The emission of chemicals into the environment has increased in a not negligible way as a result of the phenomenon of globalization and industrialization, potentially also affecting areas always considered as “uncontaminated”. In this paper, five “uncontaminated” areas were analyzed in terms of the presence of polycyclic aromatic hydrocarbons (PAHs) and heavy metals (HMs), comparing them with an “environmental blank”. Chemical analyses were carried out using standardized protocols. The ‘environmental blank’ results revealed the presence of Cu (−1), Ni (−1), and Zn (−1) as HMs and fluorene (−1) and phenanthrene (−1) as PAHs. However, regarding the results of the pollution status of the areas under study, fluorene (#S1, 0.34 ng g−1; #S2, 4.3 ng g−1; #S3, 5.1 ng g−1; #S4, 3.4 ng g−1; #S5, 0.7 ng g−1) and phenanthrene (#S1, 0. 24 ng g−1; #S2, 3.1 ng g−1; #S3, 3.2 ng g−1; #S4, 3.3 ng g−1; #S5, 0.5 ng g−1) were found in all areas, while the other PAHs investigated were detected at a concentration averaging less than 3.3 ng g−1. HMs were found in all of the investigated areas. In particular, Cd was detected in all areas with an average concentration of less than 0.036 μg g−1, while Pb was absent in area #S5, but present in the other areas with an average concentration of less than 0.018 μg g−1.

In this work we investigated the synthesis and the characterization of electrospun polyacrylonitrile (PAN) and polymethyl methacrylate (PMMA) stabilized in air, made in a 5:1 ratio, used as sorbent for the solid-phase extraction of fluoroquinolones in plasma samples and the following quantification in UHPLC-PDA. Preliminary analyses of viscosity were carried out on the polymer solution to be sure about the electrospinability. Characterizations were performed on the electrospun membrane to evaluate the morphology (SEM scanning electron microscopy and AFM atomic force microscopy), the thermal degradation behavior (TGA thermogravimetric analysis), the porosity and the surface area (BET, Brunauer Emmett Teller), and the quantitative and qualitative distribution of atomic structures (FTIR infrared analysis in Fourier transform and EDX Energy Dispersive X-ray analysis). A solid-phase extraction method was developed by studying parameters such as the amount of sorbent and the pH of the sample. Finally, a UHPLC-PDA method for the analysis of fluoroquinolones was developed and validated in accordance with the guidelines and successfully applied. The use of the prepared sorbent combined with UHPLC-PDA has allowed the development of a method whose strengths are its speed, accuracy, sensitivity, and high recoveries.

In this work, a sorbent was prepared from wastepaper samples enriched with iron oxide particles and graphene oxide and used in the solid phase extraction of antibiotics. The precursor underwent a carbothermal reduction to promote the formation of paramagnetic phases useful for the recovery of the sorbent during the analysis, and to disperse and fix graphene and the iron oxide in a durable way throughout the cellulose structure. Characterizations were carried out to evaluate the composition (Raman, XRD and EDX) and the morphological structure (SEM) of the material. A UHPLC-PDA method was developed for the simultaneous determination of antibiotics from different drug families (carbapenems, fluoroquinolones, β-lactams) using a 120 SB-C 18 poroshell column (50 × 2.1 mm I.D., 2.7 um particle size) and a mobile phase consisting of 10 mM acetate buffer at pH 5 (Line A) and acetonitrile (Line B) both containing 0.1% of triethylamine. A gradient elution was used for the separation of the analytes, while for the quantitative analysis each analyte was determined at its maximum wavelength. Several experiments were carried out to evaluate the influence of different parameters involving the dispersive magnetic solid phase extraction of these analytes. Samples were extracted using 25 mg of sorbent at pH 5 and desorbed in 5 min using methanol. We report herein on some of the outstanding advantages of using carbon-based sorbent, such as lower toxicity, scalability, improved absorption capacity, target selectivity and stability in acidic medium. Moreover, from the results obtained it is evident that, despite the use of some recycled materials, the performances obtained were comparable or even superior to the methods reported in the literature.

There are few published data on the accurate on-farm diagnosis of failure of transfer of passive immunity (FTPI) in beef–suckler calves. This observational study aimed to evaluate the diagnostic performance and differences among four types of refractometers for assessing FTPI in Chianina beef–suckler calves. Blood samples were collected from 85 Chianina calves aged 2–7 days. The serum immunoglobulin G (IgG) concentration was measured using radial immunodiffusion (RID), digital and optical serum total protein (STP) refractometers, and digital and optical serum refractometers. The diagnostic performance of the refractometers was determined based on the serum IgG threshold of 16 g/L (measured by RID). A receiver operating characteristic (ROC) curve was used to identify the optimal cutoff values for all refractometers. The RID IgG concentration was positively correlated with all four refractometers (correlation coefficient: 0.75–0.84). ROC analysis yielded optimal cutoff values for predicting FTPI of 51 g/L (sensitivity (Se)=0.63 and specificity (Sp)=0.96) and 52 g/L (Se = 0.69 and Sp = 0.90) for the digital and optical STP refractometers, respectively. At the threshold of 8.3% Brix, the Se and Sp were 0.66 and 0.92 for the optical Brix and 0.77 and 0.92 for the digital Brix refractometer, respectively. All four refractometers were useful for assessing FTPI in Chianina calves. However, the digital Brix had the highest combined diagnostic accuracy for FTPI. The on-farm use of refractometers to assess FTPI can become part of routine monitoring of the colostrum management program in beef–suckler calf herds.Highlights The cutoff values were 51 and 52 g/L for the digital and optical serum total protein refractometers, respectively. The digital Brix refractometer was the most accurate for the detection of calves with inadequate transfer of passive immunity.

