Rupture of an intracranial aneurysm leads to subarachnoid hemorrhage, a severe type of stroke. To discover new risk loci and the genetic architecture of intracranial aneurysms, we performed a cross-ancestry, genome-wide association study in 10,754 cases and 306,882 controls of European and East Asian ancestry. We discovered 17 risk loci, 11 of which are new. We reveal a polygenic architecture and explain over half of the disease heritability. We show a high genetic correlation between ruptured and unruptured intracranial aneurysms. We also find a suggestive role for endothelial cells by using gene mapping and heritability enrichment. Drug-target enrichment shows pleiotropy between intracranial aneurysms and antiepileptic and sex hormone drugs, providing insights into intracranial aneurysm pathophysiology. Finally, genetic risks for smoking and high blood pressure, the two main clinical risk factors, play important roles in intracranial aneurysm risk, and drive most of the genetic correlation between intracranial aneurysms and other cerebrovascular traits.

Visitors move between destinations differently, which creates patterns and networks that link destinations within travel itineraries. These patterns contain valuable information that can be analysed for tourism product development, to improve marketing services and assist in infrastructure and transportation development decisions. Traditionally, research on visitors' travel patterns relied on data collected from interviews, surveys, or direct observation. Normally the size of this type of data is relatively small, and processing of data collection is costly as well as time consuming. The introduction of big data analytics within tourism research has seen a diversification within data sources, and social media has become a particularly valuable source. Social media generates vast volumes of data, which in addition to text, images, and video content contains additional information in the form of metadata, such as geo-location (where posts were made) and user origins. When combined with advances in big data analytics, this type of data opens an opportunity to model temporal and spatial travel patterns and especially understand the travel pattern drivers behind the travel patterns. Due to the flexibility of social media data collection, understanding visitors' travel patterns is not limited to one destination but can be expanded to the national or global scale. This thesis took advantage of the value of social media data and big data analytics, in five independent but interconnected chapters to report, and advance knowledge on visitors travel patterns. The thesis is partly comprised of papers with a compilation of five manuscripts in addition to an introduction, an overview of the methodology, and a conclusion chapter. Manuscript one consisted of a literature review that synthesises the relevant literature relating to social media, tourism, and travel patterns. After 18F-FDG injection at 3.7 MBq/kg, 1 h early imaging and 2 h delayed imaging were performed. The metabolic and volumetric parameters such as SUVmax, SUVpeak, SULmax, SULpeak, MTV and TLG were measured. The tumor markers including CFYRA21-1, NSE, SCC-ag and the immunohistochemical biomarkers including Ki-67, P53 and CK-7 were examined. All patients were followed up for 24 months, and the 1-year and 2-year overall survival rate (OS) were recorded.Results: There were significant differences in metabolic and volumetric parameters (SUVmax, SUVpeak, SULmax, SULpeak and TLG) between adenocarcinoma and squamous cell carcinoma of NSCLC. SUVmax, SUVpeak, SULmax, SULpeak, MTV and TLG were correlated with tumor marker NSE and TNM stage. MTV and TLG were related to CYFRA21-1, and only MTV was associated with SCC-ag. The sample size effect on the deformation behaviors of metallic glasses (MGs) attracts a lot of interest. Here, a series of molecular dynamics simulations have been performed on un-notched and notched MG thin films to fully investigate the influence of sample thickness on their fracture strengths and failure mechanisms. With decreasing sample thickness, the failure strength of un-notched MG thin films decreases slightly and the failure mode changes from shear banding to homogeneous deformation. In contrast to this brittle-to-ductile transition observed in un-notched MG thin films, shear banding prevails for notched MG thin films regardless of sample thickness. Concurrently, a notch strengthening effect is observed in the notched MG thin films when the sample thickness falls close to the shear band (SB) thickness. This significant notch strengthening is due to the failure mode transition from homogeneous deformation in un-notched MGs to shear banding in notched MGs. Based on the energetic model, a theoretical analysis is proposed to rationalize the size effect on failure mechanism transitions of both un-notched and notched MG thin films. The present study offers an understanding of size effect on the un-notched and notched MG thin films, as well as useful guidelines for the design, testing, and engineering of MG for microelectromechanical applications.

