The PANDA spectrometer will be built at the FAIR facility at Darmstadt (Germany) to perform accurate tests of the strong interaction through ¯pp and ¯pA annihilation studies. The charged particle tracking at PANDA will be done using both solid state and gaseous detectors. Among the latter, two straw tube detector systems will be built [1]. The cylindrical, central straw tube tracker features a high spatial and momentum resolution for a wide range of particle momenta from about 8 GeV/c down to a few 100 MeV/c, together with particle identification in the momentum region below about 1 GeV/c by measuring the specific energy-loss. A new technique, based on self-supporting straw double layers with intrinsic wire tension developed for the COSY-TOF straw tracker [2], has been adopted for the PANDA trackers. The development of the readout electronics for the straw tubes is ongoing. Prototypes have been produced and used to instrument straw tube modules that have been tested with cosmic rays and proton beams. Design issues of the PANDA straw tubes, together with the results of the prototype tests are presented.

Deficits in memory performance have been linked to a wide range of neurological and neuropsychiatric conditions. While many studies have assessed the memory impacts of individual conditions, this study considers a broader perspective by evaluating how memory recall is differentially associated with nine common neuropsychiatric conditions using data drawn from 55 international studies, aggregating 15,883 unique participants aged 15–90. The effects of dementia, mild cognitive impairment, Parkinson’s disease, traumatic brain injury, stroke, depression, attention-deficit/hyperactivity disorder (ADHD), schizophrenia, and bipolar disorder on immediate, short-, and long-delay verbal learning and memory (VLM) scores were estimated relative to matched healthy individuals. Random forest models identified age, years of education, and site as important VLM covariates. A Bayesian harmonization approach was used to isolate and remove site effects. Regression estimated the adjusted association of each clinical group with VLM scores. Memory deficits were strongly associated with dementia and schizophrenia (p < 0.001), while neither depression nor ADHD showed consistent associations with VLM scores (p > 0.05). Differences associated with clinical conditions were larger for longer delayed recall duration items. By comparing VLM across clinical conditions, this study provides a foundation for enhanced diagnostic precision and offers new insights into disease management of comorbid disorders.

Background: Congenital absence of pericardium is a rare cardiac disorder with a reported incidence of less than 1 in 10,000. Although most of the cases are of little clinical significance, some of them are associated with serious complications, including risk of herniation and strangulation or coronary artery compression. Detailed Case Description: We report a case of a 36-year-old male referred for routine cardiovascular examination. He had a medical history of a heart murmur since childhood. Electrocardiogram (ECG) revealed sinus rhythm, normal axis, poor R-wave progression in the precordial leads and repolarization abnormalities with negative T waves in leads V1–V4. On 2D transthoracic echocardiography (TTE), an unusual heart position was noted with poor image quality from the standard acoustic windows. The parasternal long axis view gave the impression of right ventricular dilatation. The findings raised the suspicion of left to right shunt and possible atrial septal defect. For further evaluation, the patient was referred for cardiac magnetic resonance which demonstrated complete left-sided absence of the pericardium. Discussion: Due to indistinct and atypical symptoms and lack of clinical awareness, pericardial congenital absence is frequently misdiagnosed. Patients may complain of atypical chest pain. Patient’s history and physical examination are often nonspecific. In cases with complete pericardial absence, ECG findings may include right axis deviation, right bundle block and sinus bradycardia. Echocardiography findings are also not characteristic, but some may raise the clinical suspicion of this diagnosis. The imaging modalities of choice are computed tomography and cardiac magnetic resonance. Treatment depends on the type of defect and clinical symptoms.

The primary population of small ruminants in Spain is concentrated in the southern region, a critical area for the country’s livestock production. Indirect economic losses can occur when this livestock is affected by gastrointestinal parasites. This study aimed to determine the prevalence of these parasites in small ruminant herds (159 sheep and 39 goats) through coprological analyses and conducted a survey on farmers’ management practices related to gastrointestinal parasite control. The survey results revealed some important aspects: monitoring through coprological analyses is not a common practice; veterinarians are not typically involved in deworming plans; anthelmintic treatment in adults is often applied twice a year in sheep and once a year in goats; and finally, drug rotation was higher in sheep farms. Coprological analyses showed Eimeria spp. as the most common parasitic infection, followed by Strongyles infection. Other parasites like Moniezia spp., Trichuris spp., and D. dendriticum were less important, although their prevalence was higher in sheep than goats. This constitutes the first report on the epidemiological status of gastrointestinal parasites in small ruminants in southern Spain. Based on the survey findings, the introduction of certain management measures on farms could potentially mitigate parasite infections.

