Abstract

Abstract Background Triple‐negative breast cancer (TNBC) is the subtype of breast cancer with the worst prognosis, and it is highly heterogeneous. There is growing evidence that the tumor immune microenvironment (TIME) plays a crucial role in tumor development, maintenance, and treatment responses. Notably however, the full effects of the TIME on prognosis, TIME characteristics, and immunotherapy responses in TNBC patients have not been fully elucidated. Methods Gene Expression Omnibus and The Cancer Genome Atlas data were used to data analysis. Single‐cell sequencing and tissue microarray analysis were used to investigate gene expression. The concentrations and distributions of immune cell types were determined and analyzed using the CIBERSORT strategy. Tumor immune dysfunction and exclusion score and the IMvigor210 cohort were used to estimate the sensitivity of TNBC patients with different prognostic statuses to immune checkpoint treatment. Results Five immune‐related genes associated with TNBC prognosis (IL6ST, NR2F1, CKLF, TCF7L2, and HSPA2) was identified and a prognostic evaluation model was constructed based on those genes. The respective areas under the curve of the prognostic nomogram model at 3 and 5 years were 0.791 and 0.859. The group with a lower nomogram score, with a better prognosis survival status and clinical treatment benefit rate. Conclusion A prognostic model for TNBC that was closely related to the immune landscape and therapeutic responses was constructed. This model may help clinicians to make more precise and personalized treatment decisions pertaining to TNBC patients.

Abstract

[Objective] The effects of climate change on the stability of organic carbon were revealed from the perspective of the molecular structure of organic carbon in order to provide a theoretical basis for clarifying the impact of future climate change on organic carbon stability and soil fertility in mollisols. [Methods] A long-term field experiment dealing with the simulation of climate change through the use of open-top chambers (OTCs) at the Hailun Agroecological Experiment Station of the Chinese Academy of Sciences provided data for this study. We analyzed the organic carbon content and infrared spectral characteristics of various aggregates and density fractions in the 0—20 cm topsoil of black soil under three treatments: ① ambient temperature and CO2 concentration (aTaCO2); ② temperature elevated by 2 ℃ and ambient CO2 concentration (eTaCO2); ③ temperature elevated by 2 ℃ and CO2 enrichment to (700±25) μmol/mol (eTeCO2). [Results] Neither eTaCO2 nor eTeCO2 significantly affected organic carbon content of the bulk soil compared with aTaCO2 (p>0.05). However, eTaCO2 increased soil organic carbon (SOC) contents by 13.45% and 52.89% in the 0.25 mm size aggregate, but increased the -CH/C=C ratio of organic carbon in the 0.25—0.053 mm size aggregates (p

Abstract

Contact-rich manipulation tasks are commonly found in modern manufacturing settings. However, manually designing a robot controller is considered hard for traditional control methods as the controller requires an effective combination of modalities and vastly different characteristics. In this paper, we firstly consider incorporating operational space visual and haptic information into reinforcement learning(RL) methods to solve the target uncertainty problem in unstructured environments. Moreover, we propose a novel idea of introducing a proactive action to solve the partially observable Markov decision process problem. Together with these two ideas, our method can either adapt to reasonable variations in unstructured environments and improve the sample efficiency of policy learning. We evaluated our method on a task that involved inserting a random-access memory using a torque-controlled robot, and we tested the success rates of the different baselines used in the traditional methods. 187. Center-of-Mass-based Robust Grasp Planning for Unknown Objects Using Tactile-Visual Sensors

Abstract

An unstable grasp pose can lead to slip, thus an unstable grasp pose can be predicted by slip detection. A regrasp is required afterwards to correct the grasp pose in order to finish the task. In this work, we propose a novel regrasp planner with multi-sensor modules to plan grasp adjustments with the feedback from a slip detector. Then a regrasp planner is trained to estimate the location of center of mass, which helps robots find an optimal grasp pose. The dataset in this work consists of 1 025 slip experiments and 1 347 regrasps collected by one pair of tactile sensors, an RGB-D camera and one Franka Emika robot arm equipped with joint force/torque sensors. We show that our algorithm can successfully detect and classify the slip for 5 unknown test objects with an accuracy of 76.88% and a regrasp planner increases the grasp success rate by 31.0% compared to the state-of-the-art vision-based grasping algorithm., Comment: 6 pages + references

188. Towards Memory Safe Python Enclave for Security Sensitive Computation

Abstract

Intel SGX Guard eXtensions (SGX), a hardware-supported trusted execution environment (TEE), is designed to protect security-sensitive applications. However, since enclave applications are developed with memory unsafe languages such as C/C++, traditional memory corruption is not eliminated in SGX. Rust-SGX is the first toolkit providing enclave developers with a memory-language. However, Rust is considered a Systems language and has become the right choice for concurrent applications and web browsers. Many application domains such as Big Data, Machine Learning, Robotics, Computer Vision are more commonly developed in the python programming language. Therefore, Python application developers cannot benefit from secure enclaves like Intel SGX and rust-SGX. To fill this gap, we propose Python-SGX, which is a memory-safe SGX SDK providing enclave developers a memory-safe Python development environment. The key idea is to enable memory-safe Python language in SGX by solving the following key challenges: (1) defining a memory-safe Python interpreter (2)replacing unsafe elements of Python interpreter with safe ones,(3) achieving comparable performance to non-enclave Python applications, and (4) not introducing any unsafe new code or libraries into SGX. We propose to build Python-SGX with PyPy, a Python interpreter written by RPython, which is a subset of Python, and tame unsafe parts in PyPy by formal verification, security hardening, and memory safe language. We have implemented python-SGX and tested it with a series of benchmarks programs. Our evaluation results show that Python-SGX does not cause significant overhead.

191. Late sowing enhances lodging resistance of wheat plants by improving the biosynthesis and accumulation of lignin and cellulose

Abstract

Delayed sowing mitigates lodging in wheat. However, the mechanism underlying the enhanced lodging resistance in wheat has yet to be fully elucidated. Field experiments were conducted to investigate the effects of sowing date on lignin and cellulose metabolism, stem morphological characteristics, lodging resistance, and grain yield. Seeds of Tainong 18, a winter wheat variety, were sown on October 8 (normal sowing) and October 22 (late sowing) during both of the 2015–2016 and 2016–2017 growing seasons. The results showed that late sowing enhanced the lodging resistance of wheat by improving the biosynthesis and accumulation of lignin and cellulose. Under late sowing, the expression levels of key genes (TaPAL, TaCCR, TaCOMT, TaCAD, and TaCesA1, 3, 4, 7, and 8) and enzyme activities (TaPAL and TaCAD) related to lignin and cellulose biosynthesis peaked 4–12 days earlier, and except for the TaPAL, TaCCR, and TaCesA1 genes and TaPAL, in most cases they were significantly higher than under normal sowing. As a result, lignin and cellulose accumulated quickly during the stem elongation stage. The mean and maximum accumulation rates of lignin and cellulose increased, the maximum accumulation contents of lignin and cellulose were higher, and the cellulose accumulation duration was prolonged. Consequently, the lignin/cellulose ratio and lignin content were increased from 0 day and the cellulose content was increased from 11 days after jointing onward. Our main finding is that the improved biosynthesis and accumulation of lignin and cellulose were responsible for increasing the stem-filling degree, breaking strength, and lodging resistance. The major functional genes enhancing lodging resistance in wheat that are induced by delayed sowing need to be determined.
