Background A mutation in presenilin 1 (E280A) causes early-onset Alzheimer's disease. Understanding the origin of this mutation will inform medical genetics.Methods We sequenced the genomes of 102 individuals from Antioquia, Colombia. We applied identity-by-descent analysis to identify regions of common ancestry. We estimated the age of the E280A mutation and the local ancestry of the haplotype harboring this mutation.Results All affected individuals share a minimal haplotype of 1.8 Mb containing E280A. We estimate a time to most recent common ancestor of E280A of 10 (95% credible interval, 7.2-12.6) generations. We date the de novo mutation event to 15 (95% credible interval, 11-25) generations ago. We infer a western European geographic origin of the shared haplotype.Conclusions The age and geographic origin of E280A are consistent with a single founder dating from the time of the Spanish Conquistadors who began colonizing Colombia during the early 16th century.

Introduction: Hypertension is a multifactorial disease influenced by genetic and environmental components, with its prevalence varying across ethnic groups. Manifold studies on blood pressure regulatory system genes have been carried out —such as the renin-angiotensin-aldosterone system, the sympathetic nervous system, endothelial factor, and sodium balance—, but the results yielded were inconsistent among populations.Objectives: To evaluate the effect of both variants in genes AGT, AGTR1, ACE, ADRB2, DRD1, ADD1, ADD2, ATP2B1, TBXA2R PTGS2, and the result of the individual ancestry component on hypertension and blood pressure levels among population in Antioquia.Methods and materials: 107 cases and 253 controls were genotyped for 12 variants on genes AGT, AGTR1, ACE, ADRB2, DRD1, ADD1, ADD2, ATP2B1, TBXA2R y PTGS2, and for 20 ancestry informative markers. The association of polymorphisms and their interactions, and the association of ancestral genetic composition with hypertension and blood pressure levels were examined.Results: Genes ADD2, rs4852706 (OR=3.0; p=0.023); DRD1, rs686 (OR=0.38; p=0.012) and ADRB2, rs1042718 (OR=10.0; p=0.008); as well as genotypic combinations of DRD1 and AGTR1; AGT and ADD1; and ADD1 to ATP2B1 and PTGS2 were associated to hypertension. The Amerindian ancestry component was associated to some decrease in diastolic blood pressure.Conclusion: Variants on genes ADD2, DRD1, ADRB2, AGTR1, AGT, ADD1, ATP2B1 and PTGS2 individually or interacting, are associated to hypertension. La hipertensión arterial es una enfermedad multifactorial influenciada por componentes genéticos y ambientales, cuya prevalencia varía entre grupos étnicos. Se han llevado a cabo numerosos estudios en genes de sistemas reguladores de la presión arterial, como el sistema renina-angiotensina-aldosterona, el sistema nervioso simpático, los factores endoteliales, y el balance de sodio, mostrando resultados incongruentes entre poblaciones.Objetivos. Introduction. Autism spectrum disorders are severe neurodevelopmental disorders with a stronggenetic component. The potential role of the serotoninergic system in the development of autistic disorder has been based on the observation of hyperserotoninemia in autistic subjects and the results of drug treatment studies. Multiple molecules involved in serotonin metabolism and neurotransmission have been studied; however, replication studies have been inconsistent. This may be partially related to the marked genetic heterogeneity of autism in different populations.Objectives. The relationship between autism and single nucleotide polymorphisms of SLC6A4, HTR2Aand ITGB3 genes was evaluated in an urban population of northwestern Colombia.Materials and methods. In Antioquia, Colombia, 42 families with history of autism were screened for 10 SNPs in SLC6A4, HTR2A and ITGB3 genes and evaluated for associations with the transmission disequilibrium test. The interactions among these genes and autism was assessed with multidimensional reduction methods.Results. A significant main effect was seen among the SLC6A4 gene variants rs4583306 (OR=2.6,p=0.004) and rs2066713 (OR=2.2, p=0.03). No main effect of the ITGB3 or HTR2A variants was found, however, in the interaction effects, the SLC6A4 and HTR2A genes demonstrated significant evidence of association with autism (p<0.001).Conclusion. Significant association of markers were discovered within the SLC6A4 gene and the combination of SLC6A4 and HTR2A (S-A) genes to autism. These results were consistent with previous studies conducted in other populations and provide further evidence for the implication