151. Neuroimaging of depression with diffuse optical tomography during repetitive transcranial magnetic stimulation

Abstract: Abstract Repetitive transcranial magnetic stimulation (rTMS) is an effective and safe treatment for depression; however, its potential has likely been hindered due to non-optimized targeting, unclear ideal stimulation parameters, and lack of information regarding how the brain is physiologically responding during and after stimulation. While neuroimaging is ideal for obtaining such critical information, existing modalities have been limited due to poor resolutions, along with significant noise interference from the electromagnetic spectrum. In this study, we used a novel diffuse optical tomography (DOT) device in order to advance our understanding of the neurophysiological effects of rTMS in depression. Healthy and depressed subjects aged 18–70 were recruited. Treatment parameters were standardized with targeting of the left dorsolateral prefrontal cortex with a magnetic field intensity of 100% of motor threshold, pulse frequency of 10 per second, a 4 s stimulation time and a 26 s rest time. DOT imaging was simultaneously acquired from the contralateral dorsolateral prefrontal cortex. Six healthy and seven depressed subjects were included for final analysis. Hemoglobin changes and volumetric three-dimensional activation patterns were successfully captured. Depressed subjects were observed to have a delayed and less robust response to rTMS with a decreased volume of activation compared to healthy subjects. In this first-in-human study, we demonstrated the ability of DOT to safely and reliably capture and compare cortical response patterns to rTMS in depressed and healthy subjects. We introduced this emerging optical functional imaging modality as a novel approach to investigating targeting, new treatment parameters, and physiological effects of rTMS in depression.

Abstract: Summary: Microbial inoculants containing arbuscular mycorrhizal (AM) fungi are potential tools in increasing the sustainability of our food production systems. Given the demand for sustainable agriculture, the production of such inoculants has potential economic value and has resulted in a variety of commercial inoculants currently being advertised. However, their use is limited by inconsistent product efficacy and lack of consumer confidence. Here, we propose a framework that can be used to assess the quality and reliability of AM inoculants. First, we set out a range of basic quality criteria which are required to achieve reliable inoculants. This is followed by a standardized bioassay which can be used to test inoculum viability and efficacy under controlled conditions. Implementation of these measurements would contribute to the adoption of AM inoculants by producers with the potential to increase sustainability in food production systems.

Abstract: The need for redesigning existing "data-intensive" applications to execute in mobile environments has led to an enormous proliferation of systems that propose a solution to different issues. In this paper we classify these approaches and give evidence for the lack of code reuse efforts. We introduce the MoleNE system as a set of generic services that aims at providing a systematic approach to build mobile applications. Depending on the generic nature of the provided services, the MoleNE architecture is divided into a toolkit and a framework layer. The former provides device management related services. Services of the framework are application-dependent and their customization generates an instance of MoleNE that is well suited to the targeted application. Further, services design allows to easily modify their behavior depending on the application needs.

