Abstract

Other than the influence of ionizing radiation and benign thyroid disease, little is known about the risk factors for differentiated thyroid cancer (TC) which is an increasing common cancer worldwide. Consistent evidence shows that body mass is positively associated with TC risk. As excess weight is a state of chronic inflammation, we investigated the relationship between concentrations of leptin, adiponectin, C-reactive protein, interleukin (IL)-6, IL-10 and tumor necrosis factor (TNF)-α and the risk of TC. A case-control study was nested within the European Prospective Investigation into Cancer and Nutrition (EPIC) study and included 475 first primary incident TC cases (399 women and 76 men) and 1,016 matched cancer-free cohort participants. Biomarkers were measured in serum samples using validated and highly sensitive commercially available immunoassays. Odds ratios (ORs) of TC by levels of each biomarker were estimated using conditional logistic regression models, adjusting for BMI and alcohol consumption. Adiponectin was inversely associated with TC risk among women (ORT3vs.T1 = 0.69, 95% CI: 0.49-0.98, Ptrend = 0.04) but not among men (ORT3vs.T1 = 1.36, 95% CI: 0.67-2.76, Ptrend = 0.37). Increasing levels of IL-10 were positively associated with TC risk in both genders and significantly so in women (ORT3vs.T1 = 1.59, 95% CI: 1.13-2.25, Ptrend = 0.01) but not in men (ORT3vs.T1 = 1.78, 95% CI: 0.80-3.98, Ptrend = 0.17). Leptin, CRP, IL-6 and TNF-α were not associated with TC risk in either gender. These results indicate a positive association of TC risk with IL-10 and a negative association with adiponectin that is probably restricted to women. Inflammation may play a role in TC in combination with or independently of excess weight.

Abstract

Immuno-proteomic screening has identified several tumor-associated autoantibodies (AAb) that may have diagnostic capacity for invasive epithelial ovarian cancer, with AAbs to P53 proteins and cancer-testis antigens (CTAGs) as prominent examples. However, the early detection potential of these AAbs has been insufficiently explored in prospective studies. We performed ELISA measurements of AAbs to CTAG1A, CTAG2, P53 and NUDT11 proteins, for 194 patients with ovarian cancer and 705 matched controls from the European EPIC cohort, using serum samples collected up to 36 months prior to diagnosis under usual care. CA125 was measured using electrochemo-luminiscence. Diagnostic discrimination statistics were calculated by strata of lead-time between blood collection and diagnosis. With lead times 6 months, ovarian cancer detection sensitivity at 0.98 specificity (SE98) varied from 0.19 [95% CI 0.08-0.40] for CTAG1A, CTAG2 and NUDT1 to 0.23 [0.10-0.44] for P53 (0.33 [0.11-0.68] for high-grade serous tumors). However, at longer lead-times, the ability of these AAb markers to distinguish future ovarian cancer cases from controls declined rapidly; at lead times >1 year, SE98 estimates were close to zero (all invasive cases, range: 0.01-0.11). Compared to CA125 alone, combined logistic regression scores of AAbs and CA125 did not improve detection sensitivity at equal level of specificity. The added value of these selected AAbs as markers for ovarian cancer beyond CA125 for early detection is therefore limited. What's new? Could autoantibodies against tumor antigens provide an early warning system for ovarian cancer? These authors tested how well certain antibodies detected ovarian cancer. They selected four candidate antibodies, to p53, CTAG1A, CTAG2 and NUDT11 proteins, which appear in elevated levels in cancer patients. None of them performed well as a herald of burgeoning cancer. Endometrial cancer risk prediction models including lifestyle, anthropometric and reproductive factors have limited discrimination. Adding biomarker data to these models may improve predictive capacity; to our knowledge, this has not been investigated for endometrial cancer. Using a nested case-control study within the European Prospective Investigation into Cancer and Nutrition (EPIC) cohort, we investigated the improvement in discrimination gained by adding serum biomarker concentrations to risk estimates derived from an existing risk prediction model based on epidemiologic factors. Serum concentrations of sex steroid hormones, metabolic markers, growth factors, adipokines and cytokines were evaluated in a step-wise backward selection process; biomarkers were retained at p < 0.157 indicating improvement in the Akaike information criterion (AIC). Improvement in discrimination was assessed using the C-statistic for all biomarkers alone, and change in C-statistic from addition of biomarkers to preexisting absolute risk estimates. We used internal validation with bootstrapping (1000-fold) to adjust for over-fitting. Adiponectin, estrone, interleukin-1 receptor antagonist, tumor necrosis factor-alpha and triglycerides were select-ed into the model. After accounting for over-fitting, discrimination was improved by 2.0 percentage points when all evaluated biomarkers were included and 1.7 percentage points in the model including the selected biomarkers. Models including etiologic markers on independent pathways and genetic markers may further improve discrimination.

