Abstract

We report experimental results on the diffusivity of water in two ionic liquids obtained using the pulsed-gradient spin-echo NMR method. Both ionic liquids have the same cation, 1-butyl-1- methylpyrrolidinium, but different trifluoromethyl-containing anions. One has a strongly hydrophobic anion, bis(trifluoromethylsulfonyl)amide, while the second has a hydrophilic anion, trifluoromethylsulfonate. Transport of water in these ionic liquids is much faster than would be predicted from hydrodynamic laws, indicating that the neutral water molecules experience a very different friction than the anions and cations at the molecular level. Temperature-dependent viscosities, conductivities, and densities are reported as a function of water concentration to further analyze the properties of the ionic liquid-water mixtures. These results on the properties of water in ionic liquids should be of interest to researchers in diverse areas ranging from separations, solubilizing biomass and energy technologies. Abstract

Abstract Background Despite twinning being common in Africa, few prospective twin studies have been conducted. We studied twinning rate, perinatal mortality and the clinical characteristics of newborn twins in urban Guinea-Bissau. Methods The study was conducted at the Bandim Health Project (BHP), a health and demographic surveillance site in Bissau, the capital of Guinea-Bissau. The cohort included all newborn twins delivered at the National Hospital Simão Mendes and in the BHP study area during the period September 2009 to August 2011 as well as singleton controls from the BHP study area. Data regarding obstetric history and pregnancy were collected at the hospital. Live children were examined clinically. For a subset of twin pairs zygosity was established by using genetic markers. Results Out of the 5262 births from mothers included in the BHP study area, 94 were twin births, i.e. a community twinning rate of 18/1000. The monozygotic rate was 3.4/1000. Perinatal mortality among twins vs. singletons was 218/1000 vs. 80/1000 (RR = 2.71, 95% CI: 1.93-3.80). Among the 13783 hospital births 388 were twin births (28/1000). Abstract

Emergency granulopoiesis is a hematopoietic program of stem cell-driven neutrophil production used to counteract immune cell exhaustion following infection. Shigella flexneri is a Gram-negative enteroinvasive pathogen controlled by neutrophils. In this study, we use a Shigella-zebrafish (Danio rerio) infection model to investigate emergency granulopoiesis in vivo. We show that stem cell-driven neutrophil production occurs in response to Shigella infection and requires macrophage-independent signaling by granulocyte colony-stimulating factor (Gcsf). To test whether emergency granulopoiesis can function beyond homoeostasis to enhance innate immunity, we developed a reinfection assay using zebrafish larvae that have not yet developed an adaptive immune system. Strikingly, larvae primed with a sublethal dose of Shigella are protected against a secondary lethal dose of Shigella in a type III secretion system (T3SS)-dependent manner. Collectively, these results highlight a new role for emergency granulopoiesis in boosting host defense and demonstrate that zebrafish larvae can be a valuable in vivo model to investigate innate immune memory., IMPORTANCE Shigella is an important human pathogen of the gut. Emergency granulopoiesis is the enhanced production of neutrophils by hematopoietic stem and progenitor cells (HSPCs) upon infection and is widely considered a homoeostatic mechanism for replacing exhausted leukocytes. In this study, we developed a Shigella-zebrafish infection model to investigate stem cell-driven emergency granulopoiesis. We discovered that zebrafish initiate granulopoiesis in response to Shigella infection, via macrophage-independent signaling of granulocyte colony-stimulating factor (Gcsf). Strikingly, larvae primed with a sublethal dose of Shigella are protected against a secondary lethal dose of Shigella in a type III secretion system (T3SS)-dependent manner. Taken together, we show that zebrafish infection can be used to capture Shigella-mediated stem cell-driven granulopoiesis and provide a new model system to study stem cell biology in vivo. Our results also highlight the potential of manipulating stem cell-driven granulopoiesis to boost innate immunity and combat infectious disease.

160. Thermomechanical behavior of circular steel arches

Abstract

Circular steel arches are structural members curved in the plane of the applied loads, where the radius of curvature is much higher than the cross-section depth. The main objective of this work is to evaluate the behaviour of circular arches at room and elevated temperatures, like those that can arise from fires. The in plane buckling of arches is analysed numerically, using ANSYS software, considering different domains, linear elastic analysis, nonlinear elastic analysis and nonlinear geometric and material analysis, where the structural capacity of arches are studied in function of the geometric imperfections and residual stresses. The results are compared with those obtained by the simplified methods of Eurocode 3 part 1.1 and 1.2. The circular steel arches considered comprise a parametric analysis for arches with different spans, radius of curvature and opening angle at different temperature values, and subjected to an uniform radial distributed load. The values of the critical compression loads and the collapse loads were obtained for the different arcs, supports and temperatures. For the different geometries, the results allows to define the instability failure modes, symmetric or antisymmetric. It is concluded that increasing the arch height to the span ratio produces a decrease of the elastic critical load and the collapse load of the arch. The collapse mode for shorter arches is due to the formation of a plastic hinge whereas in the long span arches the structure will became instable before plastification, producing in plane instability in its stronger plane. info:eu-repo/semantics/publishedVersion