The review focuses on the recent knowledge on natural anthraquinones (AQs) of plant origin and their potential for application in an exclusive medicinal curative and palliative method named photodynamic therapy (PDT). Green approach to PDT is associated with photosensitizers (PS) from plants or other natural sources and excitation light in visible spectrum. The investigations of plants are of high research interests due to their unique health supportive properties as herbs and the high percentage availability to obtain compounds with medical value. Up-to-date many naturally occurring compounds with therapeutic properties are known and are still under investigations. Some natural quinones have already been evaluated and clinically approved as anti-tumor agents. Recent scientific interests are beyond their common medical applications but also in directions to their photo-properties as natural PSs. The study presents a systematic searches on the latest knowledge on AQ derivatives that are isolated from the higher plants as photosensitizers for PDT applications. The natural quinones have been recognized with functions of natural dyes since the ancient times. Lately, AQs have been explored due to their biological activity including the photosensitive properties useful for PDT especially towards medical problems with no other alternatives. The existing literature’ overview suggests that natural AQs possess characteristics of valuable PSs for PDT. This method is based on an application of a photoactive compound and light arrangement in oxygen media, such that the harmful general cytotoxicity could be avoided. Moreover, the common anticancer and antimicrobial drug resistance has been evaluated with very low occurrence after PDT. Natural AQs have been focused the scientific efforts to further developments because of the high range of natural sources, desirable biocompatibility, low toxicity, minimal side effects and low accident of drug resistance, together with their good photosensitivity and therapeutic capacity. Among the known AQs, only hypericin has been studied in anticancer clinical PDT. Currently, the natural PSs are under intensive research for the future PDT applications for diseases without alternative effective treatments.

Wearable health devices (WHDs) are rapidly gaining ground in the biomedical field due to their ability to monitor the individual physiological state in everyday life scenarios, while providing a comfortable wear experience. This study introduces a novel wearable biomedical device capable of synchronously acquiring electrocardiographic (ECG), photoplethysmographic (PPG), galvanic skin response (GSR) and motion signals. The device has been specifically designed to be worn on a finger, enabling the acquisition of all biosignals directly on the fingertips, offering the significant advantage of being very comfortable and easy to be employed by the users. The simultaneous acquisition of different biosignals allows the extraction of important physiological indices, such as heart rate (HR) and its variability (HRV), pulse arrival time (PAT), GSR level, blood oxygenation level (SpO2), and respiratory rate, as well as motion detection, enabling the assessment of physiological states, together with the detection of potential physical and mental stress conditions. Preliminary measurements have been conducted on healthy subjects using a measurement protocol consisting of resting states (i.e., SUPINE and SIT) alternated with physiological stress conditions (i.e., STAND and WALK). Statistical analyses have been carried out among the distributions of the physiological indices extracted in time, frequency, and information domains, evaluated under different physiological conditions. The results of our analyses demonstrate the capability of the device to detect changes between rest and stress conditions, thereby encouraging its use for assessing individuals’ physiological state. Furthermore, the possibility of performing synchronous acquisitions of PPG and ECG signals has allowed us to compare HRV and pulse rate variability (PRV) indices, so as to corroborate the reliability of PRV analysis under stationary physical conditions. Finally, the study confirms the already known limitations of wearable devices during physical activities, suggesting the use of algorithms for motion artifact correction.

Abstract Background Recent advances in CRISPR technology have enabled us to perform gene knock-in in various species and cell lines. CRISPR-mediated knock-in requires donor DNA which serves as a template for homology-directed repair (HDR). For knock-in of short sequences or base substitutions, ssDNA donors are frequently used among various other forms of HDR donors, such as linear dsDNA. However, partly due to the complexity of long ssDNA preparation, it remains unclear whether ssDNA is the optimal type of HDR donors for insertion of long transgenes such as fluorescent reporters in human cells. Results In this study, we established a nuclease-based simple method for the preparation of long ssDNA with high yield and purity, and comprehensively compared the performance of ssDNA and dsDNA donors with 90 bases of homology arms for endogenous gene tagging with long transgenes in human diploid RPE1 and HCT116 cells. Quantification using flow cytometry revealed lower efficiency of endogenous fluorescent tagging with ssDNA donors than with dsDNA. By analyzing knock-in outcomes using long-read amplicon sequencing and a classification framework, a variety of mis-integration events were detected regardless of the donor type. Importantly, the ratio of precise insertion was lower with ssDNA donors than with dsDNA. Moreover, in off-target integration analyses using donors without homology arms, ssDNA and dsDNA were comparably prone to non-homologous integration. Conclusions These results indicate that ssDNA is not superior to dsDNA as long HDR donors with relatively short homology arms for gene knock-in in human RPE1 and HCT116 cells.

