151. Design of Complex Experiments Using Mixed Integer Linear Programming

Abstract: Over the past few decades, neuroscience experiments have become increasingly complex and naturalistic. Experimental design has in turn become more challenging, as experiments must conform to an ever-increasing diversity of design constraints. In this article we demonstrate how this design process can be greatly assisted using an optimization tool known as Mixed Integer Linear Programming (MILP). MILP provides a rich framework for incorporating many types of real-world design constraints into a neuroimaging experiment. We introduce the mathematical foundations of MILP, compare MILP to other experimental design techniques, and provide four case studies of how MILP can be used to solve complex experimental design challenges.

Abstract: The unidentified very-high-energy (VHE; E $>$ 0.1 TeV) $\gamma$-ray source, HESS J1826$-$130, was discovered with the High Energy Stereoscopic System (HESS) in the Galactic plane. The analysis of 215 h of HESS data has revealed a steady $\gamma$-ray flux from HESS J1826$-$130, which appears extended with a half-width of 0.21$^{\circ}$ $\pm$ 0.02$^{\circ}_{\text{stat}}$ $\pm$ 0.05$^{\circ}_{\text{sys}}$. The source spectrum is best fit with either a power-law function with a spectral index $\Gamma$ = 1.78 $\pm$ 0.10$_{\text{stat}}$ $\pm$ 0.20$_{\text{sys}}$ and an exponential cut-off at 15.2$^{+5.5}_{-3.2}$ TeV, or a broken power-law with $\Gamma_{1}$ = 1.96 $\pm$ 0.06$_{\text{stat}}$ $\pm$ 0.20$_{\text{sys}}$, $\Gamma_{2}$ = 3.59 $\pm$ 0.69$_{\text{stat}}$ $\pm$ 0.20$_{\text{sys}}$ for energies below and above $E_{\rm{br}}$ = 11.2 $\pm$ 2.7 TeV, respectively. The VHE flux from HESS J1826$-$130 is contaminated by the extended emission of the bright, nearby pulsar wind nebula (PWN), HESS J1825$-$137, particularly at the low end of the energy spectrum. Leptonic scenarios for the origin of HESS J1826$-$130 VHE emission related to PSR J1826$-$1256 are confronted by our spectral and morphological analysis. In a hadronic framework, taking into account the properties of dense gas regions surrounding HESS J1826$-$130, the source spectrum would imply an astrophysical object capable of accelerating the parent particle population up to $\gtrsim$200 TeV. Our results are also discussed in a multiwavelength context, accounting for both the presence of nearby supernova remnants (SNRs), molecular clouds, and counterparts detected in radio, X-rays, and TeV energies., Comment: 9 Pages, 5 Figures

Abstract: Dwarf spheroidal galaxy satellites of the Milky Way are prime targets for indirect detection of dark matter with gamma rays due to their proximity, high dark matter content and absence of non-thermal emission processes. Recently, the Dark Energy Survey (DES) revealed the existence of new ultra-faint dwarf spheroidal galaxies in the southern-hemisphere sky, therefore ideally located for ground-based observations with the imaging atmospheric Cherenkov telescope array H.E.S.S. We present a search for very-high-energy ($E\gtrsim100$ GeV) gamma-ray emission using H.E.S.S. observations carried out recently towards Reticulum II, Tucana II, Tucana III, Tucana IV and Grus II satellites. No significant very-high-energy gamma-ray excess is found from the observations on any individual object nor in the combined analysis of all the datasets. Using the most recent modeling of the dark matter distribution in the dwarf galaxy halo, we compute for the first time on DES satellites individual and combined constraints from Cherenkov telescope observations on the annihilation cross section of dark matter particles in the form of Weakly Interacting Massive Particles. The combined 95% C.L. observed upper limits reach $\langle \sigma v \rangle \simeq 1 \times 10^{-23}$ cm$^3$s$^{-1}$ in the $W^+W^-$ channel and $4 \times 10^{-26}$ cm$^3$s$^{-1}$ in the $\gamma\gamma$ channels for a dark matter mass of 1.5 TeV. The H.E.S.S. constraints well complement the results from Fermi-LAT, HAWC, MAGIC and VERITAS and are currently the most stringent in the $\gamma\gamma$ channels in the multi-GeV/multi-TeV mass range., Comment: 20 pages, 12 figures

Abstract: The nearby radio galaxy Centaurus A belongs to a class of Active Galaxies that are very luminous at radio wavelengths. The majority of these galaxies show collimated relativistic outflows known as jets, that extend over hundreds of thousands of parsecs for the most powerful sources. Accretion of matter onto the central super-massive black hole is believed to fuel these jets and power their emission, with the radio emission being related to the synchrotron radiation of relativistic electrons in magnetic fields. The origin of the extended X-ray emission seen in the kiloparsec-scale jets from these sources is still a matter of debate, although Cen A's X-ray emission has been suggested to originate in electron synchrotron processes. The other possible explanation is Inverse Compton (IC) scattering with CMB soft photons. Synchrotron radiation needs ultra-relativistic electrons ($\sim50$ TeV), and given their short cooling times, requires some continuous re-acceleration mechanism to be active. IC scattering, on the other hand, does not require very energetic electrons, but requires jets that stay highly relativistic on large scales ($\geq$1 Mpc) and that remain well-aligned with the line of sight. Some recent evidence disfavours inverse Compton-CMB models, although other evidence seems to be compatible with them. In principle, the detection of extended gamma-ray emission, directly probing the presence of ultra-relativistic electrons, could distinguish between these options, but instruments have hitherto been unable to resolve the relevant structures. At GeV energies there is also an unusual spectral hardening in Cen A, whose explanation is unclear. Here we report observations of Cen A at TeV energies that resolve its large-scale jet. We interpret the data as evidence for the acceleration of ultra-relativistic electrons in the jet, and favour the synchrotron explanation for the X-rays., Comment: 32 pages, 5 figure

Abstract: The controlled modification of the electronic properties of ZnO nanorods via transition metal doping is reported. A series of ZnO nanorods were synthesized by chemical bath growth with varying Co content from 0 to 20 atomic % in the growth solution. Optoelectronic behavior was probed using cathodoluminescence, time-resolved luminescence, transient absorbance spectroscopy, and the incident photon-to-current conversion efficiency (IPCE). Analysis indicates the crucial role of surface defects in determining the electronic behavior. Significantly, Co-doping extends the light absorption of the nanorods into the visible region, increases the surface defects, shortens the non-radiative lifetimes, while leaving the radiative lifetime constant. Furthermore, for 1 atomic % Co-doping the IPCE of the ZnO nanorods is enhanced. These results demonstrate that doping can controllably tune the functional electronic properties of ZnO nanorods for applications.

