Interlocus sexual conflict (IRSC) occurs because of shared interactions that have opposite effects on male and female fitness. Typically, it is assumed that loci involved in IRSC have sex-limited expression and are thus not directly affected by selective pressures acting on the other sex. However, if loci involved in IRSC have pleiotropic effects in the other sex, intersexual selection can shape the evolutionary dynamics of conflict escalation and resolution, as well as the evolution of reproductive traits linked to IRSC loci, and vice versa. Here we used an artificial selection approach in Japanese quail (Coturnix japonica) to test if female-limited selection on reproductive investment affects the amount of harm caused by males during mating. We found that males originating from lines selected for high female reproductive investment caused more oxidative damage in the female reproductive tract than males originating from lines selected for low female reproductive investment. This male-induced damage was specific to the oviduct and not found in other female tissues, suggesting that it was ejaculate-mediated. Our results suggest that intersexual selection shapes the evolution of IRSC and that male-induced harm may contribute to the maintenance of variation in female reproductive investment.

Body condition (BC), is a measure to assess the health status of domestic and wild animals. When food resources are abundant, a decrease in BC may indicate an increase in the energetic expenditure due to the effects of growth, reproduction, or disease. BC impoverishment is one of the most common clinical effects of diseases progressing chronically, such as animal tuberculosis (TB) caused by bacteria belonging to the Mycobacterium tuberculosis complex. The Eurasian wild boar (Sus scrofa) is the main wild TB reservoir in the Mediterranean basin. The specific aims of this work were to assess the relationship between sex, age and TB severity altogether on the BC of wild boar. For this purpose, we used the kidney fat index (KFI), to assess the impact of TB progression on the BC of 1372 hunter-harvested free-ranging wild boar in seven populations in southern Spain. Surprisingly, TB had only slight effects on wild boar BC and individuals exhibiting severe TB showed greater BC than TB-free individuals. The age (adults had greater BC than juveniles) and sex (females had greater BC than males) were the main BC determinants in wild boar. Sampling population and season explained more BC variability than individual factors, suggesting that other external factors might play an important role in the BC, and probably on the impact of the disease on this wild reservoir. The low impact of TB on wild boar BC suggests that individuals with severe TB and good BC represent potential long-term super-shedders of this pathogen.

[Aim] The increasing spread of vector-borne diseases has resulted in severe health concerns for humans, domestic animals and wildlife, with changes in land use and the introduction of invasive species being among the main possible causes for this increase. We explored several ecological drivers potentially affecting the local prevalence and richness of avian malaria parasite lineages in native and introduced house sparrows (Passer domesticus) populations., [Location] Global., [Time period] 2002–2019., [Major taxa studied] Avian Plasmodium parasites in house sparrows., [Methods] We analysed data from 2,220 samples from 69 localities across all continents, except Antarctica. The influence of environment (urbanization index and human density), geography (altitude, latitude, hemisphere) and time (bird breeding season and years since introduction) were analysed using generalized additive mixed models (GAMMs) and random forests., [Results] Overall, 670 sparrows (30.2%) were infected with 22 Plasmodium lineages. In native populations, parasite prevalence was positively related to urbanization index, with the highest prevalence values in areas with intermediate urbanization levels. Likewise, in introduced populations, prevalence was positively associated with urbanization index; however, higher infection occurred in areas with either extreme high or low levels of urbanization. In introduced populations, the number of parasite lineages increased with altitude and with the years elapsed since the establishment of sparrows in a new locality. Here, after a decline in the number of parasite lineages in the first 30 years, an increase from 40 years onwards was detected., [Main conclusions] Urbanization was related to parasite prevalence in both native and introduced bird populations. In invaded areas, altitude and time since bird introduction were related to the number of Plasmodium lineages found to be infecting sparrows.

Aedes albopictus is an invasive mosquito species that can maintain and transmit several arboviruses causing disease in humans. Understanding the determinants of its ecology and population dynamics to predict its abundance was the main objective of this study. Adult mosquitoes were captured weekly between 2009 and 2016 with BG sentinel traps baited with BG-Lure outdoors at a collection site within the urban area of Palermo (southern Italy). In parallel, between 2012 and 2016, we monitored the uninterrupted weekly abundance of Ae. albopictus at four additional sites nearby over an area of about two hectares. Catches were collected three times per week and mosquitoes were identified morphologically. To identify the determinants of mosquito abundance, seasonal autoregressive integrated moving-average and Poisson regression models were fitted to the weekly abundance of Ae. albopictus with a series of weather predictors that potentially modulate its activity and population dynamics. The time lag of the influence of predictors was analysed to identify the intergenerational environmental determinants of Ae. albopictus population dynamics. A cross-validation of the predictive accuracy of the different models was carried out to select the best predictive model. Over 7 years we captured 12,152 Ae. albopictus in the first trap and another 58,710 in four years of trapping in four additional traps. Aedes albopictus abundance was highly seasonal, with activity between mid-March and late December, highest abundances between July and September, and peak abundances in autumn. The predictive potential of the best model was further externally validated with four years of data from the other four traps, showing a high predictive capacity and a very good fit of seasonality and abundance peaks. In simplified agricultural landscapes, some rodent species such as the common vole (Microtus arvalis) can reach high abundances and become agricultural pests. Crop field margins are a key structural element and, therefore, expected to play a key role in vole abundance, even within the demographic cycles that govern vole population dynamics. Here, we aim to identify i) the margin variables that determine vole population response, and ii) at which spatial scale this response can be identified and, therefore, managed. We sampled vole abundance in field margins in two replicated areas of north-western Spain during five years, including all population phases. Environmental variables related to vegetation structure, nearby crops and presence of streams or ditches were extracted at two different spatial scales: a precise small spatial scale -“trapping point” scale-, which referred to the exact location of the traps, and a broader spatial scale -“margin” scale-, that referred to the average values across the whole margin length. Using linear mixed models, we analysed the effects of the environmental variables at both spatial scales during the vole population cycle phases. The broad spatial scale accounted better for vole abundance response, being the latter dependent on the population cycle phase. The stronger effect of environmental variables consisted on vegetation structure effects during the peak phase. In this phase, margins with high cover and short vegetation promote high vole abundances, whilst margins with less cover and taller vegetation, usually associated with higher stability in margins, were related to lower peak abundances. No clear effect of nearby crops was detected in our models, when all variables were accounted for. Our results suggest that keeping stable and well vegetated field margins leads to lower abundance within crop field margins during the population outbreaks, and that any successful management strategy should be addressed to the full extent of

ApTOLL is an aptamer specifically designed to antagonize toll-like receptor 4 (TLR4), which is involved in the innate immunity that promotes inflammatory responses in several diseases, including multiple sclerosis (MS). MS is a chronic, immune, demyelinating and neurodegenerative disease of the central nervous system that represents the second most important cause of neurological disability in young adults. The drugs currently available to treat this disease are immunomodulators and, to date, there are no therapeutic remyelinating drugs available to manage MS. In this study, we show that TLR4 is located in post-mortem cortical lesions of MS patients and as a result, we evaluated the effect of its inhibition by ApTOLL in two different animal models of MS, that of experimental autoimmune encephalomyelitis (EAE) and the cuprizone model. ApTOLL administration ameliorated the clinical symptomatology of the affected mice, which was associated with better preservation and restoration of myelin and oligodendrocytes in the demyelinated lesions of these animals. This revealed not only an immunomodulatory but also a remyelinating effect of the treatment with ApTOLL which was corroborated on purified cultures of rodent and adult human oligodendrocyte precursor cells (OPCs). In summary, the molecular nature of ApTOLL and its mechanism of action strongly supports its further study and use in novel strategies to treat MS and eventually, other demyelinating diseases.

