151. An observational, prospective, multicenter study on the utilization and effectiveness of elbasvir–grazoprevir treatment association for chronic hepatitis C in France (ZEPHYR study)

Abstract

Abstract Background and Aim The efficacy and safety profiles of elbasvir–grazoprevir (EBR/GZR) has been established in more than 10 clinical trials. However, the characteristics of patients treated in routine clinical practice may differ. The present study was therefore designed to assess the real‐life effectiveness of EBR/GZR therapy in the general population and among subgroups with a high hepatitis C virus (HCV) prevalence in France. Methods The Zephyr study was designed as a French, multicentre, prospective, observational study on EBR/GZR use and effectiveness in current practice in chronic hepatitis C patients. These results are based on data regarding the adult patients who received at least one dose of EBR/GZR between December 2017 and June 2019 in 67 French hospitals and clinics. Results Overall, 478 patients were included. The Full Analysis Set corresponded to the 467 patients who met all the inclusion criteria and none of the exclusion criteria. Gender was balanced and the mean age was 55.7 ± 13.3 years. The patients were mainly treatment‐naive (89.5%) and infected with Genotype 1b (70.4%). Among the 75 patients with HCV Gt1a genotype, 56% had HCV RNA ≥ 800,000 IU/ml. F3–F4 fibrosis stage involved 24.2% of our population. Our subgroups were distributed among 110 migrants (23.6%), 58 (15.3%) using opioid agonist treatment, including people who inject drugs, 30 (6.8%) with chronic kidney disease Stages 3–5, 9 (1.9%) with an inherited blood disorder, and 4 (0.9%) coinfected with HIV. The remaining 269 (58.7%) were included in the general population subgroup. Overall, sustained virologic response 12 weeks after the end of treatment reached 98.0% and remained consistent among genotype, HCV RNA values, fibrosis stage, and the subgroup of interest. The rate of Alcohol Use Disorders Identification Test‐Consumption​​​ and Life Habit questionnaire completion was high at each visit, with data suggesting alcohol consumption decrease and an improvement in quality of life. Conclusions Using real‐world evidence data on a French population representative of HCV patients, we confirmed the results obtained during EBR/GZR development program.

Abstract

The solution chemistry of a chelator developed for 227Th targeted alpha therapy was probed. The compound of interest is an octadentate ligand comprising four N-methyl-3-hydroxy-pyridine-2-one metal-binding units, two tertiary amine groups, and one carboxylate arm appended for bioconjugation. The seven p Ka values of the ligand and the stability constants of complexes formed with Th(IV), Hf(IV), Zr(IV), Gd(III), Eu(III), Al(III), and Fe(III) were determined. The ligand exhibits extreme thermodynamic selectivity toward tetravalent metal ions with a ca. 20 orders of magnitude difference between the formation constant of the Th(IV) species formed at physiological pH, namely [ThL]-, and that of its Eu(III) analogue. Likewise, log β110 values of 41.7 ± 0.3 and 26.9 ± 0.3 (T = 25 °C) were measured for [ThL]- and [FeIIIL]2-, respectively, highlighting the high affinity and selectivity of the ligand for Th ions over potentially competing endogenous metals. Single crystal X-ray analysis of the Fe(III) complex revealed a dinuclear 2:2 metal:chelator complex crystallizing in the space group P1̅. The formation of this dimeric species is likely favored by several intramolecular hydrogen bonds and the protonation state of the chelator in acidic media. LIII edge EXAFS data on the Th(IV) complexes of both the ligand and a monoclonal antibody conjugate revealed the expected mononuclear 1:1 metal:chelator coordination environment. This was also confirmed by high resolution mass spectrometry. Finally, kinetic experiments demonstrated that labeling the bioconjugated ligand with Th(IV) could be achieved and completed after 1 h at room temperature, reinforcing the high suitability of this chelator for 227Th targeted alpha therapy.

