Abstract Brassicaceae plants have the glucosinolate–myrosinase defense system, jointly active against herbivory. However, constitutive glucosinolate (GLS) defense is observed to occur at levels that do not deter all insects from feeding. That prompts the question of why Brassicaceae plants have not evolved a higher constitutive defense. The answer may lie in the contrasting relationship between plant defense and host plant preference of specialist and generalist herbivores. GLS content increases a plant's susceptibility to specialist insects. In contrast, generalists are deterred by the plant GLSs. Although GLSs can attract the natural enemies (predators and parasitoids) of these herbivores, enemies can reduce herbivore pressure to some extent only. So, plants can be overrun by specialists if GLS content is too high, whereas generalists can invade the plants if it is too low. Therefore, an optimal constitutive plant defense can minimize the overall herbivore pressure. To explain the optimal defense theoretically, we model the contrasting host selection behavior of insect herbivores and the emergence of their natural enemies by non‐autonomous ordinary differential equations, where the independent variable is the plant GLS concentration. From the model, we quantify the optimal amount of GLSs, which minimizes total herbivore (specialists and generalists) pressure. That quite successfully explains the evolution of constitutive defense in plants from the perspective of optimality theory.

This study aimed to determine whether leaf extracts from seven Eruca vesicaria subsp. sativa cultivars and their biochemically active compounds (glucosinolates and downstream-derived products) inhibit mycelia growth of three well-known pathogenic oomycetes, Phytopythium chamaehyphon, Phytopythium vexans and Phytophthora citrophthora; being the most significant in the development of Kiwifruit Vine Decline Syndrome (KVDS). Leaf extract quantity of 10, 20 and 30 mg were inoculated in Petri dish (90 mm Ø, each 22 mL of liquid medium – Potato Dextrose Agar), for in vitro bioassays. A pathogen plug was placed in the centre of each plate and the Oomycota colony perimeter was marked 5 days after inoculation. Radial colony growth was measured from 4 marks per plate 5, 10, and 15 days after inoculation, further elaborated with Image J software image analysis. Growth rates for all strains were inhibited by around 67% after 15 days. This was most pronounced when applying the highest concentration of leaf extract. By using Liquid Chromatography Mass Spectrometry (LC-MS) and Gas Chromatography Mass Spectrometry (GC-MS), fifteen glucosinolate compounds, of which glucosativin was found in the highest quantity, were identified. Concentrations of hydrolysis products produced by leaves (erucin and sativin) were also investigated, and were significantly associated with colony radial growth, especially towards Pp. chamaehyphon and Pp. vexans. Three downstream products of glucosinolates (two pure isothiocyanates, AITC and PEITC; and one indole I3C; all commonly present in Brassicaceae) were also tested, and a statistically significant inhibition of growth was observed at the highest concentration (0.6 µL).

The impact of duckweed extracts (DEs) on the shelf-life of packaged beef burgers was evaluated through classical assays and untargeted metabolomics. Beef burgers were formulated with an antioxidants-free control (CON), 1 g/kg sodium ascorbate (ASC), and increasing levels of a DEs, namely 1 (DE1), 5 (DE5), and 10 (DE10) g/kg, packaged under modified atmosphere and stored at 4 °C for 19 days. The DEs, abundant in phytochemicals, determined no issues with the hygienic status of the product. DEs modulated the redox status, being ineffective in preserving linolenic acid from peroxidation. However, the oxidation marker 2-nonenoic acid was down-accumulated in the DE10 sample following 19 days of storage, recording a lower glutathione:glutathione disulfide ratio. The accumulation of adipate semialdehyde revealed the inefficiency of DEs in coping with protein oxidation, while DEs prevented the accumulation of biogenic amines. Therefore, this work suggests a potential pro-oxidant role of the formulated DEs.

Rocket (Eruca vesicaria subsp. sativa) is a source of sulfur-containing glucosinolates (GSLs). GSLs and their breakdown hydrolysis products (GHPs) are responsible for health-related benefits, such as anti-cancer and anti-neurodegenerative properties. Understanding how phytochemical composition changes between cultivation environments is key to developing cultivars with improved nutritional quality. Two consecutive harvests (first and second regrowth) of crops, grown in both Italy and the UK, were used to determine the phytochemical and transcriptomic differences between six lines of Eruca. Samples were taken upon delivery from field sites (D0) and after five days of cold storage (D5) for each location. Leaves were analysed for sulfur content, volatile organic compounds (VOCs), GSLs, GHPs, and sugars. Transcriptome data were associated with metabolite profiles to identify differentially expressed genes between plants grown in the two environments. VOC compounds (carbon disulfide, methyl thiocyanate) were associated with growth environment and with differences in sulfur metabolism gene expression (APR2, LSU2, LSU3, SDI1, SiR), GSL biosynthesis (MYB28, FMOGS-OX2) and GHP formation (ESM1, TGG1, TGG2). The concentrations of sugars were an order of magnitude greater in UK grown samples (up to 29.9 mg g-1 dry weight; dw). Sulfur content was significantly higher in the Italy plant samples (11.4 – 20.1 mg g-1 dw), which was in turn associated with higher concentrations of GSLs (pentyl GSL, up to 15.8 μmol g-1 dw; sinigrin, up to 0.005 μmol g-1 dw; glucoraphanin, up to 5.1 μmol g-1 dw; glucorucolamine, up to 23.6 μmol g-1 dw; neoglucobrassicin, up to 5.3 μmol g-1 dw) and hydrolysis products (sativin, up to 13.5 μmol g-1 dw; erucin, up to 1 μmol g-1 dw; sulforaphane, up to 34.7 μmol g-1 dw). VOC profiles of plants cultivated in the UK were distinct from Italy grown plants, with higher relative abundances of alkanes and esters in second cut and shelf-life (D5) samples. The data indicate a significant interaction of cultivar response with environment, highlighting the difficulty of producing Eruca crops with consistent phytochemical and postharvest traits. Genes with differential expression between plants grown in Italy and the UK could be used as markers of phytochemical quality and composition.