Taxa restricted to mountains may be vulnerable to global warming, unless localscale topographic variation and conservation actions can protect them against expected changes to the climate. We tested how climate change will affect the 19 mountain-restricted Erebia species of the Iberian Peninsula, of which 7 are endemic. To examine the scope for local topographic variation to protect against warming, we applied species distribution models (HadGEM2 and MPI) at two spatial scales (10 10 and 1 1 km) for two representative concentration pathways (RCP4.5 and RCP8.5) in 2050 and 2070. We also superimposed current and future ranges on the protected area (PA) network to identify priority areas for adapting Erebia conservation to climate change. 4. In 10 10 km HadGEM2 models, climatically suitable areas for all species decreased in 2050 and 2070 (average 95.7%). Modelled decreases at 1 1 km were marginally less drastic (95.3%), and 14 out of 19 species were still expected to lose their entire climatically favourable range by 2070. The PA network is well located to conserve the species that are expected to retain some climatically suitable areas in 2070. However, we identify 25 separate 10 10 km squares where new PAs would help to adapt the network to expected range shifts or contractions by Erebia. Based on our results, adapting the conservation of range-restricted mountain taxa to projected climate change will require the implementation of complementary in situ and ex situ measures alongside urgent climate change mitigation.

Habitat use is a virtually universal activity among animals and is highly relevant as regards designing wildlife management and conservation actions. This has led to the development of a great variety of methods to study it, of which resource selection functions combined with biologging-derived data (RSF) is the most widely used for this purpose. However this approach has some constraints, such as its invasiveness and high costs. Analytical approaches taking into consideration imperfect detection coupled with camera trap data (IDM) have, therefore, emerged as a non-invasive cost-effective alternative. However, despite the fact that both approaches (RSF and IDM) have been used in habitat selection studies, they should also be comparatively assessed. The objective of this work is consequently to assess them from two perspectives: explanatory and predictive. This has been done by analyzing data obtained from camera traps (60 sampling sites) and biologging (17 animals monitored: 7 red deer Cervus elaphus, 6 fallow deer Dama dama and 4 wild boar Sus scrofa) in the same periods using IDM and RSF, respectively, in Doñana National Park (southern Spain) in order to explain and predict habitat use patterns for three studied species. Our results showed discrepancies between the two approaches, as they identified different predictors as being the most relevant to determine species intensity of use, and they predicted spatial patterns of habitat use with a contrasted level of concordance, depending on species and scale. Given these results and the characteristics of each approach, we suggested that although partly comparable interpretations can be obtained with both approaches, they are not equivalent but rather complementary. The combination of data from biologging and camera traps would, therefore, appear to be suitable for the development of an analytical framework with which to describe and characterise the habitat use processes of wildlife.

In order to monitor wildlife populations in a manner that supports policy makers and natural resource managers, data must be collected using frameworks and methodologies that allow for comparisons between projects and across time. Though hunting statistics may represent a reliable data source for monitoring population trends in game species, a standardised framework for collecting and analysing this data has never been established in Europe, even within countries. Here we describe a case study on the use of hunting statistics in Spain in order to (i) describe the variability in big game statistics collection frameworks across mainland regions of Spain and (ii) propose a minimum common denominator for a standardised approach at the country level. The main differences in methodologies identified are that each region collects different variables, uses different spatial and temporal resolution, and follows different methodologies. We described spatial patterns by grouping regions based on similarities in the hunting data collection system and identified socio-economic factors as a potential driver of differences in methodologies among regions. Hunting effort-related variables and improved temporal resolution (to the event level) must be incorporated in order to achieve country-level standardisation of methodologies. The use of application software to collect information from the field in a standardised way is recommended, which necessitates engaging stakeholders as part of the monitoring process. Applications software should be designed intentionally, and only after clear objectives for the monitoring program have been defined. Making hunting data open access will improve collaboration and information transfer to scientific and professional sectors. Our recommendations can be adapted to other European countries, which would make hunting data more useful for population monitoring and wildlife policy-making at large spatial scales. The main drawback in developing in vitro embryo production (IVP) systems in wild species, such as the Iberian red deer, is that access to these animals is usually restricted, and long distances from the collection site to the laboratory are usually inevitable. Prolonged ovary storage is known to negatively influence the quality and developmental competence of the oocytes used for IVP. To overcome this issue, we evaluated the effect of adding a caspase-3 inhibitor, z-DEVD-fmk, to the in vitro maturation media to improve the quality and developmental potential of Iberian red deer oocytes. Oocytes were in vitro matured with and without z-DEVD-fmk, and the following parameters were analyzed: viability, early apoptosis, caspase-3 activity, DNA fragmentation, and relative abundance of mRNA transcript related to apoptosis. Moreover, oocyte maturation and blastocyst rates were also assessed. The results showed that z-DEVD-fmk decreased early apoptosis (inhibitor= 44.44 ± 3.6% vs. control= 60 ± 2.79%), DNA fragmentation (inhibitor= 57.83 ± 1.91% vs. control= 74.62 ± 1.91%), caspase-3 activity (inhibitor= 41.88 ± 3.42% vs. control= 67.10 ± 3.42%) and the relative abundance of TP53 and ITM2B transcripts, as well as increased the number of live (inhibitor= 41.48 ± 2.32% vs. control= 20 ± 1.8%) and in vitro-matured oocytes (inhibitor= 88.18 ± 1.99% vs. control= 74.01 ± 1.99%) rates. Nevertheless, the blastocyst production was not different between both experimental groups (inhibitor: 7.35 ± 2.30 vs. control: 13.77 ± 2.30). The supplementation of z-DEVD-fmk to the maturation medium improved the quality of Iberian red deer oocytes. Further research and alternative strategies are needed to evaluate if this inhibitor could still enhance the developmental potential of oocytes during prolonged ovarian transport.

Wildlife is an important reservoir of zoonotic pathogens. The objective of the present study was to assess the importance of wild ungulates in the epidemiology of Rickettsia spp. Ticks and spleen samples were collected from 262 red deer (Cervus elaphus) and 83 wild boar (Sus scrofa) hunted in southwestern Spain over a 5-year period. DNA was extracted from tick pools (n = 191) and spleens (n = 345), and two nested PCR assays targeting the rOmpA and rOmpB genes were used to detect Rickettsia DNA. Five tick species were identified (Hyalomma lusitanicum, Dermacentor marginatus, Ixodes ricinus, Rhipicephalus bursa and Haemaphysalis sulcata). Rickettsia DNA was detected in 31 (16.2%) tick pools and two red deer spleen samples (0.8%). Four validated Rickettsia species (R. slovaca, R. monacensis, R. helvetica and R. raoultii), one uncultivated species (Candidatus R. rioja) and two uncharacterized Rickettsia spp. were detected in ticks. R. helvetica and R. slovaca were also detected in spleen samples from red deer. The overall prevalence in ungulate spleen samples was lower than in tick pools suggesting that these ungulates do not play a major role in the transmission of Rickettsia spp. However, their importance as spreaders of positive ticks cannot be ruled out. The results present a challenge for the veterinary and public health communities since most of the Rickettsia spp. detected are pathogenic. Furthermore, the new Rickettsia species could be potential pathogens. For these reasons, identifying Rickettsia species present in ticks and wildlife is of particular interest to clarify their sylvatic cycle and establish appropriate control measures.

