151. In-vitro activity of cefiderocol against multidrug-resistant Enterobacterales, Pseudomonas aeruginosa and Acinetobacter baumannii isolates from the UK

Abstract

Background. Cefiderocol is a parenteral siderophore cephalosporin, with a catechol-containing 3’ side chain. We evaluated its activity against MDR Gram-negative bacteria Methods. MICs of cefiderocol, meropenem, ceftolozane-tazobactam, cefepime, ceftazidime, aztreonam, ciprofloxacin, ceftazidime-avibactam, amikacin and colistin were determined in cation-adjusted Mueller-Hinton broth; the medium was iron depleted for cefiderocol only. The panel comprised 305 Enterobacterales, 111 P. aeruginosa and 99 A. baumannii selected for carbapenemases, ESBL production or carbapenem resistance via combinations of porin-loss with AmpC or ESBL. Results. The activity of cefiderocol was unrelated to Enterobacterales species. At 4mg/L cefiderocol inhibited 92.1% of all Enterobacterales, with rates of 95-100% for isolates with AmpC+porin-loss, VIM, IMP, OXA-48-like, KPC, GES, SME or IMI. Only isolates with NDM (72.1%) or ESBL+porin-loss (88.5%) had lower rates. No comparator agent inhibited >90% of isolates at EUCAST breakpoint. Cefiderocol 4mg/L inhibited 86.5% of all P. aeruginosa, with rates of 80-100% for those with VIM, IMP, GES or VEB beta-lactamases. Lower rates were seen for those with NDM (72.7%) and PER (73.3%) enzymes. No comparator inhibited >85% of isolates at breakpoint. Cefiderocol 4mg/L also inhibited 88.9% of A. baumannii isolates with rates >85% for those with OXA-51, -23, -24, or -58. A lower rate (80%) was seen for those with NDM carbapenemases. A concentration of 16mg/L was needed to inhibit ≥90% of A. baumannii. Conclusions. Cefiderocol was widely active at low concentrations against MDR Enterobacterales, P. aeruginosa and A. baumannii. MICs for isolates with NDM enzymes nonetheless were higher than for those with other carbapenemases.

Abstract

In this paper, an inverse characterization method is proposed for measuring the flow resistivity of rigid porous materials via transmitted waves at very low frequencies. Equivalent fluid theory context is considered. Two transmission coefficient expressions have been established in the Darcy regime; the first one depends on the frequency, porosity and resistivity, while the second one is independent of the frequency and depends only on the flow resistivity. The inverse problem is solved in frequency domain. The inverted values of the resistivity are determined using the two low frequency expressions of the transmission coefficients. Tests are performed using two samples of industrial plastic foam commonly used in thermal and acoustic insulation. The reliability of the obtained results is discussed and compared with those given by classical methods.

Abstract

The present study investigates the contribution of the stochasticity of a layered soil deposit on the seismic ground motion spatial variability via the random vibration theory. The seismic response evaluation is based on the 1-D shear wave propagation through random media. The soil layers stochasticity is introduced as spatial random fields of mass density and shear modulus of the soil deposit. The stochastic variation of these two parameters results in random natural frequency and damping ratio of an equivalent single degree of freedom oscillator modelling the soil deposit. Under the assumption of stationary Gaussian random process of the incident motion, the ground surface response spectra are obtained from the cross power spectral density of the total ground surface motion. The efficiency and the validity of the present approach are conducted via some applications. Numerical results showed that, the shape of the global coherency is controlled by that of the incident motion. The stochastic aspect of the soil layers causes disturbances in the surface motion and affects the coherency and the variability of the ground surface response spectra.

Abstract

In this paper, an acoustic method is presented for measuring the porosity and the viscous tortuosity of air-saturated-porous materials at low frequencies. The proposed method is based on a temporal model of the direct and inverse problem for the reflection of low-frequency waves by homogeneous isotropic slab of porous material having a rigid frame. Reflected coefficient for a slab of porous material is derived from the responses of the medium to an incident acoustic pulse where a simple relation between flow resistivity, porosity, viscous tortuosity and the reflected waves is obtained. A numerical method and efficient tool for the estimation of the porosity and the viscous tortuosity are presented and discussed.

Abstract

International audience; Aerogel Pd/(Ce0.33Zr0.66O2)SiO2 catalysts (CeZry) were prepared with variable Ce and Zr loadings (molar ratio Ce/Zr = 1/2) by combining sol-gel and impregnation methods. First, N2-physisorption was used to investigate the texture evolution. Then, H2-chimisorption and TEM were performed to study the effect on particle dispersion. After, TPR was used to determine the catalyst reducibility. Furthermore, XPS characterization was done to identify the palladium oxidation state and to evaluate the Pd-support interaction. Finally, the prepared catalysts were tested in methane combustion to assess their catalytic activity. The obtained results showed that, when the Zr and Ce loadings are varied between 0% and 8% and between 0% and 6% respectively, the BET surface area was increased from 615 to 744 m2/g, the porosity diameter from 45.7 to 83.6 Å, the Pd particle diameter from 5.2 to 7.0 nm, the CeO2 and ZrO2 particle size from 0 to 68 nm, the reduction temperature shift reached 16°C, the Pd binding energy shift attained 0.6 eV, but an optimum amounts of Zr (4 wt.%) and Ce (3 wt.%) are needed to maximize the PdO reducibility and to enhance the catalytic activity. In effect, 100% conversion of methane was reached at around 415°C on the CeZr4 catalyst.

