En este artículo analizaremos una experiencia de educación disidente en contexto de encierro punitivo. Se trata de una serie de talleres y grupos terapéuticos que se desarrollaron desde el año 2017 al año 2019 en la Unidad N.°45 de la localidad de Melchor Romero, en el marco del Programa Masculinidades disidentes: jóvenes y salud mental en cárceles, que cuenta con dos organizaciones de la sociedad civil copartícipes: Grupo de Estudios de Educación en Cárceles (GESEC) y la Asociación Civil de Familiares de Detenidos en Cárceles Federales (ACIFAD). La experiencia tuvo como destinatarios a varones jóvenes de 18 a 23 años de edad que participaron de una experiencia de trabajo que consistía en un primer momento de reflexiones recursivas sobre temáticas vinculadas con las masculinidades, y un segundo momento, de grupo terapéutico en donde la escritura del primer momento se inscribía en el so(gra)ma (cuerpo-lenguaje) de las masculinidades en disidencia (Colanzi, 2021). A los efectos de establecer un recorte específico, trabajaremos entonces con algunos interrogantes: ¿cuál es la posición de las masculinidades en cárceles frente a los lazos sexo-afectivos? ¿cómo se vivencias los lazos sexo-afectivos entre varones privados de libertad? ¿cómo los lazos sexo-afectivos operan en tanto una estrategia de cuidado amoroso y affidamento masculino entre varones?, In this article we will analyze an experience of dissident education in the context of punitive confinement. This is a series of workshops and therapeutic groups that were developed from 2017 to 2019 in Unit No. 45 of the town of Melchor Romero, within the framework of the Dissident Masculinities Program: youth and mental health in prisons, which It has two partner civil society organizations: Prison Education Study Group (GESEC) and the Civil Association of Relatives of Detainees in Federal Prisons (ACIFAD). The experience was aimed at young men between 18 and 23 years of age who participated in a work experience that consisted in a first moment of recursive reflections on themes related to masculinities, and a second moment, of a therapeutic group in which the writing from the first moment it was inscribed in the so(gram)ma (body-language) of masculinities in dissidence (Colanzi, 2021). Background. The C-terminal-binding protein 1/brefeldin A ADP-ribosylation substrate (CtBP1/BARS) acts both as an oncogenic transcriptional co-repressor and as a fission inducing protein required for membrane trafficking and Golgi complex partitioning during mitosis, hence for mitotic entry. CtBP1/BARS overexpression, in multiple cancers, has pro-tumorigenic functions regulating gene networks associated with “cancer hallmarks” and malignant behavior including: increased cell survival, proliferation, migration/invasion, epithelial-mesenchymal transition (EMT). Structurally, CtBP1/BARS belongs to the hydroxyacid-dehydrogenase family and possesses a NAD(H)-binding Rossmann fold, which, depending on ligands bound, controls the oligomerization of CtBP1/BARS and, in turn, its cellular functions. Here, we proposed to target the CtBP1/BARS Rossmann fold with small molecules as selective inhibitors of mitotic entry and pro-tumoral transcriptional activities. Methods. Structured-based screening of drug databases at different development stages was applied to discover novel ligands targeting the Rossmann fold. Among these identified ligands, N-(3,4-dichlorophenyl)-4-{[(4-nitrophenyl)carbamoyl]amino}benzenesulfonamide, called Comp.11, was selected for further analysis. Fluorescence spectroscopy, isothermal calorimetry, computational modelling and site-directed mutagenesis were employed to define the binding of Comp.11 to the Rossmann fold. Effects of Comp.11 on the oligomerization state, protein partners binding and pro-tumoral activities were evaluated by size-exclusion chromatography, pull-down, membrane transport and mitotic entry assays, Flow cytometry, quantitative real-time PCR, motility/invasion, and colony assays in A375MM and B16F10 melanoma cell lines. Effects of Comp.11 on tumor growth in vivo were analyzed in mouse tumor model. Results. We identify Comp.11 as a new, potent and selective inhibitor of CtBP1/BARS (but not CtBP2). Comp.11 directly binds to the CtBP1/BARS Rossmann fold affecting the oligomerization state of the protein (unlike other known CtBPs inhibitors), which, in turn, hinders interactions with relevant partners, resulting in the inhibition of both CtBP1/BARS cellular functions: i) membrane fission, with block of mitotic entry and cellular secretion; and ii) transcriptional pro-tumoral effects with significantly hampered proliferation, EMT, migration/invasion, and colony-forming capabilities. The combination of these effects impairs melanoma tumor growth in mouse models. Conclusions. This study identifies a potent and selective inhibitor of CtBP1/BARS active in cellular and melanoma animal models revealing new opportunities to study the role of CtBP1/BARS in tumor biology and to develop novel melanoma treatments.