Nest predation is the main cause of reproductive failure, particularly in ground-nesting birds on farmlands. Understanding the links between nest predation and habitat change can help design effective management schemes to constrain the negative impact of predation pressure on birds. However, the mechanisms underlying the relationships between landscape attributes, predator distribution, and nest predation are still unclear. Here, we use an experimental approach to examine the effects of distance to the hedgerow as well as hedgerow and forest densities on the abundance of major mesopredators of ground nests of our study area (i.e., corvids) and on the predation rate of artificial ground nests (n = 2576). We found evidence that landscape configuration influenced predation patterns differently depending on the predator species. Nest predation by corvids was more likely in homogeneous and open agricultural landscapes with a low density of forest and hedgerows, whereas predation by other predators was more likely close to hedgerows. Nest predation by corvids and the abundance of corvids also tended to be lower in landscapes dominated by grasslands. Other variables such as road density and distance to human settlements had contrasted effects on the likelihood of a nest being depredated by corvids, i.e., no effect with proximity to human settlements and decreasing trend with road density. Altogether, our results suggest that landscape features interact with mesopredator distribution and their predation rates of ground nests. Therefore, from a conservation and management perspective, a heterogeneous agricultural landscape that includes a mixture of crops associated with patches of forests, hedgerows, and grasslands offering alternative food to generalist predators should contribute to reducing ground-nesting bird predation.

Seed treatment with pesticides is an extended agricultural practice with a high risk to granivorous birds that consume those seeds. To characterize that risk, it is necessary to understand the ecological factors that determine the exposure chances of birds to treated seeds. We investigated how pesticide uptake by red-legged partridges was related to cultivated plant ingestion and to the use of recently sown fields. We analyzed pesticide residues in 144 fecal samples from 32 flocks and determined the plant diet composition using DNA metabarcoding. Habitat use was studied through the monitoring of 15 GPS-tagged partridges. We confirmed, through the analysis of seeds, that >80% of cereal fields from the area had seeds treated with triazole fungicides. Tebuconazole was detected in 16.6% of partridges' feces. During the sowing season, cultivated plants accounted for half of the plant diet, but no association was found between cultivated plant consumption and pesticide intake. GPS tracking revealed that tebuconazole was detected in feces when partridges had recently used sown fields, whereas nonexposed partridges showed no overlap with recently sown areas. Our results highlight the need to incorporate field ecology into the characterization of pesticide exposure to improve the efficacy of environmental risk assessment.

Estimation of changes in abundances and densities is essential for the research, management, and conservation of animal populations. Recently, technological advances have facilitated the surveillance of animal populations through the adoption of passive sensors, such as camera traps (CT). Several methods, including the random encounter model (REM), have been developed for estimating densities of unmarked populations but require additional information. Hierarchical abundance models, such as the N-mixture model (NMM), can estimate abundances without performing additional fieldwork but do not explicitly estimate the area effectively sampled. This obscures the interpretation of its densities and requires its users to focus on relative measures of abundance instead. Hence, the main objective of our study is to evaluate if REM and NMM yield consistent results qualitatively. Therefore, we compare relative trends: (i) between species, (ii) between years and (iii) across years obtained from annual density/abundance estimates of three species (fox, wild boar and red deer) in central Spain monitored by a camera trapping network for five consecutive winter periods. We reveal that NMM and REM provided density estimates in the same order of magnitude for wild boar, but not for foxes and red deer. Assuming a Poisson detection process in the NMM was important to control for inflation of abundance estimates for frequently detected species. Both methods consistently ranked density/abundance across species (between species trend), but did not always agree on relative ranks of yearly estimates within a single population (between years trend), nor on its linear population trends across years (across years trend). Our results suggest that relative trends are generally consistent when the range of variability is large, but can become inconsistent when the range of variability is smaller.

Rewilding is habitat restoration that attempts to return ecosystems to their state before human alterations were imposed. Rewilding might involve actions that are not desired by society. As a result, a compromised form of rewilding has been proposed that is generally known as rewilding lite, which involves some human intervention to achieve the goals of a more natural state of ecosystems that were historically disturbed by anthropogenic influences. Rewilding lite is gaining traction in Europe where marginal agriculture areas are being abandoned. We addressed a variant that we called livestock rewilding (LR) based on the use of herbivorous livestock species to restore ecological functions and trophic complexity (functional integrity) in abandoned agricultural landscapes. Our objective was to assess societal attitudes and awareness of LR in Spain based on the answers of 1,036 respondents of a self-selecting questionnaire of 27 closed-ended questions addressed to the general public using snowball sampling, conducted between March and July 2021. Seventy-five percent of the respondents supported LR but only on the condition of imposing minimum hazards to people, crops, and property. Ranchers were the least likely societal group to support LR (58%). Women had the greatest enthusiasm for LR (79% women vs. 71% men). Horses, donkeys, and goats were the most preferred species to be included in LR programs, with pigs as the least popular after cattle and sheep. There was support for the removal of excess individuals of rewilded livestock and for the use of their meat for human consumption, especially among male respondents. Respondents were reticent to accept mortality in LR populations caused by natural stochastic events and discriminated between livestock species for population control methods. Motion-sensitive cameras are popular as non-invasive monitoring tools, and several methods have been developed to estimate population densities from camera data. These methods frequently rely on auxiliary movement data including the distance traveled by an individual in a day and the proportion of the day that an animal spends moving when individual recognition is not possible. The estimation of these movement parameters is time-consuming, which could limit the applicability of cameras to estimate population density. To investigate the relevance of measuring movement parameters for the target population, we monitored 54 wildlife populations of red deer (Cervus elaphus), fallow deer (Dama dama), roe deer (Capreolus capreolus), wild boar (Sus scrofa), and red fox (Vulpes vulpes) in different seasons through Europe with cameras. We estimated 91-day ranges and activity levels. We fitted mixed models for day range and activity level as response variables to assess if the inter-population variability in movement was explained by a set of a priori relevant geographical, environmental, biological, and management predictors. We then explored the bias in density estimates obtained in 25 independent populations when using predicted movement data. There was high intra-species variation in day range and activity level among species and populations. Only species explained a small proportion of this variability; other predictor variables did not. We observed bias in densities when predicting the day range and activity for independent populations. Considering the intra-species variability in movement parameters and the consequent unacceptable bias in density estimates, we recommend that monitoring and conservation programs estimate movement parameters for the target population and survey populations from camera data for more accurate density estimates. The management of animal tuberculosis (TB) is a priority for European Union animal health authorities. However, and despite all the efforts made to date, a significant part of Spain has as yet been unable to obtain the officially tuberculosis-free (OTF) status. Information regarding wildlife disease status is usually scarce, signifying that the role played by wildlife is usually ignored or poorly assessed in large-scale TB risk factor studies. The National Wildlife Health Surveillance Plan in Spain now provides information on infection rates in wildlife reservoirs at a national level, but there are limitations as regards the sample size, the spatio-temporal distribution of the samples, and the lack of homogeneity of the diagnostic techniques employed. The objective of the study described herein was, therefore, to employ a Bayesian approach with the intention of identifying the risk factors associated with four TB rates in cattle: prevalence, incidence, maintenance and persistence in Spain during the period 2014–2019. The modeling approach included highly informative spatio-temporal latent effects with which to control the limitations of the data. Variation partitioning procedures were carried out, and the pure effect of each factor was mapped in order to identify the most relevant factors associated with TB dynamics in cattle in each region. This made it possible to disclose that the movement of cattle, particularly from counties with herd incidence > 1%, was the main driver of the TB dynamics in cattle. The abundance of herds bred for bullfighting was retained in all four models, but had less weight than the movements. After accounting for farm-related factors, the TB prevalence in wild boar was retained in all the models and was significantly related to incidence, maintenance and persistence. Rewilding is a restoration strategy that aims to return anthropogenic ecosystems to a “self-organized” state, by reinstating trophic complexity through disturbance (e.g. predation, herbivory), dispersal and connectivity. In depopulated areas of Europe, lite versions of rewilding, that maintain but minimize the management of rewilding species (e.g. predators, large herbivores) is gaining support. Livestock rewilding (LR) is a form of rewilding-lite, that uses livestock landraces as keystone species in the restoration of herbivory (the functional integrity of ecosystems) offering ecosystem services, such as ecotourism and the sale of livestock population surpluses, that can mitigate the economic and social effects of rural depopulation. Many challenges remain to implementing LR, including (i) more empirical evidence is required of the feasibility of LR across a variety of habitats and conditions, and (ii) understanding the hurdles that legislation poses for LR, the latter being the aim of this study. To accomplish this, we reviewed the EU legislation on environmental protection, animal health and welfare, identification and traceability, and ownership and civil responsibility, to assess how this might apply to LR. Although there is no specific EU legislation prohibiting LR, the review indicates that it is not clear what legislation applies to LR, as LR’s status lies between that of livestock and wild species. As such the existing legislation can be a serious impediment to the development of LR programmes. We highlight the needs for a legal definition, and status of LR species and their ownership. We propose ways to adapt this legislation to support the application of LR programmes in abandoned areas of EU, for example, by using legal exceptions intended for livestock under extensive animal farming systems.