Growing microgreens on trays without substrate in a vertical multilayered growing unit offers several advantages over traditional agriculture methods. This study investigated the yield performance and nutritional quality of five selections of radish microgreens grown in sprouting trays, without a substrate using only water, in an indoor multilayer cultivation system using artificial light. Various parameters were measured, including fresh weight, dry matter, chlorophyll, minerals, amino acids, phenolics, flavonoids, anthocyanins, vitamin C, glucosinolates, and antioxidant activity with four different in vitro assays. After ten days, the biomass had increased by 6-10 times, and the dry matter varied from 4.75-7.65%. The highest yield was obtained from ‘Asia red’, while the lowest was from ‘Koregon red’. However, ‘Koregon red’ and ‘Asia red’ had the highest dry matter. ‘Asia red’ was found to have the highest levels of both Chls and vitamin C compared to the other cultivars, while ‘Koregon red’ exhibited the highest levels of total phenolics and flavonoids. Although variations in the levels of individual glucosinolates were observed, there were no significant differences in the total content of glucosinolates among the five cultivars. ‘Asia purple’ had the highest anthocyanin content, while ‘Asia green 2’ had the lowest. The K, Mg, and Na concentrations were significantly highest in ‘Asia green 2’, and the highest Ca was recorded in ‘Asia purple’. Overall, ‘Asia purple’ and ‘Koregon red’ were the best cultivars in terms of nutritional quality among the tested radish microgreens. These cultivars exhibited high levels of dry weight, total phenolics, flavonoids, anthocyanins, essential and total amino acids, and antioxidant activities. Moreover, the implementation of this vertical cultivation method for microgreens, which relies solely on water and seeds known for their tall shoots during the sprouting could hold promise as a sustainable approach. This method can effectively be utilized for cultivar screening and fulfilling the nutritional and functional needs of the population while minimizing the environmental impacts associated with traditional agriculture practices. Red and white cabbages (Brassica oleracea var. capitata f. alba and rubra, respectively) are two of the most commercially valued vegetables in crucifers, well-recognized for their unique sensory and nutritive attributes in addition to a myriad of health-promoting benefits. The current review addressed the differential qualitative/quantitative phytochemical make-ups for the first time for better utilization as nutraceuticals and to identify potential uses based on the chemical makeup of both cultivars (cvs.). In addition, extraction methods are compared highlighting their advantages and/or limitations with regards to improving yield and stability of cabbage bioactives, especially glucosinolates. Besides, the review recapitulated detailed action mechanism and safety of cabbage bioactives, as well as processing technologies to further improve their effects are posed as future perspectives. White and red cabbage cvs. revealed different GLSs profile which affected by food processing, including enzymatic hydrolysis, thermal breakdown, and leaching. In addition, the red cultivar provides high quality pigment for industrial applications. Moreover, non-conventional modern extraction techniques showed promising techniques for the recovery of their bioactive constituents compared to solvent extraction. All these findings pose white and red cabbages as potential candidates for inclusion in nutraceuticals and/or to be commercialized as functional foods prepared in different culinary forms. In August 2020, the Eruca sativa cultivar "Rucola della Piana del Sele" obtained from the European Union the prestigious PGI (protected geographical indication) label, which certifies the uniqueness of its characteristics and increases its prestige both nationally and, above all, internationally. This plant is recognized as a product of excellence, with a unique flavor and unmistakable aroma. Therefore, since there are no methods to characterize the PGI product, a metabolomic approach was applied to characterize E. sativa grown in the Piana del Sele and different geographical areas. As E. sativa has very wide cultivation, this study sought to compare the metabolite profiles of rocket grown in Piana del Sele, Bergamo, and Brescia, as well as in Switzerland, making a comparison also with the metabolite profile of E. sativa grown spontaneously. To determine the best procedure to distinguish "Rucola della Piana del Sele" from the others, different extraction procedures were carried out using different solvents and fresh or freeze-dried plant matrices. The different extracts were analyzed by liquid chromatography coupled with high-resolution mass spectrometry experiments, using chemometric analyses to identify biomarker metabolites that characterize the PGI product. The LC-ESI-Q-Exactive-MS/MS profiles of methanol and hydroalcoholic extracts of different cultivars of E. sativa were found to be rich in bioactive compounds such as glucosinolates, glycosylated flavonoids, fatty acids, and lipids. The LCMS data were analyzed by principal component analysis (PCA); the score scatter plot shows significant separation among Eruca samples grown in different geographical areas. In detail, loading the scatter plot revealed Eruca grown in Piana del Sele to be richer than other cultivars in glycosylated quercetin 3,3′,4′-O-triglucoside (7), quercetin-3,4′-O-diglucoside-3′-O-(6-sinapoyl-glucoside) (10), and quercetin diglucoside (30). Finally, considering the biological interest in erucin, the myrosinase product of glucoerucin, the latter was quantified in the extracts by LC-ESI/QTrap/MS/MS using the multiple reaction monitoring (MRM) method; E. sativa from Piana del Sele showed the highest content of glucoerucin. Recent technical and theoretical advances have generated an explosion in the identification of specialized metabolite pathways. In comparison, our understanding of how these pathways are regulated is relatively lagging. This and the relatively young age of specialized metabolite pathways has partly contributed to a default and common paradigm whereby specialized metabolite regulation is theorized as relatively simple with a few key transcription factors and the compounds are non-regulatory end-products. In contrast, studies into model specialized metabolites, such as glucosinolates, are beginning to identify a new understanding whereby specialized metabolites are highly integrated into the plants' core metabolic, physiological, and developmental pathways. This model includes a greatly extended compendium of transcription factors controlling the pathway, key transcription factors that co-evolve with the pathway and simultaneously control core metabolic and developmental components, and finally the compounds themselves evolve regulatory connections to integrate into the plants signaling machinery. In this review, these concepts are illustrated using studies in the glucosinolate pathway within the Brassicales. This suggests that the broader community needs to reconsider how they do or do not integrate specialized metabolism into the regulatory network of their study species. [ABSTRACT FROM AUTHOR]