Estudos recentes mostram que a evasão nos cursos da área de Computação costuma ser alta, sendo muitas vezes atribuída à dificuldade dos alunos com as disciplinas relacionadas à Matemática e à Programação. Tal problema motivou a realização deste trabalho, que investigou a evasão nos cursos de graduação da área de Computação da Universidade Estadual de Maringá. O estudo foi realizado a partir de dados obtidos junto à instituição e a partir de dados coletados com alunos que evadiram dos cursos. A partir dos dados institucionais, foram realizadas análises quantitativas e a partir das respostas dos alunos evadidos, foram realizadas análises qualitativas, incluindo as principais causas que levaram à evasão. Observou-se que o curso noturno apresentou maior taxa de evasão do que o curso em período integral e que o principal fator percebido como impactante para a evasão foi a didática adotada pelos professores. Fatores ligados à pandemia de COVID-19 também foram percebidos como impactantes.

The decision to enter mitosis requires not only the control of DNA replication but also additional and crucial preparatory steps such as, for example, partial disassembly of the Golgi complex during G2. The Golgi complex is fundamental for the processing and sorting of proteins and lipids in the secretory pathway. It is organized as stacks of cisternae laterally connected by tubules to form a continuous Golgi ribbon. During G2, the Golgi ribbon is unlinked into isolated stacks in preparation for cell division. This structural reorganization is necessary for entry into mitosis, indicating that a “Golgi mitotic checkpoint” controls the correct segregation of this organelle. To understand the physiological significance of the pre-mitotic Golgi unlinking, we devised a strategy to accumulate cells in G2 with an intact Golgi ribbon and then induce entry into mitosis. Here, we show that forcing the entry of cells into mitosis with an intact Golgi ribbon causes remarkable cell division defects, including spindle multipolarity and binucleation, favoring cell transformation. We also find that the cells entering mitosis with an intact Golgi ribbon show reduced levels at the centrosome of the kinase Aurora-A, a pivotal regulator of spindle formation. Overexpression of Aurora-A rescues spindle formation, indicating that the Golgi-dependent Aurora-A recruitment has a crucial role in spindle formation. Thus, our results show that alterations of the pre-mitotic Golgi segregation have profound consequences on the fidelity of the mitotic process, representing potential risk factors for cell transformation and cancer development.

The Software Product Line (SPL) approach is a reuse technique derived from other engineering disciplines to develop a family of products within a specific domain. An essential artifact of SPL is the Software Product Line Architecture (PLA), which identifies elements characterized by variation points, variability, and variants. PLA anticipates design decisions and aims to achieve critical features such as reusability and modularity. As a multiobjective optimization problem, the interactive multiobjective optimization approach for PLA design (iMOA4PLA) employs specific metrics to optimize the PLA problem. The iMOA4PLA is implemented through the OPLA-Tool v2.0, with the decision maker (DM) assuming a participatory role in the optimization process, guiding it through various strategies. In this context, machine learning (ML) algorithms assist the DM. We conducted two experiments to evaluate the proposed model - one quantitative and one qualitative. The results indicate that the approach facilitates the generation of PLA alternatives that align with the DM's expectations. Additionaly, strategies employed are highly efficient in assisting the DM with the PLA evaluation process. This research presents a model that helps decision-makers optimize the Software Product Line Architecture while enabling an interactive process that leverages machine learning algorithms. The scientific contribution of this work lies in providing a framework for the design and evaluation of the PLA that leverages the benefits of machine learning algorithms.hat utilizes machine learning algorithms.
