Abstract

The escalation of cancer cases globally, especially breast cancer, is of concern. Angiogenesis is hallmark of cancer pathogenesis and plays an important role in cancer progression and metastasis. Pro-angiogenic agents, secreted by tumor cells, form new blood vessels, and produce reactive oxygen species (ROS). ROS promote angiogenesis via two major pathways: namely Vascular Endothelial Growth Factor (VEGF) dependent and non-VEGF dependent pathways. As a consequence of unbalanced ROS overproduction and low antioxidants levels, oxidative stress occurs and promotes angiogenesis in breast cancer tissues. Thus, the potential use of antioxidants as a preventive therapy in breast cancer. Preclinical studies depict that vitamins A and E may counter oxidative stress resulting in reduction of metastasis and viability of breast cancer. Furthermore, clinical studies demonstrate a decline in breast cancer risk in postmenopausal women upon the consumption of antioxidants. Herein, we discuss various pro-angiogenic agents that may play an important role in breast cancer angiogenesis. Moreover, the contribution of oxidative stress in inducing the angiogenic process is extensively reviewed here. Furthermore, the findings of pre-clinical and clinical studies on the use of antioxidants, namely vitamins A and E, in breast cancer are deliberated upon, along with the role of angiogenesis in cancer therapy. Abstract

Friction stir spot welding (FSSW) process is widely used in the automobile industry for a range of applications such as battery components, standard wire connectors and terminals. This manuscript addresses two grand challenges in the arena of FSSW, hitherto, unaddressed in the extant literature: (i) lap joining of thin sheets (0.3 mm thickness) of AA 5754 alloy and (ii) lap joining of more than two sheets using FSSW. To accomplish this task, a novel pinless convex shaped tool was designed to alter the stress state while gradually advancing the tool which led to achieving stress state necessary for obtaining defect free lap joints. The weld joints were inspected by optical microscopy, SEM imaging and analysed by nanoindentation tests and Vickers microindentation tests for assessment of the quality of the weld interface (WI). Process parameters of FSSW such as torque on the tool and axially applied load were used to analytically obtain the average local measure of peak normal and axial stresses as well as the coefficient of friction in the contact zone. In samples welded at low rotational speeds, strain-hardening mechanism was seen dominating in contrast to samples welded at higher rotational speeds, which showed thermal softening. As a direct consequence of this, the samples welded at low rotational speeds showed much higher hardness at the weld surface than the samples welded at higher speeds. A strong transition of strain hardening to thermal softening was noticeable beyond an applied strain rate of 400 s-1. Abstract

Stromal tumor-infiltrating lymphocytes (sTILs) are a potential predictive biomarker for immunotherapy response in metastatic triple-negative breast cancer (TNBC). To incorporate sTILs into clinical trials and diagnostics, reliable assessment is essential. In this review, we propose a new concept, namely the implementation of a risk-management framework that enables the use of sTILs as a stratification factor in clinical trials. We present the design of a biomarker risk-mitigation workflow that can be applied to any biomarker incorporation in clinical trials. We demonstrate the implementation of this concept using sTILs as an integral biomarker in a single-center phase II immunotherapy trial for metastatic TNBC (TONIC trial, NCT02499367), using this workflow to mitigate risks of suboptimal inclusion of sTILs in this specific trial. Abstract

37th European Photovoltaic Solar Energy Conference and Exhibition; 985-988, Underwater Solar Photovoltaics have the potential to resolve the necessities of long-term power sources for underwater sensing, monitoring, and low power applications. Even though there are several challenges and limitations, however, our previous studies have demonstrated that submerged Solar cells can generate the required amount of power even with the presence of water impurities. In this work, the submerged amorphous, monocrystalline, and polycrystalline Solar cells, especially the near-infrared spectrum using the long pass glass filter, have been further studied. In order to analyze the performance of the submerged Solar cells with the infrared spectrum, the transmission of Solar radiation in water also studied. Further, the obtained results were compared to our previous studies without using the long pass filter, passing the full spectrum from the Solar simulator.

Abstract

Nematicidal efficacy of five botanicals, viz. turmeric (Curcuma longa), marwatulsi (Origanum majorana), mentha (Mentha arvensis), aonla (Phyllanthus emblica) and jatropha (Jatropha curcas) was evaluated under in vitro and in vivo conditions for their larvicidal properties against the root-knot nematode, Meloidogyne incognita in tomato during 2014–16 at Department of Nematology, CCSHAU, Hisar. Methanolic and hexane extracts of turmeric (Rhizome) showed varying degrees of lethality to M. incognita at different concentrations under in vitro conditions. Per cent larval mortality and inhibition of egg hatching were found to be directly proportional to the concentration of extracts and exposure period. In in vivo experiments, growth parameters and nematode reproduction factors (number of galls/ plant, number of egg masses/plant and final nematode population in soil) were evaluated post-treatment with chopped leaves of botanicals. The study showed that the treatment improved plant growth and reduced pathogen indices. The study suggests the representative role of botanicals in managing M. incognita in tomato plants.

Structured surfaces are attracting deep interest, as they allow tailoring the functionality via changes in the surface topography. Applications for these surfaces range greatly, including, optical surfaces for antireflective surfaces, thermal structures to assist in heat dispersion and anti-fouling surfaces to reduce micro-organisms from adhering to components. Gum metal is a relatively newer kind of beta titanium alloy that has earmarked its place as the next generation Ortheopedic implant material. In a timely effort, this work investigated the generation of micron level structured surfaces on Gum metal (Ti-30Nb – a beta titanium alloy) to explore micromilling as the robust scalable process to achieve low dimensional surfaces in titanium alloy. During micromilling, the feedrate, spindle speed, axial depth of cut and tool step over were varied to optimise these parameters for achieving superior quality of machining.