The experiment was to study the degradation effect of microbial solid-state fermentation of rapeseed cake on fiber and glucosinolates as well as improvement effect of nutrient content. In the experiment, rapeseed cake was used as fermentation material. Bacillus subtilis and Lactobacillus plantarum were used as mixed fermentation strains, and the total change rate ( Y) was used as a comprehensive index to confirm the optimal mixed bacteria ratio and the optimal fermentation conditions. The results showed that the interaction between Bacillus subtilis and Lactobacillus plantarum has a significant impact on the total change rate and the degradation rate of neutral washing fibers ( P<0.05). The best fermentation process was as follows: Inoculation amount 12%, liquid to solid ratio 1.2 mL/g, fermentation temperature 35 ℃ and fermentation time of 96 h. Under these technological conditions, the total rate of change ( Y) was the highest, and the acid-soluble protein of fermented rapesseed cake increased by 120.77%, the content of thioside and neutral detergent fiber decreased by 57.53% and 3.09%, respectively. The study indicates that mixed bacterial solid-state fermentation can increase the acid soluble protein of rapeseed cake, reduce the content of glucosides and fiber, and improve the nutritional value of rapeseed cake. Sulfur is an essential element for plant growth, development and resistance to environmental stresses. Glucosinolates (GSLs), a group of sulfur rich secondary metabolites found in Brassicaceae plants, are known for their defensive properties against pathogens and herbivores. Due to their integration of a large proportion of total sulfur, their biosynthesis and degradation are closely linked to sulfur metabolism. It has been demonstrated that GSLs can be broken down to release sulfur and facilitate the production of other thio-metabolites when the plant is under stress. However, the regulation of this process is still not fully understood. In this study, we constructed two broccoli LSU (low sulfur responsive) gene overexpressing lines, 35S::BoLSU1 and 35S::BoLSU2, to detect changes in GSL metabolism after sulfur deficiency treatment. The results showed that BoLSU1 and BoLSU2 inhibit the biosynthesis of aliphatic GSLs, while also promoting their degradation and increasing the content of glutathione (GSH), leading to the reallocation of sulfur from the GSL pool to other thio-metabolites such as GSH. Furthermore, this regulation of GSL metabolism mediated by BoLSU1 and BoLSU2 is found to be dependent on myrosinases BGLU28 and BGLU30. Mustard is a plant that is eagerly cultivated by farmers, due to its mobility across agroclimatic conditions and its high yield. Mustard seeds have thus become a valuable source of many bioactive ingredients, such as polyunsaturated fatty acids and antioxidants (carotenoids, phenolic compounds, tocopherols). The increasingly popularity of natural ingredients in food creation and other industries has made mustard a material for creating new products. The mustard-infused foods developed here have improved properties over the controls. The most common improvements relate to microbiological safety, longer shelf life, slower lipid oxidation, higher protein content and overall better product acceptability. The high content of erucic acid in its seeds, as well as of glucosinolates, mean that mustard has not been widely used in the human food industry or as animal feed. Mustard varieties with reduced levels of erucic acid and glucosinolates are now available on the market. This literature review characterizes the bioactive ingredients present in mustard seeds and presents the already developed possibilities of using mustard seeds in food and as a health-promoting product. Turnip (Brassica rapa subsp. rapa) is a cruciferous plant cultivated worldwide that serves as a source of nutrients and bioactive compounds. Most turnip studies have focused on a few compounds or on part of the plant. The establishment of a complete chemical profile of different plant parts would facilitate its use for nutritional and medicinal purposes. In the current study, mineral elements, soluble sugars, free amino acids (FAA), total phenols (TP), total flavonoids (TF), and glucosinolates (GS) were quantified in the leaves, stems, and roots. Results were compared for 20 strains of turnip. The outcomes showed significant differences between parts of the plant and strains. The leaves exhibited the highest TF, TP, indispensable FAA, and microelement levels, and they showed a higher GS. Moreover, the stems had a high content of GS and macroelements. Furthermore, the roots showed high levels of free sugars and total FAA. The findings of this work provide the basis for utilizing each part of the turnip plant based on its chemical composition. As a vegetable with high nutritional value, broccoli (Brassica oleracea var. italica) is rich in vitamins, antioxidants and anti-cancer compounds. Glucosinolates (GLs) are one of the important functional components widely found in cruciferous vegetables, and their hydrolysate sulforaphane (SFN) plays a key function in the anti-cancer process. Herein, we revealed that blue light significantly induced the SFN content in broccoli sprouts, and salicylic acid (SA) was involved in this process. We investigated the molecular mechanisms of SFN accumulation with blue light treatment in broccoli sprouts and the relationship between SFN and SA. The results showed that the SFN accumulation in broccoli sprouts was significantly increased under blue light illumination, and the expression of SFN synthesis-related genes was particularly up-regulated by SA under blue light. Moreover, blue light considerably decreased the SA content compared with white light, and this decrease was more suppressed by paclobutrazol (Pac, an inhibitor of SA synthesis). In addition, the transcript level of SFN synthesis-related genes and the activity of myrosinase (MYR) paralleled the trend of SFN accumulation under blue light treatment. Protein glycation can result in the formation of advanced glycation end products (AGEs), which pose a potential health risk due to their association with diabetic complications. Natural products are a source of drugs discovery and the search for potential natural inhibitors of AGEs is of great significance. Glucosinolates (GSLs) mainly from cruciferous plants have potential antioxidant, anti‐inflammatory, and anti‐glycation activities. In this study, the inhibitory activity of GSLs on bovine serum albumin (BSA) along with its mechanism was investigated by virtual screening and various computational simulation techniques. Virtual screening revealed that 174 GSLs were screened using Maestro based on the glide score and 89% of the compounds were found to have potential anti‐glycation ability with the docking scores less than −5 kcal/mol. Molecular docking showed that the top 10 GSLs were bound to the IIA structural domain of BSA. Among them, glucohesperin (1) and 2‐hydroxyethyl glucosinolate (2) had the lowest docking scores of −9.428 and −9.333 kcal/mol, respectively, reflecting their good binding affinity. Molecular dynamics simulations of 1 (ΔG = −43.46 kcal/mol) and 2 (ΔG = −43.71 kcal/mol) revealed that the complexes of these two compounds with proteins had good stability. Further binding site analysis suggested that the mechanism of inhibition of protein glycation by these two active ingredients might be through competitive hydrogen bonding to maintain the structural integrity of the protein, thus inhibiting glycation reaction. Moreover, the ADMET values and CYP450 metabolism prediction data were within the recommended values. Therefore, it can be concluded that 1 and 2 may act as potential anti‐glycation agents. Excessive oxidative stress and inflammatory responses are associated with the development of various diseases, including cancer. Glucosinolates (GSLs) are phytochemicals known for their antioxidant properties, and doubled haploid lines (DHLs) of Brassica rapa with high GSL contents (HGSL) were intentionally developed from two edible subspecies of Brassica rapa: B. rapa subsp. trilocularis and B. rapa subsp. chinensis. The purpose of the present study is to assess the capacity of HGSL DHLs to mitigate oxidative stress and inflammation in lipopolysaccharide (LPS)-stimulated RAW264.7 cells, compared to pak choi as a parental control. Our findings demonstrate that HGSL DH lines effectively suppressed the expression of inducible nitric oxide synthase, leading to the reduced levels of nitric oxide at non-toxic concentrations. Additionally, these lines exhibited scavenging activity against reactive oxygen species and free radicals. The enhanced antioxidant capacity of HGSL DHLs was mechanistically attributed to the upregulation of antioxidant enzymes, such as NADPH quinone oxidoreductase 1 (NQO1), the glutamate–cysteine ligase catalytic subunit (GCLC), and heme oxygenase-1 (HMOX1). Furthermore, we confirmed that these effects were mediated through the nuclear factor erythroid 2-related