The novel fluorescent sensor is proposed in this work based on the competitive interactions between the known bioactive compounds, riboflavin and catechin, which act as guests, and graphene oxide (GO) material that acts as host. Specifically, this proposal is based on an indicator displacement assay which allows the detection of GO and catechin (fluorescence quenching of riboflavin signal by GO and increase in fluorescence by catechin on the GO–riboflavin system). Three GO structures with different lateral dimensions and thickness were synthesized and tested, being able to be the three differentiated depending on the attenuation capacity of the fluorescent signal that each one possesses. The environmental analytical control of GO is more and more important, being this method sensitive and selective in the presence of other potential interferent substances. On the other hand, the other sensing capacity of the sensor also allows the determination of catechin in food samples based on the formation of riboflavin–GO complex. It is a rapid, simple and non-expensive procedure. Thus, the same 2D nanomaterial (GO) is seen to play a double role in this sensing strategy (analyte and analytical tool for the determination of another compound).

In this study, we analysed the effect of human-mediated selection on the gene pool of wild and farmed red deer populations based on genotyping-by-sequencing data. The farmed red deer sample covered populations spread across seven countries and two continents (France, Germany, Hungary, Latvia, New Zealand, Poland, and Slovakia). The Slovak and Spain wild red deer populations (the latter one in a large game estate) were used as control outgroups. The gene flow intensity, relationship and admixture among populations were tested by the Bayesian approach and discriminant analysis of principal components (DAPC). The highest gene diversity (He = 0.19) and the lowest genomic inbreeding (FHOM = 0.04) found in Slovak wild population confirmed our hypothesis that artificial selection accompanied by bottlenecks has led to the increase in overall genomic homozygosity. The Bayesian approach and DAPC consistently identified three separate genetic groups. As expected, the farmed populations were clustered together, while the Slovak and Spanish populations formed two separate clusters. Identified traces of genetic admixture in the gene pool of farmed populations reflected a strong contemporary migration rate between them. This study suggests that even if the history of deer farming has been shorter than traditional livestock species, it may leave significant traces in the genome structure., [Simple Summary] Human activities have left indelible traces in the gene pool of livestock as well as recently domesticated wild animal species which are manifested by changes in their genome structure, often accompanied by the significant loss of biodiversity, especially in the case of small local populations. Despite the benefits of deer farming, intensive artificial selection associated with domestication can lead to several negative effects, including founder effects, inbreeding depression, or rapid decline in effective population size, which are also often present in traditional livestock. This study investigates the effect of human-mediated selection on the diversity of seven farmed red deer populations compared to two wild populations through the quantification of the level of genomic heterozygosity, inbreeding, admixture, and gene flow. These results will help to improve traditional breeding schemes and assist in a more sustainable utilisation of available animal genetic resources.

La fotografía ha constituido desde su invención un medio privilegiado para registrar bienes patrimoniales y manifestaciones culturales. Este artículo estudia la obra de Tomás Camarillo (1879-1954), que testimonió de manera exhaustiva la provincia de Guadalajara, su naturaleza y cultura. Con el fin de estimar el valor de su aportación documental y creativa, se analizan fotografías y ejemplares inéditos. Dentro de su amplio repertorio visual, se presta especial atención a paisajes, vistas de pueblos, obras públicas, escenas costumbristas, retratos y manifestaciones del patrimonio inmaterial. También se reflexiona sobre los usos documentales, turísticos y políticos de la colección.

Over the last 30 years, the study of the cellular response to ionizing radiation (IR) has increased exponentially. Among the various signaling pathways affected by IR, p38 MAPK has been shown to be activated both in vitro and in vivo, with involvement in key processes triggered by IR-mediated genotoxic insult, such as the cell cycle, apoptosis or senescence. However, we do not yet have a definitive clue about the role of p38 MAPK in terms of radioresistance/sensitivity and its potential use to improve current radiotherapy. In this review, we summarize the current knowledge on this family of MAPKs in response to IR as well as in different aspects related to radiotherapy, such as their role in the control of REDOX, fibrosis, and in the radiosensitizing effect of several compounds.

Diabetes mellitus (DM) and calcific aortic stenosis (CAS) are common morbidities in the elderly, which are both chronic, progressive and often concomitant diseases. Several studies revealed that DM increases the risk of developing severe CAS, yet clear information about the relationship between both these diseases and the influence of DM on the progression of CAS is currently lacking. To evaluate the effect of DM on aortic valves and on the process of calcification, and to achieve better patient management in daily clinical practice, we analysed calcified and noncalcified valve tissue from patients with severe CAS, with or without DM. A proteomic strategy using isobaric tags was adopted and the plasma concentrations of nine proteins were studied using 3 orthogonal methods and in a separate cell model. The differentially expressed proteins identified are implicated in biological processes like endopeptidase activity, lipid metabolism, coagulation, and fibrinolysis. The results obtained provide evidence that DM provokes changes in the proteome of aortic valves, affecting valve calcification. This finding may help enhance our understanding of the pathogenesis of CAS and how DM affects the evolution of this condition, an important step in identifying targets to personalize the treatment of these patients.

