Abstract

Calcineurin inhibitors (CNIs) constitute the backbone of modern acute graft-versus-host disease (aGVHD) prophylaxis regimens but have limited efficacy in the prevention and treatment of chronic GVHD (cGVHD). We investigated the effect of CNIs on immune tolerance after stem cell transplantation with discovery-based single-cell gene expression and T cell receptor (TCR) assays of clonal immunity in tandem with traditional protein-based approaches and preclinical modeling. While cyclosporin and tacrolimus suppressed the clonal expansion of [CD8.sup.+] T cells during GVHD, alloreactive [CD4.sup.+] T cell clusters were preferentially expanded. Moreover, CNIs mediated reversible dose-dependent suppression of T cell activation and all stages of donor T cell exhaustion. Critically, CNIs promoted the expansion of both polyclonal and TCR-specific alloreactive central memory [CD4.sup.+] T cells ([T.sub.CM]) with high self-renewal capacity that mediated cGVHD following drug withdrawal. In contrast to posttransplant cyclophosphamide (PT-Cy), CSA was ineffective in eliminating IL-17A-secreting alloreactive T cell clones that play an important role in the pathogenesis of cGVHD. Collectively, we have shown that, although CNIs attenuate aGVHD, they paradoxically rescue alloantigen-specific [T.sub.CM], especially within the [CD4.sup.+] compartment in lymphoid and GVHD target tissues, thus predisposing patients to cGVHD. Abstract

Dois experimentos foram realizados com o objetivo de determinar as exigências de sódio (Na+) e cloro (Cl-) para frangos de corte na fase pré-inicial (1 a 7 dias de idade) e o melhor balanço eletrolítico da ração. Em cada experimento, 1500 pintos de corte da linhagen Cobb foram distribuídos em um delineamento inteiramente casualizado com seis tratamentos, cinco repetições e 50 aves por unidade experimental. Os tratamentos utilizados nos dois experimentos consistiram de rações com níveis de 0,10; 0,15; 0,20; 0,25; 0,30; e 0,35% de Na+ (Experimento 1) ou Cl- (Experimento 2), cada uma com diferentes valores de balanço eletrolítico. Foram utilizados os modelos quadrático e LRP para a determinação das exigências. No Experimento 1, a exigência de Na+ estimada foi em 0,26% para ganho de peso e 0,29% para conversão alimentar. Não foram observadas diferenças para consumo de ração. O melhor desempenho produtivo foi obtido com balanço eletrolítico de 304 e 319 meq/kg, No Experimento 2, a exigência de Cl- estimada foi de 0,29% para ganho de peso e 0,28% para conversão alimentar. O consumo de ração apresentou comportamento linear. O melhor balanço eletrolítico variou na faixa de 250 a 252 meq/kg.

Abstract

We report the discovery of ten short-period giant planets (TOI-2193A b, TOI-2207 b, TOI-2236 b, TOI-2421 b, TOI-2567 b, TOI-2570 b, TOI-3331 b, TOI-3540A b, TOI-3693 b, TOI-4137 b). All of the planets were identified as planet candidates based on periodic flux dips observed by NASA's Transiting Exoplanet Survey Satellite (TESS). The signals were confirmed to be from transiting planets using ground-based time-series photometry, high angular resolution imaging, and high-resolution spectroscopy coordinated with the TESS Follow-up Observing Program. The ten newly discovered planets orbit relatively bright F and G stars ($G < 12.5$,~$T_\mathrm{eff}$ between 4800 and 6200 K). The planets' orbital periods range from 2 to 10~days, and their masses range from 0.2 to 2.2 Jupiter masses. TOI-2421 b is notable for being a Saturn-mass planet and TOI-2567 b for being a ``sub-Saturn'', with masses of $0.322\pm 0.073$ and $0.195\pm 0.030$ Jupiter masses, respectively. In most cases, we have little information about the orbital eccentricities. Two exceptions are TOI-2207 b, which has an 8-day period and a detectably eccentric orbit ($e = 0.17\pm0.05$), and TOI-3693 b, a 9-day planet for which we can set an upper limit of $e < 0.052$. Abstract

Multi-planet systems are valuable arenas for investigating exoplanet architectures and comparing planetary siblings. TOI-1246 is one such system, with a moderately bright K dwarf ($\rm{V=11.6,~K=9.9}$) and four transiting sub-Neptunes identified by TESS with orbital periods of $4.31~\rm{d},~5.90~\rm{d},~18.66~\rm{d}$, and $~37.92~\rm{d}$. We collected 130 radial velocity observations with Keck/HIRES and TNG/HARPS-N to measure planet masses. We refit the 14 sectors of TESS photometry to refine planet radii ($\rm{2.97 \pm 0.06~R_\oplus},\rm{2.47 \pm 0.08~R_\oplus}, \rm{3.46 \pm 0.09~R_\oplus}$, $\rm{3.72 \pm 0.16~R_\oplus}$), and confirm the four planets. We find that TOI-1246 e is substantially more massive than the three inner planets ($\rm{8.1 \pm 1.1 M_\oplus}$, $\rm{8.8 \pm 1.2 M_\oplus}$, $\rm{5.3 \pm 1.7 M_\oplus}$, $\rm{14.8 \pm 2.3 M_\oplus}$). The two outer planets, TOI-1246 d and TOI-1246 e, lie near to the 2:1 resonance ($\rm{P_{e}/P_{d}=2.03}$) and exhibit transit timing variations. TOI-1246 is one of the brightest four-planet systems, making it amenable for continued observations. It is one of only six systems with measured masses and radii for all four transiting planets. The planet densities range from $\rm{0.70 \pm 0.24}$ to $3.21 \pm 0.44 \rm{g/cm^3}$, implying a range of bulk and atmospheric compositions. We also report a fifth planet candidate found in the RV data with a minimum mass of 25.6 $\pm$ 3.6 $\rm{M_\oplus}$. Abstract

