A simple visual ethanol biosensor based on alcohol oxidase (AOX) immobilised onto polyaniline (PANI) film for halal verification of fermented beverage samples is described. This biosensor responds to ethanol via a colour change from green to blue, due to the enzymatic reaction of ethanol that produces acetaldehyde and hydrogen peroxide, when the latter oxidizes the PANI film. The procedure to obtain this biosensor consists of the immobilization of AOX onto PANI film by adsorption. For the immobilisation, an AOX solution is deposited on the PANI film and left at room temperature until dried (30 min). The biosensor was constructed as a dip stick for visual and simple use. The colour changes of the films have been scanned and analysed using image analysis software (i.e., ImageJ) to study the characteristics of the biosensor's response toward ethanol. The biosensor has a linear response in an ethanol concentration range of 0.01%-0.8%, with a correlation coefficient (r) of 0.996. The limit detection of the biosensor was 0.001%, with reproducibility (RSD) of 1.6% and a life time up to seven weeks when stored at 4 °C. The biosensor provides accurate results for ethanol determination in fermented drinks and was in good agreement with the standard method (gas chromatography) results. Thus, the biosensor could be used as a simple visual method for ethanol determination in fermented beverage samples that can be useful for Muslim community for halal verification. Background: A novel optical sensor for the rapid and direct determination of permethrin preservatives in treated wood was designed. The optical sensor was fabricated from the immobilisation of 2,6-dichloro-p-benzoquinone-4- chloroimide (Gibbs reagent) in nafion/sol-gel hybrid film and the mode of detection was based on absorption spectrophotometry. Physical entrapment was employed as a method of immobilisation. Results: The sensor gave a linear response range of permethrin between 2.56-383.00 μM with detection limit of 2.5 μM and demonstrated good repeatability with relative standard deviation (RSD) for 10 μM at 5.3%, 100 μM at 2.7%, and 200 μM at 1.8%. The response time of the sensor was 40 s with an optimum response at pH 11. Conclusions: The sensor was useful for rapid screening of wood or treated wood products before detailed analysis using tedious procedure is performed. The validation study of the optical sensor against standard method HPLC successfully showed that the permethrin sensor tended to overestimate the permethrin concentration determined. The knowledge of how fish survive and grow at different temperatures, and how these traits vary between life stages, is essential to evaluate the effects of climate change on wild fish and implement effective strategies in aquaculture. These issues are addressed in this study through a series of experiments that evaluate the effect of temperature (23–34 °C) on the embryos and larvae of clown loach, Chromobotia macracanthus. This species is endemic to the rivers of Sumatra and Borneo, highly praised on the ornamental fish market, and has been reproduced in captivity recently. No embryo survived a 24-h exposure to 34 °C until the age of 3 days after hatching (dah); mortality was high at 32 °C at 2 and 3 dah, whereas it was low and similar from 1 to 4 dah at 23–29 °C (<10%). Yolk absorption was proportional to water temperature (Q10°C of 1.69 in the 23–32 °C range), but fish reared at cold temperatures were larger than others at the start of exogenous feeding (5.7 vs. 5.5 mm TL, at 23 and 32 °C, respectively). The survival of larvae fed Artemia nauplii ad libitum was high at 23–32 °C (80–100%), but almost null at 34 °C. Growth models at different temperatures were produced from weekly measurements in two experiments, and tested by comparing their predictions with the results of a third experiment. Throughout the larval stage, the optimal temperature for growth (T°opt) was close to 29 °C, and departures from T°opt resulted in substantial growth penalties (–30% SGR for –5.1 °C and + 3.1 °C). High survival, fast growth (0.7 mm day-1) and limited size dispersal at T°opt are encouraging perspectives for the aquaculture of clown loach. From an ecological perspective, the species has an atypical thermal biology, as it is less thermophilic than other tropical fishes, but more stenothermal than temperate fishes exhibiting similar values of T°opt , both traits being of particular concern in the context of global warming. An amperometric electrochemical biosensor has been developed for ammonium (NH4 +) ion detection by immobilising alanine dehydrogenase (AlaDH) enzyme in a photocurable methacrylic membrane made up of poly(2-hydroxyethyl methacrylate) (pHEMA) on a screen-printed carbon paste electrode (SPE). The current detected was based on the electrocatalytic oxidation of nicotinamide adenine dinucleotide reduced (NADH) that is proportional to the consumption of NH4 + ion whilst enzymatic amination of AlaDH and pyruvate is taking place. The biosensor was operated amperometrically at a potential of +0.6V and optimum pH 7. The NH4 + biosensor demonstrated linear response to NH4 + ion concentration in the range of 0.03-1.02 mg/L with a limit of detection (LOD) of 8.52 μg/L. The proposed method has been successfully applied to the determination of NH4 + ion in river water samples without any pretreatment. The levels of possible interferents in the waters were negligible to cause any interference on the proposed method. The analytical performance of the biosensor was comparable to the colorimetric method using Nesslerisation but with much lower detection limit and linear response range at ppb level. The use of multilayer of gold nanoparticles (AuNPs) attached on gold electrode surface via thiol chemistry to fabricate an ammonium (NH4+) ion biosensor based on alanine