Root-knot nematodes (RKNs) are obligate endoparasites that feed on their host plants to complete its life cycle, representing a major threat to agriculture and economy worldwide. The development of new management strategies becomes essential as effective chemical nematicides are progressively being restricted. Hence, we analysed grape pomace-derived biochars, pyrolysed at 350 °C (BC350) and 700 °C (BC700), focusing on their potential for RKN control. The thermal treatment of grape pomace caused an increase in the concentration of carbon and plant macro- and micronutrients, which were largely present in a water-soluble form. Synchrotron radiation-based Fourier transform infrared microspectroscopy data showed a general loss of carboxylic functional groups during pyrolysis, partially contributing to the alkalinisation of both biochars, mostly in BC700. Scanning electron microscopy coupled with energy-dispersive X-ray spectroscopy analysis revealed a highly porous structure filled with different crystals composed of elements such as K, Ca, Mg, P, Si or Al, which could be a suitable environment for the growth of microorganisms. Biochar-derived aqueous extracts showed phytotoxicity to tomato seedlings at high concentrations, and disappeared upon dilution, but no toxic effect was observed on the nematode’s infective stage. However, the infective and reproductive traits of a Meloidogyne javanica population in tomato were significantly reduced (i.e. egg masses and eggs per plant) in washed-biochar-treated soil in pots (0.75%; BC350W). Therefore, the large amount of grape waste generated after wine production can be transformed into a valuable product such as biochar, effective for RKNs control, thus reducing the waste management problem and contributing to a circular economy.

We present a method for human brain fixation based on simultaneous perfusion of 4% paraformaldehyde through carotids after a flush with saline. The left carotid cannula is used to perfuse the body with 10% formalin, to allow further use of the body for anatomical research or teaching. The aim of our method is to develop a vascular fixation protocol for the human brain, by adapting protocols that are commonly used in experimental animal studies. We show that a variety of histological procedures can be carried out (cyto- and myeloarchitectonics, histochemistry, immunohistochemistry, intracellular cell injection, and electron microscopy). In addition, ex vivo, ex situ high-resolution MRI (9.4T) can be obtained in the same specimens. This procedure resulted in similar morphological features to those obtained by intravascular perfusion in experimental animals, provided that the postmortem interval was under 10 h for several of the techniques used and under 4 h in the case of intracellular injections and electron microscopy. The use of intravascular fixation of the brain inside the skull provides a fixed whole human brain, perfectly fitted to the skull, with negligible deformation compared to conventional techniques. Given this characteristic of ex vivo, in situ fixation, this procedure can probably be considered the most suitable one available for ex vivo MRI scans of the brain. We describe the compatibility of the method proposed for intravascular fixation of the human brain and fixation of the donor’s body for anatomical purposes. Thus, body donor programs can provide human brain tissue, while the remainder of the body can also be fixed for anatomical studies. Therefore, this method of human brain fixation through the carotid system optimizes the procurement of human brain tissue, allowing a greater understanding of human neurological diseases, while benefiting anatomy departments by making the remainder of the body available for teaching purposes.

The funds of the Museum of Cuenca hold authentic collections of materials that come from archaeological despoilment and from donations and deposits made by private individuals. They constitute a valuable documentation of the cultural and patrimonial heritage of the province. Most of them remain unpublished. Among these materials, there is a group of metal objects, excluding coins, which are characterised by their typological heterogeneity. This volume presents a study of a total of 1.745 pieces, devoid of archaeological context. They have therefore been dated by formal parallels in the peninsular and foreign spheres, covering a chronological period between the Bronze Age and the Middle Ages., Los fondos del Museo de Cuenca custodian auténticas colecciones de materiales procedentes de la actuación de expoliadores clandestinos y de donaciones y entregas realizadas por particulares, que constituyen una valiosa documentación de la riqueza cultural y patrimonial de la provincia. La mayoría de ellos permanecen inéditos. Entre estos materiales, se encuentra un conjunto de objetos de metal, dejando fuera las monedas, que se caracterizan por su heterogeneidad tipológica. Presentamos en este volumen el estudio de un total de 1.745 piezas, desprovistas de contexto arqueológico. Por ello, han sido datadas por paralelos formales del ámbito peninsular y foráneo, que abarcan un período cronológico que se sitúa entre la Edad del Bronce y la Edad Media.

Spanish Royal Decree 865/2003 on the prevention and control of legionellosis mentions the culture method of standard ISO 11731 for testing Legionella spp., a standard which sets different parametric levels. The weaknesses that have been detected in the method of said standard (false negatives and high waiting times) have made it necessary to look for alternatives. The aim of this study is to determine whether the method of immunomagnetic separation (IMS) and quantification of Legionella spp. Legipid® Legionella Fast Detection is suitable and effective for analyzing environmental water samples in an official public health laboratory. In the absence of a standardized methodology for verifying alternative methods, we devised a method for carrying out this verification taking into account specific criteria for microbiological analysis and the ENAC’s guide for test kits. Thus, we calculated the parameters of precision and recovery of the inoculated samples, as well as the linearity from the slope and the ordinate at the origin of the absorbance-colony forming unit (CFU) relationship, and compared the experimental data we obtained with the results of the validation of the method. The results were satisfactory in all cases: a dispersion below 20 %, a recovery in the 4-71 % range, and a linearity compliant with the validation with a significance level exceeding 95 %. Based on these results, we consider the ISM method to be a valid method for detecting and quantifying Legionella spp. in an official public health laboratory., El Real Decreto 865/2003 para la prevención y control de la legionelosis menciona el método de cultivo de la norma ISO 11731 para el ensayo de Legionella spp., norma en torno a la cual, se definen límites paramétricos. Las debilidades observadas en esta ISO (falsos negativos y elevados tiempos de espera), han hecho necesario buscar métodos alternativos. En ausencia de una metodología normalizada para verificar métodos alternativos, se ha diseñado un método para realizar esta verificación, teniendo en cuenta los criterios específicos para análisis microbiológicos y la guía para kits de ensayo de ENAC. Así, se han calculado los parámetros de precisión y recuperación en las muestras inoculadas y, además, la linealidad a partir de la pendiente y la ordenada en el origen de la relación entre absorbancia y unidades formadoras de colonias, comparando los datos experimentales obtenidos con los resultados de la validación del método. Una dispersión inferior al 20 %, una recuperación del 4 - 71 % y una linealidad conforme con la validación a un nivel de significación superior al 95 %. [Background]: Although the anti-HER2 antibody trastuzumab augments patient survival in HER2+ breast cancer, a relevant number of patients progress to this treatment. In this context, novel drug combinations are needed to increase its antitumor activity. In this work, we have evaluated the efficacy of proteolysis targeting chimera (PROTAC) compounds based on BET inhibitors (BETi) to augment the activity of trastuzumab in HER2+ breast cancer models., [Methods]: BT474 and SKBR3 HER2+ breast cancer cell lines were used. The effects of trastuzumab and the BET-PROTAC MZ1 either alone or in combination, were evaluated using MTT proliferation assays, three-dimensional invasion and adhesion cultures, flow cytometry, qPCR and Western blot. In vivo studies were carried out in a xenografted model in mice. Finally, a Clariom_S_Human transcriptomic array was applied to identify deregulated genes after treatments., [Results]: MZ1 induced a higher antiproliferative effect compared to the BETi JQ1. The combination of MZ1 and -trastuzumab significantly decreased cell proliferation, the formation of three-dimensional structures and cellular invasion compared to either of the drugs alone. Evaluation of apoptosis resulted in an increase of cell death following treatment with the combination, and biochemical studies displayed modifications of apoptosis and DNA damage components. In vivo administration of agents alone or combined, to tumors orthotopically xenografted in mice, resulted in a decrease of the tumor volume only after MZ1-Trastuzumab combination treatment. Results from a transcriptomic array indicated a series of newly described transcription factors including HOXB7, MEIS2, TCERG1, and DNAJC2, that were associated to poor outcome in HER2+ breast cancer subtype and downregulated by the MZ1-trastuzumab combination., [Conclusions]: We describe an active novel combination that includes the BET-PROTAC MZ1 and trastuzumab, in HER2+ tumors. Further studies should be performed to confirm these findings and pave the way for their future clinical development.