of these rotoninergic system in the etiology of autistic disorders. doi: http://dx.doi.org/10.7705/biomedica.v32i4.593, Introducción. El espectro autista constituye un grupo de trastornos graves del neurodesarrollo, conun fuerte componente genético. Se ha sugerido un papel importante del sistema serotoninérgico en el desarrollo de este grupo de trastornos, con base en los estudios de respuesta a medicamentos y la hiperserotoninemia, característica común en el autismo. Se han implicado múltiples moléculas en el metabolismo y la neurotransmisión de la serotonina; sin embargo, los resultados de los estudios hantenido poca congruencia entre diferentes poblaciones.Objetivos. Evaluar la relación entre el autismo y el polimorfismo de nucleótido simple (SingleNucleotide Polymorphism, SNP) en los genes SLC6A4, HTR2A e ITGB3, en una muestra de la población antioqueña.Materiales y métodos. Se genotipificaron 42 núcleos familiares con autismo para 10 variantes enlos genes SLC6A4, ITGB3 y HTR2A. Se evaluó la asociación utilizando la prueba de desequilibrio enla transmisión. Se exploró el impacto de la interacción entre estos genes y el autismo, utilizando la reducción multidimensional.Resultados. Se encontró asociación de las variantes rs4583306 (OR=2,6, p=0,004) y rs2066713(OR=2,2 p=0,03), en el gen SLC6A4, y asociación de combinaciones genotípicas entre los genes SLC6A4 y HTR2A y el riesgo de autismo (p=0,0001).Conclusiones. Se encontró asociación significativa con variantes en el gen transportador de serotoninacon el autismo, al igual que interacción entre variantes en los genes HTR2A con SLC6A4. Introduction. The environmental risk factors such as food intake and physival activity, are determinants in the etiology of metabolic syndrome in overweight adolescents.Objective. To explore the association between environmental risk factors and components presence of metabolic syndrome in overweight youngsters in Medellín.Materials and methods. Adolescents between the ages of 10 and 18 were selected for a cross sectional study. Body composition by anthropometry, blood pressure, lipid profile, glucose, insulin, food intake and physical activity level were assessed in the study population.Results. The prevalence for metabolic syndrome components of hypertriglyceridemia was 40.9%; hypertension, 20.9%; low HDLc, 15.6%; high waist circumference, 4.0%, and hyperglycemia, 0.9%; the overall prevalence of metabolic syndrome was 3.1%. There was a statistical difference (p<0.005) between the consumption of calories, simple and total carbohydrates and the presence of the components; no association was found between the level of physical activity and the presence of components (p>0.05). The logistic regression model showed a higher probability of having at least one component if the youngster was male (p=0.022), with a higher BMI (Body Mass Index) (p=0.019) and was located in the fourth simple carbohydrates consumption quartile (p=0.036). Conclusions. Environmental risk factors associated with components of metabolic syndrome were the increased consumption of calories, simple and complex carbohydrates, all directly related to the BMI. In contrast, the level of physical activity, family history and personal risk factors showed no association. The metabolic syndrome only occurred in youngsters with obesity., Introducción. Factores de riesgo ambientales, como el consumo de alimentos y la actividad física, son determinantes en la etiología del síndrome metabólico y sus componentes en jóvenes con exceso de peso.Objetivo. Explorar la asociación entre factores de riesgo ambientales y presencia de componentes del síndrome metabólico en jóvenes entre 10 y 18 años, con exceso de peso en Medellín.Materiales y métodos. Se llevó a cabo un estudio descriptivo de corte transversal. En la población de estudio se evaluaron la composición corporal por antropometría, la presión arterial, el perfil lipídico, la glucemia, la insulinemia, la ingestión de alimentos y la actividad física.Resultados. La prevalencia de los componentes del síndrome metabólico fue de 40,9 % para hipertrigliceridemia; 20,9 % para hipertensión; 15,6 % para c-HDL bajas; 4,0 % para circunferencia de cintura alta, y 0,9 % para la hiperglucemia; la prevalencia de síndrome metabólico fue de 3,1 %. Se encontró diferencia estadística (p<0,005) entre el consumo de kilocalorías, carbohidratos totales y simples, y la presencia de los componentes; no se encontró asociación entre el nivel de actividad física y la presencia de componentes (p>0,05). En el modelo de regresión logística se encontró una mayor probabilidad