The photoreactivity of (3-methyl-2H-azirin-2-yl)-phenylmethanone, 1, is wavelength-dependent (Singh et al. J. Am. Chem. Soc. 1972, 94, 1199-1206). Irradiation at short wavelengths yields 2P, whereas longer wavelengths produce 3P. Laser flash photolysis of 1 in acetonitrile using a 355 nm laser forms its triplet ketone (T(1K), broad absorption with λ(max) ~ 390-410 nm, τ ~ 90 ns), which cleaves and yields triplet vinylnitrene 3 (broad absorption with λ(max) ~ 380-400 nm, τ = 2 μs). Calculations (B3LYP/6-31+G(d)) reveal that T(1K) of 1 is located 67 kcal/mol above its ground state (S(0)) and has a long C-N bond (1.58 Å), and the calculated transition state to form 3 is only 1 kcal/mol higher in energy than T(1K) of 1. The calculations show that 3 has significant 1,3-carbon iminyl biradical character, which explains why 3 reacts efficiently with oxygen and decays by intersystem crossing to the singlet surface. Photolysis of 1 in argon matrixes at 14 K produced ketene imine 7, which presumably is formed from 3 intersystem crossing to 7. In comparison, photolysis of 1 in methanol with a 266 nm laser produces mainly ylide 2 (λ(max) ~ 380 nm, τ ~ 6 μs, acetonitrile), which decays to form 2P. Ylide 2 is formed via singlet reactivity of 1, and calculations show that the first singlet excited state of the azirine chromophore (S(1A)) is located 113 kcal/mol above its S(0) and that the singlet excited state of the ketone (S(1K)) is 85 kcal/mol. Furthermore, the transition state for cleaving the C-C bond in 1 to form 2 is located 49 kcal/mol above the S(0) of 1. Thus, we theorize that internal conversion of S(1A) to a vibrationally hot S(0) of 1 forms 2, whereas intersystem crossing from S(1K) to T(1K) results in 3.

The effect of binding Tb(3+) to sodium taurocholate aggregates containing polyaromatic hydrocarbon guests was examined using pyrene and 1-ethylnaphthalene as guests that bind to the primary aggregate, and 1-naphthyl-1-ethanol as a secondary aggregate guest. Time-resolved fluorescence quenching studies were used to study the binding site properties, while laser flash photolysis quenching studies provided information on the dynamics of the guest-aggregate system. Both the primary and secondary aggregate binding sites became more compact in the presence of bound Tb(3+), while only the primary aggregate became more accessible to anionic molecules. The binding dynamics for the guest-primary aggregate system became faster when Tb(3+) was bound to the aggregate. In contrast, for the guest-secondary aggregate the presence of Tb(3+) resulted in a small decrease in the dissociation rate constant. The influence of bound Tb(3+) on the primary and secondary bile salt aggregates is significantly different, which affects how these aggregates can be used as supramolecular host systems to modify guest reactivity., (This journal is © The Royal Society of Chemistry and Owner Societies 2011)

Sodium cholate aggregates are adaptable host systems. The effect of changing the ionic strength with the addition of NaCl on the properties for guest binding to sodium cholate aggregates was investigated by using pyrene, perylene and 1-ethylnaphthalene as guests. Fluorescence, anisotropy and laser flash photolysis studies provided information on the protection efficiency of the aggregate bound guest, and provided information on the dynamics and correlation times for the host-guest system. Different trends for the protection efficiency of the bound guests were observed when the NaCl concentration was raised depending on the charge of the aqueous solubilized quencher. The increase in ionic strength was also shown to lengthen the correlation time of the aggregate bound guest and led to faster dynamics for the host-guest complex. These results show that the properties of sodium cholate aggregates as a supramolecular host system are significantly altered with changes in the ionic strength of the medium.

The photochromic properties of dihydropyrenes have been substantially improved by making use of density functional theory (DFT) activation barrier calculations, which suggested that the di-isobutenylcyclophanediene 15' should have a significant barrier to thermal isomerization to the dihydropyrene (DHP) 15, which itself should resist isomerization involving migration of the internal groups to the rearranged dihydropyrene 9 (X = -CH═C(Me)(2)). As a result of these calculations, the synthesis of the colorless cyclophanediene (CPD) 15' was undertaken and achieved from the dinitrile 28 in four steps in 37% overall yield %. The cyclophanediene 15' thermally isomerized to the dihydropyrene 15 at 100 °C with t(1/2) = 4.5 h, giving an extrapolated 20 °C t(1/2) of ∼16 y, consistent with the DFT calculations. No evidence for [1,5]-sigmatropic rearrangement in to 9 (X = -CH═C(Me)(2)) was observed on heating to 130 °C. The ring-opening isomerization quantum yields (ϕ(open)) for DHP 15 in to CPD 15' were determined in cyclohexane to be 0.12 ± 0.01, which is three times greater than for the benzoDHP 1. Friedel-Crafts naphthoylation of 15 gave 70% of purple 32, which in toluene showed the largest photochemical ring-opening isomerization quantum yields (ϕ(open)) of 0.66 ± 0.02 for any known dihydropyrene, ∼nine times greater than 1 in toluene. The thermal closing of 32' to 32, although faster than for 15', gave a useful extrapolated t(1/2) of ∼2 y at 20 °C.

