF7L2, a major transcriptional coactivator of the Wnt/β-catenin signaling, by interacting with hnRNPA2B1. hnRNPA2B1 recognized the N6-methyladenosine (m6A) site of TCF7L2 mRNA in the presence of MIR100HG. TCF7L2, in turn, activated MIR100HG transcription, forming a feed forward regulatory loop. The MIR100HG/hnRNPA2B1/TCF7L2 axis was augmented in specimens from CRC patients who either developed local or distant metastasis or had disease progression that was associated with cetuximab resistance. Conclusions MIR100HG and hnRNPA2B1 interact to control the transcriptional activity of Wnt signaling in CRC via regulation of TCF7L2 mRNA stability. Our findings identified MIR100HG as a potent EMT inducer in CRC that may contribute to cetuximab resistance and metastasis by activation of a MIR100HG/hnRNPA2B1/TCF7L2 feedback loop.

Abstract The spatial inequilibrium phenomenon is apparent during China’s rapid urbanization in the past four decades. As the fertility rate decreases and the population ages, this phenomenon will become more critical. To accurately forecast the future economic development of China, it is necessary to quantify the attractiveness of individual cities. This study introduces web search data to quantify the attractiveness of cities with a fine spatial scale (prefecture-level city) and relatively long-term span (nine years). Results confirm that the estimated city attractiveness can unravel a city’s capability to attract labor force, and suggest that tourism and health care functions of a city have a positive effect to the city’s attractiveness. Additionally, China’s north-south gap in economic development has been widened in the past decade, and 11 cities with nationwide influence have prosperous development potential. This study provides a new lens for predicting China’s economic development, as well as its spatial patterns.

The Paleogene Liushagang Formation is part of the Fushan Sag, a continental lacustrine basin located at the Southeastern margin of the Beibuwan Basin, South China Sea. Further understanding of the deep-water gravity flow deposits in this formation will be conducive to lithologic reservoir exploration in the sag. In this study, three members of the Liushagang Formation, SQEls3 SQEls2 and SQEls1, from old to young, are used with core observation, well log data, and three-dimensional seismic data to identify four deep-lacustrine gravity flow lithofacies including their vertical and lateral relationships within the depositional system. The results are then used to establish a deep-water gravity flow depositional model. Four types of gravity flow lithofacies developed in the sag: sandy debrite, turbidite, sandy slump, and bottom-current deposits. Sand-rich sub-lacustrine fan deposits with typical turbidite channels were developed mainly in the western depression, whereas distal isolated lobes formed by sandy debrite flow deposits occurred mainly in the eastern depression. The results obtained in this study will be helpful in the research of gravity flows in similar continental lacustrine environments.

Abstract Objective The present study attempted to predict blood transfusion risk in spinal tuberculosis surgery by using a novel predictive nomogram. Methods The study was conducted on the clinical data of 495 patients (167 patients in the transfusion group and 328 patients in the non-transfusion group) who underwent spinal tuberculosis surgery in our hospital from June 2012 to June 2021. The least absolute shrinkage and selection operator (LASSO) and multivariable logistic regression analyses were used to screen out statistically significant parameters, which were included to establish a novel predictive nomogram model. The receiver operating characteristic (ROC) curve, calibration curves, C-index, and decision curve analysis (DCA) were used to evaluate the model. Finally, the nomogram was further assessed through internal validation. Results The C-index of the nomogram was 0.787 (95% confidence interval: 74.6%–.82.8%). The C-value calculated by internal validation was 0.763. The area under the curve (AUC) of the predictive nomogram was 0.785, and the DCA was 0.01–0.79. Conclusion A nomogram with high accuracy, clinical validity, and reliability was established to predict blood transfusion risk in spinal tuberculosis surgery. Surgeons must prepare preoperative surgical strategies and ensure adequate availability of blood before surgery.

In order to promote the ecological environment protection and ecological disaster control of Yellow Sea, this paper analyzed the background of ecological environment and problems, and put forward suggestions for the construction of regional monitoring system based on the collection of historical data. The results showed that climate change and nutrient changes were the important factors affecting plankton community. The red tide, green tide and jellyfish bloom were the main ecological disasters of the Yellow Sea Large Marine Ecosystem (YSLME). The regional monitoring system of the YSLME should be established by strengthening various forms of monitoring and cooperation continuously.

IntroductionIn most instances, liver transplantation (LT) is the only available treatment for end‐stage liver diseases. However, LT could also induce serious liver diseases or injury, and the underlying mechanisms of LT-induced complications remain largely unknown, especially the mechanisms of the dysfunction of the immune system mediated by long noncoding RNAs (lncRNAs).MethodsIn this study, we globally analyzed the proportion of immune cells by using the transcriptome sequencing data (RNA-seq) of needle-core liver biopsies from pre- and post-transplantation recipients. Dysregulated lncRNAs were found to be correlated with the altered fractions of immune cells. We finally explored the potential targets of dysregulated lncRNAs and analyzed their functions in LT.ResultsWe found that in the samples, some immune cells changed significantly after LT, including CD4 T cells, NK cells and mast cells. The proportion of macrophages in different polarization states also changed significantly, with M0 macrophages increasing and M2 macrophages decreasing. Through weighted gene co-expression network analysis (WGCNA), 7 gene expression modules related to LT were identified. These modules were related to changes in the proportion of different immune cells. The functions of these modules represent the response modes of different functional genes after LT. Among these modules, MEtan and MEyellow modules were primarily enriched in apoptosis and inflammatory pathways. Twelve immunity-related lncRNAs were identified for the first time, and the regulatory network co-changing with immune cells was also identified. The co-expressed genes of these lncRNAs were highly enriched in apoptosis-related pathways. Many apoptosis-related genes were found to be up-regulated after LT.DiscussionIn summary, we speculated that the expression and regulation of these apoptotic genes may be related to the changes in the proportion of immune cells. Some of these lncRNAs and apoptosis-related genes have been reported to be related to cell proliferation and apoptosis. They are also potential biomarkers or therapeutic targets.