Key reactions of the synthesis include a Pd-catalyzed Suzuki–Miyaura cross-coupling reaction, a Lewis acid-catalyzed reductive Prins cyclization reaction, and a SmI2-mediated transannular pinacol coupling reaction. The synthesis provides a new avenue to access diverse relevant bioactive molecules.

- Abstract
The development of the Internet of Things (IoT) drives us to confront, manage and analyse massive and complicated data generated from various sensors. Also, social media have rapidly become very popular and can be considered as important source of data. Twitter on average generates about 6,000 tweets every second, which total to over 500 million tweets per day. Facebook has over 2 billion monthly active users. The individual posts may be trivial, however, the accumulated big data can provide diverse valuable information, which can be also correlated with IoT and enable human sentiment identification of the environment changes. This work proposes a machine learning model for correlation analysis and prediction of weather conditions and social media posts. In experimental evaluation we demonstrate that the Big Data analysis approach and machine learning techniques can be used to analyse and predict sentiment of different weather conditions.

Background: Randomised control trials and genetic analyses have demonstrated that vitamin D or 25-hydroxyvitamin D (25[OH]D) levels may not play a causal role in the development of cardiovascular disease. However, it is unclear if 25(OH)D is causally associated with cause-specific vascular disease and lipids. Therefore, we examined the causal association of 25(OH)D with myocardial infarction, stroke, ischaemic heart disease, ischaemic stroke, subarachnoid haemorrhage, intracerebral haemorrhage, and lipid levels among both Chinese and Europeans. Methods: We used a Mendelian randomisation (MR) design in the China Kadoorie Biobank, the Copenhagen City Heart Study, and the Copenhagen General Population Study. The 25(OH)D-related genetic variants in the CYP2R1 and DCHR7 genes were genotyped in 99,012 Chinese adults and 106,911 Danish adults. Results: In Chinese adults, plasma 25(OH)D levels were not significantly associated with cause-specific vascular disease or mortality, with the exception of intracerebral haemorrhage (HR, 1.09 [95% CI, 1.01,1.18] per 25 nmol/L higher plasma 25(OH)D). In Europeans, plasma 25(OH)D levels were inversely associated with all types of vascular diseases and mortality. However, MR analysis did not demonstrate causal associations of genetically increased 25(OH)D levels with cause-specific vascular diseases, or mortality in both Chinese and European adults. In addition, each 25 nmol/L higher 25(OH)D was observationally associated with lower total cholesterol and low-density lipoprotein cholesterol levels, but higher high-density lipoprotein cholesterol levels. Likewise, MR analysis showed that 25(OH)D levels were not causally associated with lipids in both Chinese and European adults after Bonferroni correction. Even before the global halt to extensive mobility due to the coronavirus crisis, there has been growing resistance amongst some groups to flying. Reflecting increasing concern about the imminent climate emergency, the phenomenon of feeling ashamed of flying and expressing related emotions through communication has resulted in the neologism "flygskam". Drawing on 14,212 Twitter posts that contained "flygskam" and a further 1037 using the English "flightshame", this research examined the spatial-temporal spread of these words from Sweden starting in 2016 to the rest of the world indicating a global phenomenon. The findings indicate that national context continues to be important in understanding the nexus of individual and social sensemaking and processing of new information. They also demonstrate, however, that global networks (facilitated through social media) might challenge the needs of physical mobility as requirements to connect, exchange views, and create identity through peer group membership. Further analysis of the content revealed key perspectives and topics, providing insight into the relatively homogeneous discussions amongst a network community. Only 6% of posts rejected the notion, whilst the majority indicated support and advanced suggestions for more sustainable alternatives. It might be too early to say whether language behaviour expressed in online communication translates into real behaviour, but the current pandemic may well provide further impetus to no-fly movements by way of a strategic alliance between different peer groups. The wide existence of saline wastewater has attracted public attention due to its environmental destructiveness. The potential of anammox to treat saline wastewater was systematically evaluated in this study. Three bioreactors with different salinity increasing strategies (R1: inhibition kinetic; R2: gradient increasing; R3: pulsed increasing) were operated to identify the optimal acclimation mode. The results showed that the inhibition of anammox activity by salinity was mainly caused by the loss of enzyme activity. Under the condition of 25.0 g NaCl L−1, the highest nitrogen removal rate of R3 (2.36 ± 0.14 kg N m−3 d−1) indicated that the pulsed strategy might be optimal. Changes in microbial community might be the primary reason lead to different acclimatization results. The relative abundance of anammox bacteria increased by 37.19% in R1 and by 46.81% in R3, but remained stable in R2 with increasing salinity. Dynamic varations in bacterial interactions, proteins, and functional genes revealed the potential resistance mechanisms of bacteria to salinity. The present work provides a novel approach and guidance for the treatment of nitrogen-rich saline wastewater by the anammox process.