Photocyclodimerization of 2-anthracenecarboxylate mediated by molecular chaperone protein was performed for the first time to afford chiral syn-head-to-tail and anti-head-to-head dimers (2 and 3) in 10% and 16% enantiomeric excess, respectively, with enhanced yields of sterically and electrostatically less-favored head-to-head dimers (3 and 4).

Bile salts form aggregates in aqueous solutions which were shown to efficiently solubilize aqueous insoluble photochromic compounds. Photochromic switching was observed for a spiropyran and a diarylethene. In the case of the spiropyran, the incorporation into the bile salt aggregate led to improved hydrolytic stability and changes in thermal rate constants.

Photolysis of 1a yields 4a in argon-saturated methanol, whereas 1b is photostable. Laser flash photolysis of 1a in acetonitrile shows formation of biradical 2a (lambda(max) = 340 nm, tau = approximately 60 ns), which undergoes intersystem crossing to form Z-3a (lambda(max) = 380 nm, tau = 270 ns) and E-3a (lambda(max) = 380 nm, tau = 300 ms). Z-3a regenerates the starting material, whereas E-3b undergoes intramolecular lactonization to release the alcohol moiety and form 4a. Similar laser flash photolysis of 1b shows formation of biradical 2b (lambda(max) = 340 nm, tau = 1.9 micros in acetonitrile), which is longer-lived than 2a is. However, 2b only undergoes intersystem crossing to form Z-3b (lambda(max) = 380 nm, tau = 4.3 micros). Calculations demonstrate that intramolecular pseudo hydrogen bonding between the OH moiety and the radical centered on the isopropyl carbon in 2b and the bulkiness of the isopropyl group prevent the necessary rotation to form E-3b. In comparison, 2a does not form an intramolecular pseudo hydrogen bond between the methylene radical center and the OH group, and as a consequence, it undergoes intersystem crossing to form both E- and Z-3a.

Pheromone-binding proteins (PBPs) in insects can bind various substances and selectively deliver the message of a signal molecule to the downstream components of the olfactory system. This can be achieved either through a ligand-specific conformational change of the C-terminal peptide of the PBP or by selectively binding/releasing the ligand. PBP may also act as a scavenger to protect the sensory neurons from saturating at high ligand doses. We have compared two PBPs from the gypsy moth (PBP1 and PBP2) and their truncated forms (TPBPs), which lack the C-terminal peptide, in this study. Stopped-flow kinetics with N-phenyl-1-naphthylamine (NPN) have revealed a diffusion-controlled collisional step, between PBP and NPN, after which the NPN relocates into a hydrophobic environment. This work supports the hypothesis that binding between PBPs and ligands occurs stepwise. With the method of tryptophan fluorescence quenching, we have shown different local conformational changes around Trp 37, induced by different ligands, manifested in changes of both the steric and electronic environment around the residue. Importantly, we have noticed a significant difference in the changes induced by the biological ligand (the pheromone) and nonbiological ligands. Therefore, we hypothesize that PBP may serve a different function in each kinetic step, displaying a unique P.L conformation.

The binding of naphthalene (Np), 1-ethylnaphthalene (EtNp), acenaphthene (AcN), and 1-naphthyl-1-ethanol (NpOH) as guests to the aggregates of sodium cholate (NaCh), taurocholate (NaTC), deoxycholate (NaDC), and deoxytaurocholate (NaTDC) was studied with the objective of determining how the structure of the bile salts affects the binding dynamics of guests and quenchers with the bile salt aggregates. Time-resolved and steady-state fluorescence experiments were used to determine the binding efficiency of the guests with the aggregates and were also employed to investigate the quenching of the singlet excited state of the guests by iodide anions. Quenching studies of the triplet excited states using laser flash photolysis were employed to determine the accessibility to the aggregate of nitrite anions, used as quenchers, and the dissociation rate constants of the guests from the bile salt aggregates. The binding efficiency of the guests to NaDC and NaTDC is higher than for NaCh and NaTC, and the protection efficiency is also higher for NaDC and NaTDC, in line with the larger aggregates formed for the latter bile salts. The formation of aggregates is in part driven by the structure of the guest, where an increased protection efficiency and residence time can be achieved by the introduction of short alkyl substituents (AcN or EtNp vs Np). NpOH was shown to be located in a very different environment in all four bile salts when compared to AcN, EtNp, and Np, suggesting that hydrogen bonding plays an important role in the formation of the aggregate around NpOH.