IntroductionEarly-stage accurate diagnosis of malignant pleural mesothelioma (MPM) has always been a formidable challenge. DNA and protein as biomarkers for the diagnosis of MPM have received considerable attention, and yet the outcomes are inconsistent.MethodsIn this study, a systematic search employing PubMed, EMBASE, and Cochrane Library to identify relevant studies from the first day of databases to October 2021. Moreover, we adopt the QUADAS-2 to evaluate the quality of eligible studies and Stata 15.0 and Review Manager 5.4 software programs to perform the meta-analysis. Additionally, bioinformatics analysis was performed at GEPIA for the purpose of exploring relationship between related genes and the survival time of MPM patients.ResultsWe included 15 studies at the DNA level and 31studies at the protein level in this meta-analysis. All results demonstrated that the diagnostic accuracy of the combination of MTAP + Fibulin-3 was the highest with the SEN 0.81 (95% CI: 0.67, 0.89) and the SPE 0.95 (95% CI: 0.90, 0.97). And the bioinformatics analysis indicated that the higher MTAP gene expression level was beneficial to enhance the survival time of MPM patients.DiscussionNonetheless, as a result of the limitations of the included samples, it may be necessary to conduct additional research before drawing conclusions.Systematic review registrationhttps://inplasy.com/inplasy-2022-10-0043/, identifier INPLASY2022100043.

IntroductionCleidocranial dysplasia (CCD) is an autosomal-dominant, heritable skeletal and dental disease, involving hypoplastic clavicles, defective ossification of the anterior fontanelle, dentin and enamel hypoplasia, and supernumerary teeth, which can seriously affect the oral and mental health of patients. Amyloid-like protein aggregation, which is established by lysozyme conjugated with polyethylene glycol (Lyso-PEG), forms a mineralized nanofilm layer on a healthy enamel surface. However, whether it can form a remineralization layer in dental tissues from CCD remains unclear.MethodsThis study evaluated deciduous teeth from healthy individuals and a patient with CCD. Because pulp and dentin are functionally closely related, stem cells from human exfoliated deciduous teeth (SHED) from CCD patients and healthy individuals were collected to compare their biological properties.ResultsThe results found that deciduous teeth from patients with CCD exhibited dentin hypoplasia. In addition, the proliferative ability and osteogenic potential of SHED from patients with CCD were lower than those of control individuals. Finally, Lyso-PEG was applied to dentin from the CCD and control groups, showing a similar remineralization-induced effect on the dentin surfaces of the two groups.ConclusionThese results extend our understanding of the dentin and SHED of patients with CCD, exhibiting good caries-preventive capacity and good biocompatibility of Lyso-PEG, thus providing a novel dental therapy for CCD and patients with tooth hypoplasia.

Background: GGPS1(geranylgeranyl diphosphate synthase 1) is a member of the prenyltransferase family. Abnormal expression of GGPS1 can disrupt the balance between protein farnesylation and geranylgeranylation, thereby affecting a variety of cellular physiologic and pathological processes. However, it is still unknown how this gene could contribute to the prognosis of oral squamous cell carcinoma (OSCC). This study aimed to explore the prognostic role of GGPS1 in OSCC and its relationship with clinical features.Methods: The RNA-seq data and clinical data were obtained from TCGA. The survival analyses, Cox regression analyses, ROC curves, nomograms, calibration curves, and gene function enrichments were established by R software.Results: The results showed that the high expression of GGPS1 in OSCC is related to poor prognosis. At the same time, multivariate Cox regression analyses showed that GGPS1 could be an independent prognostic biomarker, and its gene expression level is closely related to the histological stage of cancer. GGPS1 may promote tumorigenesis because of its metabolic function.Conclusion: This study came to a conclusion that GGPS1, whose high expression has a significantly unfavorable meaning toward the prognosis of OSCC, can act as a novel independent biomarker for OSCC.

RNA-binding proteins (RBP) have emerged as essential regulators that control gene expression and modulate multiple cancer traits. T-cell acute lymphoblastic leukemia (T-ALL) is an aggressive hematologic malignancy derived from transformation of T-cell progenitors that normally undergo discrete steps of differentiation in the thymus. The implications of essential RBP during T-cell neoplastic transformation remain largely unclear. Systematic evaluation of RBP identifies RNA helicase DHX15, which facilitates the disassembly of the spliceosome and release of lariat introns, as a T-ALL dependency factor. Functional analysis using multiple murine T-ALL models demonstrates the essential importance of DHX15 in tumor cell survival and leukemogenesis. Moreover, single-cell transcriptomics reveals that DHX15 depletion in T-cell progenitors hinders burst proliferation during the transition from doublenegative to double-positive cells (CD4-CD8- to CD4+CD8+). Mechanistically, abrogation of DHX15 perturbs RNA splicing and leads to diminished levels of SLC7A6 and SLC38A5 transcripts due to intron retention, thereby suppressing glutamine import and mTORC1 activity. We further propose a DHX15 signature modulator drug ciclopirox and demonstrate that it has prominent anti-T-ALL efficacy. Collectively, our data highlight the functional contribution of DHX15 to leukemogenesis through regulation of established oncogenic pathways. These findings also suggest a promising therapeutic approach, i.e., splicing perturbation by targeting spliceosome disassembly, may achieve considerable anti-tumor efficacy.

IntroductionThe diagnosis and treatment of ankylosing spondylitis (AS) is a difficult task, especially in less developed countries without access to experts. To address this issue, a comprehensive artificial intelligence (AI) tool was created to help diagnose and predict the course of AS.MethodsIn this retrospective study, a dataset of 5389 pelvic radiographs (PXRs) from patients treated at a single medical center between March 2014 and April 2022 was used to create an ensemble deep learning (DL) model for diagnosing AS. The model was then tested on an additional 583 images from three other medical centers, and its performance was evaluated using the area under the receiver operating characteristic curve analysis, accuracy, precision, recall, and F1 scores. Furthermore, clinical prediction models for identifying high-risk patients and triaging patients were developed and validated using clinical data from 356 patients.ResultsThe ensemble DL model demonstrated impressive performance in a multicenter external test set, with precision, recall, and area under the receiver operating characteristic curve values of 0.90, 0.89, and 0.96, respectively. This performance surpassed that of human experts, and the model also significantly improved the experts' diagnostic accuracy. Furthermore, the model's diagnosis results based on smartphone-captured images were comparable to those of human experts. Additionally, a clinical prediction model was established that accurately categorizes patients with AS into high-and low-risk groups with distinct clinical trajectories. BackgroundMultiple components of biodiversity are excellent predictors of precipitation-induced changes in ecosystem function. However, the importance of differing scales (alpha versus beta) is usually overlooked in biodiversity–ecosystem multifunctionality studies. Consequently, little is known about how precipitation regulates the relationship between multifunctionality and multiple components of alpha and beta diversity.AimsWe investigated geographic patterns of herbaceous plant diversity and ecosystem multifunctionality along a precipitation gradient spanning more than 2010 km in Northwest China.MethodsWe assessed the effects of herbaceous species, phylogenetic, and functional components at different scales on multifunctionality in drylands.ResultsThe alpha diversity of species and functional beta diversity were key components explaining the variation in multifunctionality. As the main environmental factor, MAP (mean annual precipitation) affected multifunctionality by changing the mediating variables (i.e., species alpha and functional beta diversity). More importantly, a certain precipitation threshold was detected for the relationship of multifunctionality to species alpha and functional beta diversity. MAPs of approximately 158 mm and 140 mm modulated this relationship (shifting it from uncorrelated to significantly correlated).ConclusionsOur findings provide insights into previously neglected mechanisms by which diversity in herbaceous layers at different scales affects ecosystem multifunctionality. It is highlighted that MAP regulates the relationship between diversity and multifunctionality in dryland ecosystems at different scales. Further, diversity may have substantial consequences for multifunctionality where MAP is higher. These empirical results provide a comprehensive understanding of the biodiversity–multifunctionality relationship in the context of precipitation, ultimately contributing to conservation and restoration programs for multifunctionality in drylands.

