The Bear River contributes more water to the eastern Great Basin than any other river system. It is also the most significant source of water for the burgeoning Wasatch Front metropolitan area in Northern Utah. Despite its importance for water resources for the region’s agricultural, urban, and wildlife needs, our understanding of the variability of Bear River’s stream flow derives entirely from the short instrumental record (1943-2010). Here we present a 1,200-year calibrated and verified tree-ring reconstruction of stream flow for the Bear River that explains 67% of the variance of the instrumental record over the period from 1943-2010. Furthermore, we developed this reconstruction from a species that is not typically used for dendroclimatology, Utah juniper (Juniperus osteosperma). We identify highly significant periodicity in our reconstruction at quasi-decadal (7-8 year), multi-decadal (30 year), and centennial (>50 years) scales. The latter half of the 20th century was found to be the 2nd wettest (~40-year) period of the past 1,200 years, while the first half of the 20th century marked the 4th driest period. The most severe period of reduced stream flow occurred during the Medieval Warm Period (ca. mid-1200s CE) and persisted for ~70 years. Upper-level circulation anomalies suggest that atmospheric teleconnections originating in the western tropical Pacific are responsible for the delivery of precipitation to the Bear River watershed during the October- December (OND) season of the previous year. The Bear River flow was compared to recent reconstructions of the other tributaries to the Great Salt Lake (GSL) and the GSL level. Implications for water management could be drawn from the observation that the latter half of the 20th century was the 2nd wettest in 1200 years, and that management for future water supply should take into account the stream flow variability over the past millennium.

161. Decadal Oscillation of Autumn Precipitation in Central Vietnam Modulated by the East Pacific-North Pacific (EP-NP)

Autumn precipitation over Central Vietnam is associated with an increase in the occurrence of tropical cyclones that lead to frequent flooding and pose a significant threat to lives and property. The present analyses reveal a pronounced decadal oscillation of autumn precipitation in Central Vietnam within the 8-11 year frequency band that is modulated by the East Pacific-North Pacific (EP-NP) teleconnection. The negative phase of the EP-NP pattern is associated with a positive sea surface temperature (SST) anomaly in the South China Sea (SCS) that induces low-level convergence, enhances convection, and increases precipitation over Central Vietnam and adjacent islands including Hainan (China) and the Philippines. This circulation feature around the SCS is embedded in a large-scale circulation associated with SST anomalies across the Pacific Ocean-i.e., cooling in the Eastern and Central tropical Pacific sandwiched by warming in the North and South Pacific as well as the Western Pacific Ocean. The positive phase of the EP-NP features opposite SST and circulation anomalies, with the result being reduced rainfall in Central Vietnam. This out-of-phase relationship and shared decadal spectral coherence between the EP-NP index and autumn precipitation in Central Vietnam might be useful for future climate predictions and flood management.

Extensive manufacturing and ubiquitous use in every sector of today's society has resulted in plastics being detected in all terrestrial and aquatic environments examined to date. However, the pervasiveness of small, potentially invisible, microplastics, their associated chemical additives, and organic compounds that absorb to plastic substrates are the topics of recent investigations. These micro- to nano- size plastic particles that are deliberately manufactured or were fragmented from larger plastic products are now ending up in food webs and worldwide environmental systems. Using a pyrolysis GC-MS method, plastic polymer composition was determined in samples obtained from freshwaters in urban New Jersey. Three polymers dominated the samples: polyethylene (43%), polypropylene (33%), and polystyrene (13%). The dominant polymers differed in each river. To identify Persistent Organic Pollutants sorbed to microplastic particles, headspace solid phase micro extraction coupled with gas chromatography/ion trap mass spectrometry was employed. In the majority of upriver sampling locations, Tentatively Identified Compounds were associated with both the microplastic and the water column fractions in roughly equal proportions. However, in the tidal portion of the Passaic River and in samples from Newark and Raritan Bays, the majority of organic compounds were associated with the microplastic fraction only. Based on a search of chemical databases, the possible source/use of 180 of the 223 compounds identified, whose total mass was 1 ng or more, was determined. Forty one percent of the identified compounds were natural substances, thirty five percent were identified as laboratory/research chemicals and seven percent were pharmaceutical or biomedical compounds. Twelve identified compounds are used for industrial purposes, including a plasticizer and an insecticide. Six compounds are used as cosmetic additives. The findings of this study illustrate the diversity of organic compounds associated with the presence of microplastics in aquatic media. [ABSTRACT FROM AUTHOR]