S100A7 (psoriasin) is a member of the S100 family of signaling proteins. It is implicated in and considered a therapeutic target for inflammation and cancer, yet no small molecule ligands for S100A7 have been identified. To begin the development of specific small molecule inhibitors of S100A7 function, we have used a series of surface binding fluorescent dyes to probe the surface hydrophobic sites. Two naphthalene-based dyes (2,6-ANS and 1,8-ANS) were found to bind S100A7 in a distinct cleft. We characterized the binding interaction by determining both the structure of S100A7 bound to 2,6-ANS and the structure of S100A7 bound to 1,8-ANS to 1.6 A. In both cases, two molecules of dye were docked such that the naphthalene groups were positioned in two symmetry-related grooves that are formed by the N-terminal helices of each monomer. We observed that Met12 acts as a gatekeeper to the binding cleft, adopting an "open" conformation for the more elongated 2,6-ANS while remaining in a "closed" conformation for the more compact 1,8-ANS. Steady-state fluorescence experiments revealed that S100A7 binds two copies of 2,6-ANS, each with a K(d) of 125 muM. Time-resolved fluorescence lifetime measurements indicated that the two molecules of 2,6-ANS bind in two independent binding sites with different fluorescence lifetimes, suggesting that the S100A7 homodimer is not perfectly symmetric in solution. Isothermal titration calorimetry studies demonstrate that S100A7 has a higher affinity for 2,6-ANS than 1,8-ANS. Yeast two-hybrid studies were also used to probe contributions of individual residues of an S100A7 triple mutant with respect to Jab1 binding. Mutation of Leu78, which forms part of the Met12 cleft occupied by 2,6-ANS, reduced the level of Jab1 binding, suggesting a potentially important role for the Met12 hydrophobic pocket in defining a Jab1 interface. Additional Y2H studies also delineate contributions of Gln88 and in particular Asp56 that shows the most significant abrogated binding to Jab1. Collectively, these data suggest a complex interaction between S100A7 and the much larger Jab1. These studies form the basis for the development of small molecule reporters and modifiers of S100A7 form and function.

The photochirogenesis of 2-anthracenecarboxylic acid (AC) complexed to a hydrogen-bonding template (TKS159) was investigated to obtain mechanistic information on how chirogenesis is achieved for the dimerization of AC. Complexation of AC to TKS159 leads to the shielding of one of the two surfaces of the prochiral AC molecule. The two diastereomeric AC-TKS complexes, i.e., re-AC-TKS and si-AC-TKS, were characterized by changes in the UV-vis, fluorescence, and circular dichroism spectra and excited-state lifetimes. The ee is not simply determined by the diastereomeric ratio of the re- and si-AC-TKS complexes but also depends on the relative lifetimes of the diastereomeric complexes. The relative population of the re and si complexes was calculated from the enantiomeric excess (ee) for the products, taking into account the relative lifetimes of the two complexes. These studies established a protocol that can be used to reveal the mechanism for photochirogenesis by investigating the ground state and the excited state behavior of supramolecular systems.

Understanding tissue distribution and clearance of nanomaterials following different routes of exposure is needed for risk assessment. F344 female rats received single or multiple exposures to 20 nm, 100 nm or 1000 nm latex fluorospheres by intravenous (i.v.) injection or oral pharyngeal aspiration into the airways. The presence of fluorospheres in tissues was assessed up to 90-120 days after the final dose. Blood, perfusion fluid, bone marrow, brain, eyes, feces, gut, heart, kidney, liver, lung, muscle, skin, spleen, thymus, tongue, urine and uterus plus ovaries were collected for analysis. Liver, spleen and lung were the greatest tissue depots for all particles following i.v. injection. The proportion of 100 nm and 1000 nm but not 20 nm spheres significantly increased in the spleen over time. Lung was the greatest tissue depot for all particles following single or repeat airway exposure. Greater than 95% of 1000 nm spheres that were recovered were in the lung in contrast to 70-80% of 20 nm spheres or 89-95% of 100 nm spheres. All 3 sizes were found in gut or gut+feces 1-7 days after lung exposure. The thymus was the largest extra-pulmonary depot for the particles; up to 25% of recovered 20 nm particles were in the thymus up to 4 months after exposure compared to 6% of 100 nm particles and 1-3% of 1000 nm particles. A small proportion of 20 nm particles were detected in kidney following both acute and repeat airway exposure. Low numbers of particles were found in the circulation (blood, perfusion), bone marrow, brain, heart, liver and spleen but not in eye, muscle, skin, tongue, ovaries, uterus or urine. These data show that the tissue targets of nano- and micron-sized spheres are very similar whether exposure occurs systemically or via the airways while the proportion of particles in some tissues and tissue clearance varies based on particle size.

