ABSTRACT High-temperature stress is critical for all organisms and induces a profound cellular response. For Crenarchaeota, little information is available on how heat shock affects cellular processes and on how this response is regulated. We set out to study heat shock response in the thermoacidophilic model crenarchaeon Sulfolobus acidocaldarius, which thrives in volcanic hot springs and has an optimal growth temperature of 75°C. Pulse-labeling experiments demonstrated that a temperature shift to 86°C induces a drastic reduction of the transcriptional and translational activity, but that RNA and protein neosynthesis still occurs. By combining RNA sequencing and mass spectrometry, an integrated mapping of the transcriptome and proteome was performed. This revealed that heat shock causes an immediate change in the gene expression profile, with RNA levels of half of the genes being affected, followed by a more subtle reprogramming of the protein landscape. Functional enrichment analysis indicated that nearly all cellular processes are affected by heat shock. A limited correlation was observed in the differential expression on the RNA and protein level, suggesting a prevalence of post-transcriptional and post-translational regulation. Furthermore, promoter sequence analysis of heat shock regulon genes demonstrated the conservation of strong transcription initiation elements for highly induced genes, but an absence of a conserved protein-binding motif. It is, therefore, hypothesized that histone-lacking archaea such as Sulfolobales use an evolutionarily ancient regulatory mechanism that relies on temperature-responsive changes in DNA organization and compaction induced by the action of nucleoid-associated proteins, as well as on enhanced recruitment of initiation factors. IMPORTANCE Heat shock response is the ability to respond adequately to sudden temperature increases that could be harmful for cellular survival and fitness. It is crucial for microorganisms living in volcanic hot springs that are characterized by high temperatures and large temperature fluctuations. In this study, we investigated how S. acidocaldarius, which grows optimally at 75°C, responds to heat shock by altering its gene expression and protein production processes. We shed light on which cellular processes are affected by heat shock and propose a hypothesis on underlying regulatory mechanisms. This work is not only relevant for the organism’s lifestyle, but also with regard to its evolutionary status. Indeed, S. acidocaldarius belongs to the archaea, an ancient group of microbes that is more closely related to eukaryotes than to bacteria. Our study thus also contributes to a better understanding of the early evolution of heat shock response.

We report on the neutrino mass measurement result from the first four-week science run of the Karlsruhe Tritium Neutrino experiment KATRIN in spring 2019. Beta-decay electrons from a high-purity gaseous molecular tritium source are energy analyzed by a high-resolution MAC-E filter. A fit of the integrated electron spectrum over a narrow interval around the kinematic end point at 18.57 keV gives an effective neutrino mass square value of (-1.0_{-1.1}^{+0.9})  eV^{2}. From this, we derive an upper limit of 1.1 eV (90% confidence level) on the absolute mass scale of neutrinos. This value coincides with the KATRIN sensitivity. It improves upon previous mass limits from kinematic measurements by almost a factor of 2 and provides model-independent input to cosmological studies of structure formation.

The KArlsruhe TRItium Neutrino (KATRIN) experiment aims to make a model-independent determination of the effective electron antineutrino mass with a sensitivity of 0.2 eV/c 2 . It investigates the kinematics of β-particles from tritium β-decay close to the endpoint of the energy spectrum. Because the KATRIN main spectrometer (MS) is located above ground, muon-induced backgrounds are of particular concern. Coincidence measurements with the MS and a scintillator-based muon detector system confirmed the model of secondary electron production by cosmic-ray muons inside the MS. Correlation measurements with the same setup showed that about 12% of secondary electrons emitted from the inner surface are induced by cosmic-ray muons, with approximately one secondary electron produced for every 17 muon crossings. However, the magnetic and electrostatic shielding of the MS is able to efficiently suppress these electrons, and we find that muons are responsible for less than 17% (90% confidence level) of the overall MS background.

Remotely piloted aircrafts (RPAs) are becoming well-known for monitoring forest restoration (FR). Canopy cover percentage on a restoration site is a major indicator for the early phases of FR. We present a protocol to evaluate canopy cover using low-cost RPA and a friendly computational environment. We tested the protocol in six FR projects in the Brazilian Atlantic Forest. After processing the images automatically to generate the digital surface model and the digital terrain model, the canopy cover is measured in QGIS by considering height thresholds in the canopy height model. The results had F1 values higher than 0.9, which confirms a robust methodology that does not require specific computer science or remote sensing skills. Thus, low-cost RPA proved to be accurate and user-friendly in measuring the canopy cover, which improves monitoring during most FR stages. Such finding plays an important role in FR policies and future works must study a user-friendly protocol for measuring tree height via low-cost drones.

The article analyses work on click farm platforms in Brazil and Colombia. It argues that work on these platforms updates and renews the historical informality of work in Latin America. Drawing on click farm ethnography, worker interviews and digital ethnography on WhatsApp and Facebook groups and Youtube channels, the research highlights: first, the cultural marks of Brazil and Colombia in the interactions between workers, typical of Latin American digital culture; second, the role of Youtubers as skill makers, responsible for the initiation of workers into click farm platforms and the circulation of neoliberal and entrepreneurial ideology; third, practices and discourses relating to reselling accounts, photos and bots as a new version of the historical resale markets in the region; and fourth, the boundaries between informality and illegality at work on click farm platforms. The article argues that, in addition to informal work that preceded and is connected to work on click farms, informality gains new dimensions with work on click farms, with the platformisation of labour representing an articulation between the old informality and new market practices and infrastructures.