Abstract Background Toxoplasmosis is one of the most common parasitic infections in both humans and animals. It is a frequent cause of abortion and stillbirth in intermediate hosts, especially sheep and goats but rarely causes fatal clinical form in adult animals. Case presentation In contrast, the study reports an unusual fatal case of toxoplasmosis in a young goat naturally infected with type II strain of Toxoplasma gondii. A three-month-old female goat was presented with dyspnea and died few days later. Grossly, lungs were firm, edematous and mottled with disseminated whitish areas. Generalized lymphadenopathy was found. The histopathological examination showed necrotic interstitial bronchopneumonia and necrotizing lymphadenitis with intralesional free and clustered within macrophages tachyzoites of T. gondii. DNA extracted from lungs and lymph nodes was positive for T. gondii by a fast qPCR. PCR-RFLP analysis and sequencing of GRA6 gene showed that the isolated strains belonged to type II genotype. Conclusions This is an unusual report of acute systemic toxoplasmosis caused by the type II strain of T. gondii with a fatal outcome in a young goat.

G proteins and their cognate G protein-coupled receptors (GPCRs) function as critical signal transduction molecules that regulate cell survival, proliferation, motility and differentiation. The aberrant expression and/or function of these molecules have been linked to the growth, progression and metastasis of various cancers. As such, the analysis of mutations in the genes encoding GPCRs, G proteins and their downstream targets provides important clues regarding how these signaling cascades contribute to malignancy. Recent genome-wide sequencing efforts have unveiled the presence of frequent mutations in GNA13, the gene encoding the G protein Gα13, in Burkitt's lymphoma and diffuse large B-cell lymphoma (DLBCL). We found that mutations in the downstream target of Gα13, RhoA, are also present in Burkitt's lymphoma and DLBCL. By multiple complementary approaches, we now show that that these cancer-specific GNA13 and RHOA mutations are inhibitory in nature, and that the expression of wild-type Gα13 in B-cell lymphoma cells with mutant GNA13 has limited impact in vitro but results in a remarkable growth inhibition in vivo. Thus, although Gα13 and RhoA activity has previously been linked to cellular transformation and metastatic potential of epithelial cancers, our findings support a tumor suppressive role for Gα13 and RhoA in Burkitt's lymphoma and DLBCL.

Abstract Introduction Severe pneumonia caused by multidrug-resistant Acinetobacter baumannii (MDR-AB) remains a difficult-to-treat infection. Considering the poor lung penetration of most antibiotics, the choice of the better antibiotic regimen is debated. Methods We performed a prospective, observational, multicenter study conducted from January 2017 to June 2020. All consecutive hospitalized patients with severe pneumonia due to MDR-AB were included in the study. The primary endpoint of the study was to evaluate risk factors associated with survival or death at 30 days from pneumonia onset. A propensity score for receiving therapy with fosfomycin was added to the model. Results During the study period, 180 cases of hospital-acquired pneumonia, including ventilator-associated pneumonia, caused by MDR-AB strains were observed. Cox regression analysis of factors associated with 30-day mortality, after propensity score, showed that septic shock, and secondary bacteremia were associated with death, while a fosfomycin-containing regimen was associated with 30-day survival. Antibiotic combinations with fosfomycin in definitive therapy for 44 patients were: fosfomycin + colistin in 11 (25%) patients followed by fosfomycin + carbapenem + tigecycline in 8 (18.2%), fosfomycin + colistin + tigecycline in 7 (15.9%), fosfomycin + rifampin in 7 (15.9%), fosfomycin + tigecycline in 6 (13.6%), fosfomycin + carbapenem in 3 (6.8%), and fosfomycin + aminoglycoside in 2 (4.5%). Conclusions This real-life clinical experience concerning the therapeutic approach to severe pneumonia caused by MDR-AB provides useful suggestions to clinicians, showing the use of different antibiotic regimens with a predominant role for fosfomycin. Further randomized clinical trials are necessary to confirm or exclude these observations.

The COVID-19 outbreak triggered a global crisis with long-term effects on people’s daily lives. It has altered surgical practice and education and imposed major changes in healthcare systems and resources. In order to analyze the impact the COVID-19 pandemic has had on Romanian Plastic Surgery residents, we carried out an anonymous questionnaire through Google forms. Survey items addressed aspects such as how the pandemic has affected their surgical practice, their exposure to the virus, and the protective measures that have been implemented to minimize risk. Among other results, our study found that nearly 1 in 5 respondents had contact with a coronavirus patient, that most of the patients were tested at admission, mostly using RT-PCR, and that various combinations of personal protective equipment had been used. Most participants also felt that their surgical training has been negatively affected, despite attendance at online courses and webinars. Although the pandemic has brought significant changes to the daily lives of residents and to the residency training, the Plastic Surgery community has been brought closer than ever and reminded that, through unified efforts, such obstacles can be overcome.

