Robust and accurate step counting plays an essential role in some fields, such as indoor positing, behavior recognition, and health management. Currently, there are numerous applications or methods in step counting. However, most solutions did not emphasize the elimination of false steps in the non-walking state, which still encounters the over-counting or under-counting problem. In this study, a robust and accurate step counting solution based on movement mode recognition is proposed. First, according to the characteristics of pedestrians walking, the SVM and FSM-DT classifiers were constructed to recognize the user’s motion state and phone usage mode, respectively. The purpose of classification is to solve the step-counting error of non-walking state and initialize a suitable parameter for different cases. Then, we utilized a crest-valley detection algorithm with multi-feature constrained to detect the step, and the initial threshold values for each mode can be adjusted adaptively during walking. The results indicate that the accuracy of the proposed method can reach 99.11% and 97.43% in normal and free walking. Compared with standard peak detection and an excellent method, the proposed method can improve by 44.14% and 18.71% for false walking, respectively, which is a significant improvement in the robustness and accuracy. Furthermore, we also achieve an average accuracy higher than 98% in some typically carrying modes and ways, and higher accuracy compared with four popular step-counting mobile applications in different walking states.

TANK-binding kinase 1 (TBK1) is a core enzyme downstream of several inflammatory signals. Recent studies brought the impacts of TBK1 in malignant disease to the forefront, we found aberrant TBK1 expression in CRC is correlated with CRC progression. The integrated navigation of single-frequency global navigation satellite system (GNSS) and inertial navigation system (INS) has broad application prospects with the development of multi-constellation GNSS. However, GNSS signals are susceptible to interference from a complex environment, which results in low-quality observations. The combination of frequencies cannot be applied for a single-frequency signal, so it is difficult to fix the ambiguity and process cycle slip. Therefore, the method of ambiguity resolution and cycle slip processing with INS aiding is proposed in this paper. When full ambiguities cannot be completely fixed, a subset of ambiguity is determined based on the elevation angle and posteriori residuals. The satellite is firstly eliminated one by one if the absolute value of posteriori residual is over the set threshold. And if the ambiguities still cannot be fixed, the satellite with the minimum elevation angle is eliminated each one. The short-term and high-accuracy position predicted by the INS was used as the constraint equation of the cycle slip detection. The unit weight mean square error of the cycle slip detection equation was used to identify the epoch where the cycle slip occurs, and the residual calculated by robust estimation was used to detect and repair the cycle slip. A set of data collected by a vehicle was used to analyze the proposed method. The experimental results showed that the proposed partial ambiguity resolution can obtain the optimal ambiguity fixing rate and positioning performance compared with the full ambiguity resolution and partial ambiguity resolution based on the elevation angle. The proposed algorithm can effectively detect and accurately repair the cycle slip for the simulated small and large cycle slips, and the cycle slips can still be detected when the global positioning system (GPS), global navigation satellite system of Russia (GLONASS), and BeiDou navigation satellite system (BDS) were outage for 10 s, 5 s, and 10 s without sufficient visible satellites.

This article presents an event-triggered neural-network (NN) tracking control scheme, capable of ensuring transient performance for switched nonlinear systems. A mode-dependent event-triggered communication mechanism (MDETCM) is designed, and this significantly saves communication resources without limiting the number of switches between two consecutive triggering instants. Meanwhile, to solve the impact of asynchronous switching on system performance, the information of the switching signal is considered into the event-triggered mechanism (ETM). Also, by introducing a normalized function transformation and a tan-type barrier function, the transient performance is regarded as a tracking error constraint without considering the initial conditions of system output and reference signal required in traditional prescribed performance bound (PPB) control. At the same time, the improved admissible edge-dependent average dwell time (AED-ADT) method is cleverly connected with adaptive backstepping control, and a state-feedback tracking algorithm is proposed, under which all closed-loop signals are bounded and the transient performance of the controlled plant is ensured. Finally, the superiority of the proposed scheme is demonstrated through numerical studies, and the control scheme is available for a single-link robot.