As CLD transfers from soil to animals mainly via involuntary ingestion, the consumption of foodstuffs derived from animals raised in contaminated areas may significantly contribute to exposure of humans to CLD. The present study was designed to test the efficacy of two different activated carbons (ACs) sources in limiting CLD transfer from soil to animal. Three soils (ASs) were prepared according to the OECD guideline 207. One standard soil (SS) lacking AC, and two modified preparations of SS supplemented with 2% coconut-based activated carbon (ORBO), SSO or with 2% lignite-based one (DARCO), SSD. All three soils were spiked with 10 μg of kepone per g of dry matter and aged for three weeks. This study involved 15 goat kids randomly assigned to the 3 experimental groups (n = 5/group), which were fed the experimental matrices at an exposure dose of 10 μg CLD per kg of body weight per day. After 21 d of oral exposure, CLD in adipose tissue and liver were analysed by LC-MS-MS. A significant decrease of 63.7% and 74.7% of CLD concentrations in adipose tissue and liver, respectively, were obtained from animals exposed using SS containing DARCO as compared to those receiving only SS. Decreases in CLD levels of 98.2% (adipose tissue) and 98.7% (liver) were obtained for animals exposed using SS containing ORBO. Abstract

Chlordecone (Kepone) (CLD) is a highly persistent pesticide formerly used in French West Indies. Nowadays high levels of this pesticide are still found in soils and represent a subsequent source of contamination for outdoor-reared animals. In that context, sequestering matrices such as activated carbons (ACs) are believed to efficiently decrease the bioavailability of such compounds. The present study intends to test the respective efficiency of two sequestering strategies using ACs in the animal production context: (i) ACs supplemented to feed or (ii) ACs as a soil amendment. This study involved 20 piglets randomly distributed into 5 experimental groups (4 replicates). All of them were exposed through one artificial soil to 10 µg of CLD per kg of BW. This soil was amended by 2% (mass basis) of either ORBO and DARCO, the two ACs tested, and given to 2 experimental groups. Two other groups were given additionally a feed containing 0.5% (mass basis) of one of the ACs. Only the experimental groups exposed through amended soil with one of the two ACs presented a significant decrease of concentrations of CLD in liver and adipose tissue in comparison with the control group (p< 0.001). This decrease was particularly high for a coconut shell activated carbon (ORBO) where relative bioavailability was found to be lower than 1.8% for both tissues. Overall, AC introduced in CLD contaminated soil has thye potential to strongly reduce CLD bioavailability whereas sequestration of CLD during digestive processes via supplemented feed was not efficient.