Abstract

Oxo group activation with reduction of neptunyl(vi) and plutonyl(vi) to tetravalent hydroxo species by the hydroxypyridinone siderophore derivative 3,4,3-LI-(1,2-HOPO) was investigated in the gas-phase via electrospray ionization mass spectrometry, in solution via Raman spectroscopy, and computationally via density functional theory. Dissociation of the gas-phase tetravalent complexes resulted in actinide-hydroxo bond cleavage.

Abstract

In order to reveal subtle differences between the solution chemistries of trivalent 4f and 5f elements, the physicochemical and photophysical properties of europium(iii), terbium(iii) and curium(iii) complexes formed with a 7-methoxy-coumarin appended 1,4,7,10-tetraazadodecane-1,4,7-triacid (DO3A) ligand were studied. All three complexes were found to be kinetically inert and exhibit stability constants similar to their 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) equivalents. The Cm(iii) and Eu(iii) complexes feature strong sensitised emission, while the triplet energy of the coumarin prohibits efficient sensitisation of the Tb(iii) analogue. The data presented here indicate significant differences in perturbation of the sensitising chromophore photophysics between the 4f and 5f elements. In contrast, the size of the metal center appears to not be a determining factor for the physicochemical properties of these kinetically inert Eu(iii), Tb(iii), and Cm(iii) complexes.

Abstract

The hydroxypyridinone ligand 3,4,3-LI(1,2-HOPO) is a promising agent for biological decorporation of radionuclides, and allows spectroscopic detection of many lanthanide (Ln) and actinide (An) species via sensitized luminescence. Despite the manifest uses of this ligand, the structural and thermodynamic properties of its complexes across the An series remain understudied. Theoretical investigations of the binding of An(III) and An(IV) ions, from actinium to einsteinium, by the 3,4,3-LI(1,2-HOPO) ligand, as well as experimental extended X-ray absorption fine structure (EXAFS) studies on the trivalent americium, curium, and californium complexes were employed to address the resulting structures, thermodynamic parameters, redox properties, and corresponding electronic configurations. An(IV) ions were found to form much stronger complexes than An(III) ions, consistent with experimental measurements. Complexation of both An(III) and An(IV) ions generally becomes more favorable for heavier actinides, reflecting increased energy degeneracy driven covalency and concomitant orbital mixing between the 5f orbitals of the An ions and the π orbitals of the ligand. Notably, the ability of this ligand to either accept or donate electron density as needed from its pyridine rings is found to be key to its extraordinary stability across the actinide series.

The chemistry of trivalent transplutonium ions (Am3+ , Cm3+ , Bk3+ , Cf3+ , Es3+ …) is usually perceived as monotonic and paralleling that of the trivalent lanthanide series. Herein, we present the first extended X-ray absorption fine structure (EXAFS) study performed on a series of aqueous heavy actinide chelates, extending past Cm. The results obtained on diethylenetriaminepentaacetic acid (DTPA) complexes of trivalent Am, Cm, Bk, and Cf show a break to much shorter metal-oxygen nearest-neighbor bond lengths in the case of Cf3+ . Corroborating those results, density functional theory calculations, extended to Es3+ , suggest that the shorter Cf-O and Es-O bonds could arise from the departure of the coordinated water molecule and contraction of the ligand around the metal relative to the other [MIII DTPA(H2 O)]2- (M=Am, Cm, Bk) complexes. Taken together, these experimental and theoretical results demonstrate inhomogeneity within the trivalent transplutonium series that has been insinuated and debated in recent years, and that may also be leveraged for future nuclear waste reprocessing technologies.

