As sports technology has continued to develop, monitoring athlete workloads, performance, and recovery has demonstrated boundless benefits for athlete and team success. Specifically, technologies such as global positioning systems (GPS) and heart rate (HR) monitors have granted the opportunity to delve deeper into performance contributors, and how variations may exist based upon context. A team of NCAA Division I women’s soccer athletes were monitored during games throughout one competitive season. Individual athlete, positional groups, and team external and internal workloads were explored for differences based upon game location, opponent ranking, game result, and the final score differential. Game location and opponent ranking were found to have no effect on team-wide absolute or relative external workloads, whereas game result and score differential did. Internal workloads across the team tended to only vary by game half, independent of game context; however, the HR of defenders was determined to be higher during losses as compared to wins (p = 0.0256). Notably, the games that resulted in losses also represented the games with the fewest number of substitutions. These findings suggest high value in monitoring performance and workloads that are characteristic of varying, often multifaceted, contexts. It is hoped that this information can lead to more informed approaches to vital game-time and coaching decisions.

Obtaining accurate information is critical for youth's sexual and reproductive health (SRH). Youth not in foster care often learn about SRH from their biological parents. Separated from their biological parents, youth in care depend on healthcare providers and caregivers for SRH information. However, they often receive insufficient information and feel unsupported in meeting their needs for SRH information. This study explored female African American adolescents in foster care's perspectives on effective SRH communication with caregivers to help them avoid sexual risks. A qualitative study was conducted using semi-structured interviews. We used purposive sampling to recruit 16 adolescents aged 18 to 20 years old with a history of foster care placement. The transcribed interviews underwent inductive thematic analysis. The Positive Youth Development theory underpinned this research. Two prominent themes emerged: establishing a relationship and preferred communication approach. Youth reported that for caregivers to engage in effective SRH communication, they must first establish a relationship by being aware of the youth's childhood trauma, building trust, having patience, and being vulnerable. Youth also appreciated caregivers who ensured comfortability and were honest and straightforward. Caregivers should be trained on adverse childhood experiences, trauma-informed approaches, SRH knowledge, and communication. Healthcare providers should make use of the time spent with youth and discuss SRH topics during clinical encounters. This time spent with youth may be their only chance to obtain accurate SRH information. Youth's perspectives regarding communication about SRH should be implemented in future SRH communication interventions. • Young women in foster care desire sexual health communication with caregivers. • Caregivers are often ill-equipped to engage in effective communication with youth. • Foster care agencies should provide consistent sexual health education training. • Awareness of trauma and its effect on youth should be taught to caregivers. • Healthcare providers should consistently discuss sexual health with youth. The gender gap in entrepreneurship has been observed for a long time, explained by both female-specific and gender-neutral factors, but none of these explanations is generally accepted. The aim of the paper is to assess the effect of internal economic dynamics on female entrepreneurship. Economic dynamics is a persistent process affected simultaneously by both endogenous and exogenous factors of a different time horizon, with the development trend and the business cycle as the most important time perspectives. The decomposition procedure of time series is implemented to extract trend and cyclical fluctuations, after which the Vector Error Correction Model (VECM) method is used to estimate models showing the impact of economic dynamics on female entrepreneurship in the long- and medium-run. The study concerns the countries of the Visegrad Group, including Czechia, Hungary, Poland, and Slovakia, and is based on quarterly data from the years 1998 to 2020. The results show that, although the economic dynamics impact female entrepreneurship, to some extent, it is not the most dominant factor. The impact of economic dynamics on female entrepreneurship is much stronger in the trend perspective than in the business cycle perspective. The nature of the effect of economic dynamics on female entrepreneurship is also country-specific.

