Abstract

The genotype‐first approach has been successfully applied and has elucidated several subtypes of autism spectrum disorder (ASD). However, it requires very large cohorts because of the extensive genetic heterogeneity. We investigate the alternate possibility of whether phenotype‐specific genes can be identified from a small group of patients with specific phenotype(s). To identify novel genes associated with ASD and abnormal head circumference using a phenotype‐to‐genotype approach, we performed whole‐exome sequencing on 67 families with ASD and abnormal head circumference. Clinically relevant pathogenic or likely pathogenic variants account for 23.9% of patients with microcephaly or macrocephaly, and 81.25% of those variants or genes are head‐size associated. Significantly, recurrent pathogenic mutations were identified in two macrocephaly genes (PTEN, CHD8) in this small cohort. De novo mutations in several candidate genes (UBN2, BIRC6, SYNE1, and KCNMA1) were detected, as well as one new candidate gene (TNPO3) implicated in ASD and related neurodevelopmental disorders. We identify genotype‐phenotype correlations for head‐size‐associated ASD genes and novel candidate genes for further investigation. Our results also suggest a phenotype‐to‐genotype strategy would accelerate the elucidation of genotype‐phenotype relationships for ASD by using phenotype‐restricted cohorts. [ABSTRACT FROM AUTHOR]

Abstract

BACKGROUND: Oyster polypeptides have various biofunctions, such as anti‐cancer and anti‐oxidative stress, but whether it has the protective effects to primary ovarian failure (POF) remains poorly understand. To address this issue, daily gavage of oyster polypeptides was performed to investigate their protective effect, basing on d‐galactose‐induced POF model in C57BL/6 female mice. RESULTS: Oyster polypeptides restored the irregular estrous cycles and the abnormal serum follicle stimulating hormone (FSH), luteinizing hormone (LH) and progesterone (P) levels as well as the decreased mRNA expression level of Amh that were induced by d‐galactose. The follicle development of POF mice was improved by increasing the primordial follicle ratio and decreasing the atretic follicle number after oral administration of oyster polypeptides. Moreover, in the oyster polypeptides treated mice, the total superoxide dismutase (T‐SOD) activity was significantly increased, while the malondialdehyde levels were significantly decreased. The mRNA expression levels of stress‐related genes (SOD2, SIRT1 and FOXO3a) were remarkably up‐regulated after d‐galactose induction, but the up‐regulation was weakened or disappeared by the gavage of oyster polypeptides. In addition, oyster polypeptides treatment also reduced the apoptosis of the ovarian granulosa cells and down‐regulated the mRNA expression levels of apoptosis‐related genes (p53 and Bad but not Bcl‐2). CONCLUSION: This study reveals that oyster polypeptides may protect ovary against d‐galactose‐induced POF by their anti‐oxidative stress activity to rescue d‐galactose‐induced ovarian oxidative damage and therefore to prevent ovarian cells apoptosis, thereby tipping the abnormality trigged by POF to get close to the normal levels. © 2019 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Abstract

In order to solve the problem that the mutual information function is easy to fall into local optimal values because of much local extremism in the mutual information registration method, a multi-resolution medical image registration algorithm based on firefly algorithm and Powell is put forward in this paper. The normalized mutual information is used as the similarity measure in the algorithm, and the multi-resolution strategy based on wavelet transformation is adopted in the process of searching the best value. In the lower resolution image, the firefly algorithm is used for the imprecise registration result, And in the higher resolution image, the Powell algorithm is adopted for the better registration result. The experimental results show that this algorithm can effectively overcome the problem that the mutual information function is easy to get into local optimal values and improve the precision of registration result obviously.

Abstract

In this paper, the design for a dual core FFT processor based on CORDIC algorithm is presented. This design extracts the 2-base successions as the foundation, takes the FFT butterfly-shaped arithmetical unit as the object, uses the CORDIC algorithm superiority in the vector computation to simply the revolving factor calculation, and employs the assembly line technology to enhance the turnover rate for the whole system. This FFT processor has many characteristics with the simple hardware architecture, flexible disposition, low component coupling, high precision and stable running, can perform the high speed fixed-point real-time FFT operation. Experiment carried on the gate level simulation in Altera chip EP2C35F672C6 shows that this design can satisfy the 50MHz system clock.

Abstract

This paper studies the ventilation in guest room in hotel. To air-conditioning system with fan and fresh air, the pollutants indoor are mainly removed by fresh air. Enlarging the fresh air volume can develop the indoor air quality, but goes against to the energy saving. This paper simulates the CO2 concentration indoor under different fresh air volume when the bathroom door opens and closed. The result indicates that only enlarging the fresh air volume affects the IAQ slightly, and exhaust fan can satisfy the ventilation indoor whether the bedroom door opens or closed.

Abstract

This study was conducted to evaluate the adjuvant potential of sulfated Radix Cyathulae officinalis Kuan polysaccharides (sRCPS) and their effects on specific cellular and humoral immune responses to hepatitis B subunit vaccine in mice. Our data demonstrated that sRCPS significantly promoted the rHBsAg-specific IgG, IgG1, IgG2a, and IgG2b antibody titers, the activities of natural killer cells (NK) and cytotoxic T lymphocytes (CTL), T cells proliferation, and phagocytic capacity of peritoneal macrophages. Furthermore, sRCPS increased the levels of IL-4, IL-2, and IFN-γ in CD4+T cells and the level of IFN-γ in CD8+T cells. In addition, sRCPS enhanced the expression of CD40+, CD80+, CD86+, MHC I and MHC II in dendritic cells (DCs) and upregulated the mRNA levels of MHC I, MHC II. sRCPS downregulated the frequency of CD4+CD25+Foxp3+ Treg cells. sRCPS increased both cellular and humoral immune responses by upregulating DC maturation, and suppressing the frequency of Treg cells.