We present two case reports of cardiac echinococcosis. Case 1 was a 33-year-old woman with hepatic and cardiac echinococcosis. The parasitic cyst was located intramyocardially in the free wall of the left ventricle leading to cranial dislocation of the left circumflex coronary artery (LCx). The patient was successfully operated. Case 2 was a 28-year-old woman with hepatic and cardiac echinococcosis. The parasitic cyst was located in the left ventricular myocardium in the area of the apex and manifested clinically as paroxysms of ventricular tachycardia. The ultrasound study showed a 3.2×2.8 cm cyst dislocating the papillary muscles and causing moderate mitral regurgitation.Bulgaria ranks first in the European Union in terms of the number of echinococcosis patients. Although cardiac involvement is uncommon, occurring in only 0.5%–2% of cases, it can cause a wide range of clinical symptoms. Multimodal imaging is a key step in the management of patients with cardiac involvement.

Outcome measures: uncorrected and best-corrected distance visual acuity, subjective refractive astigmatism, keratometric astigmatism, spherical equivalent, minimum corneal thickness, and corneal morphological irregularity index. Subjects were followed for 12 months post-treatment.Results: Eighteen eyes of 18 patients aged between 32 and 75 years treated with topography-guided transepithelial PRK with iRes excimer laser (iVIS Technologies, Taranto, Italy) from June 2020 to July 2021. After 12 months, the mean UDVA improved from 1.0± 0.00LogMAR pre-op to 0.15± 0.154LogMAR, and the mean CDVA improved from 0.4± 0.41LogMAR pre-op to 0.0± 0.00LogMAR. With respect to UDVA, all treated eyes (100%) showed an improvement of 6 ETDRS lines or more and with respect to CDVA, 9 out of 18 eyes (50%) showed an improvement of 6 ETDRS lines or more. The mean ablation depth was 54.7± 5.9μm. A statistically significant improvement was observed in all topographic indices. No infiltrate recurrence, post-treatment corneal haze, ocular hypertension or other side effects were observed throughout the follow-up period.Conclusion: Topography-guided PRK could be considered an effective and safe treatment option to improve visual acuity in patients affected by persistent and visually-significant subepithelial corneal infiltrates caused by EKC.Keywords: adenoviral epidemic keratoconjunctivitis, subepithelial corneal opacities, topography-guided transepithelial PRK

O processo de financeirização associado às tecnologias da informação tem impulsionado o avanço mercadológico nos diferentes setores da sociedade. A mobilidade urbana é um dos setores citadinos que mais tem sido impactado. Este trabalho objetiva discutir o conceito de micromobilidade urbana e o papel das instituições financeiras atuantes nesse setor na cidade de São Paulo (SP), com especial ênfase no caso da empresa Tembici (do Banco Itaú). A metodologia adotada neste estudo inclui revisão bibliográfica de produções científicas nacionais e internacionais, dados primários e secundários, qualitativos e quantitativos, obtidos através de trabalho de campo e relatórios. Buscamos interpretar os impactos territoriais das plataformas digitais discutidas, examinando diferentes usos do território e analisando a forma como essa empresa se insere no processo de expansão dos serviços de mobilidade. Os resultados revelam como o espaço urbano, impulsionado pelo uso da tecnologia por entidades financeiras, torna-se mercado lucrativo, afetando diretamente a mobilidade urbana.