Abstract: Understanding the emergence, co-evolution, and convergence of science and technology (S&T) areas offers competitive intelligence for researchers, managers, policy makers, and others. The resulting data-driven decision support helps set proper research and development (R&D) priorities; develop future S&T investment strategies; monitor key authors, organizations, or countries; perform effective research program assessment; and implement cutting-edge education/training efforts. This paper presents new funding, publication, and scholarly network metrics and visualizations that were validated via expert surveys. The metrics and visualizations exemplify the emergence and convergence of three areas of strategic interest: artificial intelligence (AI), robotics, and internet of things (IoT) over the last 20 years (1998-2017). For 32,716 publications and 4,497 NSF awards, we identify their conceptual space (using the UCSD map of science), geospatial network, and co-evolution landscape. The findings demonstrate how the transition of knowledge (through cross-discipline publications and citations) and the emergence of new concepts (through term bursting) create a tangible potential for interdisciplinary research and new disciplines., Comment: 10 figures

157. Prospects on high-energy source searches based on pattern recognition: Object detection in the H.E.S.S. Galactic Plane Survey and catalogue cross-matches

Abstract: The H.E.S.S. Galactic Plane Survey (HGPS) represents one of the most sensitive surveys of the Galactic Plane at very high energies (VHE, 0.1-100 TeV). However the source detection algorithm of the HGPS pipeline is not well-suited for complex regions, including sources with shell-like morphologies. As an alternative and complementary approach, we have investigated blind search methods for VHE gamma-ray source detection based on well-known and widely used image processing and pattern recognition techniques. Our goal is to build in a short amount of computational time a list of potentially valuable objects without prior case-specific morphological assumptions. We aim to classify and rank the detected objects in order to identify only the most promising source candidates for further multi-wavelength-association searches, dedicated analyses, or deeper observations. In the approach proposed, we extract sparse and pertinent structural information from the significance maps using a edge detection operator. We then apply a Hough circle transform and detect a collection of objects as local maxima in the Hough space. On the basis of morphological parameters we can characterize different object classes. Classification can be used to identify valuable source candidates sharing the characteristics of well-known sources. We show that using these pattern recognition techniques we can detect objects with partial circular symmetry irrespective of a morphological template (e.g. point-like, Gaussian-like, or shell-like). All the shell-type supernova remnants (SNRs) catalogued in the HGPS are associated with at least one detected object. Catalogue cross-matches indicate that several detected objects not catalogued in the HGPS are spatially coincident with multi-wavelength counterparts. Further investigation on the most promising candidates will be conducted in dedicated follow-up analyses.

Abstract: The history of computer science and brain sciences are intertwined. In his unfinished manuscript "The Computer and the Brain," von Neumann debates whether or not the brain can be thought of as a computing machine and identifies some of the similarities and differences between natural and artificial computation. Turing, in his 1950 article in Mind, argues that computing devices could ultimately emulate intelligence, leading to his proposed Turing test. Herbert Simon predicted in 1957 that most psychological theories would take the form of a computer program. In 1976, David Marr proposed that the function of the visual system could be abstracted and studied at computational and algorithmic levels that did not depend on the underlying physical substrate. In December 2014, a two-day workshop supported by the Computing Community Consortium (CCC) and the National Science Foundation's Computer and Information Science and Engineering Directorate (NSF CISE) was convened in Washington, DC, with the goal of bringing together computer scientists and brain researchers to explore these new opportunities and connections, and develop a new, modern dialogue between the two research communities. Specifically, our objectives were: 1. To articulate a conceptual framework for research at the interface of brain sciences and computing and to identify key problems in this interface, presented in a way that will attract both CISE and brain researchers into this space. 2. To inform and excite researchers within the CISE research community about brain research opportunities and to identify and explain strategic roles they can play in advancing this initiative. 3. Abstract: The detection of the first electromagnetic counterpart to the binary neutron star (BNS) merger remnant GW170817 established the connection between short $\gamma$-ray bursts and BNS mergers. It also confirmed the forging of heavy elements in the ejecta (a so-called kilonova) via the r-process nucleosynthesis. The appearance of non-thermal radio and X-ray emission, as well as the brightening, which lasted more than 100 days, were somewhat unexpected. Current theoretical models attempt to explain this temporal behavior as either originating from a relativistic off-axis jet or a kilonova-like outflow. In either scenario, there is some ambiguity regarding how much energy is transported in the non-thermal electrons versus the magnetic field of the emission region. Combining the VLA (radio) and Chandra (X-ray) measurements with observations in the GeV-TeV domain can help break this ambiguity, almost independently of the assumed origin of the emission. Here we report for the first time on deep H.E.S.S. observations of GW170817 / GRB 170817A between 124 and 272 days after the BNS merger with the full H.E.S.S. array of telescopes, as well as on an updated analysis of the prompt (<5 days) observations with the upgraded H.E.S.S. phase-I telescopes. Abstract: In subarctic and arctic areas, large and heavy skid-steered robots are preferred for their robustness and ability to operate on difficult terrain. State estimation, motion control and path planning for these robots rely on accurate odometry models based on wheel velocities. However, the state-of-the-art odometry models for skid-steer mobile robots (SSMRs) have usually been tested on relatively lightweight platforms. In this paper, we focus on how these models perform when deployed on a large and heavy (590 kg) SSMR. We collected more than 2 km of data on both snow and concrete. We compare the ideal differential-drive, extended differential-drive, radius-of-curvature-based, and full linear kinematic models commonly deployed for SSMRs. Each of the models is fine-tuned by searching their optimal parameters on both snow and concrete. Abstract: We report on the detection of very-high-energy (VHE; $E > 100$ GeV) $\gamma$-ray emission from the BL Lac objects KUV 00311-1938 and PKS 1440-389 with the High Energy Stereoscopic System (H.E.S.S.). H.E.S.S. observations were accompanied or preceded by multi-wavelength observations with Fermi/LAT, XRT and UVOT on board the Swift satellite, and ATOM. Based on an extrapolation of the Fermi/LAT spectrum towards the VHE $\gamma$-ray regime, we deduce a 95% confidence level upper limit on the unknown redshift of KUV 00311-1938 of z < 0.98, and of PKS 1440-389 of z < 0.53. When combined with previous spectroscopy results the redshift of KUV 00311-1938 is constrained to $0.51 \leq z < 0.98$ and for PKS 1440-389 to $0.14 \lessapprox z < 0.53$., Comment: Accepted for publication in MNRAS. 13 pages, 4 figures