Abstract: Background: Although several studies have reported genomic characterisation of primary breast tumors, little is known about the genomic alterations of the metastatic tissues. Breast cancer patients who were prospectively enrolled in a trial (SAFIR01) underwent a biopsy of metastasis. The primary aim of this biopsy was to drive the treatment according to the results of whole genome CGH arrays and sanger sequencing on two genes (PIK3CA and AKT1). The results of the clinical trial were previously reported (André et al ASCO 2013). The secondary goal of the biopsies was to perform genomic characterisation of metastases in breast cancer patients. Here we report the analyses of such metastatic samples using next generation sequencing (NGS) approaches. Patients and methods: Two approaches were applied. In order to describe the incidence of targetable genomic alterations, we performed in depth targeted sequencing on 100 genes (200x coverage) using Illumina HiSeq 2000 (DNA vision). This analysis was performed on 240 metastatic samples. The second approach was more exploratory and aimed at discovering new genes involved in the metastatic process and/or resistance to therapies. In order to achieve this goal, we performed whole exome sequencing in 100 pairs of metastatic tissue (100x) and normal DNA (Integragen Inc, Hiseq platform). Finally, phosphor-S6K staining was performed in 300 samples in order to explore the activation status of mTOR pathway in metastatic disease, and to correlate with genomic data. Results: Targeted sequencing was performed on 240 metastatic samples in order to report the prevalence of targetable genomic alterations in metastatic breast cancer. Results are available for the first 159 samples. In addition to the already reported PIK3CA (26%) and AKT1 mutations (4%), NGS identified mutations of PTEN (4%), ERBB2 (2%), K-Ras (1%), ATM (2%), CDH1 (2%), GATA3 (2%), PTPN11 (1%), PTPRD (1%), ROS1 (1%). Results on the 81 samples, together with whole exome sequencing (n = 100) and phosphoproteins are being analysed and results will be available mid-november 2013. Conclusion: This is the first large study that aims at defining the genomic landscape of metastatic samples. Results will provide insights into the prevalence of targetable genomic alterations in metastatic tissue, together with candidate genes involved in the metastatic process and resistance to therapies. Abstract: Summary form only given. The amplification of multiple signals on board telecommunication satellites is normally implemented with one TWT per channel. Thus each TWT must be able to amplify the maximum peak power required for its channel, even though it is very unlikely that all channels will require the maximum power at the same time. A flexible architecture - the multiport power amplifier (MPA) (or Butler matrix) - solves this problem by sharing the power of several TWTs between the channels. The challenge lies in the paralleling of several TWTs whose insertion phase is very sensitive to voltage drifts. The strong non-linear characteristics of the TWTs also deteriorate the isolation between the channels. Hopefully, the advent of linearized TWTs (LTWT) at 20 GHz overcomes the linearity issue. We present the main LTWTA parameters relevant for the operation in the MPA and show experimental results of a matrix of two LTWTAs.

Abstract: Thales electron devices has many years of experience in the manufacture of traveling wave tube amplifiers (TWTA) for space probes, which must send large quantities of data back to Earth. The application of TWTA have traditionally used the X band, around 8 GHz, with RF power ranging from 20 to 40 watts. However, the upcoming generation of spacecraft will also use the Ka band, around 32 GHz, at relatively high power levels of 60 watts or more. This paper presents the latest results at TWTA level and explores the different possibilities for higher power.

Abstract: Abstract Background Although repetitive transcranial magnetic stimulation (‘TMS’) is becoming a gold standard treatment for pharmacoresistant depression, we lack neural target biomarkers for identifying who is most likely to respond to TMS and why. To address this gap in knowledge we evaluate neural targets defined by activation and functional connectivity of the dorsolateral prefrontal cortex-anchored cognitive control circuit, regions of the default mode network and attention circuit, and interactions with the subgenual anterior cingulate. We evaluate whether these targets and interactions between them change in a dose-dependent manner, whether changes in these neural targets correspond to changes in cognitive behavioral performance, and whether baseline and early change in neural target and cognitive behavioral performance predict subsequent symptom severity, suicidality, and quality of life outcomes. This study is designed as a pragmatic, mechanistic trial partnering with the National Clinical TMS Program of the Veteran’s Health Administration. Methods Target enrollment consists of 100 veterans with pharmacoresistant Major Depressive Disorder (MDD). All veterans will receive a clinical course of TMS and will be assessed at ‘baseline’ pre-TMS commencement, ‘first week’ after initiation of TMS (targeting five sessions) and ‘post-treatment’ at the completion of TMS (targeting 30 sessions). Veterans will be assessed using functional magnetic resonance imaging (fMRI), a cognitive behavioral performance battery, and established questionnaires. Multivariate linear mixed models will be used to assess whether neural targets change with TMS as a function of dose (Aim 1), whether extent and change of neural target relates to and predicts extent of behavioral performance (Aim 3), and whether extent of neural target change predicts improvement in symptom severity, suicidality, and quality of life (Aim 3). For all three aims, we will also assess the contribution of baseline moderators such as biological sex and age. Discussion To our knowledge, our study will be the first pragmatic, mechanistic observational trial to use fMRI imaging and cognitive-behavioral performance as biomarkers of TMS treatment response in pharmacoresistant MDD. The results of this trial will allow providers to select suitable candidates for TMS treatment and better predict treatment response by assessing circuit connectivity and cognitive-behavioral performance at baseline and during early treatment. Abstract: The important differences between wireless and wired environments make it indispensable for applications to adapt to the type of environment in which they execute. Solutions provided by existing approaches are often embedded in the heart of the applications functionalities making them more difficult to develop and to maintain. We propose the utilization of an adaptive framework, MoleNE, which provides applications with a set of services as an abstraction of concepts usually utilized to manage mobility issues. Services are built as a set of MoleNE components that can be customized for different applications. Further a MoleNE component can be provided with a special object the Controller, which performs the adaptation of the component behavior depending on the execution conditions. This paper describes the architecture of MoleNE components and gives evidence of the easy use of the adaptation mechanisms by presenting an adaptation strategy for a transactional hospital application.