PURPOSE: In pharmacogenetic research, genetic variation in non-responders and high responders is compared with the aim to identify the genetic loci responsible for this variation in response. However, an important question is whether the non-responders are truly biologically non-responsive or actually non-adherent? Therefore, the aim of this study was to describe, within the PROspective Study of Pravastatin in the Elderly at Risk (PROSPER), characteristics of both non-responders and high responders of statin treatment in order to possibly discriminate non-responders from non-adherers.METHODS: Baseline characteristics of non-responders to statin therapy (≤10 % LDL-C reduction) were compared with those of high responders (>40 % LDL-C reduction) through a linear regression analysis. In addition, pharmacogenetic candidate gene analysis was performed to show the effect of excluding non-responders from the analysis.RESULTS: Non-responders to statin therapy were younger (p = 0.001), more often smoked (p < 0.001), had a higher alcohol consumption (p < 0.001), had lower LDL cholesterol levels (p < 0.001), had a lower prevalence of hypertension (p < 0.001), and had lower cognitive function (p = 0.035) compared to subjects who highly responded to pravastatin treatment. Moreover, excluding non-responders from pharmacogenetic studies yielded more robust results, as standard errors decreased.CONCLUSION: Our results suggest that non-responders to statin therapy are more likely to actually be non-adherers, since they have more characteristics that are viewed as indicators of high self-perceived health and low disease awareness, possibly making the subjects less adherent to study medication. We suggest that in pharmacogenetic research, extreme non-responders should be excluded to overcome the problem that non-adherence is investigated instead of non-responsiveness.

Background: Current interventions for haemorrhoidal disease include traditional haemorrhoidectomy (TH) and\ud stapled haemorrhoidopexy (SH) surgery. However, uncertainty remains as to how they compare from a clinical,\ud quality of life (QoL) and economic perspective. The study is therefore designed to determine whether SH is more\ud effective and more cost-effective, compared with TH.\ud \ud Methods/Design: eTHoS (either Traditional Haemorrhoidectomy or Stapled Haemorrhoidopexy for Haemorrhoidal\ud Disease) is a pragmatic, multicentre, randomised controlled trial. Currently, 29 secondary care centres are open to\ud recruitment. Patients, aged 18 year or older, with circumferential haemorrhoids grade II to IV, are eligible to take part.\ud The primary clinical and economic outcomes are QoL profile (area under the curve derived from the EuroQol Group’s 5\ud Dimension Health Status Questionnaire (EQ-5D) at all assessment points) and incremental cost per quality adjusted life\ud year (QALY) based on the responses to the EQ-5D at 24 months. The secondary outcomes include a comparison of the\ud SF-36 scores, pain and symptoms sub-domains, disease recurrence, complication rates and direct and indirect costs\ud to the National Health Service (NHS). A sample size of n =338 per group has been calculated to provide 90% power\ud to detect a difference in the mean area under the curve (AUC) of 0.25 standard deviations derived from EQ-5D\ud score measurements, with a two-sided significance level of 5%. Allowing for non-response, 400 participants will\ud be randomised per group. Randomisation will utilise a minimisation algorithm that incorporates centre, grade of\ud haemorrhoidal disease, baseline EQ-5D score and gender. Blinding of participants and outcome assessors is not\ud attempted.\ud \ud Discussion: This is one of the largest trials of its kind. In the United Kingdom alone, 29,000 operations for haemorrhoidal\ud disease are done annually. The trial is therefore designed to give robust evidence on which clinicians and health\ud service managers can base management decisions and, more importantly, patients can make informed choices.\ud \ud Trial registration: Current Controlled Trials ISRCTN80061723 (assigned 8 March 2010)

- Abstract
Variability in response to drug use is common and heritable, suggesting that genome-wide pharmacogenomics studies may help explain the “missing heritability” of complex traits. Here, we describe four independent analyses in 33,781 participants of European ancestry from ten cohorts that were designed to identify genetic variants modifying the effects of drugs on QT interval duration (QT). Each analysis cross-sectionally examined four therapeutic classes: thiazide diuretics (prevalence of use=13.0%), tri/tetracyclic antidepressants (2.6%), sulfonylurea hypoglycemic agents (2.9%), and QT prolonging drugs as classified by the University of Arizona Center for Education and Research on Therapeutics (4.4%). Drug-gene interactions were estimated using covariable adjusted linear regression and results were combined with fixed-effects meta-analysis. Although drug-SNP interactions were biologically plausible and variables were well-measured, findings from the four cross-sectional meta-analyses were null (Pinteraction>5.0×10−8). Simulations suggested that additional efforts, including longitudinal modeling to increase statistical power, are likely needed to identify potentially important pharmacogenomic effects.