factor 2 (NRF2) signaling pathway via p38 phosphorylation. Moreover, HGSL DHLs demonstrated inhibitory effects on pro-inflammatory cytokines and signal transducers and activators of transcription 3 (STAT3) phosphorylation. Collectively, our results indicate that HGSL DHLs possess better antioxidant and anti-inflammatory properties compared to the parental control pak choi in LPS-stimulated RAW264.7 cells, suggesting that HGSL DHLs of Brassica rapa could be considered as a beneficial daily vegetable for reducing the risk of inflammation-associated diseases. Determining plant responses to hazardous air pollutants is critical in predicting food security programs and challenges in the future. This study aimed to determine the effects of various ozone levels on plant growth responses (leaf area, dry matter, and number of leaves) and biochemical quality (photopigments and glucosinolates) on Brassica campestris L. ssp. chinensis (Pak-Choi). The experiment was conducted within test chambers under different ozone concentrations (60, 150, and 240 ppb for 2 h/day). Leaf area and dry matter were negatively correlated with increasing ozone concentrations, but the number of leaves was not affected by ozone treatment. Lycopene and chlorophylls also showed the same tendency. Even if the ambient ozone concentration was only elevated for a short time, various glucosnilates (GLS) have been diversely affected. The total aliphatic GLS content was reduced. In contrast, the total indole GLS increased at the highest ozone concentration, and the aromatic GLS significantly increased and then decreased as the ozone concentration level increased. These results provide evidence of the strong effect of ozone stress on the plant quality of Pak-Choi with respect to certain secondary plant metabolites. These findings provide an understanding of elevated ozone effects in urban horticulture sites on the growth and metabolite profiling of Brassica plants. [ABSTRACT FROM AUTHOR]

Among the Brassica oleracea L. crops, kale has gained increased global recognition in recent years as a healthy food item due to its high nutritional value and versatility. Additionally, the diversity of different kale varieties has started to be explored across large latitudes from the Mediterranean to north temperate climates. Specifically, glucosinolates are the predominant phytochemicals found in kale leaves, contributing to the specific taste of this vegetable, and they are affected by environmental factors such as temperature. To date, no study has investigated the effect of chilling on glucosinolate diversity and, thus, the taste in genetically different kale cultivars at the same time. Given the variability of glucosinolates observed among cultivars, we evaluated the impact of acclimation to cold temperatures on glucosinolate levels in curly kale, Lacinato kale, and a feral type using high-performance liquid chromatography coupled with time-of-flight mass spectrometry (HPLC-ESI-qTOF-MS). We targeted the short-term impact (after 12 h) on glucosinolates as well as the longer-term effect (after seven days) of cold acclimation. Our results revealed different molecular patterns regarding the change in glucosinolates in the feral type compared to curly kale and Lacinato-type kale. In the latter ones, primary aliphatic glucosinolates were induced (the glucoraphanin in Lacinato kale increased by more than 200%). The indole glucobrassicin was not significantly affected. Conversely, in the feral type the indole glucobrassicin was reduced by 35% after cold acclimation, whereas aliphatic glucosinolates were hardly affected. The results indicate that both genetic and environmental factors are important for the composition of glucosinolate patterns in kale. In conclusion, to obtain plants with an improved nutritional value, considering both temperature and the choice of cultivar is crucial during kale cultivation. In intensive agricultural production with narrow crop rotations, Fusarium oxysporum, a plant pathogenic fungus causing the wilt disease of pepper, frequently develops an abundant population in the soil, which threatens production profitability. Reducing the inoculum level below a critical threshold is highly demanding because of the long-lasting chlamydospores that could remain in the soil for many years. Soil disinfection with methyl bromide was the most effective way to control this pathogen. However, the ban of methyl bromide uses increased interest in control alternatives. Many studies have shown that glucosinolates, secondary metabolites of plants from the Brassicaceae family, as well as their derived biologically active products, negatively affect plant pathogenic bacteria and fungi, insects, nematodes, and weeds. The aim of this research was to determine if cabbage leaf debris, as a by-product material in cabbage production, could be effectively used in the control of Fusarium wilt in pepper. Fresh cabbage leaves were blended and immediately incorporated in sterile substrate seven days before inoculation of pepper plants at the 5-7 leaf stage. Conventional fungicide captan and biofungicide based on Trichoderma asperellum were used as reference treatments. Studying intraspecific variation in multistress responses is central for predicting and managing the population dynamics of wild plant species under rapid global change. Yet, it remains a challenging goal in this field to integrate knowledge on the complex biochemical underpinnings for the targeted 'non-model' species. Here, we studied divergence in combined drought and heat responses among Northern and Southern European populations of the dune plant Cakile maritima , by combining comprehensive plant phenotyping with metabolic profiling via FT-ICR-MS and UPLC-TQ-MS/MS. We observed pronounced constitutive divergence in growth phenology, leaf functional traits, and defence chemistry (glucosinolates and alkaloids) among population origins. Most importantly, the magnitude of growth reduction under drought was partly weaker in southern plants and associated with divergence in plastic growth responses (leaf abscission) and the modulation of primary and specialized metabolites with known central functions not only in plant abiotic but also in biotic stress responses. Our study indicates that divergent selection has shaped the constitutive and drought-/heat-induced expression of numerous morphological and biochemical functional traits to mediate higher abiotic stress resistance in southern Cakile populations, and highlights that metabolomics can be a powerful tool to explore the underlying mechanisms of local adaptation in 'non-model' species. Non-alcoholic fatty liver disease (NAFLD) is mainly characterized by excessive fat accumulation in the liver. It spans a spectrum of diseases from hepatic steatosis to non-alcoholic steatohepatitis (NASH), fibrosis, cirrhosis, and hepatocellular carcinoma (HCC). Brassica juncea is rich in glucosinolates and has been proven to possess many potential pharmacological properties, including hypoglycemic, anti-oxidation, anti-inflammatory, and anti-carcinogenic activities. This study aims to investigate whether whole-plant Brassica juncea (WBJ) and its glucosinolates extracts (BGE) have hepatoprotective effects against a high-fat diet (HFD)-induced NAFLD and further explore the mechanism underlying this process in vivo and in vitro. WBJ treatment significantly reduced body fat, dyslipidemia, hepatic steatosis, liver injury, and inflammation; WBJ treatment also reversed the antioxidant enzyme activity to attenuate oxidative stress in HFD-fed rat liver. Moreover, WBJ and BGE enhanced the activation of AMPK to reduce SREBPs, fatty acid synthase, and HMG-CoA reductase but increased the expression of CPT-I and PPARα to improve hepatic steatosis. In addition, WBJ and BGE could ameliorate NAFLD by inhibiting TNF-α and NF-κB. Based on the above results, this study demonstrates that WBJ and BGE ameliorate HFD-induced hepatic steatosis and liver injury. Epidemiological evidence from systematic research findings and clinical trials suggests that nutritional and functional dietary isothiocyanates interfere with the molecular cascades of Alzheimer's disease