Abstract

LUBAC (linear ubiquitin chain assembly complex) activates the canonical NF-?B pathway through linear polyubiquitination of NEMO (NF-?B essential modulator, also known as IKK?) and RIP1. However, the regulatory mechanism of LUBAC-mediated NF-?B activation remains elusive. Here, we show that A20 suppresses LUBAC-mediated NF-?B activation by binding linear polyubiquitin via the C-terminal seventh zinc finger (ZF7), whereas CYLD suppresses it through deubiquitinase (DUB) activity. We determined the crystal structures of A20 ZF7 in complex with linear diubiquitin at 1.70-1.98?Å resolutions. The crystal structures revealed that A20 ZF7 simultaneously recognizes the Met1-linked proximal and distal ubiquitins, and that genetic mutations associated with B cell lymphomas map to the ubiquitin-binding sites. Our functional analysis indicated that the binding of A20 ZF7 to linear polyubiquitin contributes to the recruitment of A20 into a TNF receptor (TNFR) signalling complex containing LUBAC and I?B kinase (IKK), which results in NF-?B suppression. Abstract

Recently, novel E3 ubiquitin ligases that target MHC molecules for lysosomal degradation have been discovered by several groups. All these E3s are membrane-bound and possess a variant type RING domain, termed the RING-CH or RING variant (RINGv) domain. They belong to a new E3 family designated Modulator of Immune Recognition (MIR), based on the name of the first identified family members. The discovery of the MIR family has provided fresh insight into viral pathogenesis and immune regulation. [Copyright &y& Elsevier]

Abstract

The objective of this study was to investigate the growth and light-intercepting characteristics of tomatoes when movable benches are used in their cultivation. We cultivated tomatoes in a greenhouse (168 m2) during summer (9 July–9 September 2018) under different furrow distances (F1.0 = 1.0 m and F1.6 = 1.6 m) and movable benches (M indicates that the furrow distance = 0.4–0.8 m). Compared to the other treatments, when the movable bench was used to the change furrow distance depending on the plant growth stage (M treatment), the percentage of canopy light interception increased to ~90% at the early stage of plant growth (~20 days after transplanting). The percentage of canopy light interception for different treatments increased in the order of M > F1.0 > F1.6, and it increased towards the end of cultivation. In addition, the yield per unit area exhibited the same trend. Therefore, the solar radiation inside a greenhouse can be efficiently intercepted by plants when movable benches are used. This indicated that it was possible to increase plant yield per unit area using movable benches in plant cultivation. [ABSTRACT FROM AUTHOR]