Perfluoroalkyl acids (PFAAs) are contaminants of global concern, and the inadvertent consumption of PFAA-contaminated crops may pose a threat to public health. Therefore, systematically studying their source tracing, bioaccumulation prediction and risk assessments in crops is an urgent priority. This study investigated the source apportionment and transport of PFAAs and novel fluorinated alternatives (collectively as per- and polyfluoroalkyl substances, PFASs) from factories to agricultural fields in a fluorochemical industrial region of China. Furthermore, bioaccumulation specificities and prediction of these chemicals in different vegetables were explored, followed by a comprehensive risk assessment from agricultural fields to dinner plates which considered precursor degradation. A positive matrix factorization model revealed that approximately 70 % of PFASs in agricultural soils were derived from fluorochemical manufacturing and metal processing. Alarming levels of ∑PFASs ranged 8.28–84.3 ng/g in soils and 163–7176 ng/g in vegetables. PFAS with short carbon chain or carboxylic acid group as well as branched isomers exhibited higher environmental transport potentials and bioaccumulation factors (BAFs) across a range of vegetables. The BAFs of different isomers of perfluorooctanoic acid (PFOA) decreased as the perfluoromethyl group moved further from the acid functional group. Hexafluoropropylene oxide dimer acid (GenX) showed relatively low BAFs, probably related to its ether bond with a high affinity to soil. Vegetables with fewer Casparian strips (e.g., carrot and radish), or more protein, possessed larger BAFs of PFASs. A bioaccumulation equation integrating critical parameters of PFASs, vegetables and soils, was built and corroborated with a good contamination prediction. After a total oxidizable precursors (TOP) assay, incremental perfluoroalkyl carboxylic acids (PFCAs) were massively found (325–5940 ng/g) in edible vegetable parts. Besides, precursor degradation and volatilization loss of PFASs was firstly confirmed during vegetable cooking. A risk assessment based on the TOP assay was developed to assist the protection of vegetable consumers.

BackgroundConsiderable attention has been given to how different aspects of biodiversity sustain ecosystem functions. Herbs are a critical component of the plant community of dryland ecosystems, but the importance of different life form groups of herbs is often overlooked in experiments on biodiversity-ecosystem multifunctionality. Hence, little is known about how the multiple attributes of diversity of different life form groups of herbs affect changes to the multifunctionality of ecosystems.MethodsWe investigated geographic patterns of herb diversity and ecosystem multifunctionality along a precipitation gradient of 2100 km in Northwest China, and assessed the taxonomic, phylogenetic and functional attributes of different life form groups of herbs on the multifunctionality.ResultsWe found that subordinate (richness effect) species of annual herbs and dominant (mass ratio effect) species of perennial herbs were crucial for driving multifunctionality. Most importantly, the multiple attributes (taxonomic, phylogenetic and functional) of herb diversity enhanced the multifunctionality. The functional diversity of herbs provided greater explanatory power than did taxonomic and phylogenetic diversity. In addition, the multiple attribute diversity of perennial herbs contributed more than annual herbs to multifunctionality.ConclusionsOur findings provide insights into previously neglected mechanisms by which the diversity of different life form groups of herbs affect ecosystem multifunctionality. These results provide a comprehensive understanding of the relationship between biodiversity and multifunctionality, and will ultimately contribute to multifunctional conservation and restoration programs in dryland ecosystems.

IntroductionInfluenza A virus (IAV)-induced dysbiosis may predispose to severe bacterial superinfections. Most studies have focused on the microbiota of single mucosal surfaces; consequently, the relationships between microbiota at different anatomic sites in IAV-infected mice have not been fully studied.MethodsWe characterized respiratory and gut microbiota using full-length 16S rRNA gene sequencing by Nanopore sequencers and compared the nasopharyngeal, oropharyngeal, lung and gut microbiomes in healthy and IAV-infected mice.ResultsThe oropharyngeal, lung and gut microbiota of healthy mice were dominated by Lactobacillus spp., while nasopharyngeal microbiota were comprised primarily of Streptococcus spp. However, the oropharyngeal, nasopharyngeal, lung, and gut microbiota of IAV-infected mice were dominated by Pseudomonas, Escherichia, Streptococcus, and Muribaculum spp., respectively. Lactobacillus murinus was identified as a biomarker and was reduced at all sites in IAV-infected mice. The microbiota composition of lung was more similar to that of the nasopharynx than the oropharynx in healthy mice.DiscussionThese findings suggest that the main source of lung microbiota in mice differs from that of adults. Moreover, the similarity between the nasopharyngeal and lung microbiota was increased in IAV-infected mice. We found that IAV infection reduced the similarity between the gut and oropharyngeal microbiota. L. murinus was identified as a biomarker of IAV infection and may be an important target for intervention in post-influenza bacterial superinfections.