Basal-like breast cancer is an incurable disease with limited therapeutic options, mainly due to the frequent development of anti-cancer drug resistance. Therefore, identification of druggable targets to improve current therapies and overcome these resistances is a major goal. Targeting DNA repair mechanisms has reached the clinical setting and several strategies, like the inhibition of the CHK1 kinase, are currently in clinical development. Here, using a panel of basal-like cancer cell lines, we explored the synergistic interactions of CHK1 inhibitors (rabusertib and SAR020106) with approved therapies in breast cancer and evaluated their potential to overcome resistance. We identified a synergistic action of these inhibitors with agents that produce DNA damage, like platinum compounds, gemcitabine, and the PARP inhibitor olaparib. Our results demonstrated that the combination of rabusertib with these chemotherapies also has a synergistic impact on tumor initiation, invasion capabilities, and apoptosis in vitro. We also revealed a biochemical effect on DNA damage and caspase-dependent apoptosis pathways through the phosphorylation of H2AX, the degradation of full-length PARP, and the increase of caspases 3 and 8 activity. This agent also demonstrated synergistic activity in a platinum-resistant cell line, inducing an increase in cell death in response to cisplatin only when combined with rabusertib, while no toxic effect was found on non-tumorigenic breast tissue-derived cell lines. Lastly, the combination of CHK1 inhibitor with cisplatin and gemcitabine resulted in more activity than single or double combinations, leading to a higher apoptotic effect. In conclusion, in our study we identify therapeutic options for the clinical development of CHK1 inhibitors, and confirm that the inhibition of this kinase can overcome acquired resistance to cisplatin.

Crimean–Congo haemorrhagic fever virus (CCHFV) is the causative agent of the severe tick‐borne, often fatal, zoonotic Crimean–Congo haemorrhagic fever (CCHF), which is widely distributed worldwide. The CCHFV transmission to humans occurs through tick bite, crushing of engorged ticks or contact with infected host blood. Previously, CCHFV genotype Africa III was reported in Spain. Given the emergence of CCHF and the role of ticks in pathogen maintenance and transmission, we investigated the presence and genotype identity of the virus in tick species parasitizing abundant wild host species in south‐western Spain. A total of 613 adult ticks were collected from hunter‐harvested wild ungulates in twenty locations throughout south‐western Spain. Ticks were identified, nucleic acids were extracted, RNA was analysed by a nested RT‐PCR targeting CCHFV S segment, and the amplicons were sequenced. According to the 212‐bp sequence amplified, the presence of CCHFV human genotype Europe V was detected in Hyalomma lusitanicum and Dermacentor marginatus ticks collected from red deer, fallow deer and Eurasian wild boar in different locations from south‐western Spain. Genotype Africa IV was also detected in a H. lusitanicum tick collected from a red deer. The detection of CCHFV in different tick species collected from various wild hosts and localities provided strong evidence of widespread CCHFV presence in the region, suggesting that the circulation of the virus in Spain requires more attention. Additionally, the identification of the CCHFV genotype Europe V in ticks suggested that its introduction in Spain was probably from Eastern Europe.

Oxidative stress has become a major concern in the field of spermatology, and one of the possible solutions to this acute problem would be the use of antioxidant protection; however, more studies are required in this field, as highly contradictory results regarding the addition of antioxidants have been obtained. Vitamin E is a powerful biological antioxidant, but its low stability and high hydrophobicity limit its application in spermatology, making the use of organic solvents necessary, which renders spermatozoa practically motionless. Keeping this in mind, we propose the use of hydrogels (HVEs) and nanoemulsions (NVEs), alone or in combination, as carriers for the controlled release of vitamin E, thus, improving its solubility and stability and preventing oxidative stress in sperm cells. Cryopreserved sperm from six stags was thawed and extended to 30 × 106 sperm/mL in Bovine Gamete Medium (BGM). Once aliquoted, the samples were incubated as follows: control, free vitamin E (1 mM), NVEs (9 mM), HVEs (1 mM), and the combination of HVEs and NVEs (H + N), with or without induced oxidative stress (100 µM Fe2+/ascorbate). The different treatments were analyzed after 0, 2, 5, and 24 h of incubation at 37 °C. Motility (CASA®), viability (YO-PRO-1/IP), mitochondrial membrane potential (Mitotracker Deep Red 633), lipid peroxidation (C11 BODIPY 581/591), intracellular reactive oxygen species production (CM-H2DCFDA), and DNA status (SCSA®) were assessed. Our results show that the deleterious effects of exogenous oxidative stress were prevented by the vitamin E-loaded carriers proposed, while the kinematic sperm parameters (p ˂ 0.05) and sperm viability were always preserved. Moreover, the vitamin E formulations maintained and preserved mitochondrial activity, prevented sperm lipid peroxidation, and decreased reactive oxygen species (ROS) production (p ˂ 0.05) under oxidative stress conditions. Crimean-Congo haemorrhagic fever virus (CCHFV) continues to cause new human cases in Iberia while its spatial distribution and ecological determinants remain unknown. The virus remains active in a silent tick-animal cycle to which animals contribute maintaining the tick populations and the virus itself. Wild ungulates, in particular red deer, are essential hosts for Hyalomma ticks in Iberia, which are the principal competent vector of CCHFV. Red deer could be an excellent model to understand the ecological determinants of CCHFV as well as to predict infection risks for humans because it is large, gregarious, abundant and the principal host for Hyalomma lusitanicum. We designed a cross-sectional study, analysed the presence of CCHFV antibodies in 1444 deer from 82 populations, and statistically modelled exposure risk with host and environmental predictors. The best-fitted statistical model was projected for peninsular Spain to map infection risks. Fifty out of 82 deer populations were seropositive, with individual population prevalence as high as 88%. The highest prevalence of exposure to CCHFV occurred in the southwest of the Iberian Peninsula. Climate and ungulate abundance were the most influential predictors of the risk of exposure to the virus. The highest risk regions were those where H. lusitanicum is most abundant. Eight of the nine primary human cases occurred in or bordering these regions, demonstrating that the model predicts human infection risk accurately. A recent human case of CCHF occurred in northwestern Spain, a region that the model predicted as low risk, pointing out that it needs improvement to capture all determinants of the CCHFV infection risk. In this study, we have been able to identify the main ecological determinants of CCHFV, and we have also managed to create an accurate model to assess the risk of CCHFV infection.