The mechanism for the chirogenesis in the photocyclodimerization of 2-anthracenecarboxylate (AC) bound to human serum albumin (HSA) was investigated using time-resolved fluorescence measurements in the presence of HSA inhibitors and/or an AC singlet excited state quencher. The photophysical studies were correlated with product studies to explain the high enantiomeric excess (ee) observed for the chiral photoproducts. AC binds to HSA in five different binding sites with decreasing affinities. AC bound to the sites with the highest affinity (sites 1 and 2) is unreactive, and the AC can be displaced from these sites by the use of known inhibitors. Time-resolved fluorescence studies isolated a singlet excited state AC bound to a site which exhibited moderate protection from interactions with species in the aqueous phase. This site was assigned to binding site 3, where the chiral photoproducts are formed with a high ee based on the correlation of the photophysical studies with product studies in the presence of a quencher. These results show that the use of inhibitors for multiple binding site proteins is useful to uncover the properties of binding sites for which guest binding has only moderate affinity and where the photophysical characterization of these binding sites is not possible in the absence of inhibitors.

Photolysis of gamma-azidobutyrophenone derivatives yields 1,4 ketyl biradicals via intramolecular H-atom abstraction. The 1,4 ketyl biradicals expel a nitrogen molecule to form 1,5 ketyl iminyl biradicals, which decay by ring closure to form a new carbon-nitrogen bond. The 1,5 ketyl iminyl biradicals were characterized with transient spectroscopy. In argon/nitrogen-saturated solutions, the biradicals have lambda(max) approximately 300 nm and tau = 15 micros. DFT-TD calculations were used to support the proposed mechanism for formation of the 1,5 ketyl iminyl radicals.

The pheromone-binding proteins (PBPs), which exist at a high concentration in the sensillum lymph surrounding olfactory neurons, are proposed to be important in pheromone detection and discrimination in insects. Here, we present a systematic study of PBP-ligand interaction kinetics. We find that PBP2, from the gypsy moth, Lymantria dispar, associates and dissociates slowly with its biofunctional ligands, (+)- and (-)-disparlure. Tryptophan anisotropy measurements detect PBP multimers in solution as well as an increase in the multimeric state of the protein upon long exposure to ligand. We propose a kinetic model that includes monomer/multimer equilibria and a two-step binding process: (1) external binding of the pheromone assisted by the C terminus of PBP2, and (2) slow embedding of the pheromone into the internal pocket. This experimentally derived model sheds light on the potential biological function and mechanism of PBPs as ligand scavengers.

The binding dynamics of the guests acenaphthene, phenanthrene, fluorene, and acenaphthenol with sodium cholate aggregates were studied using laser flash photolysis and fluorescence. The location of the guests in the bile salt aggregate is determined by the guest's hydrophobicity, where acenaphthene, phenanthrene, and fluorene bind to the primary aggregates, while acenaphthenol binds to the secondary bile salt aggregates. The residence time of the guests in the primary aggregates and the access of ionic species from the aqueous phase to the guest in the aggregate depend on the size and the shape of the guest. These results show that bile salt aggregates are adaptable supramolecular host systems.

A series of dihydropyrenes with appending fluorescent moieties were synthesized with the objective of increasing the photochromic efficiency for this class of compounds and to establish how suitable fluorescence would be to follow their photochromic behavior. The ring opening quantum yields of dihydropyrenes with aroyl substituents at the 4-position showed increased ring opening quantum yields without a decrease of the half-life for the thermal reversion of the less stable open isomer, the metacyclophanediene to the dihydropyrene. The fluorescence of the appending naphthoyl or pyrenoyl moieties was not suitable to follow the photochromic cycling of the dihydropyrenes. However, the emission detected above 600 nm of the closed isomer of the dihydropyrene moiety was shown to be a good monitoring method for the photochromic cycling.

The formation of micelles of an amphiphilic dendro-calixarene (1) was studied using pyrene as a guest molecule. Steady-state fluorescence experiments were performed, which showed that pyrene is readily solubilized in the micelles and senses an environment with a moderate polarity. Time-resolved fluorescence measurements showed that pyrene in the micelles have two lifetimes, suggesting some compartmentalization of the guest within the micelles. Pyrene bound to the micelles is completely protected from the interaction with anionic species in the aqueous phase, due to the repulsion between the anions and the negatively charged micelles. Compared to conventional micelles, such as those formed with sodium dodecyl sulfate, micelles of 1 form at much lower concentrations of monomer.

Bile salts form supramolecular aggregates with two binding sites with different properties. The guest binding dynamics to the aggregates and guest protection from species in the aqueous phase were investigated using fluorescence and laser flash photolysis experiments. Sodium cholate, deoxycholate and taurodeoxycholate were used as bile salts and acetonitrile or ethylene glycol were added as co-solvents to water in order to alter the binding properties of 1-ethylnaphthalene and 1-naphthyl-1-ethanol with the aggregates. The binding dynamics are faster and protection efficiencies are lower for guests bound to cholate and in the presence of either co-solvent.