The present work quantifies metal-contaminant interactions between palladium substrates and three salient chlorinated organic contaminants, namely trichloroethylene 1,3,5-trichlorobenzene (TCB), and 3,3′,4,4′-tetrachlorobiphenyl (PCB77). Given that Pd is one of the conventional catalytically active materials known for contaminant removal, maximizing catalytic efficiency through optimal adsorption dynamics reduces the cost of remediation of contaminants that are persistent water pollutants chronically affecting public health. Adsorption efficiency analyses from all-atom molecular dynamics (MD) simulations advance the understanding of reaction mechanisms available from density functional theory (DFT) calculations to an extractable feature scale that can fit the parametric design of supported metal catalytic systems and feed into high throughput catalyst selection. Data on residence time, site-specific adsorption, binding energies, packing geometries, orientation profiles, and the effect of adsorbate size show the anomalous behaviour of organic contaminant adsorption on the undercoordinated {110} surface as compared to the {111} and {100} surfaces. The intermolecular interaction within contaminants from molecular dynamics simulation exhibits refreshing results than ordinary single molecule density functional theory calculation. Since complete adsorption and dechlorination is an essential step for chlorinated organic contaminant remediation pathways, the presented profiles provide essential information for designing efficient remediation systems through facet-controlled palladium nanoparticles.

Objectives: The occurrence of OXA-232-producing carbapenem-resistant Klebsiella pneumoniae (CRKP) has been increasing in China during the last five years. The blaOXA-232-carrying CRKP strain's clonal propagation can readily lead to nosocomial epidemics. Here, we report the genome sequence of an OXA-232 and CTX-M-15 co-producing K. pneumoniae strain isolated from a gallbladder infection in China. Methods: The genome sequence of K. pneumoniae S105 was determined using the Illumina NovaSeq 6000 platform. Antimicrobial resistance genes (ARGs), multilocus sequence typing (MLST) and plasmid replicons were identified using the BacWGSTdb server. The phylogenetic relationship between S105 and other K. pneumoniae strains was analysed using the core genome multilocus sequence typing (cgMLST) strategy. Results: The genomic sequence of K. pneumoniae S105 is made up of 111 contigs with a total length of 5 748 752 bp. According to the Pasteur MLST scheme, S105 belongs to sequence type (ST) 15. Fifteen ARGs were discovered in the genome, including the beta-lactam resistance genes blaOXA-232 and blaCTX-M-15. The blaOXA-232 gene was located in a ColKP3 plasmid. KL112 was anticipated to be the capsule and lipopolysaccharide serotype. A total of 73 phylogenetically related strains were found from 19 nations across four continents; 22 of them were from China, with 21 strains harbouring the blaOXA-232 gene, and the majority of them diverged by just 6–37 cgMLST alleles. Conclusion: In summary, we reported the genomic sequencing of a K. pneumoniae ST15 clinical strain co-carrying the blaOXA-232 and blaCTX-M-15 genes. The clonal dissemination of OXA-232-producing K. pneumoniae ST15 strains in China needs our attention.

Abstract The significance of N6-methyladenosine (m6A) RNA modifications in the progression of breast cancer (BC) has been recognised. However, their potential role and mechanism of action in the tumour microenvironment (TME) and immune response has not been demonstrated. Thus, the role of m6A regulators and their downstream target gene components in BC remain to be explored. In this study, we used a series of bioinformatics methods and experiments to conduct exploratory research on the possible role of m6A regulators in BC. First, two regulatory modes of immune activation and inactivation were determined by tumour classification. The TME, immune cell infiltration, and gene set variation analysis results confirmed the reliability of this pattern. The prognostic model of the m6A regulator was established by the least absolute shrinkage and selection operator and univariate and multivariate Cox analyses, with the two regulators most closely related to survival verified by real-time quantitative reverse transcription polymerase chain reaction. Next, the prognostic m6A regulator identified in the model was crossed with the differential copy number of variant genes in invasive BC (IBC), and it was determined that YTHDF1 was a hub regulator. Subsequently, single-cell analysis revealed the expression patterns of m6A regulators in different IBC cell populations and found that YTHDF1 had significantly higher expression in immune-related IBC cells. Therefore, we selected the intersection of the BC differential expression gene set and the differential expression gene set of a cell line with knocked-down YTHDF1 in literature to identify downstream target genes of YTHDF1, in which we found IFI6, EIR, and SPTBN1. A polymerase chain reaction was conducted to verify the results. Finally, we confirmed the role of YTHDF1 as a potential prognostic biomarker through pan-cancer analysis. Furthermore, our findings revealed that YTHDF1 can serve as a new molecular marker for BC immunotherapy.

Abstract Background Drought stress severely restricts edible fungus production. The genus Auricularia has a rare drought tolerance, a rehydration capability, and is nutrient rich. Results The key genes and metabolic pathways involved in drought-stress and rehydration were investigated using a transcriptome analysis to clarify the relevant molecular mechanisms. In total, 173.93 Mb clean reads, 26.09 Gb of data bulk, and 52,954 unigenes were obtained. Under drought-stress and rehydration conditions, 14,235 and 8539 differentially expressed genes, respectively, were detected. ‘Tyrosine metabolic’, ‘caffeine metabolism’, ‘ribosome’, ‘phagosome’, and ‘proline and arginine metabolism’, as well as ‘peroxisome’ and ‘mitogen-activated protein kinase signaling’ pathways, had major roles in A. fibrillifera responses to drought stress. ‘Tyrosine’ and ‘caffeine metabolism’ might reveal unknown mechanisms for the antioxidation of A. fibrillifera under drought-stress conditions. During the rehydration process, ‘diterpenoid biosynthesis’, ‘butanoate metabolism’, ‘C5-branched dibasic acid’, and ‘aflatoxin biosynthesis’ pathways were significantly enriched. Gibberellins and γ-aminobutyric acid were important in the recovery of A. fibrillifera growth after rehydration. Many genes related to antibiotics, vitamins, and other health-related ingredients were found in A. fibrillifera. Conclusion These findings suggested that the candidate genes and metabolites involved in crucial biological pathways might regulate the drought tolerance or rehydration of Auricularia, shedding light on the corresponding mechanisms and providing new potential targets for the breeding and cultivation of drought-tolerant fungi.

