Purpose: Spontaneous intracerebral hemorrhage (ICH) is a major cause of death and disability with a huge economic burden worldwide. Cerebrolysin (CBL) has been previously used as a nootropic drug. Necroptosis is a programmed cell death mechanism that plays a vital role in neuronal cell death after ICH. However, the precise role of necroptosis in CBL neuroprotection following ICH has not been confirmed. Methods: In the present study, we aimed to investigate the neuroprotective effects and potential molecular mechanisms of CBL in ICH-induced early brain injury (EBI) by regulating neural necroptosis in the C57BL/6 mice model. Mortality, neurological score, brain water content, and neuronal death were evaluated by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining, Evans blue extravasation, Western blotting, and quantitative real-time polymerase chain reaction (PCR). Results: The results show that CBL treatment markedly increased the survival rate, neurological score, and neuron survival, and downregulated the protein expression of RIP1 and RIP3, which indicated that CBL-mediated inhibition of necroptosis, and ameliorated neuronal death after ICH. The neuroprotective capacity of CBL is partly dependent on the Akt/GSK3β signaling pathway. Conclusions: CBL improves neurological outcomes in mice and reduces neuronal death by protecting against neural necroptosis.

The Sugarcane yellow leaf virus (SCYLV) is associated with sugarcane yellow leaf disease (SCYLD) and is considered to be the most economically deleterious emerging pathogen that represents a potential threat and danger to sugarcane cultivation in China. Over the last two decades, high genetic diversity in the SCYLV genotypes was observed worldwide, with a greater chance of YLD incidence for sugarcane injury. SCYLV infection has significantly damaged its economic traits and is responsible for substantial losses in biomass production in sugarcane cultivars. This study aims to identify and comprehensively analyze sugarcane microRNAs (miRNAs) as therapeutic targets against SCYLV using plant miRNA prediction tools. Mature sugarcane miRNAs are retrieved and are used for hybridization of the SCYLV. A total of seven common sugarcane miRNAs were selected based on consensus genomic positions. The biologically significant, top ranked ssp-miR528 was consensually predicted to have a potentially unique hybridization site at nucleotide position 4162 for targeting the ORF5 of the SCYLV genome; this was predicted by all the algorithms used in this study. Then, the miRNA–mRNA regulatory network was generated using the Circos algorithm, which was used to predict novel targets. There are no acceptable commercial SCYLV-resistant sugarcane varieties available at present. Therefore, the predicted biological data offer valuable evidence for the generation of SCYLV-resistant sugarcane plants.

Sugarcane is an important sugar and energy crop worldwide. It utilises highly efficient C4 photosynthesis and accumulates sucrose in its culms. The sucrose content in sugarcane culms is a quantitative trait controlled by multiple genes. The regulatory mechanism underlying the maximum sucrose level in sugarcane culms remains unclear. We used transcriptome sequences to identify the potential regulatory genes involved in sucrose accumulation in Saccarum officinarum L. cv. Badila. The sucrose accumulating internodes at the elongation and mature growth stage and the immature internodes with low sucrose content at the mature stage were used for RNA sequencing. The obtained differentially expressed genes (DEGs) related to sucrose accumulation were analysed. Results showed that the transcripts encoding invertase (beta-fructofuranosidase, EC: 3.2.1.26) which catalyses sucrose hydrolysis and 6-phosphofructokinase (PFK, EC: 2.7.1.11), a key glycolysis regulatory enzyme, were downregulated in the high sucrose accumulation internodes. The transcripts encoding key enzymes for ABA, gibberellin and ethylene synthesis were also downregulated during sucrose accumulation. Furthermore, regulated protein kinase, transcription factor and sugar transporter genes were also obtained. This research can clarify the molecular regulation network of sucrose accumulation in sugarcane.