Photophysics and photochemistry of 2-anthracenecarboxylate (AC) bound to bovine serum albumin (BSA) were investigated in detail for the first time by electronic absorption, circular dichroism (CD), steady-state and time-resolved fluorescence, fluorescence quenching, and product analysis studies. Through the spectroscopic investigations, it was revealed that the four independent binding pockets of BSA, which are known to accommodate 1, 3, 2, and 3 AC molecules in the order of decreasing affinity, are distinctly different in hydrophobicity, chiral environment, and accessibility. Interestingly, AC bound to site 1 gave highly structured fluorescence with dual lifetimes of 4.8 and 2.1 ns in an intensity ratio of 3:2, which may be assigned to the existence of two positional or orientational isomers within the very hydrophobic site 1. In contrast, the lifetime of AC in site 2 was much longer (13.3 ns), and ACs in sites 3 and 4 have broader fluorescence spectra with lifetimes that were practically indistinguishable from that in bulk water (15.8 ns). Although each of sites 2-4 simultaneously binds multiple AC molecules, no CD exciton coupling or static fluorescence quenching was detected, indicating that ACs bound to each site are not in close proximity to each other. Quenching studies with nitromethane further confirmed the significant difference in accessibility among the binding sites; thus, ACs bound to sites 1 and 2 are highly protected from the attack of the quencher, affording 32 and 10 times smaller rate constants than that for free AC in water. Product studies in the presence and absence of nitromethane more clearly revealed the photochirogenic performance of each binding site. Although the addition of nitromethane did not greatly alter the product distribution, the enantiomeric excesses (ee's) of chiral cycloadducts 2 and 3 were critically manipulated by selectively retarding the photoreaction occurring at the more accessible binding sites. Thus, the highest ee of 38% was obtained for 2 in the presence of 18 mM nitromethane, while the highest ee of 58% was attained for 3 in the absence of nitromethane, both at [AC]/[BSA]=3.6.

The time-resolved emission properties of selected anthracene derivatives, namely anthracene (1a), 2-methylanthracene (1b), 2-chloroanthracene (1c), 2-methoxyanthracene (1d), 2-methoxy-6-methylanthracene (1e), 2-(N,N'-dicyclohexylureidocarbonyl)-6-methoxyanthrace ne (1f), 2-(6-methoxyanthr-2-yl)-4,4-dimethyl-2-oxazoline (1g), 2-(6-methoxyanthr-2-yl)-pyridine (1h), and N-cyclohexylanthracene-2-carboxamide (1i) were investigated. In contrast to anthracene (1a), 1b, and 1c, the 2-methoxy-substituted anthracene derivatives 1d-1h exhibit two emission lifetimes. The determination of the lifetimes at different emission wavelengths and additional time-resolved emission spectroscopy (TRES) reveal that the dual emission originates from two different, interconvertible emissive species, with the s-cis and s-trans conformation relative to the exocyclic C2-O bond. The energy difference between the two emissive species is very small (<0.1 eV) both in the ground and in the excited state. The larger energy difference between the conformers in the excited-state is responsible for the interconversion within the singlet excited-state lifetimes of the s-cis into the s-trans conformations leading to coupled decay kinetics. The proposed mechanism for the dual emission was qualitatively supported by theoretical studies on CASSCF and DFT level. In addition, the emission lifetimes of the fluoride- and pH-sensitive fluorescent probes 1f and 1g change upon addition of fluoride or acid, respectively, so that in these cases the detection of the fluorescence lifetime may be used complementary to the steady-state fluorimetric detection of these analytes.

Dynamics is an essential feature of supramolecular systems, and it's understanding will be central in achieving new chemical function. The methodology to obtain association and dissociation rate constants for fast binding of guests to host systems in real time is described. Examples are provided for binding of guests to cyclodextrins or bile salt aggregates with an emphasis on the type of information and mechanistic insight that can only be uncovered from kinetic studies and is not apparent in thermodynamic investigations.

Bile salt aggregates are supramolecular structures with two types of binding sites, called primary and secondary sites. The objective of this work was to explore how the nonplanarity and size of guests (biphenyl [BP], 1-1'-binaphthyl [BNP] and dibenz[b,f]oxepin [DBX]) affected their binding affinity and dynamics to sodium cholate (NaC) aggregates. Fluorescence and laser-flash photolysis experiments were performed to obtain information on the binding environment for the guests, the accessibility of quenchers to guests in the aggregate and the dissociation rate constants of the guests from the aggregates. All guests were bound to the more hydrophobic primary aggregate, showing that this site can accommodate nonplanar molecules. However, the structure of the guest affects the structure of the primary aggregates, leading to changes in the accessibility of anions to aggregate-bound guests and to changes for the guest dissociation rate constants from the aggregates.