El orden de los quirópteros es uno de los grupos de mamíferos más abundantes, diversos y extendidos geográficamente por todo el mundo. Treinta y una especies pertenecientes a este orden habitan en la Península Ibérica. La diversidad de hábitats que pueden ocupar, combinada con la capacidad de vuelo, alta longevidad, la dispersión a largas distancias, formación de colonias y adaptación a ambientes antrópicos pueden facilitar el mantenimiento y la dispersión de patógenos zoonóticos potencialmente de relevancia para la salud humana. Además, son capaces de albergar grandes cargas de ectoparásitos altamente especializados como las garrapatas, entre otros artrópodos, que pueden ser portadores de patógenos zoonóticos y probablemente actúen como vectores en las poblaciones de quirópteros. Sin embargo, el papel de los murciélagos como reservorios naturales de patógenos y los factores que determinan los patrones epidemiológicos y riesgos de transmisión con otros grupos de especies es poco conocido. En este estudio se tomaron y se analizaron muestras de sangre de veinticinco especies de quirópteros de cuatro regiones de España. El estudio se centró en la detección, identificación y caracterización molecular de patógenos pertenecientes a los órdenes bacterianos Rickettsiales y Piroplasmida. Dentro de estos grupos, un gran número de especies de Rickettsia spp., Anaplasma spp., Babesia spp. y Theileria spp. son patógenos transmitidos por garrapatas que causan enfermedades relevantes para los seres humanos y los animales silvestres y domésticos. Como base para los factores determinantes de los patrones epidemiológicos encontrados (también multi-patógeno), se analizó la relación entre la prevalencia y diversidad de estos patógenos.

This study describes chemical, physical, microbiological and technological characteristics of red deer milk and the effect of lactation on these parameters in order to know their potential aptitude to elaborate dairy products. During 18 weeks, milk from five hinds was monitored for composition, bacteriology, somatic cell count (SCC), physical properties and rennet coagulation. Mean values (g/100 g) for fat, protein, lactose and dry matter were 10.4, 7.1, 4.3 and 24.2, respectively, and for urea, 265 mg/100 mL. Except for lactose, a significant increase in these components was observed (p < 0.01) as lactation progressed. The average values for bacteriology and SCC were 5.3 log cfu/mL and 4.7 log cells/mL, respectively. Regarding physical properties, conductivity (mean: 2.8 ms/cm), viscosity (3.1 Cp), coordinates L* (89.9) and a* (−3.1) and milk fat globule diameter (D4,3: 6.1 µm) increased along with lactation while density (1.038 g/mL) decreased (p < 0.01). The pH (6.7), acidity (22.9° Dornic), coordinate b* (8.4) and ethanol stability (66.6% v/v) were stable during the study period. The stage of lactation also has a significant impact on milk coagulation properties and mean curd yield was 3.29 g/10 mL. These results suggest that red deer milk could be a potential innovative source of milk for the dairy industry.

The knowledge regarding the spatial ecology of red deer (Cervus elaphus) in different environments is crucial if effective management actions are to be designed. However, this knowledge continues to be scarce in the complex contexts of mixed land use and management circumstances. This study describes the spatial ecology of red deer monitored using GPS collars in Mediterranean ecosystems of South-Central Spain, considering the effect of individual and seasonal (food shortage period, rut, hunting season and food abundance period) factors on different land use and management scenarios, namely protected areas, mixed farms and fenced hunting estates. Our results showed less activity (ACT), a shorter daily range (DR) and a smaller home range (HR) during the food shortage period: ACT: 0.38 ± (SD) 0.12; DR: 3010.9 ± 727.3 m; and weekly HR: 122.2 ± 59.6 ha. With regard to land use, individuals were less ACT and had a smaller DR on fenced hunting estates (ACT: 0.24 ± 0.12; DR: 1946.3 ± 706.7 m) than in protected areas (ACT: 0.59 ± 0.12; DR: 4071.4 ± 1068.2 m) or on mixed farms (ACT: 0.57 ± 0.29; DR: 5431.1 ± 1939.5 m) in all the periods studied. Red deer selected land cover with forage and shelter when foraging and resting, respectively. When drive hunt events occurred (mixed farms and fenced hunting estates), the deer were more prone to select safer habitats (scrublands) and avoid open areas (crops or grasslands) than were their counterparts in protected areas. The patterns observed can be explained by sexual and seasonal differences as regards requirements, the response to disturbances and, interestingly, population management. Our results provide useful information with which to design scientifically-based species adaptive management in response to relevant and timely situations in Europe, such as the potential transmission of shared infections, vehicle collisions, and damage to crops and ecosystems.

Rhipicephalus spp. (Acari: Ixodidae) ticks are obligate hematophagous arthropods, which constitute a model for the study of vector-host interactions. The chemical composition or elementome of salivary glands (SG) and cement provides information relevant for the study of protein-based complex multifunctional tissues with a key role in tick biology. In this study, we characterized the elementome of cement cones in Rhipicephalus sanguineus collected from naturally infested dogs and in SG and cement of R. bursa collected from experimentally infested rabbits at different feeding stages. The elementome was characterized using scanning electron microscopy (SEM) combined with energy dispersive X-ray spectroscopy (EDS). The results showed the identification of up to 14 chemical elements in the cement, and suggested tick/host-driven differences in the cement elementome between tick species and between SG and cement within the same species. By still unknown mechanisms, ticks may regulate cement elementome during feeding to affect various biological processes. Although these analyses are preliminary, the results suggested that N is a key component of the cement elementome with a likely origin in SG/salivary proteins (i.e., Glycine (C2H5NO2)-rich superfamily member proteins; GRPs) and other tick/host-derived components (i.e. NAPDH). Future research should be focused on tick elementome and its functional implications to better understand cement structure and function.

[Aim]: The role of mitochondrial DNA (mtDNA) in local adaptation has been thoroughly debated, and the ancient pervasive mtDNA introgression from Lepus timidus into Lepus granatensis allows testing of this hypothesis. Here, we study the environmental niche of animals with introgressed mtDNA, to gain insights about the potential selective pressures maintaining the introgressed haplotypes., [Location]: Iberian Peninsula., [Taxon]: Lepus granatensis., [Methods]: We modelled the frequency of the L. timidus mtDNA lineage within the L. granatensis distribution area. The response variable was the presence of the L. timidus lineage at the individual level. Finally, the variation in the introgression frequencies was plotted along the environmental gradients (identified by the models), after controlling for latitude., [Results]: The results showed that (i) the niche of the introgressed populations occupy marginal environmental conditions relative to the complete niche of L. granatensis, (ii) both latitude and environment, and their overlaid effects, explain the variation in the introgression frequencies, and (iii) after controlling for latitude, clines in mtDNA introgression frequency are associated with transitions in the environmental gradients, even when the analyses were restricted to the territory where the introgressed haplotypes occur., [Main Conclusions]: While the strict demographic explanation for mtDNA introgression in L. granatensis implies that geography, resulting from the post-glacial expansion of the species, explains the gradients of introgression, our results show correlations with environmental gradients that are not geographically structured. Our results relate the prevalence of introgressed mtDNA with more extreme climatic conditions in the range of L. granatensis, suggesting that environmental selective pressures could have played a role in determining the spatial maintenance of mtDNA haplotypes originating from the cold-adapted species. These results are thus relevant to understand the response of the species to the ongoing processes of global change, which will alter the selective pressures by reducing the specific niche of the introgressed mtDNA lineage.