pathogenesis and prevent neurons from functional loss. The aim of this review is to explore the role of glucosinolates derived isothiocyanates in various molecular mechanisms involved in the progression of Alzheimer's disease and their potential in the prevention and treatment of Alzheimer's disease. Insect herbivores frequently encounter plant defense molecules, but the physiological and ecological consequences for their immune systems are not fully understood. The majority of studies attempting to relate levels of plant defensive chemistry to herbivore immune responses have used natural population or species-level variation in plant defensive chemistry. Yet, this potentially confounds the effects of plant defense chemistry with other potential plant trait differences that may affect the expression of herbivore immunity. We used an artificial diet containing known quantities of a plant toxin (4-methylsulfinylbutyl isothiocyanate; 4MSOB-ITC or ITC, a breakdown product of the glucosinolate glucoraphanin upon herbivory) to explicitly explore the effects of a plant toxin on the cellular and humoral immune responses of the generalist herbivore Trichoplusia ni (Lepidoptera: Noctuidae) that frequently feeds on glucosinolate-containing plants. Caterpillars feeding on diets with high concentrations of ITC experienced reduced survivorship and growth rates. High concentrations of ITC suppressed the appearance of several types of hemocytes and melanization activity, which are critical defenses against parasitic Hymenoptera and microbial pathogens. In terms of T. ni humoral immunity, only the antimicrobial peptide (AMP) genes lebocin and gallerimycin were significantly upregulated in caterpillars fed on diets containing high levels of ITC relative to caterpillars that were provided with ITC-free diet. Surprisingly, challenging caterpillars with a non-pathogenic strain of Escherichia coli resulted in the upregulation of the AMP gene cecropin. Feeding on high concentrations of plant toxins hindered caterpillar development, decreased cellular immunity, but conferred mixed effects on humoral immunity. Our findings provide novel insights into the effects of herbivore diet composition on insect performance demonstrating the role of specific plant defense toxins that shape herbivore immunity and trophic interactions. Recent discoveries pointed out the importance of the mutual correlation between timing of environmental stress and plant fitness. However, the internal reshaping of plant growth under daily stress sensing, and their metabolic coordination remain to be investigated. Thus, we studied the connection between time of day, growth and defence to understand how plant fitness is affected by diurnal stress inputs.We examined if simulated herbivory (leaf wounding) in the morning, at midday or the evening differentially influence plant defence state vs growth in three crop species of Brassica: broccoli (Brassica oleraceae), turnip greens (B. rapa) and rapeseed (B. napus).The data revealed that plant's tolerance of wounding stress is diurnally regulated in Brassica crops. Trade‐offs between biomass and investment in glucosinolates (GSL) and phenolics were affected by timing of leaf stress. Negative correlations between biomass and induction of defence compounds were found for plants treeated in the morning and evening. However, the correlations were positive for midday treatment. Interestingly, we revealed a new connection between plant growth and changes in aliphatic GSLs and flavonoids in response to wounding.These data suggest that metabolic stress‐dependent circadian oscillations in leaf defences could be one mechanism conferring a competitive advantage to plants to anticipate daily environmental variations by synchronizing them with growth. The ability of plants to release chemicals that affect the growth of other plants offers potential benefits for weed management and sustainable agriculture. This review explores the use of Camelina sativa as a promising cover crop with weed control potential. Camelina sativa, known for its high oil content and adaptability to diverse climatic conditions, exhibits allelopathic potential by releasing chemical compounds that inhibit weed growth. The crop's vigorous growth and canopy architecture contribute to effective weed suppression, reducing the prevalence and spread of associated pathogens. Furthermore, the chemical compounds released by camelina through the solubilization of compounds from leaves by rain, root exudation, or deriving from microbial-mediated decay of camelina's tissues interfere with the growth of neighbouring plants, indicating allelopathic interactions. The isolation and identification of benzylamine and glucosinolates as allelochemicals in camelina highlight their role in plant–plant interactions. However, the studies carried out on this species are outdated, and it cannot be excluded that other chemicals deriving from the breakdown of the glucosinolates or belonging to other classes of specialized metabolites can be involved in its allelopathic potential. Camelina sativa also demonstrates disease suppression capabilities, with glucosinolates exhibiting fungicidal, nematocidal, and bactericidal activities. Additionally, camelina cover crops have been found to reduce root diseases and enhance growth and yields in corn and soybeans. Salvia hispanica (chia) is a highly nutritious food source and has gained popularity due to its high omega-3 fatty acid content. Red spider mites are a serious problem in the production of S. hispanica. However, no study has been conducted to analyze the defensive response to the infestation of red spider mites in S. hispanica. To elucidate the molecular mechanisms of the defensive response of S. hispanica to red spider mites, we performed a transcriptomic analysis of S. hispanica when infested by red spider mites. In the comparative assessment of leaf transcriptomes, a total of 1743 differentially expressed genes (DEGs) were identified between control and mite-infested S. hispanica. From these, 1208 (69%) transcripts were upregulated and 535 (31%) were downregulated. The DEGs included transcription factors, defense hormones, and secondary metabolites that were either suppressed or activated in response to spider mite herbivory. Gene Ontology (GO) enrichment analysis revealed that plant secondary metabolites, such as glucosinolates, and signaling pathways, including the jasmonic acid signaling pathway, may play an important role in the defense against red spider mites. Glucosinolates and their metabolites from Brassicaceae plants have received widespread attention due to their anti-inflammatory effects. Glucosinolates occurs an "enterohepatic circulation" in the body, and the glucosinolates metabolism mainly happens in the intestine. Glucosinolates can be converted into isothiocyanates by intestinal bacteria, which are active substances with remarkable anti-inflammatory, anti-cancer, anti-obesity and neuroprotective properties. This biotransformation can greatly improve the bioactivities of glucosinolates. However, multiple factors in the environment can affect the biotransformation to isothiocyanates, including acidic pH, ferrous ions and thiocyanate-forming protein. The derivatives of glucosinolates under those conditions are usually nitriles and thiocyanates, which may impair the potential health benefits. In addition, isothiocyanates are extremely unstable because of an active sulfhydryl group, which limits their applications. This study aimed to investigate the influence of different thermal treatment methods (boiling, steaming) on the metabolome and volatilome of Brussels sprouts. Targeted, quantitative analysis of primary metabolites (amino acids and sugars), glucosinolates as well as untargeted analysis of volatile organic compounds (VOCs) were performed. The composition of primary metabolites was affected only by boiling, where the loss of nutrients was observed. Glucosinolate content increased after cooking, which can be explained by the loosening of the plant tissues and its better extractability. Most marked changes were noticed in the volatilome. Among the degradation products of