18 F]fluorodeoxyglucose positron emission tomography/computed tomography (18 F-FDG PET/CT) imaging in non-small cell lung cancer (NSCLC) and the predictive value for prognosis of NSCLC. Methods: Retrospective analysis was performed on 133 NSCLC patients who received18 F-FDG PET/CT imaging. After18 F-FDG injection at 3.7 MBq/kg, 1 h early imaging and 2 h delayed imaging were performed. The metabolic and volumetric parameters such as SUVmax, SUVpeak, SULmax, SULpeak, MTV and TLG were measured. The tumor markers including CFYRA21-1, NSE, SCC-ag and the immunohistochemical biomarkers including Ki-67, P53 and CK-7 were examined. All patients were followed up for 24 months, and the 1-year and 2-year overall survival rate (OS) were recorded. Results: There were significant differences in metabolic and volumetric parameters (SUVmax, SUVpeak, SULmax, SULpeak and TLG) between adenocarcinoma and squamous cell carcinoma of NSCLC. SUVmax, SUVpeak, SULmax, SULpeak, MTV and TLG were correlated with tumor marker NSE and TNM stage. MTV and TLG were related to CYFRA21-1, and only MTV was associated with SCC-ag. SUVpeak and SULmax were related to P53. In addition, early SULpeak and delayed MTV were significant prognostic factors of 1-year OS, while early SUVpeak, delayed TLG and delayed MTV were predictive factors of 2-year OS in NSCLC. Conclusion: The metabolic and volumetric parameters of18 F-FDG PET/CT were related to a variety of factors such as NSE, CFYRA21-1, SCC-ag, P53 and TNM stage, and have a predictive value in prognosis of NSCLC. This study explores naming practices among Chinese international students and their relation to personal identity during their sojourn in Japan. Although previous studies have reported that some Chinese international students in English-speaking countries adopt names of Western origin (Cotterill 2020; Diao 2014; Edwards 2006), participants in this study were found to exhibit different naming practices: either adopting names of Japanese or Western origin; or retaining both Western and Japanese names. Drawing on fifteen semi-structured interviews with Mainland Han Chinese students, this investigation examines their motivations for adopting non-Chinese names and determines how personal identities are presented through them. The qualitative analysis reveals that the practice of adopting non-Chinese names is influenced by teacher-student power relations, Chinese conventions for terms of address, pronunciation, and context-sensitivity of personal names. As will be shown in this article, through the respondents' years of selfexploration, their self-adopted non-Chinese names gradually became internalized personal identity markers that allow the bearers to explore and exhibit personality traits, which might not have been as easily displayed via their Chinese given names. Decades of research on communication strategies of English learner has offered a full picture of the field. However, previous studies are generally concerned with either daily or classroom conversation whereas little has been known concerning the language strategies in academic debate with non-native speakers of English being the debaters. Given the debater's dual identities as language learners as well, strategies utilized in debate may be influenced by a wider scope of elements that are peculiar to the situation. Centering on language-related strategies used by Japanese EFL debaters, this study made use of discourse analysis of a national parliamentary debate tournament. Combining rhetorical situation theory, the results suggest that the debaters adopted strategies including audience-oriented strategies and speech-structuring strategies that are considered unique to debate for the purpose of better conveyance of information. This indicates the debaters' awareness of a differing rhetorical situation which calls for different language-related strategies from daily conversation. Additionally, the following strategies are frequently used by debaters in the current study: fillers, code-switching, similar-sounding words, Japanese-made English, and paraphrase.