Abstract

The chlorinated polycyclic ketone pesticide chlordecone (CLD) was used from 1971 until 1993 in the French West Indies to fight against the banana black weevil (Cosmopolites sordidus). Its application resulted in a long term pollution of soils which is thought to last 5 to 7 centuries for the heaviest polluted soils. About 1/4 of the total agricultural acreage of the two French overseas departments (Guadeloupe, Martinique) are moderately to heavily polluted and national survey plans carried out since 2008 in slaughterhouses revealed unexpected contamination of animal products. A research program involving a broad research network is currently on going. It aims at evaluating local animal rearing systems in terms of livestock exposure to CLD, at characterizing the bioavailability of CLD, its behavior and metabolization in the animal organism, and at establishing strategies to bind CLD via activated carbon in the digestive tract. On the basis of the results obtained, safe local livestock rearing systems will be proposed in agricultural areas historically contaminated by CLD, and assessed in terms of economical efficiency and social acceptability. Preliminary results showed that, in tropical conditions, ingestion of soil is far from being negligible but it can be modulated by the forage offer in order to reduce livestock exposure to CLD. Regarding the potential to bind CLD in order to limit its absorption, the most promising results are obtained when activated carbon is added and maturated into contaminated soil. Where such approaches cannot be applied, there is a remaining potential to decontaminate the animals by applying a depuration period: CLD half lives in the organism are currently established. Such results are of great importance and will allow in a near future to establish innovative safe local livestock rearing systems in agricultural areas historically contaminated by CLD. The approaches implemented in this project and the proposed innovative strategies to secure the rearing systems have also to be considered as a “model approach” to be applied further in organic pollutant contaminated areas worldwide.

Because of its toxicity and considerable levels in soil, it became a major threat to address. Soil or sediments are considered as reservoir for this contaminant and soil ingestion is the main exposure route for free range animals. In order to ensure the production of safe animal foodstuffs, it is necessary to reduce the transfer from soil to animal tissues. The present study intends to test whether two different activated carbons (ACs) could limit the CLD transfer from soil to animal. Three artificial soils (ASs) were prepared according to OECD guideline 207. One standard soil (SS), devoid of organic matter (OM), and two amended versions of this SS with coconut-based activated carbon (AC1) or lignitebased one (AC2) were prepared to obtain a 2% organic carbon content (dry matter basis). This study involved 15 kids as a typical on-farm consumption animal. Animals were randomly distributed into 3 groups (n=5 replicates). During 21 days, the kids were fed AS spiked with 10 µg of CLD per g of dry matter to achieve an exposure dose of 10 µg CLD per kg of body weight per day. 175. Fertility drugs and cutaneous melanoma risk: a French prospective cohort study

Abstract

Cutaneous melanoma has been suspected to be influenced by female sex hormones. A review of the literature in 2018 indicated that fertility drug (FD) use was associated with increased melanoma risk among parous women only. However, most studies so far were based on a retrospective design and the current evidence is unclear. We sought to prospectively investigate the associations between FD use and melanoma risk in women. E3N is a prospective cohort of 98 995 French women aged 40–65 years at inclusion in 1990. Information on use of FDs, including duration and time of administration, was assessed through self-administered questionnaires. We used Cox proportional hazards regression models adjusted for age and melanoma risk factors. Over 1990–2008, about 611 melanoma cases were ascertained among 86 653 women. Compared with never use, ever use of FDs was not associated with melanoma risk overall [hazard ratio (HR) = 1.15; 95% confidence interval (CI) = 0.75–1.74], or among parous women (HR = 1.08; 95% CI = 0.67–1.73). Among ever users of FDs, duration of use and age at first use were not associated with melanoma risk. Associations were similar after adjustment for UV exposure, although FD users were more likely to report tanning bed use than never-users (odds ratio = 1.50; CI = 1.01–2.22) in a subsample with recreational UV exposure data. Our data do not support an association between FD use and melanoma risk, but underlie the importance of taking into consideration potential confounding from sun exposure in future research.