Having a good knowledge of the uncertainty in the land surface temperature (LST) product will help to encourage its use in a wide number of applications, including urban heat islands, geothermal detection, and surface energy balance. Landsat 9 was launched on 27 September 2021 and provides an LST product, which is generated by the radiative transfer equation algorithm and has a spatial resolution of 30 m. In this article, we evaluated the performance of the Landsat 9 LST product by using a temperature-based (T-based) method and cross-validation. The T-based validation results showed that the average bias at the surface radiation budget network and baseline surface radiation network sites was 0.24 K and that the corresponding root mean square error (RMSE) was 3.42 K. The Landsat 9 LST product was in good agreement with the Landsat 7/8 LSTs, with an average bias of 0.25/0.08 K, an RMSE of 0.51/1.04 K, and a mean absolute error of 0.38/0.64 K. The comparable performance of the Landsat 7/8/9 LST products can be explained by the consistent LST retrieval algorithm. The absolute differences in the LST between Landsat 9 LST and MOD11 (MOD21) LST images were between 0.01 (0.65) and 2.50 K (1.76 K), whereas the RMSE values were between 1.40 (1.80) and 3.65 K (3.26 K). The specific heat capacity and thermal inertia of the different land surface covers can explain the significant biases. The above evaluation results are consistent with the initial performance testing of thermal infrared sensor-2 (TIRS-2) by the National Aeronautics and Space Administration and the U.S. Geological Survey. Although the released Landsat 9 LST product showed good performance in the preliminary evaluation, the split-window algorithm may be a better option for Landsat 9 LST retrieval, as the TIRS-2 data addressed stray light incursion. Since there are no official validation results that have been published, this article provides a third-party performance evaluation of the Landsat 9 LST product and will benefit research fields that require Landsat series LST products.

A new type of explosive ink formulation that can be quickly cured was prepared with unsaturated polyester as binder, styrene as active monomer, 2,4,6-trimethylbenzoyl-diphenylphosphine oxide as photoinitiator, and hexanitrohexaazaisowurtzitane (CL-20) as the main explosive. Then the explosive ink direct writing technology was used to charge the micro-sized energetic devices, the curing mechanism of the explosive ink was discussed, and the microstructure, safety performance and explosive transfer performance of the explosive ink molded samples were tested and analyzed. Results indicate that the composite material has a fast curing molding speed, its hardness can reach 2H within 8 min. The crystal form of CL-20 in the molded sample is still ε type. The CL-20 based UV-curing explosive ink formulation has good compatibility, its apparent activation energy is increased by about 3.5 kJ/mol. The composite presents a significant reduction in impact sensitivity and its characteristic drop height can reach 39.8 cm, which is about 3 times higher than the raw material. When the line width of charge is 1.0 mm, the critical thickness of the explosion can reach 0.015 mm, and the explosion velocity is 7129 m/s when the charge density is 1.612 g/cm3.

Abstract In this work, lipid profile migration from muscle to juice during the tilapia muscle steaming process was revealed by a transactional analysis of data from ultra-high-performance liquid chromatography coupled with Q Exactive (UHPLC-QE) Orbitrap mass spectrometry (MS) and lipidomics. Firstly, the lipids in tilapia muscles and juices at different steaming time points were extracted and examined by UHPLC-QE Orbitrap mass spectrometry. Secondly, a transactional analysis procedure was developed to analyze the data from UHPLC-QE Orbitrap MS and lipidomics. Finally, the corrected lipidomics data and the normalized MS data were used for lipid migration analysis. The results suggested that the transactional analysis procedure was efficient to significantly decrease UHPLC-QE Orbitrap MS workloads and delete the false-positive data (22.4–36.7%) in lipidomics data, which compensated the disadvantages of the current lipidomics method. The lipid changes could be disappearance, full migration into juice, appearance in juice, appearance in muscle, appearance in both muscle and juice, and retention in the muscle. Moreover, the results showed 9 (compared with 52), 5 (compared with 116), and 10 (compared with 178) of lipid class (compared with individual lipid) variables showed significant differences among the different steaming times (0, 10, 30, and 60 min) in all the muscles, juices, and muscle-juice systems, respectively. These results showed significant lipid profile migration from muscle to juice during the tilapia steaming process.

Abstract Background Coconut is an important tropical oil and fruit crop whose evolutionary position renders it a fantastic species for the investigation of the evolution of monocot chromosomes and the subsequent differentiation of ancient plants. Results Here, we report the assembly and annotation of reference-grade genomes of Cn. tall and Cn. dwarf, whose genome sizes are 2.40 Gb and 2.39 Gb, respectively. The comparative analysis reveals that the two coconut subspecies diverge about 2–8 Mya while the conserved Arecaceae-specific whole-genome duplication (ω WGD) occurs approximately 47–53 Mya. It additionally allows us to reconstruct the ancestral karyotypes of the ten ancient monocot chromosomes and the evolutionary trajectories of the 16 modern coconut chromosomes. Fiber synthesis genes in Cn. tall, related to lignin and cellulose synthesis, are found at a higher copy number and expression level than dwarf coconuts. Integrated multi-omics analysis reveals that the difference in coconut plant height is the result of altered gibberellin metabolism, with both the GA20ox copy number and a single-nucleotide change in the promoter together leading to the difference in plant height between Cn. tall and Cn. dwarf. Conclusion We provide high-quality coconut genomes and reveal the genetic basis of trait differences between two coconuts through multi-omics analysis. We also reveal that the selection of plant height has been targeted for the same gene for millions of years, not only in natural selection of ancient plant as illustrated in coconut, but also for artificial selection in cultivated crops such as rice and maize.

Abstract Polyphagous aphids often consist of host‐specialized lineages, which have greater fitness on their native hosts than on others. The underlying causes are important for understanding of the evolution of diet breadth and host shift of aphids. The cotton‐melon aphid Aphis gossypii Glover is extremely polyphagous with many strict host‐specialized lineages. Whether and how the lineage specialized on the primary host hibiscus shifts to the secondary host cucumber remains elusive. We found that the hibiscus‐specialized lineage suffered high mortality and gave birth to very few nymphs developing into yellow dwarfs on fresh cucumber leaves, and did not inflict any damage symptoms on cucumber plants. The poor performance did not improve with prolonged exposure to cucumber; however, it did significantly improve when the cucumber leaves were pre‐infected with a biotrophic phytopathogen Pseudoperonospora cubensis. More importantly, the hibiscus‐specialized lineage with two‐generation feeding experience on pre‐infected cucumber leaves performed as well as the cucumber‐specialized lineage did on fresh cucumber leaves, and inflicted typical damage symptoms on intact cucumber plants. Electrical penetration graph (EPG) indicated that the hibiscus‐specialized lineage did not ingest phloem sap from fresh cucumber leaves but succeeded in ingesting phloem sap from pre‐infected cucumber leaves, which explained the performance improvement of the hibiscus‐specialized lineage on pre‐infected cucumber leaves. This study revealed a new pathway for the hibiscus‐specialized lineage to quickly acclimate to cucumber under the assistance of the phytopathogen. We considered that the short feeding experience on pre‐infected cucumber may activate expression of effector genes that are related to specific host utilization. We suggest to identify host‐specific effectors by comparing proteomes or/and transcriptomes of the hibiscus‐specialized lineage before and after acclimating to cucumber.