- Abstract
General cognitive function is substantially heritable across the human life course from adolescence to old age. We investigated the genetic contribution to variation in this important, health-and well-being-related trait in middle-aged and older adults. We conducted a meta-analysis of genome-wide association studies of 31 cohorts (N = 53 949) in which the participants had undertaken multiple, diverse cognitive tests. A general cognitive function phenotype was tested for, and created in each cohort by principal component analysis. We report 13 genome-wide significant single-nucleotide polymorphism (SNP) associations in three genomic regions, 6q16.1, 14q12 and 19q13.32 (best SNP and closest gene, respectively: rs10457441, P = 3.93 x 10(-9), MIR2113; rs17522122, P = 2.55 x 10(-8), AKAP6; rs10119, P = 5.67 x 10(-9), APOE/TOMM40). We report one gene-based significant association with the HMGN1 gene located on chromosome 21 (P = 1x10(-6)). These genes have previously been associated with neuropsychiatric phenotypes. Meta-analysis results are consistent with a polygenic model of inheritance. To estimate SNP-based heritability, the genome-wide complex trait analysis procedure was applied to two large cohorts, the Atherosclerosis Risk in Communities Study (N = 6617) and the Health and Retirement Study (N = 5976). The proportion of phenotypic variation accounted for by all genotyped common SNPs was 29% (s.e. = 5%) and 28% (s.e. = 7%), respectively. Using polygenic prediction analysis, similar to 1.2% of the variance in general cognitive function was predicted in the Generation Scotland cohort (N = 5487; P = 1.5 x 10(-17)). In hypothesis-driven tests, there was significant association between general cognitive function and four genes previously associated with Alzheimer's disease: TOMM40, APOE, ABCG1 and MEF2C.

- Abstract
The main challenge for gaining biological insights from genetic associations is identifying which genes and pathways explain the associations. Here we present DEPICT, an integrative tool that employs predicted gene functions to systematically prioritize the most likely causal genes at associated loci, highlight enriched pathways and identify tissues/cell types where genes from associated loci are highly expressed. DEPICT is not limited to genes with established functions and prioritizes relevant gene sets for many phenotypes.

- Abstract
Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, we conducted genome-wide association meta-analyses of waist and hip circumference-related traits in up to 224,459 individuals. We identified 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (WHRadjBMI) and an additional 19 loci newly associated with related waist and hip circumference measures (P<5×10-8). Twenty of the 49 WHRadjBMI loci showed significant sexual dimorphism, 19 of which displayed a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation, and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms.

Non-equilibrium radio-frequency driven atmospheric-pressure plasma in He/0.6%O
2 gas mixture has been used to study the reaction mechanism of plasma-generated oxygen atoms in aqueous solutions. The effluent from the plasma source operated with standard and18 O-labeled O2 gas was used to treat water in the presence of phenol as a chemical probe. Comparing the mass spectrometry and gas chromatography-mass spectrometry data of the solutions treated with plasma under normal and labeled oxygen provides clear evidence that O(aq) originating from the gas phase enters the liquid and reacts directly with phenol, without any intermediate reactions. Additionally, the atmospheric-pressure plasma source demonstrates great potential to be an effective source of O(aq) atoms without the requirement for any precursors in the liquid phase. New prognostic models are traditionally evaluated using measures of discrimination and risk reclassification, but these do not take full account of the clinical and health economic context. We propose a framework for comparing prognostic models by quantifying the public health impact (net benefit) of the treatment decisions they support, assuming a set of predetermined clinical treatment guidelines. The change in net benefit is more clinically interpretable than changes in traditional measures and can be used in full health economic evaluations of prognostic models used for screening and allocating risk reduction interventions. We extend previous work in this area by quantifying net benefits in life years, thus linking prognostic performance to health economic measures; by taking full account of the occurrence of events over time; and by considering estimation and cross-validation in a multiple-study setting. The method is illustrated in the context of cardiovascular disease risk prediction using an individual participant data meta-analysis. We estimate the number of cardiovascular-disease-free life years gained when statin treatment is allocated based on a risk prediction model with five established risk factors instead of a model with just age, gender and region. We explore methodological issues associated with the multistudy design and show that cost-effectiveness comparisons based on the proposed methodology are robust against a range of modelling assumptions, including adjusting for competing risks. Aims Atrial fibrillation/flutter (AF) is the most common arrhythmia in older people. It associates with reduced exercise capacity, increased risk of stroke, and mortality. We aimed to determine retrospectively whether pravastatin reduces the incidence of AF and whether any electrocardiographic measures or clinical conditions might be risk factors for its development. Methods and results The PROspective Study of Pravastatin in the Elderly at Risk (PROSPER) was a randomized, double-blind controlled trial that recruited 5804 individuals aged 70-82 years with a history of, or risk factors for, vascular disease. A total of 2891 were allocated to pravastatin and 2913 to placebo; mean follow-up was 3.2 years. Electrocardiograms (ECGs), which were recorded at baseline, annually thereafter, and at run-out, were processed by computer and reviewed manually. In all, 264 of 2912 (9.1%) of the placebo group and 283 of 2888 (9.8%) of the pravastatin-treated group developed AF [hazard ratio 1.08 (0.92,1.28), P = 0.35)]. Multivariate analysis showed that PR and QTc intervals, age, left ventricular hypertrophy, and ST-T abnormalities were related to development of AF after adjustment for many variables including alcohol consumption, which itself was univariately predictive of developing AF. Previous myocardial infarction on the ECG was not a risk factor. A history of vascular disease was strongly linked with developing AF but not diabetes and hypertension. Conclusion Pravastatin does not reduce the incidence of AF in older people at risk of vascular disease, at least in the short-medium term. Risk factors for AF include older age, prolongation of PR or QTc intervals, left ventricular hypertrophy, and ST-T abnormalities on the ECG.