BackgroundType 2 Diabetes Mellitus (T2DM) is a major risk factor for the development of cardiometabolic diseases. T2DM prevention is largely based on weight-loss and whole diet changes, but intervention with dietary plant bioactives may also improve metabolic health.ObjectiveTo assess whether supplementation with bilberry and grape seed extract for 12 weeks improves cardiometabolic outcomes in individuals at risk of developing T2DM, and to determine whether individual treatment response is associated with differences in gut microbiota composition and levels of phenolic metabolites in blood and feces.MethodsIn the randomized, double-blind, placebo-controlled, cross-over PRECISE intervention study, 14 participants, aged ≥45 years, with a BMI >28 kg/m2, and having an increased risk of T2DM, received a supplement containing 250 mg of bilberry plus 300 mg of grape seed extract, or 550 mg of a control extract, per day, for 12 weeks each. Blood samples were obtained for the assessment of HbA1c, fasting glucose, oral glucose tolerance tests, insulin, glucagon levels, total, LDL and HDL cholesterol, and phenolic acids. We also assessed advanced glycation end products in the skin, ambulatory 24 hours blood pressure, 7-day dietary intake by weighed food diaries, fecal levels of phenolic metabolites using LC–MS/MS and gut microbiota composition using 16S rRNA gene sequencing analysis.ResultsThe combined bilberry and grape seed extract did not affect glucose and cholesterol outcomes, but it decreased systolic and diastolic ambulatory blood pressure by 4.7 (p

Value chains that generate an increased willingness to pay among consumers by offering enhanced levels of sustainability are widely discussed as an important strategy for creating new business opportunities and fostering food system transformation. Previous research has highlighted the importance of governance arrangements to secure the trust necessary for the establishment of sustainability-based value chains. However, how different coordination designs by private and public actors along the value chain affect trust formation is not well understood. To address this question, this paper combines the concepts of hybrid governance and multidimensional trust to guide a comparative analysis of five sustainability-based agri-food value chains in Germany as exemplary case studies. The findings show that different types of governance activities are necessary to build the capacity to address four different sources of trust: dispositional, affinitive, rational and procedural trust. Building trust capacities facilitates coordination of activities along the value chain and reliable delivery of sustainability-related value propositions. In all five cases, governance arrangements and building of trust capacities were geared towards increasing willingness to pay. Sustainability-based agri-food value chains have therefore limited potential to internalize the agri-food sector’s substantial negative externalities.

Investments in "Clean water and sanitation" drive a transition from irrigation with untreated to irrigation with treated wastewater. While this transition reduces many health risks, it may decrease crop yields, and soil carbon storage, cause a release of accumulated pollutants from soils, and increase the spread of antibiotic resistance in the environment. This article analyzes how Marxist platform-dependent cultural producers in Brazil - one woman and two drag queens - navigate platform spaces in relation to their identities and everyday work. Struggling with platforms refers to the specific ways in which Marxist cultural producers seek to engage in class struggles, while also grappling with the constraints imposed by platforms. There are paradoxes, tensions and frictions in these efforts. In analytical terms, the article develops how this 'struggling with platforms' occurs in two main dimensions: (1) struggling with identities – including the implications of being a Marxist cultural producer on platforms and how one's identity is shaped and commodified in this space, and (2) struggling with everyday work – considering how the pursuit of social change is both constrained and enabled on platforms. Simple Summary: For young patients with the rare entity primary mediastinal B-cell lymphoma, a differentiated description of effects and side effects of the different therapeutic opportunities is of immense interest. We therefore conducted a planning study to compare two different radiation therapy planning variants for a cohort of these patients who were treated in a hospital cancer center. We estimated normal tissue complication probabilities for radiation side effects for the heart, the lungs and the esophagus. Introduction: Primary mediastinal B-cell lymphoma (PMBCL) is a rare form of aggressive B-cell lymphoma with a predominant onset in young patients. The minimization of potential (late) side effects is of cardinal interest for these patients. An anticipation of the individual risk profile is desirable to counsel the patient on the putative impact of radiotherapy (RT). Methods: RT plans for a cohort of 25 patients with PMBCL were prospectively designed. One plan with two parallel- opposing fields (APPA) and another with volume-modulated arc therapy (VMAT) technique with 40 Gy in 2 Gy fractions each. Normal The normal tissue complication probability (NTCP) was calculated using the Lyman-–Kutcher-–Burman model for heart, lung and oesophageal toxicity. Results: APPA planning resulted in lower median doses (Dmedian) for the heart and lungs, whereas all other dose metrics for heart, lungs and esophagus were lower in VMAT planning. A significant difference in the mean NTCPs when comparing the APPA to VMAT plans was seen for increased cardiac mortality, pneumonitis and esophagitis. PTV size correlated with increased cardiac mortality and esophagitis in both plan variations and with pneumonitis for VMAT plans. Dmean, Dmedian, and V20Gy correlated with the risk for pneumonitis, and Dmean, Dmedian, and V1% with the risk for esophagitis in both variants. Conclusions: We showed decreased risk of different NTCPs for VMAT and APPA planning for thoracic toxicities. The use of an IMRT technique like VMAT showed advantages for several DVH metrics in organs at risk and should therefore be recommended for radiation treatment of PMBCL. [ABSTRACT FROM AUTHOR]
