The objective of this article is to establish a condition by which we are able to state that an ellipsoidal fragment formed by a plane cutting the ellipsoid can always contain a sphere in any position inside in it. A method to construct a chain of mutually tangent spheres inscribed in the ellipsoidal segment has been proposed. The locus of the centroid as well as the radii of the mutually tangent spheres have been computed. The prime concern of our work is to explore some geometrical properties of such a chain of spheres which includes the condition of inscribability of a sphere in any position inside the ellipsoid along with the computation of points of tangency between consecutive spheres. Abstract Background Paraquat (1, 1-dimethyl-4, 4-bipyridium dichloride; PQ) causes neurotoxicity, especially dopaminergic neurotoxicity, and is a supposed risk factor for Parkinson's disease (PD). However, the cellular and molecular mechanisms of PQ-induced neurodegeneration are far from clear. Previous studies have shown that PQ induces neuroinflammation and dopaminergic cell loss, but the prime cause of those events is still in debate. Methods We examined the neuropathological effects of PQ not only in substantia nigra (SN) but also in frontal cortex (FC) and hippocampus of the progressive mouse (adult Swiss albino) model of PD-like neurodegeneration, using immunohistochemistry, western blots, and histological and biochemical analyses. Results PQ caused differential patterns of changes in cellular morphology and expression of proteins related to PD and neuroinflammation in the three regions examined (SN, FC and hippocampus). Coincident with behavioral impairment and brain-specific ROS generation, there was differential immunolocalization and decreased expression levels of tyrosine hydroxylase (TH) in the three regions, whereas α-synuclein immunopositivity increased in hippocampus, increased in FC and decreased in SN. PQ-induced neuroinflammation was characterized by area-specific changes in localization and appearances of microglial cells with or without activation and increment in expression patterns of tumor necrosis factor-α in the three regions of mouse brain. Expression of interleukin-1β was increased in FC and hippocampus but not significantly changed in SN. Conclusion The present study demonstrates that PQ induces ROS production and differential α-synuclein expression that promotes neuroinflammation in microglia-dependent or -independent manners, and produces different patterns of dopaminergic neurotoxicity in three different regions of mouse brain.

Abstract Background Cadmium is one of the inflammation-related xenobiotics and has been regarded as a potent carcinogen. The relationship between inflammation and cell proliferation due to chronic infection has been studied, but the mechanism is not fully clear. Though the mode of cadmium toxicity is well characterized in animal cells, still it requires some further investigations. Previously we reported that cadmium induces immune cell death in Swiss albino mice. In the present study we showed that instead of inducing cell death mechanism, cadmium in low concentration triggers proliferation in mice lung cell and our results reveals that prior to the induction of proliferation it causes severe inflammation. Methods Swiss albino mice were treated with different concentrations of cadmium to determine the LD50. Mice were subdivided (5 mice each) according to the exposure period (15, 30, 45, 60 days) and were given sub lethal dose (5 mg/Kg body weight) of cadmium chloride and ibuprofen (50 mg/Kg body weight, recommended dose) once in a week. SEM and histology were performed as evidence of changes in cellular morphology. Inflammation was measured by the expression of Cox-2 and MMPs. Expression of proinflammatory cytokines (Cox-2, IL-6), signaling and cell cycle regulatory molecules (STAT3, Akt, CyclinD1) were measured by western blot, ELISA and immunoprecipitation. Mutagenecity was evidenced by comet assay. Cell proliferation was determined by cell count, cell cycle and DNA analysis. Results Prolonged exposure of low concentration of cadmium resulted in up regulation of proinflammatory cytokines and cell cycle regulatory molecules. Though NSAIDs like Ibuprofen reduces the expression of inflammatory cytokines, but it did not show any inhibitory effect on cadmium adopted lung cell proliferation. Conclusion Our results prove that cadmium causes both inflammation and cell proliferation when applied in a low dose but proliferative changes occur independent of inflammation.

Tumor cells promote immune evasion through upregulation of programmed death-ligand 1 (PD-L1) that binds with programmed cell death protein 1 (PD1) on cytotoxic T cells and promote dysfunction. Though therapeutic efficacy of anti-PD1 antibody has remarkable effects on different type of cancers it is less effective in breast cancer (BC). Hence, more details understanding of PD-L1-mediated immune evasion is necessary. Here, we report BC cells secrete extracellular vesicles in form of exosomes carry PD-L1 and are highly immunosuppressive. Transforming growth factor beta (TGF-β) present in tumor microenvironment orchestrates BC cell secreted exosomal PD-L1 load. Circulating exosomal PD-L1 content is highly correlated with tumor TGF-β level. The later also found to be significantly associated with CD8+CD39+, CD8+PD1+ T-cell phenotype. Recombinant TGF-β1 dose dependently induces PD-L1 expression in Texos in vitro and blocking of TGF-β dimmed exosomal PD-L1 level. PD-L1 knocked down exosomes failed to suppress effector activity of activated CD8 T cells like tumor exosomes. While understanding its effect on T-cell receptor signaling, we found siPD-L1 exosomes failed to block phosphorylation of src family proteins, linker for activation of T cells and phosphoinositide phospholipase Cγ of CD8 T cells more than PD-L1 exosomes. In vivo inhibition of exosome release and TGF-β synergistically attenuates tumor burden by promoting Granzyme and interferon gamma release in tumor tissue depicting rejuvenation of exhausted T cells. Thus, we establish TGF-β as a promoter of exosomal PD-L1 and unveil a mechanism that tumor cells follow to promote CD8 T-cell dysfunction. [ABSTRACT FROM AUTHOR]