Abstract: Aims. Colliding wind binary systems have long been suspected to be high-energy (HE; 100 MeV < E < 100 GeV) {\gamma}-ray emitters. {\eta} Car is the most prominent member of this object class and is confirmed to emit phase-locked HE {\gamma} rays from hundreds of MeV to ~100 GeV energies. This work aims to search for and characterise the very-high-energy (VHE; E >100 GeV) {\gamma}-ray emission from {\eta} Car around the last periastron passage in 2014 with the ground-based High Energy Stereoscopic System (H.E.S.S.). Methods. The region around {\eta} Car was observed with H.E.S.S. between orbital phase p = 0.78 - 1.10, with a closer sampling at p {\approx} 0.95 and p {\approx} 1.10 (assuming a period of 2023 days). Optimised hardware settings as well as adjustments to the data reduction, reconstruction, and signal selection were needed to suppress and take into account the strong, extended, and inhomogeneous night sky background (NSB) in the {\eta} Car field of view. Tailored run-wise Monte-Carlo simulations (RWS) were required to accurately treat the additional noise from NSB photons in the instrument response functions. Results. H.E.S.S. detected VHE {\gamma}-ray emission from the direction of {\eta} Car shortly before and after the minimum in the X-ray light-curve close to periastron. Using the point spread function provided by RWS, the reconstructed signal is point-like and the spectrum is best described by a power law. The overall flux and spectral index in VHE {\gamma} rays agree within statistical and systematic errors before and after periastron. The {\gamma}-ray spectrum extends up to at least ~400 GeV. This implies a maximum magnetic field in a leptonic scenario in the emission region of 0.5 Gauss. Abstract: Women reliant on mostly rice-based diets can have inadequate thiamine intake, placing breastfed infants at risk of thiamine deficiency and, in turn, physical and cognitive impairments. We investigated the impact of maternal thiamine supplementation doses on infants' cognitive, motor, and language development across the first year. In this double-blind, four-parallel-arm, randomized controlled trial, healthy mothers of exclusively breastfed newborn infants were recruited in Kampong Thom, Cambodia. At 2 weeks postnatal, women (n = 335) were randomized to one of four treatment groups to consume one capsule/day with varying amounts of thiamine for 22 weeks: 0, 1.2, 2.4, and 10 mg. At 2, 12, 24, and 52 weeks of age, infants were assessed with the Mullen Scales of Early Learning (MSEL) and the Caregiver Reported Early Development Instrument (CREDI). Multiple regression and mixed effects modeling suggest that by 6 months of age, the highest maternal thiamine dose (10 mg/day) held significant benefits for infants' language development, but generally not for motor or visual reception development. Despite having achieved standardized scores on the MSEL that approximated U.S. norms by 6 months, infants showed a significant drop relative to these norms in both language domains following trial completion, indicating that nutritional interventions beyond 6 months may be necessary.

Abstract: BackgroundInfantile beriberi-related mortality is still common in South and Southeast Asia. Interventions to increase maternal thiamine intakes, and thus human milk thiamine, are warranted; however, the required dose remains unknown.ObjectivesWe sought to estimate the dose at which additional maternal intake of oral thiamine no longer meaningfully increased milk thiamine concentrations in infants at 24 wk postpartum, and to investigate the impact of 4 thiamine supplementation doses on milk and blood thiamine status biomarkers.MethodsIn this double-blind, 4-parallel arm randomized controlled dose-response trial, healthy mothers were recruited in Kampong Thom, Cambodia. At 2 wk postpartum, women were randomly assigned to consume 1 capsule, containing 0, 1.2 (estimated average requirement), 2.4, or 10 mg of thiamine daily from 2 through 24 weeks postpartum. Human milk total thiamine concentrations were measured using HPLC. An Emax curve was plotted, which was estimated using a nonlinear least squares model in an intention-to-treat analysis. Linear mixed-effects models were used to test for differences between treatment groups. Maternal and infant blood thiamine biomarkers were also assessed.ResultsIn total, each of 335 women was randomly assigned to1 of the following thiamine-dose groups: placebo (n = 83), 1.2 mg (n = 86), 2.4 mg (n = 81), and 10 mg (n = 85). The estimated dose required to reach 90% of the maximum average total thiamine concentration in human milk (191 µg/L) is 2.35 (95% CI: 0.58, 7.01) mg/d. 170. New approaches for sepsis research: host-directed therapies and predictive models

Abstract: Sepsis is defined as a dysregulated host response to infection. Decades of research have revealed many immune and metabolic mechanisms of sepsis pathogenesis, but these findings have not translated into successful therapies in the clinic. Here we work to address three major issues that have hampered this success. First, we establish a novel murine polymicrobial sepsis model that we believe addresses shortcomings of previous preclinical models: by conducting an intraperitoneal infection with the gram-negative bacterium Escherichia coli and the gram-positive bacterium Staphylococcus aureus, we combine the clinical relevance of a polymicrobial infection with the tractability of defined culturable microbes. Second, we leverage the selective serotonin reuptake inhibitor (SSRI) fluoxetine to drive immunometabolic disease tolerance during sepsis since it is widely available and affordable, has established pharmacological safety, and has demonstrated beneficial immune and metabolic effects in other contexts that translate into protective effects during sepsis. Specifically, we found that fluoxetine-mediated protection is independent of peripheral serotonin, and instead increases levels of circulating IL-10. IL-10 is necessary for protection from sepsis-induced hypertriglyceridemia and cardiac triglyceride accumulation, allowing for metabolic reprogramming of the heart. Third, we address the issue of sepsis variability – septic patients can exhibit a wide variety of outcomes, and the mechanisms that drive some individuals to survive and others to succumb are unclear. We found that in our murine polymicrobial sepsis model, cerebral cortex structure could be used to predict infection outcome – mice with thicker cortexes were more likely to survive. Together this work leverages immunometabolic mechanisms to identify a novel host-directed therapy and reveals a readily quantifiable trait that accurately trains machine learning models to predict sepsis outcome.