Hexokinase (HXK) catalyses hexose phosphorylation and senses glucose signalling to control carbon partition in plants. In sugarcane, HXK is involved in regulating source/sink carbon balance. However, the characteristics of sugarcane HXK genes remain unclear. This study revealed that the full-length cDNA sequence of the HXK gene ShHXK8 cloned from sugarcane mature leaves contains a 1560 bp open reading frame and encodes a putative protein of 519 amino acids. Amongst HXKs from monocot and dicot subjected to phylogenetic analysis, ShHXK8 was clustered into a monocot-specific group with OsHXK7, SbHXK8 and ZmHXK1. Gene expression analysis in Saccharum spp. hybrid ROC22 plants showed that ShHXK8 was expressed in leaves and stalks and expressed highly in mature leaves and young internodes. The difference in the expression of ShHXK8 was compared in high-sugar-content (HSC) and low-sugar-content (LSC) Saccharum officinarum plants. The expression of ShHXK8 in the leaves and internodes 3–24 of HSC plants was lower than that of LSC plants. The expression of ShHXK8 in the internodes 1–2 of HSC plants was higher than that of LSC plants. ShHXK8–GFP fusion was located in the cytosol by transiently expressing the corresponding vectors in rice mesophyll protoplasts. ShHXK8 might function as a cytoplasmic hexokinase involved in hexose metabolism in sugarcane mature leaves and young internodes.

Recent studies have shown that preterm delivery is a risk factor for the development of postpartum depression, which not only impairs maternal-infant interactions, leading to infant developmental delay and social interaction difficulties in affected children, but also increases the risk of depression in the mother. Hence, this article aims to study the effects of parental engagement and early interactions with preterm infants on subsequent infant development and behavior, maternal adjustment, and mother-infant relationship. A total of 151 infants/mothers were enrolled in our study. Infants were randomized either to receive early parent interaction or standard care. The early parent interaction program was performed in addition to routine standard of care, Kangaroo Mother Care, during the neonatal intensive care unit stay based on PremieStart Protocol. The behavioral competencies of preterm infants were assessed, as were their mothers' adjustment (depression and coping) and competencies (knowledge of child development). At 12 months of postnatal age, child competencies (development and behavior) were assessed, together with maternal adjustment (parenting stress and depression). Mother-infant interaction was also observed. Early parent interaction did not alter early or later infant development. Furthermore, early parent interaction did not alter early maternal adjustment or late mother-infant relationship, but it reduced the risk of late postpartum depression. Taken together, these studies provide a strong basis for interventions that support parents in the parenting role and guide parents in developmentally appropriate interactions with their preterm babies. These interventions have the potential to lessen the adverse impact of preterm birth on babies and mothers. In addition, the positive benefits of reduced stress can improve parent mental health outcomes and ultimately may further improve parents' relationships with their babies.

Resistance to anti-fungal drugs has become the main cause for increasing incidence rate of Candida infections in pediatric intensive care units (PICU). Zinc supplementation has been shown to exhibit beneficial effects on many pediatric illnesses. This study aimed to investigate the efficacy of zinc supplementation on prevalence of candidemia and candiduria infections in PICU. 724 eligible children between 1 to 5 years old admitted in PICU were randomly assigned into either zinc supplementation group or placebo group. Primary endpoints were the number of Candida infections, length of PICU stay and cases of patient death 14 days after enrollment. Secondary endpoints were the incidence rates of candidemia and candiduria. The incidences of candiduria and candidemia were significantly lower in the zinc group than the placebo group. The length of PICU stay and cases of patient death were obviously lowered in the zinc group compared to the placebo group. In conclusion, zinc supplementation shows beneficial clinical efficacy in reducing Candida infections among PICU patients on broad-spectrum antibiotics with critical illnesses.

Ethnopharmacological relevanceOsteonecrosis of the femoral head (ONFH) is still a challenge for orthopedists worldwide, which may lead to disability in patients without effective treatment. A newly developed formula of Chinese medicine, Danyu Gukang Pills (DGP), was recognized to be effective for ONFH. Nevertheless, its molecular mechanisms remain to be clarified. MethodsNetwork pharmacology was adopted to detect the mechanism of DGP on ONFH. The compounds of DGP were collected from the online databases, and active components were selected based on their OB and DL index. The potential proteins of DGP were acquired from TCMSP database, while the potential genes of ONFH were obtained from Gene Cards and Pubmed Gene databases. The function of Gene and potential pathways were researched by GO and KEGG pathway enrichment analysis. The compounds-targets and targets-pathways network were constructed in an R and Cytosacpe software. The mechanism was further investigated via molecular docking. Finally, in-vitro experiments were validated in the BMSCs. ResultsA total of 2305 compounds in DGP were gained, among which, 370 were selected as active components for which conforming to criteria. Combined the network analysis, molecular docking and in-vitro experiments, the results firstly demonstrated that the treatment effect of DGP on ONFH may be closely related to HIF-1α, VEGFA and HIF-1 signaling pathway. ConclusionThe current study firstly researched the molecular mechanism of DGP on ONFH based on network pharmacology. The results indicated that DGP may exert the effect on ONFH targeting on HIF-1α and VEGFA via HIF-1 signaling pathway.