In this paper we have studied generalized Sasakian space form admitting almost quasi-Yamabe soliton and gradient almost quasi-Yamabe soliton. It is shown that if a generalized Sasakian space form admits a closed almost quasi-Yamabe soliton, then either soliton vector field is pointwise collinear with ζ or the structure functions are connected by a relation and the manifold becomes Ricci semi-symmetric generalized Sasakian space form. Next, it is proven that, if the metric of a generalized Sasakian space form is a gradient almost quasi-Yamabe soliton, then either the gradient of ψ is pointwise collinear with ζ or the structure functions are connected by a relation. Background: Cancer-associated fibroblasts (CAFs) are known to impact on tumour behaviour, but the mechanisms controlling this are poorly understood.Methods: Breast normal fibroblasts (NFs) or CAFs were isolated from cancers by laser microdissection or were cultured. Fibroblasts were transfected to manipulate miR-222 or Lamin B receptor (LBR). The fibroblast-conditioned medium was collected and used to treat epithelial BC lines MDA-MB-231 and MDA-MB-157. Migration, invasion, proliferation or senescence was assessed using transwell, MTT or X-gal assays, respectively.Results: MiR-222 was upregulated in CAFs as compared with NFs. Ectopic miR-222 expression in NFs induced CAF-like expression profiles, while miR-222 knockdown in CAFs inhibited CAF phenotypes. LBR was identified as a direct miR-222 target, and was functionally relevant since LBR knockdown phenocopied miR-222 overexpression and LBR overexpression phenocopied miR-222 knockdown. MiR-222 overexpression, or LBR knockdown, was sufficient to induce NFs to show the CAF characteristics of enhanced migration, invasion and senescence, and furthermore, the conditioned medium from these fibroblasts induced increased BC cell migration and invasion. The reverse manipulations in CAFs inhibited these behaviours in fibroblasts, and inhibited paracrine influences on BC cells.Conclusion: MiR-222/LBR have key roles in controlling pro-progression influences of CAFs in BC. The volume of a pentahedron is calculated in terms of dihedral angles, sides, face angles of component tetrahedrons (3-simplex) using an exact computation technique (known as signed simplex decomposition). Here the signed simplex decomposition means an established technique to subdivide a polyhedra (a simplicial complex or more strictly CW complex) to component tetrahedral (3-simplex) which is used in computer graphics where meshes with pyramids, prisms or hexahedra must be subdivided into tetrahedral to use efficient algorithms for volume-rendering, iso-contouring and particle advect. One of the most practical approaches for establishing a successful microbial fuel cell (MFC) is to fasten the oxidation rate of the substrate by the microorganisms to get quick paced electron transfer between microbes and electrode. A genetically modified Escherichiacoli, overexpressing α-amylase, is constructed and applied as biocatalyst in MFC using starch as substrate. The results are compared with nonrecombinant, native E.coli. The results show better performance for the MFC containing the recombinant strain demonstrated by higher power density (PD), lower resistance, and significant electrochemical activity. Maximum PD has been recorded as 279.04 mW m−2 compared to 120.33 Mw m−2 for the MFC operated with nonrecombinant E.coli. The impedance results also suggest the effectiveness of the recombinant strain by lowering the internal resistance by more than half order as compared to the nonrecombinant one. These results affirm that the engineered strain can be used as a superior biocatalyst in contrast to the native strain and by using the technique of genetic alteration; gene of interest can be inserted based on the substrate to be treated. In this paper, we have established the existence of conformal Ricci soliton as self similar solution of conformal Ricci flow and characterized the potential function f of almost conformal gradient Ricci soliton on a Sasakian manifold in terms of the non-dynamical scalar field p. We have found the necessary condition for the potential function f to be constant. Furthermore, a relation between λ and the potential function f has been established. Finally, we prove a sufficient condition for an almost conformal Ricci soliton to be an almost conformal gradient Ricci soliton and characterize it in terms of shrinking, steady or expanding soliton.