Cylindrospermopsin (CYN) is a toxic secondary metabolite from cyanobacteria that can cause cardiovascular disease. However, the study of CYN-induced cardiovascular toxicity in vitro is very limited and the mechanism is remain to be clarified. Vascular smooth muscle cells (VMSCs) have an important function in maintaining the structural and functional integrity of the aortic wall, and are an important in vitro model for cardiovascular research. Thus, the effects of CYN exposure (2, 20, 200, and 2000 nM) on VMSCs were analyzed. In vitro study, results showed that CYN exposure decreased VMSCs viability, inhibited VMSCs migration, induced DNA damage, destroyed cytoskeleton, changed cell morphology, promoted VMSCs apoptosis, and increased intracellular reactive oxygen species (ROS) levels. In addition, CYN could induce the activities of SOD, CAT and GPX, and promote the expressions of SOD1, CAT, GPx1, p53 and Bax genes and inhibit the expression of Bcl-2 gene, leading to a higher ratio of Bax/Bcl-2. Taken together, CYN may induce ROS overproduction, leading to increased p53 expression and ultimately promoting VSMC apoptosis. Therefore, the present study demonstrates that CYN could impair VMSCs, leading to vascular developmental defects and angiocardiopathy.

Diimine (HN═NH) is a strong reducing agent, but the efficiency of diimine oxidized from hydrazine hydrate or its derivatives is still not good enough. Herein, we report an in situ neocuproine-copper complex formation method. The redox potential of this complex enable it can serve as an ideal redox catalyst in the synthesis of diimine by oxidation of hydrazine hydrate, and we successfully applied this technique in the reduction of alkynes. This reduction method displays a broad functional group tolerance and substrate adaptability as well as the advantages of safety and high efficiency. Especially, nitro, benzyl, boc, and sulfur containing alkynes can be reduced to the corresponding alkanes directly, which provides a useful complementary method to traditional catalytic hydrogenation. Besides, we applied this method in the preparation of the Alzheimer's disease drug CT-1812 and studied the mechanism.

SET protein is a multi-functional oncoprotein that is ubiquitously expressed in most tumor cells. Dysregulation of SET has been associated with many types of cancer. Due to ever-accumulating evidence of its strong correlation with both poor prognosis and drug resistance, the targeting of SET is starting to be explored. SET is currently regarded as a potential target for cancer therapy, and several inhibitors are being developed for clinical trials. In this review, the physiological and pathological functions of SET, as well as its antagonists, will be discussed along with the prospects and challenges involved with translating SET inhibitors into bona fide therapeutic options.

Colorectal cancer (CRC) is the second most common cancer-related deaths throughout the world. Ferroptosis is a recently regulated form of cell death, lately gains attention. MicroRNA-15a-3p (miR-15a-3p) plays a regulatory role in various kinds of cancers. However, the role of miR-15a-3p in cellular ferroptosis is still unclear. Here, we aimed to clarify whether miR-15a-3p could regulate the ferroptosis of CRC. Here we identified miR-15a-3p positively regulates ferroptosis via directly targeting glutathione peroxidase glutathione peroxidase 4 (GPX4) in CRC. Overexpression of miR-15a-3p repressed GPX4 through binding to the 3'-untranslated region of GPX4, resulting in increased reactive oxygen species level, intracellular Fe2+ level, and malondialdehyde accumulation in vitro and in vivo. Correspondingly, suppression of miR-15a-3p reduced the sensitivity of CRC cells to erastin and GPX4. Taken together, these data demonstrate that miR-15a-3p regulates ferroptosis through targeting GPX4 in CRC cells, illustrating the novel role of microRNA in ferroptosis.

Edge cloud computing is a new paradigm in which the computation and storage services of remote cloud data centers are moved to Edge Cloud Nodes (ECNs) in network edges. Compared to traditional cloud data centers, ECNs are geographically close to mobile users so the communication latency is significantly reduced. In this paper, we study the problem of allocating Virtual Machine (VM) resources in geo-distributed ECNs to mobile users by using the auction theory. First, we treat mobile users and ECNs as the buyers and sellers of the VM resource auction, respectively. Then, we model the VM resource allocation problem as an n-to-one weighted bipartite graph matching problem with 0-1 knapsack constraints. Since this problem is NP-hard, we design a greedy approximation algorithm to determine the winners of the auction, based on which we propose a truthful Auction-based VM resource Allocation (AVA) mechanism to solve the problem. Moreover, we prove that the AVA mechanism not only achieves an approximately optimal solution for winner selection, but also has the properties of truthfulness, individual rationality, and computational efficiency. Finally, we conduct extensive simulations on real traces to verify the significant performances of the proposed AVA mechanism.