dehydrogenase (AlaDH) was investigated. The approach of the study was based on construction of biosensor by direct deposition of AuNPs and 1,8-octanedithiol (C8-DT) onto the gold electrode surface. For the immobilisation of enzyme, 2-mercaptoethanol (2BME) was first covalently attached to AlaDH via esther bonding and then followed by chemically attached the 2BME-modified AlaDH (2BME-AlaDH) moiety onto the AuNPs electrode via the exposed thiol group of 2BME. The resulting biosensor response was examined by means of amperometry for the quantification of NH4+ ion. In the absence of enzyme attachment, the use of three layers of AuNPs was found to improve the electrochemistry of the gold electrode when compared with no AuNPs was coated. However, when more than three layers of AuNPs were coated, the electrode response deteriorated due to excessive deposition of C8-DT. When AlaDH was incoporated into the AuNPs modified electrode, a linear response to NH4+ ion over the concentration range of 0.1-0.5mM with a detection limit of 0.01mM was obtained. In the absence of AuNPs, the NH4+ ion biosensor did not exhibit any good linear response range although the current response was observed to be higher. This work demonstrated that the incorporation of AuNPs could lead to the detection of higher NH4+ ion concentration without the need of dilution for high NH4+ ion concentration samples with a rapid response time of <1 min. An artificial neural network (ANN) was applied for the determination of V(V) based on immobilized fatty hydroxamic acid (FHA) in poly(methyl methacrylate) (PMMA). Spectra obtained from the V(V)-FHA complex at single wavelengths was used as the input data for the ANN. The V(V)-FHA complex shows a limited linear dynamic range of V(V) concentration of 10 - 100 mg/L. After training with ANN, the linear dynamic range was extended with low calibration error. A three layer feed forward neural network using back-propagation (BP) algorithm was employed in this study. The input layer consisted of single neurons, 30 neurons in hidden a layer and one output neuron was found appropriate for the multivariate calibration used. The network were trained up to 10 000 epochs with 0.003% learning rate. This reagent also provided a good analytical performance with reproducibility characters of the method yielding relative standard deviation (RSD) of 9.29% and 7.09% for V(V) at concentrations of 50 mg/L and 200 mg/L, respectively. Abstract: An optical biosensor consisting of a chromoionophore (ETH5294) (CM) doped sol–gel film interfaced with another sol–gel film immobilized with acetylcholinesterase (AChE) was employed to detect the insecticide dichlorvos. The main advantage of this optical biosensor is the use of a sol–gel layer with immobilized CM that possesses lipophilic property. The highly lipophilic nature of the CM and its compatibility with the sol–gel matrix has prevented leaching, which is frequently a problem in optical sensor construction based on pH indicator dyes. The immobilization of the indicator and enzyme was simple and need no chemical modification. The CM layer is pH sensitive and detects the pH changes of the acetylcholine chloride (AChCl) substrate when hydrolyzed by AChE layer deposited above. In the absence of the AChE layer, the pH response of the CM layer is linear from pH 6 to 8 (R 2 =0.98, n =3) and it showed no leaching of the lipophilic chromoionophore. When the AChE layer is deposited on top, the optical biosensor responds to AChCl with a linear dynamic range of 40–90mM AChCl (R 2 =0.984, n =6). The response time of the biosensor is 12min. Based on the optimum incubation time of 15min, a linear calibration curve of dichlorvos against the percentage inhibition of AChE was obtained from 0.5 to 7mg/L of dichlorvos (17–85% inhibition, R 2 =0.991, n =9). The detection limit for dichlorvos was 0.5mg/L. The results of the analysis of 1.7–6.0mg/L of dichlorvos using this optical biosensor agreed well with a gas chromatography–mass spectrometry detection method. [Copyright &y& Elsevier]

The use of artificial neural networks (ANN) in optimizing salicylic acid (SA) determination is presented in this paper. A simple and rapid spectrophotometric method for salicylic acid (SA) determination was carried out based on the complexation of salicylic acid–ferric(III) nitrate, SAFe(III). The SA forms a stable purple complex with ferric(III) nitrate at pH 2.45. The useful dynamic linear range is 0.01–0.35 g/L. It has a maximum absorption at 524 nm and the stability is more than 50 hours. The results were used for artificial neural networks (ANNs) training to optimize data. For training and validation purposes, a back-propagation (BP) artificial neural network (ANN) was used. The results showed that ANN technique was very effective and useful in broadening the limited dynamic linear response range mentioned to an extensive calibration response (0.01–0.70 g/L). It was found that a network with 22 hidden neurons was highly accurate in predicting the determination of SA. This network scores a summation of squared error (SSE) skill and low average predicted error of 0.0078 and 0.00427 g/L, respectively. [ABSTRACT FROM AUTHOR]

Gallocynin immobilized in chitosan membrane has been studied as a sensor element of an optical sensor for lead using a flowing system. By using this set up, lead in solution has been determined in the concentration range from 1.0×10−1 to 1.0×103 ppm with a detection limit of 0.075 ppm. The standard deviation of the method for the repeatability of lead detection at a concentration of 100 ppm was found to be 2.10%. The response of the sensor was reproducible and can be regenerated by using acidified saturated KNO3 solution. Interference from foreign ions was also studied at 1:1 mole ratio of Pb(II):foreign ions. [Copyright &y& Elsevier]