Abstract: Tree-ring archives are one of the main sources of information to reconstruct climate variations over the last millennium with annual resolution. The links between tree-ring proxies and climate have usually been estimated using statistical approaches, assuming linear and stationary relationships. Both assumptions may be inadequate, but this issue can be overcome by ecophysiological modelling based on mechanistic understanding. In this respect, the model MAIDEN (Modeling and Analysis In DENdroecology) simulating tree-ring growth from daily temperature and precipitation, considering carbon assimilation and allocation in forest stands, may constitute a valuable tool. However, the lack of local meteorological data and the limited characterization of tree species traits can complicate the calibration and validation of such a complex model, which may hamper palaeoclimate applications. The goal of this study is to test the applicability of the MAIDEN model in a palaeoclimate context using as a test case tree-ring observations covering the 20th century from 21 Eastern Canadian taiga sites and 3 European sites. More specifically, we investigate the model sensitivity to parameter calibration and to the quality of climatic inputs, and we evaluate the model performance using a validation procedure. We also examine the added value of using MAIDEN in palaeoclimate applications compared to a simpler tree-growth model, i.e. VS-Lite. A Bayesian calibration of the most sensitive model parameters provides good results at most of the selected sites with high correlations between simulated and observed tree growth. Although MAIDEN is found to be sensitive to the quality of the climatic inputs, simple bias correction and downscaling techniques of these data improve significantly the performance of the model. The split-sample validation of MAIDEN gives encouraging results but requires long tree ring and meteorological series to give robust results. We also highlight a risk of overfitting in the calibration of model parameters that increases with short series. Finally, MAIDEN has shown higher calibration and validation correlations in most cases compared to VS-Lite. Nevertheless, this latter model turns out to be more stable over calibration and validation periods. Our results provide a protocol for the application of MAIDEN to potentially any site with tree-ring width data in the extratropical region.

Abstract: Abstract Diabetes mellitus is a chronic disease characterized by high social, economic and health burden, mostly due to the high incidence and morbidity of diabetes complications. Numerous studies have shown that optimizing metabolic control may reduce the risk of micro and macrovascular complications related to the disease, and the algorithms suggest that an appropriate and timely step of care intensification should be proposed after 3 months from the failure to achieve metabolic goals. Nonetheless, many population studies show that glycemic control in diabetic patients is often inadequate. The phenomenon of clinical inertia in diabetology, defined as the failure to start a therapy or its intensification/de-intensification when appropriate, has been studied for almost 20 years, and it is not limited to diabetes care, but also affects other specialties. In the present manuscript, we have documented the issue of inertia in its complexity, assessing its dimensions, its epidemiological weight, and its burden over the effectiveness of care. Our main goal is the identification of the causes of clinical inertia in diabetology, and the quantification of its social and health-related consequences through the adoption of appropriate indicators, in an effort to advance possible solutions and proposals to fight and possibly overcome clinical inertia, thus improving health outcomes and quality of care.

169. HETEROFOR 1.0: a spatially explicit model for exploring the response of structurally complex forests to uncertain future conditions – Part 1: Carbon fluxes and tree dimensional growth