Background: Even though in situ breast cancer (BCIS) accounts for a large proportion of the breast cancers diagnosed, few studies have investigated potential risk factors for BCIS. Their results suggest that some established risk factors for invasive breast cancer have a similar impact on BCIS risk, but large population-based studies on lifestyle factors and BCIS risk are lacking. Thus, we investigated the association between lifestyle and BCIS risk within the European Prospective Investigation into Cancer and Nutrition cohort.Methods: Lifestyle was operationalized by a score reflecting the adherence to the World Cancer Research Fund/American Institute for Cancer Research (WCRF/AICR) cancer prevention recommendations. The recommendations utilized in these analyses were the ones pertinent to healthy body weight, physical activity, consumption of plant-based foods, energy-dense foods, red and processed meat, and sugary drinks and alcohol, as well as the recommendation on breastfeeding. Cox proportional hazards regression was used to assess the association between lifestyle score and BCIS risk. The results were presented as hazard ratios (HR) and corresponding 95% confidence intervals (CI).Results: After an overall median follow-up time of 14.9 years, 1277 BCIS cases were diagnosed. Greater adherence to the WCRF/AICR cancer prevention recommendations was not associated with BCIS risk (HR = 0.98, 95% CI 0.93-1.03; per one unit of increase; multivariable model). An inverse association between the lifestyle score and BCIS risk was observed in study centers, where participants were recruited mainly via mammographic screening and attended additional screening throughout follow-up (HR = 0.85, 95% CI 0.73-0.99), but not in the remaining ones (HR = 0.99, 95% CI 0.94-1.05).Conclusions: While we did not observe an overall association between lifestyle and BCIS risk, our results indicate that lifestyle is associated with BCIS risk among women recruited via screening programs and with regular screening participation. This suggests that a true inverse association between lifestyle habits and BCIS risk in the overall cohort may have been masked by a lack of information on screening attendance. The potential inverse association between lifestyle and BCIS risk in our analyses is consistent with the inverse associations between lifestyle scores and breast cancer risk reported from previous studies. Background: Cancer antigen 125 (CA125) is the most promising ovarian cancer screening biomarker to date. Multiple studies reported CA125 levels vary by personal characteristics, which could inform personalized CA125 thresholds. However, this has not been well described in premenopausal women. Methods: We evaluated predictors of CA125 levels among 815 premenopausal women from the New England Case Control Study (NEC). We developed linear and dichotomous (=35 U/mL) CA125 prediction models and externally validated an abridged model restricting to available predictors among 473 premenopausal women in the European Prospective Investigation into Cancer and Nutrition Study (EPIC). Results: The final linear CA125 prediction model included age, race, tubal ligation, endometriosis, menstrual phase at blood draw, and fibroids, which explained 7% of the total variance of CA125. The correlation between observed and predicted CA125 levels based on the abridged model (including age, race, and menstrual phase at blood draw) had similar correlation coefficients in NEC (r = 0.22) and in EPIC (r = 0.22). The dichotomous CA125 prediction model included age, tubal ligation, endometriosis, prior personal cancer diagnosis, family history of ovarian cancer, number of miscarriages, menstrual phase at blood draw, and smoking status with AUC of 0.83. The abridged dichotomous model (including age, number of miscarriages, menstrual phase at blood draw, and smoking status) showed similar AUCs in NEC (0.73) and in EPIC (0.78). Conclusions: We identified a combination of factors associated with CA125 levels in premenopausal women. Impact: Our model could be valuable in identifying healthy women likely to have elevated CA125 and consequently improve its specificity for ovarian cancer screening. [ABSTRACT FROM AUTHOR]

