166. Why Change Management Fails in Indian Organizations: Learning from Cases.

Abstract

This paper provides evidence of what works and what does not work to make a change initiative successful from five case studies where the authors have worked, during 2009 and 2014, as change agent. The theoretical perspective of our research is interpretivism. We took the constructivists approach while studying the change management phenomenon. Hence, we relied on a qualitative method. We have used narrative inquiry based on memory as we have first-hand knowledge over a period in each case and in some cases, we both have experience in different time periods. What was happening in these organizations has been presented in a story format. From the stories, three broad themes were identified and for each organization theme wise key characteristics have been noted and from this analysis and based on our perception of success and failure of change initiative in these organizations we have presented our findings. Keeping the widely accepted change management frame work of Kotter (1995), and Aiken and Keller (2009) in the perspective, we have found six new points, which are very relevant in an Indian context. Based on the new points, a change management approach has been proposed. The suggested approach is based on SPDCA (Study, Plan, Deploy, Check, and Act) in a change management context. We strongly suggest studying the existing situation at the beginning and the proposed 12 questions for top management to ask in the planning phase. The new points that emerged from these case studies for successful implementation of change management are: Basics are in place; Strong review mechanism must exist; Knowledge on the type of initiative at the Board level must exist; and Small Group Activity is in place and should have the culture of participation in relevant public award competition. To make the article more practitioner oriented we have taken a conscious call of avoiding too much stress on theoretical aspects of change management. [ABSTRACT FROM AUTHOR]

Cells from a 'knock-in' mouse expressing a NF-[sub.K.B] p65 mutant bearing an alanine instead of serine at position 276 (S276A) displays a significant reduction of NF-[sub.K.B] dependent transcription, even though the mutant p65 forms appropriate complexes. It is demonstrated that this variegated phenotype is due to epigenetic repression.

Abstract

In addition to ligation of the T cell antigen receptor (TCR), activation of the CD28 coreceptor by the costimulatory molecule B7 is required for induction of the transcription factor NF-kappaB and robust T cell activation, although the contribution of CD28 to this process remains incompletely understood. We show here that phosphoinositide-dependent kinase 1 (PDK1) is essential for integrating the TCR and CD28 signals. After we deleted PDK1 from T cells, TCR-CD28 signals were unable to induce activation of NF-kappaB or phosphorylation of protein kinase C-theta, although T cell survival and pathways dependent on the kinases p38 and Jnk or the transcription factor NFAT were unaffected. CD28 facilitated NF-kappaB activation by regulating recruitment and phosphorylation of PDK1, which are necessary for efficient binding of PDK1 to protein kinase C-theta and the adaptor CARMA1 and thus for NF-kappaB induction.

Abstract

Asymmetric cell division (ACD) in a perpendicular orientation promotes cell differentiation and organizes the stratified epithelium. However, the upstream cues regulating ACD have not been identified. Here, we report that phosphoinositide-dependent kinase 1 (PDK1) plays a critical role in establishing ACD in the epithelium. Production of phosphatidyl inositol triphosphate (PIP3) is localized to the apical side of basal cells. Asymmetric recruitment of atypical protein kinase C (aPKC) and partitioning defective (PAR) 3 is impaired in PDK1 conditional knockout (CKO) epidermis. PDK1CKO keratinocytes do not undergo calcium-induced activation of aPKC or IGF1-induced activation of AKT and fail to differentiate. PDK1CKO epidermis shows decreased expression of Notch, a downstream effector of ACD, and restoration of Notch rescues defective expression of differentiation-induced Notch targets in vitro. We therefore propose that PDK1 signaling regulates the basal-to-suprabasal switch in developing epidermis by acting as both an activator and organizer of ACD and the Notch-dependent differentiation program. Dainichi et al. demonstrate that PDK1 plays a critical role in asymmetric cell division, wherein PDK1 regulates the activation of AKT, as well as the activation and redistribution of aPKC. PDK1 signaling from the apical side of basal keratinocytes regulates the basal-to-suprabasal switch in epithelial stratification and the Notch-dependent differentiation program.

Abstract

Southeast Asia, especially India, is well known for the highest use of smokeless tobacco. These products are known to induce oral squamous cell carcinoma. However, not all long-term tobacco-chewers develop oral squamous cell carcinoma. In addition, germline variants play a crucial role in susceptibility, prognosis, development, and progression of the disease. These prompted us to study the genetic susceptibility to oral squamous cell carcinoma among the long-term tobacco-chewers. Here, we presented a retrospective study on prolonged tobacco-chewers of Northeast India to identify the potential protective or risk-associated germline variants in tobacco-related oral squamous cell carcinoma along with HPV infection. Targeted re-sequencing (n = 60) of 170 genetic regions from 75 genes was carried out in Ion-PGM- and validation (n = 116) of the observed variants was done using Sequenom iPLEX MassARRAY- platform followed by polymerase chain reaction-based HPV genotyping and p16-immunohistochemistry study. Subsequently, estimation of population structure, different statistical and in silico approaches were undertaken. We identified one nonsensemediated mRNA decay transcript variant in the DFNA5 region (rs2237306), associated with Benzo(a)pyrene, as a protective factor (odds ratio = 0.33; p = 0.009) and four harmful (odds ratio.2.5; p\0.05) intronic variants, rs182361, rs290974, and rs169724 in SYK and rs1670661 in NELL1 region, involved in genetic susceptibility to tobacco- and HPVmediated oral oncogenesis. Among the oral squamous cell carcinoma patients, 12.6% (11/87) were HPV positive, out of which 45.5% (5/11) were HPV16-infected, 27.3% (3/11) were HPV18-infected, and 27.3% (3/11) had an infection of both subtypes. Multifactor dimensionality reduction analysis showed that the interactions among HPV and NELL1 variant rs1670661 with age and gender augmented the risk of both non-tobacco- and tobacco-related oral squamous cell carcinoma, respectively. These suggest that HPV infection may be one of the important risk factors for oral squamous cell carcinoma in this population. Finally, we newly report a DFNA5 variant probably conferring protection via nonsensemediated mRNA decay pathway against tobacco-related oral squamous cell carcinoma. Thus, the analytical approach used here can be useful in predicting the population-specific significant variants associated with oral squamous cell carcinoma in any heterogeneous population.