A series of Ru(II) complexes, tcc-RuX2(POC4Pyr-P,O)2 (X = Cl (3), Br (4), I (5)), containing the hemilabile phosphine pyrenyl ether ligand 4-{2-(diphenylphosphino)phenoxy}butylpyrene (POC4Pyr (1)) are reported. The synthesis and spectroscopic properties of both the ligand, POC4pyr (1), and ligand oxide, P(=O)OC4pyr 2, and the solid-state structure of 1 are reported. Complexes 3-5 react rapidly with CO to give complexes ttt-RuX2(CO)2(POC4pyr-P)2 (X = Cl (6), Br (7), I (8)). No pyrene excimer emission is detected from 3-5; however, different intensities of excimer emission are observed for 6-8. The intensity of excimer emission decreases through the series, with 6 showing the most intense response. The emission is solely due to intramolecular pyrene excimers at low concentrations (< or =10(-4) M). Comparison of the UV-vis and steady-state fluorescence spectra shows overlap between the low energy d-d absorption of 7 and 8 with excimer emission (480 nm), suggesting nonradiative energy transfer may be occurring. Once excess CO is removed, complexes 6-8 isomerize to cis-dicarbonyl complexes cct-RuX2(CO)2(POC4Pyr-P)2 (X = Cl (9), Br (10), I (11)). The intensity of excimer emission from 9-11 increases with respect to the excimer emission observed for 6-8, with 9 showing a significant increase in excimer intensity.

The phenomenon of beta-phenyl quenching has been examined by laser-flash photolysis in a series of alpha- and/or beta-substituted ketones 4-8 with similar excited-state characteristics. It is found that alpha-substitution markedly increases the triplet lifetimes in contrast to beta-substitution. The force field calculations for the various staggered conformers of ketones 4-6 and 8-syn show that the lowest-energy conformation in all these ketones has the carbonyl group and the beta-phenyl ring gauche to each other. Despite this geometrical requirement, the longer lifetimes observed are interpreted as being due to the influence of the alpha-substituent on the rotational freedom of the planar benzoyl moiety as a whole. The experimental results are suggestive of the attainment of what appears to be a critical geometry for quenching. This scenario may be likened to Norrish type II reactions, where the alpha-substituent has long been known to suppress the elimination pathway and promote Yang cyclization. In addition, we have shown that the diastereomers of alpha,beta-disubstituted ketones exhibit distinct lifetimes.

[structure: see text] The synthesis of the multistate photochromic switch 3 is described. This switch contains three dihydropyrene (DHP) units in the most conjugated fully closed form 3-c,c,c. The thermally stable form has the central DHP unit open and is 3-c,o,c. NMR and laser flash photolysis experiments were used to characterize the multiple states in the photoswitching of 3 by visible and UV light. Three of the possible five isomeric states of 3-c,o,c were observed. Irradiation of 3-c,o,c by visible light led to the formation of 3-o,o,o via the isomer 3-c,o,o as an intermediate, which were observed by NMR. Irradiation by UV light led to the formation of 3-c,c,c, which decays with a lifetime of 7.5 ms.

Xanthones with amino substituents were synthesized to diminish the photoreactivity of the xanthone chromophore with DNA, with the objective of using these molecules to study their binding dynamics with DNA. The aminoxanthones showed a strong solvatochromic effect on their singlet and triplet excited-state photophysics, where polar solvents led to a decrease of the energies for the excited states. Quenching of the triplet excited states by nitrite anions was used to determine the binding dynamics, and a residence time in the microsecond time domain was estimated for the bound 2-aminoxanthone with DNA. The quenching experiments performed showed that this methodology will not be applicable to study the binding dynamics of a wide variety of guests with DNA.

N,N'-Dicyclohexyl-N-(6'-methoxyanthryl-2-carbonyl)urea (1a) exhibits a strong solvatochromism with respect to its fluorescence properties. The emission maxima of 1a correlate well with the anion-stabilizing properties of the solvent. This feature allows the detection of analytes with high acceptor numbers, such as alcohols and carboxylic acids, and the detection of analytes such as fluoride ions which form strong hydrogen bonds with the amido hydrogen atom.