We determined the impacts of light intensity, blood fraction, fasting and storage time on glucose, cholesterol, triglycerides, aspartate aminotransferase (AST), alanine aminotransferase (ALT), and gamma-glutamyl transferase (GGT) concentrations in 140 broiler chickens assigned to two light intensities (5 or 20 lux/m²) and seven fasting times (0, 2, 4, 6, 8, 10, and 12 h). The collected blood was fractionated into serum or plasma and stored at -20°C for 0, 15, 30, 60, and 120 days. Serum glucose concentrations declined linearly by 1.15 mg dL-1 every 2 h of fasting, whereas plasma glucose responded quadratically (minimum point at 8 h and 13 min of fasting). Serum ALT concentrations showed a quadratic effect, with a maximum point at 3 h and 46 min of fasting. Serum GGT concentrations were not influenced by fasting time, whereas plasma decreased by 1.60 IU L-1 at each 2 h of fasting. The AST concentrations were influenced by fasting. Storage times influenced the glucose concentrations in a quadratic manner, with maximum points in serum and plasma at 95 and 72 days, respectively. Serum cholesterol and triglyceride concentrations increased linearly with storage time. Storage time caused a quadratic response on serum ALT, with a maximum point at 32 days, whereas there was a minimum point at 93 days in plasma. Concentrations of GGT increased linearly with storage. Glucose, triglycerides, AST, and ALT concentrations were highest at 20 lux. Samples for AST analysis can be stored up to 120 days at -20°C. Serum glucose, cholesterol, triglyceride, ALT, and GGT analysis is indicated with a 6 h fasting time, without freezing. The light intensity should be specified.

More than a year after the Moscow government's invasion of Ukraine, it seemed necessary to initiate a study to investigate how the New Arrivals in Italy (NAI) reception practices were implemented by Italian schools in the school year 2021/2022, following the emergency. The definition of the research theoretical framework focus on the concepts of emergency and interculture, intended as continuous processes of re-adaptation and re-organization. The data highlight the strengths and weaknesses found by the school in the reception process. The data collected about the effectiveness of the actions carried out by the school relates to what is desired and prescribed by ministerial documents. It emerges the need for a pedagogical system for the management of reception in emergency, in a circular perspective that is reflected in the theory-practice-theory nexus. L’accoglienza dei Neo Arrivati in Italia in fuga dalla guerra. Uno studio sulle pratiche adottate dalle scuole. A più di un anno dall’invasione dell’Ucraina da parte del governo di Mosca, si è sentita la necessità di avviare uno studio per indagare quali sono state le pratiche di accoglienza dei New Arrivals in Italy (NAI) messe in atto dalle scuole italiane nell’a. s. 2021/2022 a seguito dell’emergenza. La definizione del quadro teorico della ricerca ruota intorno ai concetti di emergenza e intercultura intesi come processi continui di ri-adattamento e ri-organizzazione. I dati mettono in luce i punti di forza e le criticità riscontrate dalla scuola nel processo di accoglienza. Si mettono in relazione i dati raccolti in merito all’efficacia delle azioni svolte dalla scuola con quanto auspicato e prescritto dai documenti ministeriali. Emerge la necessità di un dispositivo pedagogico per la gestione dell’accoglienza in emergenza, in un’ottica circolare che trovi riscontro nel nesso teoria-prassi-teoria.

Toxoplasmosis is a common protozoan infection that can have severe outcomes in the immunocompromised and during pregnancy, but treatment options are limited. Recently, nucleotide metabolism has received much attention as a target for new antiprotozoal agents and here we focus on pyrimidine salvage by Toxoplasma gondii as a drug target. Whereas uptake of [3H]-cytidine and particularly [3H]-thymidine was at most marginal, [3H]-uracil and [3H]-uridine were readily taken up. Kinetic analysis of uridine uptake was consistent with a single transporter with a Km of 3.3 ± 0.8 µM, which was inhibited by uracil with high affinity (Ki = 1.15 ± 0.07 µM) but not by thymidine or 5-methyluridine, showing that the 5-Me group is incompatible with uptake by T. gondii. Conversely, [3H]-uracil transport displayed a Km of 2.05 ± 0.40 µM, not significantly different from the uracil Ki on uridine transport, and was inhibited by uridine with a Ki of 2.44 ± 0.59 µM, also not significantly different from the experimental uridine Km. The reciprocal, complete inhibition, displaying Hill slopes of approximately -1, strongly suggest that uridine and uracil share a single transporter with similarly high affinity for both, and we designate it uridine/uracil transporter 1 (TgUUT1). While TgUUT1 excludes 5-methyl substitutions, the smaller 5F substitution was tolerated, as 5F-uracil inhibited uptake of [3H]-uracil with a Ki of 6.80 ± 2.12 µM (P > 0.05 compared to uracil Km). Indeed, we found that 5F-Uridine, 5F-uracil and 5F,2’-deoxyuridine were all potent antimetabolites against T. gondii with EC50 values well below that of the current first line treatment, sulfadiazine. In vivo evaluation also showed that 5F-uracil and 5F,2’-deoxyuridine were similarly effective as sulfadiazine against acute toxoplasmosis. Our preliminary conclusion is that TgUUT1 mediates potential new anti-toxoplasmosis drugs with activity superior to the current treatment.

