International audience; A fast numerical and hybrid method for the calculation of the eigenvalues and eigenvectors of waveguides that present a radius of curvature will be presented in this work, starting from the theory of propagation in electromagnetism resulting from the Maxwell's formalism [1]. This method comprises a so-called conformal transformation (CT) of the complex plane associated with a matrix formulation including a kind of virtual multilayer. The role of the conformal transformation is to avoid the use of the Jacobian transformation and to convert a curved geometry structure into a virtual straight geometry structure presenting an effective and transformed profile of the index structure. Indeed, various mathematic CT exist based on linear, inversion, rotation, homographic or more particular Schwartz-Christoffel transformations [2]. A matrix method, equivalent to those used for real multilayer of materials [3,4], is directly used into the virtual and new modified profile of CT-index in order to determine the effective propagation constants (or eigenvalues) and the profile of the optical modes (or eigenvectors) propagating in optical guides having a radius of curvature. The numerical codes have been developed and tested in various family structures (with different radii of curvature, different cladding/core index values at fixed wavelength and thicknesses) and asymptotic cases. The results of this hybrid method are in very good agreement with the commercial software COMSOL 3D Multiphysics [5]. The developed hybrid method provides a useful and fast qualitative and quantitative visualization and calculation of the wave propagation in bended waveguides and can be easily compiled on a personal computer.

Here the reader will find a practical guide that provides fast and to the point information on how to (i) deliver H2S to cells; (ii) modulate its endogenous production; and (iii) measure its levels in fluids, cells and tissues in order to gain an understanding of its role in health and disease.

The endogenous gasotransmitter hydrogen sulphide (HS) is an important regulator of the cardiovascular system, particularly of myocardial function. Moreover, HS exhibits cardioprotective activity against ischemia/reperfusion (I/R) or hypoxic injury, and is considered an important mediator of "ischemic preconditioning", through activation of mitochondrial potassium channels, reduction of oxidative stress, activation of the endogenous "anti-oxidant machinery" and limitation of inflammatory responses. Accordingly, HS-donors, i.e. pro-drugs able to generate exogenous HS, are viewed as promising therapeutic agents for a number of cardiovascular diseases. The novel HS-donor 4-carboxy phenyl-isothiocyanate (4CPI), whose vasorelaxing effects were recently reported, was tested here in different experimental models of myocardial I/R. In Langendorff-perfused rat hearts subjected to I/R, 4CPI significantly improved the post-ischemic recovery of myocardial functional parameters and limited tissue injury. These effects were antagonized by 5-hydroxydecanoic acid (a blocker of mitoK channels). Moreover, 4CPI inhibited the formation of reactive oxygen species. We found the whole battery of HS-producing enzymes to be present in myocardial tissue: cystathionine γ-lyase (CSE), cystathionine β-synthase (CBS) and 3-mercaptopyruvate sulfurtransferase (MPST). Notably, 4CPI down-regulated the post-ischemic expression of CSE. In Langendorff-perfused mouse hearts, 4CPI reduced the post-ischemic release of norepinephrine and the incidence of ventricular arrhythmias. In both rat and mouse hearts, 4CPI did not affect the degranulation of resident mast cells. In isolated rat cardiac mitochondria, 4CPI partially depolarized the mitochondrial membrane potential; this effect was antagonized by ATP (i.e., the physiological inhibitor of K channels). Moreover, 4CPI abrogated calcium uptake in the mitochondrial matrix. Finally, in an in vivo model of acute myocardial infarction in rats, 4CPI significantly decreased I/R-induced tissue injury. In conclusion, HS-donors, and in particular isothiocyanate-based H2S-releasing drugs like 4CPI, can actually be considered a suitable pharmacological option in anti-ischemic therapy.

S1P (sphingosine 1-phosphate) represents one of the key latest additions to the list of vasoactive substances that modulate vascular tone. PAR-2 (proteinase activated receptor-2) has been shown to be involved in cardiovascular function. In the present study, we investigated the involvement of PAR-2 in S1P-induced effect on vascular tone. The present study has been performed by using isolated mouse aortas. Both S1P and PAR-2 agonists induced endothelium-dependent vasorelaxation. L-NAME (NG-nitro-L-arginine methyl ester) and wortmannin abrogated the S1P-induced vasorelaxatioin, while significantly inhibiting the PAR-2-mediated effect. Either ENMD1068, a PAR-2 antagonist, or gabexate, a serine protease inhibitor, significantly inhibited S1P-induced vasorelaxation. Aortic tissues harvested from mice overexpressing PAR-2 displayed a significant increase in vascular response to S1P as opposed to PAR-2-null mice. Immunoprecipitation and immunofluorescence studies demonstrated that S1P1 interacted with PAR-2 and co-localized with PAR-2 on the vascular endothelial surface. Furthermore, S1P administration to vascular tissues triggered PAR-2 mobilization from the plasma membrane to the perinuclear area; S1P-induced translocation of PAR-2 was abrogated when aortic rings were pre-treated with ENMD1068 or when caveolae dysfunction occurred. Similarly, experiments performed in cultured endothelial cells (human umbilical vein endothelial cells) showed a co-localization of S1P1 and PAR2, as well as the ability of S1P to induce PAR-2 trafficking. Our results suggest that S1P induces endothelium-dependent vasorelaxation mainly through S1P1 and involves PAR-2 transactivation.

Introduction: S1P systemic administration leads to an increased airway hyper-reactivity. Aim: To address the mechanism(s) involved in S1P-induced airway hyper-reactivity. Methods/Results: Balb/c mice receiving s.c. injection of S1P (10ng) display a dose- and time-dependent increase in airway reactivity. Airway hyper-reactivity was coupled to an enhanced inflammatory response. In particular, lungs harvested from S1P-treated animals showed an increased cell infiltration, loss of the alveolar structure and mucous cell metaplasia. We also found a significant increase in serum levels of both PGD2 and IgE. FACS analysis on lung-derived cells evidenced that mast cells were prominent cell type. Proliferation assay of peripheral lymph nodes-derived cells showed that lymphocytes (CD4+) obtained from S1P-sensitized mice displayed a higher proliferation rate compared to cells from vehicle group. In order to address mast cells or T cells contribution in S1P effect mast cell knock-out mice (MCKO) and nude athymic mice were used. In MCKO mice S1P-induced hyper-reactivity was lost, while, still displayed an ongoing inflammatory response. Furthermore IgE serum levels were still increased in MCKO. Conversely, the absence of T cells first dampened S1P-induced hyper-responsiveness and, more interestingly, abrogated inflammatory response. Conclusions. Our data demonstrate that S1P alters lung morphology leading towards an asthma-like environment in which mucus and prostaglandins together with IgE are highly produced. Mast cells are mainly responsible for the airway increased reactivity but do not play a key role in the inflammatory effect elicited by S1P and driven by T cells.