- Abstract
Zearalenone (ZEA), a fungal mycotoxin, and its metabolite zeranol (ZAL) are known estrogen agonists in mammals, and are found as contaminants in food. Zeranol, which is more potent than ZEA and comparable in potency to estradiol, is also added as a growth additive in beef in the US and Canada. This article presents the development and application of a Physiologically-Based Toxicokinetic (PBTK) model for ZEA and ZAL and their primary metabolites, zearalenol, zearalanone, and their conjugated glucuronides, for rats and for human subjects. The PBTK modeling study explicitly simulates critical metabolic pathways in the gastrointestinal and hepatic systems. Metabolic events such as dehydrogenation and glucuronidation of the chemicals, which have direct effects on the accumulation and elimination of the toxic compounds, have been quantified. The PBTK model considers urinary and fecal excretion and biliary recirculation and compares the predicted biomarkers of blood, urinary and fecal concentrations with published in vivo measurements in rats and human subjects. Additionally, the toxicokinetic model has been coupled with a novel probabilistic dietary exposure model and applied to the Jersey Girl Study (JGS), which involved measurement of mycoestrogens as urinary biomarkers, in a cohort of young girls in New Jersey, USA. A probabilistic exposure characterization for the study population has been conducted and the predicted urinary concentrations have been compared to measurements considering inter-individual physiological and dietary variability. The in vivo measurements from the JGS fall within the high and low predicted distributions of biomarker values corresponding to dietary exposure estimates calculated by the probabilistic modeling system. - Abstract
Using genome-wide data from 253,288 individuals, we identified 697 variants at genome-wide significance that together explained one-fifth of the heritability for adult height. By testing different numbers of variants in independent studies, we show that the most strongly associated 1/42,000, 1/43,700 and 1/49,500 SNPs explained 1/421%, 1/424% and 1/429% of phenotypic variance. Furthermore, all common variants together captured 60% of heritability. The 697 variants clustered in 423 loci were enriched for genes, pathways and tissue types known to be involved in growth and together implicated genes and pathways not highlighted in earlier efforts, such as signaling by fibroblast growth factors, WNT/I 2-catenin and chondroitin sulfate-related genes. We identified several genes and pathways not previously connected with human skeletal growth, including mTOR, osteoglycin and binding of hyaluronic acid. Our results indicate a genetic architecture for human height that is characterized by a very large but finite number (thousands) of causal variants.