In order to obtain the full impedance characteristics of a lithium-ion capacitor as a function of temperature, the authors proposed the use of dynamic electrochemical impedance spectroscopy. Impedance tests were carried out under wide range of dynamic temperature changes for lithium-ion supercapacitors. Significant differences in electrochemical processes were observed as a result of working temperature. Moreover, the quality of fitting of the equivalent circuits most frequently used in impedance analysis of lithium-ion capacitors was discussed. The proposed methodology allows for a comprehensive characterization of the performance of these devices and provides key information for their optimization in wide range of operations.

Entrepreneurship is believed to be shaped by institutions; however, the paper assumes that trust in institutions is a fundamental prerequisite for the impact of institutions on entrepreneurship. The aims of the paper are to determine (i) whether trust in European institutions affects the level of entrepreneurship in European Union countries, and (ii) whether there are any differences in this impact regarding the types of institutions and (iii) a country’s government budget size. Based on yearly panel data for 27 European Union countries in the years 2004–2019 and estimations of panel regression models, the results show that confidence in institutions is a significant factor in explaining entrepreneurship. However, institutional trust has no homogeneous effect on entrepreneurship, as the impact depends on the kind of institutions related to their functions and values as well as on the country’s characteristics in relation to the size of the government’s budget. Practical implications suggest the possibilities of supporting entrepreneurship—especially in countries with a relatively lower public redistribution—by raising the level of confidence in the European Central Bank. The originality of the paper is related to distinguishing institutional trust based on the type of institutions and the country’s characteristics of governmental budget size.

Water management is a key factor affecting the efficiency of proton exchange membrane fuel cells (PEMFCs). The currently used monitoring methods of PEMFCs provide limited information about which processes or components that humidity has a significant impact upon. Herein, we propose the use of a novel approach of impedance measurements using a multi-sinusoidal perturbation signal, which enables impedance measurements under dynamic operating conditions. The manuscript presents the effect of the relative humidity (RH) of the reactants on the instantaneous impedance of the middle cell in the PEMFC stack as a function of the current load. Analysis of changes in the values of equivalent circuit elements was carried out to determine which process determines the stack’s performance depending on the load range of the fuel cell during operation. Comprehensive impedance analysis showed that to ensure optimal cell operation, the humidity of the reactants should be adjusted depending on the load level. The results showed that at low-current loads, the humidity of gases should be at least 50%, while at high-current loads, the cell should operate optimally at a gas humidity of 30% or lower. The presented methodology provides an important tool for optimizing and monitoring the operation of fuel cells.

Utilization of natural shale formations for the creation of annular barriers in oil and gas wells is currently discussed as a mean of simplifying cumbersome plugging and abandonment procedures. Shales that are likely to form annular barriers are shales with high content of swelling clays and relatively low content of cementation material (e.g., quartz, carbonates). Shales with large content of quartz and low content of swelling clays will be rather brittle and not easily deformable. In this paper we ask the question whether and to what extent it is possible to modify the mechanical properties of relatively brittle shales by chemically removing some cementation material. To answer this question, we have leached out carbonates from Pierre I shale matrix using hydrochloric acid and we have compared mechanical properties of shale before and after leaching. We have also followed leaching dynamics using X-ray tomography. The results show that removal of around 4–5 wt% of cementation material results in 43% reduction in Pierre I shale shear strength compared to the non-etched shale exposed to sodium chloride solution for the same time. The etching rate was shown to be strongly affected by the volume of fluid staying in direct contact with the shale sample.