Neurotrophin-4 (NT-4) is a neurotrophic factor that plays important roles in maintaining nerve cell survival, regulating neuronal differentiation and apoptosis, and promoting nerve injury repair. However, the source of sufficient NT-4 protein and efficient delivery of NT-4 remain a challenge. This study aims to express an activated human NT-4 protein in a large scale by genetically engineering silk gland bioreactor of silkworm as a host. We showed that the expression of human NT-4-functionalized silk material could promote proliferation of mouse HT22 cells when compared to the natural silk protein, and no obvious cytotoxicity was observed under the conditions of different silk materials. Importantly, this functional silk material was able to induce the potential differentiation of HT22 cells, promote peripheral neural cell migration and neurite outgrowth of chicken embryo dorsal root ganglion (DRG). All these results demonstrated a high bioactivity of human NT-4 protein produced in silk gland. Therefore, based on the silkworm model, the further fabrication of different silk materials-carrying active NT-4 protein with good mechanical properties and great biocompatibility will give promising applications in tissue engineering and neurons regeneration.

BackgroundTuberculosis (TB) is a chronic infectious disease. Bone and joint TB is a common type of extrapulmonary TB and often occurs secondary to TB infection. In this study, we aimed to find the difference in the blood examination results of patients with bone and joint TB and patients with TB by using machine learning (ML) and establish a diagnostic model to help clinicians better diagnose the disease and allow patients to receive timely treatment.MethodsA total of 1,667 patients were finally enrolled in the study. Patients were randomly assigned to the training and validation cohorts. The training cohort included 1,268 patients: 158 patients with bone and joint TB and 1,110 patients with TB. The validation cohort included 399 patients: 48 patients with bone and joint TB and 351 patients with TB. We used three ML methods, namely logistic regression, LASSO regression, and random forest, to screen the differential variables, obtained the most representative variables by intersection to construct the prediction model, and verified the performance of the proposed prediction model in the validation group.ResultsThe results revealed a great difference in the blood examination results of patients with bone and joint TB and those with TB. Infectious markers such as hs-CRP, ESR, WBC, and NEUT were increased in patients with bone and joint TB. Patients with bone and joint TB were found to have higher liver function burden and poorer nutritional status. The factors screened using ML were PDW, LYM, AST/ALT, BUN, and Na, and the nomogram diagnostic model was constructed using these five factors. In the training cohort, the area under the curve (AUC) value of the model was 0.71182, and the C value was 0.712. In the validation cohort, the AUC value of the model was 0.6435779, and the C value was 0.644.ConclusionWe used ML methods to screen out the blood-specific factors—PDW, LYM, AST/ALT, BUN, and Na+—of bone and joint TB and constructed a diagnostic model to help clinicians better diagnose the disease in the future.

The intake safety of nuclear power cooling-water systems (NPCSs) is an important aspect of operational safety of nuclear power plants (NPPs). The blockages caused by aberrant outbreaks of various aquatic organisms have seriously affected operational safety. Large jellyfish constitute the main groups of marine organisms responsible for these blockages. The processes of aggregation and the relationships of two major disaster-causing scyphozoan jellyfish species, Nemopilema nomurai and Aurelia coerulea, with four environmental factors at the intake area of an NPCS in Eastern Liaodong Bay, China, were investigated in 2019 and 2020. The findings revealed that A. coerulea ephyrae were present in the surrounding ports in mid-May; however, N. nomurai ephyrae were absent during the survey period in this study, and the medusae of N. nomurai started appearing from late May. The individual growth and relative biomass (RB) of the jellyfish increased rapidly from late June to July and decreased rapidly thereafter, in September. The RB of N. nomurai was highly correlated to the sea surface temperature (SST) and levels of dissolved oxygen (DO) in the region. The RB increased with increasing SST and decreased at increasing DO levels. The RB of A. coerulea was significantly negatively correlated with that of N. nomurai, and the peak biomass of the two species alternated over time, which could be attributed to the fact that the jellyfish species share similar ecological niches. The bell diameters were significantly positively correlated with the individual wet weights, and the value of one could be inferred from the value of the other. Although the processes of jellyfish aggregation are attributed to several factors, including interactions with environmental factors and human activities, such as fishing, the results obtained in this study would serve as an important reference and provide a basis for the prevention of jellyfish blooms in waters adjacent to NPPs. The prevention and control of jellyfish disasters at the intake area of NPCSs are not only local concerns. Therefore, remediation from the source combined with the maximum utilization of social resources for monitoring and early warning would immensely improve the efficacy of such preventive strategies.

IntroductionNanopore sequencing has been widely used in clinical metagenomic sequencing for pathogen detection with high portability and real-time sequencing. Oxford Nanopore Technologies has recently launched an adaptive sequencing function, which can enrich on-target reads through real-time alignment and eject uninteresting reads by reversing the voltage across the nanopore. Here we evaluated the utility of adaptive sequencing in clinical pathogen detection.MethodsNanopore adaptive sequencing and standard sequencing was performed on a same flow cell with a bronchoalveolar lavage fluid sample from a patient with Chlamydia psittacosis infection, and was compared with the previous mNGS results.ResultsNanopore adaptive sequencing identified 648 on-target stop receiving reads with the longest median read length(688bp), which account for 72.4% of all Chlamydia psittaci reads and 0.03% of total reads in enriched group. Brain organoids have been considered as an advanced platform for in vitro disease modeling and drug screening, but numerous roadblocks exist, such as lack of large-scale production technology and lengthy protocols with multiple manipulation steps, impeding the industrial translation of brain organoid technology. Here, we describe the high-speed and large-scale production of midbrain organoids using a high-throughput screening-compatible platform within 30 days. Micro midbrain organoids (µMOs) exhibit a highly uniform morphology and gene expression pattern with minimal variability. Notably, µMOs show dramatically accelerated maturation, resulting in the generation of functional µMOs within only 30 days of differentiation. Furthermore, individual µMOs display highly consistent responsiveness to neurotoxin, suggesting their usefulness as an in vitro high-throughput drug toxicity screening platform. Collectively, our data indicate that µMO technology could represent an advanced and robust platform for in vitro disease modeling and drug screening for human neuronal diseases.