Abstract

The acoustic characterization of porous materials with rigid structure plays an important role in the prediction of the acoustic behavior of these media. In order to obtain an accurate prediction of its response as a function of frequency, several parametric models have been proposed. However, semi-phenomenological models are the most used to describe the acoustic behavior of porous materials with rigid or flexible structures according to a certain number of macroscopic physical properties related to the internal structure of the material and essential for their valuation. Among these models, the one proposed by Johnson et al. to describes the complex density of an acoustic porous material with an immobile skeleton having arbitrary pore shapes. The objective of this work is to study the effect of these parameters involved in Johnson's model as well as other new parameters, recently introduced by Sadouki [ Phys. Fluids 33, (2021)), on the transmitted signal in the low-frequency regime of ultrasound.

Abstract

In this work, a simulated study on the effects of physical and mechanical parameters describing human cancellous bone on reflected ultrasound waves is proposed. The visco-inertial exchanges between the structure and the saturating fluid are described by the Biot theory modified by the Johnson model. A simplified expression of the reflection coefficient at the first interface is obtained in the frequency domain. This expression depends on three physical parameters which are porosity, tortuosity, and viscous characteristic length as well as the mechanical parameters; Young's modulus and Poisson's ratio. The reflected signal is calculated in the frequency domain by the product of the spectrum of the incident signal with the reflection coefficient. In the time domain, the reflected wave is obtained by taking the inverse Fourier transform of the reflected frequency signal. The effect of each parameter on the reflected signal is studied. The results obtained are discussed and compared with those given in the literature.

Abstract

An acoustic reflectivity method is proposed for measuring the permeability or flow resistivity of air-saturated porous materials. In this method, a simplified expression of the reflection coefficient is derived in the Darcy's regime (low frequency range), which does not depend on frequency and porosity. Numerical simulations show that the reflection coefficient of a porous material can be approximated by its simplified expression obtained from its Taylor development to the first order. This approximation is good especially for resistive materials (of low permeability) and for the lower frequencies. The permeability is reconstructed by solving the inverse problem using waves reflected by plastic foam samples, at different frequency bandwidths in the Darcy regime. The proposed method has the advantage of being simple compared to the conventional methods that use experimental reflected data, and is complementary to the transmissivity method, which is more adapted to low resistive materials (high permeability). Abstract

An improved acoustic method is proposed for measuring the tortuosity and the viscous and thermal characteristic lengths of air-saturated porous materials via low-frequency ultrasonic transmitted waves (70 kHz–110 kHz). The equivalent fluid model is considered. The interaction between the saturated fluid and the structure is taken into account in two frequency response factors: the dynamic tortuosity of the medium and the dynamic compressibility of the air which are described by their high-frequency expansion in powers of the viscous and thermal skin depth, either limiting the expansion to the first two leading terms or the first three leading terms with the introduction of two new viscous and thermal shape factors. These two new factors play an important role in stabilizing and improving the inverted parameters of the tortuosity and the viscous and thermal characteristic lengths in the frequency band considered. The inverse problem is solved numerically in the frequency domain by minimizing the spectrum of simulated and experimental transmitted signals. Optimized parameters of tortuosity, viscous and thermal characteristic lengths, and viscous and thermal shape factors are obtained simultaneously. The tests are carried out with three samples of plastic foam. The results obtained by the two procedures are discussed and compared to those given in the literature.

Abstract

A numerical approach, named TransChlor, is proposed to simulate transport phenomena of various substances in concrete. This approach is a theoretical model based on finite elements and finite differences methods. The model consists of coupled nonlinear partial differential equations based on Fick's diffusion law and on kinematics equations. Simulation results from a parametrical study highlight the influence of microclimatic conditions, exposure to deicing salts and concrete cover permeability and thickness on chloride ingress in concrete. The results show that the chloride ion concentration increases quickly in concrete cover when a structure is exposed to deicing salts at a mountainous location; whereas permeability of concrete cover is an insignificant parameter when the concrete is in direct or splash water contact.

In this paper, an inverse characterization method is proposed for measuring the flow resistivity of rigid porous materials via transmitted waves at very low frequencies. Equivalent fluid theory context is considered. Two transmission coefficient expressions have been established in the Darcy regime; the first one depends on the frequency, porosity and resistivity, while the second one is independent of the frequency and depends only on the flow resistivity. The inverse problem is solved in frequency domain. The inverted values of the resistivity are determined using the two low frequency expressions of the transmission coefficients. Tests are performed using two samples of industrial plastic foam commonly used in thermal and acoustic insulation. Abstract

Objective Monocyte Chemoattractant Protein-1 (MCP-1/CCL2), a key player in immune-mediated responses against Mycobacterium tuberculosis, is encoded by a polymorphic gene. Functionally relevant polymorphic variations in the MCP-1 gene have been associated with both susceptibility to and protection against tuberculosis-related disorders. Here, we investigated the potential impact of some of these polymorphisms on Pott’s disease risk in a patient cohort from Algeria. Methods DNA from 132 Algerian patients with exclusive Pott’s disease and 204 healthy controls, included under a case-control design, were analyzed for the MCP1 -2518A/G (rs1024611), -362G/C (rs2857656) and int1del554-567 (rs3917887) polymorphisms. PHASE software was used for haplotype reconstruction. Genetic associations were examined using chi-square tests. Results We found that the rs1024611 -2518 GG, rs2857656 -362 CC and rs3917887 int1del554-567 del/del homozygous genotypes each were significantly more prevalent in patients than in controls (respective corrected p value [Pc] = 0.01, 0.04 and 0.04) Haplotype distribution profile further confirmed this, as the homozygous combination of GCdel haplotype was also found with raised susceptibility to Pott’s disease (Pc = 0.03). Conclusion Our findings confirm and replicate the recent data from China (which dealt essentially with rs1024611 and rs2857656) and also reinforce them by providing trans-ethnic evidence and extending the genetic association to the rs3917887.