Primary biliary cholangitis (PBC) is a chronic autoimmune liver disease characterized by immune-mediated destruction of intrahepatic bile ducts and the presence of specific antibodies. The aim of the study was to examine the diagnostic significance of antibodies against promyelocytic leukemia nuclear body (PML NB) components such as Sp100, Sp140, and PML in a cohort of PBC patients and compare the results with biochemical and histological parameters. Serum samples were collected from 93 PBC patients. Anti-Sp100 and anti-PML antibodies were assessed using commercially available kits, anti-Sp140 using developed “in-house” ELISA test. Anti-Sp140, anti-Sp100, and anti-PML antibodies were present in 25 (27%), 37 (40%), and 29 (31%) PBC patients, respectively. Anti-PML NB positive patients also showed increased concentration of bilirubin and alkaline phosphatase (p < 0.05). In the group with the presence of at least two types of these antibodies, more frequent deaths or transplantations were observed. A correlation between the presence of antibodies and histological grade (OR = 2.55 p = 0.039) was established. Patients with bilirubin > 1.1 mg/dL at the time of diagnosis had a significantly shorter time of survival than patients with bilirubin ≤ 1.1 mg/dL (HR 5.7; 95% C.I., 2.7, 12.3; p < 0.001). Our data confirm very high specificity of anti-PML NB antibodies, which can expand the laboratory diagnostic capabilities of PBC. We found an association between positive reactivity of autoantibodies directed against components of PML nuclear bodies and higher concentrations of bilirubin and alkaline phosphatase, but the main prognostic marker of survival remains serum bilirubin.

MicroRNAs (miRNAs, miRs) are small non-coding RNA (ncRNA) molecules capable of regulating post-transcriptional gene expression. Imbalances in the miRNA network have been associated with the development of many pathological conditions and diseases, including cancer. Recently, miRNAs have also been linked to the phenomenon of multidrug resistance (MDR). MiR-7 is one of the extensively studied miRNAs and its role in cancer progression and MDR modulation has been highlighted. MiR-7 is engaged in multiple cellular pathways and acts as a tumor suppressor in the majority of human neoplasia. Its depletion limits the effectiveness of anti-cancer therapies, while its restoration sensitizes cells to the administered drugs. Therefore, miR-7 might be considered as a potential adjuvant agent, which can increase the efficiency of standard chemotherapeutics.

Multiple sclerosis (MS) is a chronic inflammatory disease of the CNS and has a varying disease course as well as variable response to treatment. Biomarkers may therefore aid personalized treatment. We tested whether in vitro activation of MS patient-derived CD4+ T cells could reveal potential biomarkers. The dynamic gene expression response to activation was dysregulated in patient-derived CD4+ T cells. By integrating our findings with genome-wide association studies, we constructed a highly connected MS gene module, disclosing cell activation and chemotaxis as central components. Changes in several module genes were associated with differences in protein levels, which were measurable in cerebrospinal fluid and were used to classify patients from control individuals. In addition, these measurements could predict disease activity after 2 years and distinguish low and high responders to treatment in two additional, independent cohorts. While further validation is needed in larger cohorts prior to clinical implementation, we have uncovered a set of potentially promising biomarkers.

Good shear bonding and hydraulic bonding between cement and steel play a crucial role in well integrity of oil and gas wells. In this experimental study, we investigate the effect that constant electric field may have on the bonding at cement-steel interfaces. Constant voltage (18 V) was applied between two stainless-steel electrodes immersed into a cement slurry. It was found that bonding was significantly improved at the positive electrode, while it was significantly worse at the negative electrode. The effect was due to the negatively-charged cement particles being attracted to the positive electrode. The effect may potentially be used for manipulation and control of casing-cement and reinforcement-concrete bonding strengths in oil & gas and construction industries, respectively. Side-effects that might reduce the applicability of this technology, are gas production at both electrodes (and especially at the negative one) and significant corrosion at the positive electrode due to electrochemical reactions at metal surfaces. Poor bonding at the negative electrode may potentially be used for cleaning of cement equipment, such as cement pumps, pipes, tanks, and mixers used on the rigs to perform well cementing jobs in oil & gas industry.