Molecular linkers have emerged as an effective strategy to improve electronic transport properties on solution-processed layered materials via defect functionalization. However, a detailed discussion on the microscopic mechanisms behind the beneficial effects of functionalization is still missing. Here, by first-principles calculations based on density functional theory, we investigate the effects on the electronic properties of interconnected MoS$_2$ model flakes systems upon functionalization with different thiol molecule linkers, namely thiophenol, 1,4-benzenedithiol, 1,2-ethanedithiol, and 1,3-propanedithiol. The bonding of benzene- and ethanedithiol bridging adjacent armchair MoS$_2$ nanoflakes leads to electronic states just above or at the Fermi level, thus forming a molecular channel for electronic transport between flakes. In addition, the molecular linker reduces the potential barrier for thermally activated hopping between neighboring flakes, improving the conductivity as verified in experiments. The comprehension of such mechanisms helps in future developments of solution-processed layered materials for use on 2D electronic devices.

Here, we demonstrate that the expression of the C/G oncogenic fusion protein promotes the transformation of human cord blood hematopoietic stem and progenitor cells (CB HSPCs) in an endothelial cell coculture system that recapitulates the transcriptome, morphology, and immunophenotype of C/G acute myeloid leukemia (AML) and induces highly aggressive leukemia in xenograft models. Interrogating the transcriptome of C/G-CB cells and primary C/G AML identified a library of C/G-fusion-specific genes that are potential targets for therapy. We developed chimeric antigen receptor (CAR) T cells directed against one of the targets, folate receptor α (FOLR1), and demonstrated their preclinical efficacy against C/G AML using in vitro and xenograft models. FOLR1 is also expressed in renal and pulmonary epithelium, raising concerns for toxicity that must be addressed for the clinical application of this therapy. Our findings underscore the role of the endothelial niche in promoting leukemic transformation of C/G-transduced CB HSPCs. Furthermore, this work has broad implications for studies of leukemogenesis applicable to a variety of oncogenic fusion-driven pediatric leukemias, providing a robust and tractable model system to characterize the molecular mechanisms of leukemogenesis and identify biomarkers for disease diagnosis and targets for therapy.

Abstract

Obesity is a global pandemic that is causally linked to many life-threatening diseases. Apart from some rare genetic conditions, the biological drivers of overeating and reduced activity are unclear. Here, we show that neurotensin-expressing neurons in the mouse interstitial nucleus of the posterior limb of the anterior commissure (IPAC), a nucleus of the central extended amygdala, encode dietary preference for unhealthy energy-dense foods. Optogenetic activation of IPACNts neurons promotes obesogenic behaviors, such as hedonic eating, and modulates food preference. Conversely, acute inhibition of IPACNts neurons reduces feeding and decreases hedonic eating. Chronic inactivation of IPACNts neurons recapitulates these effects, reduces preference for sweet, non-caloric tastants and, furthermore, enhances locomotion and energy expenditure; as a result, mice display long-term weight loss and improved metabolic health and are protected from obesity. Thus, the activity of a single neuronal population bidirectionally regulates energy homeostasis. Our findings could lead to new therapeutic strategies to prevent and treat obesity.

Abstract

We report the detection and characterization of the transiting sub-Neptune TOI-1759 b, using photometric time-series from TESS and near infrared spectropolarimetric data from SPIRou on the CFHT. TOI-1759 b orbits a moderately active M0V star with an orbital period of $18.849975\pm0.000006$ d, and we measure a planetary radius and mass of $3.06\pm0.22$ R$_\oplus$ and $6.8\pm2.0$ M$_\oplus$. Radial velocities were extracted from the SPIRou spectra using both the CCF and the LBL methods, optimizing the velocity measurements in the near infrared domain. We analyzed the broadband SED of the star and the high-resolution SPIRou spectra to constrain the stellar parameters and thus improve the accuracy of the derived planet parameters. A LSD analysis of the SPIRou Stokes $V$ polarized spectra detects Zeeman signatures in TOI-1759. We model the rotational modulation of the magnetic stellar activity using a GP regression with a quasi-periodic covariance function, and find a rotation period of $35.65^{+0.17}_{-0.15}$ d. We reconstruct the large-scale surface magnetic field of the star using ZDI, which gives a predominantly poloidal field with a mean strength of $18\pm4$ G. Finally, we perform a joint Bayesian MCMC analysis of the TESS photometry and SPIRou RVs to optimally constrain the system parameters. At $0.1176\pm0.0013$ au from the star, the planet receives $6.4$ times the bolometric flux incident on Earth, and its equilibrium temperature is estimated at $433\pm14$ K. TOI-1759 b is a likely gas-dominated sub-Neptune with an expected high rate of photoevaporation. Therefore, it is an interesting target to search for neutral hydrogen escape, which may provide important constraints on the planetary formation mechanisms responsible for the observed sub-Neptune radius desert., Comment: Accepted for publication in the 10.