glucosinolates, isothiocyanates were the main chemical group in the raw and boiled vegetable. In contrast, steaming favored the formation of nitriles. Another dominant group of VOCs were aldehydes, which were the most abundant in raw Brussels sprouts and their level significantly decreased after thermal treatment. It is well known that isothiocyanates are substances with widely proven bioactive properties. Therefore, their formation in the glucosinolate degradation process is favorable. It is becoming increasingly challenging to maintain crop yields and quality as the global climate changes. The aim of this study was to determine whether and how the profile of health-promoting and taste-related compounds of radishes changes within a growing season. A total of 16 radish (Raphanus sativus L.) genotypes that are commercially available on the Czech market were assessed by means of chemical analysis. Radishes were cultivated in three independent growing cycles under controlled conditions, and the effects of the genotype and growing cycle, as well as their interactions, on the chemical traits were evaluated. Most of the variability in chemical composition was associated with the growing cycle, which accounted for 51.53% of total variance, followed by the genotype (26% of total variance). The interaction between the growing cycle and genotype explained 22.47% of total variance. The growing cycle had the strongest effect on amino acid profiles. More specifically, the amino acids that are known to contribute to overall taste (glycine, along with glutamic and aspartic acids) showed the highest degree of variation, while the amino acids related to glucosinolate biosynthesis (methionine, isoleucine, tryptophan, and phenylalanine) showed relatively low variability. Toxic breakdown products of young Camelina sativa (L.) Crantz, glucosinolates can eliminate microorganisms in the soil. Since microorganisms are essential for phosphate cycling, only insensitive microorganisms with phosphate-solubilizing activity can improve C. sativa's phosphate supply. In this study, 33P-labeled phosphate, inductively coupled plasma mass spectrometry and pot experiments unveiled that not only Trichoderma viride and Pseudomonas laurentiana used as phosphate-solubilizing inoculants, but also intrinsic soil microorganisms, including Penicillium aurantiogriseum, and the assemblies of root-colonizing microorganisms solubilized as well phosphate from apatite, trigger off competitive behavior between the organisms. Driving factors in the competitiveness are plant and microbial secondary metabolites, while glucosinolates of Camelina and their breakdown products are regarded as key compounds that inhibit the pathogen P. aurantiogriseum, but also seem to impede root colonization of T. viride. On the other hand, fungal diketopiperazine combined with glucosinolates is fatal to Camelina. The results may contribute to explain the contradictory effects of phosphate-solubilizing microorganisms when used as biofertilizers. Sprouts are increasingly present in the human diet, being tasty and healthy foods high in antioxidant compounds. Although there is a body of literature on the sprouting of many plant species, Camelina sativa (L.) Crantz has not yet been studied for this purpose. This study aimed to characterize the main bioactive compounds and antioxidant potential of seeds and sprouts of five different Camelina cultivars (ALBA, CO46, CCE43, JOELLE, and VERA). In particular, the contents of phenolic compounds (PCs), phenolic acids (PAs), and glucosinolates (GLSs) were investigated. PCs, PAs, GLSs, and the antioxidant activity of seeds differed among cultivars and were greatly increased by sprouting. A PCA analysis underlined both the effect of the cultivar (PC2) and the germination (PC1) on the nutritional properties of Camelina. The best nutritional properties of seeds were observed for ALBA and CCE43, while the best nutritional properties of sprouts were recorded for CCE43 and JOELLE, since the latter cultivar showed a greater enhancement in phytochemical content and antioxidant activity with sprouting. Finally, a UHPLC-UV procedure for the analysis of GLSs in Camelina was developed and validated. The need for soil fumigants of natural origin such as glucosinolates (GSLs) has increased due to the general prevention of manmade soil fumigants. GSLs and other phytochemicals (vitamin C and phenols) present in Brassica vegetables such as turnips, arugula, and mustard have antioxidant properties, and hence have important health attributes. The study examined how different soil amendments (chicken manure CM, vermicompost Vermi, horse manure HM, sewage sludge SS, elemental inorganic fertilizer Inorg, organic fertilizer Org, and biochar) impact the concentrations of glucosinolates (GSLs), vitamin C, phenols, and reducing sugars in three varieties of turnips (Purple Top White Globe PTWG, Scarlet Queen Red SQR, and Tokyo Cross TC), arugula, and mustard greens grown under field conditions. The results showed that mustard greens contained higher concentrations of GSLs (974 µg g−1 fresh shoots) than arugula (651 µg g−1 fresh shoots), and the TC variety of turnip had the highest concentrations of GSLs, vitamin C, and sugars. Additionally, amending the soil with SS, CM, and HM significantly increased the vitamin C content in mustard shoots by 82%, 90%, and 31%, respectively, and the total phenols by 77%, 70%, and 36%, respectively, compared to the control treatment. Cabbage (Brassica oleracea L. var. capitata) is an excellent source of glucosinolates (GLS) that could reduce the risk of chronic diseases. The purpose of this study was to investigate the biological traits, pigment contents, color, and GLS content of 13 cabbage varieties. This study found that there were significant differences in the GLS content for various developmental stages of cabbage varieties, and the accumulation of GLS in young leaves was higher than that in mature stages. In most of the samples, the GLS content accumulated in different parts and changed as inner leaf > middle leaf > condensed stem > root. Double haploids of the M18-15 variety may be good candidates for future breeding programs and consumers, due to their high GLS content (ranging from 201.10 to 396.25 nmol mg−1 FW). GLS also act as a defense substance, and the data related to GLS accumulation patterns in different leaf locations and root parts may be useful for understanding leaf defense mechanisms and potential source–sink relationships. Matthiola longipetala subsp. livida is an annual herb in Brassicaceae that has received little attention despite the family's high reputation for health benefits, particularly cancer prevention. In this study, UPLC-HRMS-MS analysis was used for mapping the chemical constituents of different plant parts (i.e., flowers, leaves, and roots). Also, spectral similarity networks via the Global Natural Products Social Molecular Networking (GNPS) were employed to visualize their chemical differences and similarities. Additionally, the cytotoxic activity on HCT-116, HeLa, and HepG2 cell lines was evaluated. Throughout the current analysis, 154 compounds were annotated, with the prevalence of phenolic acids, glucosinolates, flavonol glucosides, lipids, peptides, and others. Predictably, secondary metabolites (phenolic acids, flavonoids, and glucosinolates) were predominant in flowers and leaves, while the roots were characterized by primary metabolites (peptides and fatty acids). Four diacetyl derivatives tentatively assigned as O-acetyl O-malonyl glucoside of quercetin (103), kaempferol (108 and 112), and isorhamnetin (114) were detected for the first time in nature. The flowers and leaves extracts showed significant inhibition of HeLa cell line propagation with LC50 values of 18.1 ± 0.42 and 29.6 ± 0.35 µg/mL, respectively, whereas the flowers extract inhibited HCT-116 with LC50 24.8 ± 0.45 µg/mL, compared to those of Doxorubicin (26.1 ± 0.27 and 37.6 ± 0.21 µg/mL), respectively. Glucosinolates are key defense compounds of plants in Brassicales order, and their accumulation in seeds is essential for the protection of the next generation. Recently, members of the Usually Multiple