The work begins by reviewing publications on the use of optical-disc technology in cataloging departments. This section includes descriptive information on specific products and comparative considerations on the value of the CD genre. Most commentary to date seems to find cost advantages to the optical format for smaller libraries but fewer attractions for larger institutions who would lose online immediacy. Metabolomic data come in many forms depending on what was collected. In a study looking for biomarkers of exposure to H2S, whole blood was collected from participants and subsequently analyzed by GC/ITMS. The data from chromatograms were sorted using principal component analysis. A key portion of the chromatograph was used to demonstrate differences between no exposure, a low and high exposure. True difference mass spectra were used to differentiate urine samples from mice before and after they ocnsumed green tea. MS data from control subjects looking at MTBE exposure sensitivity, showed a time course difference from a single subject, following exposure. All of this data came from mass spectral data points but the pattern may not be appropriate for storage in a database that should be searchable. The patterns created are the most effective way to verify biological differences in conditions and identify potential biomarkers, but is not useful for comparing results from one study to another. Data in the raw phase (mass spectral data) cannot be compared to other raw data (NMR) and cannot be linked directly to metabolic pathways.

Parkinson’s disease (PD) is a neurodegenerative disease that is difficult to diagnose because there are no specific diagnostic biomarkers. It cannot be diagnosed solely on pathology. A reliable animal model may assist in identification of novel biomarkers. Paraquat (PQ) has been identified both epidemiologically and experimentally as a possible risk factor for PD in humans and mice. It has been demonstrated to preferentially kill dopaminergic neurons in the substantia nigra in mice and rats and its effects have also been found to be persistent with an apparent half-life of ~ 28 days. Its neurotoxic affects have been increased with concomitant doses of MPTP or other pesticides. One of the keys to determine it’s efficacy as a model for PD is to measure its concentration and disposition effectively in the brain tissue of the animal.

This proof of concept study was undertaken to test methodologies to characterize potential environmental risk associated with the presence of microplastics in surface waters. The goals of the study were to determine whether urban New Jersey freshwaters contained microplastic pollutants, and if so, to test analytic techniques that could potentially identify chemical compounds associated with this pollution. A third objective was to test whether identified associated compounds might have physiological effects on an aquatic organism. Using field collected microplastic samples obtained from the heavily urbanized Raritan and Passaic Rivers in New Jersey, microplastic densities, types, and sizes at 15 sampling locations were determined. Three types of plastic polymers were identified using pyrolysis coupled with gas chromatography (Pyr-GC/MS). Samples were further characterized using solid phase micro extraction coupled with headspace gas chromatography/ion trap mass spectrometry (HS-SPME-GC/ITMS) to identify organic compounds associated with the: (i) solid microplastic fraction, and (ii) site water fraction. Identical retention times for GC peaks found in both fractions indicated compounds can move between the two phases, potentially available for uptake by aquatic biota in the dissolved phase. Patterns of tentatively identified compounds were similar to patterns obtained in Pyr-GC/MS. Embryonic zebrafish exposed to PyCG/MS- identified pure polymers in the 1-10 ppm range exhibited altered growth and heart defects. Using two analytic methods (SPME GC/MS and Pyr-GC/MS) allows unambiguous identification of compounds associated with microplastic debris and characterization of the major plastic type(s). Specific 'fingerprint' patterns can categorize the class of plastics present in a waterbody and identify compounds associated with the particles. Superstorm Sandy provided an opportunity to study filamentous fungi (molds) associated with winter storm damage. We collected 36 morphologically distinct fungal isolates from flooded buildings. By combining traditional morphological and cultural characters with an analysis of ITS sequences (the fungal DNA barcode), we identified 24 fungal species that belong to eight genera: Penicillium (11 species), Fusarium (four species), Aspergillus (three species), Trichoderma (two species), and one species each of Metarhizium, Mucor, Pestalotiopsis, and Umbelopsis. Then, we used a Drosophila larval assay to assess possible toxicity of volatile organic compounds (VOCs) emitted by these molds. When cultured in a shared atmosphere with growing cultures of molds isolated after Hurricane Sandy, larval toxicity ranged from 15 to 80%. VOCs from Aspergillus niger 129B were the most toxic yielding 80% mortality to Drosophila after 12 days. The VOCs from Trichoderma longibrachiatum 117, Mucor racemosus 138a, and Metarhizium anisopliae 124 were relatively non-toxigenic. A preliminary analysis of VOCs was conducted using solid-phase microextraction-gas chromatography-mass spectrometry from two of the most toxic, two of the least toxic, and two species of intermediate toxicity. The more toxic molds produced higher concentrations of 1-octen-3-ol, 3-octanone, 3-octanol, 2-octen-1-ol, and 2-nonanone; while the less toxic molds produced more 3-methyl-1-butanol and 2-methyl-1-propanol, or an overall lower amount of volatiles. Our data support the hypothesis that at certain concentrations, some VOCs emitted by indoor molds are toxigenic. [ABSTRACT FROM AUTHOR]