Abstract: Given the multiple abiotic and biotic stressors resulting from global changes, management systems and practices must be adapted in order to maintain and reinforce the resilience of forests. Among others, the transformation of monocultures into uneven-aged and mixed stands is an avenue to improve forest resilience. To explore the forest response to these new silvicultural practices under a changing environment, one needs models combining a process-based approach with a detailed spatial representation, which is quite rare. We therefore decided to develop our own model (HETEROFOR for HETEROgeneous FORest) according to a spatially explicit approach, describing individual tree growth based on resource sharing (light, water and nutrients). HETEROFOR was progressively elaborated within Capsis (Computer-Aided Projection for Strategies in Silviculture), a collaborative modelling platform devoted to tree growth and stand dynamics. This paper describes the carbon-related processes of HETEROFOR (photosynthesis, respiration, carbon allocation and tree dimensional growth) and evaluates the model performances for three broadleaved stands with different species compositions (Wallonia, Belgium). This first evaluation showed that HETEROFOR predicts well individual radial growth (Pearson's correlation of 0.83 and 0.63 for the European beech and sessile oak, respectively) and is able to reproduce size–growth relationships. We also noticed that the net to gross primary production (npp to gpp) ratio option for describing maintenance respiration provides better results than the temperature-dependent routine, while the process-based (Farquhar model) and empirical (radiation use efficiency) approaches perform similarly for photosynthesis. To illustrate how the model can be used to predict climate change impacts on forest ecosystems, we simulated the growth dynamics of the mixed stand driven by three IPCC climate scenarios. Abstract: Informe sobre interacció agonística en granotes de Malàisia del gènere Gephyromantis (Anura, Mantellidae) Hi ha informacions sobre el comportament territorial de diverses famílies d’amfibis, però n’hi ha molt poques de referents a granotes de Madagascar, malgrat la diversitat extremadament alta d’espècies existent al país. Aquí es descriu l’enfrontament agonístic de dos mascles del gènere Gephyromantis, endèmic de Madagascar. El combat físic va anar precedit d’un canvi en la crida d’advertiment, que va passar a un to més agressiu, per part del mascle resident davant l’aproximació de l’intrús. Tot i que fins ara no s’havia informat d’interacció física, el canvi en el tipus de crida es similar a l’observat en altres Gephyromantis. El combat va durar aproximadament un minut i durant aquest temps cadascun dels mascles va tractar de sotmetre l’altre, fins que l’intrús es va retirar.

171. HETEROFOR 1.0: a spatially explicit model for exploring the response of structurally complex forests to uncertain future conditions – Part 2: Phenology and water cycle

Abstract: Climate change affects forest growth in numerous and sometimes opposite ways, and the resulting trend is often difficult to predict for a given site. Integrating and structuring the knowledge gained from the monitoring and experimental studies into process-based models is an interesting approach to predict the response of forest ecosystems to climate change. While the first generation of models operates at stand level, one now needs spatially explicit individual-based approaches in order to account for individual variability, local environment modification and tree adaptive behaviour in mixed and uneven-aged forests that are supposed to be more resilient under stressful conditions. The local environment of a tree is strongly influenced by the neighbouring trees, which modify the resource level through positive and negative interactions with the target tree. Among other things, drought stress and vegetation period length vary with tree size and crown position within the canopy. In this paper, we describe the phenology and water balance modules integrated in the tree growth model HETEROFOR (HETEROgenous FORest) and evaluate them on six heterogeneous sessile oak and European beech stands with different levels of mixing and development stages and installed on various soil types. More precisely, we assess the ability of the model to reproduce key phenological processes (budburst, leaf development, yellowing and fall) as well as water fluxes. Two two-phase models differing regarding their response function to temperature during the chilling period (optimum and sigmoid functions) and a simplified one-phase model are used to predict budburst date. The two-phase model with the optimum function is the least biased (overestimation of 2.46 d), while the one-phase model best accounts for the interannual variability (Pearson's r=0.68). For the leaf development, yellowing and fall, predictions and observations are in accordance. Regarding the water balance module, the predicted throughfall is also in close agreement with the measurements (Pearson's r=0.856; bias =-1.3 %), and the soil water dynamics across the year are well reproduced for all the study sites (Pearson's r was between 0.893 and 0.950, and bias was between −1.81 and −9.33 %). The model also reproduced well the individual transpiration for sessile oak and European beech, with similar performances at the tree and stand scale (Pearson's r of 0.84–0.85 for sessile oak and 0.88–0.89 for European beech). The good results of the model assessment will allow us to use it reliably in projection studies to evaluate the impact of climate change on tree growth in structurally complex stands and test various management strategies to improve forest resilience.

Abstract: Alcohol use disorder (AUD) continues to be challenging to treat despite the best available interventions, with two-thirds of individuals going on to relapse by 1 year after treatment. Recent advances in the brain-based conceptual framework of addiction have allowed the field to pivot into a neuromodulation approach to intervention for these devastative disorders. Small trials of repetitive transcranial magnetic stimulation (rTMS) have used protocols developed for other psychiatric conditions and applied them to those with addiction with modest efficacy. Recent evidence suggests that a TMS approach focused on modulating the salience network (SN), a circuit at the crossroads of large-scale networks associated with AUD, may be a fruitful therapeutic strategy. The anterior insula or dorsal anterior cingulate cortex may be particularly effective stimulation sites given emerging evidence of their roles in processes associated with relapse.