Abstract Background Qiangji Jianli Fang (QJF) has been used for treatment of myasthenia gravis (MG) in China. However, our understanding of the effects of QJF against MG at the molecular level is limited. This study aims to investigate the effects of QJF treatment of MG patients on the protein, peptide and metabolite levels in serum. Methods High-throughput proteomic, peptidomic and metabolomic techniques were applied to investigate serum samples from 21 healthy individuals and 47 MG patients before and after QJF treatment via two-dimensional gel electrophoresis, matrix-assisted laser desorption/ionization time of flight mass spectrometry and liquid chromatography Fourier transform mass spectrometry, respectively. Results After QJF treatment, the expression levels of peptides m/z 1865.019, 2021.128 and 1211.668 of complement C3f increased (P = 0.004, P = 0.001 and P = 0.043, respectively), while that of peptide m/z 1739.931 of component C4b decreased (P = 0.043), in the serum of MG patients. The levels of γ-aminobutyric acid (P = 0.000) and coenzyme Q4 (P = 0.000) resumed their normal states. Conclusion QJF could inhibit the activity of the complement system and restore the normal levels of metabolites.

Esophageal squamous cell carcinoma (ESCC) is one of the most common and serious malignancies in China. However, the exact mechanisms of tumor formation and progression are unclear. As late diagnosis and poor therapeutic efficacy result in lower survival rates, identifying biomarkers for early detection, prognostic evaluation, and recurrence monitoring of ESCC is necessary. Here we analyzed 10 protein expression profiles of ESCC core tissues and paired normal esophageal epithelial tissues using two-dimensional gel electrophoresis. We excised 29 protein spots with two-fold or greater differential expression between cancer and normal tissues and identified them using matrix-assisted laser desorption/ionization-time-of-flight/time-of-flight mass spectrometry. The role of PA28β in ESCC cell was confirmed using cell growth, colony formation and soft agar in TE-1 cells pre- and post- PA28β transfection. Compared to their expression in the adjacent normal epithelia, 12 proteins, including transgelin (TAGLN), were upregulated in ESCC tissues; 17 proteins, including proteasome activator 28-beta subunit (PA28β), were downregulated (p < 0.05). Western blotting and immunohistochemistry confirmed that PA28β was significantly underexpressed in ESCC tissues. The functional assays demonstrate that PA28β inhibited cell growth, proliferation and malignancy of TE-1 cells. Among the differentially expressed proteins, PA28β is a potential tumor inhibitor.

Background Breastfeeding confers substantial benefits to child health and has also been associated with lower risk of maternal cardiovascular diseases ( CVDs ) in later life. However, the evidence on the effects of CVD is still inconsistent, especially in East Asians, in whom the frequency and duration of breastfeeding significantly differ from those in the West. Methods and Results In 2004–2008, the nationwide China Kadoorie Biobank recruited 0.5 million individuals aged 30 to 79 years from 10 diverse regions across China. During 8 years of follow‐up, 16 671 incident cases of coronary heart disease and 23 983 cases of stroke were recorded among 289 573 women without prior CVD at baseline. Cox regression yielded adjusted hazard ratios ( HR s) and 95% CIs for incident CVD by breastfeeding. Overall, ≈99% of women had given birth, among whom 97% reported a history of breastfeeding, with a median duration of 12 months per child. Compared with parous women who had never breastfed, ever breastfeeding was associated with a significantly lower risk of CVD , with adjusted HR s of 0.91 (95% CI, 0.84–0.99) for coronary heart disease and 0.92 (95% CI, 0.85–0.99) for stroke. Women who had breastfed for ≥24 months had an 18% ( HR, 0.82; 0.77–0.87) lower risk of coronary heart disease and a 17% ( HR, 0.83; 0.79–0.87) lower risk of stroke compared with women who had never breastfed. Among women who ever breastfed, each additional 6 months of breastfeeding per child was associated with an adjusted HR of 0.96 (95% CI, 0.94–0.98) for coronary heart disease and 0.97 (95% CI, 0.96–0.98) for stroke. Conclusions Among Chinese women, a history of breastfeeding was associated with an ≈10% lower risk of CVD in later life and the magnitude of the inverse association was stronger among those with a longer duration of breastfeeding.