Abstract

Background Soy-based dietary supplements have been promoted as natural alternatives to menopausal hormone therapy, but their potential effect on breast cancer development is controversial. Objectives We examined the relation between the consumption of soy supplements and the risk of breast cancer, overall and by tumor hormone receptor status, among women aged >50 y. Methods In total, 76,442 women from the Etude Epidemiologique aupres de Femmes de la Mutuelle Generale de l'Education Nationale (E3N) cohort, born between 1925 and 1950, were followed from 2000 to 2011 (11.2 y on average, starting at a mean age of 59.5 y; 3608 incident breast cancers), with soy supplement use assessed every 2–3 y. HRs of breast cancer were estimated with the use of multivariable Cox models. Results Compared with never using soy supplements, the HRs associated with current use of soy supplements were 0.92 (95% CI: 0.76, 1.11) for all, 0.78 (95% CI: 0.60, 0.99) for estrogen receptor (ER)–positive, and 2.01 (95% CI: 1.41, 2.86) for ER-negative breast cancers. There was no association between past use of soy supplements and breast cancer. HRs for current use were 1.36 (95% CI: 0.95, 1.93) and 0.82 (95% CI: 0.65, 1.02) among women with and without a family history of breast cancer, respectively (P- interaction = 0.03) and 1.06 (95% CI: 0.87, 1.30) ≥5 y after menopause compared with 0.50 (95% CI: 0.31, 0.81) in premenopause or ≤5 y postmenopause (P- interaction = 0.04). Conclusions In this cohort of women aged >50 y, we report opposing associations of soy supplements with ER-positive and ER-negative breast cancer risk. Our results also caution against the use of these supplements in women with a family history of breast cancer. Whether the risk profile of soy supplements could be more favorable among premenopausal or recently postmenopausal women deserves further investigation. [ABSTRACT FROM AUTHOR]

Abstract

Estrogen-progestagen menopausal hormone therapy (MHT), indicated for the treatment of troublesome climacteric symptoms, is recognized as carcinogenic to humans. We present the available epidemiologic evidence concerning its link with the risks of breast, ovarian, endometrial and colorectal cancers.While results on cancers other than breast cancer are still imprecise or even contradictory, there is a clear increase in breast cancer risk associated with the use of estrogen-progestagen MHT. Attempts to play down in the public opinion the risk of breast cancer associated with these treatments evoke a better prognosis of tumors induced by MHT, the fact that MHT would only accelerate the presentation of pre-existing breast tumors that would otherwise have appeared latter, or that MHT would be devoid of risks when initiated close to menopause onset. We show that these assertions are not supported by the available epidemiological evidence., Parmi les médicaments reconnus comme cancérogènes pour l'homme figurent les traitements hormonaux de la ménopause (THM) estroprogestatifs, indiqués pour le traitement des troubles gênants du climatère. Nous présentons ici les connaissances épidémiologiques concernant leur lien avec les risques de cancers du sein, de l'ovaire, de l'endomètre, et de cancer colorectal. Si les résultats concernant les localisations autres que le sein sont encore imprécis ou parfois contradictoires, l'augmentation du risque de cancer du sein avec l'utilisation de THM estroprogestatifs est claire. Des tentatives pour minimiser auprès du public le risque de cancer du sein associé à ces traitements évoquent un meilleur pronostic des cancers du sein développés sous THM, le fait que les THM ne feraient qu'accélérer l'apparition de tumeurs préexistantes qui seraient de toute façon survenues plus tard, ou que les THM seraient dénués de risques lorsqu'ils sont instaurés dès la survenue de la ménopause. Nous montrons que ces affirmations ne sont pas étayées par les données épidémiologiques actuelles.

Abstract

International audience; The current evaluation of the benefit/risk ratio associated with menopausal hormone therapy (MHT) use is largely based on clinical trials which investigated the effects of oral treatments. Would MHT with transdermal estrogens be associated with a more favourable benefit/risk ratio? We reviewed the available epidemiologic evidence on that question. Epidemiologic studies were considered if they provided risk estimates of conditions which carry an important weight among menopausal women, and for which epidemiologic evidence of a possible link with MHT use is convincing: cardiovascular diseases, breast cancer, diabetes, colorectal cancer and hip fracture. We did not include studies with only surrogate measures. We found that the available information on the potential impact of the route of administration of MHT on the risk of our selected outcomes is limited. To date, epidemiologic data suggest that it has no impact on the risk of breast cancer and hip fracture. Results on the risk of coronary heart disease and colorectal cancer are inconsistent. Studies on stroke and diabetes risk are too few to allow meaningful conclusions. There is a suggestion that transdermal MHT may be less deleterious than oral MHT regarding venous thromboembolism which needs to be confirmed. The issue of the route of administration of MHT should remain an active area of research as part of an attempt to identify treatment modalities that would have the least potential for exerting adverse effects.