5-methylcytosine (5mC) is converted to 5-hydroxymethylcytosine (5hmC) by the TET family of enzymes as part of a recently discovered active DNA de-methylation pathway. 5hmC plays important roles in regulation of gene expression and differentiation and has been implicated in T cell malignancies and autoimmunity. Here, we report early and widespread 5mC/5hmC remodeling during human CD4+ T cell differentiation ex vivo at genes and cell-specific enhancers with known T cell function. We observe similar DNA de-methylation in CD4+ memory T cells in vivo, indicating that early remodeling events persist long term in differentiated cells. Underscoring their important function, 5hmC loci were highly enriched for genetic variants associated with T cell diseases and T-cell-specific chromosomal interactions. Extensive functional validation of 22 risk variants revealed potentially pathogenic mechanisms in diabetes and multiple sclerosis. Our results support 5hmC-mediated DNA de-methylation as a key component of CD4+ T cell biology in humans, with important implications for gene regulation and lineage commitment.

Within the EU-SPIDIA project (www.spidia.eu), the quality parameters of blood genomic DNA were defined [SPIDIA-DNA: an External Quality Assessment for the pre-analytical phase of blood samples used for DNA-based analyses – [1]; Influence of pre-analytical procedures on genomic DNA integrity in blood samples: the SPIDIA experience – [2]; Combining qualitative and quantitative imaging evaluation for the assessment of genomic DNA integrity: the SPIDIA experience – [3]. DNA quality parameters were used to evaluate the laboratory performance within an External Quality Assessment (EQA) [Second SPIDIA-DNA External Quality Assessment (EQA): Influence of pre-analytical phase of blood samples on genomic DNA quality – [4]. These parameters included DNA purity and yield by UV spectrophotometric measurements, the presence of PCR interferences by Kineret software and genomic DNA integrity analysis by Pulsed Field Gel Electrophoresis.Here we present the specific laboratory report of the 2nd SPIDIA-DNA EQA as an example of data and performances evaluation.

Metabotropic glutamate subtype 5 (mGlu5) receptors are implicated in various forms of synaptic plasticity, including drugs of abuse. In drug-addicted individuals, associative memories can drive relapse to drug use. The present study investigated the potential of the mGlu5 receptor positive allosteric modulator (PAM), VU-29 (30 mg/kg, i.p.), to inhibit the maintenance of a learned association between ethanol and environmental context by using conditioned place preference (CPP) in rats. The ethanol-CPP was established by the administration of ethanol (1.0 g/kg, i.p. ×10 days) using an unbiased procedure. Following ethanol conditioning, VU-29 was administered at various post-conditioning times (ethanol free state at the home cage) to ascertain if there was a temporal window during which VU-29 would be effective. Our experiments indicated that VU-29 did not affect the expression of ethanol-induced CPP when it was given over two post-conditioning days. However, the expression of ethanol-CPP was inhibited by 10-day home cage administration of VU-29, but not by first 2-day or last 2-day injection of VU-29 during the 10-day period. These findings reveal that VU-29 can inhibit the maintenance of ethanol-induced CPP, and that treatment duration contributes to this effect of VU-29. Furthermore, VU-29 effect was reversed by pretreatment with either MTEP (the mGlu5 receptor antagonist), or MK-801 (the N-methyl-D-aspartate-NMDA receptor antagonist). Thus, the inhibitory effect of VU-29 is dependent on the functional interaction between mGlu5 and NMDA receptors. Because a reduction in ethanol-associated cues can reduce relapse, mGlu5 receptor PAM would be useful for therapy of alcoholism. Future research is required to confirm the current findings.