Penelitian ini menggunakan 4 teknik pengumpulan data yaitu teknik bebas libat cakap (SBLC), rekam, transkrip data, dan catat. Hasil penelitian berupa 1) bentuk tindak tutur deklaratif lebih banyak ditemukan sekitar 20 data, sedangkan data tindak tutur yang paling sedikit yaitu tindak tutur interogatif dengan 1 data. Bentuk tindak tutur imperatif ada 2 data bentuk tindak tutur imperatif berupa perintah dan bentuk tindak tutur interogatif berupa pertanyaan. 2) Fungsi tindak tutur representatif paling banyak ditemukan sebanyak 18 data, sedangkan fungsi tindak tutur direktif hanya ditemukan 7 data. 3) Relevansi tindak tutur slogan iklan kopi bisa diimplikasikan pada KD 3.3 dan 4.3 pada muatan ajar bahasa Indonesia di SMP kelas VIII. Implementasi tindak tutur bisa dilakukan dengan menambahkan kajian tindak tutur slogan iklan sebagai materi penguat atau sumber belajar tambahan bagi siswa sebagai pembanding model tindak tutur di ranah digital visual. Abstract: Abstract Background While some research indicates that individuals can accurately judge smile authenticity of enjoyment and masking smile expressions, other research suggest modest judgment rates of masking smiles. The current study explored the role of emotion-related individual differences in the judgment of authenticity and recognition of negative emotions in enjoyment and masking smile expressions as a potential explanation for the differences observed. Methods Specifically, Experiment 1 investigated the role of emotion contagion (Doherty in J Nonverbal Behav 21:131–154, 1997), emotion intelligence (Schutte et al. in Personality Individ Differ 25:167–177, 1998), and emotion regulation (Gratz and Roemer in J Psychopathol Behav Assess 26:41–54, 2004) in smile authenticity judgment and recognition of negative emotions in masking smiles. Experiment 2 investigated the role of state and trait anxiety (Spielberger et al. in Manual for the state-trait anxiety inventory, Consulting Psychologists Press, Palo Alto, 1983) in smile authenticity judgment and recognition of negative emotions in the same masking smiles. In both experiments, repeated measures ANOVAs were conducted for judgment of authenticity, probability of producing the expected response, for the detection of another emotion, and for emotion recognition. A series of correlations were also calculated between the proportion of expected responses of smile judgement and the scores on the different subscales. Results Results of the smile judgment and recognition tasks were replicated in both studies, and echoed results from prior studies of masking smile judgment: participants rated enjoyment smiles as happier than the masking smiles and, of the masking smiles, participants responded “really happy” more often for the angry-eyes masking smiles and more often categorized fear masking smiles as “not really happy”. Conclusions Overall, while the emotion-related individual differences used in our study seem to have an impact on recognition of basic emotions in the literature, our study suggest that these traits, except for emotional awareness, do not predict performances on the judgment of complex expressions such as masking smiles. These results provide further information regarding the factors that do and do not contribute to greater judgment of smile authenticity and recognition of negative emotions in masking smiles.

Abstract: Complex natural tasks likely recruit many different functional brain networks, but it is difficult to predict how such tasks will be represented across cortical areas and networks. Previous electrophysiology studies suggest that task variables are represented in a low-dimensional subspace within the activity space of neural populations. Here we develop a voxel-based state space modeling method for recovering task-related state spaces from human fMRI data. We apply this method to data acquired in a controlled visual attention task and a video game task. We find that each task induces distinct brain states that can be embedded in a low-dimensional state space that reflects task parameters, and that attention increases state separation in the task-related subspace. Our results demonstrate that the state space framework offers a powerful approach for modeling human brain activity elicited by complex natural tasks.

Abstract: Background Successful containment strategies for SARS-CoV-2, the causative virus of the COVID-19 pandemic, have involved widespread population testing that identifies infections early and enables rapid contact tracing. In this study, we developed a rapid and inexpensive RT- qPCR testing pipeline for population-level SARS-CoV-2 detection, and used this pipeline to establish a clinical laboratory dedicated to COVID-19 testing at the University of California San Diego (UCSD) with a processing capacity of 6,000 samples per day and next-day result turnaround times. Methods and findings Using this pipeline, we screened 6,786 healthcare workers and first responders, and 21,220 students, faculty, and staff from UCSD. Additionally, we screened 6,031 preschool-grade 12 students and staff from public and private schools across San Diego County that remained fully or partially open for in-person teaching during the pandemic. Between April 17, 2020 and February 5, 2021, participants provided 161,582 nasal swabs that were tested for the presence of SARS-CoV-2. Overall, 752 positive tests were obtained, yielding a test positivity rate of 0.47%. While the presence of symptoms was significantly correlated with higher viral load, most of the COVID-19 positive participants who participated in symptom surveys were asymptomatic at the time of testing. The positivity rate among preschool-grade 12 schools that remained open for in-person teaching was similar to the positivity rate at UCSD and lower than that of San Diego County, with the children in private schools being less likely to test positive than the adults at these schools. Conclusions Most schools across the United States have been closed for in-person learning for much of the 2020-2021 school year, and their safe reopening is a national priority. However, as there are no vaccines against SARS-CoV-2 currently available to the majority of school-aged children, the traditional strategies of mandatory masking, physical distancing, and repeated viral testing of students and staff remain key components of risk mitigation in these settings. The data presented here suggest that the safety measures and repeated testing actions taken by participating healthcare and educational facilities were effective in preventing outbreaks, and that a similar combination of risk-mitigation strategies and repeated testing may be successfully adopted by other healthcare and educational systems.

Abstract: Three decades of concentrated research on the topic of cheating in college has shown that there is no magic bullet to stopping or reducing cheating. The goal of this study was to exam the potential of nudges other than integrity pledges to incentivize, encourage, or entice students to be honest, even when it is easy and advantageous to be dishonest. The authors' goal was to find, in the context of actual classes with actual stakes (grades and enforcement), salient messages that could positively influence students' attitudes and behaviors away from cheating and toward integrity. The project deemphasized the integrity pledge approach, instead looking for other messaging approaches that might resonate more with students, not lead them to suspect more cheating, and remind them to act with integrity.