An experiment was performed in the DIII-D lower divertor to validate numerical SOL tungsten (W) impurity erosion and redeposition simulations against experimental data. The net and gross erosion of W were calculated as a function of the voltage (or bias) applied to the exposed material. Five samples were inserted into the DIII-D lower divertor using the Divertor Material Evaluation System (DiMES) manipulator and exposed to constant L-mode attached plasma conditions. Each sample was partially coated with W. During plasma shots, samples were biased with respect to the machine vessel ground, ranging from −60 V to 25 V. The ERO2.0 code was used to numerically simulate the experiment aiming to compare the numerical results with experimental measures. A good agreement is found between estimated and measured tungsten erosion at least for negative biases.

Abstract Studies and innovations on alternative feed additives, especially on homeopathic remedies have been highlighted in order to replace or reduce the use of antibiotics in pig production. This paper aimed to assess the addition of homeopathic products in pig diet and their effects on the growth performance, serum metabolites, nutrient and energy digestibility, carcass traits and meat quality. A total of 60 immunocastrated male pigs, weighing on average 30.91 ± 0.95 kg, were distributed in two treatments, 10 replicates and three animals/experimental unit. There was no effect (P≥0.05) of treatment on the growth performance and serum metabolites. The percentage of acid-insoluble ash recovered in the diet was greater (P≤0.01) in diets containing homeopathic products. The apparent digestible energy of diets containing homeopathic products was reduced (P≤0.01) in the growing phase and reduced (P≤0.01) the apparent digestibility coefficients of dry matter, crude protein, soluble neutral and acid detergent fibers, and gross energy in the growing and finishing phases. Pig that received diets with homeopathic products had higher (P≤0.05) amount of meat, percentage of meat and marbling. The use of homeopathic products in diets improves the percentage and quality of meat, as well as the marbling of the pig carcass, maintaining the performance.

AbstractMumps is an acute, contagious, viral vaccine-preventable disease caused by mumps virus (MuV). The measles, mumps, rubella (MMR) vaccine that is used in many countries is considered highly effective with decreased MuV incidence, but suboptimal MuV long-term immunity. This study assessed the MuV seropositivity and antibody titre among vaccinated children in Bulgaria to provide evidence for a better understanding of MuV circulation and immunity in Bulgaria. The samples from 734 immunized children (369 females and 365 males) aged 1 to 16, divided into four age groups (≥1, 2–6, 7–11, and 12–16) were tested. Qualitative and quantitative indirect enzyme-linked immunosorbent assay (Anti-Mumps IgG ELISA, Euroimmun, Germany) was performed to determine the mumps IgG antibody levels in sera. Among all participants, protective MuV immunity was identified in 93%. MuV IgG antibody positivity ranged between 87% in the age group between 1 and 2 years and 96% in the age group 12–16 years, but no statistically significant difference was found among age groups. At the same time, there was a statistically significant difference (p = 0.01) between seropositivity in male and female participants, with male participants having an overall seropositivity of 90% and female 95%. Among the antibody-positive samples, the quantitative measurements of median anti-MuV IgG concentrations showed that titres decreased with increasing age. A slightly waning immunity following vaccination was observed, but positivity remained high among vaccinated children over the years. Similar studies show that maintaining high immunity is crucial to prevent mumps outbreaks.

Avian influenza viruses (AIV) have been classified on the basis of 16 subtypes of hemagglutinin (HA) and 9 subtypes of neuraminidase. Here we describe genomic evidence for a new candidate HA subtype, nominally H19, with a large genetic distance to all previously described AIV subtypes, derived from a cloacal swab sample of a Common Pochard (Aythya ferina) in Kazakhstan, in 2008. Avian influenza monitoring in wild birds especially in migratory hotspots such as central Asia is an important approach to gain information about the circulation of known and novel influenza viruses. Genetically, the novel HA coding sequence exhibits only 68.2% nucleotide and 68.5% amino acid identity with its nearest relation in the H9 (N2) subtype. The new HA sequence should be considered in current genomic diagnostic AI assays to facilitate its detection and eventual isolation enabling further study and antigenic classification.