Abstract Background COVID-19, the disease caused by the highly infectious and transmissible coronavirus SARS-CoV-2, has quickly become a morbid global pandemic. Although the impact of SARS-CoV-2 infection in children is less clinically apparent, collecting high-quality biospecimens from infants, children, and adolescents in a standardized manner during the COVID-19 pandemic is essential to establish a biologic understanding of the disease in the pediatric population. This biorepository enables pediatric centers world-wide to collect samples uniformly to drive forward our understanding of COVID-19 by addressing specific pediatric and neonatal COVID-19-related questions. Methods A COVID-19 biospecimen collection study was implemented with strategic enrollment guidelines to include patients seen in urgent care clinics and hospital settings, neonates born to SARS-CoV-2 infected mothers, and asymptomatic children. The methodology described here, details the importance of establishing collaborations between the clinical and research teams to harmonize protocols for patient recruitment and sample collection, processing and storage. It also details modifications required for biobanking during a surge of the COVID-19 pandemic. Results Considerations and challenges facing enrollment of neonatal and pediatric cohorts are described. A roadmap is laid out for successful collection, processing, storage and database management of multiple pediatric samples such as blood, nasopharyngeal and oropharyngeal swabs, sputum, saliva, tracheal aspirates, stool, and urine. Using this methodology, we enrolled 327 participants, who provided a total of 972 biospecimens. Conclusions Pediatric biospecimens will be key in answering questions relating to viral transmission by children, differences between pediatric and adult viral susceptibility and immune responses, the impact of maternal SARS-CoV-2 infection on fetal development, and factors driving the Multisystem Inflammatory Syndrome in Children. The specimens in this biorepository will allow necessary comparative studies between children and adults, help determine the accuracy of current pediatric viral testing techniques, in addition to, understanding neonatal exposure to SARS-CoV-2 infection and disease abnormalities. The successful establishment of a pediatric biorepository is critical to provide insight into disease pathogenesis, and subsequently, develop future treatment and vaccination strategies.

Three years after the earthquake that struck central Italy, a number of pressing points need to be addressed as part of a far-ranging discussion that seeks to identify the steps to be taken in response, including: the widespread agreement on the need for reconstruction efforts which ensure a high level of security; the importance of preserving the urban fabric of ‘minimal’ population centres that are unquestionably intertwined with their surrounding landscapes; the need to acknowledge the most valuable features of historic downtown areas, including their undeniable fact of their intrinsic fragility in the face of seismic events, even though construction techniques developed and refined over time have provided them with a certain resilience. While use is made of a frankly contemporary idiom, when needed to remedy shortcomings, a reconstruction grounded in a critical understanding of the ‘sense of place’ must guarantee that the identifying features of historic downtown areas remain in place (at least in terms of the lay of the land and spatial relations) while, at the same time, ensuring that the constantly evolving memories which render such areas unique are also preserved, so as to allow the past to play its rightful role in the planning of the future.

Compact spectrometers can be useful in many applications and many sophisticated architectures have been proposed. In this work, the authors show that with an evaporating droplet on a fiber tip, spectrometry can be robustly and accurately performed with a simple and passive microfluidic system.

Introdução/Objetivo: O SARS-COV-2 mostrou-se uma grave ameaça à saúde global. O grande número de infectados desencadeou altas taxas de mortalidade e sobrecarga do sistema de saúde. O prognóstico da doença é muito variável e dependente de diversos fatores. Dessa forma, neste trabalho buscou-se identificar a mortalidade dos pacientes por covid-19 admitidos nas Unidades de Terapia Intensiva em um hospital brasileiro, bem como suas características clínicas e epidemiológicas. Métodos: Trata-se de pesquisa observacional, transversal, retrospectiva, descritiva e pretende analisar o desfecho de mortalidade em pacientes COVID-19, no Hospital Geral da Fundação Universidade de Caxias do Sul. Os critérios de inclusão foram período de internação entre 1 de abril de 2020 e 30 de abril de 2021, idade maior de 18 anos, internação em UTI adulto por no mínimo 24 horas e testagem positiva para COVID-19. Foram avaliados dados como o sexo, comorbidades prévias e tempo de internação em UTI. Resultados: Foram avaliados 170 pacientes, sendo que 55,5% do sexo masculino. A idade média foi 59 anos - 57 para mulheres e 61 para homens. 55% evoluiram a óbito em decorrência de complicações da infecção, 33% do homens e 21% mulheres. A idade média para mulheres foi de 62 anos e para os homens, 64 anos. O tempo de internação em UTI até o óbito foi em média 16 dias (13 para mulheres e 16 para homens). Desses pacientes, 87% possuiam comorbidades, sendo as três mais prevalentes a hipertensão arterial sistêmica (59%), a obesidade (41%) e o diabetes mellitus (40%). Conclusão: Podemos inferir que o desfecho mais prevalente foi o óbito. Desses, o perfil mais prevalente foi de homens idosos. Os pacientes do sexo masculino que necessitaram de internação em UTI e foram a óbito, possuíam idade mais avançada do que os pacientes do sexo feminino. Concluímos que uma elevada porcentagem de pacientes com o desfecho de óbito possuíam pelo menos uma comorbidade associada, sendo as mais frequentes a hipertensão arterial, a obesidade e a diabetes.