Palmatine (PALM) and berberine (BERB) are widely identified isoquinoline alkaloids among the representatives of the Berberidaceae botanical family. The antiseizure activity of BERB was shown previously in experimental epilepsy models. We assessed the effect of PALM in a pentylenetetrazole (PTZ)-induced seizure assay in zebrafish, with BERB as an active reference compound. Both alkaloids were isolated from the methanolic root extract of Berberis sibirica by counter-current chromatography, and their ability to cross the blood–brain barrier was determined via quantitative structure–activity relationship assay. PALM exerted antiseizure activity, as confirmed by electroencephalographic analysis, and decreased c-fos and bdnf levels in PTZ-treated larvae. In a behavioral assay, PALM dose-dependently decreased PTZ-induced hyperlocomotion. The combination of PALM and BERB in ED16 doses revealed hyperadditive activity towards PTZ-induced hyperlocomotion. Notably, we have indicated that both alkaloids may exert their anticonvulsant activity through different mechanisms of action. Additionally, the combination of both alkaloids in a 1:2.17 ratio (PALM: BERB) mimicked the activity of the pure extract, which indicates that these two active compounds are responsible for its anticonvulsive activity. In conclusion, our study reveals for the first time the anticonvulsant activity of PALM and suggests the combination of PALM and BERB may have higher therapeutic value than separate usage of these compounds.

In carbon storage activities, and in shale oil and gas extraction (SOGE) with carbon dioxide (CO2) as stimulation fluid, CO2 comes into contact with shale rock and its pore fluid. As a reactive fluid, the injected CO2 displays a large potential to modify the shale’s chemical, physical, and mechanical properties, which need to be well studied and documented. The state of the art on shale−CO2 interactions published in several review articles does not exhaust all aspects of these interactions, such as changes in the mechanical, petrophysical, or petrochemical properties of shales. This review paper presents a characterization of shale rocks and reviews their possible interaction mechanisms with different phases of CO2. The effects of these interactions on petrophysical, chemical and mechanical properties are highlighted. In addition, a novel experimental approach is presented, developed and used by our team to investigate mechanical properties by exposing shale to different saturation fluids under controlled temperatures and pressures, without modifying the test exposure conditions prior to mechanical and acoustic measurements. This paper also underlines the major knowledge gaps that need to be filled in order to improve the safety and efficiency of SOGE and CO2 storage.

Conservation has long focused on preserving or restoring pristine ecosystems. However, understanding and managing novel ecosystems has grown in importance as they outnumber pristine ecosystems worldwide. While non-native species may be neutral or detrimental in pristine ecosystems, it is possible that even notorious invaders could play beneficial or mixed roles in novel ecosystems. We examined the effects of two long-established non-native species—Philippine deer (Rusa marianna) and feral pigs (Sus scrofa)—in Guam, Micronesia, where native vertebrate frugivores are functionally absent leaving forests devoid of seed dispersers. We compared the roles of deer and pigs on seedling survival, seed dispersal and plant community structure in limestone karst forests. Deer, even at low abundances, had pronounced negative impacts on forest communities by decreasing seedling and vine abundance. By contrast, pigs showed no such relationship. Also, many viable seeds were found in pig scats, whereas few were found in deer scats, suggesting that pigs, but not deer, provide an ecosystem function—seed dispersal—that has been lost from Guam. Our study presents a discrepancy between the roles of two non-native species that are traditionally managed as a single entity, suggesting that ecological function, rather than identity as a non-native, may be more important to consider in managing novel systems.

Stem cells (SCs) may constitute a perspective alternative to pharmacological treatment in neurodegenerative diseases. Although the safety of SC transplantation has been widely shown, their clinical efficiency in amyotrophic lateral sclerosis (ALS) is still to be proved. It is not only due to a limited number of studies, small treatment groups, and fast but nonlinear disease progression but also due to lack of objective methods able to show subtle clinical changes. Preliminary guidelines for cell therapy have recently been proposed by a group of ALS experts. They combine clinical, neurophysiological, and functional assessment together with monitoring of the cytokine level. Here, we describe a pilot study on transplantation of autologous adipose-derived regenerative cells (ADRC) into the spinal cord of the patients with ALS and monitoring of the results in accordance with the current recommendations. To show early and/or subtle changes within the muscles of interest, a wide range of clinical and functional tests were used and compared in order to choose the most sensitive and optimal set. Additionally, an analysis of transplanted ADRC was provided to develop standards ensuring the derivation and verification of adequate quality of transplanted cells and to correlate ADRC properties with clinical outcome.