Abstract: Abstract Many biotechnologies require direct contact between biomolecules and (semi)solid substrates. However, little information is available regarding site‐directed covalent surface immobilization of heterodimeric proteins. Integrins are heterodimeric, conformationally dependent membrane adhesion‐receptors, which are important in the (patho)biology of almost all human diseases. In this study, a biomimetic‐platform is developed for covalent and site‐directed immobilization of oriented integrin heterodimers onto most hydroxyl bearing surfaces, herein demonstrated on both glass‐slides and silica‐particles. This platform consists of a self‐assembled monolayer, upon which C‐terminal modified αIIbβ3 integrin extracellular domains are oriented and covalently anchored. As with cell surface integrins, the conformation of immobilized αIIbβ3 is switchable and can be modulated to the active ligand‐binding conformation by divalent cations. Furthermore, the αIIbβ3‐coupled silica particles display platelet‐mimetic hemostat function and co‐aggregate with platelets from both wild‐type and fibrinogen/von Willebrand factor double deficient mice, facilitating αIIbβ3‐non‐classical ligand discoveries. This work provides a biomaterial platform for functional multimeric protein‐substrate coupling, which should have broad impact on multiple fields of biology, biotechnology, and clinical diagnosis/therapy.

Abstract: Extreme event attribution is a rapidly growing field of climate science with important implications for public and government understanding of human-induced climate change. However, there is substantial variation in how well events can be attributed to human-induced climate change, depending on the nature of the event. Focusing on Australia: at one end of the scale, large-scale heat events on both the land and in the ocean are well suited to attribution studies because climate models simulate them reasonably well, there are high-quality observations available and our understanding of the processes that lead to extreme heat events is reasonably well developed. At the other end of the scale, very important phenomenon such as changes in east coast lows, severe convective storms and long-term droughts are less well observed, are beyond our current capability to robustly simulate in climate models and the complex mechanisms that lead to intensification are not well understood. Thus, some important extreme events can be linked to human-induced climate change, with a high degree of confidence, while others cannot. We review the state of the science relevant to event attribution with a focus on Australia. We highlight where progress can be made, focusing on observations, physical understanding, and realistic climate modelling.

Abstract: This study aimed to estimate associations between weight control status (trying to lose, gain or maintain weight) and lifestyle behaviors (moderate-to-vigorous physical activity (MVPA), screen time, and the consumption of breakfast, fast food, fruits and vegetables, and sugar-sweetened beverages (SSB)) in adolescents.Data from 919 adolescents in the MATCH study, in New Brunswick, Canada, who self-reported their weight control status at least once within 24 data collection cycles over 8 years (from 2011 to 2019) and from 812 who provided data at least once over the 7 cycles on eating behaviors were used. Generalized estimating equations were used.At the first cycle, mean age was 11.3 (SD = 1.2) years old and 56% were girls. Trying to gain (β = 0.47, CI = [0.15, 0.79]) and maintain weight (β = 0.35, CI = [0.12, 0.57]) were positively associated with MVPA. Trying to lose weight was negatively associated with breakfast (IRR = 0.90, CI = [0.85, 0.94]) and positively associated with screen time (β = 0.62, CI = [0.15, 1.10]), fruit and vegetable (IRR = 1.12, CI = [1.01, 1.25]) and SSB (IRR = 1.42, CI = [1.10, 1.84]). Changes from one weight control status to trying to lose weight were associated with increases in fast food consumption (β = 0.49, CI = [0.15, 0.84]).Weight control status was associated with healthy and unhealthy behaviors in adolescents. Trying to gain or maintain weight was generally associated with more favorable health-related behaviors. Education on healthy weight management behaviors is needed to improve adolescents’ health.

Abstract: Abstract. Objectives:. Patient engagement in the design and implementation of clinical trials is necessary to ensure that the research is relevant and responsive to patients. The PREP-IT trials, which include 2 pragmatic trials that evaluate different surgical preparation solutions in orthopaedic trauma patients, followed the patient-centered outcomes research (PCOR) methodology throughout the design, implementation, and conduct. We conducted a substudy within the PREP-IT trials to explore participants' experiences with trial participation. Methods:. At the final follow-up visit (12 months after their fracture), patients participating in the PREP-IT trials were invited to participate in the substudy. After providing informed consent, participants completed a questionnaire that asked about their experience and satisfaction with participating in the PREP-IT trials. Descriptive statistics are used to report the findings. Results:. Four hundred two participants participated in the substudy. Most participants (394 [98%]) reported a positive experience, and 376 (94%) participants felt their contributions were appreciated. The primary reasons for participation were helping future patients with fracture (279 [69%]) and to contribute to science (223 [56%]). Two hundred seventeen (46%) participants indicated that their decision to participate was influenced by the minimal time commitment. Conclusions:. Most participants reported a positive experience with participating in the PREP-IT trials. Altruism was the largest motivator for participating in this research. Approximately half of the participants indicated that the pragmatic, low-participant burden design of the trial influenced their decision to participate. Meaningful patient engagement, a pragmatic, and low-burden protocol led to high levels of participant satisfaction.

183. Development of the Nova Scotia Potential Donor Audit (PDA) Tool and 2020 Historic Performance Database: Lessons Learned From the First 1000 Medical Record Reviews

Abstract: Background:. Legislation and accountability frameworks are key components of high-performing deceased-donation systems. In 2021, Nova Scotia (NS), Canada, became the first jurisdiction in North America to enact deemed consent legislation and concurrently implemented mandatory referral legislation similar to that found in other Canadian provinces. Frontline financial resources were provided by the government to support the development of program infrastructure, including implementation of means to evaluate system performance. Methods:. The Organ Donation Program (ODP), in collaboration with other stakeholders, developed a Potential Donor Audit (PDA) tool and database for referral intake and manual performance audits. Medical record reviews of deaths in the year before legislative change were conducted to pilot and revise the PDA and evaluate missed donation opportunities. Results:. The NS PDA was piloted on 1028 patient deaths. Of 518 patients (50.4%) who met clinical triggers for referral to the ODP, 72 (13.9%) were referred (86.1% missed referral rate). One hundred sixty-three patients met the NS definition of a potential donor; 53 (32.5%) were referred (110 missed potential donors). Referral consent rates reached 71.7% (n = 38 of 53 approaches). The actualized donation rate reported by Canadian Blood Services was 29.9 donors per million population (n = 34 donors). Discussion:. We documented high rates of missed referrals and missed potential donors before the enactment of mandatory referral and deemed consent legislation. Conclusions:. The ODP has intentionally broadened clinical criteria for referral to shift the responsibility of identifying medically suitable potential donors from bedside clinicians to organ donation specialists. Lessons learned from our experience developing a PDA include the importance of early involvement of multiple stakeholders and ongoing modification of fields and workflow based on data availability and utility for clinical, educational, research, and reporting purposes.