Abstract

Several MRI contrast agent clinical formulations are now known to leave deposits of the heavy metal gadolinium in the brain, bones, and other organs of patients. This persistent biological accumulation of gadolinium has been recently recognized as a deleterious outcome in patients administered Gd-based contrast agents (GBCAs) for MRI, prompting the European Medicines Agency to recommend discontinuing the use of over half of the GBCAs currently approved for clinical applications. To address this problem, we find that the orally-available metal decorporation agent 3,4,3-LI(1,2-HOPO) demonstrates superior efficacy at chelating and removing Gd from the body compared to diethylenetriaminepentaacetic acid, a ligand commonly used in the United States in the GBCA Gadopentetate (Magnevist). Using the radiotracer 153Gd to obtain precise biodistribution data, the results herein, supported by speciation simulations, suggest that the prophylactic or post-hoc therapeutic use of 3,4,3-LI(1,2-HOPO) may provide a means to mitigate Gd retention in patients requiring contrast-enhanced MRI.

Abstract

The toxicity of heavy metals such as lead (Pb), cadmium (Cd) and tin (Sn) has long been known but accidental exposures of large populations to these elements remain unfortunately a topical issue. Chelating agents against Pb, Cd, or Sn poisoning are still limited to classical ligands such as ethylenediamine tetraacetic acid (EDTA). Here, we evaluate the ability of 3,4,3-LI(1,2-HOPO) (L4-), an octadentate ligand currently under evaluation for actinide decorporation, to bind Pb, Cd and Sn metal ions in aqueous solutions. This ligand forms 1:1 complexes with Pb(ii), Cd(ii) and Sn(iv) as well as bimetallic 2:1 species with Pb(ii) and Cd(ii), all of which were characterized by high resolution mass spectrometry and spectrophotometric titrations. The 3,4,3-LI(1,2-HOPO) ligand exhibits an extreme affinity for Sn(iv) ions (logβ110 > 40), with the complex remaining stable from highly acidic conditions to alkaline media (from 3 M HCl to pH 8). Single crystals of the neutral complex [SnIV3,4,3-LI(1,2-HOPO)·3H2O] were obtained and its structure determined, revealing a chiral conformation. Although not initially designed for hexacoordinated metals such as Pb(ii) and Cd(ii), the octadentate ligand shows promise for the in vitro or in vivo sequestration of toxic heavy metals, as evidenced by decorporation experiments performed in mice contaminated with 210Pb(ii) and treated with either EDTA or 3,4,3-LI(1,2-HOPO).

Abstract

Targeted alpha therapy is an oncological treatment, where cytotoxic doses of alpha radiation are locally delivered to tumor cells, while the surrounding healthy tissue is minimally affected. This therapeutic strategy relies on radiopharmaceuticals made of medically relevant radionuclides chelated by ligands, and conjugated to targeting vectors, which promote the drug accumulation in tumor sites. This review discusses the state-of-the-art in the development of radiopharmaceuticals for targeted alpha therapy, breaking down their key structural components, such as radioisotope, targeting vector, and delivery formulation, and analyzing their pros and cons. Moreover, we discuss current drawbacks that are holding back targeted alpha therapy in the clinic, and identify ongoing strategies in field to overcome those issues, including radioisotope encapsulation in nanoformulations to prevent the release of the daughters. Lastly, we critically discuss potential opportunities the field holds, which may contribute to targeted alpha therapy becoming a gold standard treatment in oncology in the future.

Abstract

We theoretically predict that a true bilayer exciton condensate,characterised by off-diagonal long range order and global phase coherence, can be created in one-dimensional solid state electron systems. The mechanism by which this happens is to introduce a single particle hybridization of electron and hole populations, which locks the phase of the relevant mode and hence invalidates the Mermin--Wagner theorem. Electron--hole interactions then amplify this tendency towards off-diagonal long range order, enhancing the condensate properties by more than an order of magnitude over the noninteracting limit. Abstract