In order to effectively improve the impact resistance performance of the roadway anti-impact hydraulic support and reduce the loss caused by rock burst to a certain extent, a torsion plate energy-absorbing member was designed for using with the hydraulic column. By using theoretical analysis and numerical simulation methods and combining them with the parameters of the energy-absorbing characteristics evaluation index, single-factor tests were designed. Under the premise of constant impact velocity, the influence law of torsion angle, plate thickness, and fillet radius of the pressure plate on the energy-absorbing and anti-impact characteristics of energy-absorbing members were analysed. The results show that the deformation process and resistance trend of the torsion plate correspond to energy absorption. The resistance stability is high in the reaction force platform stage, and the energy increases linearly. With the increase of the torsion angle, the mean value of the reaction force increases, the fluctuation of the reaction force platform stage becomes more obvious, the specific energy absorption increases linearly, and the standard deviation reaches the minimum at 480°. Both the tensile length and the length of the reaction force platform stage increase continuously. With the increase of the plate thickness, the flatness of the torsion plate shows a trend of bad-good-bad, the mean value of the reaction force gradually increases, the specific energy absorption grows faster and then slower, and the tensile length and the length of the reaction force platform stage increase accordingly. With the increase of the fillet radius, the flatness of the torsion plate is better and the reaction force has a small improvement. The fluctuation of reaction force is most significant when the fillet radius is 2 mm, and the energy-absorbing member has the largest swing. The specific energy absorption is highest when the fillet radius is 2.5 mm, the standard deviation shows a tendency to develop firstly decreasing and then stabilising, the tensile length of the torsion plate decreases slightly, and the length of the reaction force platform firstly increases and then tends to stabilise. The torsion plate energy-absorbing member is a more ideal energy-absorbing and anti-impact member, which plays a positive role in the energy-absorbing and anti-impact of the hydraulic support.

Commonly used clinical and surgical methods include autologous/allogeneic bone and cartilage transplantation, vascularized bone transplantation, autologous chondrocyte implantation, mosaicplasty, and joint replacement. 3D bio printing technology to construct implants by layer-by-layer printing of biological materials, living cells, and other biologically active substances in vitro, which is expected to replace the repair mentioned above methods. Researchers use cells and biomedical materials as discrete materials. 3D bio printing has largely solved the problem of insufficient organ donors with the ability to prepare different organs and tissue structures. This paper mainly discusses the application of polymer materials, bio printing cell selection, and its application in bone and cartilage repair.

ObjectiveThe aim of this study was to investigate the efficacy and complications of stent-assisted coiling in comparison with flow diversion for wide-necked intracranial aneurysms.MethodsPatients with wide-necked intracranial aneurysms who were treated with stent-assisted coiling or flow diversion were respectively, enrolled into the stent-assisted coiling or flow diversion treatment group. The clinical and angiographic data were analyzed.ResultsA total of 61 patients with intracranial aneurysms underwent stent-assisted coiling, including 35 (57.4%) female and 26 (42.6%) male patients with 21 (34.4%) ruptured and 40 (65.6%) unruptured aneurysms. Also, 53 patients underwent deployment of flow-diverting devices, including 30 (56.6%) female and 23 (43.4%) male patients with 25 (47.2%) ruptured and 28 (52.8%) unruptured aneurysms. Stent-assisted coiling was performed successfully in 60 patients with 63 stents deployed, and immediate aneurysm occlusion was complete occlusion in 38 (62.3%) aneurysms, residual neck in 12 (19.7%), and residual aneurysm in 10 (16.4%). Procedure-related complications included in-stent thrombosis in three (4.9%) patients, coil protrusion in three (4.9%), and re-rupture of one (1.6%) aneurysm, with a total complication rate of 11.5%. In the flow diversion group, a pipeline embolization device alone was deployed in each of the 24 (45.3%) patients, adjunctive coiling combined with a pipeline device in 29 (54.7%), and double pipeline devices in each of the 6 (11.3%) patients. Immediately after treatment, complete occlusion was achieved in 3 (5.7%) patients with adjunctive coiling, residual neck in 3 (5.7%), and residual aneurysm in 47 (88.7%). Procedure-related complications included aneurysm rebleeding in one patient (1.9%). Clinical and angiographic follow-up was performed 13–49 months (median 29) after the procedure for 49 (80.3%) patients with stent-assisted coiling, with complete aneurysm occlusion in 27 (55.1%) aneurysms, residual neck in 3 (6.1%), residual aneurysm in 5 (10.2%), and recurrence in 14 (28.6%). Follow-up was performed for 14–37 (median 25) months in 45 (84.9%) patients with flow diversion treatment, with complete occlusion in 39 (86.7%) patients, residual neck in 5 (11.1%), residual aneurysm in 1 (2.2%), and no aneurysm recurrence.ConclusionsStent-assisted coiling comes with more complications but fewer permanent aneurysm occlusions than flow diverters, and flow diverters are superior to stent-assisted coiling in the treatment of wide-necked intracranial aneurysms, especially in the long-term effect.

Using supercritical CO2 as a heat transfer fluid in microchannel receivers is a promising alternative for tower concentrating solar power plants. In this paper, the heat transfer and flow characteristics of supercritical CO2 in microchannels at high temperature are investigated by numerical simulations. The effects of microchannel structure, mass flow rate, heat flux, pressure, inlet temperature and radiation are analyzed and discussed. The results show that higher mass flow rate obtains poorer heat transfer performance with larger flow resistance of supercritical CO2 in microchannels at high temperature. The fluid and wall temperatures, average heat transfer coefficient and pressure drop all increase nearly linearly with the increases in heat flux and inlet temperature in the high-temperature region. Moreover, high pressure contributes to great hydraulic performance with approximate thermal performance. The effect of radiation on thermal performance is more pronounced than that on hydraulic performance. Furthermore, the optimized structures of inlet and outlet headers, as well as those of the multichannel in the microchannels, are proposed to obtain good temperature uniformity in the microchannels with relatively low pressure drop. The results given in the current study can be conducive to the design and application of microchannel receivers with supercritical CO2 as a heat transfer fluid in the third generation of concentrating solar power plants.