DNA-polyacrylamide hybrid hydrogels designed with covalent and double-stranded (dsDNA) crosslinks respond to specific single-stranded DNA (ssDNA) probes by adapting new equilibrium swelling volume. The ssDNA probes need to be designed with a base pair sequence that is complementary to one of the strands in a dsDNA supported network junction. This work focuses on tuning the hydrogel swelling kinetics by introducing polyethylene glycol (PEG) as a pore-forming agent. Adding PEG during the preparation of hydrogels, followed by removal after polymerization, has been shown to improve the swelling dynamics of DNA hybrid hydrogels upon specific ssDNA probe recognition. The presence of porogen did not influence the kinetics of osmotic pressure-driven (2-acrylamido-2-methylpropane sulfonic acid)-co-acrylamide (AMPSA-co-AAm) hydrogels’ swelling, which is in contrast to the DNA-sensitive hydrogels. The difference in the effect of using PEG as a porogen in these two cases is discussed in view of processes leading to the swelling of the gels.

The aim of this work is to compare the electrical properties of BaCe0.85Y0.15O3−δ (BCY15), BaCe0.70In0.30O3−δ (BCI30) and a composite material consisting of 30%vol. BCY15 and 70%vol. Ce0.85Y0.15O2−δ (YDC15). BCY15 and YDC15 were synthesized by co-precipitation, whereas BCI30 was obtained using the solid-state reaction method. Pellets were initially formed from powders at 5 MPa, after which they were isostatically pressed at 250 MPa and sintered at 1500°C. Electrochemical impedance spectroscopy (EIS) was used to determine the electrical properties of the samples in both air (pO2 = 0.021 MPa) and Ar-5%H2 atmospheres. In the temperature range 200-400°C in air atmosphere the highest conductivity values were determined for BCY15 (5,22·10−5 − 2.74·10−3 S/cm). On the other hand, the electrical conductivity values obtained for Y70B30 in both atmospheres between 200 and 550°C are in the order of magnitude of 10−7 − 10−3 S/cm. Consequently, it can be concluded that the compounds exhibit significant H+ and O2− electrical conductivity at temperatures above 500°C, which indicates the possibility for their potential use as ceramic fuel cell electrolytes.

Recent technological advancements have made time-resolved, quantitative, multi-omics data available for many model systems, which could be integrated for systems pharmacokinetic use. Here, we present large-scale simulation modeling (LASSIM), which is a novel mathematical tool for performing large-scale inference using mechanistically defined ordinary differential equations (ODE) for gene regulatory networks (GRNs). LASSIM integrates structural knowledge about regulatory interactions and non-linear equations with multiple steady state and dynamic response expression datasets. The rationale behind LASSIM is that biological GRNs can be simplified using a limited subset of core genes that are assumed to regulate all other gene transcription events in the network. The LASSIM method is implemented as a general-purpose toolbox using the PyGMO Python package to make the most of multicore computers and high performance clusters, and is available at https://gitlab.com/Gustafsson-lab/lassim. As a method, LASSIM works in two steps, where it first infers a non-linear ODE system of the pre-specified core gene expression. Second, LASSIM in parallel optimizes the parameters that model the regulation of peripheral genes by core system genes. We showed the usefulness of this method by applying LASSIM to infer a large-scale non-linear model of naïve Th2 cell differentiation, made possible by integrating Th2 specific bindings, time-series together with six public and six novel siRNA-mediated knock-down experiments. ChIP-seq showed significant overlap for all tested transcription factors. Next, we performed novel time-series measurements of total T-cells during differentiation towards Th2 and verified that our LASSIM model could monitor those data significantly better than comparable models that used the same Th2 bindings. In summary, the LASSIM toolbox opens the door to a new type of model-based data analysis that combines the strengths of reliable mechanistic models with truly systems-level data. We demonstrate the power of this approach by inferring a mechanistically motivated, genome-wide model of the Th2 transcription regulatory system, which plays an important role in several immune related diseases.