The abundance of CH+ and OH and excitation are predicted to be enhanced by the presence of vibrationally excited H2 or hot gas (~500-1000 K) in PDRs with high incident FUV radiation field. The excitation may also originate in dense gas (>10^5 cm-3) followed by nonreactive collisions. Previous observations suggest that the CH+ and OH correlate with dense and warm gas, and formation pumping contributes to CH+ excitation. We examine the spatial distribution of the CH+ and OH emission in the Orion Bar to establish their physical origin and main formation and excitation mechanisms. We present spatially sampled maps of the CH+ J=3-2 transition at 119.8 {\mu}m and the OH {\Lambda}-doublet at 84 {\mu}m in the Orion Bar over an area of 110"x110" with Herschel (PACS). We compare the spatial distribution of these molecules with those of their chemical precursors, C+, O and H2, and tracers of warm and dense gas. We assess the spatial variation of CH+ J=2-1 velocity-resolved line profile observed with Herschel (HIFI). The OH and CH+ lines correlate well with the high-J CO emission and delineate the warm and dense molecular region. While similar, the differences in the CH+ and OH morphologies indicate that CH+ formation and excitation are related to the observed vibrationally excited H2. This indicates that formation pumping contributes to the excitation of CH+. Interestingly, the peak of the rotationally excited OH 84 {\mu}m emission coincides with a bright young object, proplyd 244-440, which shows that OH can be an excellent tracer of UV-irradiated dense gas. The spatial distribution of CH+ and OH revealed in our maps is consistent with previous modeling studies. Both formation pumping and nonreactive collisions in a UV-irradiated dense gas are important CH+ J=3-2 excitation processes. Abstract

Astrophysical images issued from different instruments and/or spectral bands often require to be processed together, either for fitting or comparison purposes. However each image is affected by an instrumental response, also known as PSF, that depends on the characteristics of the instrument as well as the wavelength and the observing strategy. Given the knowledge of the PSF in each band, a straightforward way of processing images is to homogenise them all to a target PSF using convolution kernels, so that they appear as if they had been acquired by the same instrument. We propose an algorithm that generates such PSF-matching kernels, based on Wiener filtering with a tunable regularisation parameter. This method ensures all anisotropic features in the PSFs to be taken into account. We compare our method to existing procedures using measured Herschel/PACS and SPIRE PSFs and simulated JWST/MIRI PSFs. Significant gains up to two orders of magnitude are obtained with respect to the use of kernels computed assuming Gaussian or circularised PSFs., Comment: 7 pages, 6 figure. 178. Robustness of topologically protected transport in graphene-boron nitride lateral heterostructures

Abstract

Previously, graphene nanoribbons set in lateral heterostructures with hexagonal boron nitride were predicted to support topologically protected states at low energy. We investigate how robust the transport properties of these states are against lattice disorder. We find that forms of disorder that do not couple the two valleys of the zigzag graphene nanoribbon do not impact the transport properties at low bias, indicating that these lateral heterostructures are very promising candidates for chip-scale conducting interconnects. Forms of disorder that do couple the two valleys, such as vacancies in the graphene ribbon, or substantial inclusions of armchair edges at the graphene-hexagonal boron nitride interface will negatively affect the transport. Abstract

Topological states of matter are characterized by global topological invariants which change their value across a topological quantum phase transition. It is commonly assumed that the transition between topologically distinct noninteracting gapped phases of fermions is necessarily accompanied by the closing of the band gap as long as the symmetries of the system are maintained. We show that such a quantum phase transition is possible without closing the gap in the case of a three-dimensional topological band insulator. We demonstrate this by calculating the free energy of the minimal model for a topological insulator, the Bernevig-Hughes-Zhang model, and show that as the band curvature continuously varies, a jump between the band gap minima corresponding to the topologically trivial and nontrivial insulators occurs. Therefore, this first order phase transition is a generic feature of three-dimensional topological band insulators. For a certain parameter range we predict a re-entrant topological phase transition. Abstract