de tener, al menos, un componente si el joven era de sexo masculino (p=0,022), tenía un mayor índice de masa corporal (IMC) (p=0,019) y si se ubicaba en el cuarto cuartil de consumo de carbohidratos simples (p=0,036).Conclusiones. Los factores de riesgo ambientales asociados con los componentes del síndrome metabólico en este estudio fueron el mayor consumo de calorías, carbohidratos complejos y simples, todos relacionados directamente con el IMC; por el contrario, el nivel de actividad física, los antecedentes familiares y los personales no mostraron ninguna asociación. In Antioquia, Colombia, four families have been reported with juvenile Parkinson´s disease and carrying the C212Y mutation in the Parkin gene. Many cognitive alterations associated with idiopathic, late-onset Parkinson´s disease have been described; however, little attention has been paid to the description of neuropsychological profiles in families carrying mutations in genes associated with juvenile Parkinson´s disease. For this study we selected a group of ten homozygous carriers of Parkin mutation C212Y with the clinical and a group of molecular diagnosis of Parkinson´s disease, and ten healthy relatives as controls. The neuropsychological evaluation revealed statistically significant differences between the two groups (p < 0.05) in Minimental State Examination and in tests evaluating working memory and attention in which prolonged execution times and marked slowing down of information processing were observed. We suggest that the observed alterations could be considered as neuropsychological features of patients with the C212Y mutation in the Parkin gene, the phenotypic expression of which seems to be associated in this population with slow evolution, mild cognitive impairment and functional involvement., En Antioquia (Colombia) hay reportadas cuatro grandes familias afectadas con enfermedad de Parkinson Familiar Juvenil portadoras de la mutación C212Y en el gen Parkin. Nosotros seleccionamos un grupo de 10 sujetos con diagnóstico de enfermedad de Parkinson (EP), portadores de la mutación C212Y y un grupo de 10 familiares sanos (no portadores). En el ámbito neuropsicológico se evidencian diferencias significativas entre los grupos (p<0.05) en el minimental state, y en pruebas que evalúan la memoria inmediata y la atención, observandose tiempos prolongados de ejecución y marcada lentificación. Esta investigación sugiere que las alteraciones observadas podrían ser consideradas como rasgos neuropsicológicos de los pacientes con la mutación C212Y en el gen Parkin, cuya expresión fenotípica en esta población parece estar asociada con una lenta evolución y a deterioro cognitivo leve (DCL).

Introduction. The uncoupling proteins belong to the family of anion transporting proteins which uncouple the ATP production from the mitochondrial respiration, cause proton leakage through the inner mitochondrial membrane, and release energy as heat. Although uncoupling protein function has not been well established, specific polymorphisms in these proteins have been associated with type 2 diabetes mellitus, obesity and insulin resistance.Objective. The association was assessed between the polymorphisms in uncoupling protein genes 1, 2 and 3 genes and type 2 diabetes mellitus.Materials and methods. In a northwestern Colombian population, 545 diabetes cases and 449 controls were investigated for presence of 14 polymorphisms in uncoupling protein genes (3826A/G, ID 45, 2723T/A, 1957G/A, 866G/A, and 55C/T) by PCR and PCR-RFLP. Single associations were evaluated by chi-square test, and bayesian logistic regression analysis was done including as covariates the individual admixture estimates obtained by 54 informative markers for European, African and Amerind ancestry.Results. Association between type 2 diabetes mellitus and the polymorphisms 3826A (OR=0.78; 95%CI=0.63-0.97; p=0.02) and 55C (OR=1.41; 95%CI=1.04-1.92; p=0.03) and the haplotype D45, 866G, 1957G, 2723T, and 55C (OR=1.26; 95%CI=1.02-1.56; p=0.03) were found. These associations remained after adjustment using individual genetic admixture estimates.Conclusion. Some alleles of uncoupling protein genes 1, 2 and 3, and their haplotypes confer risk to type 2 diabetes in a northwestern Colombian population.,  Introducción. Las proteínas desacoplantes pertenecen a la familia de proteínas transportadoras de aniones que desacoplan la producción de ATP de la respiración mitocondrial, causando pérdida de protones a través de la membrana mitocondrial interna y disipando la energía en forma de calor. Aunque su función no ha sido bien