ABSTRACT Integrated crop-livestock systems (ICLS) foster synergistic relationships to increase nitrogen (N) cycling and soil organic carbon (SOC) accrual in agricultural setups. This study evaluated how the grazing intensity and N fertilization (rates and timing) affect both SOC and N fractions, and soil organic matter chemical composition in an ICLS managed under no-tillage in an Oxisol, six years after initiation. The ICLS was compared to a nearby pasture (PA) and a native forest (NF). The treatments consisted of two grazing intensities: Low Sward Height (LH) and High Sward Height (HH) were maintained with high and low stocking rates, respectively. The HH varied between 0.20 and 0.60 m, and LH between 0.10 and 0.30 m according to the plant forage species throughout the experiment. Fertilization using 200 kg ha-1 N-urea, not splitting up, was conducted at two timings, either at the winter pasture establishment (autumn), about 35 days after sowing or during the summer cash crop cycle (spring). Total N amount per year, including both phases, pasture and cash crop was the same for all treatments. The SOC and N contents were assessed in soil and particulate organic matter (POM), while carbon (C) and N stocks were specifically determined in the soil. Soil organic matter composition was characterized by FTIR. The combination of HH and N fertilization during the pasture phase increased the content of C from 36.1 to 39.9 ± 0.7 g C kg-1 and of N from 2.7 to 3.2 ± 0.1 g N kg-1. The SOC stocks varied from 37.3 to 41.1 ± 0.7 Mg C ha-1, and the N stocks from 2.1 to 3.3 ± 0.1 Mg N ha-1 at 0.0-0.10 m soil layer. The SOC content of the POM and the soil organic matter chemical composition determined by FTIR were mainly affected by the grazing intensity. The HH led to an increased in C content within the POM fraction, reaching values of 51.6 ± 1 and 49.2 g C kg-1, respectively to N crop fertilization and N pasture fertilization. Land-use changed how organic functional groups were stored in soil organic matter fractions. The NF had a greater abundance of aliphatic and phenol in the MAOM, while pasture and ICLS systems had greater aliphatic in the POM fraction. In ICLS, SOC accrual was positively associated with more recalcitrant organic functional groups of phenol, aromatic, and carbonyl C-O. The HH increases SOC accrual, while N-fertilization on pasture ensures adequate nutrition of plants and animals during the winter ICLS phase, at the same time as providing greater residual N for subsequent cash crops through enhanced catalyzed by ruminants. Therefore, grazing and fertilization management strategies should be considered to promote sustainable agriculture intensification with ICLS.

RESUMO Esta pesquisa busca investigar conformações das práticas equestres de corrida no espaço social de Santa Cruz do Sul/Rio Grande do Sul na primeira década do século XX. Com base em preceitos históricos e socioculturais, foram analisados indícios em documentos impressos, especialmente o jornal Kolonie. A partir do cotejamento e interpretação das evidências, pondera-se sobre uma configuração cunhada pelas dependências dinâmicas de pessoas que se organizavam em torno de práticas centradas no cavalo, estabelecendo relações socioculturais, equilibradas em aproximações e distanciamentos sobre noções de identidade e classe.

The taxonomy of the Neotropical Carcharodini genus Viuria Grishin, 2019 (Lepidoptera: Hesperiidae: Pyrginae) is presented. Two new species are described: Viuria inanna sp. nov. from south-southeastern habitats Brazil and Paraguay, and Viuria acadia sp. nov. from Brazil and Ecuador. Illustrations of adults, including morphology of secondary sexual organs in males, male and female genitalia, diagnoses, remarks, and updated geographical distributions are also provided for all species.