Abstract

Porous ionic liquids are non‐volatile, versatile materials that associate porosity and fluidity. New porous ionic liquids, based on the ZIF‐8 metal–organic framework and on phosphonium acetate or levulinate salts, were prepared and show an increased capacity to absorb carbon dioxide at low pressures. Porous suspensions based on phosphonium levulinate ionic liquid absorb reversibly 103 % more carbon dioxide per mass than pure ZIF‐8 at 1 bar and 303 K. We show how the rational combination of MOFs with ionic liquids can greatly enhance low pressure CO2 absorption, paving the way towards a new generation of high‐performance, readily available liquid materials for effective low pressure carbon capture. Abstract

The glass transition pressure at room temperature, Pg, of six ionic liquids based on 1-alkyl-3- methylimidazolium cations and the anions [BF4]-, [PF6]-, and bis(trifluromethanesulfonyl)imide, [NTf2]-, has been obtained from the pressure dependence of the bandwidth of the ruby fluorescence line in diamond anvil cells. Molar volume, Vm(Pg), has been estimated by a group contribution model (GCM) developed for the ionic liquids. A density scaling relation, TVγ, has been considered for the states Vm(Pg, 295 K) and Vm(Tg, 0.1 MPa) using the simplifying condition that the viscosity at the glass transition is the same at Pg at room temperature and at atmospheric pressure at Tg. Assuming a constant γ over this range of density, a reasonable agreement has been found for the γ determined herein and that of a previous density scaling analysis of ionic liquids viscosities under moderate conditions. Further support for the appropriateness of extrapolating the GCM equation of state to the GPa pressure range is provided by comparing the GCM and an equation of state previously derived in the power law density-scaling regime. Abstract

Polymer mixtures based on native corn flour and synthetic biodegradable polyester are prepared in one processing step. Melt mixing is performed in a laboratory micro-compounder. Starch contained in native corn flour is plasticized either by commonly used glycerol/water mixture or by two types of ionic liquids, 1-allyl-3-methylimidazolium chloride, and 1-butyl-3-methylimidazolium chloride. Efficiency of plasticizers on the compatibility of native corn flour with polyester and starch plasticizing ability is evaluated and compared. The structure of mixtures is examined using scanning electron microscopy and wide-angle X-ray scattering. It is found that the plasticizer applied has a crucial effect on the resulting morphology. Abstract

The opportunistic pathogen Burkholderia cenocepacia is particularly life-threatening for cystic fibrosis (CF) patients. Chronic lung infections with these bacteria can rapidly develop into fatal pulmonary necrosis and septicaemia. We have recently shown that macrophages are a critical site for replication of B. cenocepacia K56-2 and the induction of fatal pro-inflammatory responses using a zebrafish infection model. Here, we show that ShvR, a LysR-type transcriptional regulator that is important for biofilm formation, rough colony morphotype and inflammation in a rat lung infection model, is also required for the induction of fatal pro-inflammatory responses in zebrafish larvae. ShvR was not essential, however, for bacterial survival and replication in macrophages. Temporal, rhamnose-induced restoration of shvR expression in the shvR mutant during intramacrophage stages unequivocally demonstrated a key role for ShvR in transition from intracellular persistence to acute fatal pro-inflammatory disease. ShvR has been previously shown to tightly control the expression of the adjacent afc gene cluster, which specifies the synthesis of a lipopeptide with antifungal activity. Mutation of afcE, encoding an acyl-CoA dehydrogenase, has been shown to give similar phenotypes as the shvR mutant. We found that, like shvR, afcE is also critical for the switch from intracellular persistence to fatal infection in zebrafish. The closely related B. cenocepacia H111 has been shown to be less virulent than K56-2 in several infection models, including Galleria mellonella and rats. Interestingly, constitutive expression of shvR in H111 increased virulence in zebrafish larvae to almost K56-2 levels in a manner that absolutely required afc. These data confirm a critical role for afc in acute virulence caused by B. cenocepacia that depends on strain-specific regulatory control by ShvR. The insect repellency and insecticidal properties of Anthemis sp., Coriandrum sativum and Piper nigrum against the maize weevil, Sitophilus zeamais Motschulsky, 1855 (Coleoptera: Curculionidae), were investigated in order to find possible natural sources to allow the protection of stored grains. Two experiments were installed with a completely randomized design, using two cultivars of hybrid maize (AS 1551 YG and AS 1555 YG) as food substrates. The first experiment aimed to verify the mortality of the pest after 7 days of exposure to the studied plants at 0.5g/treatment, with a factorial arrangement (control, three plant species, two maize cultivars). In the second experiment, the pest repellency promoted by each plant species was evaluated in an arena. After 24 hours, the percentage of adults repelled (% AR) was calculated. The study showed that P. nigrum presented insecticidal properties, observing a mean mortality of 49%. Anthemis sp. presented a repellent effect (I.R./I.D. = 0.44) on S. zeamais when the substrate used was the hybrid AS 1555 YG, with 26.5% adults repelled. As for the hybrid AS 1551 YG, P. nigrum showed a repellent effect (I.R./I.D. = 0.68 and 31% of adults repelled), while C. sativum and Anthemis sp. were considered neutral against the maize weevil., Plantas podem apresentar propriedades bioativas inseticidas como mecanismo de defesa natural contra algumas espécies de insetos-praga. Visando investigar possíveis fontes naturais que contribuam para a proteção de grãos armazenados, foram avaliadas as propriedades de repelência e inseticida de macerados de Anthemis sp., Coriandrum sativum e Piper nigrum sobre o gorgulho-do-milho, Sitophilus zeamais Motschulsky, 1855 (Coleoptera: Curculionidae). Dois experimentos foram instalados com delineamento inteiramente casualizado, utilizando-se dois cultivares de milho híbrido (AS 1551 YG e AS 1555 YG) como substratos alimentares. O primeiro experimento objetivou verificar a mortalidade da praga após sete dias de exposição aos vegetais estudados, com delineamento em arranjo fatorial (testemunha; três espécies vegetais; dois cultivares de milho). No segundo experimento, avaliou-se a repelência da praga promovida pelas espécies vegetais, testadas isoladamente em uma arena. Após 24 horas, a porcentagem de adultos repelidos (% AR) foi calculada. O estudo permitiu verificar que P. nigrum apresenta propriedade inseticida, observando uma mortalidade média de 49%. Anthemis sp. demonstrou efeito repelente (I.R./I.D. = 0,44) sobre S. zeamais quando o substrato utilizado foi o híbrido AS 1555 YG, com 26,5% de adultos repelidos. Já para o híbrido AS 1551 YG, P. nigrum mostrou efeito repelente (I.R./I.D. = 0,68 e 31% de adultos repelidos), enquanto C. sativum e Anthemis sp. foram consideradas neutras.