Abstract: Summary: Genomic and transcriptomic analysis has furthered our understanding of many tumors. Yet, thyroid cancer management is largely guided by staging and histology, with few molecular prognostic and treatment biomarkers. Here, we utilize a large cohort of 251 patients with 312 samples from two tertiary medical centers and perform DNA/RNA sequencing, spatial transcriptomics, and multiplex immunofluorescence to identify biomarkers of aggressive thyroid malignancy. We identify high-risk mutations and discover a unique molecular signature of aggressive disease, the Molecular Aggression and Prediction (MAP) score, which provides improved prognostication over high-risk mutations alone. The MAP score is enriched for genes involved in epithelial de-differentiation, cellular division, and the tumor microenvironment. The MAP score also identifies aggressive tumors with lymphocyte-rich stroma that may benefit from immunotherapy. Future clinical profiling of the stromal microenvironment of thyroid cancer could improve prognostication, inform immunotherapy, and support development of novel therapeutics for thyroid cancer and other stroma-rich tumors.

Abstract: BackgroundPediatric emergency departments (ED) in many countries are implementing electronic tools such as kiosks, mobile apps, and electronic patient portals, to improve the effectiveness of discharge communication. ObjectiveThis study aimed to survey nurse and physician readiness to adopt these tools. MethodsAn electronic, cross-sectional survey was distributed to a convenience sample of currently practicing ED nurses and physicians affiliated with national pediatric research organizations in Canada, Australia, and New Zealand. Survey development was informed by the nonadoption, abandonment, scale-up, spread, sustainability framework. Measures of central tendency, and parametric and nonparametric tests were used to describe and compare nurse and physician responses. ResultsOut of the 270 participants, the majority were physicians (61%, 164/270), female (65%, 176/270), and had 5 or more years of ED experience (76%, 205/270). There were high levels of consensus related to the value proposition of electronic discharge communication tools (EDCTs) with 82% (221/270) of them agreeing that they help parents and patients with comprehension and recall. Lower levels of consensus were observed for organizational factors with only 37% (100/270) agreeing that their staff is equipped to handle challenges with communication technologies. Nurses and physicians showed significant differences on 3 out of 21 readiness factors. Abstract: PSR B1259-63/LS 2883 is a gamma-ray binary system consisting of a pulsar in an eccentric orbit around a bright Oe stellar-type companion star that features a dense circumstellar disc. The high- and very-high-energy (HE, VHE) gamma-ray emission from PSR B1259-63/LS 2883 around the times of its periastron passage are characterised, in particular, at the time of the HE gamma-ray flares reported to have occurred in 2011, 2014, and 2017. Spectra and light curves were derived from observations conducted with the H.E.S.S.-II array in 2014 and 2017. A local double-peak profile with asymmetric peaks in the VHE light curve is measured, with a flux minimum at the time of periastron $t_p$ and two peaks coinciding with the times at which the neutron star crosses the companion's circumstellar disc ($\sim t_p \pm 16$ d). A high VHE gamma-ray flux is also observed at the times of the HE gamma-ray flares ($\sim t_p + 30$ d) and at phases before the first disc crossing ($\sim t_p - 35$ d). PSR B1259-63/LS 2883 displays periodic flux variability at VHE gamma-rays without clear signatures of super-orbital modulation in the time span covered by H.E.S.S. observations. In contrast, the photon index of the measured power-law spectra remains unchanged within uncertainties for about 200 d around periastron. Lower limits on exponential cut-off energies up to $\sim 40$ TeV are placed. At HE gamma-rays, PSR B1259-63/LS 2883 has now been detected also before and after periastron, close to the disc crossing times. Repetitive flares with distinct variability patterns are detected in this energy range. Such outbursts are not observed at VHEs, although a relatively high emission level is measured. Abstract: Gamma-ray bursts (GRBs) are brief flashes of gamma rays, considered to be the most energetic explosive phenomena in the Universe. The emission from GRBs comprises a short (typically tens of seconds) and bright prompt emission, followed by a much longer afterglow phase. During the afterglow phase, the shocked outflow -- produced by the interaction between the ejected matter and the circumburst medium -- slows down, and a gradual decrease in brightness is observed. GRBs typically emit most of their energy via gamma-rays with energies in the kiloelectronvolt-to-megaelectronvolt range, but a few photons with energies of tens of gigaelectronvolts have been detected by space-based instruments. However, the origins of such high-energy (above one gigaelectronvolt) photons and the presence of very-high-energy (more than 100 gigaelectronvolts) emission have remained elussive. Here we report observations of very-high-energy emission in the bright GRB 180720B deep in the GRB afterglow -ten hours after the end of the prompt emission phase, when the X-ray flux had already decayed by four orders of magnitude. Two possible explanations exist for the observed radiation: inverse Compton emission and synchrotron emission of ultrarelativistic electrons. Our observations show that the energy fluxes in the X-ray and gamma-ray range and their photon indices remain comparable to each other throughout the afterglow. This discovery places distinct constraints on the GRB environment for both emission mechanisms, with the inverse Compton explanation alleviating the particle energy requirements for the emission observed at late times. Abstract: Flat-spectrum radio-quasars (FSRQs) are rarely detected at very-high-energies (VHE; E>100 GeV) due to their low-frequency-peaked SEDs. At present, only 6 FSRQs are known to emit VHE photons, representing only 7% of the VHE extragalactic catalog. Following the detection of MeV-GeV gamma-ray flaring activity from the FSRQ PKS 0736+017 (z=0.189) with Fermi, the H.E.S.S. array of Cherenkov telescopes triggered ToO observations on February 18, 2015, with the goal of studying the gamma-ray emission in the VHE band. H.E.S.S. ToO observations were carried out during the nights of February 18, 19, 21, and 24, 2015. Together with Fermi-LAT, the multi-wavelength coverage of the flare includes Swift observations in soft-X-rays and optical/UV, and optical monitoring (photometry and spectro-polarimetry) by the Steward Observatory, the ATOM, the KAIT and the ASAS-SN telescope. VHE emission from PKS 0736+017 was detected with H.E.S.S. during the night of February 19, 2015, only. Fermi data indicate the presence of a gamma-ray flare, peaking at the time of the H.E.S.S. detection, with a flux doubling time-scale of around six hours. The gamma-ray flare was accompanied by at least a 1 mag brightening of the non-thermal optical continuum. No simultaneous observations at longer wavelengths are available for the night of the H.E.S.S. detection. The gamma-ray observations with H.E.S.S. and Fermi are used to put constraints on the location of the gamma-ray emitting region during the flare: it is constrained to be just outside the radius of the broad-line-region with a bulk Lorentz factor $\simeq 20$, or at the level of the radius of the dusty torus with Gamma > 60. PKS 0736+017 is the seventh FSRQ known to emit VHE photons and, at z=0.189, is the nearest so far. Abstract: The Crab nebula is one of the most studied cosmic particle accelerators, shining brightly across the entire electromagnetic spectrum up to very high-energy gamma rays. It is known from radio to gamma-ray observations that the nebula is powered by a pulsar, which converts most of its rotational energy losses into a highly relativistic outflow. This outflow powers a pulsar wind nebula (PWN), a region of up to 10~light-years across, filled with relativistic electrons and positrons. These particles emit synchrotron photons in the ambient magnetic field and produce very high-energy gamma rays by Compton up-scattering of ambient low-energy photons. While the synchrotron morphology of the nebula is well established, it was up to now not known in which region the very high-energy gamma rays are emitted. Here we report that the Crab nebula has an angular extension at gamma-ray energies of 52 arcseconds (assuming a Gaussian source width), significantly larger than at X-ray energies. This result closes a gap in the multi-wavelength coverage of the nebula, revealing the emission region of the highest energy gamma rays. These gamma rays are a new probe of a previously inaccessible electron and positron energy range. We find that simulations of the electromagnetic emission reproduce our new measurement, providing a non-trivial test of our understanding of particle acceleration in the Crab nebula., Comment: 23 pages, 5 figures