It remains challenging in chronic lymphocytic leukemia (CLL) to distinguish between patients with favorable and unfavorable time-to-first treatment (TTFT). Additionally, the downstream protein correlates of well-known molecular features of CLL are not always clear. To address this, we selected 40 CLL patients with TTFT ≤24 months and compared their B cell intracellular protein expression with 40 age- and sex-matched CLL patients with TTFT >24 months using mass spectrometry. In total, 3268 proteins were quantified in the cohort. Immunoglobulin heavy-chain variable (IGHV) mutational status and trisomy 12 were most impactful on the CLL proteome. Comparing cases to controls, 5 proteins were significantly upregulated, whereas 3 proteins were significantly downregulated. Of these, only THEMIS2, a signaling protein acting downstream of the B cell receptor, was significantly associated with TTFT, independently of IGHV and TP53 mutational status (hazard ratio, 2.49 [95% confidence interval, 1.62-3.84]; P < 0.001). This association was validated on the mRNA and protein level by quantitative polymerase chain reaction and ELISA, respectively. Analysis of 2 independently generated RNA sequencing and mass spectrometry datasets confirmed the association between THEMIS2 expression and clinical outcome. In conclusion, we present a comprehensive characterization of the proteome of untreated CLL and identify THEMIS2 expression as a putative biomarker of TTFT.

Summary: Successful explicit memory encoding is associated with inferior temporal activations and medial parietal deactivations, which are attenuated in aging. Here we used dynamic causal modeling (DCM) of functional magnetic resonance imaging data to elucidate effective connectivity patterns between hippocampus, parahippocampal place area (PPA), and precuneus during encoding of novel visual scenes. In 117 young adults, DCM revealed pronounced activating input from the PPA to the hippocampus and inhibitory connectivity from the PPA to the precuneus during novelty processing, with both being enhanced during successful encoding. This pattern could be replicated in two cohorts (N = 141 and 148) of young and older adults. In both cohorts, older adults selectively exhibited attenuated inhibitory PPA-precuneus connectivity, which correlated negatively with memory performance. Our results provide insight into the network dynamics underlying explicit memory encoding and suggest that age-related differences in memory-related network activity are, at least partly, attributable to altered temporo-parietal neocortical connectivity.

Rapid climate warming is altering Arctic and alpine tundra ecosystem structure and function, including shifts in plant phenology. While the advancement of green up and flowering are well-documented, it remains unclear whether all phenophases, particularly those later in the season, will shift in unison or respond divergently to warming. Here, we present the largest synthesis to our knowledge of experimental warming effects on tundra plant phenology from the International Tundra Experiment. We examine the effect of warming on a suite of season-wide plant phenophases. Results challenge the expectation that all phenophases will advance in unison to warming. Instead, we find that experimental warming caused: (1) larger phenological shifts in reproductive versus vegetative phenophases and (2) advanced reproductive phenophases and green up but delayed leaf senescence which translated to a lengthening of the growing season by approximately 3%. Patterns were consistent across sites, plant species and over time. The advancement of reproductive seasons and lengthening of growing seasons may have significant consequences for trophic interactions and ecosystem function across the tundra.

Owing to their ultra-high accelerating gradients, combined with injection inside micrometer-scale accelerating wakefield buckets, plasma-based accelerators hold great potential to drive a new generation of free-electron lasers (FELs). Indeed, the first demonstration of plasma-driven FEL gain was reported recently, representing a major milestone for the field. Several groups around the world are pursuing these novel light sources, with methodology varying in the use of wakefield driver (laser-driven or beam-driven), plasma structure, phase-space manipulation, beamline design, and undulator technology, among others. This paper presents our best attempt to provide a comprehensive overview of the global community efforts towards plasma-based FEL research and development.

