Abstract

Nowadays, there is an increasing need to understand the behaviour of geotechnical structures during earthquakes. The damage caused by the recent earthquakes has shown that the local geology and the geotechnical characteristics of the foundation soil can influence significantly the seismic response of structures. So, in order to correctly predict the behaviour of a structure subjected to an earthquake, it is necessary to focus attention on the dynamic soil behaviour. In general, very simple soil constitutive models are implemented in commercial codes. Several studies have shown that when shear strains in the soil are small, it is possible to use the elastic-linear model; for medium strains it is convenient to use equivalent linear or nonlinear models. Elastic-plastic models, incrementally nonlinear models or hypoplastic models can more accurately capture response for sites that experience higher strains. A recent elasto-plastic constitutive model including both isotropic and kinematic hardening has been implemented in a FEM code. The numerical results achieved by the new version of the FEM code, are discussed and validated by means of the comparison with laboratory experimental results involving sands of different relative densities. An interesting parametric analysis is also presented, in order to investigate the effects of the implemented constitutive model parameter variation in the soil cyclic behaviour. Abstract

This paper describes analytical and experimental research into the structure and properties of the near wake of generic shapes at supersonic flight conditions. Three axisymmetric model configurations were constructed and tested in the aeroballistic research facility at Eglin Air Force Base, Florida at Mach numbers from 3 to 5. Laser interferograms were taken of the near wake of the configurations at sea level conditions. Computational interferograms were then developed at identical conditions and were correlated against the experimental data. This correlation will help calibrate high speed CFD codes in this complex flow region. Keywords: Aeroballistics, Near wake; Recirculation zone; Interferogram.

196. Effects of vascular risk factors on balance assessed by computerized posturography in the elderly.

Abstract

AIMS: The purpose of this study was to evaluate the effects of vascular risk factors, isolated or in association, on balance, as assessed by posturographic platform. METHODS: One hundred and seven elderly subjects (mean age 73.8+/-5.8), with no cognitive impairment (MMSE>24), able to perform self-care activities and to walk independently for at least 400 meters, free from major diseases. Subjective complaints were assessed by means of the Sickness Impact Profile Questionnaire. The following cardiovascular risk factors were considered: hypercholesterolemia, smoking, hypertension, glucose intolerance, and obesity. Balance tests were performed in three standardized positions (side-by-side, semi-tandem, tandem) on a vertical force platform, from which center of foot pressure positions and displacements were recorded. RESULTS: A large percentage of enrolled subjects (35.5%) complained of unsteadiness, dizziness or vertigo, but only a few (24=22.4%) reported at least one fall in the 6 months before enrolment in the study. Among the cardiovascular risk factors taken into account, only glucose intolerance and, to a lesser extent, obesity, were associated with worse performance in stabilometric tests, independent of age and sex. Subjects with 3 or more risk factors, compared with those with 2 or less showed worse performance in medio-lateral sway (p=0.001), track length (p=0.05) and elliptical area (p=0.005), in tandem position. CONCLUSIONS: The cumulative presence of cardiovascular risk factors may contribute to impairment of balance in the elderly. Abstract

Background: One strategy for improving anti-tuberculosis (TB) vaccination involves the use of recombinant bacille Calmette-Guérin (rBCG) overexpressing protective TB antigens. rBCG30, which overexpresses the Mycobacterium tuberculosis secreted antigen Ag85b, was the first rBCG shown to induce significantly greater protection against TB in animals than parental BCG.Methods: We report here the first double-blind phase 1 trial of rBCG30 in 35 adults randomized to receive either rBCG30 or parental Tice BCG intradermally. Clinical reactogenicity was assessed, and state-of-the-art immunological assays were used to study Ag85b-specific immune responses induced by both vaccines.Results: Similar clinical reactogenicity occurred with both vaccines. rBCG30 induced significantly increased Ag85b-specific T cell lymphoproliferation, interferon (IFN)-gamma secretion, IFN-gamma enzyme-linked immunospot responses, and direct ex vivo intracellular IFN-gamma responses. Additional flow cytometry studies measuring carboxyfluorescein succinimidyl ester dilution and intracellular cytokine production demonstrated that rBCG30 significantly enhanced the population of Ag85b-specific CD4(+) and CD8(+) T cells capable of concurrent expansion and effector function. More importantly, rBCG30 significantly increased the number of Ag85b-specific T cells capable of inhibiting intracellular mycobacteria.Conclusions: These results provide proof of principal that rBCG can safely enhance human TB immunity and support further development of rBCG overexpressing Ag85b for TB vaccination.