Introdução/Objetivo: A Covid-19 pode apresentar-se de forma assintomática até quadros críticos de insuficiência respiratória aguda com complicações sistêmicas. A mortalidade associada é significativa, com necessidade de internação em unidade de terapia intensiva. Assim como a população que evolui ao óbito, os sobreviventes à doença grave merecem atenção no intuito de fornecer conhecimento para ações que ajudem a reduzir a mortalidade. Métodos: Este é um projeto de pesquisa observacional, transversal, retrospectivo, descritivo e que pretende analisar o desfecho sobrevida em pacientes COVID-19, no Hospital Geral da Fundação Universidade de Caxias do Sul, entre 01 de abril de 2020 e 30 de abril de 2021. Os critérios de inclusão adotados foram: idade maior de 18 anos, internação em UTI adulto por no mínimo 24 horas, testagem positiva para COVID-19. A população estudada foi a de pacientes com infecção por COVID-19 que sobreviveram após a internação na UTI. Foram avaliados os seguintes dados: Sobrevivência global, Idade, comorbidades, Tempo de internação, Ventilação mecânica e Ventilação em posição prona, Complicações, e Realização de traqueostomia. Resultados: Foram avaliados 192 pacientes, 53% faleceram e 47% sobreviveram. A idade média dos sobreviventes foi 55 para homens e 52 para mulheres. Comorbidades se apresentaram em 79 pacientes, 34 nos homens e 45 nas mulheres. A incidência de comorbidades foi: HAS, 47%; DM 25%; Sobrepeso 26%; Obesos, 44%; DPOC, 7%; Cardiopatia isquêmica 2%; ICC 4%; Valvulopatia 1%; Uso anticoagulante 4%; Doença reumática 4%; Insuficiência Renal, 4% pacientes. Em 68% dos casos utilizou-se ventilação mecânica. A VM em posição pronada foi aplicada 38%. Traqueostomia foi realizada em 32%. A incidência de complicações foi 130. 13 casos de tromboembolia pulmonar (TEP), 41 de BCP, 26 de insuficiência renal aguda, 26 de escaras, 15 de derrame pleural, 5 de pneumotórax e 1 de isquemia periférica. 6 pacientes necessitaram de hemodiálise e 2 de diálise peritoneal. Conclusão: A incidência de comorbidades entre os sobreviventes que necessitaram de internação em UTI foi maior entre as mulheres. 71% dos sobreviventes possuíam IMC elevado e 87% apresentavam alguma comorbidade, sendo as de maiores incidências HAS (47%) e DM (25%). O tempo total de internação em UTI foi maior entre o sexo feminino, associado também a maior necessidade de VM e prona se comparado ao sexo masculino. A complicação mais prevalente foi a BCP, seguida por escaras e TEP.

Fused deposition modelling is one of the most widely used additive manufacturing techniques and the diffusion of 3D printers has increased in popularity even further in recent times. Since high precision is required in 3D printing, a good control over the extrusion process is necessary. In this regard, a crucial phenomenon to be accounted for is the die or extrudate swell, i.e., the enlargement of the cross-section of the strand when coming out of the printer nozzle. While this phenomenon has been studied in large scale extruders, it has not yet been investigated in depth for 3D printing processes. In this work, the die swell phenomenon observed in a printed PLA filament is studied by experiments and fluid dynamic simulations. A novel, easy-to-use, accurate and fast procedure for measuring the value of the die swell ratio during the printing process is developed, accounting for typical errors related to a non-constant strand diameter and possible oscillations of the filament with respect to the extrusion direction. As the printing velocity is increased, a linearly increasing swelling ratio is observed at low printing speeds. The trend flattens at moderate speed values. A sudden increase is found at high printing velocities. The swelling ratio measured with the proposed technique is compared with the results of multi-mode viscoelastic simulations at different temperatures. A fair agreement between the experimental measurements and the numerical predictions is found for printing velocities that are typically employed in commercial 3D printers, supporting the reliability of the developed procedure.

This study evaluates the feasibility of a local action program for HCV micro-elimination in highly endemic areas. Retrospective analysis: administrative and laboratory data (Local Health Unit, southern Italy) were integrated to quantize the anti-HCV-positive subjects not RNA tested and untreated HCV-infected subjects (2018–2022). Prospective analysis: all subjects admitted to a division of the LHU largest hospital (2021–2022) were tested for HCV, with linkage of active-infected patients to care. Overall, 49287 subjects were HCV-Ab tested: 1071 (2.2%) resulted positive without information for an HCV RNA test and 230 (0.5%) had an active infection not yet cured. Among 856 admitted subjects, 54 (6.3%) were HCV-Ab+ and 27 (3.0%) HCV RNA+. Of HCV-infected patients, 22.2% had advanced liver disease, highlighting the need for earlier diagnosis; 27.7% were unaware of HCV infection; and 20.4% were previously aware but never referred to a clinical center. Of these, 26% died and 74% received treatment. Our study emphasizes the value of an active HCV hospital case-finding program to enhance diagnosis in patients with several comorbidities and to easily link them to care. Our data strongly suggest extending this program to all hospital wards/access as a standard of care, particularly in highly endemic areas, to help HCV disease control and take steps in achieving the elimination goals.

