Abstract

A genetic contribution to the development of systemic lupus erythematosus (SLE) is well established. Several genome-wide linkage scans have identified a number of putative susceptibility loci for SLE, some of which have been replicated in independent samples. This study aimed to identify the regions showing the most consistent evidence for linkage by applying the genome scan meta-analysis (GSMA) method. The study identified two genome-wide suggestive regions on 6p21.1-q15 and 20p11-q13.13 (P-value=0.0056 and P-value=0.0044, respectively) and a region with P-value<0.01 on 16p13-q12.2.The region on chromosome 6 contains the human leukocyte antigen cluster, and the chromosome 16 and 20 regions have been replicated in several cohorts. The potential importance of the identified genomic regions are also highlighted. These results, in conjunction with data emerging from dense single nucleotide polymorphism typing of specific regions or future genome-wide association studies will help guide efforts to identify the actual predisposing genetic variation contributing to this complex genetic disease.

Abstract

Recent interest in atherosclerosis has focused on the genetic determinants of low-density lipoprotein (LDL) particle size, because of (i) the association of small dense LDL particles with a three-fold increased risk for coronary artery disease (CAD) and (ii) the recent report of linkage of the trait to the LDL receptor (chromosome 19). By utilizing nonparametric quantitative sib-pair and relative-pair analysis methods in CAD families, we tested for linkage of a gene or genes controlling LDL particle sizes with the genetic loci for the major apolipoproteins and enzymes participating in lipoprotein metabolism. We confirmed evidence for linkage to the LDL receptor locus (P=.008). For six candidate gene loci, including apolipoprotein(apo)B, apoAII, apo(a), apoE-CI-CII, lipoprotein lipase, and high-density lipoprotein-binding protein, no evidence for linkage was observed by sib-pair linkage analyses (P values ranged from .24 to .81). However, in addition, we did find tentative evidence for linkage with the apoAI-CIII-AIV locus (chromosome 11) (P=.06) and significant evidence for linkage of the cholesteryl ester transfer protein locus (chromosome 16) (P=.01) and the manganese superoxide dismutase locus (chromosome 6) (P=.001), thus indicating multilocus determination of this atherogenic trait.

Abstract

Tuberous sclerosis (TSC) is a dominantly inherited disorder characterized by hamartomas and hamartias in one or more organs, most often in skin, brain, and kidneys. Analysis of the basic genetic defect in tuberous sclerosis would be greatly expedited by definitive determination of the chromosomal location of the TSC gene or genes. We have carried out genetic linkage studies in 15 TSC families, using 34 polymorphic markers including protein markers and DNA markers. Pairwise lod scores were calculated using LIPED, and multipoint analyses were carried out using MENDEL. In the pairwise linkage analysis, using a penetrance value of 90%, a significant positive lod score was obtained with MCT128.1 (D11S144), 11q22-11q23, Zmax 3.26 at theta = 0.08. The tyrosinase probe TYR (11q14-11q22) gave a maximum lod score of 2.88 at theta = 0. In the multipoint analyses the most likely order is (TYR,TSC)-MCT128.1-HHH172. Homogeneity analysis was carried out using the USERM9 subprogram of MENDEL, which conducts the admixture test of C. Smith (1963, Ann. Hum. Genet. 27: 175-182). This test provided no evidence for genetic heterogeneity (that is, non-11-linked families) in this data set.