174. Patterns of relapse of N2 nonsmall-cell lung carcinoma patients treated with preoperative chemotherapy: should prophylactic cranial irradiation be reconsidered?

Abstract: Although it induces a relevant reduction in the risk of both visceral metastases and locoregional recurrences, the combination of chemotherapy and surgery only marginally improves the survival of patients with Stage IIIA(N2) (International Union Against Cancer staging and classification system) nonsmall-cell lung carcinoma (NSCLC). The purpose of the current study was to analyze the patterns of relapse in these patients.In this study, the authors compared the patterns of relapse in 81 patients with clinically detectable N2 NSCLC who had been treated with preoperative chemotherapy with the relapse patterns of 186 comparable patients who had been treated with primary surgery. Clinically detectable N2 (cN2) denotes mediastinal lymph node enlargement on computed tomography scan, which then is confirmed by mediastinoscopy.Overall 20% of patients developed a locoregional recurrence. Chemotherapy decreased the risk of visceral metastasis as 28% of the patients preoperatively treated and 38%of those not treated with preoperative chemotherapy presented a visceral metastasis (P0.05). Preoperative chemotherapy and adenocarcinoma subtypes were associated with a higher rate of brain metastasis (P0.05). Thirty-two percent of the patients treated preoperatively and 18% of those not treated with preoperative chemotherapy presented a brain metastasis (P0.05), which was isolated in 22% and 11% of the patients, respectively (P0.05).The current study found that preoperative chemotherapy for cN2 decreases the risk of visceral metastasis but is associated with a high rate of isolated brain metastases. Prophylactic cranial irradiation may need to be reinvestigated in clinical trials, especially in patients who present with an adenocarcinoma.

176. Migration properties of the human ovarian adenocarcinoma cell line IGROV1: importance of alpha(v)beta3 integrins and vitronectin

Abstract: Cell migration of ovarian tumoral cells is essential for cell dissemination and for invasion of the submesothelial extracellular matrix (ECM). We have conducted a study of the migratory properties of an ovarian adenocarcinoma cell line (IGROV1) by using 2 distinct methods for the evaluation of cell migration. We found that in a short-term transfilter migration assay, IGROV1 cells migrated toward vitronectin, fibronectin, type IV collagen and laminin in an integrin-dependent manner. When migration was evaluated in a wound healing assay, the restitution of the wounded area was stimulated solely by added, exogenous vitronectin and was almost totally dependent on alpha(v)beta3 integrin function. Moreover, we demonstrated that alpha(v)beta3 was localized in focal contacts restricted to the leading edge of migrating cells, whereas vitronectin notably localized with actin stress fibers and cortical actin. On the other hand, several kinase inhibitors were found to impede migration of IGROV1 induced by vitronectin. It thus appears that alpha(v)beta3-vitronectin interactions lead to the activation of multiple signaling pathway including activation of protein kinase C, phosphatidyl-inositol-3-phosphate kinase and protein tyrosine kinase. The "alpha(v)beta3-vitronectin system" is therefore essential to the migration of human ovarian carcinoma cells.

Abstract: The SAFIR02-BREAST trial enrolled 1462 patients with HER2-non-overexpressing mBC to undergo genomic profiling, of whom 238 were randomised in two trials comparing maintenance treatment with targeted therapy matched to genomic alteration. PFS was improved with targeted therapy matched to genomics when alterations were classified as level I or II according to ESCAT (ESMO Scale for Clinical Actionability of Molecular Targets; adjusted HR 0.41; 90% CI 0.27-0.61), but not when alterations were unselected using ESCAT (0.77; 95% CI 0.56-1.06); there was no significant PFS improvement with targeted therapies for patients presenting with ESCAT alterations beyond level I or II. Olaparib was associated with significant benefit among 49 participants with a germline BRCA1 or BRCA2 mutation (respective HRs 0.36; 90% CI 0.14-0.89 and 0.37; 95% CI 0.17-0.78). Abstract: A 40-year-old man experiencing early satiety, a 25-1b weight loss, and intermittent nausea underwent an esophagogastroduodenoscopy, which demonstrated an ulcerated mass of the stomach extending from 43 cm to 53 cm from the incisors occupying the gastroesophageal junction, gastric cardia, fundus, and body. A biopsy of the gastric body showed diffuse infiltration of the lamina propria by poorly differentiated signet-ring cell adenocarcinoma. F-18 fluorodeoxyglucose (FDG) positron emission tomography (PET) with computed tomography demonstrated minimally increased FDG uptake relative to normal tissues. Further study to determine the use of FDG PET in the evaluation of patients with gastric signet-ring cell carcinoma is warranted.