The aim of the study was to assess plasma concentration of catecholamines and asymmetric dimethyl arginine levels and a possible relationship to predict the mortality rates among hemodialysis patients. The study population comprised 27 subjects, aged 65-70 years. Each patient underwent dialysis thrice a week. Furthermore, the median duration of hemodialysis was 3.5 years. Based on the conducted research, it can be concluded that the concentrations of adrenaline and the level of asymmetric dimethylarginine have predictive value of mortality among hemodialysis patients. Of note, lowering plasma asymmetric dimethylarginine concentration may represent therapeutic target for prevention of progressive renal damage.

Considering the nutritional values, breadstuff plays a big part in covering human nourishment needs and constitutes a base of all day diet. Moreover, bread is an excellent source of numerous vitamins and minerals the abundance of which depends on the degree of grinding. Thus, it seems to be very important to know the composition and level of bio-elements. That is why the main target of this study was to evaluate the concentration of selected trace elements: chromium (Cr), nickel (Ni), iron (Fe) and manganese (Mn) in wheat grain, wheat bran, different wheat and rye flour types and variety of breadstuff also with addition of grains and seeds from different bakeries and mills. Another task was to analyze if the technological process has an influence on secondary despoil of bread goods with heavy metal elements. The analyzed trace elements were measured with a precise and accurate atomic absorption spectrophotometric method (AAS) and the results were expressed in mg/kg of selected sample. Obtained results show that bread and grain products are a good source of trace elements like chromium, nickel, iron and manganese. However, the higher levels of chromium and nickel in bread goods could rather be an effect of impurity caused by a technological process in mill and bakeries.

This paper presents a quite simple, fully automatized single droplet generator, which can be an alternative for more expensive and complicated microfluidic devices. The simple generation nozzle connected to the pressure cells and cheap peristaltic pumps, synchronized via developed software with simple GUI (graphical user interface) implemented into the Raspberry Pi microcomputer allows precise control over the single droplet diameter and detachment frequency. The generator allows the formation of droplets of quite wide range of diameters without the need of orifice diameter replacements. Free control over time available for adsorption of surface active-substances over the surface of immobilized droplet, before its detachment from the orifice, is an advantage of the developed device.

Multiuser virtual environments (MUVEs) like Second Life present unparalleled opportunities to help students connect knowledge by description to knowledge by experience; in a MUVE, students can experience phenomena rather than only reading about them. Baba Kofi Weusijana, Vanessa Svihla, Drue Gawel, and John Bransford describe their use of a maze constructed in Second Life to help students experience firsthand the phenomena described in their educational psychology course. Their use of Second Life is particularly notable in its use of MUVE-based movies and other strategies to leverage Second Life's interactive powers for exploration despite restricted access to technology. The examples they present can hopefully lead to new designs and uses of virtual environments that allow students to experience relevant phenomena and enable researchers to conduct additional experiments of virtual, experience-rich additions to traditional ways to teach and learn. Specific immunotherapy (SIT) reverses the symptoms of seasonal allergic rhinitis (SAR) in most patients. Recent studies report type I interferons shifting the balance between type I T helper cell (Th1) and type II T helper cells (Th2) towards Th2 dominance by inhibiting the differentiation of naive T cells into Th1 cells. As SIT is thought to cause a shift towards Th1 dominance, we hypothesized that SIT would alter interferon type I signaling. To test this, allergen and diluent challenged CD4+ T cells from healthy controls and patients from different time points were analyzed. The initial experiments focused on signature genes of the pathway and found complex changes following immunotherapy, which were consistent with our hypothesis. As interferon signaling involves multiple genes, expression profiling studies were performed, showing altered expression of the pathway. These findings require validation in a larger group of patients in further studies.