Aims. We investigate the spatial distribution of a collection of absorbing gas clouds, some associated with the dense, massive star-forming core NGC6334 I, and others with diffuse foreground clouds. For the former category, we aim to study the dynamical properties of the clouds in order to assess their potential to feed the accreting protostellar cores. Methods. We use spectral imaging from the Herschel SPIRE iFTS to construct a map of HF absorption at 243 micron in a 6x3.5 arcmin region surrounding NGC6334 I and I(N). Results. The combination of new, spatially fully sampled, but spectrally unresolved mapping with a previous, single-pointing, spectrally resolved HF signature yields a 3D picture of absorbing gas clouds in the direction of NGC6334. Toward core I, the HF equivalent width matches that of the spectrally resolved observation. The distribution of HF absorption is consistent with three of the seven components being associated with this dense star-forming envelope. For two of the remaining four components, our data suggest that these clouds are spatially associated with the larger scale filamentary star-forming complex. Our data also implies a lack of gas phase HF in the envelope of core I(N). Using a simple description of adsorption onto and desorption from dust grain surfaces, we show that the overall lower temperature of the envelope of source I(N) is consistent with freeze-out of HF, while it remains in the gas phase in source I. Conclusions. We use the HF molecule as a tracer of column density in diffuse gas (n(H) ~ 10^2 - 10^3 cm^-3), and find that it may uniquely trace a relatively low density portion of the gas reservoir available for star formation that otherwise escapes detection. At higher densities prevailing in protostellar envelopes (>10^4 cm^-3), we find evidence of HF depletion from the gas phase under sufficiently cold conditions., Comment: 14 pages, 10 figures. The aim of this paper is to compare the performances of the optimal strategy under parameters mis-specification and of a technical analysis trading strategy. The setting we consider is that of a stochastic asset price model where the trend follows an unobservable Ornstein-Uhlenbeck process. For both strategies, we provide the asymptotic expectation of the logarithmic return as a function of the model parameters. Finally, numerical examples find that an investment strategy using the cross moving averages rule is more robust than the optimal strategy under parameters mis-specification.

Abstract

Dust grains play a key role in the physics of star-forming regions, even though they constitute only $\sim$1 % of the mass of the interstellar medium. The derivation of accurate dust parameters such as temperature ($T_{dust}$), emissivity spectral index ($\beta$) and column density requires broadband continuum observations at far-infrared wavelengths. We present Herschel-SPIRE Fourier Transform Spectrometer (FTS) measurements of three starless cores: L1521E, L1521F and L1689B, covering wavelengths between 194 and 671 $\mu$m. This paper is the first to use our recently updated SPIRE-FTS intensity calibration, yielding a direct match with SPIRE photometer measurements of extended sources. In addition, we carefully assess the validity of calibration schemes depending on source extent and on the strength of background emission. The broadband far-infrared spectra for all three sources peak near 250 $\mu$m. Our observations therefore provide much tighter constraints on the spectral energy distribution (SED) shape than measurements that do not probe the SED peak. The spectra are fitted using modified blackbody functions, allowing both $T_{dust}$ and $\beta$ to vary as free parameters. This yields $T_{dust}$ of 9.8$\pm$0.2 K, 15.6$\pm$0.5 K and 10.9$\pm$0.2 K and corresponding $\beta$ of 2.6$\mp$0.9, 0.8$\mp$0.1 and 2.4$\mp$0.8 for L1521E, L1521F and L1689B respectively. The derived core masses are 1.0$\pm$0.1, 0.10$\pm$0.01 and 0.49$\pm$0.05 $M_{\odot}$, respectively. The core mass/Jeans mass ratios for L1521E and L1689B exceed unity indicating that they are unstable to gravitational collapse, and thus pre-stellar cores. Abstract

The Draco nebula is a high Galactic latitude interstellar cloud likely to have been formed by the collision of a Galactic halo cloud entering the disk of the Milky Way. Such conditions are ideal to study the formation of cold and dense gas in colliding flows of warm gas. We present Herschel-SPIRE observations that reveal the fragmented structure of the interface between the infalling cloud and the Galactic layer. This front is characterized by a Rayleigh-Taylor instability structure. From the determination of the typical length of the periodic structure (2.2 pc) we estimated the gas kinematic viscosity and the turbulence dissipation scale (0.1 pc) that is compatible with that expected if ambipolar diffusion is the main mechanism of energy dissipation in the WNM. The small-scale structures of the nebula are typical of that seen in some molecular clouds. The gas density has a log-normal distribution with an average value of $10^3$ cm$^{-3}$. The size of the structures is 0.1-0.2 pc but this estimate is limited by the resolution of the observations. The mass ranges from 0.2 to 20 M$_{\odot}$ and the distribution of the more massive clumps follows a power law $dN/d\log(M) \sim M^{-1.4}$. We identify a mass-size relation with the same exponent as that found in GMCs ($M\sim L^{2.3}$) but only 15% of the mass of the cloud is in gravitationally bound structures. We conclude that the increase of pressure in the collision is strong enough to trigger the WNM-CNM transition caused by the interplay between turbulence and thermal instability as self-gravity is not dominating the dynamics., Comment: 16 pages, A&A, in press