Abstract

We will present our work on the development of ultra-high-resolution detectors for alpha particle spectrometry. These detectors, based on superconducting transition-edge sensors, offer energy resolution that is five to ten times better than conventional silicon detectors. Using these microcalorimeter detectors, the isotopic composition of mixed-actinide samples can be determined rapidly without the need for actinide separation chemistry to isolate each element, or mass spectrometry to separate isotopic signatures that can not be resolved using traditional alpha spectrometry (e.g. Pu-239/Pu-240, or Pu-238/Am-241). This paper will cover the detector and measurement system, actinide source preparation, and the quantitative isotopic analysis of a number of forensics- and safeguards-relevant radioactive sources. Abstract

Microcalorimeters have been shown to yield unsurpassed energy resolution for alpha spectrometry, up to 1.06 keV FWHM at 5.3 MeV. These detectors use a superconducting transition-edge sensor (TES) to measure the temperature change in an absorber from energy deposited by an interacting alpha particle. Our system has four independent detectors mounted inside a liquid nitrogen/liquid helium cryostat. An adiabatic demagnetization refrigerator (ADR) cools the detector stage to its operating temperature of 80 mK. Temperature regulation with ∼15-μK peak-to-peak variation is achieved by PID control of the ADR. The detectors are voltage-biased, and the current signal is amplified by a commercial SQUID readout system and digitized for further analysis. Abstract

We are developing the next generation of high-resolution high-speed X-ray detectors based on superconducting tunnel junctions (STJs). They consist of 112-pixel arrays of 208 $$\mu $$ m $$\times $$ 208 $$\mu $$ m Ta-Al- $${\text {AlO}}_x$$ -Al-Ta tunnel junctions whose Ta absorber increases the detection efficiency compared to earlier Nb-based STJs. To read out these medium size detector arrays we have also developed a compact and scalable 32-channel preamplifier with an input voltage noise $$<$$ 1 nV/ $$\surd $$ Hz and a dc voltage bias for stable STJ operation between Fiske mode resonances. The pixels have a uniform response across the array, an energy resolution between 7.5 and 9.5 eV FWHM at 525 eV, and can be operated at several 1,000 counts/s per pixel. [ABSTRACT FROM AUTHOR]

Abstract

We are developing a cryogen-free Ta-based superconducting tunnel junction (STJ) detector for soft X-ray spectroscopy at synchrotrons. With an energy resolution 10 times higher than conventional solid-state X-ray detectors and count-rate capabilities above 5 kHz/pixel, STJ detectors offer potentially increased sensitivity for fluorescence-yield X-ray absorption spectroscopy (FY-XAS). We have developed 36-pixel arrays of 208 $$\times $$ 208 $$\upmu $$ m Ta STJs with an energy resolution of $$\sim $$ 9 eV FWHM at the 525 eV oxygen K line. Compared to earlier Nb-based STJs, Ta-STJs offer improved energy resolution and absorption efficiency and extend the operating range to several keV. Here we describe the integration of the 36-pixel arrays into a cryogen-free, user-friendly X-ray spectrometer. A computer-controlled adiabatic demagnetization refrigerator coupled to a two-stage pulse tube refrigerator allows operation below 100 mK. The detector chip is located at the end of a 42 cm shielded snout for insertion into the analysis chamber. The system is currently being commissioned at the Advanced Light Source synchrotron. Abstract

Studying genetic determinants of intermediate phenotypes is a powerful tool to increase our understanding of genotype-phenotype correlations. Metabolic traits pertinent to the central nervous system (CNS) constitute a potentially informative target for genetic studies of intermediate phenotypes as their genetic underpinnings may elucidate etiological mechanisms. We therefore conducted a genome-wide association study (GWAS) of monoamine metabolite (MM) levels in cerebrospinal fluid (CSF) of 414 human subjects from the general population. In a linear model correcting for covariates, we identified one locus associated with MMs at a genome-wide significant level (standardized β=0.32, P=4.92 × 10−8), located 20 kb from SSTR1, a gene involved with brain signal transduction and glutamate receptor signaling. By subsequent whole-genome expression quantitative trait locus (eQTL) analysis, we provide evidence that this variant controls expression of PDE9A (β=0.21; Punadjusted=5.6 × 10−7; Pcorrected=0.014), a gene previously implicated in monoaminergic transmission, major depressive disorder and antidepressant response. A post hoc analysis of loci significantly associated with psychiatric disorders suggested that genetic variation at CSMD1, a schizophrenia susceptibility locus, plays a role in the ratio between dopamine and serotonin metabolites in CSF. The presented DNA and mRNA analyses yielded genome-wide and suggestive associations in biologically plausible genes, two of which encode proteins involved with glutamate receptor functionality. These findings will hopefully contribute to an exploration of the functional impact of the highlighted genes on monoaminergic transmission and neuropsychiatric phenotypes. [ABSTRACT FROM AUTHOR]