establecida, algunos polimorfismos en estas proteínas se han asociado con diabetes mellitus tipo 2, obesidad y resistencia a la insulina.Objetivo. Evaluar la asociación entre las variantes -3826A/G, ID 45, -2723T/A, -1957G/A, -866G/A, -55C/T de los genes de las proteínas desacoplantes 1, 2 y 3 con diabetes mellitus tipo 2 en una población del nordeste colombiano.Materiales y métodos. Se tipificaron 545 casos y 449 controles para 14 variantes de los genes de las proteínas desacoplantes por medio de PCR y PCR-RFLP. Se hicieron pruebas de asociación simples con ji al cuadrado y se corrigieron en un análisis de regresión logística bayesiana, incluyendo los estimados de mezcla individual obtenidos mediante 54 marcadores informativos de ascendencia europea, africana y amerindia.Resultados. Las variantes -3826A (OR=0,78; IC95% 0,63-0,97; p=0,02), -55C (OR=1,41; IC95% 1,04-1,92; p=0,03) de las proteínas desacoplantes 1 y 3, respectivamente, y el haplotipo D45, -866G, -1957G, -2723T, -55C (OR=1,26; IC95% 1,02-1,56; p=0,03) se asociaron con diabetes tipo 2. Estas asociaciones se conservaron después de ajustar por la mezcla genética individual.Conclusión. Algunas variantes de las proteínas desacoplantes 1, 2 y 3, y sus haplotipos, confieren riesgo para diabetes mellitus tipo 2 en una población del nordeste colombiano.&nbsp

The large and diverse population of Latin America is potentially a powerful resource for elucidating the genetic basis of complex traits through admixture mapping. However, no genome-wide characterization of admixture across Latin America has yet been attempted. Here, we report an analysis of admixture in thirteen Mestizo populations (i.e. in regions of mainly European and Native settlement) from seven countries in Latin America based on data for 678 autosomal and 29 X-chromosome microsatellites. We found extensive variation in Native American and European ancestry (and generally low levels of African ancestry) among populations and individuals, and evidence that admixture across Latin America has often involved predominantly European men and both Native and African women. An admixture analysis allowing for Native American population subdivision revealed a differentiation of the Native American ancestry amongst Mestizos. This observation is consistent with the genetic structure of pre-Columbian populations and with admixture having involved Natives from the area where the Mestizo examined are located. Our findings agree with available information on the demographic history of Latin America and have a number of implications for the design of association studies in population from the region.

Introduction. The E280A mutation of the presenilin 1 gene has been found to be the most common associate in Alzheimer's patients with a family history of this disease. Presenilin 1 is a critical component of the g-secretase complex and plays an essential role in the production of amyloid-b peptide. This peptide has been strongly associated with the physiopathology of the disease.Objective. The E280A mutation in the presenilin 1 was investigated for increased production of amyloid-b, as a response to gain in g-secretase function.Materials and methods. Levels of systemic amyloid-b were measured with congo red staining and immuno-histochemistry of the tissues of affected cadavers, compared with non-affected cadavers. The 40 and 42 amino acid amyloid-b levels were quantified by ELISA assay in CHO cell cultures. The amyloid precursor protein expressed by the cultures was detected by transfection with the cDNAs of presenilin 1 carrying the M146L, E280A, DE9 y L392V mutations.Results. Protein deposits were found in all tissues investigatged, but only a few with b-amyloid deposition. No differences were observed in the amount or location of amyloid-b between affected and unaffected cadavers. Not increase was noted in the production of amyloid-b from the CHO cells transfected with cDNA from any of the mutations of presenilin 1.Conclusions. The E280A mutation in the presenilin 1 gene was not associated with the increased production of amyloid-b in non-neuronal peripheral tissues, or in the in vitro model. This is in contrast to the expectation in a g-secretase gain of function., Introducción. La mutación E280A en el gen de presenilina 1 se encuentra asociada al grupo familiar más grande del mundo con enfermedad de Alzheimer. La presenilina 1 es un componente esencial del complejo g-secretasa, responsable de la producción del péptido b-amiloide, considerado clave en la fisiopatogenia de la enfermedad.Objetivo. Determinar si la mutación E280A