We report the preparation of complexes in which ruthenium(II) bis(bipyridyl) groups are coordinated to oligothiophenes via a diphenylphosphine linker and a thienyl sulfur (P,S bonding) to give [Ru(bpy)(2)PT(3)-P,S](PF(6))(2) (bpy = 2,2'-bipyridyl, PT(3) = 3'-(diphenylphosphino)-2,2':5',2' '-terthiophene), [Ru(bpy)(2)PMeT(3)-P,S](PF(6))(2) (PMeT(3) = 3'-(diphenylphosphino)-5-methyl-2,2':5',2' '-terthiophene), [Ru(bpy)(2)PMe(2)T(3)-P,S](PF(6))(2) (PMe(2)T(3) = 5,5' '-dimethyl-3'-(diphenylphosphino)-2,2':5',2' '-terthiophene), and [Ru(bpy)(2)PDo(2)T(5)-P,S](PF(6))(2) (PDo(2)T(5) = 3,3' '' '-didodecyl-3' '-diphenylphosphino-2,2':5',2' ':5' ',2' '':5' '',2' '' '-pentathiophene). These complexes react with base, resulting in the complexes [Ru(bpy)(2)PT(3)-P,C]PF(6), [Ru(bpy)(2)PMeT(3)-P,C]PF(6), [Ru(bpy)(2)PMe(2)T(3)-P,C]PF(6), and [Ru(bpy)(2)PDo(2)T(5)-P,C]PF(6), where the thienyl carbon is bonded to ruthenium (P,C bonding). The P,C complexes revert back to the P,S bonding mode by reaction with acid; therefore, metal-thienyl bonding is reversibly switchable. The effect of interaction of the metal groups in the different bonding modes with the thienyl backbone is reflected by changes in alignment of the thienyl rings in the solid-state structures of the complexes, the redox potentials, and the pi --> pi transitions in solution. Methyl substituents attached to the terthiophene groups allow observation of the effect of these substituents on the conformational and electronic properties and aid in assignments of the electrochemical data. The PT(n)() ligands bound in P,S and P,C bonding modes also alter the electrochemical and spectroscopic properties of the ruthenium bis(bipyridyl) group. Both bonding modes result in quenching of the oligothiophene luminescence. Weak, short-lived Ru --> bipyridyl MLCT-based luminescence is observed for [Ru(bpy)(2)PDo(2)T(5)-P,S](PF(6))(2), [Ru(bpy)(2)PT(3)-P,C]PF(6), [Ru(bpy)(2)PMeT(3)-P,C]PF(6), and [Ru(bpy)(2)PMe(2)T(3)-P,C]PF(6), and no emission is observed for the alternate bonding mode of each complex.

[reaction: see text] Photolysis of 3 in argon-saturated 2-propanol led to formation of 5 via intermolecular H-atom abstraction followed by lactonization. Irradiation of 4 in 2-propanol gave compounds 6 and 7 that also come from intermolecular H-atom abstraction. In contrast, photolysis of an oxygen-saturated solution of 3 in 2-propanol yields products 8, 9, and 10, which were all formed from intramolecular H-atom abstraction and trapping of the corresponding biradical with oxygen. Laser flash photolysis of 3 in methanol showed formation of biradical 3BR (lambda(max) 330 nm, and tau = 50 ns) via intramolecular H-atom abstraction as the main photoreactivity of 3. Biradical 3BR decayed into photoenols 3Z and 3E (lambda(max) 390 nm, tau = 6.5 micros and tau = 162 micros, respectively). In comparison, laser flash photolysis of 4 yielded photoenols 4Z and 4E (lambda(max) 390 nm, tau = 15 micros and tau = 3.6 ms, respectively). Thus photoenol 3E is unusually short-lived, and therefore it does not undergo the intramolecular lactonization as we have observed for the analogous photoenol 1E. Photoenol 3Z decays back to 3 via an intramolecular 1,5-H shift, whereas photoenol 3E reforms 3 efficiently via the solvent with the aid of the ortho ester group. The intramolecular lactonization of photoenols 1E and 3E must be a slow process, presumably because the photoenols are rigid and the hydroxyl group is inhibited, by intramolecular hydrogen bonding, from acquiring the correct geometry for lactonization. Thus only photoenols that are resistant to reformation of their ketone via the solvent are long-lived enough to undergo lactonization and release the alcohol moiety.

Mechanistic investigations of the photoinduced DNA damage by acridizinium salts (4a-azonia-anthracene derivatives) are presented. Irradiation of 9-bromoacridizinium in the presence of defined double- and single-stranded DNA oligomers under aerobic conditions leads to both frank strand breaks and alkali-labile sites as determined by polyacrylamide gel electrophoresis (PAGE). The extent of the DNA damage increases significantly in D(2)O and occurs selectively at guanosine residues. These observations reveal the formation of singlet oxygen ((1)O(2)) as reactive species, which oxidizes the DNA bases, above all the guanine bases. Further evidence for (1)O(2) formation was obtained from laser-flash spectroscopic investigations, which show intersystem crossing (S(1) to T(1)) of the excited states of the parent acridizinium and of the 9-bromo- and 9-amino-substituted derivatives. The resulting triplet state is efficiently quenched by oxygen (k(q) > 10(9) s(-)(1)M(-)(1)) to yield (1)O(2). Under anaerobic conditions, no significant alkali-labile lesions are observed, but frank strand breaks are induced; however, to lesser extent than under aerobic conditions. The DNA damage is suppressed in the presence of a radical scavenger, namely t-BuOH, and hydroxyl radicals are shown to be the reactive intermediates by trapping experiments with terephthalic acid. Moreover, the intercalated acridizinium molecules are not involved in the DNA damage reactions. The intercalated acridizinium salt leads to a primary PET reaction with the DNA bases; however, a fast BET transfer is proposed that regains the dye and the DNA, so that the excited intercalated dye does not contribute significantly to the overall DNA damage.

