Abstract

G-quadruplex DNA stabilization mediated by small molecules is an attractive approach to modulate the transcription of guanine-rich sequences and contrast unregulated cell proliferation. Natural alkaloids have been reported to target this macromolecular arrangement, and such mechanism may be among these underlying the antiproliferative effect of some flavonoids. The binding affinity for G-quadruplex and double stranded DNA of 4 isoflavones from Maclura pomifera, namely osajin, pomiferin, scandenone and auriculasin, was evaluated using electrospray mass spectrometry (ESI-MS). The experiments pointed out that osajin and scandenone preferentially bind G-quadruplex. Moreover, since G-quadruplex stabilization is a crucial aim for triggering biological effects such as gene expression, collision-induced dissociation (CID) assays were performed to study the relative gas-phase kinetic stability of the DNA/ligand complexes. Osajin was identified as the best G-quadruplex stabilizer of the set, and in silico docking studies indicate that stacking is the preferred interaction motif of this natural compound. Abstract

In spite of the constant technological improvements in the field of detector development, X-ray fluorescence (XRF) in the soft X-ray regime remains a challenge. The low intrinsic fluorescence yield for energies below 2 keV indeed renders the applicability of low-energy XRF still difficult. Here, we report on a new multi-element multi-tile detection system currently under development, designed to be integrated into a soft X-ray microscopy end station. The system will be installed at the TwinMic beamline of Elettra synchrotron (Trieste, Italy) in order to increase the detected count rate by up to an order of magnitude. The new architecture is very versatile and can be adapted to any XRF experimental setup. Even though the first results of the previous version of such a multi-element system were encouraging, several issues still needed to be addressed. The system described here represents a further step in the detector evolution. It is based on four trapezoidal-shaped monolithic silicon drift detector tiles (matrices) with six hexagonal elements each equipped with a custom ultra-low noise application-specific integrated circuit readout. The whole signal processing chain has been improved leading to an overall increase in performances, namely, in terms of energy resolution and acquisition rates. The design and development of this new detection system will be described, and recent results obtained at the TwinMic beamline at Elettra will be presented. Future perspectives and improvements will also be discussed.

Abstract

In 1998, sildenafil was marketed as the first FDA-approved oral drug for the treatment of erectile dysfunction (ED). During the last two decades, the commercialization of other synthetic phosphodiesterase 5 (PDE5) inhibitors has been paralleled by the rise of remedies based on natural molecules from different chemical classes (flavonoids, polyphenols and alkaloids in general). In this work, a set of in silico tools were applied to study a panel of 30 natural compounds claimed to be effective against ED in the scientific literature or in folk medicine. First, pharmacokinetic properties were analysed to exclude the compounds lacking in specific drug-like features. Estimated binding energy for PDE5 and selectivity towards other PDE isoforms were then considered to highlight some promising molecules. Finally, a detailed structural investigation of the interaction pattern with PDE in comparison with sildenafil was conducted for the best performing compound of the set. [ABSTRACT FROM AUTHOR]

Abstract

Developing and deploying software systems for data acquisition and experiment control in a beamline laboratory can be a very challenging task. In certain cases there is the need to replace and modernize an existing system in order to accommodate substantial beamline upgrades. DonkiOrchestra is a TANGO-based framework for data acquisition and experiment control developed at Elettra Sincrotrone Trieste. The framework is based on an advanced software trigger-driven paradigm developed in-house. DonkiOrchestra is meant to be general and flexible enough to be adapted to the development needs of different laboratories and their data acquisition requirements. This presentation outlines the upgrade of the LabVIEW-based TwinMic beamline control system which hosts a unique soft X-ray transmission and emission microscope. Other than the technical demanding tasks of interfacing and controlling old and new instrumentation with DonkiOrchestra, this presentation discusses the various challenges of upgrading the software in a working synchrotron beamline., Proceedings of the 16th Int. Abstract