Abstract

Generalist predators have the potential advantage to control more than one pest and to be more persistent than specialist predators because they can survive on different foods. Moreover, their population growth rate may be elevated when offered a mixture of prey species. We studied a generalist predatory mite Balaustium sp. that shows promise for biological control of thrips and whiteflies in protected rose cultures in Colombia. Although starting its life in the soil, this predator makes excursions onto plants where it feeds on various arthropods. We quantified life history parameters of the predator, offering high densities of three pest species: first-instar larvae of Frankliniella occidentalis, eggs of Trialeurodes vaporariorum and Tetranychus urticae, either alone or in combination. The predators completed their life cycle on each diet. The egg-to-egg period was c. 2 months. All eggs were laid in one batch in 1–2 days, indicating a pronounced semelparous reproduction pattern. In general, females reproduced earlier and laid more eggs on mixed diets, and these early reproducers consequently had higher population growth rates than late reproducers. The best diet in terms of egg-to-egg period and juvenile survival was the combination of eggs from whiteflies and spider mites. Spider mite eggs alone and western flower thrips larvae alone were the worst diets. It remains to be investigated whether mixed diets promote the population growth rate of Balaustium sufficiently for biocontrol of whiteflies and thrips in the presence of alternative prey, such as spider mites, to become effective. Despite a growing number of children awaiting heart transplant (HT), annual number of HT is relatively unchanged and waitlist mortality is high especially in infants. Donor heart discard rate is also high with ventricular dysfunction as a major reason. Use of donors with low ejection fraction (DLEF) is rare (<15%) in pediatric HT. We evaluated the use of DLEF in pediatric HT using the Pediatric Heart Transplant Society (PHTS) Database. PHTS Database was queried from 1/1993 to 12/2020. DLEF was defined as: donor with left ventricular ejection fraction (LVEF) on pre-procurement echo <54% with wall motion abnormality or LVEF <50% irrespective of wall motion abnormality Donor and recipient characteristics, graft survival and trends of DLEF use across regions and eras were compared. Of 7520 pediatric HT, 7237 (96%) had a donor echo report but only 6332 (84%) had a LVEF reported. Of these, 143 (2.3%) were DLEF with median LVEF 47% [IQR 44-50%]. Focal or diffuse wall motion abnormality was seen in 35 (24%) and 23 (16%) had abnormal septal motion. Recipient characteristics between groups including age, diagnosis, end-organ function, use of VAD and/or ECMO at listing or transplant were similar. Mechanical ventilation at HT (p=.0082), earlier era (p<.0001), and regional variation (p<.0001) were associated with increased use of DLEF. Donor characteristics were similar with the exception that DLEF were more likely to be on epinephrine (20.7% vs 9.9%, p=<.0001) and/or vasopressin (73% vs 54%, p=.0018). DLEF use decreased over time (1993-2002: 4.26%; 2003-12: 2.18%; 2013-20: 1.6%). Differences were seen across UNOS regions (Region 4 vs 10: 0.9% vs 4.2%, p=<.0001). Graft survival at 3 months post-HT was similar for DLEF vs non-DLEF donors (91% vs. 94%, logrank p=0.1) (Fig 1). Use of DLEF in pediatric HT is limited and lower than previously reported. Use of DLEF has decreased over time. DLEF had comparable graft survival at 3 months post-HT, supporting efforts to expand the use of DLEF in pediatric HT. [ABSTRACT FROM AUTHOR]