Abstract: The flat spectrum radio quasar 3C 279 is known to exhibit pronounced variability in the high-energy ($100\,$MeV$100\,$GeV) $\gamma$-ray domain. While the observation in 2014 provides an upper limit, the observation in 2015 results in a signal with $8.7\,\sigma$ significance above an energy threshold of $66\,$GeV. No VHE variability has been detected during the 2015 observations. The VHE photon spectrum is soft and described by a power-law index of $4.2\pm 0.3$. The H.E.S.S. data along with a detailed and contemporaneous multiwavelength data set provide constraints on the physical parameters of the emission region. The minimum distance of the emission region from the central black hole is estimated using two plausible geometries of the broad-line region and three potential intrinsic spectra. The emission region is confidently placed at $r\gtrsim 1.7\times10^{17}\,$cm from the black hole, i.e., beyond the assumed distance of the broad-line region. Time-dependent leptonic and lepto-hadronic one-zone models are used to describe the evolution of the 2015 flare. Neither model can fully reproduce the observations, despite testing various parameter sets. Abstract: Young core-collapse supernovae with dense-wind progenitors may be able to accelerate cosmic-ray hadrons beyond the knee of the cosmic-ray spectrum, and this may result in measurable gamma-ray emission. We searched for gamma-ray emission from ten supernovae observed with the High Energy Stereoscopic System (H.E.S.S.) within a year of the supernova event. Nine supernovae were observed serendipitously in the H.E.S.S. data collected between December 2003 and December 2014, with exposure times ranging from 1.4 hours to 53 hours. In addition we observed SN 2016adj as a target of opportunity in February 2016 for 13 hours. No significant gamma-ray emission has been detected for any of the objects, and upper limits on the $>1$ TeV gamma-ray flux of the order of $\sim$10$^{-13}$ cm$^{-2}$s$^{-1}$ are established, corresponding to upper limits on the luminosities in the range $\sim$2 $\times$ 10$^{39}$ erg s$^{-1}$ to $\sim$1 $\times$ 10$^{42}$ erg s$^{-1}$. These values are used to place model-dependent constraints on the mass-loss rates of the progenitor stars, implying upper limits between $\sim$2 $\times 10^{-5}$ and $\sim$2 $\times 10^{-3}$M$_{\odot}$yr$^{-1}$ under reasonable assumptions on the particle acceleration parameters., Comment: Accepted for publication in A&A

Abstract: PKS 1830-211 is a known macrolensed quasar located at a redshift of z=2.5. Its high-energy gamma-ray emission has been detected with the Fermi-LAT instrument and evidence for lensing was obtained by several authors from its high-energy data. Observations of PKS 1830-211 were taken with the H.E.S.S. array of Imaging Atmospheric Cherenkov Telescopes in August 2014, following a flare alert by the Fermi- LAT collaboration. The H.E.S.S observations were aimed at detecting a gamma-ray flare delayed by 20-27 days from the alert flare, as expected from observations at other wavelengths. More than twelve hours of good quality data were taken with an analysis threshold of $\sim67$ GeV. The significance of a potential signal is computed as a function of the date as well as the average significance over the whole period. Data are compared to simultaneous observations by Fermi-LAT. No photon excess or significant signal is detected. An upper limit on PKS 1830-211 flux above 67 GeV is computed and compared to the extrapolation of the Fermi-LAT flare spectrum., Comment: Accepted for publication in MNRAS