Amino acids Move In and Out Transporter (UMAMIT) family were shown to be essential for facilitating transport of seed-bound glucosinolates from site of synthesis within the reproductive organ to seeds. Here, we set out to identify amino acid residues responsible for glucosinolate transport activity of the main seed glucosinolate exporter UMAMIT29 in Arabidopsis thaliana. Based on a predicted model of UMAMIT29, we propose that the substrate transporting cavity consists of 51 residues, of which four are highly conserved residues across all the analyzed homologs of UMAMIT29. A comparison of the putative substrate binding site of homologs within the brassicaceous-specific, glucosinolate-transporting clade with the non-brassicaceous-specific, non-glucosinolate-transporting UMAMIT32 clade identified 11 differentially conserved sites. When each of the 11 residues of UMAMIT29 was individually mutated into the corresponding residue in UMAMIT32, five mutant variants (UMAMIT29#V27F, UMAMIT29#M86V, UMAMIT29#L109V, UMAMIT29#Q263S, and UMAMIT29#T267Y) reduced glucosinolate transport activity over 75% compared to wild-type UMAMIT29. This suggests that these residues are key for UMAMIT29-mediated glucosinolate transport activity and thus potential targets for blocking the transport of glucosinolates to the seeds. Glucosinolates (GSLs) and their degradation products in radish confer plant defense, promote human health, and generate pungent flavor. However, the intact GSLs in radish have not been investigated comprehensively yet. Here, an accurate qualitative and quantitative analyses of 15 intact GSLs from radish, including four major GSLs of glucoraphasatin (GRH), glucoerucin (GER), glucoraphenin (GRE), and 4-methoxyglucobrassicin (4MGBS), were conducted using UHPLC-HRMS/MS in combination with UHPLC-QqQ-MS/MS. Simultaneously, three isomers of hexyl GSL, 3-methylpentyl GSL, and 4-methylpentyl GSL were identified in radish. The highest content of GSLs was up to 232.46 mmol/g DW at the 42 DAG stage in the 'SQY' taproot, with an approximately 184.49-fold increase compared to the lowest content in another sample. That the GSLs content in the taproots of two radishes fluctuated in a similar pattern throughout the five vegetative growth stages according to the metabolic profiling, whereas the GSLs content in the '55' leaf steadily decreased over the same period. Additionally, the proposed biosynthetic pathways of radish-specific GSLs were elucidated in this study. Our findings will provide an abundance of qualitative and quantitative data on intact GSLs, as well as a method for detecting GSLs, thus providing direction for the scientific progress and practical utilization of GSLs in radish. Consequently, plants also need systems allowing correct spatiotemporal reactivation of such metabolites, for instance under pathogen or herbivore attack.Via co‐expression analysis with public transcriptomes, we determined that the model legume Medicago truncatula has evolved a two‐component system composed of a β‐glucosidase, denominated G1, and triterpene saponins, which are physically separated from each other in intact cells.G1 expression is root‐specific, stress‐inducible, and coregulated with that of the genes encoding the triterpene saponin biosynthetic enzymes. However, the G1 protein is stored in the nucleolus and is released and united with its typically vacuolar‐stored substrates only upon tissue damage, partly mediated by the surfactant action of the saponins themselves. Subsequently, enzymatic removal of carbohydrate groups from the saponins creates a pool of metabolites with an increased broad‐spectrum antimicrobial activity.The evolution of this defense system benefited from both the intrinsic condensation abilities of the enzyme and the bioactivity properties of its substrates. Moringa oleifera Lam. is a tropical tree that has been traditionally used for food, medicine and industrial purposes. All parts of the tree have been utilized and have biological activities, including anti-inflammatory, antibacterial, antioxidant, antidiabetic, wound healing, cardiovascular, anti-asthmatic, hepatoprotective, protection of the central nervous system and more. The leaves and seeds of moringa are particularly rich in phytochemicals, including glucosinolates and isothiocyanates (ITCs). Glucosinolates are inactive precursors that are hydrolyzed into bioactive ITCs. Isothiocyanates, such as 4-(α-L-rhamnopyranosyloxy) benzyl isothiocyanate (moringa isothiocyanate 1, or MIC-1), are known for their diverse biological activities, including anti-inflammatory, antioxidant and anticancer properties. The unique sugar moiety in moringa's ITCs contributes to its stability and distinct properties compared to other cruciferous vegetables. In vitro models, such as the TIM-1 model and in vivo murine models demonstrate MIC-1's bioaccessibility and bioavailability, with no statistical difference in state. Inflammation, as a defense mechanism, is implicated in a wide range of diseases such as diabetes, cancer, obesity, cardiovascular disease, arthritis and multiple sclerosis. In vivo models were used to evaluate the efficacy of MIC-1 against acute skin inflammation, which inhibited the inflammatory pathway. Cold stress is one of the most common abiotic stresses in leaf B. juncea; however, the metabolic response pattern of GSLs in leaf B. juncea under cold stress has not yet been reported. In the present study, we analyzed the GSLs content of leaf B. juncea under cold stress and found that it increased and subsequently decreased. According to RNA-seq data, genes related to the synthesis of aliphatic GSLs were significantly upregulated following 24 h of cold stress; genes related to the synthesis of indole GSLs were significantly upregulated following 48 h of cold stress; and BjBGLU25 and BjBGLU27 were significantly upregulated. Further analysis of the correlation between transcription factors and GSLs content revealed that MYB, ERF, IQD, and bHLH may be involved in regulating the GSLs response pattern in leaf B. juncea under cold stress. In particular, an unreported transcription factor, BjMYBS3 (BjuVA05G33250), was found to play a possible role in the synthesis of aliphatic GSLs. This study evaluated the effects of inclusion of raw, hydrobarothermally treated and fermented rapeseed cake (RRC, HRC and FRC, respectively) in diets fed to laying hens. Hydrobarothermal treatment decreased the hydroxyglucobrassicin and neoglucobrassicin content of rapeseed cake (RC), whereas fermentation significantly reduced the content of all glucosinolates (GLS) and phytate phosphorus (PP) concentration. Both HRC and FRC increased the serum concentrations of total protein (TP) and globulin (GLB), compared with group C. Group HRC hens had desirably lowest blood triacylglycerol (TAG) levels and aspartate aminotransferase (AST) activity. Group FRC hens were characterized by the lowest alkaline phosphatase (ALP) activity, and the highest concentrations of phosphorus (P) and triiodothyronine (T3). Regardless of its form, RC improved ether extract (EE) digestibility, and decreased dry matter (DM) digestibility and calcium (Ca) retention. In comparison with group C, RRC, HRC and FRC decreased cecal digesta weight and enhanced the activity of bacterial α-galactosidase and ß-galactosidase; HRC also increased ammonia concentration in the ceca and reduced β-xylosidase activity. The activity of α-glucosidase and α-arabinopiranosidase was highest, and the activity of bacterial β-glucuronidase was lowest in the ceca of group FRC hens. The cecal concentrations of acetic acid, propionic acid, and total short-chain fatty acids (SCFA s) were highest in birds fed a diet containing RRC and lowest in group HRC. In conclusion, RC fermentation considerably reduces the concentrations of GLS and PP. A diet containing 20% FRC is more recommended than diets containing RRC and HRC because it exerted a beneficial effect on metabolic parameters and intestinal function in laying hens. Mashua (Tropaeolum tuberosum) is an Andean tuber rich in phenolic compounds and glucosinolates, known for its consumption that allows the prevention and/or reduction of getting diseases. The present