Polyphenols are secondary metabolites of interest due to their potential application in various fields. This study is supposed to analyse the content of total polyphenols, total tannins, condensed tannins and antioxidant activity of ten wild plant species of nutritive interest to better understand their potential applications. Furthermore, the effect of heavy metals on the production of the investigated secondary metabolites was analysed. The different phenolic compounds were determined in methanol extracts obtained from edible plants collected during three sampling periods (June, September and November–December) in four areas of the Central Italy. Analyses were carried out by applying standard methodologies. In particular, total polyphenols were determined by the Folin–Ciocalteu method, total tannins by the polyvinylpolypyrrolidone (PVPP) reagent and condensed tannins by the 4-(dimethylamino)cinnamaldehyde (DMCA). Antioxidant activity was determined by assessing the scavenging capacity of 2,2-diphenyl-1-picrylhydrazyl (DPPH). The results showed a satisfactory content of phenolic compounds and antioxidant activity for all species analysed. Furthermore, the existence of a negative correlation between the presence of heavy metals and phenolic compounds was found. Results proved the potential use of these plants for balanced feeding of ruminants.

We present the status of the art of the Archimedes experiment, devoted to measuring the debated interaction of quantum vacuum fluctuations and gravity. The method is essentially the weighing of the transition energy of a layered superconductor where the contribution of vacuum energy to the transition energy is expected to be relevant. The transition is obtained by modulating the temperature of the superconducting sample at a frequency of about 10 mHz and the expected change of weight is measured with a suitably designed high sensitivity cryogenic beam balance. In this paper, we present an overview of the experiment, discussing the expected signal to be measured, and presenting in particular the result of a prototype balance operated in our present laboratory. In the frequency range of the measurement, the sensitivity is affected mainly by seismic, thermal, sensor, and control noise. We discuss these points showing in particular the design of the cryogenic apparatus, the final balance, and the quiet seismic site that will host the final measurement.

Carbonaceous aerosol is the most significant contributor to the particulate matter in the atmosphere. It is composed of a complex mixture of compounds containing carbon atoms and is usually classified into two main fractions: black carbon (BC) and organic carbon (OC). BC is essentially a primary pollutant emitted in particulate form, and its chemical stability excludes chemical transformations during its lifetime in the atmosphere. Therefore, it should be considered a tracer for the long-range transport of anthropogenic air pollution. OC has both primary and secondary origins: primary OC particulate is formed during combustion and emitted mainly as submicron particles, whereas secondary OC particulate originates from gas-to-particle conversion processes. This SI aims to deepen the state of the art on this important topic ranging from theory to the development of new instrumentation useful for this determination, to the chemical–physical problems in the atmosphere, to the health and toxicological implications related to exposure to these pollutants as well as papers showing historical data series both in urban, rural, and remote areas are also appreciated.

Palazzo Valentini, the institutional head office of Città Metropolitana di Roma Capitale, stands in in a crucial position in the Roman archaeological and urban contexts, exactly between the Fora valley, Quirinal Hill slopes, and Campus Martius. It stands on a second-century A.D. complex to which belong, between other archeological remains, two richly decorated aristocratic domus. One of these buildings, the domus A, presents an outward porticoed room with a fourth-century AD central impluvium (open air part of the atrium designed to carry away rainwater) with a black/white tiled mosaic pavement, the preservation status of which is compromised by an incoherent degradation product that has caused gradual detachment of the mosaic tiles. To identify the product and determine the causes of degradation, samples of the product were taken and subjected to SEM-EDS, XRF, NMR, FT-IR and GC-MS analyses. The findings reported in this study can help restorers, archaeologists and conservation scientists in order to improve knowledge about the Roman mosaic, its construction phases, conservation problems and proper solutions.

A direct sandwich enzyme-linked immunosorbent assay (sELISA) was developed for the detection of the atypical β2-toxin (CPB2) of Clostridium perfringens. Polyclonal (PAbs) and monoclonal (MAbs) antibodies were previously obtained employing recombinant CPB2 produced in the baculovirus system as antigen. In the current study, PAbs were used as capture molecules, while purified MAbs conjugated to horseradish peroxidase (MAbs-HRP) were used for the detection of atypical CPB2 toxin. MAbs 5C11E6 and 2G3G6 showed high reactivity, sensitivity and specificity when tested on 232 C. perfringens cell culture isolates. In addition, a reactivity variation among different strains producing atypical CPB2 toxin was observed using the conformation-dependent MAb 23E6E6, suggesting the hypothesis of high instability and/or the existence of different three-dimensional structures of this toxin. Results obtained by sELISA and Western blotting performed on experimentally CPB2-contaminated feces revealed a time-dependent proteolytic degradation as previously observed with the consensus allelic form of CPB2. Finally, the sELISA and an end-point PCR, specific for the atypical cpb2 gene, were used to test field samples (feces, rectal swabs and intestinal contents) from different dead animal species with suspected or confirmed clostridiosis. The comparison of sELISA data with those obtained with end-point PCR suggests this method as a promising tool for the detection of atypical CPB2 toxin.