Abstract: Background: Rare cancers sometimes present clonal evolution. Genomic characterisation of these rare entities could lead to identify new genes involved in the progression of more frequent cancers. In the present study, we have performed whole exome sequencing of three rare and aggressive histological subtypes in order to identify new targets that could subsequently be validated in common breast cancers. Methods: Whole exome sequencing was applied in ten pleiomorphic lobular carcinoma, seven micropapillary BC and eight metaplastic BC. Each cancer sample was matched with normal DNA from the same patient. Overall 50 samples were therefore sequenced. Tumor samples were selected to present >30% cancer cells. Histological diagnosis was confirmed by a BC pathologist. Whole exome sequencing (WES) was performed by BGI (Bejing Genomic Institute, Hong-Kong, China) using the Illumina Hiseq2000 platform (90bp paired-end reads, depth ≥80x). Genes recurrently mutated (with somatic mutations in >2 cases) were selected for validation. The validation by IonTorrent target sequencing was performed on additional formalin-fixed paraffin-embedded (FFPE) tumor samples (>30% of tumor cells). Results: Complete results of both WES and targeted resequencing validation are available for pleiomorphic lobular carcinoma. Nine of the 10 DNA pairs were eligible for WES. Genes already implicated in BC were significantly mutated: PIK3CA (3/9), TP53 (3/9) and CDH1 (3/9). Novel significantly mutated genes were identified: PYGM (2/9), EMR1 (2/9), ALDH1A2 (2/9), and DCLK1 (2/9). The validation using target gene sequencing was performed on the same 9 cases and on 19 additional confirmed pleiomorphic lobular BC cases (n = 28). For 8 out of the 9 cases with WES data, the results were reproducible on FFPE samples. Validation set confirmed recurrent mutations on PYGM genes (25%). PYGM is involved in glycogen metabolism. Further analyses on gene expression array revealed that PYGM is downregulated in most of the common BC samples, as compared to normal tissue, suggesting a role for this gene in the metabolism of common BC. WES data are available for micropapillary and metaplastic BC. HSPA8 (coding for a Heat shock protein) was found mutated in 3/7 of micropapillary samples. Validation on additional FFPE samples is ongoing and will be presented. WES of metaplastic BC revealed a high frequency of TP53 and PIK3CA mutations (50% in both cases). Conclusions: A novel mutated gene was identified in PLBC. PYGM is a gene involved in glycogen metabolism and was found downregulated in most of the common breast cancer. WES of micropapillary cancers suggests mutations on HSPA8, a gene encoding for a chaperon protein. Validation is ongoing. Deeper sequencing is ongoing for metaplastic BC that may help identifying new mutated genes in this multiclonal disease. Results will be presented. Abstract: Carp spermatozoa are immotile in seminal plasma or in saline solution of high osmolality (> 400 mosmol kg-1). These ‘quiescent’ spermatozoa initiate a progressive forward motility when transferred in freshwater or in saline solution with low osmolality (< 160 mosmol kg-1). In this study we investigated ‘in vitro’ the relationship between sperm ATP content (measured by bioluminescence) and sperm motility (analysed by videomicroscopy). Sperm ATP content remained high in the immobilizing medium (200 mM KCl, Tris 30 mM, pH 8.0) where no flagellar movement occurs. Dilution of these spermatozoa in the activating medium (45 mM NaCl, 5 mM KCl, Tris 30 mM, pH 8.0) triggered forward motility which varied with temperature. At 20 degrees C, sperm ATP content decreased rapidly during the progressive forward motility phase from 12 to 4 nmol/10(8) spermatozoa, concomitantly with decreases in velocity (130 to 10 microns s-1) and the beat frequency (50 to 7 Hz). An inhibitor of mitochondrial respiration (KCN 10 mM) produced a drop in sperm ATP content irrespective of the incubation medium (activating or immobilizing). A second phase of sperm motility in the activating medium was induced following a previous transfer of spermatozoa into a medium of high osmolality for a few minutes prior to the second phase. Within 10 minutes, spermatozoa recover 90% of the initial ATP level as well as forward motility. These results suggest that motility of carp spermatozoa depends on sperm ATP synthesized by mitochondrial respiration mainly stored before activation. In low osmolality conditions, the mitochondrial oxidative phosphorylation is unable to compensate for the ATP hydrolysis required to sustain motility.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract: This work aims to prepare low-cost nanocomposite and microcomposite with lower molar ratio CaO/SiO2 (0.4). Nano-silica, rice husk ash and calcined chicken eggshell have been used as silica and calcium sources. Metakaolin has been separately replaced by 0, 10 and 20 wt% of each composite in order to study their behavior on the properties of geopolymers. The hardener used is sodium waterglass from rice husk ash. The surface area of nano-silica and rice husk ash was 54.40 and 4.08 m2/g, respectively. The cumulative volumes of the control geopolymer, the ones containing 10 wt% of microcomposite and nanocomposite are 119.71, 89.92 and 110.49 mm3/g, respectively. The compressive strength of the control specimen is around 64.02 MPa. The one using 10 wt% of microcomposite was 68.97 MPa. It drops to 42.88 MPa when metakaolin was replaced by 20 wt % of microcomposite. Whereas the one using 10 wt% of nanocomposite was 30.03 MPa and it decreases to 26.05 MPa when metakaolin was substituted by 20 wt% of nanocomposite. It can be concluded that 10 wt% of microcomposite could be mixed to metakaolin for strength development and nanocomposite does not recommend to use as an additive for producing high strength of geopolymer cements.