en presenilina 1 incrementa la producción de b-amiloide, como reflejo de la ganancia de función g-secretasa.Materiales y métodos. Se evaluaron, por la tinción con rojo congo e inmunohistoquímica, depósitos sistémicos de b-amiloide en tejidos de cadáveres afectados, portadores de la mutación E280A en la presenilina 1 y cadáveres de no afectados. Se cuantificó por la técnica de ELISA el b-amiloide de 40 y 42 aminoácidos en cultivos de células CHO que expresan la proteína precursora de amiloide, transfectadas con los cADN de presenilina 1 portando las mutaciones M146L, E280A, DE9 y L392V.Resultados. Se encontraron depósitos proteicos en todos los tejidos estudiados y escasos depósitos de b-amiloide. No se encontró diferencia en la cantidad de depósitos, ni en su localización. No se observó incremento en la producción de b-amiloide en las células transfectadas con los cADN de presenilina 1 con las mutaciones comparadas con los controles. Conclusiones. La mutación E280A en presenilina 1 no aumentó la producción de b-amiloide en tejidos periféricos no neuronales o en el modelo in vitro, contrario a lo que sucede en una ganancia de función de g-secretasa.

Introduction. Genetic studies of Trypanosoma cruzi have tried to establish relations between genetic variants and their biological characteristics, such as clinical manifestations, host or geographic origin. However, much controversy exists on the associations between the commonly used DNA markers with group, clinical characteristics and disease epidemiology.Objective. In this study determined the variability of the genes that code for the proteins trypanothione reductase and cruzipain, both involved in the infection and survival of the parasite in the mammalian host, was studied and the association between genetic polymorphism and biological and geographic sources in Colombian T. cruzi strains was examined.Materials and methods. The genotypes for each of six SNPs (single nucleotide polymorphisms) for trypanothione reductase and eight SNPs for cruzipain genes were identified by polymerase chain reaction-restriction fragment length polymorphism.in 36 T. cruzi Colombian stocks from several regions and biological origins. Results. Three genotypes were identified for trypanothione reductase with Acy I and Hae III enzymes and six genotypes for cruzipain with the Rsa I, Ban I and Bsu 36I enzymes. Conclusion. For trypanothione reductase ,an association was not established with biological or geographical origin; however, alleles at positions 102 and 210 allowed discrimination with groups I and II. For cruzipain, specific genotypes were associated with group, biological and geographic origin. The usefulness of molecular markers on these genes was demonstrated for the determination and differentiation of genetic varieties in T. cruzi., Introducción. Los estudios genéticos en Trypanosoma cruzi han buscado establecer asociaciones de variantes genéticas del parásito con manifestaciones clínicas de la enfermedad, origen biológico y geográfico de los aislamientos; sin embargo, los resultados de asociación con los marcadores comúnmente usados en estos estudios han generado mucha controversia, principalmente en la asociación de grupos con características clínicas y epidemiológicas de la enfermedad.Objetivo. Se planteó determinar la variabilidad de genes que codifican para las proteínas tripanotión reductasa y cruzipaína, involucradas en mecanismos de infección y supervivencia del parásito en el hospedador mamífero, y probar la asociación de variantes génicas con origen biológico y geográfico de cepas colombianas de T. cruzi.Materiales y métodos. Se tipificaron por reacción en cadena de la polimerasa- polimorfismo de longitud del fragmento de restricción seis SNPs (Single Nucleotide Polymorphisms) en tripanotión reductasa y ocho SNPs en cruzipaína en 36 cepas colombianas de T. cruzi de diferentes regiones y origen biológico.Resultados. Con las enzimas Acy I y Hae III se determinaron tres genotipos para tripanotión reductasa. Para cruzipaína se identificaron seis genotipos con las enzimas Rsa I, Ban I y Bsu 36I.Conclusiones. Para tripanotión reductasa no fue posible establecer una asociación con el origen biológico o geográfico; sin embargo los alelos producidos en las posiciones 102 y 210, permitieron discriminar los grupos tradicionales I y II. Con los genotipos obtenidos para cruzipaína se establecieron relaciones a estos grupos, origen biológico y geográfico. Los resultados sugieren la utilidad de estos genes como marcadores moleculares para determinar y diferenciar variedades genéticas en T. cruzi.