This study investigated the effect of crude protein (CP) and non-essential amino acid (NEAA) supplementation on the growth performance, blood profile, intestinal morphology, mRNA relative abundance of inflammatory and antioxidant markers, and tight junction proteins in piglets over the first 2 weeks after weaning. Ninety 21-day-old piglets (7.55 ± 0.72 kg) were assigned in a randomized block design to one of three dietary treatments: (1) high CP, a diet with 24% CP; (2) low CP, a diet with 18% CP; and (3) low CP + NEAA, a diet with 18% CP supplemented with 5 g/kg Arg (L-arginine; purity >99%) and 10 g/kg Glu + Gln (minimum 10% L-glutamine and minimum 10% L-glutamate). ABSTRACT The objective of this study was to examine the effects of replacement of an antibiotic (ANT, chlorohydroxyquinoline) in the diet of weaned piglets with a prebiotic composed of β-glucans, glucomannans, and mannan-oligosaccharides on growth performance, diarrhea occurrence (DO), hematological parameters, pH of digestive content, organ weights, intestinal epithelium morphology, and intestinal bacterial counts. A total of 120 piglets (weaned at 21 d old; 6.32±0.10 kg BW) were allocated in a randomized block design, with six pens replicates and four piglets per pen as experimental unit. Treatments consisted of diets with 0, 1,000, 2,000, or 3,000 mg kg−1 of prebiotic and a basal diet with 120 mg kg−1 ANT. No differences of treatments were observed on performance, DO, pH of digestive content, organ weights, and hematological parameters. Duodenal villus density (VD) increased linearly with the prebiotic levels. In the jejunum, the prebiotic level of 2,000 mg kg−1 resulted in a greater VD compared with the ANT. Enterobacteriaceae count in the ileum and total and lactic acid bacteria counts in the cecum responded quadratically to increasing prebiotic levels. The equations estimated the prebiotic levels of 2,125, 1,167, and 1,500 mg kg−1 to provide reduced counts of the respective bacteria (ileal enterobacteria: 6.47 log CFU g−1; total cecal bacteria: 6.70 log CFU g−1; and cecal lactic acid: 7.29 log CFU g−1). Salmonella spp. was not detected, whereas Escherichia coli and Lactobacillus spp. gene copies were unaffected by the treatments. Prebiotic levels or ANT do not enhance performance or health of weanling pigs, but the effects observed on VD and microbiota profile might be an indication of the potential benefits of these products in commercial farming conditions.

However, the availability of psychometrically-evaluated assessments of job interviewing skills is limited for autism services providers and researchers.ObjectiveWe analyzed the psychometric properties of the Mock Interview Rating Scale that was adapted for research with autistic transition-age youth (A-MIRS; a comprehensive assessment of video-recorded job interview role-play scenarios using anchor-based ratings for 14 scripted job scenarios).MethodsEighty-five transition-age youth with autism completed one of two randomized controlled trials to test the effectiveness of two interventions focused on job interview skills. All participants completed a single job interview role-play at pre-test that was scored by raters using the A-MIRS. We analyzed the structure of the A-MIRS using classical test theory, which involved conducting both exploratory and confirmatory factor analyzes, Rasch model analysis and calibration techniques. We then assessed internal consistency, inter-rater reliability, and test–retest reliability. Pearson correlations were used to assess the A-MIRS’ construct, convergent, divergent, criterion, and predictive validities by comparing it to demographic, clinical, cognitive, work history measures, and employment outcomes.ResultsResults revealed an 11-item unidimensional construct with strong internal consistency, inter-rater reliability, and test–retest reliability. Background: Lung cancer patients diagnosed following emergency admission often present with advanced disease and poor performance status, leading to suboptimal treatment options and outcomes. This study aimed to investigate the clinical and molecular characteristics, treatment initiation, and survival outcomes of these patients. Methods: We retrospectively analyzed data from 124 patients diagnosed with lung cancer following emergency admission at a single institution. Clinical characteristics, results of molecular analyses for therapeutic purpose, systemic treatment initiation, and survival outcomes were assessed. Correlations between patients’ characteristics and treatment initiation were analyzed. Results: Median age at admission was 73 years, and 79.0 % had at least one comorbidity. Most patients (87.1 %) were admitted due to cancer-related symptoms. Molecular analyses were performed in 89.5 % of advanced non-small cell lung cancer (NSCLC) cases. In this subgroup, two-thirds (66.2 %) received first-line therapy. Median overall survival (OS) was 3.9 months for the entire cohort, and 2.9 months for patients with metastatic lung cancer. Among patients with advanced NSCLC, OS was significantly longer for those with actionable oncogenic drivers and those who received first-line therapy. Improvement of performance status during hospitalization resulted in increased probability of receiving first-line systemic therapy. Discussion: Patients diagnosed with lung cancer following emergency admission demonstrated poor survival outcomes. Treatment initiation, particularly for patients with actionable oncogenic drivers, was associated with longer OS. These findings highlight the need for proactive medical approaches, including improving access to molecular diagnostics and targeted treatments, to optimize outcomes in this patient population.