Abstract: Amiodarone (Am) pneumonitis is currently a common and potentially severe adverse reaction, the accurate diagnosis of which remains difficult to establish.To determine the contribution of bronchoalveolar lavage (BAL) in the diagnostic workup of patients suspected of having Am pneumonitis.Diagnosis of Am pneumonitis was established on the basis of (1) development of recent symptoms and pulmonary opacities while receiving the drug, (2) exclusion of other possible causes, and (3) improvement following cessation of Am and/or steroid therapy. (4) Confirmatory changes were obtained by histopathologic examination in eight cases. BAL was performed in each patient at the time of initial evaluation.Am pneumonitis was diagnosed in 15 consecutive patients between 1985 and 1991. The disease was associated with significant morbidity and mortality. Six patients died; four died of Am pneumonitis. A neutrophilic BAL was found in nine patients (average PMN = 26.6 percent). A mixed pattern (lymphocytic + neutrophilic) was seen in four patients (average: Ly = 19.9 percent; PMN = 11.9 percent). Two patients had a normal BAL. No patient had a lymphocytic pattern. A low CD4+/CD8+ ratio was seen in two patients. A literature survey indicated 70 cases of Am pneumonitis with detailed information on BAL. The BAL pattern was mixed in 23 (33 percent), neutrophilic in 18 (26 percent), lymphocytic in 15 (21 percent), and normal in 14 (20 percent). No correlation was found between BAL pattern and prognosis. Also, BAL pattern was related neither to daily or total dose of Am nor to duration of treatment with Am.The cellular profile of BAL in Am pneumonitis is highly variable, and no cellular pattern of BAL seems to be predictive of a detrimental outcome or of irreversible fibrosis. Aside from excluding other illnesses, and due to its extreme variability, the contribution of BAL differential in the initial workup of patients suspected of having Am pneumonitis is limited.

