Abstract

INTRODUCCIÓN Y OBJETIVOS: Los tumores vesicales con un patrón morfológico no transicional, constituyen un reto diagnóstico al patólogo y terapéutico al urólogo, pues aunque son poco frecuentes (menos del 5% de los tumores malignos de vejiga), presentan un comportamiento biológico diferente que obliga a un abordaje terapéutico particular. Las variedades anatomopatológicas más importantes son: el carcinoma epidermoide, el adenocarcinoma y el sarcoma primarios. Analizamos nuestra experiencia en este tipo de tumores. MATERIAL Y MÉTODOS: Estudio retrospectivo de los tumores vesicales inusuales tratados en nuestro Hospital entre 1988- 2001. Analizamos su comportamiento biológico y tratamiento aplicado. En todos ellos se descartó el origen extravesical de la tumoración. RESULTADOS: Encontramos 21 casos, cuyas variedades anatomopatológicas fueron: 13 Carcinomas Epidermoides, 7 Adenocarcinomas (3 de Uraco) y un Sarcoma. La edad media fue muy diferente entre los carcinomas epidermoides (69,2 ± 9,1 años) y los adenocarcinomas (50,3 ± 8,1). En las tres variedades histológicas el comportamiento fue muy agresivo. En el momento del diagnóstico 19 pacientes presentaban estadios locales avanzados (≥T2). Aunque el tratamiento electivo fue la cistectomía, sólo se pudo realizar en 10 casos. Quimioterapia sistémica (la más frecuente M-VAC) y/o RT local se aplicó en 7 casos. 14 pacientes han fallecido, con una supervivencia media de 15,7 ± 11,6 meses. CONCLUSIONES: El diagnóstico tardío de estos tumores y su agresivo comportamiento biológico comportan un pronóstico sombrío. Sólo un diagnóstico precoz y un tratamiento radical podrían mejorar su pronóstico. INTRODUCTION AND OBJECTIVES: Non-transitional cell tumours of the bladder are both a diagnostic challenge for the pathologist and a therapeutic challenge for the urologist, because although uncommon (less than 5% of all malignancies of the bladder) they show different biological behaviours each requiring a unique approach. The most significant pathoanatomical types are: primary epidermoid carcinoma, primary adenocarcinoma and primary sarcoma. This paper presents an analysis of our experience in these types of tumours. MATERIAL AND METHODS: A retrospective study of unusual cases of cancer of the bladder seen in our hospital between 1988-2001. Their biological behaviour and the therapies applied are analysed. The extravesical origin of the tumour was ruled out in all cases. RESULTS: We found 21 cases of the following pathoanatomical varieties: 13 epidermoid carcinomas, 7 adenocarcinomas (3 urachal) and 1 sarcoma. Mean age was very different between epidermoid carcinomas (69.2 ± 9.1 years) and adenocarcinomas (50.3 ± 8.1). Tumour behaviour was very aggressive in all three histological varieties. At the time of diagnosis 19 patients had locally advanced stages (≥T2). Although elective therapy was cystectomy, this was only feasible in 10 cases. Systemic chemotherapy (most frequently M-VAC) and/or local radiotherapy was used in 7 cases. 14 patients have died after a mean survival of 15.7 ± 11.6 months. CONCLUSIONS: Late diagnosis of these tumours and their aggressive biological behaviour involve a gloomy prognosis. Only early diagnosis and radical therapy could improve the prognosis.

158. Genetic similarities among Spanisch populations of Agropyron, Elymus and Thinopyrum, using PRC-based markers

Abstract

Arbitrary oligonucleotides were used as primers to amplify genomic DNA of 48 wild Spanish populations of Agropyron cristatum, Elymus hispanicus, E. caninus, E. repens, Thinopyrum curvifolium, Th. junceum and Th. intermedium. Genetic diversity was analysed using nineteen primers. The number of amplified products ranged from 8 to 18 per primer and a total of 240 markers were scored. Different levels of intraspecific genetic diversity were found, the allogamous species E. repens and Th. intermedium being the most variable. Jaccard's similarity coefficients for internal measure within and between populations were used to produce a cluster diagram. The results demonstrate differences in the degree of similarity between taxonomic units. Interpopulational variability and interspecific genomic relationships of these species are discussed.

Abstract

The use of wild species in plant improvement is greatly favored if their genetic variability and taxonomical relationships are known. A study was made of the genetic variability of fifteen Spanish populations of E. caninus and E. hispanicus. The relationships among these species and E. panormitanus were also investigated. Intra- and interpopulational variation was determined by electrophoresis of endosperm proteins. DNA polymorphisms generated by PCR amplification using arbitrary and specific primers were used to determine interpopulational variability and interspecific relationships. The variability observed was similar to that reported for other autogamous species. The reduced variability observed in some populations of E. caninus was attributed to increased distance from the species center of distribution and to the founder effect. Endosperm proteins and random amplified polymorphic DNAs (RAPDs) provided complementary results and differentiated most populations of E. caninus. The interspecific relationships observed were more in agreement with the classification of E. hispanicus as a different species to E. panormitanus than as a variant form of the same taxon.

192. Evaluation and characterisation of a collection of wild Spanish populations of the genera Elymus and Thinopyrum using morphological and agronomical traits

Abstract

Quantification of the variability in wild species is a necessary step before determining their possible future use in plant breeding. A collection of 29 wild Spanish populations of the genera Elymus and Thinopyrum was characterised using 22 morphological and agronomical characteristics. Field studies were performed over two years, 1994 and 1995. Frequencies were calculated for qualitative and quantitative traits. Pearson's product-moment correlation coefficient was used to analyse inter-population variation using frequency data. Means and coefficients of variation were calculated for quantitative characteristics. Analysis of variance was performed for each species to study differences between intra- and inter-populational variability. Similar degrees of variability were observed between quantitative and qualitative traits in all the species studied (T. junceum, T. junceiforme, E. caninus and E. hispanicus). Differences in variability were found at the level of species and population. The relationships between these differences and biological and environmental factors are discussed.

199. Genetic variation in Spanish populations of the genus Aegilops revealed by RAPDs

Abstract

Random amplified polymorphic DNA (RAPD) markers were used to analyse genetic variability and relationships in wild species of the genus Aegilops. Fifty natural populations, which included the species Aegilops biuncialis (UUMM), Ae. neglecta (UUMMNN), Ae. ovata (UUMM), Ae. ventricosa (DDNN) and Ae. triuncialis (UUCC) were selected. These populations are widely distributed in the Iberian peninsula and Balearic islands. Twenty primers were used to generate DNA amplification data. Genetic relationships were established by correspondence analysis. In the resulting dendrogram, Ae. ventricosa appears to be segregated from the other species, probably owing to the influence of the D genome. Ae. biuncialis and Ae. ovata are clearly separated suggesting that the superindex system should be used to differentiate the M genomes of both species. The separation of the M and N genomes was also confirmed, as was the proximity of N to U and C to M. In addition, correlation was found between RAPD markers and ecogeographical factors.