To date no direct methods are available for the subunit quantification of pheomelanin, the sulfur-containing melanin form whose structure increases melanoma risk. Here we apply Raman spectroscopy combined with chemometrics (MCR-ALS method) to quantify benzothiazine (BT) and benzothiazole (BZ), the constituent monomers of pheomelanin. The Raman spectra of synthetic pheomelanins only composed of BT or BZ were markedly different from those of natural pheomelanins composed of mixed BT-BZ, indicating the necessity of using a multivariate curve resolution method to extract the pure profiles of these monomers in complex samples. MCR-ALS with correlation constraint was applied to the Raman spectra of pheomelanized feathers from 26 bird species to predict BT-BZ concentrations. The methodology was able to quantify BT with R ​= ​0.91 and a relative error of 9%, for the range of 100–1000 ​ng/mg. However, the model did not perform well for BZ quantification. In addition, the MCR-ALS model could retrieve a pure profile associated to keratin, a major component (interference) in feathers. The deconvolution of Raman spectra, which predicts total pheomelanin content, combined with MCR-ALS, which predicts BT content, can therefore provide a complete pheomelanin determination at the monomeric level. This represents a direct and non-destructive analytical method for pheomelanin subunits, opening the door to investigate the differential cytotoxicity of these subunits in a high diversity of biological samples, including melanoma.

Abandoned mine lands (AMLs), which are considered some of the most dangerous anthropogenic activities in the world, are a source of hazards relating to potentially toxic elements (PTEs). Traditional reclamation techniques, which are expensive, time‐consuming and not well accepted by the general public, cannot be used on a large scale. However, plant‐based techniques have gained acceptance as an environmentally friendly alternative over the last 20 years. Plants can be used in AMLs for PTE phytoextraction, phytostabilization, and phytovolatilization. We reviewed these phytoremediation techniques, paying particular attention to the selection of appropriate plants in each case. In order to assess the suitability of plants for phytoremediation purposes, the accumulation capacity and tolerance mechanisms of PTEs was described. We also compiled a collection of interesting actual examples of AML phytoremediation. On‐site studies have shown positive results in terms of soil quality improvement, reduced PTE bioavailability, and increased biodiversity. However, phytoremediation strategies need to better characterize potential plant candidates in order to improve PTE extraction and to reduce the negative impact on AMLs.

The Tablas de Daimiel National Park (TDNP) is a floodplain ecosystem in central Spain with a potential risk of heavy metal and metalloid pollution. The objective of this study was to know the accumulation of arsenic (As), cadmium (Cd), mercury (Hg), lead (Pb), and selenium (Se) in muscle of six species of freshwater fish from the floodplain of TDNP. We obtained muscle samples of Cyprinus carpio (n = 89); Squalius pyrenaicus (n = 16); Ameiurus melas (n = 9); Lepomis gibbosus (n = 8); Micropterus salmoides (n = 6) and Carassius auratus (n = 5). A. melas, which is a predatory species, had significantly higher Hg concentrations than omnivorous or herbivorous species (i.e. C. carpio). On the contrary, A. melas showed lower concentrations of As, Pb and Se than omnivorous species (i.e. S. pyrenaicus and L. gibbosus ). The concentration of Hg was positively associated with fish size in C. carpio and A. melas. Some individuals of C. carpio (5.7%) and S. pyrenaicus (12.5%) showed Pb muscle concentrations above the maximum residue levels established by the European legislation for human consumers. The observed muscle Se concentrations can be associated with adverse effects on fish such as blood changes, reduced growth, mortality of juveniles and reproductive failure. The accumulation of Se in this floodplain located in a seleniferous area and the contamination produced by spent Pb shot pellets used for hunting in the past are discussed as potential sources of the elevated levels of these two elements in fish from this floodplain wetland.

Wildlife management and conservation requires monitoring of species distribution and population indicators, especially when the unbalanced demographic changes of some species can affect the whole ecosystem functioning. The populations of wild boar (Sus scrofa) have, over the past few decades, undergone an expansion around the world, reaching situations of overabundance that can cause serious economic, ecological and health problems. This numerical increase of wild boar and its new spatial invasion can affect certain vulnerable species in sensitive ecological zones, such as the main inland wetland complex in SW Europe. In this context, we aim to (i) examine the association of wild boar abundance and that of lagomorphs, and waterbird productivity, controlling the possible effects of wild carnivores and other environmental predictors, and (ii) applying a survey method useful for managers to evaluate expected effect of wild boar abundance. Overall, the presence of wild boar has been detected in 80.8% of the monitored wetlands (n = 26), but a high variation of abundance rates was found. Wild boar abundance negatively associated with the productivity of the entire community of waterbirds, a priori highly vulnerable, breeding on shores or islands, both colonially (genus Gelochelidon, Himantopus, Recurvirostra, Sternula, Glareola, Tringa and Phoenicopterus) or solitary (genus Anas, Spatula, Mareca, Charadrius, Vanellus, Anser, Ardea, Aythya, Fulica, Netta, Oxyura and Tadorna). In addition, we evidenced a clear negative association of wild boar and wild rabbit (Oryctolagus cuniculus) population abundance in the monitored wetlands. Rabbits are a keystone species in the study area and represent stable prey for many endangered predators. Our results suggest that potential impacts caused by high wild boar abundance may be already reaching unsustainable levels for some wetlands. Se presenta el estudio integral de un soporte decorado con una forma lineal simple recuperado en el abrigo de Buendía, en un contexto magdaleniense datado en ±18000 cal BP. Se estudia el soporte y la forma decorada recurriendo al análisis microscópico a fin de comprender la cadena operativa, concretada en la captación del soporte en el ámbito cercano, la preparación de la superficie antes de la ejecución gráfica, la realización de una decoración lineal simple y la modificación posterior de la superficie tras el dibujo, lo que plantea la pérdida del valor simbólico original. Considerando las evidencias arqueológicas asociadas a la pieza decorada, se concluye que la acción simbólica se llevó a cabo en un contexto multifuncional. La búsqueda de referentes similares en el arte mueble del sudoeste europeo pone de manifiesto la existencia de motivos similares magdalenienses. La comprensión de las cadenas operativas y la contextualización del arte mueble permitirán avanzar en el conocimiento de la función y uso del simbolismo gráfico., We present the systematic study of a blank decorated with a simple linear shape from the Buendía rockshelter, in a Magdalenian context dated at ±18000 cal BP. The blank and its decoration are studied microscopically, in order to understand the operative sequence, which includes the procurement of the blank in the surroundings, preparation of its surface prior to the graphic execution, drawing of a simple linear motif, and the subsequent modification of the surface after the drawing was made. This sequence suggests the loss of its original symbolic value. Considering the archaeological evidence associated with the decorated piece, we conclude that the symbolic action was conducted in a multifunctional context. Similar motives are reported in other Southwest European Magdalenian contexts. A better understanding of portable art operational sequences and their context will enable moving forward in our knowledge of the use and function of graphic symbolism.