IntroductionAcute kidney injury (AKI) in cancer patients receiving immune checkpoint inhibitors (ICIs) may recognize multiple causes. Here, we reviewed cases of biopsy-proven acute tubulointerstitial nephritis (ATIN) to describe the clinical characteristics and outcomes of this condition.MethodWe conducted a pooled analysis of clinical cases of ICI-related biopsy-proven ATIN up to 1 May 2022. We collected data on clinical characteristics, AKI, biopsy findings, laboratory examinations, and renal outcomes.ResultsEighty-five patients (61.4 ± 19 years, 56 male) were evaluated. Melanoma was the most prevalent diagnosis (51%), followed by non-small cell lung cancer (30%). ICI treatment consisted of PD-1, PDL-1 (nivolumab, pembrolizumab, atezolizumab), and CTLA-4 inhibitors (i) (ipilimumab) or combination PD-1i+CTLA4i. Renal toxicity developed after a median of four cycles of therapy. Fifty-one patients (65.5%) developed the most severe form of AKI- stage 3, including five patients requiring dialysis. The amyloidoses constitute a group of diseases occurring in humans and animals that are characterized by abnormal deposits of aggregated proteins in organs, affecting their structure and function. In the Abyssinian cat breed, a familial form of renal amyloidosis has been described. In this study, multi-omics analyses were applied and integrated to explore some aspects of the unknown pathogenetic processes in cats. Whole-genome sequences of two affected Abyssinians and 195 controls of other breeds (part of the 99 Lives initiative) were screened to prioritize potential disease-associated variants. Proteome and miRNAome from formalin-fixed paraffin-embedded kidney specimens of fully necropsied Abyssinian cats, three affected and three non-amyloidosis-affected were characterized. While the trigger of the disorder remains unclear, overall, (i) 35,960 genomic variants were detected; (ii) 215 and 56 proteins were identified as exclusive or overexpressed in the affected and control kidneys, respectively; (iii) 60 miRNAs were differentially expressed, 20 of which are newly described. With omics data integration, the general conclusions are: (i) the familial amyloid renal form in Abyssinians is not a simple monogenic trait; (ii) amyloid deposition is not triggered by mutated amyloidogenic proteins but is a mix of proteins codified by wild-type genes; (iii) the form is biochemically classifiable as AA amyloidosis.

The global burden of mortality and morbidity caused by traumatic brain injury (TBI) is significant, and the heterogeneity of TBI patients and the relatively small sample sizes of most current neuroimaging studies is a major challenge for scientific advances and clinical translation. The ENIGMA (Enhancing NeuroImaging Genetics through Meta-Analysis) Adult moderate/severe TBI (AMS-TBI) working group aims to be a driving force for new discoveries in AMS-TBI by providing researchers world-wide with an effective framework and platform for large-scale cross-border collaboration and data sharing. Based on the principles of transparency, rigor, reproducibility and collaboration, we will facilitate the development and dissemination of multiscale and big data analysis pipelines for harmonized analyses in AMS-TBI using structural and functional neuroimaging in combination with non-imaging biomarkers, genetics, as well as clinical and behavioral measures. Ultimately, we will offer investigators an unprecedented opportunity to test important hypotheses about recovery and morbidity in AMS-TBI by taking advantage of our robust methods for large-scale neuroimaging data analysis. In this consensus statement we outline the working group's short-term, intermediate, and long-term goals.