Abstract

BACKGROUND: The higher risk of death resulting from excess adiposity may be attenuated by physical activity (PA). However, the theoretical number of deaths reduced by eliminating physical inactivity compared with overall and abdominal obesity remains unclear.OBJECTIVE: We examined whether overall and abdominal adiposity modified the association between PA and all-cause mortality and estimated the population attributable fraction (PAF) and the years of life gained for these exposures.DESIGN: This was a cohort study in 334,161 European men and women. The mean follow-up time was 12.4 y, corresponding to 4,154,915 person-years. Height, weight, and waist circumference (WC) were measured in the clinic. PA was assessed with a validated self-report instrument. The combined associations between PA, BMI, and WC with mortality were examined with Cox proportional hazards models, stratified by center and age group, and adjusted for sex, education, smoking, and alcohol intake. Center-specific PAF associated with inactivity, body mass index (BMI; in kg/m(2)) (>30), and WC (≥102 cm for men, ≥88 cm for women) were calculated and combined in random-effects meta-analysis. Life-tables analyses were used to estimate gains in life expectancy for the exposures.RESULTS: Significant interactions (PA × BMI and PA × WC) were observed, so HRs were estimated within BMI and WC strata. The hazards of all-cause mortality were reduced by 16-30% in moderately inactive individuals compared with those categorized as inactive in different strata of BMI and WC. Avoiding all inactivity would theoretically reduce all-cause mortality by 7.35% (95% CI: 5.88%, 8.83%). Corresponding estimates for avoiding obesity (BMI >30) were 3.66% (95% CI: 2.30%, 5.01%). The estimates for avoiding high WC were similar to those for physical inactivity.CONCLUSION: The greatest reductions in mortality risk were observed between the 2 lowest activity groups across levels of general and abdominal adiposity, which suggests that efforts to encourage even small increases in activity in inactive individuals may be beneficial to public health.

Abstract

Laying hens are likely to be exposed to a wide range of persistent organic pollutants (POPs), including polychlorinated biphenyls (PCBs). To improve the safety of poultry farming systems in terms of POPs, the present research focused on assessing the impact of physiological characteristics of the hen on the transfer of ingested PCBs to eggs. Modeling was used as a research tool to explore the impact of some physiological characteristics on the transfer of PCBs in the laying hen. The mathematical model simulates the dynamics of the size of the lipid compartments in the animal and the frequency of laying, with the PCB concentrations in egg yolk and adipose tissue being model outputs. Simulations were run to assess effects of animal characteristics on the transfer of PCBs to eggs. Laying rate proportionally influenced the PCB level of eggs and adipose tissue at steady state. Body fat diluted absorbed PCBs in the absence of laying and significantly influenced the decontamination rate of tissues during depuration after an exposure period. Abstract

International audience; Objectives Menopausal hormone therapy (MHT) use is characterized by important geographical and temporal variations. Knowledge of these variations is necessary to interpret the results of epidemiological studies on MHT use and disease risk, and can contribute to understand national changes in diseases’ incidence. Methods We used data from a cohort study of women with information on lifetime MHT use collected in 2003-2004 to describe the characteristics and recent evolution of MHT consumption in Sweden. Results Among the 17,450 postmenopausal participants, prevalence of use declined by approximately 30% between 2001 and 2003. MHT was almost no longer used to prevent osteoporosis, and worry about side effects was often cited as a reason for stopping treatment. MHT mostly consisted in estradiol used alone or opposed with either norethisterone acetate or medroxyprogesterone acetate, or in low-potency estrogens. An estimated 30% of participants initiated MHT before menopause. The cumulative monthly dose of progestagen was generally doubled in continuous-combined compared with sequential combinations. Conclusions In this cohort, both prevalence of use and reasons for initiating MHT have varied between 2001 and 2003, while the modalities of use remained essentially the same.