Abstract Background Occupational or environmental exposure to asbestos fibres is associated with pleural and parenchymal lung diseases. A histopathologic hallmark of exposure to asbestos is the presence in lung parenchyma of the so-called asbestos bodies. They are the final product of biomineralization processes resulting in deposition of endogenous iron and organic matter (mainly proteins) around the inhaled asbestos fibres. For shedding light on the formation mechanisms of asbestos bodies it is of fundamental importance to characterize at the same length scales not only their structural morphology and chemical composition but also to correlate them to the possible alterations in the local composition of the surrounding tissues. Here we report the first correlative morphological and chemical characterization of untreated paraffinated histological lung tissue samples with asbestos bodies by means of soft X-ray imaging and X-Ray Fluorescence (XRF) microscopy, which reveals new features in the elemental lateral distribution. Results The X-ray absorption and phase contrast images and the simultaneously monitored XRF maps of tissue samples have revealed the location, distribution and elemental composition of asbestos bodies and associated nanometric structures. The observed specific morphology and differences in the local Si, Fe, O and Mg content provide distinct fingerprints characteristic for the core asbestos fibre and the ferruginous body. The highest Si content is found in the asbestos fibre, while the shell and ferruginous bodies are characterized by strongly increased content of Mg, Fe and O compared to the adjacent tissue. The XRF and SEM-EDX analyses of the extracted asbestos bodies confirmed an enhanced Mg deposition in the organic asbestos coating. Conclusions The present report demonstrates the potential of the advanced synchrotron-based X-ray imaging and microspectroscopy techniques for studying the response of the lung tissue to the presence of asbestos fibres. The new results obtained by simultaneous structural and chemical analysis of tissue specimen have provided clear evidence that Mg, in addition to Fe, is also involved in the formation mechanisms of asbestos bodies. This is the first important step to further thorough investigations that will shed light on the physiopathological role of Mg in tissue response to the asbestos toxicity.

Abstract

Cannabidiol (CBD) is a non-psychotropic phytocannabinoid which represents one of the constituents of the “phytocomplex” of Cannabis sativa. This natural compound is attracting growing interest since when CBD-based remedies and commercial products were marketed. This review aims to exhaustively address the extractive and analytical approaches that have been developed for the isolation and quantification of CBD. Recent updates on cutting-edge technologies were critically examined in terms of yield, sensitivity, flexibility and performances in general, and are reviewed alongside original representative results. As an add-on to currently available contributions in the literature, the evolution of the novel, efficient synthetic approaches for the preparation of CBD, a procedure which is appealing for the pharmaceutical industry, is also discussed. Moreover, with the increasing interest on the therapeutic potential of CBD and the limited understanding of the undergoing biochemical pathways, the reader will be updated about recent in silico studies on the molecular interactions of CBD towards several different targets attempting to fill this gap. Computational data retrieved from the literature have been integrated with novel in silico experiments, critically discussed to provide a comprehensive and updated overview on the undebatable potential of CBD and its therapeutic profile.

Abstract

Synchrotron-based soft-X-ray imaging and microspectroscopy with sub-micrometer spatial resolution are used to investigate the electrodeposition of Mn-Cu-ZnO, a prospective active material for supercapacitors. This study is focused on the correlation of the local current density and the spatial distribution of the composition and chemical-state in electrodeposits grown potentiostatically at -0.7V (vs a saturated calomel electrode), as well as the optimal potential for the achievement of high specific capacitance and cycling stability. The morphology, elemental distribution, and the local chemical state of both the electrode deposits and the grown dendrite structures are followed by using X-ray imaging, X-ray fluorescence mapping, and X-ray absorption microspectroscopy. For transmission soft-X-ray measurements, a thin-layer Hull-type microcell is developed and fabricated by using electron-beam lithography. The information obtained for the spatial distribution of the deposit is complemented by using electrochemical measurements and numerical simulations.

Abstract

In this paper we report on the electrochemical reconstruction of a Tarentum hemiobolus Ag coin, severely corroded in marine environment. As assessed by conventional analytical tools, most of the initially metallic Ag coin had been converted to AgCl by exposure to the aggressive coastal burial conditions. X-ray computed microtomography proved that only small portions of the artefact had preserved their metallic nature. Since the engraving was preserved partly in the corrosion product bulk and partly in the metallic rests, electrodeposition of Ag from the AgCl layer, under controlled conditions ensuring shape preservation, resulted in the reconstruction of the coin surface with full recovery of the original engraving. Such optimal electrodeposition conditions were identified by a combination of electrochemical and quasi-in situ X-ray microtomography experiments, carried out with artificially corrored engraved Ag wires. Microtomography of the reconstructed coin confirmed the compaction of the external Ag layer and disclosed that the central core of the coin still contains unconverted AgCl. The presence of such a mineralised core does not however impact the numismatic use of the coin and the safeguard of the original engraving.