We report the design of a microcapillary rheometer (μCR) that allows to perform experiments rapidly and in a broad range of shear rates (i.e., from 0.1 to 1000 s−1), using small amounts of material (i.e., just few milligrams). Additionally, multiple measurement parallelization makes it suitable for High-Throughput Rheological Experimentation of polymer melts (HT-Rheo-E).The novel rheometer consists of a set of three cylindrical microcapillaries in which the fluid flows driven by a controlled pressure. A camera, placed at the capillary exit, records the fluid motion to measure its flow rate, from which the fluid viscosity can be determined. The optimization of the setup allowed for reliable and fast viscosity measurements using ca. 10 mg of material. The current work reports the design of the rheometer and validation measurements on several model fluids. The microfabricated μCR is of potential interest for applications in quality control and research where rapid and repeated measurements using limited milligrams of polymer are required, as well as for High-Throughput-Experimentation of complex fluids (e.g., biological systems).

Background: During the COVID-19 pandemic, telemedicine use has increased within community pediatrics. This trend runs counter to reluctance to adaptation of the new mode of healthcare that existed prior to the pandemic. Little is known about what we can expect after the pandemic: if physicians will opt for telemedicine modalities and if tele-pediatrics will continue to be a significant mode of community pediatric care.Objective: The goal of this study was to survey primary pediatric care providers as to their experiences and clinical decision making with telemedicine modalities prior to and during the COVID-19 pandemic, as well as their projected use after the pandemic ends.Material and methods: Using the EAPRASnet database we surveyed pediatricians throughout Europe, using a web-based questionnaire. The survey was performed during the COVID-19 pandemic (June–July 2020), assessed telemedicine use for several modalities, prior to and during the pandemic as well as predicted use after the pandemic will have resolved. Participants were also surveyed regarding clinical decision making in two hypothetical clinical scenarios managed by telemedicine.Results: A total of 710 physicians participated, 76% were pediatricians. The percentage of respondents who reported daily use for at least 50% of all encounters via telemedicine modalities increased during the pandemic: phone calls (4% prior to the pandemic to 52% during the pandemic), emails (2–9%), text messages (1–6%), social media (3–11%), cell-phone pictures/video (1–9%), and video conferencing (1–7%) (p < 0.005). The predicted post-pandemic use of these modalities partially declined to 19, 4, 3, 6, 9, and 4%, respectively (p < 0.005), yet demonstrating a prospectively sustained use of pictures/videos after the pandemic. Reported high likelihood of remotely treating suspected pneumonia and acute otitis media with antibiotics decreased from 8 to 16% during the pandemic to an assumed 2 and 4% after the pandemic, respectively (p < 0.005).Conclusions: This study demonstrates an increased utilization of telemedicine by pediatric providers during the COVID-19 pandemic, as well as a partially sustained effect that will promote telemedicine use as part of a hybrid care provision after the pandemic will have resolved.

Severe acute respiratory syndrome-related coronavirus 2 (SARS-CoV-2) has infected millions of individuals around the World. Hypertension (HT), chronic heart disease (CHD), and diabetes mellitus (DM), particularly in the elderly, increase susceptibility to SARS-CoV-2 infection. However, conflicting results [such as coronavirus 2019 (COVID-19) disease vulnerability, case fatality, etc.] have been reported about the response to infection and COVID-19 outcomes in men and women. Therefore, understanding predictors of Intensive Care Unit (ICU) admission might help future planning and management of the disease. We conducted a multicenter survey about COVID-19 involving internists from Internal Medicine Wards. This survey indirectly allowed us to analyze the information of 2400 patients hospitalized in 35 wards of Internal Medicine of the Campania Region between July and October 2020. Our investigation has detected that the infection is more frequent in males, and the number of male patients hospitalized in ICU is also higher than females, with a large proportion of hypertensive patients. Extensive prospective studies are required to confirm this finding and explore the mechanisms for which hypertensive males are exposed to a higher proportion of admission to ICU and higher case fatality rates.

The midbody is an organelle that forms between the two daughter cells during cytokinesis. It co-ordinates the abscission of the nascent daughter cells and is composed of a multitude of proteins that are meticulously arranged into distinct temporal and spatial localization patterns. However, very little is known about the mechanisms that regulate the localization and function of midbody proteins. Here, we analyzed the temporal and spatial profiles of key midbody proteins during mitotic exit under normal conditions and after treatment with drugs that affect phosphorylation and proteasome-mediated degradation to decipher the impacts of post-translational modifications on midbody protein dynamics. Our results highlighted that midbody proteins show distinct spatio-temporal dynamics during mitotic exit and cytokinesis that depend on both ubiquitin-mediated proteasome degradation and phosphorylation/de-phosphorylation. They also identified two discrete classes of midbody proteins: ‘transient’ midbody proteins—including Anillin, Aurora B and PRC1—which rapidly accumulate at the midbody after anaphase onset and then slowly disappear, and ‘stable’ midbody proteins—including CIT-K, KIF14 and KIF23—which instead persist at the midbody throughout cytokinesis and also post abscission. These two classes of midbody proteins display distinct interaction networks with ubiquitylation factors, which could potentially explain their different dynamics and stability during cytokinesis.

