Abstract

SUMMARY: Undaria pinnatifida, the brown macroalga, is a major commercial edible seaweed, and there is increasing interest in breeding new, improved cultivars for market expansion. In this study, we attempted to establish mutagenesis in U. pinnatifida using Ar and C ion beams as mutagens to meet future demands. To screen irradiated generations for mutants, U. pinnatifida zoospores irradiated with Ar and C ion beams were cultivated in plastic Petri dishes. Some gametophytes derived from the irradiated zoospores showed growth arrest or cell death at the initial developmental stage. Although the growth inhibition and lethal effects were observed at high doses of each ion irradiation, the Ar ion irradiation had high biological effects on cell division and growth. The gametophytes that showed a reduction in cell elongation were designated as an inhibited cell elongation mutant. A comparison of the mutant induction frequencies revealed that the C ion beam showed a higher frequency than the Ar ion beam. The highest frequency was 0.83% at 12.5 Gy of the C ion beam. We determined the total number of sporophytes and embryos per female gametophyte after sporophyte induction. High‐dose irradiation with the Ar ion beam decreased the embryo and sporophyte formation, suggesting that the Ar ion beam also has exhibited high biological effects on the fertilization or embryogenesis processes or both. The developed heavy ion mutagenesis and mutant screening methods would be useful for mutation breeding and constructing specific mutant libraries in brown algae, and not only in U. pinnatifida. Abstract

ABSTRACTChalky grains of rice are increased due to high temperature (HT) during a ripening period. However, the underlying mechanisms of the chalkiness are not well known, seemingly due to the obtuse response of wild type and lack of effective mutants. In this study, we isolated and characterized the flo11-2mutant, which showed higher number of chalky grains than wild type under HT but quite small number of chalkiness under cool temperature as well as similar growth, development, and yield to that of the wild type. Using this high sensitivity of the flo11-2mutant, we identified the most critical meteorological factor and developmental stage affecting chalkiness with 5 days HT treatments over 4 consecutive years. The results demonstrated that daily maximum temperature was more causative than daily mean or minimum temperatures which have been regarded as important factors before this. Besides, the developmental stage around 20 days after flowering (DAF) was most sensitive to HT rather than the early stage up to 15 DAF. In addition, we found that the flo11-2mutant with a high chalky ratio was vulnerable to preharvest sprouting, which has never been reported before for chalky grains, but could cause significant yield and quality loss after extremely hot and dry summers followed by rainy cool autumns. Abstract

RIKEN RI Beam Factory is one of the world-leading heavy-ion facilities where nuclear physics research and application research have been conducted. One important application, performed more than twenty years at RIBF, is breeding plants and others where energetic heavy ions are used as very effective mutagens. Our previous main accelerator RIKEN Ring Cyclotron has been used for this purpose but available beam energies and resultant ion ranges are limited for medium heavy ions such as argon and iron ions. Hence, a new beam transport line delivering beams accelerated by one of our new-generation cyclotrons, RIKEN Intermediate Ring Cyclotron, to E5H experimental vault, where biological samples are irradiated in air, has been constructed. We will report design of the new beam line and the results obtained by the beam-commissioning test performed using a 160 MeV/nucleon argon beam in January 2015. We obtained performances as designed, for example, nearly 100% transmission efficiency, and a threefold increase of ion ranges. Abstract

We previously used heavy-ion beam irradiation to generate a self-incompatible mutant of Verbena×hybrida ‘Temari Coral Pink’ (SC), which exhibits a late-acting self-incompatibility system. In the present study, the behavior of pollen tubes and seed productivity after self-pollination were comparatively investigated in SC and wild species Verbena peruviana (VP), one of the parental species of Verbena×hybrida. Although reciprocal cross-pollination between VP and SC produced seeds with high frequencies, namely 57.1% for SC×VP and 59.7% for VP×SC, self-pollinated VP flowers produced no seeds. In the latter, almost all of the pollen germinated on the stigma, but further growth of the pollen tube was inhibited at the upper part of the style. These observations of pollen tube behavior may indicate that the SI system of VP was different from that of SC. VP may possess a homomorphic gametophytic SI system, which is characterized by the inhibition of pollen tube growth at style. We propose a set of SC and VP as a novel model plants for genetic analysis of the SI mechanism in the Verbenaceae family.

Abstract

A poly (3-hydroxybutyrate) (PHB) depolymerase was purified 4, 400-fold from the supernatant fraction of Ralstonia eutropha cells through fractionation with ammonium sulfate and chromatography with Toyopearl DEAE, Q Sepharose, and Toyopearl Ether. The partially purified preparation showed a high level of specific activity to hydrolyze amorphous PHB, comparable to PhaZd from R. eutropha. The degradation was inhibited by diisopropylphosphate (10mM) and dithiothreitol (100mM). Most of the degradation products were 3-hydroxybutyrate oligomers. The properties of the enzyme closely resembled those of PhaZd, but some differences were observed. This distinction from PhaZd was discussed.

