Cell growth is controlled by a lysosomal signalling complex containing Rag small GTPases and mammalian target of rapamycin complex 1 (mTORC1) kinase. Here, we carried out a microscopy-based genome-wide human short interfering RNA screen and discovered a lysosome-localized G protein-coupled receptor (GPCR)-like protein, GPR137B, that interacts with Rag GTPases, increases Rag localization and activity, and thereby regulates mTORC1 translocation and activity. High GPR137B expression can recruit and activate mTORC1 in the absence of amino acids. Furthermore, GPR137B also regulates the dissociation of activated Rag from lysosomes, suggesting that GPR137B controls a cycle of Rag activation and dissociation from lysosomes. GPR137B-knockout cells exhibited defective autophagy and an expanded lysosome compartment, similar to Rag-knockout cells. Like zebrafish RagA mutants, GPR137B-mutant zebrafish had upregulated TFEB target gene expression and an expanded lysosome compartment in microglia. Thus, GPR137B is a GPCR-like lysosomal regulatory protein that controls dynamic Rag and mTORC1 localization and activity as well as lysosome morphology.

Vinegar is one of the most widely used acidic condiments. Recently, rapid advances have been made in the area of vinegar research. Different types of traditional vinegar are available around the globe and have many applications. Vinegar can be made either naturally, through alcoholic and then acetic acid fermentation, or artificially, in laboratories. Vinegar is the product of acetic acid fermentation of dilute alcoholic solutions, manufactured by a two-step process. The first step is the production of ethanol from a carbohydrate source such as glucose, which is carried out by yeasts. The second step is the oxidation of ethanol to acetic acid, which is carried out by acetic acid bacteria. Acetic acid bacteria are not only producers of certain foods and drinks, such as vinegar, but they can also spoil other products such as wine, beer, soft drinks, and fruits. Various renewable substrates are used for the efficient biological production of acetic acid, including agro and food, dairy, and kitchen wastes. Numerous reports on the health advantages associated with vinegar ingredients have been presented. Fresh sugarcane juice was fermented with wine yeast and LB acetate bacteria to develop a high-quality original sugarcane vinegar beverage. To facilitate the current study, the bibliometric analysis method was adopted to visualize the knowledge map of vinegar research based on literature data. The present review article will help scientists discern the dynamic era of vinegar research and highlight areas for future research.