International audience; PURPOSETo investigate whether the relation between estrogen-progestagen menopausal hormone therapy (EP-MHT) and breast cancer risk varies according to the delay between menopause onset and treatment initiation.MATERIALS AND METHODSBetween 1992 and 2005, 1,726 invasive breast cancers were identified among 53,310 postmenopausal women from the French E3N cohort (mean duration of follow-up: 8.1 years). Hazard ratios (HRs) and confidence intervals (CI) were estimated using Cox models, with MHT never-users as the reference.RESULTSAmong recent users of EP-MHT, the risk of breast cancer varied according to the timing of treatment initiation. This variation was confined to short durations of use (≤2 years): the HR was 1.54 (95% CI 1.28-1.86) for short treatments initiated in the 3-year period following menopause onset and 1.00 (95% CI 0.68-1.47) for short treatments initiated later (p=0.04 for homogeneity). This pattern of risks was however not observed in users of EP-MHT containing progesterone, among whom there was no significantly increased risk associated with short durations of use (HR 0.87, 95% CI 0.57-1.32 for treatments initiated ≤3 years after menopause and 0.90, 95% CI 0.45-1.81 for treatments initiated later). Longer durations of EP-MHT use were generally associated with increases in breast cancer risk, whatever the gap time.CONCLUSIONOur results suggest that, for some EP-MHT, the timing of treatment initiation transiently modulates the risk of breast cancer; and that, when initiated close to menopause, even short durations of use are associated with an increased breast cancer risk. Estrogen-progesterone combinations might be an exception in this regard.

International audience; Young children with their hand-to-mouth activity may be exposed to contaminated soils. However few studies assessing exposure of organic compounds sequestrated in soil were realized. The present study explores the impact of different organic matters on retention of NDL-PCBs during digestive processes using commercial humic substances in a close digestive model of children: the piglet. Six artificial soils were used. One standard soil, devoid of organic matter, and five amended versions of this standard soil with either fulvic acid, humic acid, Sphagnum peat, activated carbon or a mix of Sphagnum peat and activated carbon (95:5) (SPAC) were prepared. In order to compare the different treatments, we use spiked oil and negative control animals. Forty male piglets were randomly distributed in 7 contaminated and one control groups (n = 5 for each group). During 10 days, the piglets were fed artificial soil or a corn oil spiked with 19 200 ng of Aroclor 1254 per g of dry matter (6 000 ng.g(-1) of NDL-PCBs) to achieve an exposure dose of 1 200 ng NDL-PCBs.Kg(-1) of body weight per day. NDL-PCBs in adipose tissue were analyzed by GC-MS. Fulvic acid reduced slightly the bioavailability of NDL-PCBs compared to oil. Humic acid and Sphagnum peat reduced it significantly higher whereas activated carbon reduced the most. Piglets exposed to soil containing both activated carbon and Shagnum peat exhibited a lower reduction than soil with only activated carbon. Therefore, treatment groups are ordered by decreasing value of relative bioavailability as following: oil >= fulvic acid > Sphagnum peat >= Sphagnum peat and activated carbon >= Humic acid >> activated carbon. This suggests competition between Sphagnum peat and activated carbon. The present study highlights that quality of organic matter does have a significant effect on bioavailability of sequestrated organic compounds.