Daylighting structures, including solar cells and building windows, utilize sunlight whilst suffering from undesired solar heat and outdoor dust contamination. A radiative cooling system that is transparent to sunlight and has a superhydrophobic surface would cool and clean the daylighting structures in a sustainable manner. However, the majority of the current daytime radiative cooling systems were designed to fully reflect the incident sunlight to maximize the cooling power. In this work, we optimized both the sunlight transmission and infrared thermal irradiation by modeling the size-dependent scattering and absorption of light by SiO2 spheres embedded in a polymer matrix, we found that the use of nanospheres (20 nm) enabled both high sunlight transmittance (> 90%) and infrared emissivity (~0.85). This theoretical prediction was confirmed by experimental measurements of a solution-processed nanocomposite film. When coated on a solar cell, the as-prepared film not only preserved the power conversion efficiency of the cell (14.71%, uncoated cell has an efficiency of 14.79%) but also radiatively cooled the cell by up to 5 °C under direct sunlight. This reduction of the operating temperature of the solar cell further enhanced its electrical power output, evidenced by an increase in the equilibrium temperature of the LED load by about 14 °C. The nanoscale textured surface formed by the nanospheres further led to superhydrophobicity and thus excellent self-cleaning performance (efficient removal of dust by wind and/or water droplets).

In Alzheimer's disease (AD), damaged Aβ clearance contributes to elevated levels of Aβ that cause a series of cytotoxic cascade reactions. Thus, targeting Aβ clearance has now been considered a valid therapeutic approach for AD. Cellular uptake and degradation are important mechanisms for Aβ clearance, which are mainly performed by the endosomal-autophagic-lysosomal (EAL) pathway. Our previous study showed that OAB-14, a novel small molecule designed with bexarotene as the lead compound, treatment for 3 months significantly alleviated cognitive disorders and remarkably reduced the deposition of Aβ without affecting its production in APP/PS1 transgenic mice. Here, we further revealed that enhancement of the EAL activity is one of the mechanisms that increases Aβ clearance after OAB-14 administration for 3 months. OAB-14 facilitates receptor-mediated endocytosis and restores autophagy flux via the AMPK/mTOR pathway. Meanwhile, OAB-14 enhances the lysosomal activity, and reduced Aβ accumulation in lysosomes was observed in OAB-14-treated AD mice. These results suggest that OAB-14 may promote Aβ clearance in lysosomes by alleviating the EAL dysfunction in AD mice.

Background Colon cancer is one of the most common malignant tumors, with high rates of incidence and death. The tumor mutational burden (TMB), which is characterized by microsatellite instability, has been becoming a powerful predictor which can show tumor behavior and response to immunotherapy. Methods In this study, we analyzed 437 mutation data of colon cancer samples obtained from The Cancer Genome Atlas (TCGA) and divided patients into low- and high-TMB groups according to the TMB value. Then we identified differentially-expressed genes (DEGs), conducted immune cell infiltration and survival analyses between groups. Results The higher TMB of the patients with colon cancer predicts a poorer prognosis. Functional analysis was performed to assess the prognostic value of the top 30 core genes. The CIBER-SORT algorithm was used to investigate the correlation between the immune cells and TMB subtypes. An immune prognosis model was constructed to screen out immune genes related to prognosis, and the tumor immunity assessment resource (TIMER) was then used to determine the correlation between gene expression and the abundance of tumor-infiltrating immune cell subsets in colon cancer. We observed that APC, TP53, TTN, KRAS, MUC16, SYNE1, PIK3CA have higher somatic mutations. DEGs enrichment analysis showed that they are involved in the regulation of neuroactive ligand-receptor interaction, the Cyclic adenosine monophosphate (cAMP) signaling pathway, the calcium signaling pathway, and pantothenate and Coenzyme A (CoA) biosynthesis. The difference in the abundance of various white blood cell subtypes showed that Cluster of Differentiation 8 (CD8) T cells (P=0.008), activated CD4 memory T cells (P=0.019), M1 macrophages (P=0.002), follicular helper T cells (P=0.034), activated Natural killer (NK cell) cells (P=0.017) increased remarkably, while M0 macrophages significantly reduced (P=0.025). The two immune model genes showed that secretin (SCT) was negatively correlated with survival, while Guanylate cyclase activator 2A (GUCA2A) was positively correlated. Conclusions This study conducted a systematically comprehensive analysis of the prediction and clinical significance of TMB in colon cancer in identification, monitoring, and prognosis of colon cancer, and providing reference information for immunotherapy.