Abstract: The blazar Mrk 501 (z=0.034) was observed at very-high-energy (VHE, $E\gtrsim 100$~GeV) gamma-ray wavelengths during a bright flare on the night of 2014 June 23-24 (MJD 56832) with the H.E.S.S. phase-II array of Cherenkov telescopes. Data taken that night by H.E.S.S. at large zenith angle reveal an exceptional number of gamma-ray photons at multi-TeV energies, with rapid flux variability and an energy coverage extending significantly up to 20 TeV. This data set is used to constrain Lorentz invariance violation (LIV) using two independent channels: a temporal approach considers the possibility of an energy dependence in the arrival time of gamma rays, whereas a spectral approach considers the possibility of modifications to the interaction of VHE gamma rays with extragalactic background light (EBL) photons. The non-detection of energy-dependent time delays and the non-observation of deviations between the measured spectrum and that of a supposed power-law intrinsic spectrum with standard EBL attenuation are used independently to derive strong constraints on the energy scale of LIV ($E_{\rm{QG}}$) in the subluminal scenario for linear and quadratic perturbations in the dispersion relation of photons. For the case of linear perturbations, the 95% confidence level limits obtained are $E_{\rm{QG},1} > 3.6 \times 10^{17} \ \rm{GeV} $ using the temporal approach and $E_{\rm{QG},1} > 2.6 \times 10^{19} \ \rm{GeV}$ using the spectral approach. For the case of quadratic perturbations, the limits obtained are $E_{\rm{QG},2} > 8.5 \times 10^{10} \ \rm{GeV} $ using the temporal approach and $E_{\rm{QG},2} > 7.8 \times 10^{11} \rm{ GeV}$ using the spectral approach., Comment: Published in ApJ

Abstract: The H.E.S.S. experiment in Namibia is a high-energy gamma-ray telescope sensitive in the energy range from 30 GeV to a several tens of TeV, that uses the atmospheric Cherenkov technique to detect showers developed within the atmosphere. The elastic lidar, installed on the H.E.S.S. site, allows to reduce the systematic errors related to the atmospheric composition uncertainties thanks to the estimation of the extinction profile for the Cherenkov light (300-650 nm). The latter has a direct impact on the reconstructed parameters, such as the photon energy and the source flux. In this paper we report on physics results obtained on the Crab Nebula spectrum using the lidar profiles obtained at the H.E.S.S. site.

Abstract: Growing tumors are dynamic and nonlinear ecosystems, wherein cancer cells adapt to their local microenvironment, and these adaptations further modify the environment, inducing more changes. From nascent intraductal neoplasms to disseminated metastatic disease, several levels of evolutionary adaptations and selections occur. Here, we focus on one example of such an adaptation mechanism, namely, "niche construction" promoted by adaptation to acidosis, which is a metabolic adaptation to the early harsh environment in intraductal neoplasms. The avascular characteristics of ductal carcinoma in situ (DCIS) make the periluminal volume profoundly acidic, and cancer cells must adapt to this to survive. Based on discovery proteomics, we hypothesized that a component of acid adaptation involves production of collagen by pre-cancer cells that remodels the extracellular matrix (ECM) and stabilizes cells under acid stress. The proteomic data were surprising as collagen production and deposition are commonly believed to be the responsibility of mesenchymally derived fibroblasts, and not cells of epithelial origin. Subsequent experiments in 3D culture, spinning disk and second harmonic generation microscopy of DCIS lesions in patients' samples are concordant. Collagen production assay by acid-adapted cells in vitro demonstrated that the mechanism of induction involves the RAS and SMAD pathways. Secretome analyses show upregulation of ECM remodeling enzymes such as TGM2 and LOXL2 that are collagen crosslinkers. These data strongly indicate that acidosis in incipient cancers induces collagen production by cancer cells and support the hypothesis that this adaptation initiates a tumor-permissive microenvironment promoting survival and growth of nascent cancers.

Abstract: Experience influences behavior, but little is known about how experience is encoded in the brain, and how changes in neural activity are implemented at a network level to improve performance. Here we investigate how differences in experience impact brain circuitry and behavior in larval zebrafish prey capture. We find that experience of live prey compared to inert food increases capture success by boosting capture initiation. In response to live prey, animals with and without prior experience of live prey show activity in visual areas (pretectum and optic tectum) and motor areas (cerebellum and hindbrain), with similar visual area retinotopic maps of prey position. However, prey-experienced animals more readily initiate capture in response to visual area activity and have greater visually-evoked activity in two forebrain areas: the telencephalon and habenula. Consequently, disruption of habenular neurons reduces capture performance in prey-experienced fish. Together, our results suggest that experience of prey strengthens prey-associated visual drive to the forebrain, and that this lowers the threshold for prey-associated visual activity to trigger activity in motor areas, thereby improving capture performance.

Abstract: Bathymetry (seafloor depth), is a critical parameter providing the geospatial context for a multitude of marine scientific studies. Since 1997, the International Bathymetric Chart of the Arctic Ocean (IBCAO) has been the authoritative source of bathymetry for the Arctic Ocean. IBCAO has merged its efforts with the Nippon Foundation-GEBCO-Seabed 2030 Project, with the goal of mapping all of the oceans by 2030. Here we present the latest version (IBCAO Ver. 4.0), with more than twice the resolution (200 × 200 m versus 500 × 500 m) and with individual depth soundings constraining three times more area of the Arctic Ocean (∼19.8% versus 6.7%), than the previous IBCAO Ver. 3.0 released in 2012. Modern multibeam bathymetry comprises ∼14.3% in Ver. 4.0 compared to ∼5.4% in Ver. 3.0. Thus, the new IBCAO Ver. 4.0 has substantially more seafloor morphological information that offers new insights into a range of submarine features and processes; for example, the improved portrayal of Greenland fjords better serves predictive modelling of the fate of the Greenland Ice Sheet.

Abstract: PurposeWe investigated the value of transcriptome sequencing (RNAseq) in ascertaining the consequence of DNA variants on RNA transcripts to improve the diagnostic rate from exome or genome sequencing for undiagnosed Mendelian diseases spanning a wide spectrum of clinical indications.MethodsFrom 234 subjects referred to the Undiagnosed Diseases Network, University of California-Los Angeles clinical site between July 2014 and August 2018, 113 were enrolled for high likelihood of having rare undiagnosed, suspected genetic conditions despite thorough prior clinical evaluation. Exome or genome sequencing and RNAseq were performed, and RNAseq data was integrated with genome sequencing data for DNA variant interpretation genome-wide.ResultsThe molecular diagnostic rate by exome or genome sequencing was 31%. Integration of RNAseq with genome sequencing resulted in an additional seven cases with clear diagnosis of a known genetic disease. Thus, the overall molecular diagnostic rate was 38%, and 18% of all genetic diagnoses returned required RNAseq to determine variant causality.ConclusionIn this rare disease cohort with a wide spectrum of undiagnosed, suspected genetic conditions, RNAseq analysis increased the molecular diagnostic rate above that possible with genome sequencing analysis alone even without availability of the most appropriate tissue type to assess.