Abstract: Background:FGFR1 is a tyrosine kinase receptor involved in cell proliferation, migration and differentiation. FGFR1 gene is amplified in about 10% of breast cancer. Several studies have previously reported that FGFR1 inhibition could lead to a decrease in cell proliferation. Recent data obtained in prostate cancer have shown that FGFR1 activation could mediate epithelial to mesenchymal transition. In the present study, we evaluated the rational to target FGFR1 in clinical setting.Material and methods:We first assessed whether FGFR1 amplification was associated with a specific breast cancer phenotype. We assessed FGFR1 amplification by CGH array on 235 samples and by FISH (Z-2072, Spec FGFR1/Cen8 dual color probe, Zytovision®) on 260 samples, not included in the CGH part. We then assessed the functional effects of FGFR1 inhibitions in vitro. The antiproliferative effect of E3810 (FGFR1 inhibitor) was assessed both on FGFR1-non amplified (T47D/MCF7) and FGFR1 amplified (SUM44PE/MDA134) cell lines. Effect of FGFR1 inhibition was assessed on gene expression profiling.Results:When assessed by FISH, FGFR1 amplification was observed in 23/260 cases. Most of the samples (62%) exhibited an ER+/PR- phenotype, suggesting activation of intracellular kinase. FGFR1 amplification was observed in 24 out of 235 samples (10%). Interestingly, FGFR1 amplification was associated with gene gain in FGF3/FGF4/FGF19 (11q13) (55% versus 22%, p=0.05). FGFR1 inhibition by E-3810 inhibits MAPk phosphorylation, cell proliferation. A differential IC 50 for cell proliferation was observed between amplified cell lines and control cell lines. Preliminary data on gene expression profiling suggest that FGFR1 inhibition on breast cancer cells could lead to significant decrease in the Sox9 expression, a protein involved in epithelial to mesenchymal transition.Conclusion:FGFR1-amplified breast cancers present a discrete phenotype, including a high rate of ER+/PR- and high rate of FGF3/4/19 gains. Pharmacologic inhibition of FGFR1 leads to cell proliferation decrease and modulation of genes involved in EMT. 195. Clinical Subtype-Derived p53 Gene Signature Is Predictive of Prognosis and Response to Chemotherapy in ER-Positive but Not in ER-Negative Breast Cancers

Abstract: BackgroundBreast cancer is a collection of molecularly distinct neoplastic diseases and therefore, we hypothesized that p53 gene mutations may lead to different transcriptional changes in the different molecular subtypes and these may translate into subtype-dependent prognostic and predictive values.MethodsWe developed gene expression-based predictors of p53 status separately for estrogen receptor-positive (ER+) and -negative (ER-) breast cancers from a publicly available data set with known p53 mutation status (n=251). We validated the two signatures on an independent cohort of cancers (n=103) with known p53 functional status and tested their prognostic and predictive values on two other cohorts of breast cancers that received no systemic adjuvant therapy (n=255; n=198), and on one cohort of ER+ patients treated with adjuvant tamoxifen (n=277). We also examined if the p53 signatures were associated with chemotherapy sensitivity in ER+ and ER- cancers, respectively in two separate neoadjuvant data sets (n=233; n=103).ResultsWe developed a 39-gene p53 signature derived from 213 ER+ and a 30-gene p53 signature derived from 38 ER- breast cancers with no overlapping genes. External validation showed a sensitivity and specificity of 89% and 54%, respectively for the 39-gene signature in ER+ breast cancers; and 82% and 61%, respectively for the 30-gene signature in ER- cancers. The 39-gene signature was predictive of worse distant metastasis free survival (DMFS) in ER+ cancers with p53 dysfunction in both prognostic data sets (Hazard ratio (HR): 2.3 (95% confidence interval (CI):1.25-4.23, p=0.005 and HR:2.17 (95%CI:0.85-5.56, p=0.09). Abstract: Abstract #2029 Background: Fine needle aspiration (FNA) for breast cancer diagnosis, a simple, quick and cheap procedure, is limited by a significant number of inconclusive exams. In the present study, we have analyzed whether the expression of a set of exonic probes could accurately diagnose breast cancer of FNA biopsies. Patients and methods: cDNA from 165 breast lesions (120 cancers), obtained by FNA in 2006-2007, and for which RIN score was > 6, was hybridized on splice arrayTM in two separate series (training/validation)(data set Andre et al SABCS 2008). We focused on 703 680 evidenced probe sets. The actual diagnosis of breast cancer was established by pathological examination at surgery. Associations between exonic probes and diagnosis were assessed by correlation coefficients (using t-tests) and a nearest centroid prediction rule was determined on the training set of 94 samples (70 cancers) in order to classify lesions as malignant or benign. Using the validation set of 71 samples (50 cancers), the performance of the prediction rule was evaluated and the proportion of explained variation (PEV) estimated in a logistic regression model (including only variables significant by a likelihood ratio test).