This study aimed to prepare the sugar industry for the possible introduction of genetically modified (GM) sugarcane and derived retail sugar products and to address several potential public concerns regarding the characteristics and safety of these products. GM sugarcane lines with integrated Cry1Ab and EPSPS foreign genes were used for GM sugar production. Traditional PCR, real-time fluorescent quantitative PCR (RT-qPCR), and enzyme-linked immunosorbent assay (ELISA) were performed in analyzing leaves, stems, and other derived materials during sugar production, such as fibers, clarified juices, filter mud, syrups, molasses, and final GM sugar product. The toxicity of GM sugar was examined with a feeding bioassay using Helicoverpa armigera larvae. PCR and RT-qPCR results showed that the leaves, stems, fibers, juices, syrups, filter mud, molasses, and white granulated sugar from GM sugarcane can be distinguished from those derived from non-GM sugarcane. The RT-qPCR detection method using short amplified product primers was more accurate than the traditional PCR method. Molecular analysis results indicated that trace amounts of DNA residues remain in GM sugar, and thus it can be accurately characterized using molecular analysis methods. ELISA results showed that only the leaves, stems, fibers, and juices sampled from the GM sugarcane differed from those derived from the non-GM sugarcane, indicating that filter mud, syrup, molasses, and white sugar did not contain detectable Cry1Ab and EPSPS proteins. Toxicity analysis showed that the GM sugar was not toxic to the H. armigera larvae. The final results showed that the GM sugar had no active proteins despite containing trace amounts of DNA residues. This finding will help to pave the way for the commercialization of GM sugarcane and production of GM sugar.

The properties of lignocellulosic substrates obtained from different pretreatments have a big impact on downstream saccharification based on both the fungal cellulase system and the cellulosome-based whole-cell biocatalysis system. However the corresponding effect of these two distinct saccharification strategies has not been comparatively analyzed. In this work, three ammonium sulfite (AS)-based pretreatment combinations (i.e., AS + hydrothermal (HT) pretreatment, AS + xylanase (X) pretreatment, and HT + AS pretreatment) were conducted to treat wheat straw. The obtained pretreated substrates with different properties were saccharified using fungal cellulase or an engineered Clostridium thermocellum strain as the whole-cell biocatalyst, and the ability to release sugar was comparatively evaluated. It was found that for the whole-cell saccharification, the total sugar digestibility of AS + HT/X pretreated wheat straw was 10% higher than that of HT + AS pretreated wheat straw. However, for fungal cellulase-based saccharification, the enzymatic hydrolysis efficiency was less susceptible to the sequence of pretreatment combinations. Hence, the whole-cell biocatalysis system was more sensitive to substrate accessibility compared to the free enzymes. In addition, the characterization and analyses showed that AS + HT/X pretreatment could remove more lignin, generating a more accessible surface with a larger external surface and lower surface lignin coverage, compared to the HT + AS pretreatment. Therefore, the AS + HT/X pretreatment was more compatible with the cellulosome-based whole-cell saccharification.

Sugarcane streak mosaic virus (SCSMV, genus Poacevirus) causes sugarcane mosaic disease and leads to considerable economic losses. However, current methods for SCSMV diagnosis are limited. In this study, a novel isothermal assay was established on the basis of reverse transcription–recombinase polymerase amplification (RT-RPA) and evaluated in terms of its ability to detect SCSMV. The established RT-RPA assay with high specificity and reliability was 100-fold more sensitive than the RT-polymerase chain reaction. The evaluation of the temperature limit revealed that the assay had a flexible reaction temperature (26–46 °C), thereby indicating that the reaction mixture can be incubated with simple heating equipment, ambient temperature, or even human body heat. Time limit assessment results revealed that SCSMV can be detected in 5 min at 38 °C and in 20 min at 26 °C, thereby suggesting that the assay was rapid for SCSMV detection. Overall, the RT-RPA assay was specific, sensitive, reliable, and rapid for SCSMV detection, with a flexible and easily reachable reaction temperature. Thus, it can be an ideal method for SCSMV detection in sugarcane.