Plasma wakefield accelerators are capable of sustaining gigavolt-per-centimeter accelerating fields, surpassing the electric breakdown threshold in state-of-the-art accelerator modules by 3-4 orders of magnitude. Beam-driven wakefields offer particularly attractive conditions for the generation and acceleration of high-quality beams. However, this scheme relies on kilometer-scale accelerators. Here, we report on the demonstration of a millimeter-scale plasma accelerator powered by laser-accelerated electron beams. We showcase the acceleration of electron beams to 128 MeV, consistent with simulations exhibiting accelerating gradients exceeding 100 GV m-1. This miniaturized accelerator is further explored by employing a controlled pair of drive and witness electron bunches, where a fraction of the driver energy is transferred to the accelerated witness through the plasma. Such a hybrid approach allows fundamental studies of beam-driven plasma accelerator concepts at widely accessible high-power laser facilities. It is anticipated to provide compact sources of energetic high-brightness electron beams for quality-demanding applications such as free-electron lasers.

Abstract The rapid ongoing changes in the Central Arctic Ocean call for baseline information on the pelagic fauna. However, sampling for motile organisms which easily escape vertically towed nets is challenging. Here, we report the species composition and catch weight of pelagic fishes and larger zooplankton from 12 trawl hauls conducted in ice covered waters in the Central Arctic Ocean beyond the continental slopes in late summer. Combined trawl catches with acoustics data revealed low amounts of fish and zooplankton from the advective influenced slope region in the Nansen Basin in the south to the ice-covered deep Amundsen Basin in the north. Both arctic and subarctic-boreal species, including the ones considered as Atlantic expatriate species were found all the way to 87.5o N. We found three fish species (Boreogadus saida, Benthosema glaciale and Reinhardtius hippoglossoides), but the catch was limited to only seven individuals. Euphausiids, amphipods and gelatinous zooplankton dominated the catch weight in the Nansen Basin in the mesopelagic communities. Euphausiids were almost absent in the Amundsen Basin with copepods, amphipods, chaetognaths and gelatinous zooplankton dominating. We postulate asymmetric conditions in the pelagic ecosystems of the western and eastern Eurasian Basin caused by ice and ocean circulation regimes.

A20 is a ubiquitin-modifying protein that negatively regulates NF-[kappa]B signaling. Mutations in A20/TNFAIP3 are associated with a variety of autoimmune diseases, including multiple sclerosis (MS). We found that deletion of A20 in central nervous system (CNS) endothelial cells (ECs) enhances experimental autoimmune encephalomyelitis (EAE), a mouse model of MS. [A20.sup.[DELTA]CNS-EC] mice showed increased numbers of CNS-infiltrating immune cells during neuroinflammation and in the steady state. While the integrity of the blood-brain barrier (BBB) was not impaired, we observed a strong activation of CNS- ECs in these mice, with dramatically increased levels of the adhesion molecules ICAM-1 and VCAM-1. We discovered ICOSL to be expressed by A20-deficient CNS-ECs, which we found to function as adhesion molecules. Silencing of ICOSL in CNS microvascular ECs partly reversed the phenotype of [A20.sup.[DELTA]CNS-EC] mice without reaching statistical significance and delayed the onset of EAE symptoms in WT mice. In addition, blocking of ICOSL on primary mouse brain microvascular ECs impaired the adhesion of T cells in vitro. Longitudinal electron-beam diagnostics play a critical role in the operation and control of x-ray free-electron lasers, which rely on parameters such as the current profile, the longitudinal phase space, or the slice emittance of the particle distribution. On the one hand, the femtosecond-scale electron bunches produced at these facilities impose stringent requirements on the resolution achievable with the diagnostics. On the other, research and development of novel accelerator technologies such as beam-driven plasma-wakefield accelerators (PWFA) demand unprecedented capabilities to resolve the centroid offsets in the full transverse plane along the longitudinal bunch coordinate. We present the beam-based commissioning of an advanced X-band transverse-deflection rf structure (TDS) system with the new feature of providing variable polarization of the deflecting force: the PolariX-TDS. By means of a comprehensive campaign of measurements conducted with the prototype, key parameters of the rf performance of the system are validated and a phase-space characterization of an electron bunch is accomplished with a time resolution of 3.3 fs. Furthermore, an analysis of second-order effects induced on the bunch from its passage through the PolariX-TDS is presented.