International audience; Bacteria of the Burkholderia cepacia complex (Bcc) can cause devastating pulmonary infections in cystic fibrosis (CF) patients, yet the precise mechanisms underlying inflammation, recurrent exacerbations and transition from chronic stages to acute infection and septicemia are not known. Bcc bacteria are generally believed to have a predominant extracellular bio-film life style in infected CF lungs, similar to Pseudomonas aeruginosa, but this has been challenged by clinical observations which show Bcc bacteria predominantly in macro-phages. More recently, Bcc bacteria have emerged in nosocomial infections of patients hospitalized for reasons unrelated to CF. Research has abundantly shown that Bcc bacteria can survive and replicate in mammalian cells in vitro, yet the importance of an intracellular life style during infection in humans is unknown. Here we studied the contribution of innate immune cell types to fatal pro-inflammatory infection caused by B. cenocepacia using zebra-fish larvae. In strong contrast to the usual protective role for macrophages against microbes, our results show that these phagocytes significantly worsen disease outcome. We provide new insight that macrophages are critical for multiplication of B. cenocepacia in the host and for development of a fatal, pro-inflammatory response that partially depends on Il1-signalling. In contrast, neutrophils did not significantly contribute to disease outcome. In subcuta-neous infections that are dominated by neutrophil-driven phagocytosis, the absence of a functional NADPH oxidase complex resulted in a small but measurably higher increase in bacterial growth suggesting the oxidative burst helps limit bacterial multiplication; however, neutrophils were unable to clear the bacteria. We suggest that paradigm-changing approaches are needed for development of novel antimicrobials to efficiently disarm intra-cellular bacteria of this group of highly persistent, opportunistic pathogens. Abstract

Arches are metal structural members that are curved in the plane of the applied loads, where the radius of curvature is much higher than the cross-section depth. Due to their geometry, they have different mechanical properties than conventional straight beams. The main objective of this work is to evaluate the behavior of circular arches at room and elevated temperatures, like those that can arise from fires. The in plane buckling of arches plane was analyzed numerically, using ANSYS software and the results compared with those obtained by the simplified methods of Eurocode 3 part 1.1 and 1.2. The critical buckling load is further compared to the classical buckling theory. Linear elastic analysis, nonlinear elastic analysis and nonlinear geometric and material analysis were considered. The analyzed arches comprise a parametric analysis for arches with different spans, radius of curvature and opening angle for different temperature values. The arches are subjected to uniform radial pressure. The values of the critical compression loads and the collapse loads were obtained for the different arcs, supports and temperatures. The results also allows to define the instability modes, symmetrical or antisymmetric. It is concluded that increasing the arch height to the span ratio produces a decrease of the elastic critical load and the collapse load of the arch. The collapse mode for shorter arches is due to the formation of a plastic hinge whereas in the long span arches the structure will became instable before plastification, producing in plane instability in its stronger plane. info:eu-repo/semantics/publishedVersion
