157. Ultrasound microbubble-mediated delivery of the siRNAs targeting MDR1 reduces drug resistance of yolk sac carcinoma L2 cells

Abstract

Abstract Background MDR1 gene encoding P-glycoprotein is an ATP-dependent drug efflux transporter and related to drug resistance of yolk sac carcinoma. Ultrasound microbubble-mediated delivery has been used as a novel and effective gene delivery method. We hypothesize that small interfering RNA (siRNA) targeting MDR1 gene (siMDR1) delivery with microbubble and ultrasound can down-regulate MDR1 expression and improve responsiveness to chemotherapeutic drugs for yolk sac carcinoma in vitro. Methods Retroviral knockdown vector pSEB-siMDR1s containing specific siRNA sites targeting rat MDR1 coding region were constructed and sequence verified. The resultant pSEB-siMDR1 plasmids DNA were encapsulated with lipid microbubble and the DNA release were triggered by ultrasound when added to culture cells. GFP positive cells were counted by flow cytometry to determine transfection efficiency. Quantitative real-time PCR and western blot were performed to determine the mRNA and protein expression of MDR1. P-glycoprotein function and drug sensitivity were analyzed by Daunorubicin accumulation and MTT assays. Results Transfection efficiency of pSEB-siMDR1 DNA was significantly increased by ultrasound microbubble-mediated delivery in rat yolk sac carcinoma L2 (L2-RYC) cells. Ultrasound microbubble-mediated siMDR1s delivery effectively inhibited MDR1 expression at both mRNA and protein levels and decreased P-glycoprotein function. Silencing MDR1 led to decreased cell viability and IC50 of Vincristine and Dactinomycin. Conclusions Our results demonstrated that ultrasound microbubble-mediated delivery of MDR1 siRNA was safe and effective in L2-RYC cells. MDR1 silencing led to decreased P-glycoprotein activity and drug resistance of L2-RYC cells, which may be explored as a novel approach of combined gene and chemotherapy for yolk sac carcinoma.

Abstract

We discuss the dynamic traffic network equilibrium system problem. We introduce the equilibrium definition based on Wardrop's principles when there are some internal relationships between different kinds of goods which transported through the same traffic network. Moreover, we also prove that the equilibrium conditions of this problem can be equivalently expressed as a system of evolutionary variational inequalities. By using the fixed point theory and projected dynamic system theory, we get the existence and uniqueness of the solution for this equilibrium problem. Finally, a numerical example is given to illustrate our results.

Abstract Background Tumor-specific cytotoxic T cells and infiltrating lymphocytes are frequently found in tumor tissues in patients with nasopharyngeal carcinoma (NPC). Most patients with NPC, however, especially those with advanced stages, have a poor clinical prognosis despite conventional immunotherapy. The aim of this work was to examine the effect of indoleamine 2,3-dioxygenase (IDO), an immunosuppressive enzyme, on the lymphocyte function in NPC. Methods The NPC cell line CNE2 was treated by interferon-γ (IFNγ) and the levels of IDO expression was analyzed by Western blotting and reverse phase high-performance liquid chromatography (HPLC). Lymphocytes from health human exposed to the milieu created by IDO-positive CNE2 cells and the lymphocyte cytotoxicity to target tumor cells was analyzed by standard lactate dehydrogenase (LDH) release assay. Additionally, expression of IDO was determined by Immunohistochemical assay in the tumor tissues form clinically evaluated NPC. Results IDO expression was acutely induced in the NPC cell line CNE2 by low dose interferon-γ (IFNγ) or by co-incubation with activated lymphocytes. Exposure to the milieu created by IDO-positive CNE2 cells did not promote lymphocyte death, but lymphocyte cytotoxicity against target tumor cells was impaired. The suppression of lymphocyte cytotoxic function was fully restored when the conditioned medium was replaced by fresh medium for 24 h. In additionally, the IDO-positive cells were found scattered in the tumor tissues from patients with NPC. Conclusion Altogether, these findings suggest that IDO-mediated immunosuppression may be involved in the tumor immune evasion, and that blocking IDO activity in tumor cells may help to re-establish an effective anti-tumor T cell response in NPC.

Abstract

Abstract Background Protein sumoylation is an essential dynamic, reversible post translational modification that plays a role in dozens of cellular activities, especially the regulation of gene expression and the maintenance of genomic stability. Currently, the complexities of sumoylation mechanism can not be perfectly solved by experimental approaches. In this regard, computational approaches might represent a promising method to direct experimental identification of sumoylation sites and shed light on the understanding of the reaction mechanism. Results Here we presented a statistical method for sumoylation site prediction. A 5-fold cross validation test over the experimentally identified sumoylation sites yielded excellent prediction performance with correlation coefficient, specificity, sensitivity and accuracy equal to 0.6364, 97.67%, 73.96% and 96.71% respectively. Additionally, the predictor performance is maintained when high level homologs are removed. Conclusion By using a statistical method, we have developed a new SUMO site prediction method – SUMOpre, which has shown its great accuracy with correlation coefficient, specificity, sensitivity and accuracy.