The Nramp1 gene has been associated with natural resistance to intracellular microorganisms in several species including bovine. Recent evidence suggests an association between polymorphism in the 3' untranslated region (3' UTR) of this gene with resistance/susceptibility (R/S) to Brucella abortus as determined in vivo and in vitro. In this study we tested for the variability of the short tandem repeat (STR) within the 3' UTR of Nramp1 in six breeds of Colombian creole cattle (CCC) and compared the genotypes with those of Holstein and Brahman, which were recently introduced into this country. In CCC as well as in Holstein we found the allele 175 fixed in all populations. In Brahman, 175 allele was also present with a frequency of 0.467 but additionally, in this breed there appeared five other alleles and among them two previously unreported: 183 y 185; also was found the allele 189 in the Colombian creole Harton del Valle cattle, which is not previously reported. Together these results suggest that the 175 allele in the 3' UTR Nramp1 may be an ancestral allele in cattle and if this is true the association previously reported with the R/S trait requires further evaluation.

Introduction. Surname frequency (isonymy) is used as a marker of paternal lineage and is used to characterize human population structure. Principles of isonymy were used to determine the genetic structure, migration rates, ancestry relations and origins of populations. This analysis was applied to two historically related local populations which currently are considered to be genetically isolated.Objective. The genetic relationships and influence zones of the Aranzazu and Marinilla populations were assessed by means of surname frequency analysis.Materials and methods. Data originated from database with the title "System of Identification of Beneficiaries of the Social Programs" database or Sisben. Population parameters such as a priori kinship (fii), population homogeneity with B and C estimators, and Cavalli-Sforza's genetic distance were calculated for (a) three towns of Marinilla and its influence zone and (b) Aranzazu. The Rionegro population served as an external, comparison population.Results. The Aranzazu and Marinilla populations showed the higher homogeneity (B value between 0.25 and 0.5) in contrast with Rionegro (B = 0.159), as well as greater a priori kinship values (fii between 0.003 and 0.010). The lowest distances were found between Marinilla and Aranzazu.Conclusions. Aranzazu is a population with characteristics similar to those of Marinilla and its influence zone. The close similarity of genetic characteristics for these populations is due probably to a founder effect. La utilización de la frecuencia de apellidos como marcadores de linajes paternos ha permitido caracterizar poblaciones. Los principios de isonimia se han empleado para determinar el grado de estructuración genética, las tasas de migraciones y las relaciones de ancestría y origen entre poblaciones. Este análisis se aplicó a dos poblaciones históricamente relacionadas y consideradas como aislados genéticos.Objetivo. Evaluar las relaciones genéticas y de origen entre Aranzazu y Marinilla y su zona de influencia por medio de análisis de frecuencia de apellidos.Materiales y métodos. A partir de la base de datos del Sistema de Identificación de Beneficiarios de los Programas Sociales, Sisbén, se calcularon los parámetros poblacionales de coeficiente de parentesco (fii), la homogeneidad poblacional con los estimadores B (porcentaje de la población que comparte los siete apellidos más frecuentes) y C (15 apellidos más frecuentes) y la distancia genética de Cavalli-Sforza en tres poblaciones del núcleo fundador de Marinilla y Rionegro como población externa.Resultados. Marinilla y Aranzazu, al igual que las poblaciones de Marinilla y su zona de influencia, mostraron los mayores valores de homogeneidad (valores B entre 0,25 y 0,5) comparados con Rionegro (B = 0,159) y también mayores valores de parentesco intrapoblacional (valores fii entre 0,0034 y 0,01). Las menores distancias se encontraron entre Marinilla y Aranzazu.Conclusiones. Aranzazu es una población con características similares a las de Marinilla y su zona de influencia y debido al efecto fundador, estas poblaciones pueden presentar características genéticas similares. Por lo tanto, las enfermedades genéticas, principalmentelas de herencia compleja, podrían tener la misma etiología genética en ambas poblaciones, lo que garantizaría las condiciones óptimas para estudios de cartografía genética.