BACKGROUND AND PURPOSE The spinal cord central echo complex (SCCEC) is a special ultrasonography-based intramedullary structure, but its clinical significance in degenerative cervical myelopathy (DCM) is undefined. This study aimed to explore the potential of the SCCEC in predicting postoperative neurological recovery in DCM. METHODS Thirty-two DCM patients who underwent intraoperative ultrasonography-guided French-door laminoplasty were prospectively enrolled. The modified Japanese Orthopaedic Association (mJOA) score was evaluated preoperatively and 12 months postoperatively. SCCEC width (SCCEC-W), and anteroposterior diameter (APD) and transverse diameter (TD) of the spinal cord were measured on transverse ultrasonographic images, while the tissue widths from anterior and posterior borders of the spinal cord to the SCCEC were measured on sagittal ultrasonographic images. The APD of the spinal cord and occupying rate of the spinal canal were measured on preoperative magnetic resonance imaging (MRI). RESULTS All patients achieved improvements in mJOA scores, with an average recovery rate (RR) of 68.69 ± 20.22%. Spearman correlation analysis revealed that SCCEC-W, and ratios between the SCCEC-W and APD/TD based on ultrasonography, correlated moderately with mJOA score RR, with coefficients of -0.527, -0.605 and -0.514, respectively. The ratio between SCCEC-W and ultrasonographic TD correlated moderately with preoperative APD of the spinal cord. The MRI measurements and ultrasonography-based tissue widths showed no significant correlation with mJOA score RR. CONCLUSIONS The SCCEC may have predictive potential as an intraoperative indicator of neurological recovery in treating DCM. SCCEC-W may be related to spinal cord compression in DCM.

This article outlines the research progress on radiocarbon (14C) dating of the Erlitou site. The Erlitou site, belonging to the Bronze Age, located in Yanshi, Henan province, China, was discovered by archaeologists in 1959 when they investigated the Xia people’s remains in the area where the Xia people lived according to the records of ancient documents. Since then, there has been a standing debate about whether the site belongs to the Xia or Shang dynasty. By the mid-1990s, several hundred discussion articles on the issue had been published, but the question was still unresolved. Therefore, evidence from the chronology has attracted a great amount of attention. The dating of the Erlitou site began in the 1970s, and since the Xia-Shang-Zhou Chronology Project began in the mid-1990s, by application of wiggle-matching on the basis of improving the dating accuracy, the date of the Erlitou site has gradually become clear, which provides a basis for the archaeological research on the Xia and Shang dynasties.

The acoustic echo cannot be entirely removed by linear adaptive filters due to the nonlinear relationship between the echo and the far-end signal. Usually, a post-processing module is required to further suppress the echo. In this paper, we propose a residual echo suppression method based on the modification of dual-path recurrent neural network (DPRNN) to improve the quality of speech communication. Both the residual signal and the auxiliary signal, the far-end signal or the output of the adaptive filter, obtained from the linear acoustic echo cancelation are adopted to form a dual-stream for the DPRNN. We validate the efficacy of the proposed method in the notoriously difficult double-talk situations and discuss the impact of different auxiliary signals on performance. We also compare the performance of the time domain and the time-frequency domain processing. Furthermore, we propose an efficient and applicable way to deploy our method to off-the-shelf loudspeakers by fine-tuning the pre-trained model with little recorded-echo data.