Immobilisation plays an important role in the industrial application of enzymes. The stabilisation and reusability of immobilised enzymes reduce the cost of the catalyst and facilitate their use in continuously operated reactors. For this purpose, an applicable type of immobilisation needs to be identified. In this study, we investigate the conversion of CDP and PolyP to CTP by NDP polyphosphate phosphotransferase 3 from Ruegeria pomeroyi (RpPPK2-3) and describe the covalent immobilisation of RpPPK2-3. In order to select a suitable carrier for the immobilisation of RpPPK2-3, a screening with different amino methacrylate (glutaraldehyde-pre-activated) and epoxy methacrylate carriers was carried out. The epoxy methacrylate carrier ECR8209M (Purolite®) was found to be the most suitable. With a half-life of 462 d when stored at 6 °C and a 50-fold reusability with a 93% residual activity, the immobilised enzyme showed a higher stability compared to the soluble enzyme with a half-life of 0.04 d. Although the half-life of the soluble enzyme could be increased to 32 d by adding PPi, it could not reach the stability of the immobilisate. Due to the resilience of the immobilisate, it is suitable for application in continuous reactor set-ups, e.g., packed-bed reactors.

The Accelerator Research Experiment at Sinbad (ARES) linac at DESY aims to generate and characterize ultrashort electron bunches (fs to sub-fs duration) with high momentum and arrival time stability for the purpose of applications related to accelerator research and development, e.g., development of advanced and compact diagnostics and accelerating structures, test of new accelerator components, medical applications studies, machine learning, etc. During its commissioning phase, the bunch duration characterization of the electron bunches generated at ARES has been performed with an rf-phasing technique relying on momentum spectra measurements, using only common accelerator elements (rf accelerating structures and magnetic spectrometers). The sensitivity of the method allowed highlighting different response times for Mo and Cs_{2}Te cathodes. The measured electron bunch duration in a wide range of machine parameters shows excellent agreement overall with the simulation predictions, thus demonstrating a very good understanding of the ARES operation on the bunch duration aspect. The importance of a precise in situ experimental determination of the phase velocity of the first traveling wave accelerating structure after the electron source, for which we propose a simple new beam-based method precise down to a variation of one part per ten thousand respective to the speed of light in vacuum, is emphasized for this purpose. A minimum bunch duration of 20 fs rms, resolution-limited by the space charge forces, is reported. This is, to the best of our knowledge, around 4 times shorter than what has been previously experimentally demonstrated based on rf-phasing techniques with a single rf structure. The present study constitutes a strong basis for future time characterization down to the sub-fs level at ARES, using dedicated X-band transverse deflecting structures.

Siberia experienced a prolonged heatwave in the spring of 2020, resulting in extreme summer drought and major wildfires in the North-Eastern Siberian lowland tundra. In the Arctic tundra, plants play a key role in regulating the summer land surface energy budget by contributing to land surface cooling through evapotranspiration. Yet we know little about how drought conditions impact land surface cooling by tundra plant communities, potentially contributing to high air temperatures through a positive plant-mediated feedback. Here we used high-resolution land surface temperature and vegetation maps based on drone imagery to determine the impact of an extreme summer drought on land surface cooling in the lowland tundra of North-Eastern Siberia. We found that land surface cooling differed strongly among plant communities between the drought year 2020 and the reference year 2021. Further, we observed a decrease in the normalized land surface cooling (measured as water deficit index) in the drought year 2020 across all plant communities. This indicates a shift towards an energy budget dominated by sensible heat fluxes, contributing to land surface warming. Overall, our findings suggest significant variation in land surface cooling among common Arctic plant communities in the North-Eastern Siberian lowland tundra and a pronounced effect of drought on all community types. Based on our results, we suggest discriminating between functional tundra plant communities when predicting the drought impacts on energy flux related processes such as land surface cooling, permafrost thaw and wildfires.