research evaluated the effect of different types of post-harvest management (sunny, bleached, and freeze-dried) on glucosinolate content and its relationship with myrosinase activity. It is also important to note that when plant cells are damaged and glucosinolates meet the plant's myrosinase, which hydrolyzes them and degrades them to other compounds. In the present research, the types of glucosinolates, the content of glucosinolates and the activity of myrosinase of the purple variety of fresh raw mashua and in mashua treated by several types of processing were evaluated, including the previous treatment (sunny and unscrewed), the boiling treatment and the microwave treatment, with whitening times of 3, 4, 5 and 6 minutes, being the condition of the tuber at the time of bleaching in whole form and by cutting, to then be subsequently subjected to lyophilization treatments. Five glucosinolatos (GSL) were identified: 5-methylsulfinilpentile, 4-hydroxybenzyl, 4-hydroxy-3-indolylmethyl, benzyl and 4- methoxybenzyl, of which 4-methoxybenzyl glucosinolate is the most abundant, symbolizing 98% and 95% of the total GSL in the purple ecotype. According to the total concentration of glucosinolates found in the studied variety (purple), it must be 74.22 and 110.60 micromoles per gram of dry base (μmol/g dry weight), so mashua can be considered, the edible plant with the highest existing GSL content. The tripeptide glutathione plays important roles in many cell processes, including differentiation, proliferation, and apoptosis; in fact, disorders in glutathione homeostasis are involved both in the etiology and in the progression of several human diseases, including cancer. Natural compounds have been found to modulate glutathione levels and function beyond their role as mere antioxidants. For example, certain compounds can upregulate the expression of glutathione-related enzymes, increase the availability of cysteine, the limiting amino acid for glutathione synthesis, or directly interact with glutathione and modulate its function. These compounds may have therapeutic potential in a variety of disease states where glutathione dysregulation is a contributing factor. On the other hand, flavonoids' potential to deplete glutathione levels could be significant for cancer treatment. Overall, while natural compounds may have potential therapeutic and/or preventive properties and may be able to increase glutathione levels, more research is needed to fully understand their mechanisms of action and their potential benefits for the prevention and treatment of several diseases. In this review, particular emphasis will be placed on phytochemical compounds belonging to the class of polyphenols, terpenoids, and glucosinolates that have an impact on glutathione-related processes, both in physiological and pathological conditions. Background and aimsWe investigate patterns of evolution of genome size across a morphologically and ecologically diverse clade of Brassicaceae, in relation to ecological and life history traits. While numerous hypotheses have been put forward regarding autecological and environmental factors that could favour small vs. large genomes, a challenge in understanding genome size evolution in plants is that many hypothesized selective agents are intercorrelated.MethodsWe contribute genome size estimates for 47 species of Streptanthus Nutt. and close relatives, and take advantage of many data collections for this group to assemble data on climate, life history, soil affinity and composition, geographic range and plant secondary chemistry to identify simultaneous correlates of variation in genome size in an evolutionary framework. We assess models of evolution across clades and use phylogenetically informed analyses as well as model selection and information criteria approaches to identify variables that can best explain genome size variation in this clade.Key resultsWe find differences in genome size and heterogeneity in its rate of evolution across subclades of Streptanthus and close relatives. We show that clade-wide genome size is positively associated with climate seasonality and glucosinolate compounds. Model selection and information criteria approaches identify a best model that includes temperature seasonality and fraction of aliphatic glucosinolates, suggesting a possible role for genome size in climatic adaptation or a role for biotic interactions in shaping the evolution of genome size. We find no evidence supporting hypotheses of life history, range size or soil nutrients as forces shaping genome size in this system.ConclusionsOur findings suggest climate seasonality and biotic interactions as potential forces shaping the evolution of genome size and highlight the importance of evaluating multiple factors in the context of phylogeny to understand the effect of possible selective agents on genome size.

Plants produce diverse metabolites to cope with the challenges presented by complex and ever-changing environments. These challenges drive the diversification of specialized metabolites within and between plant species. However, we are just beginning to understand how frequently new alleles arise controlling specialized metabolite diversity and how the geographic distribution of these alleles may be structured by ecological and demographic pressures. Here, we measure the variation in specialized metabolites across a population of 797 natural Arabidopsis thaliana accessions. We show that a combination of geography, environmental parameters, demography and different genetic processes all combine to influence the specific chemotypes and their distribution. This showed that causal loci in specialized metabolism contain frequent independently generated alleles with patterns suggesting potential within-species convergence. This provides a new perspective about the complexity of the selective forces and mechanisms that shape the generation and distribution of allelic variation that may influence local adaptation.

The research is devoted to the study on adaptation, verification and implementation of the method for determining sinigrin in mustard seeds and products of its processing, as well as in other types of food raw materials of plant origin. The relevance of the research is determined, on the one hand, by the need to control sinigrin in the technological processes of mustard processing, and, on the other hand, by the lack of effective standardized methods for determining sinigrin in the Russian Federation. The aim of the research is to modify and (or) adapt the most effective methods for the identification and quantification of sinigrin, presented in the publications of foreign researchers. The objectives of the study included construction of calibration graphs; modification of the sample preparation technique to reduce the complexity and duration of the analysis while maintaining the required parameters of accuracy and reproducibility; adaptation of the analysis method using high performance liquid chromatography; verification and approbation of the proposed method using real production samples, i.e. mustard seeds.High performance liquid chromatography has been used as the basic method.As a result of the research, the sample preparation technique has been modified. It has made it possible to reduce labor intensity and reduce the time for its implementation while maintaining the required metrological characteristics. The calibration dependence has been determined and a method for the identification and quantitative analysis of sinigrin in raw materials of plant origin using a high-performance liquid chromatograph equipped with a diode array and mass spectrometric detectors proposed. As a result of verification, the metrological characteristics of the proposed method have been determined: the measurement range is from 0.0005 to 10%, the accuracy index is ± 0.01%. The method has been tested on real objects, mustard seeds, which are the main sources of sinigrin. The proposed method for the identification and quantification of sinigrin can be recommended for use in research and production laboratories equipped with the necessary measuring instruments and testing equipment.
