Abstract

Chitins and chitosans are some of the most abundant natural polysaccharide materials, and are used to increase innate immune response and disease resistance in humans and animals. In this work, chitin and chitosan from housefly, Musca domestica, pupa shells were obtained by treatment with HCl and NaOH. For chitin extraction, 2 N HCl and 1.25 N NaOH solutions were used to achieve decalcification and deproteinization, respectively. For chitosan extraction, 50% NaOH solution was used to achieve deacetylation. The yields of chitin and chitosan from pupa shells of M. domestica were 8.02% and 5.87%, respectively. The deacetylations of chitosan (from chitin C1 and C2) were 89.76% and 92.39%, respectively, after the first alkali treatment with 50% NaOH (w/w) solution at 105 °C for 3 h and 5 h, respectively. The viscosities of the chitosans (from chitin C1 and C2) were 33.6 and 19.2 cP, respectively. [ABSTRACT FROM AUTHOR]

Abstract

Target of rapamycin ( TOR) is an evolutionarily conserved serine-threonine kinase that affects various cellular functions including growth, development, ageing, immunity and autophagy. Inhibition in TOR activity stimulates autophagy, a self-eating process that ensures survival by maintaining metabolic homeostasis and energy retention at both cellular and organismal levels. To understand the interplay between TOR and autophagy during development and immunity, we screened and identified a TOR homologue from the coleopteran insect, T enebrio molitor ( T m TOR). The full-length cDNA of TmTOR contained an open reading frame (ORF) of 7,197 bp encoding a protein of 2,398 amino acids, and a 5′- and 3′- untranslated region (UTR) of 156 and 457 bp, respectively. Deduced amino acid sequence of T m TOR shows characteristic Huntington, EF3A, ATM, TOR ( HEAT) repeat, focal adhesion kinase targeting ( FAT), rapamycin-binding, phosphatidylinositol 3-/4-kinase and FRAP, ATM and TRRAP C-terminal ( FATC) domains known to be conserved among TOR orthologs. T m TOR showed high sequence identity (92%) with T ribolium castaneum TOR ( T c TOR) and the two were placed in the same cluster of the phylogenetic tree. T m TOR shows high m RNA expression level in the fat body and integument of T . molitor larvae. Significantly, a reduced expression of T m TOR m RNA during pupation correlated to an increase in the area of autolysosomes. Intriguingly, T . molitor larvae showed an increase in Tm TOR transcripts upon a challenge with various bacterial pathogens. Abstract

Grassland butterflies are considered representative indicators of biodiversity and ecosystem health. Their dramatic decline caused by habitat destruction, intensifying agriculture and global warming has prompted concern for conservation. While ecological indices of butterflies have been documented, there is a lack of genomic resources relative to the biological importance of this group. Here, we report, first whole-transcriptomic resource for F abriciana nerippe, a brush-footed butterfly member that is endangered in Korea. Approximately, 241.3 million clean reads were obtained from paired-end Illumina sequencing of adult whole-body RNA. The de novo assembly resulted in 114 405 unigenes with length ranging from 133 to 33 218 bp. We found 41 868 assembled unigenes homologous to sequences in locally curated PANM-DB ( Protostome DB). We assigned gene ontology ( GO) and Kyoto Encyclopedia of Genes and Genomes ( KEGG) orthology terms to 18 085 and 5779 unigenes, respectively. Inter Pro Scan predicted 6691 protein domains for the assembled unigene sequences, with the reverse transcriptase and zinc finger domains found to be the most predominant. A total of 315 282 simple sequence repeats ( SSRs) were identified from the assembled unigenes, when considering dinucleotide to octanucleotide repeats with a minimum of three repeat units. AT/ AT, AAT/ ATT, and AAAT/ ATTT represented the most prominent SSR repeat types in the transcriptome. The unigene annotation profile and microsatellites generated in the study can serve as a reference resource for closely related species and gene functional analysis studies. Abstract

Vespa mandarinia found in the forests of East Asia, including Korea, occupies the highest rank in the arthropod food web within its geographical range. It serves as a source of nutrition in the form of Vespa amino acid mixture and is listed as a threatened species, although no conservation measures have been implemented. Here, we performed de novo assembly of the V. mandarinia transcriptome by Illumina HiSeq 4000 sequencing. Over 60 million raw reads and 59,184,811 clean reads were obtained. After assembly, a total of 66,837 unigenes were clustered, 40,887, 44,455, and 22,390 of which showed homologous matches against the PANM, Unigene, and KOG databases, respectively. A total of 15,675 unigenes were assigned to Gene Ontology terms, and 5,132 unigenes were mapped to 115 KEGG pathways. The zinc finger domain (C2H2-like), serine/threonine/dual specificity protein kinase domain, and RNA recognition motif domain were among the top InterProScan domains predicted for V. mandarinia sequences. Among the unigenes, we identified 534,922 cDNA simple sequence repeats as potential markers. This is the first transcriptomic analysis of the wasp V. mandarinia using Illumina HiSeq 4000.