The binding dynamics with bile salt aggregates for a series of naphthalene derivatives of different polarities was studied using fluorescence and laser flash photolysis. Fluorescence was employed to determine the nature of the binding site for each guest and the accessibility of the bound guest to quenchers. Laser flash photolysis was employed to study the mobility of the triplet states of the naphthalenes between the sodium cholate aggregates and the aqueous phase. Primary aggregates, which provide an environment protected from quenchers in the aqueous phase, bind 1- and 2-ethylnaphthalene as guests. The complexation dynamics with this type of aggregate is slow. 1- and 2-Naphthyl-1-ethanol, and 1- and 2-acetonaphthone bind to the secondary aggregates, which provide moderate protection from quenching and faster binding dynamics. The addition of salts lowered the cholate concentration at which primary aggregates were formed, but did not influence the formation of secondary aggregates.

The photochemistry of several photochromic arene [e]-annelated dimethyldihydropyrenes (DHPs) was studied. These compounds have much larger photochemical ring opening quantum yields than the simple DHPs, making them potentially more useful as building blocks for photoswitchable materials.

The photochemistry of ninhydrin in benzene and water was studied by laser flash photolysis and electron paramagnetic resonance. Its photochemistry was shown to be dependent on the solvent. In benzene, a triplet excited state was observed, which underwent hydrogen abstraction reactions or reduction to the radical anion. In water, the radical anion of ninhydrin was formed within the laser pulse (15 ns) at neutral pH, whereas the neutral ketyl radical was formed by protonation of the radical anion at low pH. A pKa of 0.77 was determined for the protonation equilibrium. The formation of hydrindantin is proposed to occur through the dimerization of the ketyl radical or the radical anion (or both). In addition, ninhydrin was shown to be a poor precursor for the photogeneration of hydroxyl radicals.

A series of dimethyldihydropyrene derivatives was studied to elucidate the photochemical mechanism associated with the switching between the dimethyldihydropyrene (DHP, closed) and metacyclophanediene (CPD, open) forms of the molecule. Quantum yields of ring opening and closure, fluorescence quantum yields and lifetimes, as well as laser flash photolysis studies were performed to establish the effect of substituents on the switching efficiency. Ring opening of the DHPs occurs from the first singlet excited state. The low quantum yields for the ring opening reaction observed (< or =0.042) are a consequence of the low rate constant (< or =1.7 x 10(7) s(-1)) for this process. The quantum yields for ring closure of the CPD were determined for select compounds and were of the order of 0.1-0.4. These results show that the efficiency for ring opening of this class of compounds is intrinsically low, but can be modulated to some extent by the introduction of substituents. These properties should be taken into account when considering what type of photoswitching devices DHPs might be useful for.

The complexation dynamics of radical cations with cyclodextrins (CD) was studied using photophysical techniques. Radical cations of 4-vinylanisole and trans-anethole were formed within alpha- and beta-CD cavities by two-photon photolysis of the respective styrene precursors. Exit of the radical cations from alpha-CD complexes with 1: 1 and 1:2 (guest: CD) stoichiometries and beta-CD complexes with 1:1 stoichiometries occurred with lifetimes shorter than 100 ns. Most of the radical cations formed escape from the CD cavities, but a small portion do react with alpha-CD when this host is present in high concentrations.

Short lived reaction intermediates such as triplet states and free radicals can be detected in vivo using laser photolysis techniques with time-resolved diffuse reflectance detection. This novel approach is illustrated for bacterial suspensions of Bacillus subtilis.

The reaction of isobutyraldehyde with dissolved oxygen catalyzed by horseradish peroxidase has been studied from the standpoint of determining the rate-limiting factor under a variety of conditions. Chemiluminescence from the product triplet acetone and rate of oxygen uptake were determined in simultaneous experiments. The reaction is initiated by the peracid obtained from the uncatalyzed autoxidation of the aldehyde; and under certain conditions the amount of peracid is also rate-limiting in the steady-state portion of the reaction. Under other conditions the total amount of enzyme and under still others the rate of formation of enol from the parent aldehyde controls the rate.

A Michelson interferometer spectrometer cooled to 10 degrees by liquid helium was flown into an IBC class III aurora on 1 April 1976 from Poker Flat, Alas. The sensor, HIRIS, covered the spectral range 455-2500 wave numbers (4-22 microm) with a spectral resolution of 1.8 cm(-1) and an NESR of 5 x 10-12 W/cm2 scrm(-1) at 1000 cm(-1). An atmospheric emission spectrum was obtained every 0.7 sec over an altitude range of 70-125 km. Atmospheric spectra were obtained of CO2 (nu3), NO (Deltanu = 1), O3 (nu3) and CO2 (nu2). Auroral produced excitations were observed for each band, this being the first known measurement of auroral enhancements of O3 (nu3), 9.6 microm, and CO2 (nu2), 15 microm, emissions.

Micelle-solubilized chlorophyll efficiently detects electronically excited species generated in enzymatic systems. In most, if not all, systems the chemiexcited species is formed in the triplet state; chlorophyll fluorescence is observed as result of energy transfer. Red emission can also be elicited from chlorophyll in chloroplasts or bound to microsomes.