The placenta is a crucial interface between the fetus and the maternal environment. It allows for nutrient absorption, thermal regulation, waste elimination, and gas exchange through the mother’s blood supply. Furthermore, the placenta determines important adjustments and epigenetic modifications that can change the phenotypic expression of the individual even long after birth. Polyethylene glycol (PEG) is a polyether compound derived from petroleum with many applications, from medicine to industrial manufacturing. In this study, for the first time, an integration of ultra-high-performance liquid chromatography (UHPLC) coupled with mass spectrometry (MS) was used to detect suites of PEG compounds in human placenta samples, collected from 12 placentas, originating from physiological pregnancy. In 10 placentas, we identified fragments of PEG in both chorioamniotic membranes and placental cotyledons, for a total of 36 samples.

Azobenzene is one of the most investigated photo-responsive liquid crystalline molecules. It can isomerize between two different isoforms, trans (E) and cis (Z) configurations, when stimulated by light. It is used as a molecular engine in photo-mobile materials (PMPs). The use of liquid crystals (LCs) as building blocks enhances the mechanical properties of the PMPs. It is not easy to obtain PMPs with monodomain configurations when the LCs are 100% azobenzene. In this work, we studied three LC mixtures, describing the thermo/mechanical phenomena that regulate the actuation of such materials. The nematic temperature of the LC elastomers was measured and the PMPs carefully characterized for their bending and speed capability. Our finding suggests that the ratio between linear and cross-linker monomer greatly influences the nematic temperature of the mixture. Furthermore, 100% azobenzene materials polymerized using dicumyl peroxide can be useful to design polarization-selective switches.

Bacterial conjunctivitis significantly impacts public health, including more than one-third of eye diseases reported worldwide. It is an infection caused by various aerobic and anaerobic bacteria and is highly contagious. Therefore, it has a high incidence of bacterial resistance to the antibiotics commonly used for treatment. Among the most recent antibiotics, besifloxacin is a fourth-generation fluoroquinolone antibiotic indicated exclusively for topical ophthalmic use. Due to its importance in treating bacterial conjunctivitis and its low solubility in water, limiting its efficacy, a nanotechnology-based drug delivery preparation was developed to overcome this hurdle. Besifloxacin nanocrystals were prepared by small-scale wet milling and response surface methodology, using Povacoat® as a stabilizer. The particle’s average hydrodynamic diameter (Z-ave) was approximately 550 nm (17 times smaller than raw material), with a polydispersity index (PdI) of less than 0.2. The saturation solubility increased about two times compared to the raw material, making it possible to increase the dissolution rate of this drug substance, potentially improving its bioavailability and safety. The optimized preparation was stable under an accelerated stability study (90 days). The Z-ave, PZ, PdI, and content did not alter significantly during this period. Furthermore, the 0.6% m/m besifloxacin nanocrystals at the maximum dose and the Povacoat® stabilizer did not show toxicity in Galleria mellonella larvae. The innovative ophthalmic preparation minimum inhibitory concentration (MIC) was 0.0960 µg/mL and 1.60 µg/mL against Staphylococcus aureus and Pseudomonas aeruginosa, respectively, confirming in vitro efficacy. Therefore, besifloxacin nanocrystals revealed the potential for reduced dosing of the drug substance, with a minor occurrence of adverse effects and greater patient adherence to treatment.

Trimethylamine-N-oxide (TMAO) is a gut microbiota-derived metabolite recognized as strongly related to cardiovascular diseases (CVD), mainly increasing the risk of atherosclerosis development. Currently, no pharmacological approaches have been licensed for reduction of TMAO serum levels and conventional anti-atherosclerosis treatments only target the traditional risk factors, and the cardiovascular risk (CVR) still persist. This underlines the need to find novel targeted strategies for management of atherosclerosis. In this study we tested the ability of a novel nutraceutical formulation based on grape pomace polyphenols (Taurisolo®) in reducing both the serum levels of TMAO and oxidative stress-related biomarkers in humans (n = 213). After chronic treatment with Taurisolo® we observed significantly reduced levels of TMAO (−49.78 and −75.80%, after 4-week and 8-week treatment, respectively), oxidized LDL (oxLDL; −43.12 and −65.05%, after 4-week and 8-week treatment, respectively), and reactive oxygen species (D-ROMs; −34.37 and −49.68%, after 4-week and 8-week treatment, respectively). On the other hand, no significant changes were observed in control group. Such promising, the results observed allow indicating Taurisolo® as an effective nutraceutical strategy for prevention of atherosclerosis.Clinical Trial Registration: This study is listed on the ISRCTN registry with ID ISRCTN10794277 (doi: 10.1186/ISRCTN10794277).