Abstract

Ralstonia eutropha H16 mobilizes intracellularly accumulated poly(3-hydroxybutyrate) with intracellular poly(3-hydroxybutyrate) depolymerases. Different types of depolymerase, PhaZ1 and its homologs, account for the mobilization significantly but incompletely, and it is proposed that further unknown depolymerases or related enzymes complement the mobilization. A novel hydrolase was therefore investigated. The amino acid sequence of the intracellular 3HB-oligomer hydrolase of Acidovoraxsp. SA1 and the genome sequence of Ralstonia solanacearum, a close relative of R. eutropha, revealed a candidate for the hydrolase gene. The DNA sequence of its hydrolase was used for the design of a pair of primes. With chromosomal DNA of R. eutropha as a template, these primers were used to amplify about a 1.3-kbp fragment including the equivalent gene, phaZc. Then the phaZc gene was cloned by Southern hybridization using the amplified DNA fragment as a probe. The hydrolase (PhaZc) was purified from Escherichia coli overexpressing phaZc and characterized. Immunoblot analysis revealed that PhaZc exists mainly in the cytosolic fraction of R. eutropha. PhaZc degraded various 3-hydroxybutyrate-oligomers at a high rate and artificial amorphous poly (3-hydroxybutyrate) at a lower rate. A phaZ-rmLl mutation enhanced the deposition of PHB in the logarithmic phase in a nutrient-rich medium. PhaZc differs from the previously reported hydrolases in R. eutropha and is a novel type of intracellular 3HB-oligomer hydrolase, which participates in the mobilization of PHB along with other hydrolases.

183. Effects of Ion Irradiation on Seedlings Growth Monitored by Ultraweak Delayed Luminescence

Abstract

The optical technique based on the measurement of delayed luminescence emitted from the biological samples has demonstrated its ability to provide valid and predictive information on the functional status of various biological systems. We want to extend this technique to study the effect of ionizing radiation on biological systems. In particular we are interested in the action of ion beams, used for therapeutic purposes or to increase the biological diversity. In general, the assessment of the damage that radiation produces both in the target objects and in the surrounding tissues, requires considerable time because is based on biochemical analysis or on the examination of the evolution of the irradiated systems. The delayed luminescence technique could help to simplify this investigation. We have so started our studies performing irradiations of some relatively simple vegetable models. In this paper we report results obtained from mung bean (Vigna radiata) seeds submitted to a 12C ion beam at the energy of 62 MeV/nucleon. The dry seeds were irradiated at doses from 50 to 7000 Gy. The photoinduced delayed luminescence of each seed before and after ion irradiation was measured. The growth of seedlings after irradiation was compared with that of untreated seeds. A growth reduction on increasing the dose was registered. The results show strong correlations between the ion irradiation dose, seeds growth and delayed luminescence intensity. In particular, the delayed luminescence intensity is correlated by a logistic function to the seedlings elongation and, after performing a suitable measurement campaign based on blind tests, it could become a tool able to predict the growth of seeds after ion irradiation. Moreover these results demonstrate that measurements of delayed luminescence could be used as a fast and non-invasive technique to check the effects of ion beams on relatively simple biological systems.

199. Possibility of polyploidy breeding using cryptic species in the marine crop <italic>Pyropia yezoensis</italic> (Bangiales, Rhodophyta).

Abstract

In the marine crop Pyropia yezoensis, a cross-fertilization experiment between a cultivated strain and cryptic species was carried out to investigate the possibility of polyploidy breeding using cryptic species. To perform the cross-fertilization, we isolated a green mutant strain (IBYC-G1) of the cryptic species by heavy-ion beam irradiation. The green mutant was crossed with a wildtype strain (HG-4) of P. yezoensis f. narawaensis. The F1 heterozygous conchocelis developed normally and released numerous conchospores. However, almost all the conchospore germlings did not survive past the four-cell stage or thereabout, and a few germlings developed into gametophytic blades. These results indicated that reproductive isolation occurred during meiosis and that P. yezoensis and cryptic species were different species using the biological species concept. Almost all the archeospores that were released from the surviving F1 blades developed into normal gametophytic blades. The result of nuclear DNA marker analysis revealed that the F1 blades that developed from both conchospores and archeospores were allodiploids in the haploid gametophytic phase. Although their F1 blades were extremely slender, their blade color was a deeper reddish brown as compared with that of P. yezoensis f. narawaensis, and the rate of blade length increase was remarkably higher in the blades of F1 than in those of the cryptic species. Abstract

Tricholoma matsutake is an ectomycorrhizal agaricomycete that produces the prized mushroom 'matsutake' in Pinaceae forests. Currently, there are no available cultivars or cultivation methods that produce fruiting bodies. Heavy-ion beams, which induce mutations through double-stranded DNA breaks, have been used widely for plant breeding. In the present study, we examined whether heavy-ion beams could be useful in isolating T. matsutake mutants. An argon-ion beam gave a suitable lethality curve in relation to irradiation doses, accelerating killing at 100-150 Gy. Argon-ion beam irradiation of the agar plate cultures yielded several transient mutants whose colony morphologies differed from that of the wild-type strain at the first screening, but which did not persist following culture transfer. It also generated a mutant whose phenotype remained stable after repeated culture transfers. The stable pleiotropic mutant not only exhibited a different colony morphology to the wild type, but also showed increased degradation of dye-linked water-insoluble amylose and cellulose substrates. Thus, heavy-ion beams may be useful for isolating mutants of T. matsutake, although precautions may be required to maintain the mutants, without phenotypic reversion, during repetitive culture of their mycelia.