Abstract

The rat hepatic stellate cell line PAV-1 was established two decades ago and proposed as a cellular model to study aspects of hepatic retinoic acid metabolism. This cell line exhibits a myofibroblast-like phenotype but also has the ability to store retinyl esters and synthesize retinoic acid from its precursor retinol. Importantly, when cultured with palmitic acid alone or in combination with retinol, the cells switch to a deactivated phenotype in which the proliferation and expression of profibrogenic marker genes are suppressed. Despite these interesting characteristics, the cell line has somehow fallen into oblivion. However, based on the fact that working with in vivo models is becoming increasingly complicated, genetically characterized established cell lines that mimic aspects of hepatic stellate cell biology are of fundamental value for biomedical research. To genetically characterize PAV-1 cells, we performed karyotype analysis using conventional chromosome analysis and multicolor spectral karyotyping (SKY), which allowed us to identify numerical and specific chromosomal alteration in PAV-1 cells. In addition, we used a panel of 31 species-specific allelic variant sites to define a unique short tandem repeat (STR) profile for this cell line and performed bulk mRNA-sequencing, showing that PAV-1 cells express an abundance of genes specific for the proposed myofibroblastic phenotype. Finally, we used Rhodamine-Phalloidin staining and electron microscopy analysis, which showed that PAV-1 cells contain a robust intracellular network of filamentous actin and process typical ultrastructural features of hepatic stellate cells.

In recent decades, cultural heritage survey practices have significantly evolved due to the increasing use of digitization tools providing quick and easy access to faithful copies of study objects. While these digital data have clear advantages, especially in terms of geometric characterization, they also introduce a paradigm shift by outsourcing ex situ most of the analysis activities. This break between real and virtual working environments now raises new issues, both in terms of data dispersion and knowledge correlation in multidisciplinary teams. Benefiting from the fields of information systems and augmented reality, we proposed an integrated approach allowing the fusion of geometric, visual and semantic features in a single platform. Today, this proof of concept leads to new perspectives for the production of semantically enriched digital data. Abstract

Recent efforts to activate the strong uranium-oxygen bonds in the dioxo uranyl cation have been limited to single oxo-group activation through either uranyl reduction and functionalization in solution, or by collision induced dissociation (CID) in the gas-phase, using mass spectrometry (MS). Here, we report and investigate the surprising double activation of uranyl by an organic ligand, 3,4,3-LI(CAM), leading to the formation of a formal U6+ chelate in the gas-phase. The cleavage of both uranyl oxo bonds was experimentally evidenced by CID, using deuterium and 18O isotopic substitutions, and by infrared multiple photon dissociation (IRMPD) spectroscopy. Density functional theory (DFT) computations predict that the overall reaction requires only 132 kJ/mol, with the first oxygen activation entailing about 107 kJ/mol. Combined with analysis of similar, but unreactive ligands, these results shed light on the chelation-driven mechanism of uranyl oxo bond cleavage, demonstrating its dependence on the presence of ligand hydroxyl protons available for direct interactions with the uranyl oxygens.