Micro-scale light-emitting diodes (micro-LEDs) have received widespread attention in recent years for applications in display and optical communication. Compared with conventional quantum well active regions, quantum dots (QDs) can increase the carrier concentration at the same current density, which is beneficial for improving the efficiency and bandwidth of LEDs at low current densities. This is exactly what micro-LEDs need for display and communication applications. In this Perspective, we give a general introduction to InGaN QDs and provide an overview of the growth of InGaN QDs by metal-organic chemical vapor deposition. We then discuss the advances in green and red micro-LEDs based on InGaN QDs for display applications. This is followed by recent progress on high-speed blue micro-LEDs, which have great potential for use in chip-to-chip optical interconnections. With the increasing complexity of societal and environmental problems in the Anthropocene, the use of both classification-based approaches, which provide in-depth understanding within disciplinary boundaries, and citation-based approaches, which provide interdisciplinary research, has been encouraged. However, there are limited comparisons of the knowledge networks produced between these two approaches, which compromises our capacity to manage technological development. This paper aims to investigate the similarities and differences of river technology networks produced using classification-based and citation-based approaches. The World Intellectual Property Organization (WIPO) database from 1863 to 2020 was used as the data source. River technology systems contained three interactive subsystems: water demand, water supply, and water management, and the structure was measured using network-based metrics. It was found that river technology systems constructed using the classification-based and the citation-based approaches developed similarly in terms of their temporal, spatial, and compositional features. The structural differences were attributed to the addition of an external system that draws upon interdisciplinary knowledge beyond water resources. Both approaches can be used for guiding technology management, with the classification-based approach being more effective for understanding the content of innovations and the citation-based approach being more effective in gathering information beyond the water resource discipline. Aiming at the problems of power attenuation and spot expansion of pulsed laser transmission in snowfall environments, based on ray scattering and Fraunhofer diffraction theories, a model of laser transmission attenuation in snowfall environments is established. The model considers the influence of multiple scattering effects by introducing a forward correction coefficient. The Monte Carlo multiple scattering model was used to numerically simulate the attenuation characteristics of a 905-nm pulsed laser in snowfall environments, and an experimental platform for the attenuation characteristics was constructed. The experimental, simulation, and theoretical results show that, in three snowfall environments of 0 < SR < 1, 1 < SR < 2, and 2 < SR < 3, the laser transmittance decreases with the increase of transmission distance, and when the transmission distance is the same, the laser transmittance decreases with the increase in snowfall rate. The influence of transmission distance, simulated photon number, and asymmetry factor on spot extension was studied. The simulation results show that the spot radius increases with the increase in transmission distance, remains almost constant with the increase in the simulated photon number, and decreases with the increase in the asymmetry factor in three snowfall environments. Previous research assumed deep buried loess as the initial composition of the overlying paleosol and failed to address the long-term continuous pedogenic weathering history in the deep loess-paleosol sequence, which was attributed to little understanding on the difference between loess and paleosol in the long-term deep loess-paleosol sequence. To distinguish between the loess and paleosol, in the long-term deep loess-paleosol sequence in northeast China, the morphology, dust deposition fluxes, geochemical characteristics, magnetic susceptibility, and grain size distributions were investigated. Results showed that the loess layers buried at depth could be differentiated from the paleosol by their poor pedogenic development. The presence of coarser grains in the loess as well as lesser amounts of clay and Fe–Mn coatings compared to paleosol indicated weaker weathering of the loess. Also, optical iron clay films deposited on the surface of the skeleton particles were less visible in the loesses than in the paleosols. From the loess evolution perspective, the pedogenic formation process of the newly formed loess soils should be considered as important as that of the reddish paleosol layer based on criteria of formation time, depth within profile, and morphological characteristics. The formation of the reddish or yellowish paleosol layer was constrained by pedogenic environments associated with climatic change and by the relative rates of deposition and pedogenesis. The terms “loessal paleosol” and “loessal paleosol sequence” are suggested to aid in systematically and consistently addressing the long-term pedogenic weathering evolution recorded in the complex formation of deep loess and paleosol sequences in pedology research. The long-term deep loessal sediments of Northeast China are loessal paleosols, which cannot be simply used as a reference for the overlying paleosol and be deducted from pedogenesis consideration.

Abstract Background Autophagy is a conserved catabolic process, which plays an important role in regulating tumor cell motility and degrading protein aggregates. Chemotherapy-induced autophagy may lead to tumor distant metastasis and even chemo-insensitivity in the therapy of hepatocellular carcinoma (HCC). Therefore, a vast majority of HCC cases do not produce a significant response to monotherapy with autophagy inhibitors. Results In this work, we developed a biomimetic nanoformulation (TH-NP) co-encapsulating Oxaliplatin (OXA)/hydroxychloroquine (HCQ, an autophagy inhibitor) to execute targeted autophagy inhibition, reduce tumor cell migration and invasion in vitro and attenuate metastasis in vivo. The tumor cell-specific ligand TRAIL was bioengineered to be stably expressed on HUVECs and the resultant membrane vesicles were wrapped on OXA/HCQ-loaded PLGA nanocores. Especially, TH-NPs could significantly improve OXA and HCQ effective concentration by approximately 21 and 13 times in tumor tissues compared to the free mixture of HCQ/OXA. Moreover, the tumor-targeting TH-NPs released HCQ alkalized the acidic lysosomes and inhibited the fusion of autophagosomes and lysosomes, leading to effective blockade of autophagic flux. In short, the system largely improved chemotherapeutic performance of OXA on subcutaneous and orthotopic HCC mice models. Importantly, TH-NPs also exhibited the most effective inhibition of tumor metastasis in orthotopic HCCLM3 models, and in the HepG2, Huh-7 or HCCLM3 metastatic mice models. Finally, we illustrated the enhanced metastasis inhibition was attributed to the blockade or reverse of the autophagy-mediated degradation of focal adhesions (FAs) including E-cadherin and paxillin. Conclusions TH-NPs can perform an enhanced chemotherapy and antimetastatic effect, and may represent a promising strategy for HCC therapy in clinics. Graphical Abstract