Benign prostatic hyperplasia (BPH) is a common condition in adult men. Especially in Europe, increasing attention has been focused on E. angustifolium extracts (EAEs), which are widely used for their positive effects on the symptoms of BPH, although human clinical trials are limited. The aim of this monocentric, randomized, double-blind, placebo-controlled clinical trial is to evaluate if a daily intake of hard, gastric-resistant capsules containing a chemically characterized EAE (500 mg) for 6 months may allow a significant improvement in symptoms in subjects with BPH. This study was conducted in 128 adult men, randomly assigned to receive either EAE food supplement (N = 70) or placebo (N = 58), who underwent four visits (baseline = t0, after 15 days = t1, after 2 months = t2 and after 6 months = t3) in an outpatient setting to evaluate post-void residual (PVR) and prostate volume (PV) by means of prostate ultrasound, prostate-specific antigen (PSA) and neutrofile/lymphocyte ratio (N/L), nocturia before the clinical visits and International Prostate Specific Score (IPSS) registered by the physicians. EAE food supplement induced a significant decrease in the PVR and consequently nocturia improving the quality of life as suggested by the decrease of IPSS. No subjects reported adverse effects related to oral intake of EAE food supplement. Moreover, EAE food supplement did not show hepatic or renal toxicity. In conclusion, EAE food supplements can be used in subjects with BPH, to improve their quality of life and general renal function.

OBJECTIVE: The aim of this study is to assess whether the touch of osteopathic manipulative treatment (OMT) can affect the endogenous production of oxytocin in fullterm pregnant women and the assessment of well-being following the treatment. PATIENTS AND METHODS: In this study have been enrolled 57 pregnant women at full-term pregnancy (37th-41st week) for evaluation of the concentration of salivary oxytocin 2 minutes before and 2 minutes after a single session of OMT by an osteopath lasting for 30 minutes. Pre-OMT and post-OMT saliva samples were collected with the use of Salivette® salivary swabs. 7 salivary swabs were excluded from the analysis. 50 samples were analyzed with an appropriate ELISA kit. RESULTS: The mean OT salivary concentration pre-OMT was 89.98±16.39, and post-OMT was 100.60±19.13 tends to increase with p=0.0000051. In multivariate analysis, two subgroups show interesting data in the mean difference in OT salivary concentration post-OMT: women with painful contractions (p=0.06) and women under 35 years (p=0.09). CONCLUSIONS: The results of this study demonstrate that the effectiveness of OMT-increasing endogenous oxytocin is statistically significant in full-term pregnant women. The sensation of well-being found in most women indicates that there has been a predominantly central rather than peripheral oxytocin release after OMT. [ABSTRACT FROM AUTHOR]

Resumen: La evaluación histológica de la Enfermedad Inflamatoria Intestinal (EII) tiene como finalidad corroborar el diagnóstico de EII. En este artículo revisaremos múltiples conceptos histológicos relacionados con EII. Definiremos los términos de colitis aguda y crónica, las características macroscópicas y microscópicas de EII (incluyendo las características histológicas distintivas entre Colitis Ulcerativa Idiopática y Enfermedad de Crohn), y otras formas de colitis no relacionadas a EII que actúan como diagnósticos diferenciales (colitis infecciosa, colitis microscópica y colitis diverticular, entre otras). Así mismo, haremos referencia al riesgo de los pacientes con EII a desarrollar cáncer colorrectal y a las características histológicas de displasia en EII, incluyendo la última terminología recomendada para la descripción de la apariencia endoscópica de lesiones en seguimiento de displasia en EII. In this article, we will review multiple histological concepts related to IBD. We will define the terms of acute and chronic colitis, the macroscopic and microscopic characteristics of IBD (including the distinctive histological features between Idiopathic Ulcerative Colitis and Crohn's Disease) and other forms of colitis not related to IBD that act as differential diagnoses (infectious, microscopic and diverticular colitis, among others). Likewise, we will talk about the risk of IBD patients to develop colorectal cancer and the histological features of dysplasia in IBD, including the latest terminology recommended for the description of the endoscopic appearance of lesions in the follow-up of dysplasia in IBD. Abstract We review the physics opportunities of the Future Circular Collider, covering its e+e-, pp, ep and heavy ion programmes. We describe the measurement capabilities of each FCC component, addressing the study of electroweak, Higgs and strong interactions, the top quark and flavour, as well as phenomena beyond the Standard Model. We highlight the synergy and complementarity of the different colliders, which will contribute to a uniquely coherent and ambitious research programme, providing an unmatchable combination of precision and sensitivity to new physics.

Abstract Cophylogeny is the congruence of phylogenetic relationships between two different groups of organisms due to their long‐term interaction. We investigated the use of tree shape distance measures to quantify the degree of cophylogeny. We implemented a reverse‐time simulation model of pathogen phylogenies within a fixed host tree, given cospeciation probability, host switching, and pathogen speciation rates. We used this model to evaluate 18 distance measures between host and pathogen trees including two kernel distances that we developed for labeled and unlabeled trees, which use branch lengths and accommodate different size trees. Finally, we used these measures to revisit published cophylogenetic studies, where authors described the observed associations as representing a high or low degree of cophylogeny. Our simulations demonstrated that some measures are more informative than others with respect to specific coevolution parameters especially when these did not assume extreme values. For real datasets, trees’ associations projection revealed clustering of high concordance studies suggesting that investigators are describing it in a consistent way. Our results support the hypothesis that measures can be useful for quantifying cophylogeny. This motivates their usage in the field of coevolution and supports the development of simulation‐based methods, i.e., approximate Bayesian computation, to estimate the underlying coevolutionary parameters.