Abstract

The potential consequences of a major radiological event are not only large-scale external radiation exposure of the population, but also uncontrolled dissemination of, and internal contamination with, radionuclides. When planning an emergency response to radiological and nuclear incidents, one must consider the need for not only post-exposure treatment for contaminated individuals, but also prophylactic measures to protect the workforce facing contaminated areas and patients in the aftermath of such events. In addition to meeting the desired criteria for post-exposure treatments such as safety, ease of administration, and broad-spectrum efficacy against multiple radionuclides and levels of challenge, ideal prophylactic countermeasures must include rapid onset; induce minimal to no performance-decrementing side effects; be compatible with current military Chemical, Biological, Radiological, Nuclear, and Explosive countermeasures; and require minimal logistical burdens. Hydroxypyridinone-based actinide decorporation agents have shown the most promise as decorporation strategies for various radionuclides of concern, including the actinides plutonium and americium. The studies presented here probe the extent of plutonium decorporation efficacy for two chelating agents, 3,4,3-LI(1,2-HOPO) and 5-LIO(Me-3,2-HOPO), from early pre-exposure time points to a delay of up to 7 days in parenteral or oral treatment administration, i.e., well beyond the initial hours of emergency response. Despite delayed treatment after a contamination event, both ligands clearly enhanced plutonium elimination through the investigated 7-day post-treatment period. In addition, a remarkable prophylactic efficacy was revealed for 3,4,3-LI(1,2-HOPO) with treatment as early as 48 h before the plutonium challenge. This work provides new perspectives in the indication and use of experimental actinide decorporation treatments.

Abstract

Berkelium (Bk) has been predicted to be the only transplutonium element able to exhibit both +III and +IV oxidation states in solution, but evidence of a stable oxidized Bk chelate has so far remained elusive. Here we describe the stabilization of the heaviest 4+ ion of the periodic table, under mild aqueous conditions, using a siderophore derivative. The resulting Bk(IV) complex exhibits luminescence via sensitization through an intramolecular antenna effect. This neutral Bk(IV) coordination compound is not sequestered by the protein siderocalin-a mammalian metal transporter-in contrast to the negatively charged species obtained with neighbouring trivalent actinides americium, curium and californium (Cf). The corresponding Cf(III)-ligand-protein ternary adduct was characterized by X-ray diffraction analysis. Combined with theoretical predictions, these data add significant insight to the field of transplutonium chemistry, and may lead to innovative Bk separation and purification processes.

Abstract

Most low-mass stars form in stellar clusters that also contain massive stars, which are sources of far-ultraviolet (FUV) radiation. Theoretical models predict that this FUV radiation produces photodissociation regions (PDRs) on the surfaces of protoplanetary disks around low-mass stars, which affects planet formation within the disks. We report James Webb Space Telescope and Atacama Large Millimeter Array observations of a FUV-irradiated protoplanetary disk in the Orion Nebula. Emission lines are detected from the PDR; modeling their kinematics and excitation allowed us to constrain the physical conditions within the gas. We quantified the mass-loss rate induced by the FUV irradiation and found that it is sufficient to remove gas from the disk in less than a million years. This is rapid enough to affect giant planet formation in the disk.

Abstract

Mimivirus is the prototype of the Mimiviridae family of giant dsDNA viruses. Little is known about the organization of the 1.2 Mb genome inside the membrane-limited nucleoid filling the ~0.5 µm icosahedral capsids. Cryo-electron microscopy, cryo-electron tomography, and proteomics revealed that it is encased into a ~30-nm diameter helical protein shell surprisingly composed of two GMC-type oxidoreductases, which also form the glycosylated fibrils decorating the capsid. The genome is arranged in 5- or 6-start left-handed super-helices, with each DNA-strand lining the central channel. This luminal channel of the nucleoprotein fiber is wide enough to accommodate oxidative stress proteins and RNA polymerase subunits identified by proteomics. Such elegant supramolecular organization would represent a remarkable evolutionary strategy for packaging and protecting the genome, in a state ready for immediate transcription upon unwinding in the host cytoplasm. The parsimonious use of the same protein in two unrelated substructures of the virion is unexpected for a giant virus with thousand genes at its disposal.