Abstract

International audience; The relative bioavailability of sediment-bound polychlorinated biphenyls (PCBs 138, 153, and 180) from a local contaminated site was examined using an in vivo carp model. Surface sediment from the PCB-contaminated site and spiked canola oil containing equivalent masses of PCBs were respectively incorporated in the sediment-dosed diets and oil-dosed diets at 3 dose levels resulting in 6 experimental diets. Juvenile carps (n=90) were divided in 18 tanks (5 fishx6 treatmentsx3 tanks). Fish were fed the control diet during the adaptation period (15 d). Next, 1 fish was sampled in each tank and muscle tissues were combined in control groups. During the exposure period (15 d), the remaining fish were fed with 1 of the 6 experimental diets. At the end of the experiment, fish were sampled and muscle tissues were combined for each tank. The PCBs were monitored in feed and fish muscle. For both the contaminated sediment and spiked canola oil groups, concentrations of PCBs 138, 153, and 180 in muscle linearly increased with concentrations in food, with similar intercepts and slopes. In the present study, the sediment-bound PCBs were as bioavailable as those spiked into canola oil and fed to carp in a standard diet.

Abstract

Background: In addition to HPV, high parity and hormonal contraceptives have been associated with cervical cancer (CC). However, most of the evidence comes from retrospective case-control studies. The aim of this study is to prospectively evaluate associations between hormonal factors and risk of developing cervical intraepithelial neoplasia grade 3 (CIN3)/carcinoma in situ (CIS) and invasive cervical cancer (ICC). Methods and Findings: We followed a cohort of 308,036 women recruited in the European Prospective Investigation into Cancer and Nutrition (EPIC) Study. At enrollment, participants completed a questionnaire and provided serum. After a 9-year median follow-up, 261 ICC and 804 CIN3/CIS cases were reported. In a nested case-control study, the sera from 609 cases and 1,218 matched controls were tested for L1 antibodies against HPV types 11,16,18,31,33,35,45, 52,58, and antibodies against Chlamydia trachomatis and Human herpesvirus 2. Multivariate analyses were performed to estimate hazard ratios (HR), odds ratios (OR) and corresponding 95% confidence intervals (CI). The cohort analysis showed that number of fullterm pregnancies was positively associated with CIN3/CIS risk (p-trend = 0.03). Duration of oral contraceptives use was associated with a significantly increased risk of both CIN3/CIS and ICC (HR = 1.6 and HR = 1.8 respectively for >= 15 years versus never use). Ever use of menopausal hormone therapy was associated with a reduced risk of ICC (HR = 0.5, 95% CI: 0.4-0.8). A non-significant reduced risk of ICC with ever use of intrauterine devices (IUD) was found in the nested case-control analysis (OR = 0.6). Analyses restricted to all cases and HPV seropositive controls yielded similar results, revealing a significant inverse association with IUD for combined CIN3/CIS and ICC (OR = 0.7). Abstract

The role of hormonal factors in the etiology of lymphoid neoplasms remains unclear. Previous studies have yielded conflicting results, have lacked sufficient statistical power to assess many lymphoma subtypes, or have lacked detailed information on relevant exposures. Within the European Prospective Investigation Into Cancer and Nutrition cohort, we analyzed comprehensive data on reproductive factors and exogenous hormone use collected at baseline (1992–2000) among 343,458 women, including data on 1,427 incident cases of B-cell non-Hodgkin lymphoma (NHL) and its major subtypes identified after a mean follow-up period of 14 years (through 2015). We estimated hazard ratios and 95% confidence intervals using multivariable proportional hazards modeling. Overall, we observed no statistically significant associations between parity, age at first birth, breastfeeding, oral contraceptive use, or ever use of postmenopausal hormone therapy and risk of B-cell NHL or its subtypes. Women who had undergone surgical menopause had a 51% higher risk of B-cell NHL (based on 67 cases) than women with natural menopause (hazard ratio = 1.51, 95% confidence interval: 1.17, 1.94). Given that this result may have been due to chance, our results provide little support for the hypothesis that sex hormones play a role in lymphomagenesis. [ABSTRACT FROM AUTHOR]