Abstract

OBJECTIVES:The aim of CATS (Cardiotoxicity of Adjuvant Trastuzumab Study) was to prospectively assess clinical, biochemical, and genomic predictors of trastuzumab-related cardiotoxicity (TRC). BACKGROUND:Cardiac dysfunction is a common adverse effect of trastuzumab. Studies to identify predictive biomarkers for TRC have enrolled heterogeneous populations and yielded mixed results. METHODS:A total of 222 patients with early-stage human epidermal growth factor receptor 2-positive breast cancer scheduled to receive adjuvant anthracyclines followed by 12 months of trastuzumab were prospectively recruited from 17 centers. Left ventricular ejection fraction (LVEF), troponin T, and N-terminal prohormone of brain natriuretic peptide were measured at baseline, post-anthracycline, and every 3 months during trastuzumab. Germline single-nucleotide polymorphisms in ERBB2, FCGR2A, and FCGR3A were analyzed. 185. Effects of fumonisin B1-contaminated feeds on weanling angora goats

Abstract

Two diets containing no (< 1.0 mg/ kg) or 95 mg of fumonisin [B.sub.1] (F[B.sub.1])/kg were fed to eight weanling Angora goats for 112 d. Dry matter intake, apparent nutrient digestibilities, serum chemistry profiles, sphingolipid concentrations, and persistency of F[B.sub.1] in tissues were evaluated. No differences (P > .10) were found between control and treated goats in terms of DMI, apparent nutrient digestibilities, or ADG. Elevated concentrations (P < .10) of blood-borne enzymes such as aspartate aminotransferase, lactate dehydrogenase, and gamma glutamyl transpeptidase and increased concentrations of cholesterol and triglycerides indicated mild liver damage and kidney dysfunction in treated goats. Linear relationships (P < .10) were observed between these serum constituents and duration of F[B.sub.1] exposure. The sphingolipid analysis of liver, kidney, and heart tissues showed elevated free sphinganine:free sphingosine ratios in.the treated group. The elevated sphingolipid ratios were mainly due to increased concentrations of free sphinganine in tissues. However, without serum profile and sphingolipid analyses, fumonisin toxicosis would not have been recognized because treated animals showed no clinical signs of toxicosis throughout the trial. No measurable [FB.sub.1] was present in liver, kidney, and heart tissues (detection limit of 1 ppm). However, further research is needed to analyze tissues for F[B.sub.1] or its metabolites with a lower detection limit. In conclusion, goats can be fed for up to 112 d with diets containing 95 mg [FB.sub.1]/kg of diet without any overt signs of toxicosis and also without any effect on live weight gain. Abstract

[EN] This work analyses the viability of the theory of critical distances (TCD) using mesh control for fretting fatigue lifetime assessment. More than seven hundred sets of simulations were performed by taking seventy different experimental tests reported previously in the literature. The outcome of the present study suggests that the TCD mesh control method can be extended to fretting fatigue problems by the reasonable assumption of setting the right element size proportional to critical distance. In this study, a significant computational time reduction of up to 97% was obtained. Thus, this study provides a simple method to design complex 3D industrial components subjected to fretting fatigue phenomena using finite element analysis efficiently without requiring complex remeshing techniques

Abstract

Angioid streaks (AS) are irregular crack-like dehiscences in Bruch's membrane that are often associated with atrophic degeneration of the overlying retinal pigment epithelium. We herein report multimodal imaging of AS. Multicolor imaging highlighted AS in dark orange color. AS were better visualized in infrared reflectance as compared to green reflectance and blue reflectance. Peau d'orange appearance was seen as alternating dark and bright patches on color fundus photography with corresponding hyporeflective and hyperreflective patches on infrared reflectance. Comet lesions showed increased signal on infrared reflectance and hyperautofluorescence. Multicolor imaging is a non-invasive imaging modality which helps in clearly delineating these lesions.

This recognition has come about thanks to standardized visual reporting guidelines, which helped to reduce inter-reader variability. Now, there are ripe opportunities to employ computational methods that extract spatio-morphologic predictive features, enabling computer-aided diagnostics. We detail the benefits of computational TILs assessment, the readiness of TILs scoring for computational assessment, and outline considerations for overcoming key barriers to clinical translation in this arena. Specifically, we discuss: 1. ensuring computational workflows closely capture visual guidelines and standards; 2. challenges and thoughts standards for assessment of algorithms including training, preanalytical, analytical, and clinical validation; 3. perspectives on how to realize the potential of machine learning models and to overcome the perceptual and practical limits of visual scoring.

Abstract

Stromal tumor-infiltrating lymphocytes (sTILs) are important prognostic and predictive biomarkers in triple-negative (TNBC) and HER2-positive breast cancer. Incorporating sTILs into clinical practice necessitates reproducible assessment. Previously developed standardized scoring guidelines have been widely embraced by the clinical and research communities. We evaluated sources of variability in sTIL assessment by pathologists in three previous sTIL ring studies. We identify common challenges and evaluate impact of discrepancies on outcome estimates in early TNBC using a newly-developed prognostic tool. Discordant sTIL assessment is driven by heterogeneity in lymphocyte distribution. Additional factors include: technical slide-related issues; scoring outside the tumor boundary; tumors with minimal assessable stroma; including lymphocytes associated with other structures; and including other inflammatory cells. Small variations in sTIL assessment modestly alter risk estimation in early TNBC but have the potential to affect treatment selection if cutpoints are employed. Scoring and averaging multiple areas, as well as use of reference images, improve consistency of sTIL evaluation.