Multispectral and hyperspectral image fusion (MHF) aims to reconstruct high-resolution hyperspectral images by fusing spatial and spectral information. Unlike the traditional canonical polyadic decomposition and Tucker decomposition models, the block-term tensor decomposition model is able to improve the quality of fused images using known endmember and abundance information. This paper presents an improved hyperspectral image fusion algorithm. Firstly, the two abundance matrices are combined into a single bulk matrix to promote structural sparsity by introducing the L2,1-norm to eliminate the scaling effects present in the model. Secondly, the counter-scaling effect is eliminated by adding the L2-norm to the endmember matrix. Finally, the chunk matrix and the endmember matrix are coupled together, and the matrix is reorganized by adding the L2,1-norm to the matrix to facilitate chunk elimination and solved using an extended iterative reweighted least squares (IRLS) method, focusing on the problem of the inability to accurately estimate the tensor rank in the chunk-term tensor decomposition model and the noise/artifact problem arising from overestimation of rank. Experiments are conducted on standard and local datasets, and the fusion results are compared and analyzed in four ways: visual result analysis, metric evaluation, time of the algorithm, and classification results, and the experimental results show that the performance of the proposed method is better than the existing methods. An extensive performance evaluation of the algorithms is performed by conducting experiments on different datasets. The experimental results show that the proposed algorithm achieves significant improvements in terms of reconstruction error, signal-to-noise ratio, and image quality compared with the existing methods. Especially in the case of a low signal-to-noise ratio, the proposed algorithm shows stronger robustness and accuracy. These results show that the proposed algorithm has significant advantages in dealing with multispectral high-resolution hyperspectral data.

Nitrogen-vacancy (NV) centers in diamonds are promising solid-state magnetic sensors with potential applications in power systems, geomagnetic navigation, and diamond NV color center current transformers, in which both high bandwidth and high magnetic field resolution are required. The wide bandwidth requirement often necessitates high laser power, but this induces significant laser fluctuation noise that affects the detection magnetic field resolution severely. Therefore, enhancement of the magnetic field resolution of wide-bandwidth NV center magnetic sensors is highly important because of the reciprocal effects of the bandwidth and magnetic field resolution. In this article, we develop a common mode rejection (CMR) model to eliminate the laser noise effectively. The simulation results show that the noise level of the light-detected magnetic resonance signal is significantly reduced by a factor of 6.2 after applying the CMR technique. After optimization of the laser power and modulation frequency parameters, the optimal system bandwidth was found to be 75 Hz. Simultaneously, the system’s detection magnetic field resolution was enhanced significantly, increasing from 4.49 nT/Hz1/2 to 790.8 pT/Hz1/2, which represents an improvement of nearly 5.7 times. This wide-bandwidth, high-magnetic field resolution NV color center magnetic sensor will have applications including power systems, geomagnetic navigation, and diamond NV color center current transformers.

Excessive fertilizer application, majorly nitrogen- and phosphorus-based fertilizers, in farmland has intensified environmental pollution of rivers, lakes, and other surface water bodies worldwide by agricultural non-point sources, especially the highly-mobile nitrogen. To solve nitrogen pollution in sugarcane areas, exploring the nitrogen output characteristics of agricultural watersheds in crop fields becomes necessary. Therefore, the objective of the study was to evaluate the characteristics of nitrogen output during typical rainfall events in different sugarcane growth stages in a southern tropical watershed in China. Dynamic monitoring of runoff and nitrogen concentration was carried out for four rainfall events and compared among four sugarcane growth stages (Establishment; Vegetative growth; Grand growth; Ripening) during the growing season of 2018 in the Nala watershed, Kelan Reservoir, Guangxi, China. The results showed that the total dissolved nitrogen flux of the 4 rainfall events ranged from 0.08 to 9.88 kg·hm−2 for the different growth stages. Nitrate nitrogen was the main component of the total flux, accounting between 75.7 and 92.1% of the total dissolved nitrogen while ammonium nitrogen accounted between 1.80 and 5.26% of the total flux for the 4 rainfall events. Total dissolved nitrogen and nitrate-nitrogen were significantly and negatively correlated with runoff (p < 0.05), while total dissolved nitrogen concentration did not correlate with runoff. The incipient scouring effect of total dissolved nitrogen and nitrate-nitrogen was not noticeable. The concentration of total dissolved nitrogen in the Nala watershed was inferior to class V water quality standard, indicating water eutrophication danger. The study showed that nitrogen nutrient inflow into the river was promoted by N-fertilization time and rainfall. Therefore, reasonably reducing N-fertilization dose and post-rain fertilization could effectively reduce nitrogen inflow into rivers and avoid the intensification of eutrophication in sugarcane areas. We recommend multiple years of studies to verify the possible impacts of differences in weather conditions.

This paper presents a novel design for uplift piles incorporating a composite-anchor system. The composite-anchor system consists of steel strands, a non-expansion grouting body, and a high-strength steel pile. The aim of this design is to enhance the mechanical performance, durability, and economic efficiency of uplift piles. To evaluate the performance of the new pile, three sets of full-scale load tests were conducted, focusing on their in situ capacity, deformation, and stress characteristics. Despite a significantly lower reinforcement ratio of 0.75% compared to conventional piles with a ratio of 3.84%, the new uplift piles exhibit an exceptional uplift bearing performance. The utilization of the lateral friction resistance of the lower pile body is significantly improved, leading to enhanced load distribution and stress transfer mechanisms. Furthermore, a numerical model was developed and validated against the experimental results, demonstrating its reliability in simulating the bearing characteristics of the new uplift piles. The multi-interface design of the composite-anchor system ensures the efficient transmission of internal forces induced by external uplift loads, resulting in an improved stress state within the pile body. Moreover, the multi-layer structure of the composite main bar enhances the durability of the uplift piles. In comparison to conventional piles, the new uplift pile design offers substantial advantages, including an 80% reduction in reinforcement ratio, a 65% reduction in reinforcement cage welding, a cost reduction of approximately 30%, and a shortened construction time by around 20%. These findings highlight the potential of the new composite-anchor-pile design to revolutionize the field of uplift pile applications, offering improved efficiency and effectiveness.

Wherever there is group life, there has been a social division of labor and resource allocation, since ancient times. Examples include ant colonies, bee colonies, and wolf colonies. Different roles are responsible for different tasks. The same is true of human beings. Human beings are the largest social group in nature, among whom there are intricate social networks and interest networks between individuals. In such a complex relationship, how do decision makers allocate resources or tasks to individuals in a fair way? This is a topic worthy of further study. In recent decades, fair allocation has been at the core of research in economics, mathematics and other fields. The fair allocation problem is to assign a set of items to a set of agents so that each agent’s allocation is as fair as possible to satisfy each agent. The fairness measurements followed in current research include envy-freeness, proportionality, equitability, maximin share fairness, competitive equilibrium, maximum Nash social diswelfare, and so on. In this paper, the main concern is the allocation of chores. We discuss this problem in two parts: divisible and indivisible. We comprehensively review the existing results, algorithms, and approximations that meet various fairness criteria in chronological order. The relevant results of achieving fairness and efficiency are also discussed. In addition, we propose some open questions and future research directions for this problem based on existing research.