Abstract

User-generated item lists are popular on many platforms. Examples include video-based playlists on YouTube, image-based lists (or"boards") on Pinterest, book-based lists on Goodreads, and answer-based lists on question-answer forums like Zhihu. As users create these lists, a common challenge is in identifying what items to curate next. Some lists are organized around particular genres or topics, while others are seemingly incoherent, reflecting individual preferences for what items belong together. Furthermore, this heterogeneity in item consistency may vary from platform to platform, and from sub-community to sub-community. Hence, this paper proposes a generalizable approach for user-generated item list continuation. Complementary to methods that exploit specific content patterns (e.g., as in song-based playlists that rely on audio features), the proposed approach models the consistency of item lists based on human curation patterns, and so can be deployed across a wide range of varying item types (e.g., videos, images, books). A key contribution is in intelligently combining two preference models via a novel consistency-aware gating network - a general user preference model that captures a user's overall interests, and a current preference priority model that captures a user's current (as of the most recent item) interests. In this way, the proposed consistency-aware recommender can dynamically adapt as user preferences evolve. Evaluation over four datasets(of songs, books, and answers) confirms these observations and demonstrates the effectiveness of the proposed model versus state-of-the-art alternatives. 183. A Hierarchical Self-Attentive Model for Recommending User-Generated Item Lists

Abstract

User-generated item lists are a popular feature of many different platforms. Examples include lists of books on Goodreads, playlists on Spotify and YouTube, collections of images on Pinterest, and lists of answers on question-answer sites like Zhihu. Recommending item lists is critical for increasing user engagement and connecting users to new items, but many approaches are designed for the item-based recommendation, without careful consideration of the complex relationships between items and lists. Hence, in this paper, we propose a novel user-generated list recommendation model called AttList. Two unique features of AttList are careful modeling of (i) hierarchical user preference, which aggregates items to characterize the list that they belong to, and then aggregates these lists to estimate the user preference, naturally fitting into the hierarchical structure of item lists; and (ii) item and list consistency, through a novel self-attentive aggregation layer designed for capturing the consistency of neighboring items and lists to better model user preference. Through experiments over three real-world datasets reflecting different kinds of user-generated item lists, we find that AttList results in significant improvements in NDCG, Precision@k, and Recall@k versus a suite of state-of-the-art baselines. Abstract

How evolution favors cooperation is a fundamental issue in social and economic systems. In the business world, actively selecting a suitable project is usually helpful for a businessman to be in an advantageous position. By incorporating project selection mechanism into the threshold public goods game, we have investigated the coupling effect of mutation and imitation in updating one's preferred project scale on the evolution of cooperation. Compared with the situation where there is no project selection mechanism, the existence of project selection may suppress or promote cooperation depending upon different updating rules. There exists a critical ratio of the imitators in the population, below which cooperation is suppressed while above which cooperation is promoted. With the coevolving of individual strategies and preferred project scales, a higher level of cooperation corresponds to a larger average value of the preferred project scales. A theoretical analysis indicates that, as most of the individuals are mutants, the coevolving process is governed by the mutation process, which leads to a smaller average value of the preferred project scales and a lower level of cooperation. As most of the individuals are imitators, the coevolving process is governed by the coupling of the mutation and imitation processes, which leads to a larger average value of the preferred project scales and a higher level of cooperation. As all the individuals are imitators, the coevolving process is governed by the imitation process, which leads to an intermediate average value of the preferred project scales and an intermediate level of cooperation.

185. Voltage Control of Perpendicular Exchange Bias in Multiferroic Heterostructures

Abstract

Perpendicular exchange bias (EB), which combines the perpendicular magnetic anisotropy and the ferromagnetic (FM) - antiferromagnetic (AFM) exchange coupling, is extremely important in the high-density AFM spintronics. However, the effective modulation of EB remains challenging, since the alternant spins at the AFM/FM interface are strongly pinned by the AFM layer. Voltage tuning of EB through the magnetoelectric coupling provides a potential way to achieve a rapid magnetization switching in an energy-efficient manner. Nevertheless, the interfacial strain mediation of perpendicular EB induced by E-field remains unexplored. In this work, we obtain perpendicular EB nanostructure by room-temperature fabrication process, and demonstrate the voltage tunable perpendicular EB in Pt/IrMn/(Co/Pt)2/Ta/(011) Pb(Mg1/3Nb2/3)O3-PbTiO3 multiferroic heterostructure. To enhance the voltage control effect on perpendicular EB, we further investigate both strain-mediated magnetoelectric coupling and ionic liquid gating method in the thinned EB system with the structure of Pt/IrMn/Co/Pt/Ta. As a result, the voltage induced lattice distortion effectively transmits to the AFM/FM interface, while the charge accumulation in gating method generates a relatively large hysteresis loop offset that has not been observed before at room temperature.
